The synthesis of 3,4‐didehydroionone isomers 4 , (+)‐ 6 , and (?)‐ 6 and of 3,4‐didehydro‐7,8‐dihydroionone isomers 5 , (+)‐ 7 , and (?)‐ 7 was accomplished starting from commercially available racemic α‐ionone ( 1 ). Their preparation of the racemic forms 4 – 7 was first achieved by mean of a number of chemo‐ and regioselective reactions (Schemes 1 and 2). The enantio‐ and diastereoselective lipase‐mediated kinetic acetylation of 4‐hydroxy‐γ‐ionone ( 10a / 10b ) provided 4‐hydroxy‐γ‐ionone (+)‐ 10a /(±)‐ 10b and (+)‐4‐(acetyloxy)‐γ‐ionone ((+) 12b ) (Scheme 3). The latter compounds were used as starting materials to prepare the 3,4‐didehydro‐γ‐ionones (+)‐ and (?)‐ 6 and the 3,4‐didehydro‐7,8‐dihydro‐γ‐ionones (+)‐ and (?)‐ 7 in enantiomer‐enriched form. The absolute configuration of (+)‐ 12b was determine by chemical correlation with (+)‐(6S)‐γ‐ionone ((+)‐ 3 ) and with (?)‐(6S)‐α‐ionone ((?)‐ 1 ) therefore allowing to assign the (S)‐configuration to (+)‐ 6 and (+)‐ 7 . Olfactory evaluation of the above described 3,4‐didehydroionone isomers shows a significant difference between the enantiomers and regioisomers both in fragrance feature and in detection threshold (Table).  相似文献

The two epimers (?)‐ 1a and (?)‐ 1b of the macrocyclic lactam alkaloid 3‐hydroxycelacinnine with the (2R,3R) and (2R,3S) absolute configurations, respectively, were synthesized by an alternative route involving macrocyclization with the regio‐ and stereoselective oxirane‐ring opening by the terminal amino group (Schemes 2 and 6). Properly N‐protected chiral trans‐oxirane precursors provided (2R,3R)‐macrocycles after a one‐pot deprotection‐macrocyclization step under moderate dilution (0.005–0.01M ). The best yields (65–85%) were achieved with trifluoroacetyl protection. Macrocyclization of the corresponding cis‐oxiranes was unsuccessful for steric reasons. Inversion at OH? C(3) via nucleophilic displacement of the cyclic sulfamidate derivative with NaNO2 led to (2R,3S)‐macrocycles. The synthesized (?)‐(2R,3S)‐3‐hydroxycelacinnine ((?)‐ 1b ) was identical to the natural alkaloid.  相似文献

The synthesis of the marine sesquiterpene quinone (+)‐hyatellaquinone ( 1 ) was achieved starting from the sesquiterpene aldehyde (+)‐albicanal ((+)‐ 3 ) (Schemes 3 and 4). Coupling of (+)‐albicanal with 2,3,5,6‐tetramethoxyphenyllithium led to the aryl‐sesquiterpene system, which was modified to the target molecule. Furthermore, the first total synthesis of the marine compound spongiaquinone ( 2 ) was carried out starting from ((?)‐albicanal (?)‐ 3 ) in a reaction sequence encompassing a stereoselective C?C bond hydrogenation and a one‐pot AcOH elimination/demethylation reaction (Schemes 7 and 10). The occurrence of 1,2‐ and 1,4‐benzoquinone forms of 1 and 2 depends on the pH of the solvent system.  相似文献

To complete our panorama in structure–activity relationships (SARs) of sandalwood‐like alcohols derived from analogues of α‐campholenal (= (1R)‐2,2,3‐trimethylcyclopent‐3‐ene‐1‐acetaldehyde), we isomerized the epoxy‐isopropyl‐apopinene (?)‐ 2d to the corresponding unreported α‐campholenal analogue (+)‐ 4d (Scheme 1). Derived from the known 3‐demethyl‐α‐campholenal (+)‐ 4a , we prepared the saturated analogue (+)‐ 5a by hydrogenation, while the heterocyclic aldehyde (+)‐ 5b was obtained via a Bayer‐Villiger reaction from the known methyl ketone (+)‐ 6 . Oxidative hydroboration of the known α‐campholenal acetal (?)‐ 8b allowed, after subsequent oxidation of alcohol (+)‐ 9b to ketone (+)‐ 10 , and appropriate alkyl Grignard reaction, access to the 3,4‐disubstituted analogues (+)‐ 4f,g following dehydration and deprotection. (Scheme 2). Epoxidation of either (+)‐ 4b or its methyl ketone (+)‐ 4h , afforded stereoselectively the trans‐epoxy derivatives 11a,b , while the minor cis‐stereoisomer (+)‐ 12a was isolated by chromatography (trans/cis of the epoxy moiety relative to the C2 or C3 side chain). Alternatively, the corresponding trans‐epoxy alcohol or acetate 13a,b was obtained either by reduction/esterification from trans‐epoxy aldehyde (+)‐ 11a or by stereoselective epoxidation of the α‐campholenol (+)‐ 15a or of its acetate (?)‐ 15b , respectively. Their cis‐analogues were prepared starting from (+)‐ 12a . Either (+)‐ 4h or (?)‐ 11b , was submitted to a Bayer‐Villiger oxidation to afford acetate (?)‐ 16a . Since isomerizations of (?)‐ 16 lead preferentially to β‐campholene isomers, we followed a known procedure for the isomerization of (?)‐epoxyverbenone (?)‐ 2e to the norcampholenal analogue (+)‐ 19a . Reduction and subsequent protection afforded the silyl ether (?)‐ 19c , which was stereoselectively hydroborated under oxidative condition to afford the secondary alcohol (+)‐ 20c . Further oxidation and epimerization furnished the trans‐ketone (?)‐ 17a , a known intermediate of either (+)‐β‐necrodol (= (+)‐(1S,3S)‐2,2,3‐trimethyl‐4‐methylenecyclopentanemethanol; 17c ) or (+)‐(Z)‐lancifolol (= (1S,3R,4Z)‐2,2,3‐trimethyl‐4‐(4‐methylpent‐3‐enylidene)cyclopentanemethanol). Finally, hydrogenation of (+)‐ 4b gave the saturated cis‐aldehyde (+)‐ 21 , readily reduced to its corresponding alcohol (+)‐ 22a . Similarly, hydrogenation of β‐campholenol (= 2,3,3‐trimethylcyclopent‐1‐ene‐1‐ethanol) gave access via the cis‐alcohol rac‐ 23a , to the cis‐aldehyde rac‐ 24 .  相似文献

(+)-cis-Achillene ( 10 ) and (?)-trans-achillenol (7), two monoterpenes recently isolated [1] from the essential oil of Achilleafilipendulina, were synthesized, together with their stereoisomers (?)-(9) and (+)-(8), starting from (S)-(+)-2,6-trans-dimethylocta-1,3, 7-triene ( 1 ). The isomeric ß-hymen thereties ((?)- 3 and (+)-4), often quoted [2] [3] [4] but never isolated, were obtained as intermediates. The mode of synthesis chosen establishesis (R)-chirality for naturally occurring (?)-trans-achillenol (7) and (+)-cis-achillene ( 10 ) as well as for the purely synthetic 4, 7-diene derivatives described in this paper.  相似文献

Photochemical Reaction of Optically Active 2-(1′-Methylallyl)anilines with Methanol It is shown that (?)-(S)-2-(1′-methylallyl)aniline ((?)-(S)- 4 ) on irradiation in methanol yields (?)-(2S, 3R)-2, 3-dimethylindoline ((?)-trans- 8 ), (?)-(1′R, 2′R)-2-(2′-methoxy-1′-methylpropyl)aniline ((?)-erythro- 9 ) as well as racemic (1′RS, 2′SR)-2-(2′-methoxy-1′-methylpropyl) aniline ((±)-threo- 9 ) in 27.1, 36.4 and 15.7% yield, respectively (see Scheme 3). By deamination and chemical correlation with (+)-(2R, 3R)-3-phenyl-2-butanol ((+)-erythro- 13 ; see Scheme 4) it was found that (?)-erythro- 9 has the same absolute configuration and optical purity as the starting material (?)-(S)- 4 . Comparable results are obtained when (?)-(S)-N-methyl-2-(1′-methylallyl)aniline ((?)-(S)- 7 ) is irradiated in methanol, i.e. the optically active indoline (+)-trans- 10 and the methanol addition product (?)-erythro- 11 along with its racemic threo-isomer are formed (cf. Scheme 3). These findings demonstrate that the methanol addition products arise from stereospecific, methanol-induced ring opening of intermediate, chiral trans, -(→(?)-erythro-compounds) and achiral cis-spiro [2.5]octa-4,6-dien-8-imines (→(±)-threo-compounds; see Schemes 1 and 2).  相似文献

Starting from inexpensive (E)‐β‐farnesene ( 1 ), an eight‐step enantioselective synthesis of the olfactively precious Ambrox® ((?)‐ 2a ) has been performed. The crucial step is the catalytic asymmetric isomerization of (2E,6E)‐N,N‐diethylfarnesylamine ( 3 ) to the corresponding enamine (?)‐(R,E)‐ 4a , applying Takasago's well‐known industrial methodology. The resulting dihydrofarnesal ((+)‐(R)‐ 5 ) (90% yield, 96% ee), obtained after in situ hydrolysis (AcOH, H2O), was then cyclized under catalytic SnCl4 conditions, via its corresponding unreported enol acetate (?)‐(R)‐ 4b , to afford trans‐decalenic aldehyde (+)‐ 6a . Subsequent transformations furnished bicyclic ketone (?)‐ 8a and unsaturated nitrile (+)‐ 11 , both reported as intermediates to access to (?)‐ 2a .  相似文献

A short and efficient protocol for the asymmetric synthesis of cis‐ and trans‐3,4‐dihydro‐2,4,8‐trihydroxynaphthalen‐1(2H)‐one ( 1 and 2 , resp.) is described, with a phthalide annulation as the key step. Introduction of a OH substituent at position 2 was performed by Sharpless dihydroxylation of a silyl enol ether or by means of an N‐sulfonyloxaziridine. The absolute configuration of each isomer was determined via Mosher‐ester derivatives. By comparison with previously recorded CD spectra of our natural sample, we established that the natural trans‐ and cis‐isomers from Ceratocystis fimbriata sp. platani were the (?)‐(2S,4S)‐isomer (?)‐ 2 and the (+)‐(2S,4R)‐isomer (+)‐ 1 , respectively.  相似文献

The 2,2′‐methylenebis[furan] ( 1 ) was converted to 1‐{(4R,6S))‐6‐[(2R)‐2,4‐dihydroxybutyl]‐2,2‐dimethyl‐1,3‐dioxan‐4‐yl}‐3‐[(2R,4R)‐tetrahydro‐4,6‐dihydroxy‐2H‐pyran‐2‐yl)propan‐2‐one ((+)‐ 18 ) and its (4S)‐epimer (?)‐ 19 with high stereo‐ and enantioselectivity (Schemes 13). Under acidic methanolysis, (+)‐ 18 yielded a single spiroketal, (3R)‐4‐{(1R,3S,4′R,5R,6′S,7R)‐3′,4′,5′,6′‐tetrahydro‐4′‐hydroxy‐7‐methoxyspiro[2,6‐dioxabicyclo[3.3.1]nonane‐3,2′‐[2H]pyran]‐6′‐yl}butane‐1,3‐diol ((?)‐ 20 ), in which both O‐atoms at the spiro center reside in equatorial positions, this being due to the tricyclic nature of (?)‐ 20 (methyl pyranoside formation). Compound (?)‐ 19 was converted similarly into the (4′S)‐epimeric tricyclic spiroketal (?)‐ 21 that also adopts a similar (3S)‐configuration and conformation. Spiroketals (?)‐ 20 , (?)‐ 21 and analog (?)‐ 23 , i.e., (1R,3S,4′R,5R,6′R)‐3′,4′,5′,6′‐tetrahydro‐6′‐[(2S)‐2‐hydroxybut‐3‐enyl]‐7‐methoxyspiro[2,6‐dioxabicyclo[3.3.1]nonane‐3,2′‐[2H]pyran]‐4′‐ol, derived from (?)‐ 20 , were assayed for their cytotoxicity toward murine P388 lymphocytic leukemia and six human cancer cell lines. Only racemic (±)‐ 21 showed evidence of cancer‐cell‐growth inhibition (P388, ED50: 6.9 μg/ml).  相似文献

A broadly applicable route to trans‐2,5‐disubstituted pyrrolidines has been developed. Key steps are an asymmetric iridium‐catalyzed allylic amination, a Suzuki–Miyaura coupling, and an intramolecular aza‐Michael addition. Enantiomeric excesses in the range of 93–99 % ee have been achieved. Total syntheses of the alkaloids (?)‐ 225 C , (+)‐ and (?)‐ 223 H (xenovenine), (+)‐ 223 AB , (+)‐ 195 B , and (+)‐ 223 R have been carried out as applications.  相似文献

The reactions of (+)‐car‐2‐ene ( 1 ) and (+)‐car‐3‐ene ( 2 ) with aldehydes in the presence of montmorillonite clay were studied for the first time (Schemes 3 and 5). The major products of these reactions are optically active, substituted hexahydroisobenzofurans, probably formed as a result of an attack of the protonated aldehyde at the cyclopropane ring. Quite unexpectedly, the products are cis‐configured at the ring‐fusion site; the fact was established by means of quantum‐chemical calculations and NMR data. It appeared that the behavior of the 2 : 3 mixture 1 / 2 in reactions with aldehydes in the presence of K10 clay differed substantially from the reactivities of the corresponding individual monoterpenes.  相似文献

A concise asymmetric (>99:1 e.r.) total synthesis of (+)‐anti‐ and (?)‐syn‐mefloquine hydrochloride from a common intermediate is described. The key asymmetric transformation is a Sharpless dihydroxylation of an olefin that is accessed in three steps from commercially available materials. The Sharpless‐derived diol is converted into either a trans or cis epoxide, and these are subsequently converted into (+)‐anti‐ and (?)‐syn‐mefloquine, respectively. The synthetic (+)‐anti‐ and (?)‐syn‐mefloquine samples were derivatized with (S)‐(+)‐mandelic acid tert‐butyldimethylsilyl ether, and a crystal structure of each derivative was obtained. These are the first X‐ray structures for mefloquine derivatives that were obtained by coupling to a known chiral, nonracemic compound, and provide definitive confirmation of the absolute stereochemistry of (+)‐anti‐ as well as (?)‐syn‐mefloquine.  相似文献

Discrepancies between chiroptical data from the literature and our determination of the structure of the title compounds (+)‐ 5 and (+)‐ 9a were resolved by an unambiguous assignment of their absolute configuration. Accordingly, the dextrorotatory cis‐3‐hydroxy esters have (3R,4R)‐ and the laevorotatory enantiomers (3S,4S)‐configuration. The final evidences were demonstrated on both enantiomers (+)‐ and (?)‐ 5 by biological reduction of 4 by bakers' yeast and stereoselective [RuII(binap)]‐catalyzed hydrogenations of 4 (Scheme 2), by the application of the NMR Mosher method on (+)‐ and (?)‐ 5 (Scheme 3), as well as by the transformation of (+)‐ 5 into a common derivative and chiroptical correlation (Scheme 4).  相似文献

LC/MS3‐Guided biotransformation of p‐coumaric acid (=(2E)‐3‐(4‐hydroxyphenyl)prop‐2‐enoic acid; CA) with H2O2/Momordica charantia peroxidase at pH 5.0 and 45° in the presence of acetone has resulted in the isolation of three CA trimers, triCA1 ( 1 ), triCA2 (trans‐ 2 ), and triCA3 (cis‐ 2 ), and seven CA dimers, diCA1–diCA7, i.e., 3 – 9 , among which seven (triCA1–triCA3 and diCA1–diCA4) are new compounds and three (diCA5–diCA7) are known compounds. The structures were established by 2D‐NMR such as HSQC, HMBC, and NOESY measurements. The possible mechanism for the formation of the products is also discussed (Schemes 1–3). This is the first time that the biotansformation of p‐coumaric acid catalyzed by peroxidase in vitro was achieved. Compounds triCA3 (cis‐ 2 ), diCA1 ( 3 ), diCA5 ( 7 ), and diCA7 ( 9 ) exhibit a stronger antioxidative activity than the parent CA.  相似文献

ent‐Erythramine ((?)‐ 1 ), the enantiomer of the alkaloid erythramine, was prepared in 15 steps from known compounds. The first of three pivotal bond‐forming steps in the synthesis was a Suzuki–Miyaura cross‐coupling reaction of the starting materials to give a bis‐silyl ether. The second involved silver(I)‐induced electrocyclic ring opening of the gem‐dichlorocyclopropane formed in the next step and trapping of the ensuing π‐allyl cation by the tethered nitrogen atom to give, following cleavage of the allyloxycarbonyl protecting group, an approximately 5:6 mixture of the chromatographically separable diastereoisomeric spirocyclic products. In the third critical bond‐forming reaction, the iodide formed from one of the diastereoisomers underwent a radical‐addition/elimination reaction sequence that led to (?)‐ 1 in 89 % yield. The application of the same sequence of transformations to the other diastereoisomer afforded 3‐epi‐(+)‐erythramine (3‐epi‐(+)‐ 1 ).  相似文献

Novel cis‐ and trans‐2‐(p‐bromophenyl)‐5‐methylthiazolidin‐4‐ones, S,N‐containing heterocyclic compounds, were provided in a cis‐stereocomplementary and trans‐stereocomplementary synthetic manner. cis‐Selective cyclo‐condensation proceeded between 2‐sulfanylpropanoic acid (thiolactic acid) and an imine derived from 4‐bromobenzaldehyde and methylamine, whereas Ti(OiPr)4 and Ti(OiBu)4‐promoted trans‐selective cyclo‐condensation proceeded between benzyl 2‐sulfanylpropanoate and the imine. The obtained cis‐ and trans ‐ 2‐(p‐bromophenyl)‐5‐methylthiazolidin‐4‐ones were successfully converted to 2‐(3‐furyl)phenyl derivatives and bis(pinacolato)diborane derivatives utilizing Suzuki–Miyaura and Miyaura–Ishiyama cross‐coupling reactions, respectively, in an umpolung manner.  相似文献

Enantioselective iridium‐catalyzed allylic substitutions were used to prepare N‐allyl hydroxamic acid derivatives that were suitable for ring‐closing metathesis, giving N‐methoxylactams. Reactions of these derivatives with Grignard or organolithium compounds gave hemiaminals, which could be reduced diastereoselectively via acyliminium intermediates to give cis‐piperidines or cis‐pyrrolidines with substituents in the 2,6‐ or 2,5‐positions, respectively. In addition, compounds with a quaternary carbon center could be synthesized by corresponding reactions with potassium cyanide/AcOH. The procedures were applied in the syntheses of alkaloids (?)‐209D and (+)‐prosophylline.  相似文献

During the studies of hydrolysis of epoxides in water, we found that the hydrolysis of (?)‐α‐pinene oxide at 20 °C gave enantiomerically pure trans‐(?)‐sobrerol, whereas the same reaction in water heated at reflux unexpectedly gave a racemic mixture of trans‐ and cis‐sobrerol (trans/cis=6:4). We have examined this remarkable difference in detail and found that hot water, whose behavior is quite different compared with room‐ or high‐temperature water, could promote SN1 solvolysis reactions of allylic alcohols and thus caused the racemization of trans‐(?)‐sobrerol. The effect of reaction temperature, the addition of organic co‐solvent, and the concentration of the solute on the rate of the racemization of trans‐(?)‐sobrerol were further examined to understand the role that hot water played in the reaction. It was proposed that the catalytic effects of hot water are owing to its mild acidic characteristic, thermal activation, high ionizing power, and better solubility of organic reactant. Further investigation showed that the racemization of other chiral allylic/benzylic alcohols could efficiently proceed in hot water.  相似文献

The reactions of thiobenzamide 8 with diazo compounds proceeded via reactive thiocarbonyl ylides as intermediates, which underwent either a 1,5‐dipolar electrocyclization to give the corresponding five membered heterocycles, i.e., 4‐amino‐4,5‐dihydro‐1,3‐thiazole derivatives (i.e., 10a, 10b, 10c , cis‐ 10d , and trans‐ 10d ) or a 1,3‐dipolar electrocyclization to give the corresponding thiiranes as intermediates, which underwent a SNi′‐like ring opening and subsequent 5‐exo‐trig cyclization to yield the isomeric 2‐amino‐2,5‐dihydro‐1,3‐thiazole derivatives (i.e., 11a, 11b, 11c , cis‐ 11d , and trans‐ 11d ). In general, isomer 10 was formed in higher yield than isomer 11 . In the case of the reaction of 8 with diazo(phenyl)methane ( 3d ), a mixture of two pairs of diastereoisomers was formed, of which two, namely cis‐ 10d and trans‐ 10d , could be isolated as pure compounds. The isomers cis‐ 11d and trans‐ 11d remained as a mixture. In the reactions of the thioxohydrazone 9 with diazo compounds 3b and 3d , the main products were the alkenes 18 and 23 , respectively. Their formation was rationalized by a 1,3‐dipolar electrocyclization of the corresponding thiocarbonyl ylide and subsequent desulfurization of the intermediate thiiran. As minor products, 2,5‐dihydro‐1,3‐thiazol‐5‐amines 21 and 24 were obtained, which have been formed by 1,5‐dipolar electrocyclization of the thiocarbonyl ylide, followed by a 1,3‐shift of the dimethylamino group.  相似文献
