In this study, the antioxidant activity of aqueous and ethanol extracts of four plants from the genus Cassia were evaluated by various antioxidant assays, including ferric reducing antioxidant power (FRAP), DPPH free radical scavenging, metal chelating activity, phosphomolybdenum reducing power, hydrogen peroxide radical scavenging, hydroxyl radical scavenging, deoxyribose degradation and β-carotene bleaching assay. The various antioxidant activities were compared to standard antioxidant such as ascorbic acid. All the extracts showed antioxidant activity in the tested methods. Among the four species, Cassia auriculata has been found to possess highest activity in most of the tested models. In addition to the antioxidant activity, the total phenolics and flavonoids were measured in the extracts. The ethanolic extract exhibited highest phenolics and flavonoid contents and had also shown potent antioxidant activity in comparison to the aqueous extracts. The possible antioxidant mechanism of the ethanol extract can be due to its hydrogen or electron donating and direct free radical scavenging properties. Hence, the ethanol extract represents a source of potential antioxidants that could be used in pharmaceutical industries.  相似文献

Swertia chirayita is one of the potential medicinal plants of the family Gentianaceae in traditional medicine. Due to its high demand and scarcity, trade of chirayita is affected by adulterants. Swertia species from Western Ghats were compared with S. chirayita for phytochemical characterisation and antioxidant activities by using different extracts. This study revealed that acetone is the best extraction solvent of phenolic and flavonoid compounds with antioxidant properties as compared with other extracts. S. chirayita showed better antioxidant activity than other species with highest content of phenolics and flavonoids. Among the species from Western Ghats, Swertia minor has better antioxidant properties with higher content of phenolics and flavonoids when compared with S. chirayita. Gallic acid was detected in all species under study by using HPLC analysis. The Swertia species under study showed similar phytochemical properties and antioxidant potential and hence their use as substitute to S. chirayita needs to be further investigated.  相似文献

This study reports the antioxidant properties of three wild medicinal plants from Bahrain, namely Aizoon canariense L., Asphodelus tenuifolius Cav., and Emex spinosus (L.) Campdera. Antioxidant and antiradical activities of dried materials of these plants were investigated using FRAP, DPPH and ABTS assays. Total phenolics, free phenolics and total flavonoids were also determined. E. spinosus was ranked by the assays as the plant possessing the highest antioxidant and antiradical activities with an average FRAP value of 1.84 mmol/g and IC50 of 10.7 and 7.75 mg/ml for DPPH and ABTS assays, respectively. A. tenuifolius ranked second with a mean FRAP value of 0.69, IC50 DPPH of 1.72 and ABTS of 0.36. A. canariense possessed the lowest activities with a mean FRAP value of 0.6, and averaged IC50 of 103.8 and ABTS of 14.6. E. spinosus possessed the highest content of free phenolics (mg/100 g) (64.64) followed by A. tenuifolius (45.21) and A. canariense (32.23). E. spinosus also exhibited the highest total flavonoids with an average 82.71 mg/100 g followed by A. canariense (55.92) and A. tenuifolius (49.10). The studied medicinal plants possess considerable antioxidant activities and may contribute to the well-being of individuals who consume them.  相似文献

The aerial parts of Epilobium plants are widely used as folk medicine and food around the world. The present study was aimed to investigate the antioxidant activities and active chemical constituents from Epilobium angustifolium L. The results revealed that the EtOAc extract, rich in phenolic compounds and flavonoids (16.81 ± 0.67 g GAE/100 g extract and 4.95 ± 0.21 g QE/100 g extract, respectively), possessed significantly antioxidant activities in reducing power, DPPH radical scavenging activity, ABTS radical scavenging activity and highly in inhibiting lipid peroxidation activity. Simultaneously, active fractions F to H from EtOAc extracts showing potent in vitro antioxidant activities also contained high content of total phenolic and flavonoid. Twenty-eight compounds were identified as phenolic compounds and flavonoids by LC-MS/MS. The results illustrate that the E. angustifolium L., which is rich in phenolics, could be used as a natural resource of antioxidant ingredient.  相似文献

The antioxidant, antiproliferative and cytotoxic activities against different human cancer cells were investigated in local and recently introduced plants of Mentha sp., Rosmarinus officinalis L. (ROL) and Origanum majorana L. (OML). ROL exhibited the highest antioxidant activities (IC50 8.4 ± 0.2 μg/mL) followed by OML and mint species such as Mentha suaveolens ‘apple mint’ and Mentha longifolia L. exhibiting moderate antioxidant activities. HPLC analysis of leaf extract revealed that rosmarinic acid is the main component followed by caffeic acid. Herbal leaf extracts varied in their proliferation inhibition and cytotoxicity against HeLa, MCF-7 and Jurkat cancer cells in a dose-dependent matter. The highest antiproliferative inhibition and cytotoxic activity were detected in ROL and OML followed by mint. Local herbs might have a potential role as anticancer natural medicines in addition to their high antioxidant activities due to the presence of different phenolics in their aqueous tea extracts.  相似文献

A range of conventional, i.e. maceration, percolation, ultrasonic assisted, Soxhlet and Soxtec extraction (STE), to advanced extraction techniques of accelerated solvent extraction (ASE) was utilized for the first time in order to optimize the extract yield and recovery of phenolics—gallic acid (GA), rutin (RT) and quercetin (QT)—quantified via ultra-high performance liquid chromatography with diode array detector (UHPLC–DAD). The effect of solvents (n-hexane, dichloromethane and methanol) and temperature (60, 80 and 100°C) upon extraction yield, phenolic content and antioxidant activity (DPPH, ABTS and DPPH) was studied, and the method was validated in commercial food samples from Saudi Arabia, China and India. A high extract yield with percentage recovery was observed for STE (1221.10 mg/5 g; 24.42%) and ASE techniques (91.50 mg/1 g; 9.15%) in methanol at 100°C. UHPLC–DAD showed retention times (min) of 0.67, 1.93 and 1.90 for GA, RT and QT, respectively in the shortest runtime of 3 min. The yield for phenolics was higher for STE/ASE (ppm): 15.27/15.29 (GA), 85.24/37.56 (RT) and 52.20/33.40 (QT), respectively. In terms of antioxidant activities, low IC50 values (μg/ml) of 1.09/1.18 (DPPH), 2.11/5.32 (ABTS) and 4.35/7.88 (phenazine methosulfate–nicotinamide adenine dinucleotide) were observed for STE and ASE, respectively. Multivariate analysis for STE showed a significant (P = 0.000) correlation for extraction type vs. extract yield and phenolics content; however, there was no significance for antioxidant activities vs. extraction type. ASE showed a positive correlation for solvent vs. extraction yield, phenolics and antioxidant activity; however, there was no correlation for extraction yield and DPPH activity. Principal component analysis for STE showed a major variability (52.02%) for extraction yield and phenolics in PC1 followed by PC2 (38.30%) for antioxidant activities. For ASE, PC1 (48.68%) showed a positive correlation for solvent vs. extraction yield and phenolics while PC2 (33.12%) showed a positive correlation for temperature and antioxidant activities. STE and ASE were the optimized extraction techniques for the garlic food sample while a significant effect of solvent and temperature was observed upon extraction yield, phenolics and antioxidant activity.  相似文献

ABSTRACT

A chemiluminescence (CL) biosensing system for antioxidants was developed based on luminol and hematin co-immobilized on a cellulose membrane disc. The concentration of the antioxidant was quantified through the measurement of the inhibition of the CL emitted when hydrogen peroxide was introduced into the reagent phase. The instrumentation employed in the measurement was a fabricated luminometer employing optical fibers and a UV-enhanced photodiode transducer. Under optimum conditions, linear calibration curves were obtained for antioxidant concentrations ranging from 1 × 10-4 M to 0.10 M, with an average relative standard deviation of about 5%. The minimum detectable concentration was 100 μM, and the response time was less than 60 seconds. The sensor response correlated well (r = 0.9979) with the results of a standard colorimetric method for a specific antioxidant (propyl gallate). The sensor was used to assess the antioxidant capacity of water infusions prepared from the dried leaves of some Philippine medicinal plants.  相似文献

ABSTRACT

A widely used method for obtaining silver nanoparticles uses plant extracts for reduction because of the presence of phytochemicals such as terpenoids, tannins, and flavonoids. Extracts of Flores sambuci, Hypericum perforatum, Lavandula angustifolia, Origanum vulgare, Rosmarinus officinalis, and Salvia officinalis were used for generating silver nanoparticles. The ultraviolet–visible spectra of silver nanoparticle solutions were correlated with variations of phytochemical characteristics to evaluate the plant extracts. These parameters were the antioxidant activity, total flavonoids, total tannins, total terpenoids, and total phenolics. Correlations between measurements of extracts’ phytoreductive characteristics were explained using Pearson coefficients. The results showed medium linear positive correlations for total tannins with the spectra of silver nanoparticle solutions. The antioxidant activity and total terpenoids presented medium linear negative correlations. Pearson coefficients between total phenolics and relative areas from ultraviolet–visible spectra from 350 to 600?nm were close to zero indicating no linear correlation.  相似文献

Physalis ixocarpa Brot. ex Hornem. and Physalis angulata L. are two edible species of the family Solanaceae, which have an important variety of antioxidant compounds present in their roots, stems, leaves, calyces, and fruits. This work reports the development of multivariate models based on the use of partial least square (PLS) analysis and Fourier transform infrared (FTIR) spectroscopy for the quantitative determination of total phenolics, total flavonoids, free radical scavenging activity, total antioxidant capacity, and reducing power in the extracts of roots, stems, and leaves of both P. ixocarpa and P. angulata. Standard chromatographic and colorimetric techniques were used to determine the quantitative actual values (references) in the extracts, which served as input data to develop the multivariate PLS models. Optimized FTIR-PLS models were realized by cross-validation procedures, obtaining the determination coefficients for prediction between 0.792 and 0.905 for P. ixocarpa, and between 0.756 and 0.893 for P. angulata. In this form, FTIR spectroscopy with multivariate analysis could represent a versatile tool to evaluate quantitatively concentrations of bioactive compounds and antioxidant properties in the extracts of both species, requiring a very short time at low cost.  相似文献

Abstract

A chemiluminometric methodology for fluoxetine determination based on its antioxidant scavenger effect on the fast chemiluminescence reaction between luminol and hypochlorite, is proposed. The developed procedure was implemented in a fully automated multi‐pumping flow system, combining the excellent mixing characteristics of the pulsed flow with chemiluminescence detection, resulting on a sensitive, simple, and fast procedure for fluoxetine determination in pharmaceutical formulations. Linear calibration plots for fluoxetine hydrochloride concentrations of up to 10 mg l?1 (r=0.9995, n=7) were obtained, with an r.s.d<2% (n=10). Detection limit (3 σ) was 0.31 mg l?1 and the sampling rate was about 136 determinations per h.  相似文献

The aim of this study was to characterize aqueous and alcoholic extracts [30%, 50% and 70% (w/v)] obtained from medicinal herbs (Calendula officinalis, Hypericum perforatum, Galium verum, and Origanum vulgare) used in traditional medicine from our country. Samples were examined for total and individual content of phenolics and antioxidant activities. The highest content of total polyphenols (9.9 ± 0.02 mg gallic acid equivalents (GAE) L–1 extract) and antioxidant activities expressed as Trolox Equivalent Antioxidant Capacity [307.51 TEAC mmol g–1DW by the ABTS (2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) method and 20.90 TEAC mmol g–1 DW by the DPPH (1,1-diphenyl-2-picrylhydrazyl) method] was found in Origanum vulgare (50%) extract. The aim of this study was to examine the influence of storage conditions and duration on composition and antioxidant activity of supercritical fluid (SCF) extracted essential oil of rosemary (Rosmarinus officinalis L.). Supercritical extraction was carried out sequentially by using SCF carbondioxide in the first two steps and with 5% ethanol as entrainer in the third step. The compositions of the extracts were determined by gas chromatography/mass spectrometry. The total phenolics were analyzed using Folin-Ciocalteau assay. Abstract

The concentration and antioxidant activity of the known phenolics macluraxanthone, rutin, chlorogenic acid, and psoralen present in M. pomifera, F. carica, and F. elastica extracts were determined to evaluate the plant extracts potency as new antioxidant suppliers.

The quantification of the compounds was obtained by HPLC-DAD. The antioxidant activity of plant extracts and compounds was evaluated using DPPH and ABTS methods. The results seem to indicate that these extracts can be promising as new antioxidant suppliers, particularly F. carica and F. elastica leaves because only about 30% and about 50%, respectively, of the total activity was due to the phenolics determined.  相似文献

In this paper is described the applicability of a flow injection system, operating with an amperometric detector, for measurement in rapid and simple way the antioxidant power of honey, propolis and royal jelly. The proposed method evaluates the reducing power of selected antioxidant compounds and does not require the use of free radicals or oxidants. Twelve honey, 12 propolis and 4 royal jelly samples of different botanical and geographical origin were evaluated by the electrochemical method and the data were compared with those obtained by the DPPH assay. Since a good correlation was found (R2 = 0.92) the proposed electrochemical method can be successfully employed for the direct, rapid and simple monitoring of the antioxidant power of honeybee products. Furthermore, the total phenolic content of samples was determined by the Folin-Ciocalteau procedure and the characteristic antioxidant activities showed a good correlation with phenolics (R2 = 0.96 for propolis and 0.90 for honey).  相似文献

The antioxidant activity, total phenolic content, and total flavonoid content of 10 edible mushrooms species, including cultivated (Pleurotus ostreatus, Agaricus bisporus white and brown) and wild (P. ostreatus, Macrolepiota procera, Cantharellus cibarius, Russula vesca, Russula alutacea, Boletus edulis and Agaricus campestris), were determined. The extraction was performed using water and 50% water–ethanol and the caps and stipe were investigated separately. Water was the most appropriate solvent for phenolic compounds regardless of the sample. In contrast, the presence of ethanol in the solvent increased the extraction of flavonoids for cultivated P. ostreatus (caps and stipe), and wild R. alutacea, R. vesca, A. campestris, P. ostreatus (only caps) and C. cibarius, M. procera (only stipe). Significant differences between the antioxidant activities of the samples were registered in relationship with the different solvents. The antioxidant activity of water extract of dried A. bisporus brown (cultivated) showed the strongest 1,1-diphenyl-2-picrylhydrazyl radicals-scavenging assay (88.64%), while the B. edulis hydroalcoholic extract contained 74.93%. A detailed investigation into the functional group of phenolics and other organic compounds responsible with the antioxidant activity has been performed using Fourier transform infrared spectroscopy. The infrared spectra showed that the solvent-type directly influences the extraction process and, hence, the antioxidant activity. The present study contributes to information concerning mushrooms as sources of biologically active compounds. To investigate the correlations between phytochemical characteristics (i.e., phenolics, flavonoids and antioxidant activity) closely related to nature of solvents, the statistical analysis was performed.  相似文献

Hibiscus manihot L. is a kind of healthy plant with edible value and health benefits, which possesses multiple pharmacological activities that are closely related to antioxidant and anti-inflammatory activities. The dynamic changes of main active components and biological activities in Hibiscus manihot L. flower (HMLF) during its flowering period were systematically studied to determine the appropriate harvest time. Chemopreventive efficacies of the investigated HMLF extracts, by means of their anti-inflammatory and antioxidant activities, were assessed. The sample harvested on early August had the supreme total flavonoid content, total phenolic content and the strongest antioxidant activity (DPPH radical scavenging activity (IC50 0.160 mg/mL), ABTS radical scavenging activity (1.570 mmol/g Trolox), reducing power (IC50 0.101 mg/mL) and FRAP (3.644 mmol FeSO4/g)). The results of principal component analysis indicated that the primary active components included hyperin, isoquercetin, hibifolin and quercetin-3′-O-glucoside, which were strongly associated with the antioxidant activity in the early August sample, while neochlorogenic acid, chlorogenic acid and caffeic acid were associated with the anti-inflammatory activity. The extracts significantly inhibited lipopolysaccharide-induced nitric oxide production in RAW264.7 cells, especially the samples harvested around August, which was only 3.569 μΜ with the inhibition ratio of>50%. This study indicated that HMLF harvested on the early August possessed the highest antioxidant and anti-inflammatory potential and could be used as high bioactive resources for healthy production.  相似文献

The antioxidant activities of crude extract fractions using Hexane, Chloroform, Ethyl Acetate, Butanol and Water of Clematis orientalis and Clematis ispahanica were investigated. 1,1-diphenyl-2-picryl-hydrazyl (DPPH) assay and the ferric reducing/antioxidant potential (FRAP) were used to evaluate the antioxidant capacity. The total phenolics were found to be 4.37–9.38 and 1.32–11.37 mg gallic acid equivalents (GAE)/g in different fractions for C. orientalis and C. ispahanica, respectively. The ethyl acetate fraction of C. orientalis and chloroform fraction of C. ispahanica showed the highest DPPH and FRAP activities at a concentration of 300 μg/mL. The predominant phenolic compounds identified by HPLC in C. orientalis were Resorcinol (603.5 μg/g DW) in chloroform fraction and Ellagic acid (811.7 μg/g DW) in chloroform fraction of C. ispahanica.  相似文献

Novel methods for the determination of polyphenolic antioxidants present in extracts from inflorescences of Cirsium vulgare (Savi) Ten. based on ultra‐high performance liquid chromatography with photodiode array and chemiluminescence detection have been developed. Under the optimized conditions of chromatographic separation the analytical characteristic of the method was performed. The proposed method was successfully applied to the determination of ten polyphenols present in inflorescences of Cirsium vulgare . A comparison of the contents of analytes in extracts prepared by using various extraction media (methanol, ethanol, 70% methanol, 70% ethanol, and water) was carried out for the first time. For the postcolumn detection of scavenging activity of polyphenolic antioxidants against reactive oxygen species (H2O2, OH, O2• −) three systems based on chemiluminescence of luminol were used. A review of the current scientific literature shows that this is the first report on the application of luminol‐based postcolumn detection for the on‐line investigation of OH scavenging activity. The main compound determined in extracts from inflorescences of Cirsium vulgare was apigenin 7‐O‐glucuronide, whereas the highest antioxidant activity was observed for chlorogenic acid, luteolin 7‐O‐glucoside, and apigenin.  相似文献

The effect of continuous UV radiation and hydrogen peroxide on destruction and antioxidant properties of synthetic DOPA-melanin (prepared by oxidation of 3,4-dihydroxyphenylalanine (DOPA)) and melanosomes isolated from cells of the retinal pigment epithelium (RPE) was investigated. The kinetics of melanin destruction was recorded based on the accumulation of fluorescent low-molecular-weight reaction products, the antiradical activity of melanin was determined by chemiluminescence method, the concentration of free radical products was measured by electron paramagnetic resonance, and the antioxidant activity of melanins was estimated by their inhibitory effect on lipid peroxidation. It was shown that UVC—UVA irradiation (up to 5 hours) of DOPA-melanin and melanosomes of retinal pigment epithelium decreased neither the latency period of luminol chemiluminescence nor the inhibitory action of pigments on Fe2+- and UV-induced peroxidation of cardiolipin liposomes. However, very long UV irradiation gave rise to fluorescent destruction products, decreased the concentration of paramagnetic centers in the pigment (especially light-dependent ones), and decreased the antiradical and antioxidant activities. For example, UV irradiation of DOPA-melanin during 52 h resulted in approximately a 2-fold decrease in the concentration of paramagnetic centers and decline of antiradical and antioxidant activities. However, even with such a hard irradiation the pigment retained significant inhibitory activity against lipid peroxidation. The oxidative destruction of DOPA-melanin in the presence of hydrogen peroxide in the dark resulted in complete destruction of the polymer and loss of its protective properties. It is assumed that destruction of RPE cell melanin is caused mainly by oxidative processes.  相似文献

Abstract

Species of Garcinia (Guttiferae) are used for flavoring curries, as a supplement, and to treat various diseases. This study describes the comparison and discrimination of Garcinia cambogia, Garcinia indica, Garcinia mangostana and Garcinia atroviridis fruits by analyzing their major phytochemicals, elemental content, antioxidant, antidiabetic, and anticholinesterase enzymes activities. For phytochemical and elemental profiling, ultraviolet (UV), near infrared/infrared (NIR/IR), inductively coupled plasma-optical emission spectroscopy (ICP-OES) and ICP-mass spectrometric (ICP-MS) techniques were used. The chemometric multivariate tests of linear discriminant and principal component analyses (LDA, PCA) were used to discriminate the subject fruit samples. Spectroscopic data showed resonances of phenolics and flavonoidal constituents present in the fruits. G. mangostana exhibited the highest phenolics (721.6 to 2815.3?µM GAE/g), whereas G. cambogia was rich in flavonoids (51.9 to 2709.2?µM QE/g). Anthocyanin (cyanidin-3-O-glucoside) evaluated by high performance liquid chromatographic was 9.01?mg/kg in G. mangostana fruit. In the analyzed fruits, Ca, K and Na were high, trace essential elements were at appreciable contents, whereas the toxic elements As, Cd, Tl, and Pb were within the safe limits. G. mangostana contained potent free radicals and cholinesterase enzyme inhibitors, whereas G. cambogia inhibited α-amylase enzyme more significantly. PCA and LDA discriminated the fruit samples with distinct classification and variability indices. The analyzed fruits were shown to be good sources of free radicals, cholinesterase, and α-amylase enzymes inhibition, mineral and essential elements, and safe for human consumption.  相似文献
