The ability of proteins to provide stability in foams is greatly influenced by their interfacial dilatational rheological properties. Surface tension response of a pulsatingbubble with an adsorbed layer of beta-lactoglobulin was measured for different frequencies and protein concentrations using a pulsating bubble tensiometer. A methodology, accounting for adsorption/desorption as well as variation of surface concentration due to expansion/contraction, was developed for the evaluation of surface dilatational elasticity and viscosity at different frequencies from these measurements. The adsorption rate constants were inferred from the surface pressure dynamics of protein adsorption using a Langmuir minitrough. The desorption rates were shown to be negligible for beta-lactoglobulin from the surface pressure response of a spread monolayer when subjected to compression in a Langmuir minitrough. The proposed model was employed to infer the interfacial dilatational viscosity and elasticity of an adsorbed beta-lactoglobulin layer at the air-water interface from experimental pulsating bubble data for protein concentrations in the range of 0.01-0.5 wt % at pH 7. As expected, the interfacial dilatational rheological properties were found to be higher at higher protein concentrations, this effect being less pronounced for dilatational elasticity. Heating at 80 degrees C for 30 min was found to result in higher interfacial dilatational viscosity and lower interfacial dilatational elasticity though this difference was within experimental error. The traditional approach for the inference of interfacial dilatational rheological properties is found to overpredict the interfacial dilatational elasticity whereas the viscosity values do not differ significantly from those obtained using the current analysis.  相似文献

This report aims at (i) presenting a quantitative interpretation of interfacial dilational moduli (|E|) for four proteins at three different interfaces and (ii) identifying the main parameters responsible. The proteins were adsorbed from aqueous solution against air, n-tetradecane and sunflower seed oil, as a function of protein concentration and adsorption time.Experimentally, a dynamic drop tensiometer is a convenient instrument to generate the required sinusoidal oscillations for compression/expansion of interfaces (Benjamins et al., 1996 [1]).Theoretically, a simple two-dimensional solution model with a constant molecular area of the protein described the data only at fairly low pressures. Much better agreement over the entire elastic range was found with a recent extension of the model. This extension accounted for adsorbed proteins adopting smaller molecular areas with increasing surface pressure.Three factors dominated the values of the dilational modulus: (i) rigidity of protein molecules, (ii) degree of interfacial non-ideality and (iii) tension of the clean interface (Benjamins et al., 2006 [2]). The last factor is clearly of great relevance to food emulsions.For each protein at different interfaces, the elasticity increased with the enthalpy parameter (ΗS) of the equation of state. Elasticity and ΗS both increased with the clean-interface tension, γ0, i.e., with decreasing polarity of the interface (Benjamins et al., 2006 [2]; Fainerman et al., 2003 [3]). The elasticity of the different proteins also increased with increasing rigidity of the molecules, indicating a lower compressibility of the molecular area at the interface.Pure viscosities were never observed in our experience. However, viscoelastic behaviour was found at high pressures, i.e., in densely packed surfaces. The measured viscous phase angles strongly decreased at still higher pressures, indicating that the active relaxation mechanism slowed down with increasing molecular packing density. Specific kinetic models are yet to be developed for such mechanisms.  相似文献

The adsorption behaviour of proteins and systems mixed with surfactants of different nature is described. In the absence of surfactants the proteins mainly adsorb in a diffusion controlled manner. Due to lack of quantitative models the experimental results are discussed partly qualitatively. There are different types of interaction between proteins and surfactant molecules. These interactions lead to protein/surfactant complexes the surface activity and conformation of which are different from those of the pure protein. Complexes formed with ionic surfactants via electrostatic interaction have usually a higher surface activity, which becomes evident from the more than additive surface pressure increase. The presence of only small amounts of ionic surfactants can significantly modify the structure of adsorbed proteins. With increasing amounts of ionic surfactants, however, an opposite effect is reached as due to hydrophobic interaction and the complexes become less surface active and can be displaced from the interface due to competitive adsorption. In the presence of non-ionic surfactants the adsorption layer is mainly formed by competitive adsorption between the compounds and the only interaction is of hydrophobic nature. Such complexes are typically less surface active than the pure protein. From a certain surfactant concentration of the interface is covered almost exclusively by the non-ionic surfactant. Mixed layers of proteins and lipids formed by penetration at the water/air or by competitive adsorption at the water/chloroform interface are formed such that at a certain pressure the components start to separate. Using Brewster angle microscopy in penetration experiments of proteins into lipid monolayers this interfacial separation can be visualised. A brief comparison of the protein adsorption at the water/air and water/n-tetradecane shows that the adsorbed amount at the water/oil interface is much stronger and the change in interfacial tension much larger than at the water/air interface. Also some experimental data on the dilational elasticity of proteins at both interfaces measured by a transient relaxation technique are discussed on the basis of the derived thermodynamic model. As a fast developing field of application the use of surface tensiometry and rheometry of mixed protein/surfactant mixed layers is demonstrated as a new tool in the diagnostics of various diseases and for monitoring the progress of therapies.  相似文献

Adsorption and desorption dynamics of lysozyme and β-casein at the air/water interface were investigated through stress relaxation experiments. The resulting surface tension changes due to a step-type surface area disturbance, as a function of time, were measured through a capillary wave probe. The adsorption data, obtained after a surface area expansion, can be well fitted to a diffusion-controlled adsorption model. However, desorption relaxation following a surface compression is much slower and cannot be modeled by the diffusion theory. Characteristic diffusion frequency and high-frequency dilational elasticity for protein layers were also obtained and found to be consistent with data reported in the literature.  相似文献

Various experimental methods were used to investigate interaction between polymer and anionic/nonionic surfactants and mechanisms of enhanced oil recovery by anionic/nonionic surfactants in the present paper. The complex surfactant molecules are adsorbed in the mixed micelles or aggregates formed by the hydrophobic association of hydrophobic groups of polymers, making the surfactant molecules at oil-water interface reduce and the value of interfacial tension between oil and water increase. A dense spatial network structure is formed by the interaction between the mixed aggregates and hydrophobic groups of the polymer molecular chains, making the hydrodynamic volume of the aggregates and the viscosity of the polymer solution increase. Because of the formation of the mixed adsorption layer at oil and water interface by synergistic effect, ultra-low interfacial tension (~2.0?×?10?3 mN/m) can be achieved between the novel surfactant system and the oil samples in this paper. Because of hydrophobic interaction, wettability alteration of oil-wet surface was induced by the adsorption of the surfactant system on the solid surface. Moreover, the studied surfactant system had a certain degree of spontaneous emulsification ability (D50?=?25.04?µm) and was well emulsified with crude oil after the mechanical oscillation (D50?=?4.27?µm).  相似文献

The surface properties of mixed system containing gemini anionic surfactant 1,2,3,4-butanetetracarboxylic sodium, 2,3-didodecyl ester and partly hydrolyzed polyacrylamide were investigated by surface tension measurements and oscillating bubble methods. The influences of surfactant concentration, dilational frequency, temperature, pH, as well as salts on dilational modulus were explored. Meanwhile, the interfacial tension relaxation method was employed to obtain the characteristic time of surface relaxation process. The polymers play important roles in changing the interfacial properties especially at lower surfactant concentration. The possible mechanism of the polymer in changing the interfacial properties is proposed. Both the hydrophobic and electrostatic interaction among the surfactants and polymers dominate the surface properties of mixed system. These dynamic properties are of fundamental interest in understanding the structure of adsorption layers, dynamics of surfactant molecules, and their interaction with polymers at the surface.  相似文献

Interfacial tension changes during protein adsorption at both the solid-liquid and the liquid-vapor interface were measured simultaneously by ADSA-P from sessile droplets of protein solutions on fluoroethylenepropylene-Teflon. Four globular proteins of similar size, viz. lysozyme, ribonuclease, -lactalbumin and Ca2+-free -lactalbumin, and one larger protein, serum albumin, were adsorbed from phosphate solutions at varying pH values (pH 3-12). The kinetics of the interfacial tension changes were described using a model accounting for diffusion-controlled adsorption of protein molecules and conformational changes of already adsorbed molecules. The contribution of conformational changes to the equilibrium interfacial pressure was shown to be relatively small and constant with respect to pH when compared to the contribution of adsorption of the protein molecules. The model also yields the diffusion relaxation time and the rate constant for the conformational changes at the interface. Around the isoelectric point of a protein the calculated diffusion relaxation time was minimal, which is ascribed to the absence of an energy barrier to adsorption. Energy barriers to adsorption become larger at pH values away from the isoelectric point and can therefore become rate-limiting for the adsorption process. The rate constants for conformational changes at the liquid-vapor interface were maximal around the isoelectric point of a protein, suggesting a smaller structural stability of the adsorbed protein. At the solid-liquid interface the rate constants were smaller and independent of pH. indicating that conformational changes more readily occur at the liquid-vapor than at the solid-liquid interface.  相似文献

We examined the adsorption, on hydrophobic and hydrophilic surfaces, of 4 rake-type poly(dimethyl siloxane) (PDMS) copolymers varying the amount of poly(ethylene glycol) (PEG) graft arms from 41 to 72%. The copolymers formed large aggregates in solution, complicating their adsorption kinetics and layer structures. We found the adsorption process always to be dominated by the adsorption of large aggregates, with strongly bound layers resistant to rinsing in adsorbing buffer. Adsorbed amounts were nearly independent of the substrate. However, subtleties in the adsorption kinetics suggested different layer structures for the different systems. On hydrophilic silica, aggregates adsorbed at the transport limited rate until surface saturation, and associated interfacial structures were likely retained. On the hydrophobic surface, a subset of the copolymers exhibited retarded late stage adsorption kinetics suggestive of brush formation. This work demonstrates how subtle differences in adsorption kinetics provide insight into potential interfacial layer structures.  相似文献

The interfacial film of physically adsorbed ionic amphiphilic molecules on submicron particles dispersed in water was studied by a combination of surface tension measurements, laser light scattering (LLS) and high-shear experiments in a microchannel. General features in the structure and morphology of the molecular film are identified and understood in the framework of the two-step Langmuir adsorption model deduced from the adsorption isotherm. On the basis of this approach, the phase transitions and structural ordering of the film at the solid-liquid interface are analyzed in detail. A novel methodology based on high-shear aggregation experiments subsequently analyzed by means of LLS is proposed and turns out to be able to provide significant information on the phase transitions and structural arrangements of the adsorbed molecules (in substantial agreement with the adsorption isotherm model) as well as on the resulting interactions. Particularly important for applications is the result that, with no added salt, the films on two particles can adhere/fuse, leading to aggregation as long as an uncovered (hydrophobic) patch is present (unsaturated molecular layers). In the opposite case of fully developed layers, by analyzing the mechanism of shear aggregation of charged particles in the low-salt limit, we show that, when the hydrophobic attraction is absent, short-range hydration repulsive forces dominate over Derjaguin-Landau-Verwey-Overbeek (DLVO) forces and adhesion can never be achieved even upon application of very high collision energies. Consistently, a lower limiting boundary for the hydration interaction is calculated and found to be in agreement with data in the literature.  相似文献

Dynamic interfacial tensions and surface dilational moduli were measured for four proteins at three fluid interfaces, as a function of time and concentration. The proteins-beta-casein, beta-lactoglobulin, bovine serum albumin, and ovalbumin-were adsorbed from aqueous solution against air, n-tetradecane, and a triacylglycerol oil. The sinusoidal interfacial compression/expansion, at frequencies ranging from 0.005 to 0.5 Hz, was effected in a dynamic drop tensiometer suited to viscous oil phases. Generally, at interfacial pressures up to 15 mN/m, dilational moduli were purely elastic at frequencies from 0.1 Hz. In this elastic range, in-surface relaxation either was essentially completed or had not yet started within a time on the order of 10 s. Within this time span, protein exchange with the bulk solution was negligible. In cases where in-surface relaxation was completed in the imposed time, the moduli depended only on the equilibrium Pi(Gamma) relationship. We interpret these results in terms of a simple two-dimensional solution model, based on a Gibbs dividing surface, accounting for nonideal mixing to the first order with respect to both entropy and enthalpy. Interfacial mixing enthalpy is shown to have a major effect on the elasticity, with both quantities increasing in the sequence triacylglycerol < tetradecane < air. We also suggest a strong correlation between enthalpy and clean-interface tension that increases in the same order. At each interface, the enthalpy increases with increasing molecular rigidity: beta-casein < beta-lactoglobulin < bovine serum albumin < ovalbumin. Best agreement with the experimental data was obtained with a recently extended version of the model accounting for proteins adopting smaller molecular areas with increasing surface pressure. For interfacial pressures above 15 mN/m, the moduli were generally no longer purely elastic, with viscous loss angles ranging up to 36 degrees. In this range of high pressures, the moduli depended on relaxation mechanisms for which specific kinetic models must be developed.  相似文献

This study investigated the roles of gamma-globulin in the dynamic interfacial behavior of dipalmitoyl phosphatidylcholine (DPPC)/gamma-globulin monolayers at air/liquid interfaces at 25 degrees C. The surface tension behavior demonstrated that gamma-globulin had a large adsorption time scale. Moreover, the surface pressure-area hysteresis behavior of adsorbed gamma-globulin monolayers suggested that no significant desorption occurred during the compression stage, and the respreading of gamma-globulin molecules at the interface during the expansion stage was slow. From the hysteresis behavior of adsorbed gamma-globulin monolayers with spread DPPC molecules, it was found that gamma-globulin molecules were expelled from the interface as DPPC molecules were in a condensed state. The squeeze-out of gamma-globulin molecules seemed to induce the loss of DPPC molecules at the interface with the extent depending on the initial gamma-globulin surface concentration. Furthermore, the expelled gamma-globulin molecules re-entered the monolayer and participated in the surface pressure increase during the following expansion stage. The exclusion of gamma-globulin associated with the removal of DPPC during monolayer compression and the re-entry of gamma-globulin during subsequent monolayer expansion represented a mechanism for DPPC depletion and gamma-globulin enrichment at the interface, which may explain the inhibitory effect of certain proteins on the surface activity of DPPC. The properties and structure of different types of interfacial layers obtained from aqueous dispersions of nanometric silica and palmitic acid (PA) have been studied and characterized by different diagnostics and measurements. The investigations concern PA monolayers spread on the silica dispersions, dispersions in contact with PA solutions in oil and silica dispersions containing PA, aiming at elucidating the role of the PA interaction with the particles and investigating the surface-activity of the originated silica-PA complexes. Drop shape tensiometry was utilized to measure the dynamic surface and interfacial tension while a Langmuir trough apparatus was used to obtain compression isotherms of the spread PA layers and to measure the dilational viscoelasticity according to the oscillating barrier method. Brewster angle microscopy and ellipsometry were utilized to investigate the lateral and vertical structure of the interfacial layers. From this multifold approach emerges a complex picture of the features of these interfacial layers that can be rationalized on the basis of the adsorption of PA on the particle surface. The results evidence a threshold in PA adsorption above which particles change from hydrophilic to partially hydrophobic, promoting their incorporation into the interfacial layer.  相似文献

Adsorbed films of proteins at the air-water interface have been imaged using Brewster angle microscopy (BAM). The proteins beta-lactoglobulin (beta-L) and ovalbumin (OA) were studied at a range of protein concentrations and surface ages at 25.0 degrees C and two pH values (7 and 5) in a Langmuir trough. The adsorbed films were periodically subjected to compression and expansion cycles such that the film area was typically varied between 125% and 50% of the original film area. With beta-L on its own, no structural changes were observable at pH 7. When a low-area fraction (less than 0.01%) of 20 mum polystyrene latex particles was spread at the interface before adsorption of beta-L, the particles became randomly distributed throughout the interface, but after protein adsorption and compression/expansion, the particles highlighted notable structural features not visible in their absence. Such features included the appearance of long (several hundred micrometers or more) folds and cracks in the films, generally oriented at right angles to the direction of compression, and also aggregates of protein and/or particles. Such structuring was more visible the longer the film was aged or at higher initial protein concentrations for shorter adsorption times. At pH 5, close to the isoelectric pH of beta-L, such features were just noticeable in the absence of particles but were much more pronounced than at pH 7 in the presence of particles. Similar experiments with OA revealed even more pronounced structural features, both in the absence and presence of particles, particularly at pH 5 (close to the isoelectric pH of OA also), producing striking stripelike and meshlike domains. Changes in the dilatational elasticity of the films could be correlated with the variations in the structural integrity of the films as observed via BAM. The results indicate that interfacial area changes of this type, typical of those that occur in food colloid processing, will lead to highly inhomogeneous adsorbed protein layers, with implications for the stability of the corresponding foams and emulsions stabilized by such films. Overall, the experimental results are in broad agreement with the sorts of trends predicted by earlier computer simulations of protein films subjected to such compression and expansion.  相似文献

The formation of protein layers during competitive adsorption was studied with ellipsometry. Single, binary, and ternary protein solutions of human serum albumin (HSA), IgG, and fibrinogen (Fgn) were investigated at concentrations corresponding to blood plasma diluted 1/100. As a model surface, hydrophobic hexamethyldisiloxane (HMDSO) plasma polymer modified silica was used. By using multiambient media measurements of the bare substrate prior to protein adsorption the adsorbed amount as well as the thickness and refractive index of the adsorbed protein layer could be followedin situand in real time. Under conditions used in these experiments neither IgG nor fibrinogen could fully displace serum albumin from the interface. The buildup of the protein layer occurred via different mechanisms for the different protein systems. Fgn adsorbed in a rather flat orientation at low adsorbed amounts, while at higher surface coverage the protein reoriented to a more upright orientation in order to accommodate more molecules in the adsorbed layer. IgG adsorption proceeded mainly end-on with little reorientation or conformational change on adsorption. Finally, for HSA an adsorbed layer thickness greater than the molecular dimensions was observed at high concentrations (although not at low), indicating that aggregates or multilayers formed on HMDSO plasma polymer surfaces. For all protein mixtures the adsorbed layer structure and buildup indicated that Fgn was the protein dominating the adsorbed layer, although HSA partially blocked the adsorption of this protein. At high surface concentration, HSA/Fgn mixtures show an abrupt change in both adsorbed layer thickness and refractive index suggesting, e.g., an interfacial phase transition of the mixed protein layer. A similar but less pronounced behavior was observed for HSA/IgG. For IgG/Fgn and HSA/IgG/Fgn a buildup of the adsorbed layer similar to that displayed by Fgn alone was observed.  相似文献

The dilational properties of partly hydrolyzed polyacrylamide (HPAM) and 4,5-diheptyl-2-propylbenzene sulfonate (377) mixed systems in the absence or presence of electrolyte or oleic acid at the oil-water interface have been described by means of the oscillating barriers method and the interfacial tension relaxation method. The polymer plays different roles in influencing the nature of polymer-surfactant adsorbed layers at different surfactant concentrations. At low surfactant concentration, the addition of polymer perhaps weakens the “entanglement” of long alkyl chains, which decreases strikingly the dilational modulus of the adsorbed layer. At high surfactant concentration, the addition of the polymer increases the dilational modulus due to the hydrophobic interactions between polymer and surfactant molecules. On the case of adding electrolyte, the frequency dependence of dilational modulus increases due to the enhancement of exchange process of surfactant molecules and bivalent cation has more obvious effect than Na ion. Oleic acid plays dual roles in controlling interfacial dilational properties of mixed adsorption films: a small quantity of oleic acid increases the dilational modulus by forming densely packed mixed adsorption layer with surfactant molecules, while the superfluous addition of oleic acid could decrease the dilational modulus mainly due to the weakening of the “entanglement” among long alkyl chains in surfactant molecules.  相似文献

The kinetics of adsorption of lysozyme and alpha-lactalbumin from aqueous solution on silica and hydrophobized silica has been studied. The initial rate of adsorption of lysozyme at the hydrophilic surface is comparable with the limiting flux. For lysozyme at the hydrophobic surface and alpha-lactalbumin on both surfaces, the rate of adsorption is lower than the limiting flux, but the adsorption proceeds cooperatively, as manifested by an increase in the adsorption rate after the first protein molecules are adsorbed. At the hydrophilic surface, adsorption saturation (reflected in a steady-state value of the adsorbed amount) of both proteins strongly depends on the rate of adsorption, but for the hydrophobic surface no such dependency is observed. It points to structural relaxation ("spreading") of the adsorbed protein molecules, which occurs at the hydrophobic surface faster than at the hydrophilic one. For lysozyme, desorption has been studied as well. It is found that the desorbable fraction decreases after longer residence time of the protein at the interface.  相似文献

The air-BSA solution interface has been investigated by various techniques for years. From these studies we know that BSA molecules segregate at the BSA solution-air interface, and the surface coverage increases with the increase of the bulk solution concentration. However, questions still remain as to whether the protein changes conformation, orientation, or a combination of the two upon adsorption. In this paper, by using sum frequency generation (SFG) vibrational spectroscopy we found that the conformation of interfacial BSA molecules changes dramatically at the solution-air interface, compared to that of the native BSA in solution. The hydrophobic methyl groups of BSA molecules at this interface tend to align along the surface normal. The degree of such conformational changes of surface BSA molecules depend on the surface coverage, indicating that the protein-protein interaction plays a very important role in determining the conformation of interfacial protein molecules. At very low surface concentration, the adsorbed BSA molecules unfold substantially. Our results can provide a molecular interpretation of results obtained from other studies such as protein layer thickness and surface tension measurements of protein solution.  相似文献

We investigated the characteristics of heterogeneous layers composed of linear hydrolyzed polyvinylamine and branched polyethyleneimine adsorbed at silica/water interfaces. The studies also included heterogeneous layers where branched polyethyleneimine was replaced by polyethyleneimine modified by grafting with C12-C22 alkyl chains. Surface area exclusion chromatography was used to determine the interfacial relaxation and surface affinity of the polymer molecules within homogeneous layers. The relaxation of bare and grafted polyethyleneimine was found to be small and of equal extent but to develop at different rates. Comparatively, the relaxation of hydrolyzed polyvinylamine was faster and of greater extent. Within heterogeneous layers composed of polyvinylamine and bare or grafted polyethyleneimine, the relaxation of the different molecules was strongly increased as compared to that prevailing in homogeneous layers. The chromatographic method was then used to determine the mode of layer establishment. The polymer coating profiles on successive glass fiber filters were found to depend on the sequence of injection of the two polymers, due to the interfacial stability or instability of the initially established layer. It was shown that a previously established extremely thin layer of bare or grafted polyethyleneimine molecules strongly modified the adsorption profile of subsequently adsorbed polyvinylamine molecules.  相似文献

The dilatational rheological properties of cross-linked protein layers adsorbed at the oil-water interface were investigated with help of a modified drop tensiometer allowing successive replacements of the external phase. This setup enables one to perform cross-linking reactions at the interface only, that is, without any contact between the cross-linking agent and protein molecules in solution, under continuous monitoring of the interfacial tension. The mechanical properties of the resulting interface were investigated with dilatational large strain experiments. Measured rheological properties were related to the expected stability of an emulsion against disproportionation by considering the ratio of the interfacial elasticity to the interfacial tension. In an attempt to increase this ratio to improve the resistance against disproportionation, experiments were performed with densified protein layers obtained via reduction of the droplet area prior to cross linking. To highlight the influence of the protein morphology on the dilatational rheological properties of the cross-linked adsorbed layers, experiments were performed with random coil (beta-casein) as well as globular (beta-lactoglobulin) proteins. Glutaraldehyde was used as a cross-linking agent. Experiments were performed at 55 degrees C and pH 7.0 in 20 mM imidazole buffer for later comparison with enzymatically cross-linked adsorbed protein layers. The present work demonstrated substantial qualitative and quantitative differences in the interfacial rheological properties of cross-linked random coil and globular proteins.  相似文献
