A novel method for the simultaneous determination of phenolic compounds such as salicylic acid, resorcinol, phloroglucinol, p-hydroxybenzoic acid, 2,4-dihydroxybenzoic acid, and m-nitrophenol by high-performance liquid chromatography (HPLC) coupled with chemiluminescence (CL) detection was developed. The procedure was based on the chemiluminescent enhancement by phenolic compounds of the cerium(IV)-Tween 20 system in a sulfuric acid medium. The separation was carried out with an isocratic elution or with a gradient elution using a mixture of methanol and 1.5% acetic acid. For six phenolic compounds, the detection limits (3σ) were in the range 1.40-5.02 ng/ml and the relative standard deviations (n=11) for the determination of 0.1 μg/ml compounds were in the range 1.9-2.9%. The CL reaction was well compatible with the mobile phase of HPLC, no baseline drift often occurred in HPLC-CL detection was observed with a gradient elution. The method has been successfully applied to the determination of salicylic acid and resorcinol in Dermatitis Clear Tincture and p-hydroxybenzoic acid in apple juices.  相似文献

A 3-step analytical procedure was developed and optimized for the simultaneous determination of 6 phenols (phenol, o-, m-, p-cresol, catechol and resorcinol) and 19 chlorophenols (all mono-, di-, tri-, and tetrachlorophenol isomers and pentachlorophenol) from environmental water samples. The analytical scheme consists of (1) solid-phase extraction (SPE) carried out on hypercrosslinked styrene-divinylbenzene (Isolute ENV+) cartridge; (2) derivatization with trimethylsilyl-N,N-dimethylcarbamate (TMSDMC); (3) analysis of the derivatives with capillary gas chromatography-mass spectrometry, in the selective ion monitoring mode. Ethyl acetate, ethyl acetate/acetic acid (5 v/v%) mixture, dichloromethane and acetonitrile were compared as to their ability to elute the phenols and chlorophenols from the ENV + sorbent in the smallest solvent volume possible. The optimized extraction step uses a minimal amount of organic solvent (4 mL ethyl acetate). Derivatization of the phenols and chlorophenols with TMSDMC was studied with respect to conversion, reagent excess, medium, temperature and the stability of the trimethylsilyl derivatives. If the reagent is applied in sufficient excess, the reaction takes place instantaneously at room temperature, and the derivatives remain stable for 24 h, making the procedure simple, fast and convenient. The overall method gave detection limits of 0.05-100 ng/L for all compounds except resorcinol which could not be retained on the SPE cartridge. The complete optimized analytical scheme was applied to ground water and river water samples collected in Hungary.  相似文献

This work describes an analytical procedure for vanadium determination in human hair slurries by electrothermal AAS using longitudinal heating (LHGA) and transversal heating (THGA) graphite furnace atomizers. The samples were powdered using cryogenic grinding and the hair slurries containing 0.2% (m/v) were prepared in three different media for determination of vanadium: 0.14 mol L−1 HNO3, 0.1% (v/v) Triton X-100 and 0.1% (v/v) water soluble tertiary amines (CFA-C, pH 8). The limits of detection (LOD), limits of quantification (LOQ), and characteristic masses obtained were 0.28, 0.95 μg L−1 and 35 pg (LHGA) and 0.34, 1.13 μg L−1 and 78 pg (THGA), respectively. The accuracy of the analytical results obtained by the proposed procedure in both equipments was confirmed by a paired t-test at the 95% confidence level and compared with a conventional procedure based on acid digestion.  相似文献

The effect of 36 aromatic compounds on the luminol-dimethylsulfoxide-OH chemiluminescence (CL) was systematically studied. It was found that dihydroxybenzenes, and ortho- and para-substituted aminophenols and phenylenediamines inhibited the CL and phenols with three or more than three hydroxyls except phloroglucin tended to enhance the CL. The CL inhibition and enhancement was proposed to be dependent on whether superoxide anion radical (O2) was competitively consumed by compounds in the CL system. Trihydroxybenzenes were capable of generating superoxide anion radical, leading to the CL enhancement, whereas dihydroxybenzenes were superoxide anion radical scavenger, causing the CL inhibition. Based on the inhibited CL, a novel method for the simultaneous determination of p-phenylenediamine, o-phenylenediamine, p-aminophenol, o-aminophenol, resorcinol and hydroquinone by high-performance liquid chromatography coupled with chemiluminescence detection was developed. The method has been successfully applied to determine intermediates in oxidative hair dyes and wastewater of shampooing after hair dyed.  相似文献

We report here, for the first time, application of batch injection analysis (BIA) with amperometric detection for determination of the phenolic antioxidant butylated hydroxyanisole (BHA) in biodiesel. A sample plug was directly injected onto a boron-doped diamond electrode immersed in 50% v/v hydroethanolic solution with 0.1 mol L−1 HClO4 using an electronic micropipette. Importantly, the only preparation step required for biodiesel analysis is dilution in the same hydroethanolic electrolyte solution. Our proposed method has several advantages for routine biodiesel analysis, including: a low relative standard deviation between injections (0.29%, n = 20), high analytical frequency (120 h−1), adequate recovery values (93-101%) for spiked samples, satisfactory accuracy (based on comparative determinations by high-performance liquid-chromatography), and a low detection limit (100 ng of BHA per g of biodiesel). Finally, our method can be adapted for the determination of other antioxidants in biodiesel samples.  相似文献

By use of the high separation capability of capillary electrophoresis (CE) and ultrasensitive chemiluminescence (CL) detection, a CE-CL method was proposed for the ultrasensitive determination of trace cobalt in a single hair, which is potentially useful in construction of the fingerprint of trace elements in the hair collected from crime scenes. In this work, the CE experimental conditions, CL experimental conditions and the digestion methods for the analysis of a single hair sample for cobalt were investigated in detail. The relative limit of detection (LOD, 3σ) was 0.01 ng/mL, and the absolute LOD was 2.4 × 10− 16 g considering the sampling volume of 24 nL. Using a dry digestion method, the analytical results for certified reference hair samples by the proposed method were in good agreement with the certified values. Finally, this method was successfully used to detect trace cobalt in a single hair from three adults. It has potential applications in forensic analysis.  相似文献

Spectrophotometric and high performance liquid chromatographic (HPLC) methods have been developed for the determination of cis-platin and carboplatin based on the pre-column derivatization of platinum(II) with 2-acetylpyridine-4-phenyl-3-thiosemicarbazone. The complex was extracted in chloroform with molar absorptivity of 2.2 × 104 L mol−1 cm−1 at 380 nm. The complex eluted from a Phenomenex C-18 (150 mm × 4.6 mm i.d.) column with methanol:water:acetonitrile:tetrabutyl ammonium bromide (1 mM) (44:30:25:1, v/v/v/v) with a flow rate of 1 ml/min and UV detection at 260 nm. Ruthenium(IV) and selenium(IV) also separated completely. The linear calibration curve was with 0.5-12.5 μg/ml and detection limit of 10 ng/ml platinum(II).The analysis of cis-platin and carboplatin injections by spectrophotometric and HPLC methods indicated relative standard deviation (R.S.D.) of 0.66-2.1%. The method was used for the determinations of cis-platin in serum and urine of cancer patients after chemotherapy and platinum contents were found 148-444 and 50-90 ng/ml with R.S.D. of 0.3-3.0 and 0.6-2.4% for the serum and urine, respectively. The recovery of platinum(II) from serum was 97% with R.S.D. 2.2%.  相似文献

Two polar benzo[c]phenanthridine alkaloids, chelerythrine (CHE) and dihydrochelerythrine (DHCHE), were extracted at 35 °C and 10 MPa (15 MPa for real samples) from real and spiked plasma samples with acceptable recoveries (95.1% and 81.0%, respectively) using near-critical CO2 modified with aqueous (1:1, v/v) methanol. The alkaloids were quantified by a liquid chromatographic/electrospray mass spectrometric (LC/ESI-MS) method on a Zorbax SB-CN column (75 mm × 4.6 mm, 3.5 μm particle size) using methanol (organic phase) and 50 mM ammonium formiate (aqueous phase) as a mobile phase. A linear gradient 0-1 min, isocratic at 60% organic phase (v/v); from 1.0 to 7.0 min, 60-71% organic phase (v/v); from 7.0 to 18.0 min, 71-60% organic phase (v/v) was applied. The limit of detection was 1.22 ng (3.50 pmol) for CHE and 0.95 ng (2.72 pmol) for DHCHE per 1 ml of the sample. The linearity of the calibration curves was satisfactory as indicated by coefficients of determination 0.9979 and 0.9995 for CHE and DHCHE, respectively. Repeatability and intermediate precision (average R.S.D.s) were 1.0-1.5%, accuracy was in the range 99.7-100.3%. Average recovery was 100.1% for both, standard solutions and spiked plasma extracts. Three samples of real rat plasma were extracted and analysed to test the method.  相似文献

The separation and determination of aminophenols and phenylenediamines were investigated by liquid chromatography (LC) and micellar electrokinetic chromatography (MEKC) in this study. Aminophenols and phenylenediamines are commonly used components in commercial hair colorants. The problem of tailing peaks in LC was improved by the technique of using mobile phase containing 15 mM triethylamine at pH 8.0. The analysis of o-aminophenol was not succeeded with LC even though the modifier of triethylamine was added. But it could be quantitative successfully by MEKC. The optimum separation condition of MEKC was achieved by employing 55 mM cetyltrimethyl ammonium chloride in 50 mM borate buffer (pH 9.2) with electric field strength of −145 V cm−1. Finally, the commercial hair dyes were analyzed by developing methods of LC and MEKC. From both the results, there is no significant difference presence at 99.5% confidence level. These two methods could give the complementary results.  相似文献

Aminopyrene was convalently anchored onto the surface of mesoporous MCM-41 silica by post-grafting. This organic-inorganic hybrid has been applied as sensing material to phenols determination. Experimental results reveal that the functionalized material presents good sensitivity and selectivity towards resorcinol and can be used for resorcinol determination in water at pH 6.0. The fluorescence intensity of aminopyrene functionalized mesoporous silica decreases proportionally to the logarithm of resorcinol concentration in water. The linear range for resorcinol detection lies in 4.79-163 μM with a detection limit of 2.86 μM (S/N = 3).  相似文献

A rapid and practical solid sampling electrothermal atomic absorption spectrometric method was described for the determination of lead in scalp hair. Hair samples were washed once with acetone; thrice with distilled-deionized water and again once with acetone and dried at 75 °C. Typically 0.05 to 1.0 mg of dried samples were inserted on the platforms of solid sampling autosampler. The effects of pyrolysis temperature, atomization temperature, the amount of sample as well as addition of a modifier (Pd/Mg) and/or auxiliary digesting agents (hydrogen peroxide and nitric acid) and/or a surfactant (Triton X-100) on the recovery of lead were investigated. Hair samples were washed once with acetone; thrice with distilled-deionized water and again once with acetone and dried at 75 °C. Typically 0.05 to 1.0 mg of dried samples were inserted on the platforms of solid sampling autosampler. The limit of detection for lead (3σ, N = 10) was 0.3 ng/g The addition of modifier, acids, oxidant and surfactant hardly improved the results. Due to the risk of contamination and relatively high blank values, the lead in hair were determined directly without adding any reagent(s). Finally, the method was applied for the segmental determination of lead concentrations in hair of different persons which is important to know when and how much a person was exposed to the analyte. For this purpose, 0.5 cm of pieces were cut along the one or a few close strands and analyzed by solid sampling.  相似文献

A sensitive and specific liquid chromatography-electrospray ionization-mass spectrometry (LC-ESI-MS) method for the separation and analysis of d- and l-thyroxine was developed using R(−)/S(+)-4-(3-isothiocyanatopyrrolidin-1-yl)-7-(N,N-dimethylaminosulfonyl-2,1,3-benzoxadiazole, [R(−)/S(+)-DBD-PyNCS] as a chiral derivatization reagents. The T4 derivatives with R(−)-DBD-PyNCS were efficiently separated on a reversed-phase column with water-acetonitrile containing 0.1% formic acid (41:59, v/v) as the eluent and analyzed using ESI-MS with negative selected ion monitoring (SIM) mode. The calibration curves of both the d-T4 and l-T4 were linear over the concentration range of 0.13-13 μg/ml. The detection limits (S/N = 3) were 28 ng/ml for d-T4 and 40 ng/ml for l-T4, respectively. The relative standard deviations (RSD, n = 5) were less than 3.6% at 1.3 μg/ml for both T4 enantiomers. The proposed method was applied to the determination of l-T4 enantiomer in a pharmaceutical formulation.  相似文献

In this study, a CE-MS method using a monolithic sol-gel concentrator for in-line solid-phase extraction (SPE) is evaluated for the analysis of methionine enkephalin in biological samples. Operational SPE parameters such as sample pH, loading volume, elution volume and composition have been studied. After optimization of the in-line preconcentration methodology, a 40-fold preconcentration was demonstrated for a methionine enkephalin test solution using a loading volume of 3200 nL. The method was linear in the range from 62.5 to 1000 ng/mL (R2 > 0.99). R.S.D. values for migration times and peak areas were 1.2% and 8.4%, respectively. Finally, the analysis of cerebrospinal fluid samples spiked with methionine enkephalin and deproteinized with perchloric acid (1:1, v/v) showed a detection limit (S/N = 3) of approximately 1 ng/mL (ca. 5 nM). The recoveries of methionine enkephalin for three concentration levels (100, 10 and 1 ng/mL) were in the range of 74-91%, demonstrating the promising potential of the methodology for the analysis of biological samples.  相似文献

A simple and sensitive high performance liquid chromatography method with photodiode array detection (HPLC-DAD) was developed for simultaneous determination of eight bioactive constituents (schisandrin, schisandrol B, schisantherin A, schisanhenol, anwulignan, deoxyshisandrin, schisandrin B and schisandrin C) in the ripe fruit of Schisandra sphenanthera and its traditional Chinese herbal preparations Wuzhi-capsule by optimizing the extraction, separation and analytical conditions of HPLC-DAD. The chemical fingerprint of S. sphenanthera was established using raw materials of 15 different origins in China. The chromatographic separations were obtained by an Agilent Eclipse XDB-C18 reserved-phase column (250 mm × 4.6 mm i.d., 5 μm) using gradient elution with water-formic acid (100:0.1, v/v) and acetonitrile, at a flow rate of 1.0 mL min−1, an operating temperature of 35 °C, and a wavelength of 230 nm. The constituents were confirmed by (+) electrospray ionization LC-MS. The new method was validated and was successfully applied to simultaneous determination of components in 13 batches of Wuzhi-capsule. The results indicate that this multi-component determination method in combination with chromatographic fingerprint analysis is suitable for quantitative analysis and quality control of S. sphenanthera.  相似文献

The development of a rapid method for the determination of perchlorate in rain and drinking waters is presented. In the optimised method, an on-line preconcentration technique was employed utilising a 10 mm × 4.6 mm Phenomenex Onyx monolithic guard cartridge coated with (N-dodecyl-N,N-dimethylammonio)undecanoate for selective preconcentration, with subsequent elution into a fixed volume injection loop (‘heart-cut’ of the concentrator column eluate) and separation using an IonPac AS16 (250 mm × 2 mm) anion exchange column and a potassium hydroxide concentration gradient. Off-line optimisation studies showed that the coated monolith displayed near quantitative recovery up to 50 μg/L perchlorate level from standards prepared in reagent water. On-line preconcentration of perchlorate obtained detection limits down to 56 ng/L in reagent water, between 70 and 80 ng/L in rainwater samples and 2.5 μg/L in non-pretreated drinking water. After an additional sample sulphate/carbonate removal step, low ng/L perchlorate concentrations could also be observed in drinking water. The complete on-line method exhibited reproducibility for n = 10 replicate runs of R.S.D. ≤ 3% for peak height/area and R.S.D. = 0.08% for retention time. The optimised method, of 20 min total duration, was applied to the determination of perchlorate by standard addition in 10 rainwater samples and one drinking water sample. Concentrations of perchlorate present ranged from below the detection limit for four rainwater samples, with another three samples showing perchlorate present at between 70 and 100 ng/L, and one sample showing perchlorate present at 2.8 μg/L. Levels of 1.1 μg/L in the drinking water sample were also recorded.  相似文献

A simple and rapid liquid chromatographic method for enantioselective separation and determination of R-(+) and S-(−) enantiomers of rabeprazole in drugs and pharmaceuticals using photo diode array (PDA) and polarimetric detectors connected in series was developed. Chiralpak AD-H (250 mm × 4.6 mm) 5 μm column packed with amylose tris(3,5-dimethylphenyl carbamate) as a stationary phase and the mobile phase containing n-hexane:ethanol:2-propanol(75:15:10, v/v/v) in an isocratic mode has yielded baseline separation with resolution greater than 3.0 at 40 °C. Effects of ethanol, 2-propanol and temperature on separation were studied for optimum resolution. Lansoprazole sulphone was used as an internal standard (IS) for quantitative determination of individual enantiomers in bulk drugs as well as pharmaceutical formulations. The method was validated in terms of accuracy, precision and linearity according to ICH guidelines. The linearity of the method was studied in the range of 0.5-50 μg/ml and the r2 was >0.9997. The inter- and intra-day precision of assay were determined (R.S.D. < 1%) and the recoveries were in the range of 99.63-100.22% with <1% R.S.D. The limits of detection (LOD) and quantification (LOQ) were 0.02 μg/ml and 0.07 μg/ml for both the enantiomers, respectively.  相似文献

A simple and sensitive automated method for determination of aflatoxins (B1, B2, G1, and G2) in nuts, cereals, dried fruits, and spices was developed consisting of in-tube solid-phase microextraction (SPME) coupled with liquid chromatography–mass spectrometry (LC–MS). Aflatoxins were separated within 8 min by high-performance liquid chromatography using a Zorbax Eclipse XDB-C8 column with methanol/acetonitrile (60/40, v/v): 5 mM ammonium formate (45:55) as the mobile phase. Electrospray ionization conditions in the positive ion mode were optimized for MS detection of aflatoxins. The pseudo-molecular ions [M+H]+ were used to detect aflatoxins in selected ion monitoring (SIM) mode. The optimum in-tube SPME conditions were 25 draw/eject cycles of 40 μL of sample using a Supel-Q PLOT capillary column as an extraction device. The extracted aflatoxins were readily desorbed from the capillary by passage of the mobile phase, and no carryover was observed. Using the in-tube SPME LC–MS with SIM method, good linearity of the calibration curve (r > 0.9994) was obtained in the concentration range of 0.05–2.0 ng/mL using aflatoxin M1 as an internal standard, and the detection limits (S/N = 3) of aflatoxins were 2.1–2.8 pg/mL. The in-tube SPME method showed >23-fold higher sensitivity than the direct injection method (10 μL injection volume). The within-day and between-day precision (relative standard deviations) at the concentration of 1 ng/mL aflatoxin mixture were below 3.3% and 7.7% (n = 5), respectively. This method was applied successfully to analysis of food samples without interference peaks. The recoveries of aflatoxins spiked into nuts and cereals were >80%, and the relative standard deviations were <11.2%. Aflatoxins were detected at <10 ng/g in several commercial food samples.  相似文献

A simple and sensitive method for the determination of patulin in fruit juice and dried fruit samples was developed using a fully automated method consisting of in-tube solid-phase microextraction (SPME) coupled with liquid chromatography–mass spectrometry (LC–MS). Patulin was separated within 5 min by high-performance liquid chromatography using a Synergi MAX-RP 80A column and water/acetonitrile (80/20, v/v) as the mobile phase. Electrospray ionization conditions in the negative ion mode were optimized for MS detection of patulin. The pseudo-molecular ion [M−H] was used to detect patulin in selected ion monitoring (SIM) mode. The optimum in-tube SPME conditions were 25 draw/eject cycles of 40 μL of sample using a Carboxen 1006 PLOT capillary column as an extraction device. The extracted patulin was readily desorbed from the capillary by passage of the mobile phase, and no carry-over was observed. Using the in-tube SPME LC–MS with SIM method, good linearity of the calibration curve (r = 0.9996) was obtained in the concentration range of 0.5–20 ng/mL using 13C3-patulin as an internal standard, and the detection limit (S/N = 3) of patulin was 23.5 pg/mL. The in-tube SPME method showed >83-fold higher sensitivity than the direct injection method (10 μL injection volume). The within-day and between-day precision (relative standard deviations) were below 0.8% and 5.0% (n = 6), respectively. This method was applied successfully for the analysis of fruit juice and dried fruit samples without interference peaks. The recoveries of patulin spiked into apple juice were >92%, and the relative standard deviations were <4.5%. Patulin was detected at ng/mL levels in various commercial apple juice samples.  相似文献

In the study, we developed a simple, rapid and sensitive method for the determination of tiopronin (TP) in human plasma, which was based on derivatization with p-bromophenacyl bromide (p-BPB) followed by liquid-liquid extraction and reverse-phase HPLC-UV detection. For the first time, the p-BPB was introduced into the derivatization of TP. The thiol group of TP was trapped with p-BPB to form a TP-p-BPB adduct, which can be very suitable for UV detection. From acidified plasma samples, the derivatized TP was extracted with 5 mL dichloromethane. Effective chromatographic separation was achieved using a C18 column (DIAMONSIL 150 mm × 4 mm i.d., 5 μm) based on an acetonitrile-water-trifluoroacetic acid (40:59.88:0.12, v/v/v) elution at a flow-rate of 1 mL/min. The IS and the derivatized TP were detected at 263 nm. No endogenous substances were found to interfere. The limit of quantification for derivatized TP (TP-p-BPB) in plasma was 40 ng/mL. The calibration curve for the derivatized TP showed linearity in the range 0.04-4 μg/mL with a regression coefficient corresponding to 0.9991 and the coefficient of the variation of the points of the calibration curve being lower than 10%. Extraction recoveries of the derivatized TP in plasma were greater than 72%. The method was suitably validated and successfully applied to determination of TP in human plasma samples.  相似文献

A new method for the measurement of N-nitrosamines in part-per-trillion concentrations from water samples without preconcentration steps has been developed. This method is based on online UV irradiation after high-performance liquid chromatographic separation and subsequent luminol chemiluminescence detection without addition of an oxidant. It was confirmed that N-nitrosamines in basic aqueous solution were transformed to peroxynitrite by UV irradiation. The detection limits for this method were 1.5 ng/L, 2.9 ng/L, 3.0 ng/L, and 2.7 ng/L for N-nitrosodimethylamine, N-nitrosomorpholine, N-nitrosomethylethylamine, and N-nitrosopyrrolidine, respectively, at a signal-to-noise ratio of 3. The calibration graphs were linear in the range of 5–1000 ng/L for these N-nitrosamines. This method was used for the determination of N-nitrosamines in tap water, river water, and industrial plant effluent samples. The recoveries of N-nitrosodimethylamine, N-nitrosomorpholine, N-nitrosomethylethylamine, and N-nitrosopyrrolidine present in tap water sample at a concentration of 10 ng/L (mean ± standard deviation, n = 4) were (94.8 ± 2.7)%, (102.0 ± 6.9)%, (99.3 ± 3.9)%, and (102.8 ± 2.5)%, respectively. These results indicate that our proposed method can be applied satisfactorily to the determination of N-nitrosamines in water samples.  相似文献
