A new automated spectrophotometric method for the determination of total sulfite in white and red wines is reported. The assay is based on the reaction of o-phthalaldehyde (OPA) and ammonium chloride with the analyte in basic medium under SI conditions. Upon on-line alkalization with NaOH, a blue product is formed having an absorption maximum at 630 nm. The parameters affecting the reaction - temperature, pH, ionic strength, amount concentration and volume of OPA, amount concentration of ammonium chloride, flow rate and reaction coil length - and the gas-diffusion process - sample and HCl volumes, length of mixing coil, donor flow rate - were studied. The proposed method was validated in terms of linearity (1-40 mg L−1, r = 0.9997), limit of detection (cL = 0.3 mg L−1) and quantitation (cQ = 1.0 mg L−1), precision (sr = 2.2% at 20 mg L−1 sulfite, n = 12) and selectivity. The applicability of the analytical procedure was evaluated by analyzing white and red wine samples, while the accuracy as expressed by recovery experiments ranged between 96% and 106%.  相似文献

An artificial neural network (ANN) procedure was used in the development of a catalytic spectrophotometric method for the determination of Cu(II) and Ni(II) employing a stopped-flow injection system. The method is based on the catalytic action of these ions on the reduction of resazurin by sulfide. ANNs trained by back-propagation of errors allowed us to model the systems in a concentration range of 0.5-6 and 1-15 mg l−1 for Cu(II) and Ni(II), respectively, with a low relative error of prediction (REP) for each cation: REPCu(II) = 0.85% and REPNi(II) = 0.79%. The standard deviations of the repeatability (sr) and of the within-laboratory reproducibility (sw) were measured using standard solutions of Cu(II) and Ni(II) equal to 2.75 and 3.5 mg l−1, respectively: sr[Cu(II)] = 0.039 mg l−1, sr[Ni(II)] = 0.044 mg l−1, sw[Ni(II)] = 0.045 mg l−1 and sw[Ni(II)] = 0.050 mg l−1. The ANNs-kinetic method has been applied to the determination of Cu(II) and Ni(II) in electroplating solutions and provided satisfactory results as compared with flame atomic absorption spectrophotometry method. The effect of resazurin, NaOH and Na2S concentrations and the reaction temperature on the analytical sensitivity is discussed.  相似文献

A spectrophotometric method is reported for the determination of bismuth in pharmaceutical products using sequential injection analysis. Methylthymol blue (MTB) was used as a color forming reagent and the absorbance of the Bi(III)-MTB complex was monitored at 548 nm. The various chemical and physical variables that affected the reaction were studied. A linear calibration graph was obtained in the range 0.0-75.0 mg l−1 Bi(III) at a sampling frequency of 72 h−1. The reagent consumption was considerably reduced compared to conventional flow injection systems, as only 150 μl of MTB were consumed per run. The precision was very satisfactory (sr=0.5%, at 50.0 mg l−1 Bi(III), n=12) and the limit of detection, cL, was 0.250 mg l−1. The developed method was applied successfully to the analysis of various pharmaceutical products containing Bi(III). The relative errors er, were <1.5% in all cases and were evaluated by comparison of the obtained results with those found using atomic absorption spectrometry as the reference method.  相似文献

A reversed-phase high performance liquid chromatographic method for the successful separation and determination of 13 synthetic food colorants (Tartrazine E 102, Quinoline Yellow E 104, Sunset Yellow E 110, Carmoisine E 122, Amaranth E 123, Ponceau 4R E 124, Erythrosine E 127, Red 2G E 128, Allura Red AC E 129, Patent Blue V E 131, Indigo Carmine E 132, Brilliant Blue FCF E 133 and Green S E 142) was developed. A C18 stationary phase was used and the mobile phase contained an acetonitrile-methanol (20:80 v/v) mixture and a 1% (m/v) ammonium acetate buffer solution at pH 7.5. Successful separation was obtained for all the compounds using an optimized gradient elution within 29 min. The diode-array detector was used to monitor the colorants between 350 and 800 nm. The method was thoroughly validated. Detection limits for all substances varied between 1.59 (E 142) and 22.1 (E 124) μg L−1. The intra-day precision (as R.S.D.r) ranged from 0.37% (E 122 in fruit flavored drink at a concentration of 100 mg L−1) to 4.8% (E 142 in icing sugar at a level of 0.9 mg kg−1). The inter-day precision (as R.S.D.R) was between 0.86% for E 122 in fruit flavored drink at 100 mg L−1 and 10% for E142 in jam at a concentration of 9 mg kg−1. Satisfactory recoveries, ranging from 94% (E 142 in jam) to 102% (E 131 in sweets), were obtained. The method was applied to the determination of colorants in various water-soluble foods, such as fruit flavoured drinks, alcoholic drinks, jams, sugar confectionery and sweets, with simple pre-treatment (dilution or water extraction).  相似文献

This study reports a sensitive solvent extraction flow-injection (FI) method for the simultaneous spectrophotometric determination of free cyanide and thiocyanate in human saliva and pralidoxime solutions. Cyanide and thiocyanate form colored (λmax=540 nm) ternary complexes with copper and 2,2′-dipyridyl-2-quinolylhydrazone (DPQH) that are extractable into chloroform. The determination of thiocyanates in the presence of cyanides is accomplished after on-line masking of the latter with formaldehyde through a binary inlet static mixer (BISM). Total thiocyanates and cyanides are determined in a second run, without the use of the masking agent. The proposed method allows the determination of the analytes in the range of 0-4 mg l−1 thiocyanates and 0-3 mg l−1 cyanides, with the 3σ detection limits being 0.007 and 0.004 mg l−1, respectively. The precision of the method (sr<1.0% at 1 mg l−1 CN or SCN, n=12 in both cases) and the sampling rates were quite satisfactory (60 injections per hour). The method was applied to the analysis of human saliva and pralidoxime solutions and gave recoveries in the range of 98.0-102.2% for both analytes whereas the mean relative error was er=1.7%.  相似文献

This paper describes selenium determination based on Se0 preconcentration in the imprinted polymer (synthesized with 2.25 mmol SeO2, 4-vinylpyridine and 1-vinylimidazole) with subsequent detection on-line in HG-FAAS. During the synthesis, SeO2 is reduced to Se (0). Therefore, there are no MIP neither IIP in the present work, thus we denominated: AIP, i.e., atomically imprinted polymers. For the optimization of analytical parameters Doehlert design was used. The method presented limit of detection and limit of quantification of 53 and 177 ng L−1, respectively, and linear range from 0.17 up to 6 μg L−1 (r = 0.9936). The preconcentration factor (PF), consumptive index (CI) and concentration efficiency (CE) were 232; 0.06 mL and 58 min−1 respectively. The proposed method was successfully applied to determine Se in Brazil nuts (0.33 ± 0.03 mg kg−1), apricot (0.46 ± 0.02 mg kg−1), white bean (0.47 ± 0.03 mg kg−1), rice flour (0.47 ± 0.02 mg kg−1) and milk powder (0.22 ± 0.01 mg kg−1) samples. It was possible to do 12 analyzes per hour. Accuracy was checked and confirmed by analyzing certified reference material (DORM-2, dogfish muscle), and samples precision was satisfactory with RSD lower than 10%.  相似文献

A simple, specific and sensitive sequential injection analysis (SIA) system based on non-immunoassay fluorescent detection has been developed for the determination of urinary albumin. The specific binding of the dye Albumin Blue 580 (AB 580) to albumin in urine generated high emission fluorescent signals. The excitation and emission wavelengths were set at 590 and 610 nm, respectively. The analytical range was obtained from 1 to 100 mg L−1, with a detection limit of 0.3 mg L−1 (S/N = 3). The SIA system gave high precision with relative standard deviations (R.S.D.s) of 0.9% and 1.4% when evaluated with 15 and 100 mg L−1 albumin (n = 15), respectively. The method exhibited good reproducibility, as assessed by performing four analytical curves on different days, and intra-run CVs (2.3-3.3%) and inter-run CVs (3.8%) were obtained. Rapid operation was achieved with a sample throughput of 37 h−1. This method was successfully applied to the determination of urinary albumin, and the method was highly correlated with the immunoturbidimetric method (r2 = 0.965; n = 72).  相似文献

The present work reports the first sequential injection (SI) method for the spectrophotometric determination of Ti(IV). The method is based upon the reaction of Ti(IV) with chromotropic acid (CA) in acidic medium to form a water-soluble complex (λmax = 420 nm). The chemical and instrumental variables of the system that affected the reaction were studied. Selectivity was greatly enhanced using ascorbic acid. A linear calibration graph was obtained in the range 0.2-10.0 mg l−1 Ti(IV) at a sampling frequency of 24 h−1. The precision was satisfactory (sr = 1.5% at 5.0 mg l−1 Ti(IV), n = 12) and the 3σ limit of detection, cL, was 0.7 mg l−1 (n = 10). The developed method proved to be adequately selective and was applied successfully to the analysis of real samples (dental implant and natural Moroccan phosphate rock) giving accurate results based on recovery studies (98-105%).  相似文献

A spot test was implemented in a flow-injection system for the spectrophotometric determination of zinc in digests of plant materials. It is based on the influence of Zn2+ on the oxidation rate of 1-naphthylethylenediamine (NED) by hexacyanoferrate(III) under acidic conditions. In order to control the precipitate formation and to maintain the resulting suspension, a micellar medium was established by adding Triton X-100. The proposed system handles about 65 samples per hour, meaning 72 μg NED and 9.0 mg K3[Fe(CN)6] per determination. Baseline drift is usually <0.01 absorbance per hour and the analytical signals for 0.5-2.5 mg l−1 Zn range within ca. 0.07-0.45 absorbance. Linearity of the analytical curve is fair (r>0.999, n=6) and detection limit was estimated as 0.2 mg l−1 Zn. Results are precise (R.S.D.<1%, n=10) and in agreement with flame atomic absorption spectrometry and with certified values of standard reference materials.  相似文献

Nitrate determination in waters is generally carried out with cadmium filings and carcinogenic reagents or by reaction with phenolic compounds in highly concentrated sulfuric acid medium. In this work, it was developed a green analytical procedure for nitrate determination in natural waters based on direct spectrophotometric measurements in ultraviolet, using a flow-injection system with an anion-exchange column for separation of nitrate from interfering species. The proposed method employs only one reagent (HClO4) in a minimum amount (equivalent to 18 μL concentrated acid per determination), and allowed nitrate determination within 0.50-25.0 mg L−1, without interference of up to 200.0 mg L−1 humic acid; 1.0 mg L−1 NO2; 200.0 mg L−1 PO43−; 75.0 mg L−1 Cl; 50.0 mg L−1 SO42− and 15.0 mg L−1 Fe3+. The detection limit (99.7% confidence level) and the coefficient of variation (n = 20) were estimated as 0.1 mg L−1 and 0.7%, respectively. The results obtained for natural water samples were in agreement with those achieved by the reference method based on nitrate reduction with copperized cadmium at the 95% confidence level.  相似文献

Inductively coupled plasma mass spectrometry (ICP-MS) is a suitable tool for multi-element analysis at low concentration levels. Rare earth element (REE) determinations in standard reference materials and small volumes of molten ice core samples from Antarctica have been performed with an ICP-time of flight-MS (ICP-TOF-MS) system. Recovery rates for REE in e.g. SPS-SW1 amounted to ∼103%, and the relative standard deviations were 3.4% for replicate analysis at REE concentrations in the lower ng L−1 range. Analyses of REE concentrations in Antarctic ice core samples showed that the ICP-TOF-MS technique meets the demands of restricted sample mass. The data obtained are in good agreement with ICP-Quadrupole-MS (ICP-Q-MS) and ICP-Sector Field-MS (ICP-SF-MS) results. The ICP-TOF-MS system determines accurately and precisely REE concentrations exceeding 5 ng L−1 while between 0.5 and 5 ng L−1 accuracy and precision are element dependent.  相似文献

Mycophenolic acid (MPA) is an immunosuppressant drug which powerfully inhibits lymphocyte proliferation. Since the early 1990s it has been used to prevent rejection in organ transplantation. The requirement of therapeutic drug monitoring shown in previous studies raises the necessity of acquiring accurate and sensitive methods to measure MPA and its major metabolite mycophenolic acid glucuronide (MPAG).The authors developed a sample cleanup-free, rapid, and highly specific method for simultaneous measurement of MPA and MPAG in human plasma and serum using the novel technology of ultra-performance liquid chromatography-electrospray ionization tandem mass spectrometry. MPA- and MPAG-determinations were performed during a 2.0-min run time. Multiple calibration curves for the analysis of MPA and MPAG exhibited consistent linearity and reproducibility in the range of 0.05-100 (r > 0.999) mg L−1 and 4-4000 mg L−1 (r > 0.999), respectively. Limits of Detection were 0.014 mg L−1 for MPA and 1.85 mg L−1 for MPAG. Lower Limits of Quantification were 0.05 mg L−1 for MPA and 2.30 mg L−1 for MPAG. Interassay imprecision was <10% for both substances. Mean recovery was 103.6% (range 78.1-129.7%) for MPA and 111.1% (range 73.0-139.6%) for MPAG. Agreement was good for MPA and MPAG between the presented method and a validated HPLC-MS/MS method. The Passing-Bablok regression line for MPA and MPAG was HPLC-MS/MS = 1.14 UPLC-MS/MS—0.14 [mg L−1], r = 0.96, and HPLC-MS/MS = 0.77 UPLC-MS/MS + 0.50 [mg L−1], r = 0.97, respectively. This sample cleanup-free and robust LC-MS/MS assay facilitates the rapid, accurate and simultaneous determination of MPA and MPAG in human body fluids.  相似文献

A highly selective and simple flow injection method is reported for the determination of Au(III) in jewel samples. The method is based on the catalytic effect of Au(III) on the oxidation of 4-amino-4′-methoxydiphenylamine hydrochloride (Variamine Blue B base, VB) by KIO3. The colored reaction product was monitored spectrophotometrically at 546 nm. A volume fraction of 40% N,N-dimethylformamide (DMF) greatly enhances the selectivity of the method. The chemical (pH and concentrations of reagents) and instrumental variables (sample injection volume, reagents flow rates, reaction coil length) affecting the determination were studied and optimized. Under the selected values, the analyte could be determined in the range of 0.1-12.0 mg L−1 (r = 0.9997), at a sampling rate of 120 h−1. The proposed assay was precise (sr = 0.8% at 5.0 mg L−1 Au(III), n = 12) and adequately sensitive with a 3σ limit of detection of 0.03 mg L−1. The method was successfully applied to the analysis of jewel samples. The obtained results were favorably compared to flame atomic absorption spectrometry (FAAS) used as a reference method.  相似文献

The present work describes the development of a fast and robust sequential injection fluorimetric procedure for the determination of Sn in juices of canned fruits. The developed automatic methodology is based on the complexation of Sn with 8-hydroxyquinoline-5-sulfonic acid (HQSA) to form a fluorimetric product (λexc = 354 nm; λem = 510 nm). The influence of dimethylsulfoxide (DMSO) and cetylpyridinium bromide (CPB) on the sensitivity of the fluorimetric determination was evaluated.Linear calibration plots were obtained for Sn concentrations between 1 and 10 mg L−1, with a detection limit of 0.38 mg L−1. In each analytical cycle 0.006 mg of HQSA and 0.47 mg of CPB were consumed and 1.5 mL of effluent was generated.The developed methodology was applied to the determination of Sn in juices of canned fruits and the results complied with those furnished by an electrothermal atomic absorption spectrometry comparison procedure, with relative deviations lower than 5.2%.The automatic procedure exhibited good precision (R.S.D. < 1.4%) and the sampling rate was about 70 determinations per hour.  相似文献

A sensitive fluorimetric method for determination of phytic acid in human urine samples was described. The method was based on a fluorimetric replacement reaction, in which the added phytic acid replaced the Cu2+ ion from Cu2+-gelatin complex, liberating the fluorescent gelatin molecule. The fluorescence of the solution was accordingly recovered proportionally to the amount of the foreign phytic acid. The excitation wavelength was 273.5 nm and the characteristic emission wavelength was 305.0 nm, respectively. The calibration graph was obtained by plotting the recovered fluorescent intensity at maximum 305.0 nm against the added standard phytic acid, and was divided into two sections. One section was linear over the range of 0.40-2.40 mg L−1 with a linear regression equation of If = −0.895 + 15.146c (R2 > 0.9993), and the other over the range of 2.40-9.20 mg L−1 with a linear regression equation of If = −29.526 + 26.113c (R2 > 0.9996), respectively. The relative standard deviation (R.S.D.) at 95% confidence degree for a 2.0 mg L−1 of standard phytic acid within 1 month was less than 1.26% (n = 5), indicating the procedure is reproducible. The detection and the quantification limits of phytic acid were estimated to be 0.23 and 0.40 mg L−1, respectively. The proposed method was applied to the determination of phytic acid in urine samples and the found concentrations of phytic acid in urine were in the range of 0.49-0.75 mg L−1 with recoveries of 96.2-108.8%. Comparison of the obtained results with the reported HPLC was performed, indicating the proposed method was reliable.  相似文献

A novel simple, fast and efficient ultra-high performance supercritical fluid chromatography (UHPSFC) method was developed and validated for the separation and quantitative determination of eleven illegal dyes in chili-containing spices. The method involved a simple ultrasound-assisted liquid extraction of illegal compounds with tetrahydrofuran. The separation was performed using a supercritical fluid chromatography system and CSH Fluoro-Phenyl stationary phase at 70 °C. The mobile phase was carbon dioxide and the mixture of methanol:acetonitrile (1:1, v/v) with 2.5% formic acid as an additive at the flow rate 2.0 mL min−1. The UV–vis detection was accomplished at 500 nm for seven compounds and at 420 nm for Sudan Orange G, Butter Yellow, Fast Garnet GBC and Methyl Red due to their maximum of absorbance. All eleven compounds were separated in less than 5 min. The method was successfully validated and applied using three commercial samples of chili-containing spices – Chili sauce (Indonesia), Feferony sauce (Slovakia) and Mojo sauce (Spain). The linearity range of proposed method was 0.50–9.09 mg kg−1 (r ≥ 0.995). The detection limits were determined as signal to noise ratio of 3 and were ranged from 0.15 mg kg−1 to 0.60 mg kg−1 (1.80 mg kg−1 for Fast Garnet) for standard solution and from 0.25 mg kg−1 to 1.00 mg kg−1 (2.50 mg kg−1 for Fast Garnet, 1.50 mg kg−1 for Sudan Red 7B) for chili-containing samples. The recovery values were in the range of 73.5–107.2% and relative standard deviation ranging from 0.1% to 8.2% for within-day precision and from 0.5% to 8.8% for between-day precision. The method showed potential for being used to monitor forbidden dyes in food constituents. The developed UHPSFC method was compared to the UHPLC-UV method. The orthogonality of Sudan dyes separation by these two methods was demonstrated. Benefits and drawbacks were discussed showing the reliability of both methods for monitoring of studied illegal dyes in real food constituents.  相似文献

A novel on-line microdialysis sampling coupled with flame atomic absorption spectrometry (FAAS) with an attractive application is reported. Microdialysates perfused through implanted microdialysis probes were directly introduced into the flame atomizer of a FAAS system using 0.2% HNO3 as carrier solution at a nebulizer uptake flow rate of 6 ml min−1. The interval for each determination was 90 s (60 s sampling time, 10 s read time and 20 s washing time). The analytical characteristics of the on-line microdialysis-FAAS system were validated as follows: linearity range, 0-300 mg l−1; detection limit (3σ, n = 7), 0.53 mg l−1; precision (R.S.D., n = 50), 4.1%. By comparing Mg levels in the blood of living rabbits with the results obtained from in vivo no net flux (NNF) method, the accuracy of the proposed on-line method was found to be good. The present method can be successfully applied to the in vivo monitoring of diffusible Mg in the blood of living rabbits after magnesium sulfate (MgSO4) administration with a temporal resolution of 1.5 min.  相似文献

Ethyl Lauroyl Arginate (LAE) is a cationic tensoactive compound, soluble in water, with a wide activity spectrum against moulds and bacteria. LAE has been incorporated as antimicrobial agent into packaging materials for food contact and these materials require to comply with the specific migration criteria. In this paper, one analytical procedure has been developed and optimized for the analysis of LAE in food simulants after the migrations tests. It consists of the formation of an ionic pair between LAE and the inorganic complex Co(SCN)42− in aqueous solution, followed by a liquid–liquid extraction in a suitable organic solvent and further UV–Vis absorbance measurement. In order to evaluate possible interferences, the ionic pair has been also analyzed by high performance liquid chromatography with UV–Vis detection. Both procedures provided similar analytical characteristics, with linear ranges from 1.10 to 25.00 mg kg−1, linearity higher than 0.9886, limits of detection and quantification of 0.33 and 1.10 mg kg−1, respectively, accuracy better than 1% as relative error and precision better than 3.6% expressed as RSD. Optimization of analytical techniques, thermal and chemical stability of LAE, as well as migration kinetics of LAE from experimental active packaging are reported and discussed.  相似文献

This work describes development of a flow injection (FI) system for determination of iodide, based on the chemiluminescence (CL) reaction between iodine and luminol. Iodide in the sample zone is oxidized to iodine. Employment of a gas-diffusion (GD) unit allows for selective detection of the generated CL (425 nm). Preliminary results showed for concentrations of less than 2 mg L−1, that signals were irreproducible and that the calibration was not linear.In order to solve these problems, a method of ‘membrane conditioning’ was investigated, in which iodide stream was continuously merged with oxidant to generate I2 that conditioned the GD membrane and tubing. This minimized surface interaction between the active surface and the I2 generated from the samples, thus improving both precision and sensitivity. By employing membrane conditioning, it has been possible to reliably detect concentrations down to 0.1 mg L−1.At the optimized condition, an excellent linear calibration (r2 = 0.999) was obtained from 0.1 to 1.0 mg L−1. The method was successfully applied to determine iodide in some pharmaceutical products such as potassium iodide tablets and a liquid patent medicine. However, for vitamin tablets, ascorbic acid was found to interfere seriously by causing a negative signal.  相似文献

In this work, for the first time, capillary zone electrophoresis (CZE) technique combined with microwave-assisted extraction (MAE) was developed for the fast quantification of chlorogenic acid (CA) in tobacco residues. CA in tobacco residue samples were extracted by MAE technique, and then analyzed by CZE. As a new sample preparation method for tobacco residues, the MAE procedure is optimized, validated and compared with conventional methods including ultrasonic extraction (USE) and reflux extraction (RE). It is found that MAE gives the best result due to the highest extraction efficiency within shortest extraction time (only 4.0 min). Here, CA is determined by CZE based on the calibration curve of its authentic standard. The method linearity, detection limit, precision and recovery are studied. The results show that the combined MAE and CZE method has a linearity (R2 0.991, 0.003-0.5 mg ml−1), a limit of detection (0.003 mg ml−1), a limit of quantification (0.01 mg ml−1), good precision (R.S.D. = 4.28%) and a finer recovery (89.0%). The proposed method was successfully applied to the analysis of CA in tobacco residue samples. The experiment results have demonstrated that the CZE combined with MAE is a convenient, fast, economical and reliable method for the determination of CA in tobacco residues.  相似文献
