A controlled drug‐delivery system has been developed based on mesoporous silica nanoparticles that deliver anticancer drugs into cancer cells with minimized side effects. The copolymer of two oligo(ethylene glycol) macromonomers cross‐linked by the disulfide linker N,N′‐bis(acryloyl)cystamine is used to cap hollow mesoporous silica nanoparticles (HMSNs) to form a core/shell structure. The HMSN core is applied as a drug storage unit for its high drug loading capability, whereas the polymer shell is employed as a switch owing to its redox/temperature dual responses. The release behavior in vitro of doxorubicin demonstrated that the loaded drugs could be released rapidly at higher temperature or in the presence of glutathione (GSH). Thus, the dual‐stimulus polymer shell exhibiting a volume phase transition temperature higher than 37 °C can effectively avoid drug leakage in the bloodstream owing to the swollen state of the shell. Once internalized into cells, the carriers shed the polymer shell because of cleavage of the disulfide bonds by GSH, which results in the release of the loaded drugs in cytosol. This work may prove to be a significant development in on‐demand drug release systems for cancer therapy.  相似文献

The synthesis and characterization of two new capped silica mesoporous nanoparticles for controlled delivery purposes are described. Capped hybrid systems consist of MCM‐41 nanoparticles functionalized on the outer surface with polymer ε‐poly‐L ‐lysine by two different anchoring strategies. In both cases, nanoparticles were loaded with model dye molecule [Ru(bipy)3]2+. An anchoring strategy involved the random formation of urea bonds by the treatment of propyl isocyanate‐functionalized MCM‐41 nanoparticles with the lysine amino groups located on the ε‐poly‐L ‐lysine backbone (solid Ru‐rLys‐S1 ). The second strategy involved a specific attachment through the carboxyl terminus of the polypeptide with azidopropyl‐functionalized MCM‐41 nanoparticles (solid Ru‐tLys‐S1 ). Once synthesized, both nanoparticles showed a nearly zero cargo release in water due to the coverage of the nanoparticle surface by polymer ε‐poly‐L ‐lysine. In contrast, a remarkable payload delivery was observed in the presence of proteases due to the hydrolysis of the polymer’s amide bonds. Once chemically characterized, studies of the viability and the lysosomal enzyme‐controlled release of the dye in intracellular media were carried out. Finally, the possibility of using these materials as drug‐delivery systems was tested by preparing the corresponding ε‐poly‐L ‐lysine capped mesoporous silica nanoparticles loaded with cytotoxic drug camptothecin (CPT), CPT‐rLys‐S1 and CPT‐tLys‐S1 . Cellular uptake and cell‐death induction were studied. The efficiency of both nanoparticles as new potential platforms for cancer treatment was demonstrated.  相似文献

The amphiphilic polymer‐grafted silica was newly prepared as a stationary phase in high‐performance liquid chromatography. Poly(4‐vinylpyridine) with a trimethoxysilyl group at one end was grafted onto porous silica particles and the pyridyl side chains were quaternized with 1‐bromooctadecane. The obtained poly(octadecylpyridinium)‐grafted silica was characterized by elemental analysis, diffuse reflectance infrared Fourier transform spectroscopy and Brunauer–Emmett–Teller analysis. The degree of quaternization of the pyridyl groups on the obtained stationary phase was estimated to be 70%. The selective retention behaviors of polycyclic aromatic hydrocarbons including some positional isomers were investigated using poly(octadecylpyridinium)‐grafted silica as an amphiphilic polymer stationary phase in high‐performance liquid chromatography and results were compared with commercially available polymeric octadecylated silica and phenyl‐bonded silica columns. The results indicate that the selectivity toward polycyclic aromatic hydrocarbons exhibited by the amphiphilic polymer stationary phase is higher than the corresponding selectivity exhibited by a conventional phenyl‐bonded silica column. However, compared with the polymeric octadecylated silica phase, the new stationary phase presents similar retention behavior for polycyclic aromatic hydrocarbons but different retention behavior particularly for positional isomers of disubstituted benzenes as the aggregation structure of amphiphilic polymers on the surface of silica substrate has been altered during mobile phase variation.  相似文献

Particle monolayer formation at the air–water interface by polymer‐grafted colloidal silica was investigated. Methyl methacrylate (MMA) was polymerized from initiative bromide groups at colloidal silica surface by atom transfer radical polymerization. We obtained polymer‐grafted silica particle (SiO2‐PMMA) with relative narrow polydispersity of PMMA. For the polymer‐grafted particle with high graft density, particle monolayer formation was confirmed by π‐A isotherm measurement and SEM observation. Interparticle distance was controllable by surface pressure. Furthermore, grafted polymer chains were suggested to be fairly extended at the air–water interface. However, for the polymer‐grafted particle with low graft density, monolayer structure on substrate showed aggregation and voids. © 2006 Wiley Periodicals, Inc. Acrylic polymer/silica hybrids were prepared by emulsifier‐free emulsion polymerization and the sol–gel process. Acrylic polymer emulsions containing triethoxysilyl groups were synthesized by emulsifier‐free batch emulsion polymerization. The acrylic polymer/silica hybrid films prepared from the acrylic polymer emulsions and tetraethoxysilane (TEOS) were transparent and solvent‐resistant. Atomic force microscopy studies of the hybrid film surface suggested that the hybrid films did not contain large (e.g., micrometer‐size) silica particles, which could be formed because of the organic–inorganic phase separation. The Si? O? Si bond formed by the cocondensation of TEOS and the triethoxysilyl groups on the acrylic polymer increased the miscibility between the acrylic polymer component and the silica component in the hybrid films, in which the nanometer‐size silica domains (particles) were dispersed homogeneously in the acrylic polymer component. © 2005 Wiley Periodicals, Inc. Silica–polystyrene core‐shell particles were successfully prepared by surface‐mediated reversible addition fragmentation chain transfer (RAFT) polymerization of styrene monomer from the surfaces of the silica‐supported RAFT agents. Initially, macro‐RAFT agents were synthesized by RAFT polymerization of γ‐methacryloxypropyltrimethoxysilane (MPS) in the presence of chain transfer agents (CTAs). Immobilization of CTAs onto the silica surfaces was then performed by reacting silica with macro‐RAFT agents via a silane coupling. Grafting of polymer onto silica forms core‐shell nanostructures and shows a sharp contrast between silica core and polymer shell in the phase composition. The thickness of grafted‐polymer shell and the diameter of core‐shell particles increase with the increasing ratio of monomer to silica. A control experiment was carried out by conventional free radical emulsion copolymerization of MPS‐grafted silica and styrene under comparable conditions. The resulting data provide further insight into the chemical composition of grafted‐polymers that are grown from the silica surface through RAFT process. © 2008 Wiley Periodicals, Inc. Summary: Carbon nanotubes (CNTs) have been grown on MCM‐41 supported Fe nanoparticles and the as‐prepared (no further purification) CNT‐silica hybrid was directly incorporated into nylon‐6 (PA6) by simple melt‐compounding. The urchin‐shaped CNT‐silica hybrid filler was observed to be homogeneously dispersed throughout the matrix by scanning electron and transmission electron microscopy. Compared with neat PA6, the tensile modulus and strength of the composite are greatly improved by about 110%, with incorporation of only 1 wt.‐% CNT‐silica filler.

Mesoporous silica nanoparticles (MSN) were coated by pH‐responsive polymer chitosan‐poly (methacrylic acid) (CS‐PMAA). This nano drug delivery system showed good application prospects and the polymer‐coated microspheres were promising site‐specific anticancer drug delivery carriers in biomedical field. A continuous detection of pH‐responsive drug delivery system in cells in situ, utilizing MSN/CS‐PMAA composite microspheres, was proposed. Two kinds of different cell lines, tumor cell line (Hela) and normal somatic cells (293T), were used to investigate the behaviours of the drug loaded system in the cells. Conclusions could be drawn from the fluorescent images obtained by confocal laser scanning microscopy (CLSM), modified drug‐loaded microspheres (MSN/CS‐PMAA) were ingested into cells more easily, the uptake of DOX@FITC‐MSN/CS‐PMAA by HeLa/293T cells were performed at pH 7.4/pH 6.8, DOX was released during the ingestion process, fluorescence intensity decreased with time because of efflux transport and photo‐bleaching. Fluoresence detection by flow cytometry was performed as comparison. The continuous fluorescent observation in situ could be widely used in the pH‐responsive releasing process of drug delivery system in the cells.  相似文献

The objective of this study is to utilize the pH sensitivity of modified mesoporous silica nanoparticles (MSN) for oral drug delivery. In the first time, a pH‐sensitive ionic liquid was synthesized through the quaternization of 3‐aminopropyltrimethoxysilane (3‐ATMS) with sodium monochloroacetate (SMCA). Then, silica nanoparticle was modified by this pH‐sensitive ionic liquid and converted to a pH‐sensitive positive‐charge silica nanoparticle (PCSN). The nanoparticle was characterized by FTIR and SEM. Naproxen as anionic drug molecules was entrapped in this pH‐sensitive positive‐charge silica nanoparticles (PCSN) and the in vitro release profiles were established separately in both (SGF, pH 1) and (SIF, pH 7.4).  相似文献

Scale‐up synthesis of hyperbranched poly(amidoamine)‐grafted ultrafine silica was successfully achieved by using dendrimer synthesis methodology in solvent‐free dry‐system. The poly(amidoamine) was allowed to grow from silica surface by repeating two steps: (1) Michael addition of methyl acrylate (MA) to amino group on the surface and (2) amidation of terminal ester group with ethylenediamine (EDA). MA was sprayed onto silica having amino group and the silica agitated at 300 rpm at 50 °C. After the reaction, unreacted MA was removed under vacuum. Then EDA was sprayed and the reaction was conducted at 50 °C with agitation. After the reaction, unreacted EDA was also removed under vacuum at 50 °C and MA was sprayed again. The percentage of poly(amidoamine) grafting onto the surface was determined to be 141% with repeated reaction cycles of eight‐times. However, the value was considerably smaller than that of the theoretical value. This indicates that the propagation of poly(amidoamine) dendron from silica surface was not achieved theoretically and hyperbranched poly(amidoamine) was grafted onto the surface because of steric hindrance of grafted polymer. In addition, the effect of initial amino group content on the growth of poly(amidoamine) from the surface was investigated. It was concluded that the method is suitable for the scale‐up synthesis of hyperbranched poly(amidoamine)‐grafted silica. Copyright © 2002 John Wiley & Sons, Ltd.  相似文献

The effects of filler loading and a new silane coupling agent 3‐octanoylthio‐1‐ propyltriethoxysilane (NXT silane) on the polymer‐filler interaction and mechanical properties of silica‐filled and carbon black‐filled natural rubber (NR) compounds were studied. Silica (high dispersion silica7000GR, VN2, and VN3) and carbon black (N330) were used as the fillers, and the loading range was from 0 to 50 phr. The loading of NXT silane was from 0 to 6 phr. Experimental results show that the maximum and minimum torques of silica and carbon black‐filled NR increase with increasing filler loading. With increasing filler loading, the scorch time and optimum cure time decrease for carbon black‐filled NR, but increase for silica‐filled NR. The minimum torque, scorch time, and optimum cure time decrease because of the presence of NXT silane. For the carbon black and silica‐filled NR, the tensile strength and elongation at break have maximum values, but the hardness, M300, M100, and tear strength keep increasing with filler loading. The mechanical properties of silica‐filled NR were improved in the presence of NXT silane. With increasing filler loading, the storage modulus of filled NR increases, but the loss factor decreases. The effects of radicals on silica surface, which were formed by γ‐ray irradiation, on the polymerization of vinyl monomers were investigated. It was found that the polymerization of styrene was remarkably retarded in the presence of γ‐ray‐irradiated silica above 60 °C, at which thermal polymerization of styrene is readily initiated. During the polymerization, a part of polystyrene formed was grafted onto the silica surface but percentage of grafting was very small. On the other hand, no retardation of the polymerization of styrene was observed in the presence of γ‐ray‐irradiated silica below 50 °C; the polymerization tends to accelerate and polystyrene was grafted onto the silica surface. Poly(vinyl acetate) and poly(methyl methacrylate) (MMA) were also grafted onto the surface during the polymerization in the presence of γ‐ray‐irradiated silica. The grafting of polymers onto the silica surface was confirmed by thermal decomposition GC‐MS. It was considered that at lower temperature, the grafting based on the propagation of polystyrene from surface radical (“grafting from” mechanism) preferentially proceeded. On the contrary, at higher temperature, the coupling reaction of propagating polymer radicals with surface radicals (“grafting onto” mechanism) proceeded to give relatively higher molecular weight polymer‐grafted silica. © 2006 Wiley Periodicals, Inc. Adsorbed poly(ethylene‐stat‐vinyl acetate) (PEVAc) on fumed silica was studied using temperature‐modulated differential scanning calorimetry (TMDSC) and FT‐IR spectroscopy. The properties of the copolymers were compared with poly(vinyl acetate) (PVAc) and low density polyethylene (LDPE) as references. TMDSC analysis of the copolymer‐silica samples in the glass transition region was complicated for the copolymers because of the ethylene crystallinity. Nevertheless, examination of the glass transition region for small adsorbed amounts of these copolymers indicated the presence of tightly‐ and loosely‐bound polymer segments, similar to other polymers which have an attraction to silica. Compared with bulk polymers with the same composition, the tightly‐bound polymers showed an increased glass transition temperature (Tg) and a loosely‐bound fraction with a lower Tg than bulk. FT‐IR spectra of the surface copolymers indicated that the fraction of bound carbonyls (p) increased as the fraction of vinyl acetate in the copolymers decreased, consistent with the notion that the carbonyls from vinyl acetate preferentially find their way to the silica surface. Spectra from samples with different adsorbed amounts of polymer were used to obtain the amount of bound polymer (Mb) and the ratio of molar absorption coefficients of bound carbonyls to free carbonyls (X). The copolymers had very large p values (up to 0.8) at small adsorbed amounts and dependent on the composition of the polymer. However, an analysis of the bound fractions, based on only the vinyl acetate groups, superimposed the data, suggesting that the ethylene units simply dilute the vinyl acetate groups in the surface polymer. The sample with the smallest fraction of vinyl acetate did not show this behavior and may be considered to be “carbonyl poor.” © 2014 Wiley Periodicals, Inc. J. Polym. Sci., Part B: Polym. Solution impregnations, pulltrusion and film stacking are widely used methods to prepare thermoplastic composite materials. Extruders are used to melt the polymer and to incorporate fibers into the polymer in order to modify physical properties. In this article, the compounding of colloidal silica nanoparticles filled polyamide‐6 (PA‐6) is achieved using a twin‐screw extruder, which has a significant market share due to its low cost and easy maintenance. The experiments were performed at 250 rpm and the bulk throughput was 6 kg h?1 with a pump pressure of 30 bars. The composites were characterized with nuclear magnetic resonance (NMR), wide angle X‐ray diffraction (WAXD), differential scanning calorimetry (DSC) and transmission electron microscopy (TEM). As determined by WAXD, the PA‐6 showed higher amounts of γ‐phase when compared to other synthesis methods such as in situ polymerization. TEM pictures showed that the silica particles aggregated nevertheless, upon addition of 14% (w/w) silica the E‐modulus increased from 2.7 to 3.9 GPa indicating that an effective mechanical coupling with the polymer was achieved. The behavior, illustrated with dynamic mechanical analysis (DMA) curves, indicated that in general when a filled system is compared to unfilled material, the values of the moduli (E′ and E″) increased and tan δ decreased. Determination of molecular mass distribution of the samples by means of size exclusion chromatography (SEC) coupled to a refractive index (RI), viscosity (DV) and light scattering (LS) detector revealed that the addition of silica did not decrease the average molecular weight of the polymer matrix, which is of importance for composite applications. The development of silica‐filling elastomers with high mechanical performance and good processability is still a great challenge. In this study, we fabricated siloxane‐grafted atactic 1,2‐polybutadiene (1,2‐PB) rubber through grafting poly(1,3‐butadiene)‐block‐(dimethylsiloxane) (PB‐b‐PDMS) onto 1,2‐PB molecular chains by coordination polymerization using a molybdenum (Mo)‐based catalyst system. The PB‐b‐PDMS with active double bonds was synthesized by anionic polymerization. Fourier transform infrared analysis (FTIR), elementary analysis, and GPC‐MALLS‐viscometer analyses verified the incorporation of PB‐b‐PDMS and the grafting structure in the resulting polymer. Scanning electron microscope (SEM), bound rubber testing, and dynamic mechanical analysis demonstrated that the graft‐modification with PB‐b‐PDMS improved silica dispersity in the 1,2‐PB matrix because the incorporation of siloxane groups provided stronger interfacial interaction with silica. Meanwhile, the graft‐modified 1,2‐PB exhibited lower Mooney viscosity, higher tensile strength, and lower heat build‐up than unmodified 1,2‐PB. This concept provides novel inspiration for the preparation of advanced rubber with promoted silica compatibility and mechanical performance.  相似文献

A versatile one‐pot strategy for the preparation of reversibly cross‐linked polymer‐coated mesoporous silica nanoparticles (MSNs) via surface reversible addition–fragmentation chain transfer (RAFT) polymerization is presented for the first time in this paper. The less reactive monomer oligo(ethylene glycol) acrylate (OEGA) and the more reactive cross‐linker N,N′‐cystaminebismethacrylamide (CBMA) are chosen to be copolymerized on the external surfaces of RAFT agent‐functionalized MSNs to form the cross‐linked polymer shells. Owing to the reversible cleavage and restoration of disulfide bonds via reduction/oxidation reactions, the polymer shells can control the on/off switching of the nanopores and regulate the drug loading and release. The redox‐responsive release of doxorubicin (DOX) from this drug carrier is realized. The protein adsorption, in vitro cytotoxicity assays, and endocytosis studies demonstrate that this biocompatible vehicle is a potential candidate for delivering drugs. It is expected that this versatile grafting strategy may help fabricate satisfying MSN‐based drug delivery systems for clinical application.

Bromoacetate‐substituted [3‐(2‐O‐β‐cyclodextrin)‐2‐hydroxypropoxy]propylsilyl‐appended silica particles (BACD‐HPS), an important and useful synthetic intermediate for preparation of novel types of macrocycles‐capped β‐CD‐bonded silica particles including crown ether/cyclam/calix[4]arene‐capped β‐CD‐bonded silica particles, have been prepared and used as chiral stationary phase for HPLC. This synthetic stationary phase is characterized by means of elemental analysis. For the first time, the chromatographic behavior of BACD‐HPS was systematically evaluated with several disubstituted benzenes and some chiral drug compounds under both normal and RP conditions in HPLC. The results show that BACD‐HPS has excellent selectivity for the separation of aromatic positional isomers and chiral isomers of some drug compounds when used as stationary phase in HPLC.  相似文献

The synthesis of polymer–drug conjugates from prodrug monomers consisting of a cyclic polymerizable group that is appended to a drug through a cleavable linker is achieved by organocatalyzed ring‐opening polymerization. The monomers polymerize into well‐defined polymer prodrugs that are designed to self‐assemble into nanoparticles and release the drug in response to a physiologically relevant stimulus. This method is compatible with structurally diverse drugs and allows different drugs to be copolymerized with quantitative conversion of the monomers. The drug loading can be controlled by adjusting the monomer(s)/initiator feed ratio and drug release can be encoded into the polymer by the choice of linker. Initiating these monomers from a poly(ethylene glycol) macroinitiator results in amphiphilic diblock copolymers that spontaneously self‐assemble into micelles with a long plasma circulation, which is useful for systemic therapy.  相似文献

Crystallization in a series of variable crosslink density poly(dimethyl‐diphenyl)siloxanes random block copolymers reinforced through a mixture of precipitated and fumed silica fillers has been studied by differential scanning calorimetry (DSC), dynamic mechanical analysis (DMA), nuclear magnetic resonance (NMR), and X‐ray diffraction (XRD). The silicone composite studied was composed of 94.6 mol % dimethoylsiloxane, 5.1 mol % diphenylsiloxane, and 0.3 mol % methyl‐vinyl siloxane (which formed crosslinking after peroxide cure). The polymer was filled with a mixture of 21.6 wt % fumed silica and 4.0 wt % precipitated silica previously treated with 6.8 wt % ethoxy‐end‐blocked siloxane processing aid. Molecular weight between crosslinks and filler–polymer interaction strength were modified by exposure to γ‐irradiation in either air or in vacuo. Isothermal DMA experiments illustrated that crystallization at ?85 °C occurred over a 1.8 hour period in silica‐filled systems and 2.2–2.6 hours in unfilled systems. The crystallization kinetics for irradiated samples were found to be dependent on crosslink density. Irradiation in vacuo resulted in faster overall crystallization rates compared to air irradiation for the same crosslink density, likely due to a reduction in the interaction between the polymer chains and the silica filler surface for samples irradiated in air. © 2006 Wiley Periodicals, Inc. The main goal of the present study was to produce insights from the atomistic modeling into the structural changes in elastomer‐based polymer nanocomposites caused by the surface modifications of the filler particles, and by the crosslinking of the participating polymer matrix. The fully atomistic molecular‐dynamics computer simulations of crosslinked (1,4) cis‐polyisoprene (PI) films capped by amorphous silica substrates was set‐up, in the presence of realistic coupling and covering agents. The PI film stratified structure has been studied in the proximity of the corresponding glass transition, by varying the degree of crosslinking and the PI film thickness. Some monomer ordering induced by the pristine (bare) silica disappeared almost completely in films with modified surfaces. The average monomer density increased with degree of crosslinking. As compared with PI bulk, the glass‐transition temperature was slightly larger for highly crosslinked PI films with bare silica and surfaces with coupling agents, and increased with increasing confinement. © 2015 Wiley Periodicals, Inc.
