A new biochemical oxygen demand (BOD) sensing method employing a double-mediator (DM) system coupled with ferricyanide and a lipophilic mediator, menadione and the eukaryote Saccharomyces cerevisiae has been developed. In this study, a stirred micro-batch-type microbial sensor with a 560 μL volume and a two-electrode system was used. The chronamperometric response of this sensor had a linear response between 1 μM and 10 mM hexacyanoferrate(II) (r2 = 0.9995, 14 points, n = 3, average of relative standard deviation and R.S.D.av = 1.3%). Next, the optimum conditions for BOD estimation by the DM system (BODDM) were investigated and the findings revealed that the concentration of ethanol, used to dissolve menadione, influenced the sensor response and a relationship between the sensor output and glucose glutamic acid concentration was obtained over a range of 6.6-220 mg O2 L−1 (five points, n = 3, R.S.D.av 6.6%) when using a reaction mixture incubated for 15 min. Subsequently, the characterization of this sensor was studied. The sensor responses to 14 pure organic substances were compared with the conventional BOD5 method and other biosensor methods. Similar results with the BOD biosensor system using Trichosporon cutaneum were obtained. In addition, the influence of chloride ion, artificial seawater and heavy metal ions on the sensor response was investigated. A slight influence of 20.0 g L−1 chloride ion and artificial seawater (18.4 g L−1 Cl) was observed. Thus, the possibility of BOD determination for seawater was suggested in this study. In addition, no influence of the heavy metal ions (1.0 mg L−1 Fe3+, Cu2+, Mn2+, Cr3+ and Zn2+) was observed. Real sample measurements using both river water and seawater were performed and compared with those obtained from the BOD5 method. Finally, stable responses were obtained for 14 days when the yeast suspension was stored at 4 °C (response reduction, 93%; R.S.D. for 6 testing days, 9.1%).  相似文献

The performance of a micropump operating on evaporation and capillary effects, developed for microfluidic (lab-on-a-chip) systems, was studied employing it as the fluid drive in a microfluidic flow injection (FI) system, with chemiluminescence (CL) detection. The micropump featured simple structure, small dimensions, low fabrication cost and stable and adjustable flow-rates during long working periods. Using a micropump with 6.6 cm2 evaporation area, with the ambient temperature and relative humidity fluctuating within 2 h in the ranges 20-21 °C and 30-32%, respectively, an average flow-rate of 3.02 μL/min was obtained, with a precision better than 1.2% R.S.D. (n = 61). When applied to the microchip FI-CL system using the luminol/hexacyanoferrate/H2O2 reaction, a precision of 1.4% R.S.D. (n = 11) was obtained for luminol at a sampling frequency of 30 h−1.  相似文献

Alkaline-phosphatase (ALP) catalyses the hydrolysis of 1-naphthyl phosphate to fluorescent 1-naphthol (λex=346 nm, λem=463 nm). This enzymatic reaction was investigated in presence of inhibitors: organochlorine (tetradifon), carbamate (metham-sodium) and organophosphorus pesticides (fenitrothion), heavy metal (Ag+) and CN. The fluorescent signal, which is inversely dependent on the inhibitor concentration, is related to the amount of the inhibitor. Detection limits between 4.1 μM for tetradifon and 91.2 μM for metham-sodium were found. The relative standard deviation (R.S.D.) was between 2.6 and 6.2%.Sol-gel matrices derived from tetramethyl orthosilicate were doped with ALP using microencapsulation. The response of the biosensor based ALP sol-gel encapsulated to 1-naphthyl phosphate was reproducible (R.S.D.=6.6%). Inhibition plots obtained for test pesticides (metham-sodium and tetradifon) display linear calibration in the ranges 194-774 μM and 3.5-28 μM, detection limits of 4.9 and 292.3 μM and R.S.D. of 3.9 and 7.3% for metham-sodium and tetradifon, respectively. The results show that the system is able to detect class compounds such as pesticides and inorganic compounds.  相似文献

Capability of fast analysis of a novel miniaturized capillary electrophoresis with carbon disk electrode amperometric detection (mini-CE-AD) system was demonstrated by determining acetaminophen and p-aminophenol in dosage forms. Factors influencing the separation and detection processes were examined and optimized. Under the optimum conditions, the end-capillary 300 μm carbon disc electrode amperometric detector offered favorable signal-to-noise characteristics at a relatively low potential (+600 mV versus Ag/AgCl) for detecting acetaminophen and p-aminophenol. Two analytes can been separated within 150 s in a 8.5 cm length capillary at a separation voltage of 2000 V using a Na2B4O7-KH2PO4 running buffer (pH 7.2). Acetaminophen and p-aminophenol could be detected down to the 1.4 × 10−6-5.9 × 10−7 mol L−1 level with linearity up to the 1.0 × 10−3 mol L−1 level examined. The inter-day repeatability for analytes in peak current (R.S.D. ≤ 2.3%) and migration times (R.S.D. ≤ 1.3%) were excellent. The proposed mini-CE-AD system should find a wide range of analytical applications in pharmaceutical formulations as an alternative to conventional CE and μ-CE.  相似文献

Methods for short-term BOD analysis (BODst) based on ferricyanide mediator reduction have succeeded in overcoming some problems associated with the standard BOD test analysis (BOD5) such as long-term incubations (5 days), the need to dilute samples and low reproducibility. Here we present a bioassay where a Klebsiella pneumoniae environmental strain successfully reduces ferricyanide without de-aeration of the samples with linear BOD5 ranges between 30 and 500 mg L−1 or 30 and 200 mg L−1, using glucose-glutamic acid solution (GGA) or OECD standards respectively. We further propose a new assay termination solution that allows higher reproducibility and standardization of the cell-based assay, employing formaldehyde (22.7 g L−1) or other compounds in order to stop ferricyanide reduction without affecting the amperometric detection and therefore replace the centrifugation step normally used to stop microbial-driven reactions in ferricyanide-mediated bioassays. These improvements led to an accurate determination of real municipal wastewater samples.  相似文献

A flow-injection method for measuring the peroxide value (PV, mequiv. O2 kg−1) in edible oils is described. The technique is based on spectrophotometric monitoring at 660 nm of methylene blue (MB), generated from leucomethylene blue (LMB) oxidation with peroxides present in oil samples. After being optimized, the method was validated in terms of linearity, precision sensitivity and recovery.Linear calibration graph was obtained in the range 0.1-5 mequiv. O2 kg−1, with a detection limit (S/N  =  3) of 0.014 mequiv. O2 kg−1. The precision of the method (R.S.D., n = 9) for within and between-days is better than 1.5% and 2.2%, respectively at 0.4 mequiv. O2 kg−1. The method was applied successfully to the determination of PV in six edible oil samples, and compared to the classical official method. Using the linear regression test, Student's t-test and variance ratio F-test, there was no significant difference between the compared methods. The proposed method is accurate, simple, cheap and could be used to control edible oil rancidity with a high sample throughputs (30 samples h−1).  相似文献

A highly sensitive optical humidity probe based on reflectance measurements has been developed using Nafion®-crystal violet (CV) films. This sensor can be used to calibrate relative humidity (RH) in the range 0-0.25% with a detection limit (blank signal + 3σb, where σb = the standard deviation (S.D.) of the blank signal) of 0.018% RH (∼4.37 ppm) and exhibited low hysteresis. The sensor films were fully reversible in dry nitrogen and reversal times were shown to be dependent on exposure time and % RH. The response to 1% RH was highly reproducible (S.D. = 1.67%, number of samples (n) = 5). Hydrogen chloride gas did not interfere with the response of the sensor to RH but did reduce sensor reversal times. This sensor displayed sufficient sensitivity that it could be used to detect ppm levels of moisture in process gases such as nitrogen and HCl.  相似文献

The use of menadione (MD) as a pre-column reagent for high performance liquid chromatography (HPLC) analysis of aliphatic thiols is proposed. The reaction was carried out for 5 min at room temperature and pH 8.5. The developed method was applied to the N-acetylcysteine (NAC) analysis of alimentary supplements and pharmaceutical formulations. The effect of the complex matrix was evaluated by the study of the thiol derivatization reaction both in standard and in placebo solutions. The yield of NAC-MD adduct was found to be quantitative at a reagent to thiol molar ratio of about 4 in comparison with an authentic specimen of synthesized NAC adduct, which was characterized by 1H NMR, IR and UV. The routine chromatographic separations were performed on a Synergi MAX-RP column using a mobile phase consisting of methanol/triethylammonium (TEA) phosphate buffer (pH 3; 0.05 mol L−1) 70:30 (v/v) at a flow-rate of 0.4 mL min−1. UV-diode array detection was used setting the wavelength at λ = 260 nm. The validation parameters such as linearity, sensitivity, accuracy, precision, selectivity and ruggedness were found to be highly satisfactory. Similar linear responses were observed by standard and placebo solutions (determination coefficient: 0.9996). Limit of detection was about 0.019 μg g−1. Intra-day precision (relative standard deviation, R.S.D.) was ≤0.81% for NAC to internal standard (IS) peak area ratio, ≤0.28% and ≤0.32%, respectively, for NAC and IS retention times (tR), without significant differences between intra- and inter-day data. NAC recovery studies gave good results (100.12%) with R.S.D. = 1.05%.  相似文献

The usefulness of zone capillary electrophoresis (CZE) in combination with isotachophoresis (cITP) as on-line preconcentration technique was examined for analysis of tripolyphosphate (STPP) in meat and meat products. The mean concentrations of STPP in different types of meat products varied from 39 mg P2O5/100 g to 219 mg P2O5/100 g, these values are below the legal requirements. The detection (LOD) and quantification (LOQ) limits for STPP in extracted solutions were 0.80 mg P2O5/dm3 and 2.69 mg P2O5/dm3, respectively. Obtained results were compared with the Kjeldahl method. Accuracy (97.4-98.3%) was determined using recovery assay based on standard additions method. Precision was evaluated by within-day R.S.D. (1.40-2.19%), between-days R.S.D. (3.00-3.82%) and demonstrates the benefit of using this procedure for the routine analysis of STPP in meat and their products. The F-Snedecor test was employed to compare the precision of the used methods and calculated F-test values (4.00, 6.13) were less than the theoretical (6.39).  相似文献

A sequential injection analysis (SIA) methodology for the fluorimetric determination of aminocaproic acid in pharmaceutical formulations is proposed. The developed analytical procedure is based on the derivatisation reaction of the aminocaproic primary amine with o-phthalaldehyde (OPA) and N-acetylcysteine (NAC) and fluorimetric detection of the formed product (λex = 350 nm; λem = 450 nm). The implementation of a SIA flow system allowed for the development of a simple, fast and versatile automated methodology, which exhibits evident advantages regarding the US Pharmacopoeia 24 (USP 24) reference procedure. By combining the SIA time-based sample insertion with a subsequent zone sampling approach, which permitted to select for detection of a well-defined sample zone, it was possible to implement an on-line dilution strategy that enabled the expansion of the analytical working range of the methodology, and thus its application in dissolution studies, without manifold re-configuration.Linear calibration plots were obtained for aminocaproic acid concentrations up to 6 × 10−5 mol l−1. The developed methodology exhibit a good precision, with a R.S.D. < 2.0% (n = 15) and the detection limit was 2.5 × 10−7 mol l−1. The obtained results complied with those furnished by the reference procedure with a relative deviation lower than 1.2%. No interference was found.  相似文献

A novel biochemical oxygen demand (BOD) sensing method employing a ferricyanide (FC) mediator immobilized in an ion-exchangeable polysiloxane was developed. The ion-exchangeable polysiloxane containing alkylammonium groups (PAPS-Cl) was synthesized by sol-gel reaction of 3-(aminopropyl)trimethoxysilane (APTMOS) catalyzed by hydrochloric acid. FC was combined in PAPS-Cl via ion-association and the product was labeled as PAPS-FC, which was employed for a modified glassy carbon electrode. The apparent diffusion coefficient (Dapp) of FC on the modified glassy carbon electrode was 9.8 × 10−11 cm2 s−1. In a three-electrode system, a linear relationship between the anodic current responses and glucose/glutamate (GGA) concentration was obtained up to 40 mg O2 L−1 (r = 0.994) when the reaction mixture was incubated for 30 min. Single sensor and piece-to-piece reproducibility were less than 3.8 and 7.7%, respectively. The effects of dissolved oxygen, pH, temperature and co-existing substances on the BOD responses were studied. The sensor responses to nine pure organic substances were compared with the conventional BOD5 method and other biosensor methods. Detection results of seawater samples were compared with those obtained from the BOD5 method and showed a good correlation (r = 0.988).  相似文献

A simple and rapid method using micellar electrokinetic capillary chromatography (MEKC) was developed for the separation and determination of acrylamide in potato chips at low levels for the first time. The experimental conditions for the separation and quantification of acrylamide were optimized at first. The optimized conditions were: 50 mmol/L Na2B4O7 and 40 mmol/L SDS at pH 10.0, 12 kV applied voltage, 76 cm total length (67 cm effective length) and 75 μm i.d. capillary, 198 nm wavelength, 15 cm high 25 s hydrodynamics sample injection, 20 °C air-cooling. The linear response of acrylamide concentration ranges from 0.5 to 100 μg/mL with high correlation coefficient (r = 0.9986, n = 9). The LOD and LOQ were estimated to be 0.1 and 0.33 μg/mL based on S/N = 3 and 10. The precision values (expressed as R.S.D.) of intra- and inter-day were 0.86-4.35% and 2.61-9.65%, respectively. Recoveries spiked at levels 2, 20, 60 μg/mL ranged between 90.86% and 99.6% with R.S.D. less than 6.5%. Finally, the developed method has been applied to the analysis of real samples and has achieved satisfactory results. All of these indicated that it was a reliable method for the quantification of acrylamide in potato chips.  相似文献

A novel and sensitive gas sensor was proposed for the determination of carbon tetrachloride based on its cataluminescence (CTL) by oxidation in the air on the surface of nanosized ZnS. The luminescence characteristics and the optimal conditions were investigated in detail. Under the optimized conditions, the linear range of the CTL intensity versus the concentration of carbon tetrachloride was 0.4-114 μg mL−1, with a correlation coefficient (R) of 0.9986 and a limit of detection (S/N = 3) of 0.2 μg mL−1. The relative standard deviation (R.S.D.) for 5.9 μg mL−1 carbon tetrachloride was 2.9% (n = 5). There was no or weak response to common foreign substances including methanol, ethanol, benzene, acetone, formaldehyde, acetaldehyde, dichloromethane, xylene, ammonia and trichloromethane. There was no significant change of the catalytic activity of the sensor for 40 h over 4 days, with a R.S.D. of less than 5% by collecting the CTL intensity once an hour. The proposed method was simple and sensitive, with a potential of detecting carbon tetrachloride in environment and industry grounds. The possible mechanism was also discussed briefly.  相似文献

A new method for solid-phase extraction and preconcentration of trace amounts Hg(II) from environmental samples was developed by using sodium dodecyle sulphate-coated magnetite nanoparticles (SDS-coated Fe3O4 NPs) as a new extractant. The procedure is based on the adsorption of the analyte, as mercury-Michler's thioketone [Hg2(TMK)4]2+ complex on the negatively charged surface of the SDS-coated Fe3O4 NPs and then elution of the preconcentrated mercury from the surface of the SDS-coated Fe3O4 NPs prior to its determination by flow injection inductively coupled plasma-optical emission spectrometry. The effects of pH, TMK concentration, SDS and Fe3O4 NPs amounts, eluent type, sample volume and interfering ions on the recovery of the analyte were investigated. Under optimized conditions, the calibration curve was linear in the range of 0.2-100 ng mL−1 with r2 = 0.9994 (n = 8). The limit of detection for Hg(II) determination was 0.04 ng mL−1. Also, relative standard deviation (R.S.D.) for the determination of 2 and 50 ng mL−1 of Hg(II) was 5.2 and 4.7% (n = 6), respectively. Due to the quantitative extraction of Hg(II) from 1000 mL of the sample solution an enhancement factor as large as 1230-fold can be obtained. The proposed method has been validated using a certified reference materials, and also the method has been applied successfully for the determination of Hg(II) in aqueous samples.  相似文献

A simple, rapid, and accurate method for the separation and determination of alpinetin and cardamonin in Alpinia katsumadai Hayata was developed by combination of flow injection (FI)-micellar electrokinetic chromatography (MEKC) for the first time. The analysis was carried out using an unmodified fused-silica capillary (50 μm i.d.; total length 13.6 cm; effective length 10.3 cm) and direct ultraviolet (UV) detection at 214 nm. The sample throughput was 11-24 samples per hour using the background electrolyte (BGE) containing 4 mM sodium borate-8 mM NaH2PO4 (pH 8.1)-8 mM sodium dodecyl sulfate (SDS)-19% (v/v) ethanol. The repeatabilities (n = 4) reached relative standard deviation values (R.S.D.) of 3.0% and 2.5% for the peak areas and 2.5% and 3.1% for peak heights of alpinetin and cardamonin, respectively. Regression equations revealed linear relationships (r2: 0.9993-0.9994) between the peak area of each analyte and the concentration. Recoveries were in the range 90-92% and 99-105% for alpinetin and cardamonin, respectively.  相似文献

A novel voltammetric sensor for O,O-dimethyl-(2,4-dichlorophenoxyacetoxyl)(3′-nitrophenyl)methinephosphonate (Phi-NO2) based on molecularly imprinted polymer (MIP) film electrode is constructed by using sol-gel technology. The sensor responds linearly to Phi-NO2 over the concentration range of 2.0 × 10−5 to 1.0 × 10−8 mol L−1 and the detection limit is 1.0 × 10−9 mol L−1 (S/N = 3). This sensor provides an efficient way for eliminating interferences from coexisting substances in the solution. The high sensitivity, selectivity and stability of the sensor demonstrates its practical application for a simple and rapid determination of Phi-NO2 in cabbage samples.  相似文献

A capillary zone electrophoresis method was developed for simultaneous determination of nine flavonoids, including two rare flavonols, in Tibetan medicine Anaphalis margaritacea. Baseline separation was performed at pH 9.6 with 25 mM Na2B4O7 and 10 mM NaH2PO4 buffer solution, 20 kV as driving voltage and 275 nm as detection wavelength. Repeatability tests showed that the R.S.D. of both intra- and inter-day migration times and peak areas were less than 5%. Recovery results ranged from 87.9% to 106.1%. Samples of A. margaritacea extracts were analyzed using the validated method, which is useful for its quality control.  相似文献

A highly sensitive and virtually specific method has been developed for the trace and ultra trace 5 ng ml−1-1 μg ml−1 fluorimetric analysis of nitrite. The method is based on the quenching action of nitrite on the native fluorescence of murexide (ammonium purpurate) [λex=349.0 nm, λem=444.5 nm] in the acid range of 0.045-0.315 (M) H2SO4. The method is very precise and accurate (S.D.=±0.4877 and R.S.D.=0.4878% for the determination of 0.1 μg ml−1 of nitrite in 11 replicates). Relatively large excesses of over 35 cations and anions do not interfere. The proposed technique has been successfully applied for the determination of nitrite and nitrate in ground water, surface water and sea water, nitrite in soil and nitrate in forensic samples. The method has also been extended for the analysis of NOx in air.  相似文献

Nanostructured titania-based solid-phase microextraction (SPME) fibers were fabricated through the in situ oxidation of titanium wires with H2O2 (30%, w/w) at 80 °C for 24 h. The obtained SPME fibers possess a ∼1.2 μm thick nanostructured coating consisting of ∼100 nm titania walls and 100-200 nm pores. The use of these fibers for headspace SPME coupled with gas chromatography with electron capture detection (GC-ECD) resulted in improved analysis of dichlorodiphenyltrichloroethane (DDT) and its degradation products. The presented method to detect DDT and its degradation products has high sensitivity (0.20-0.98 ng L−1), high precision (relative standard deviation R.S.D. = 9.4-16%, n = 5), a wide linear range (5-5000 ng L−1), and good linearity (coefficient of estimation R2 = 0.991-0.998). As the nanostructured titania was in situ formed on the surface of a titanium wire, the coating was uniformly and strongly adhered on the titanium wire. Because of the inherent chemical stability of the titania coating and the mechanical durability of the titanium wire substrate, this new SPME fiber exhibited long life span (over 150 times).  相似文献
