A complete library of poly(2‐oxazoline) block copolymers was synthesized via cationic ring opening polymerization for the characterization by two different soft ionization techniques, namely matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI‐TOF MS) and electrospray ionization quadrupole time‐of‐flight mass spectrometry (ESI‐Q‐TOF MS). In addition, a detailed characterization was performed by tandem MS to gain more structural information about the block copolymer composition and its fragmentation behavior. The fragmentation of the poly(2‐oxazoline) block copolymers revealed the desired polymer structure and possible side reactions, which could be explained by different mechanisms, like 1,4‐ethylene or hydrogen elimination and the McLafferty +1 rearrangement. Polymers with aryl side groups showed less fragmentation due to their higher stability compared to polymers with alkyl side groups. These insights represent a further step toward the construction of a library with fragments and their fragmentation pathways for synthetic polymers, following the successful examples in proteomics. © 2010 Wiley Periodicals, Inc. Fingerprinting of the degradation product patterns by electrospray ionization mass spectrometry (ESI‐MS) was evaluated as a tool to monitor the degree of degradation in polyester‐ether networks. Four different crosslinked caprolactone (CL) and/or 1,5‐dioxepan‐2‐one (DXO) networks were subjected to hydrolytic degradation in aqueous solution at 37 °C for up to 147 days. After predetermined time periods, the water‐soluble degradation products were analyzed by ESI‐MS and tandem ESI‐MS. In addition, changes in pH, mass loss, and copolymer composition were determined. In the case of more slowly hydrolyzed CL‐rich (co)polymers, CL and/or DXO oligomers terminated by hydroxyl and carboxyl end groups were predominantly formed as degradation products. However, on prolonged hydrolysis oligomers with attached crosslinking agent dominated the degradation product patterns of more easily hydrolyzed DXO‐rich (co)polymers. It was shown that in the recorded mass spectra the variation of intensities in the series of ions corresponding to DXO and CL/DXO oligomers with or without attached crosslinking agent could be utilized to monitor the extent of hydrolytic degradation in the polyester matrix and the disruption of the network structure. © 2008 Wiley Periodicals, Inc. A new two‐step synthesis of polyisobutylene (PIB) with precisely one thymine functionality per chain (PIB‐T) is reported. The primary hydroxyl‐functionalized PIB (PIB‐OH) precursor was prepared by direct functionalization via living carbocationic polymerization of isobutylene initiated by the α‐methylstyrene epoxide/TiCl4 system. Matrix assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI‐ToF MS) of a low molecular weight PIB‐OH precursor demonstrated the effectiveness of direct functionalization by this method. A PIB‐acrylate precursor (PIB‐Ac) was obtained from such a PIB‐OH, and the PIB‐T was subsequently prepared by Michael addition of thymine across the acrylate double bond. Summary: Biodegradable poly(1,5‐dioxepan‐2‐one) (PDXO) was grown directly from Si OH groups of a silica nanoparticle by surface‐initiated, ring‐opening polymerization (SI‐ROP) of 1,5‐dioxepan‐2‐one (DXO). The direct SI‐ROP of DXO was achieved by heating a mixture of Sn(Oct)2, DXO, and the silica nanoparticles (316 nm in diameter) in anhydrous toluene. The resulting silica/PDXO hybrid nanoparticles were characterized by means of 1H NMR spectroscopy, IR spectroscopy, thermogravimetric analysis, and field‐emission scanning electron microscopy.

Morphology is presented as a powerful tool to control the in vitro degradation and drug release characteristics of novel drug delivery microspheres prepared from homopolymer blends of 1,5‐dioxepan‐2‐one, DXO, and L ‐lactide, L‐LA. Their performance in this respect was compared to analogous P(L‐LA‐co‐DXO) microspheres. Blends formed denser and less porous microspheres with a higher degree of matrix crystallinity than copolymers of corresponding L‐LA:DXO composition. The morphology differences of blends and copolymers, further adjustable by means of component ratio, are shown to have a vital impact on the in vitro performance. Sustained drug delivery was obtained from both copolymers and blends. Molecular weight loss was retarded and diffusion‐mediated release was inhibited in the latter case, further delaying the release process. The effects of storage on the physicochemical properties of these systems were evaluated under desiccated and moist conditions for 5 months. Storage‐induced physicochemical changes, such as matrix crystallization and molecular weight decrease, were accelerated at higher relative humidities. P(L‐LA‐co‐DXO) demonstrated higher moisture sensitivity than a PLLA‐PDXO blend of corresponding composition. The more crystalline and dense morphology of blend microspheres may thus be considered an improvement of the storage stability. © 2000 John Wiley & Sons, Inc. The molar mass determination of block copolymers, in particular amphiphilic block copolymers, has been challenging with chromatographic techniques. Therefore, methoxy poly(ethylene glycol)‐b‐poly(styrene) (mPEG‐b‐PS) was synthesized by atom transfer radical polymerization (ATRP) and characterized in detail not only by conventional chromatographic techniques, such as size exclusion chromatography (SEC), but also by matrix‐assisted laser/desorption ionization tandem mass spectrometry (MALDI‐TOF MS/MS). As expected, different molar mass values were obtained in the SEC measurements depending on the calibration standards (either PEG or PS). In contrast, MALDI‐TOF MS/MS analysis allowed the molar mass determination of each block, by the scission of the weakest point between the PEG and PS block. Thus, fragments of the individual blocks could be obtained. The PEG block showed a depolymerization reaction, while for the PS block fragments were obtained in the monomeric, dimeric, and trimeric regions as a result of multiple chain scissions. The block length of PEG and PS could be calculated from the fragments recorded in the MALDI‐TOF MS/MS spectrum. Furthermore, the assignment of the substructures of the individual blocks acquired by MALDI‐TOF MS/MS was accomplished with the help of the fragments that were obtained from the corresponding homopolymers. © 2010 Wiley Periodicals, Inc. High‐resolution matrix‐assisted laser desorption/ionization (MALDI) time‐of‐flight mass spectrometry (TOF MS) was used for the analysis of the low‐molecular‐weight products from the photo‐oxidation of poly(3‐hexylthiophene) (P3HT) in solution and thin film. Eight new peak series were observed in the low‐mass range of the mass spectra of the products degraded in solution, and the formulas of the eight components were determined from the accurate mass. From SEC/MALDI‐TOF MS, two components were identified as the degraded products, and the other six components were derived from the fragmentation of the degraded products during the MALDI process. A mechanism for the formation of these components was proposed on the basis of the results of MALDI‐TOF MS. For the thin film degradation, a part of products in the solution degradation were observed, which supports that the oxidation of P3HT in solution and thin film proceeded in the same mechanism. This study shows that high‐resolution MALDI‐TOF MS is effective for the analysis of the low‐molecular‐weight products from P3HT photo‐oxidation and expected to be feasible for the degradation analyses of other polymers. To predict long‐term polymer behavior during a nuclear waste storage time, radiation effects on a segmented aromatic poly(ether‐urethane) induced by high‐energy radiation under oxygen atmosphere were investigated. To obtain a predictive model of polymer radio‐oxidation during several centuries, the first step consists to elaborate the elementary degradation mechanisms. Thus, electron paramagnetic resonance (EPR), Fourier transform infra‐red spectroscopy (FT‐IR), electrospray ionisation‐mass spectrometry (ESI‐MS), and gas mass spectrometry were carried out to identify radicals, chemical modifications, and gases to reach the radio‐oxidative mechanism at doses inferior than 1000 kGy. Degradation mainly occurs at urethane bonds and in polyether soft segments that produces stable oxidative products as formates, alcohols, carboxylic acids and H2, CO2 and CO gases. Predominant degradation occurred at polyether soft segments and crosslinking is in competition with scission. To avoid organometallic catalysts in the synthesis of poly(1,5‐dioxepan‐2‐one), the enzymatic ring‐opening polymerization of 1,5‐dioxepan‐2‐one (DXO) was performed with lipase CA (derived from Candida antarctica) as a biocatalyst. A linear relationship between the number‐average molecular weight and monomer conversion was observed, and this suggested that the product molecular weight could be controlled by the stoichiometry of the reactants. The monomer consumption followed a first‐order rate law with respect to the monomer, and no chain termination occurred. Water acted as a chain initiator, but it could cause polymer hydrolysis when it exceeded an optimum level. An initial activation via the heating of the enzyme was sufficient to start the polymerization, as the monomer conversion occurred when samples were left at room temperature after an initial heating at 60 °C. A high lipase content led to a high monomer conversion as well as a high molecular weight. An increase in the monomer conversion and molecular weight was observed when the polymerization temperature was increased from 40 to 80 °C. A further increase in the polymerization temperature led to a decrease in the monomer conversion and molecular weight because of the denaturation of the enzyme at elevated temperatures. The polymerization behavior of DXO under lipase CA catalysis was compared with that of ε‐caprolactone (CL). The rate of monomer conversion of DXO was much faster than that of CL, and this may have been due to differences in their specificity toward lipase CA. © 2005 Wiley Periodicals, Inc. In order to investigate gas‐phase fragmentation reactions of phosphorylated peptide ions, matrix‐assisted laser desorption/ionization (MALDI) and electrospray ionization (ESI) tandem mass (MS/MS) spectra were recorded from synthetic phosphopeptides and from phosphopeptides isolated from natural sources. MALDI‐TOF/TOF (TOF: time‐of‐flight) spectra of synthetic arginine‐containing phosphopeptides revealed a significant increase of y ions resulting from bond cleavages on the C‐terminal side of phosphothreonine or phosphoserine. The same effect was found in ESI‐MS/MS spectra recorded from the singly charged but not from the doubly charged ions of these phosphopeptides. ESI‐MS/MS spectra of doubly charged phosphopeptides containing two arginine residues support the following general fragmentation rule: Increased amide bond cleavage on the C‐terminal side of phosphorylated serines or threonines mainly occurs in peptide ions which do not contain mobile protons. In MALDI‐TOF/TOF spectra of phosphopeptides displaying N‐terminal fragment ions, abundant b–H3PO4 ions resulting from the enhanced dissociation of the pSer/pThr–X bond were detected (X denotes amino acids). Cleavages at phosphoamino acids were found to be particularly predominant in spectra of phosphopeptides containing pSer/pThr–Pro bonds. A quantitative evaluation of a larger set of MALDI‐TOF/TOF spectra recorded from phosphopeptides indicated that phosphoserine residues in arginine‐containing peptides increase the signal intensities of the respective y ions by almost a factor of 3. A less pronounced cleavage‐enhancing effect was observed in some lysine‐containing phosphopeptides without arginine. The proposed peptide fragmentation pathways involve a nucleophilic attack by phosphate oxygen on the carbon center of the peptide backbone amide, which eventually leads to cleavage of the amide bond. New star‐shaped and photocrosslinked poly(1,5‐dioxepan‐2‐one) (PDXO) has been synthesized through ring‐opening polymerization initiated by SnOct2/pentaerythritol. The star‐shaped PDXO was end‐functionalized by acrolyol chloride to form acrylate end groups. The end‐functionalized PDXO was photocrosslinked initiated by 2,2‐dimethoxy‐2‐phenylacetophenone. The gel content ranged from 80 to 99%, indicating a high degree of crosslinking. The thermal properties of the star‐shaped PDXO and the photocrosslinked PDXO were analyzed by differential scanning calorimetry. The glass‐transition temperature was determined to approximately ?32 °C for the crosslinked PDXO. The viscosity numbers were determined for star‐shaped PDXO, with reference to linear homologues. The star‐shaped PDXO had lower viscosity numbers than the linear counterparts. The crosslinked PDXO showed a rather hydrophilic surface as compared with other resorbable polyesters. New amphiphilic triblock copoly(2‐oxazoline)s, containing hydrophobic domains with fluorine‐containing blocks, were synthesized. Using microwave radiation as heating source, triblock copolymers with narrow molar mass distributions were obtained by the sequential addition of 2‐ethyl‐2‐oxazoline, 2‐(1‐ethylheptyl)‐2‐oxazoline, and 2‐(2,6‐difluorophenyl)‐2‐oxazoline. The polymers obtained were characterized by size exclusion chromatography, 1H NMR spectroscopy and matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI‐TOF MS). To investigate the incorporation of all three monomers into the triblock copolymers, a model polymer was prepared with shorter blocks exhibiting a suitable length to be measured in the reflector mode of a MALDI‐TOF MS. In addition, kinetic investigations on the homopolymerizations of all monomers were performed in nitromethane at 140 °C, yielding the polymerization rates under these conditions. DSC measurements of poly(2‐(1‐ethylheptyl)‐2‐oxazoline) and poly(2‐(2,6‐difluorophenyl)‐2‐oxazoline)) revealing glass transitions at about 33 and 120 °C, respectively. The thermal analysis of a blend of the two polymers showed two glass transitions revealing demixing, which could be an indicating for the immiscibility of the two components in the block copolymers. © 2010 Wiley Periodicals, Inc. Poly(pyridine ether)s were prepared in two ways: the polycondensation of silylated 1,1,1‐tris(4‐hydroxyphenyl)ethane (THPE) with 2,6‐difluoropyridine (method A) and the polycondensation of free THPE with 2,6‐dichloropyridine (method B). With method A, the THPE/difluoropyridine feed ratio was varied from 1.0:1.0 to 1.0:1.6. Cycles, bicycles, and multicycles were the main reaction products, and crosslinking was never observed. When ideal stoichiometry was used exclusively, multicycles free of functional groups were obtained. These multicycles were detectable in matrix‐assisted laser desorption/ionization time‐of‐flight (MALDI‐TOF) mass spectra up to B38C76 with a mass of approximately 32,000 Da. With method B, the reaction conditions were varied at a fixed feed ratio to achieve an optimum for the preparation of multicyclic polyethers, but because of the lower reactivity of 2,6‐dichloropyridine, a quantitative conversion was not achieved. 3‐Ethyl‐3‐hydroxymethyloxetane (EOX) polymerizes readily to branched multihydroxyl polyethers. Molecular weights of the polymers are, however, limited, and macromolecules are predominantly cyclic. This indicates that intramolecular chain transfer to polymer (back‐biting) proceeds in the system. Repeating units in poly‐EOX contain two nucleophilic sites that may participate in back‐biting, namely ether groups and hydroxyl groups. Analysis of matrix‐assisted laser desorption/ionization time‐of‐flight (MALDI‐TOF) mass spectra of poly‐EOX prepared in the presence of analogous polyether that does not contain HO? groups (poly(3,3‐dimethyloxetane)‐poly‐DMOX) shows that the ether group in the repeating unit of poly‐DMOX does not participate in chain transfer to the polymer. However, when DMOX was polymerized in the presence of poly‐EOX, clear evidence of participation of HO? groups in intramolecular chain transfer was obtained. © 2003 Wiley Periodicals, Inc. An analytical workflow involving high resolution mass analysis, collision‐induced dissociation and ion mobility was implemented to structurally characterize polymeric by‐products detected in lieu of intact species when performing matrix‐assisted laser desorption/ionization (MALDI) of polystyrenes with fragile end groups. Studied samples were prepared by atom transfer radical polymerization, reversible addition–fragmentation transfer polymerization and nitroxide‐mediated polymerization. Spectral resolution enabled by orthogonal injection of MALDI ions into a reflectron time‐of‐flight mass analyzer allowed a thorough inventory of species, including some with the same nominal m/z value but different elemental composition. Individual end‐group mass determination was achieved in MS/MS experiments, implementing an additional separative dimension based on ion mobility prior to CID to assist precursor ion selection in case of interferences. Besides validating commonly reported polystyrene chains terminated with either endo‐ or exo‐double bond, this multidimensional approach permitted to show that initiating moiety could also be affected by the MALDI process. © 2016 Wiley Periodicals, Inc. The thermal stability of several commonly used crystalline matrix‐assisted ultraviolet laser desorption/ionization mass spectrometry (UV‐MALDI‐MS) matrices, 2,5‐dihydroxybenzoic acid (gentisic acid; GA), 2,4,6‐trihydroxyacetophenone (THA), α‐cyano‐4‐hydroxycinnamic acid (CHC), 3,5‐dimethoxy‐4‐hydroxycinnamic acid (sinapinic acid; SA), 9H‐pirido[3,4‐b]indole (nor‐harmane; nor‐Ho), 1‐methyl‐9H‐pirido[3,4‐b]indole (harmane; Ho), perchlorate of nor‐harmanonium ([nor‐Ho + H]+) and perchlorate of harmanonium ([Ho + H]+) was studied by heating them at their melting point and characterizing the remaining material by using different MS techniques [electron ionization mass spectrometry (EI‐MS), ultraviolet laserdesorption/ionization‐time‐of‐flight‐mass spectrometry (UV‐LDI‐TOF‐MS) and electrospray ionization‐time‐of‐flight‐mass spectrometry (ESI‐TOF‐MS)] as well as by thin layer chromatography analysis (TLC), electronic spectroscopy (UV‐absorption, fluorescence emission and excitation spectroscopy) and 1H nuclear magnetic resonance spectroscopy (1H‐NMR). In general, all compounds, except for CHC and SA, remained unchanged after fusion. CHC showed loss of CO2, yielding the trans‐/cis‐4‐hydroxyphenylacrilonitrile mixture. This mixture was unambiguously characterized by MS and 1H‐NMR spectroscopy, and its sublimation capability was demonstrated. These results explain the well‐known cluster formation, fading (vanishing) and further recovering of CHC when used as a matrix in UV‐MALDI‐MS. Commercial SA (SA 98%; trans‐SA/cis‐SA 5 : 1) showed mainly cis‐ to‐trans thermal isomerization and, with very poor yield, loss of CO2, yielding (3′,5′‐dimethoxy‐4′‐hydroxyphenyl)‐1‐ethene as the decarboxilated product. These thermal conversions would not drastically affect its behavior as a UV‐MALDI matrix as happens in the case of CHC. A sample pretreatment was evaluated to enable the production of intact cationic species of synthetic polymers holding a labile end‐group using matrix‐assisted laser desorption/ionization (MALDI) mass spectrometry. More specifically, polymers obtained by nitroxide‐mediated polymerization involving the MAMA‐SG1 alkoxyamine were stirred for a few hours in trifluoroacetic acid (TFA) to induce the substitution of a tert‐butyl group on the nitrogen of nitroxide end‐group by a hydrogen atom. Nuclear magnetic resonance, electrospray ionization tandem mass spectrometry, and theoretical calculations were combined to scrutinize this sample pretreatment from both mechanistic and energetic points of view. The substitution reaction was found to increase the dissociation energy of the fragile C? ON bond to a sufficient extent to prevent this bond to be spontaneously cleaved during MALDI analysis. This TFA treatment is shown to be very efficient regardless of the nature of the polymer, as evidenced by reliable MALDI mass spectrometric data obtained for poly(ethylene oxide), polystyrene and poly(butylacrylate).  相似文献

Matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI‐TOF MS) coupled with CID (collision‐induced dissociation) has been used for the detailed characterization of two poly(2‐ethyl‐2‐oxazoline)s as part of a continuing study of synthetic polymers by MALDI‐TOF MS/MS. These experiments provided information about the variety of fragmentation pathways for poly(oxazoline)s. It was possible to show that, in addition to the eliminations of small molecules, like ethene and hydrogen, the McLafferty rearrangement is also a possible fragmentation route. A library of fragmentation pathways for synthetic polymers was also constructed and such a library should enable the fast and automated data analysis of polymers in the future. Poly(2‐ethyl‐2‐oxazoline), a synthetic polymer was analysed by mass spectrometry using different ion sources. Two distributions could be identified in the mass spectra which related to two different polymer series (one with hydrogen and hydroxyl end‐groups and the other with methyl and hydroxyl end‐groups). The fragmentation behaviour of the protonated oligomers was studied in a quadrupole time‐of‐flight mass spectrometer (MS) with electrospray, atmospheric pressure chemical ionization and direct analysis in real time soft ionization techniques. Three product ion series were identified in the MS/MS spectra independently of the ion source used. Based on the results, a mechanism was proposed for the dissociation by means of the accurate mass of the product ions, pseudo MS3 experiments and the energy dependence of the product ion intensity, i.e. breakdown curves. The copolymerization of 1,5‐dioxepan‐2‐one (DXO) and ε‐caprolactone, initiated by a five‐membered cyclic tin alkoxide initiator, was performed in chloroform at 60 °C. Copolymers with different molar ratios of DXO (25, 40, and 60%) were synthesized and characterized. 13C NMR spectroscopy of the carbonyl region revealed the formation of copolymers with a blocklike structure. Differential scanning calorimetry measurements showed that all the copolymers had a single glass transition between ?57 and ?49 °C and a melting temperature in the range of 30.1–47.7 °C, both of which were correlated with the amount of DXO. An increase in the amount of DXO led to an increase in the glass‐transition temperature and to a decrease in the melting temperature. Dynamic mechanical thermal analysis measurements confirmed the results of the calorimetric analysis, showing a single sharp drop in the storage modulus in the temperature region corresponding to the glass transition. Tensile testing demonstrated good mechanical properties with a tensile strength of 27–39 MPa and an elongation at break of up to 1400%. The morphology of the copolymers was examined with polarized optical microscopy and atomic force microscopy; the films that crystallized from the melt showed a short fibrillar structure (with a length of 0.05–0.4 μm) in contrast to the untreated solution‐cast films. © 2003 Wiley Periodicals, Inc.
