Synthesis of derivatives of pyrido[2,3-d]pyrimidines condensed with tetrahydropyran and tetrahydrothiopyran

Synthesis of new pyrido[2,3-d]pyrimidine derivatives

The mutual condensation of 4-aminouracil or 2,4-diamino-6-hydroxypyrimidine with bisacetonitrile and aldehydes was used to synthesize 2,4-dioxo-5-R-7-methyl-6-cyano-1,2,3,4,5,8-hexahydropyrido[2,3-d]pyrimidine and 2-amino-4-oxo-5-R-7-methyl-6-cyano-3,4,5,8-tetrahydropyrido[2,3-d]pyrimidine derivatives, which were oxidized with chromic anhydride to the corresponding 2,4-dioxo-5-R-7-inethyl-6-eyano-1,2,3,4-tetrahydropyrido[2,3-d]pyrimidine and 2-amino-4-oxo-5-R-7-methyl-6-cyano-3,4-dihydro[2,3-d]pyrimidines. 5-Acetyl-6-amino-4-methylthio-2-phenylpyrimidine and its use in the synthesis of functionalized pyrido[2,3-d]pyrimidines and pyrimido-[4,5-d]pyrimidines

Schemes for the synthesis of MeS-substituted pyrido[2,3-d]pyrimidin-5-one, pyrimido[4,5-d]pyrimidine, and 4-methylenepyrimido[4,5-d]pyrimidin-2-one based on 5-acetyl-6-amino-4-methylthio-2-phenylpyrimidine, which was prepared from the adduct of benzoyl isothiocyanate with acetylketeneN-benzoylaminal, were suggested.Translated fromIzvestiya Akademii Nauk. One-Pot Synthesis of Novel Spiro Pyrano[2,3-d][1,3]thiazolo[3,2-a]pyrimidine Derivatives

In a one-pot synthesis, 1′-methyl-2,3″-dioxo-5″-aryl-1,2,5a″,7″,8″,9a″-hexahydro-5″H,6″H-dispiro[indole-3,2′-pyrrolidine-3′,2″-pyrano[2,3-d][1,3]thiazolo[3,2-a]pyrimidine]-4′-carboxylic acid methyl ester was prepared via the sequential reaction of 4-aryl-octahydro-pyrano[2,3-d]pyrimidine-2-thione, dimethyl acetylenedicarboxylate (DMAD), and a mixture of isatin and sarcosine. All the novel spiro compounds, in moderate yields, were characterized thoroughly by infrared, NMR, mass spectromentry, and elemental analysis together with x-ray crystallographic analysis.     

新型碱性离子液体催化剂高效催化合成吡喃酮[2,3-d]嘧啶酮和[2,3-d]嘧啶衍生物

制备了碱性离子液体1,1'-(丁烷-1,4-二基)双(1,4-二氮杂二环[2.2.2]辛烷-1-ium)羟化物,并采用红外光谱、1H核磁共振谱和pH值分析对其进行了表征.然后将它用于室温研磨条件下高效催化合成吡喃酮[2,3-d]嘧啶酮和[2,3-d]嘧啶.该法步骤简单,反应时间短,产物收率高,无需柱色谱分离,原料易得,且可回收利用.     

Pyrido[2,3-d]pyrimidines 4. Synthesis and some transformations of oxo(hydroxy)pyrido[2,3-d]pyrimidines

Synthesis and transformations of some pyrido[2,3-d]pyrimidines

A new synthetic approach for pyrido[2,3-d]pyrimidine 3-oxides and other pyrido[2,3-d]pyrimidines has been developed. This bicyclic system readily undergoes ring opening to give a variety of products, depending on the reagent and reaction conditions.

Synthesen und Umwandlungen einiger Pyrido[2,3-d]pyrimidine

Zusammenfassung Synthesen von Pyrido[2,3-d]pyrimidin-3-oxiden und einigen anderen Pyrido[2,3-d]pyrimidinen werden beschrieben. Das bicyclische System reagiert leicht unter Ringöffnung, wobei entsprechend den Reaktionsbedingungen und Reagenzien verschiedene Produkte entstehen.

     

A new base-induced ring transformation of pyrido[2,3-d]pyrimidines

8-Substituted 5,8-dihydro-5-oxopyrido[2,3-d]pyrimidine-6-carboxylates ( 3 ) rearranged to 8-substituted 7,8-dihydro-5-hydroxy-7-oxopyrido[2,3-d]pyrimidine-6-carboxaldehydes ( 5 ) when treated with sodium ethoxide in an aprotic polar solvent at room temperature. The 6-cyano analogue ( 18 ) also underwent ring transformation under the same mild conditions giving 7-amino-8-ethyl-5,8-dihydro-5-oxopyrido[2,3-d]pyrimidine-6-carboxaldehyde ( 21 ). However, the ring transformations of the pyrido[2,3-d]pyrimidine bearing no N₈-substituent ( 12 ), ethyl 1-ethyl-1,4-dihydro-4-oxo-1,8-naphthyridine- ( 14 ) and -quinoline-3-carboxylates ( 16 ) failed to occur. A mechanism is discussed.     

Thiazolo(oxazolo)thieno[b]pyrimidines. Preparation from 2-allylthio(oxy)thieno[2,3-d]pyrimidines and hydrolysis

Heterocyclization of 5,6-disubstituted 2-allylthio(oxy)-4-oxothieno[2,3-d]pyrimidines by treatment with halogens affords thieno[3,2-e]thiazolo[3,2-a]pyrimidine iminium salts. Pyrido[2,3-d]pyrimidines 3. Synthesis and properties of 7-chloro- and 6-nitro-7-chloropyrido[2,3-d]pyrimidine-2,4,5-triones

7-Chloropyridopyrimidine was obtained by diazotization of 1,3-dimethyl-7-amino-8H-pyrido[2,3-d]pyrimidine-2,4,5-trione in HCl, and its nitration and reactions with nucleophilic reagents were studied. Pyrylia[2,3-d]pyrimidine salts

Catalyst free synthesis of fused pyrido[2,3-d]pyrimidines and pyrazolo[3,4-b]pyridines in water

A one-pot,three-component condensation reaction of an aldehyde,benzoyl acetonitrile(3-oxo-3-phenylpropane nitrile) and 6- amino-1,3-dimethylpyrimidine-2,4(1H,3H)-dione or 3-methyl-1-phenyl-1H-pyrazol-5-amine in water to give fused pyrido[2,3- d]pyrimidines and pyrazolo[3,4-b]pyridines in high yields without any catalyst,is described.

以非经典叶酸拮抗剂2,4-二氨基-6-(4-甲基苯基)乙基吡啶并[3,2-d]嘧啶(wm-5b)及其侧链简化产物2,4-二氨基吡啶并[3,2-d]嘧啶为先导化合物, 选取具有抗肿瘤活性的基团, 通过微波法高效合成了2-位或4-位取代吡啶并嘧啶类非经典叶酸拮抗剂, 研究了2-位及4-位取代基对抗肿瘤活性的影响, 为非经典叶酸拮抗剂的设计合成提供了更多的理论依据. 目标化合物的结构均经核磁共振波谱(NMR)和质谱(MS)确证. 生物活性测定结果表明, 所有目标化合物均具有抗肿瘤活性, 其中, 6-(4-甲基苯基)乙基-4-氨基-2-(3-氯-4-氟苯基)氨基吡啶并[3,2-d]嘧啶(6b)对HL-60细胞的IC₅₀=(4.09±0.48) μmol/L, 对A549细胞的IC₅₀=(17.99±7.20) μmol/L, 而对HCT116细胞的IC₅₀=(14.52±4.74) μmol/L; 部分目标化合物具有二氢叶酸还原酶抑制活性. 此外, 对部分目标化合物和先导物进行了二氢叶酸还原酶晶体结构的分子对接, 对活性结果和构效关系从分子水平上进行解释.     

A clean and efficient cyclocondensation to pyrido[2,3-d]pyrimidine derivatives in aqueous media

A series of pyrido[2,3-d]pyrimidine derivatives were easily constructed by cyclocondensation reactions of 6-aminouracils or 6- aminothiouracil withα,β-unsaturated carbonyl compounds（aldehyde,ketone and ester） possessing a leaving group on theβposition,in H₂O under reflux conditions.     

Syntheses of some new azolopyrido‐[4′,3′:4,5]thieno[2,3‐d]pyrimidines

Diethyl 2‐[(ethoxythioxomethyl)amino]‐4,5,6,7‐tetrahydrothieno[2,3‐c]‐pyridine‐3,6‐dicarboxylate 2 , prepared from diethyl 2‐isothiocyanato‐4,5,6,7‐tetrahydrothieno[2,3‐c]pyridine‐3,6‐dicarboxylate 1 by boiling in anhydrous ethanol, was converted into pyrido[4′,3′:4,5]thieno[2,3‐d]pyrimidine derivatives 3, 4 by treatment with hydrazine hydrate. The tetracyclic systems imidazo[1,2‐a]pyrido‐[4′,3′:4,5]thieno[2,3‐d]pyrimidine 9 and pyrido[4′,3′:4,5]thieno[2,3‐d][1,3]thiazolo‐[3,2‐a]pyrimidine 10 were synthesized by the reaction of 2 with 1,2‐diaminoethane and aminoethanethiol, respectively. The hydrazino derivative 4 underwent cyclization reactions with orthoesters and nitrous acid to give the corresponding pyrido[4′,3′:4,5]thieno[2,3‐d][1,2,4]triazolo[1,5‐a]pyrimidines 5, 6 and pyrido[4′,3′:4,5]thieno[3,2‐e][1,2,3,4]tetrazolo[1,5‐a]pyrimidine 8 , respectively. Moreover, reactions of 3 with cyanogen bromide, N‐carbethoxyhydrazine, carbon disulfide, and ethylchloroformate resulted in the formation of the new pyrido[4′,3′:4,5]thieno[2,3‐d][1,3,4]thiadiazolo[3,2‐a]pyrimidine derivatives 12–15 . © 2002 Wiley Periodicals, Inc. Mass spectrometry of nitrogen heterocycles. 4. Mass spectral behavior of triazolo[1,5-a]pyrido[2,3-d]pyrimidines

The electron impact mass spectra of substituted 9-imino- and 9-oxotriazolo[1,5-a]pyrido[2,3-d]pyrimidines were studied. The mass spectral data obtained permitted us to determine the order of the fusion of the triazole, pyrimidine, and pyridine rings as well as the position of the hydrogen atoms at the heteroatoms.
