We investigated the morphological transitions induced by alkyne/azide Huisgen 1,3‐dipolar cycloaddition reaction in a series of poly(ethylene oxide)‐block‐poly(n‐butyl methacrylate‐random‐propargyl methacrylate) (PEO‐b‐P(nBMA‐r‐PgMA)) diblock copolymers. Studies on the phase behavior of neat diblock copolymers revealed that the interactions between the PEO block and the terminal alkyne groups in the P(nBMA‐r‐PgMA) block significantly affected the miscibility between the two blocks and the crystallization of the PEO block. Phase‐mixed diblock copolymers underwent disorder‐to‐order transitions by blending with Rhodamine B azide and annealing at elevated temperatures. Different morphologies were achieved, not only by controlling the composition of the block copolymer but also by blending the diblock copolymer with different amount of azides. Microphase separated PEO‐b‐P(nBMA‐r‐PgMA) diblock copolymer also exhibited reactivity toward azides, and order‐to‐order transitions were observed. © 2011 Wiley Periodicals, Inc. In this study, an equimolar mixture of oxanorbornenyl‐anthracene (ONB‐anthracene), oxanorbornenyl‐bromide (ONB‐Br), and oxanorbornenyl tosylate (ONB‐OTs) was polymerized via ring opening metathesis polymerization using the first generation Grubbs' catalyst in CH2Cl2 at room temperature to form poly(ONB‐anthracene‐co‐ONB‐Br‐co‐ONB‐OTs)10 copolymer as a main backbone. Next, this main backbone was sequentially clicked with a furan protected maleimide‐terminated poly(methyl methacrylate), 2,2,6,6‐tetramethyl‐1‐piperidinyloxy‐terminated poly(ethylene glycol), and alkyne‐terminated poly(ε‐caprolactone) (PCL20‐alkyne) via Diels–Alder, nitroxide radical coupling, and copper‐catalyzed azide‐alkyne cycloaddition, respectively, to yield a poly(ONB‐g‐PMMA‐co‐ONB‐g‐PEG‐co‐ONB‐g‐PCL)10 heterograft brush copolymer © 2012 Wiley Periodicals, Inc. Comb‐like amphiphilic poly(poly((lactic acid‐co‐glycolic acid)‐block‐poly(ethylene glycol)) methacrylate (poly((PLGA‐b‐PEG)MA)) copolymers were synthesized by radical polymerization. (PLGA‐b‐PEG)MA macromonomer was prepared by ring‐opening bulk polymerization of DL ‐lactide and glycolide using purified poly(ethylene glycol) monomethacrylate (PEGMA) as an initiator. (PLGA‐b‐PEG)MA macromonomer was copolymerized with PEGMA and/or acrylic acid (AA) by radical polymerization to produce comb‐like amphiphilic block copolymers. The molecular weight and chemical structure were investigated by GPC and 1H NMR. Poly((PLGA‐b‐PEG)MA) copolymer aqueous solutions showed gel–sol transition behavior with increasing temperature, and gel‐to‐sol transition temperature decreased as the compositions of the hydrophilic PEGMA and AA increased. The gel‐to‐sol transition temperature of the terpolymers of the poly((PLGA‐b‐PEG)MA‐co‐PEGMA‐co‐AA) also decreased when the pH was increased. The effective micelle diameter obtained from dynamic light scattering increased with increasing temperature and with increasing pH. The critical micelle concentration increased as the composition of the hydrophilic monomer component, PEGMA and AA, were increased. The spherical shape of the hyperbranched polymers in aqueous environment was observed by atomic force microscopy. © 2008 Wiley Periodicals, Inc. A new class of temperature and pH dual‐responsive and injectable supramolecular hydrogel was developed, which was formed from block copolymer poly(ethylene glycol)‐block‐poly[(2‐dimethylamino)ethyl methacrylate] (PEG‐b‐PDMAEMA) and α‐cyclodextrin (α‐CD) inclusion complexes (ICs). The PEG‐b‐PDMAEMA diblock copolymers with different ratio of ethylene glycol (EG) to (2‐dimethylamino)ethyl methacrylate (DMAEMA) (102:46 and 102:96, respectively) were prepared by atom transfer radical polymerization (ATRP). 1H NMR measurement indicated that the ratio of EG unit to α‐CD in the resulted ICs was higher than 2:1. Thermal analysis showed that thermal stability of ICs was improved. The rheology studies showed that the hydrogels were temperature and pH sensitive. Moreover, the hydrogels were thixotropic and reversible. The self‐assembly morphologies of the ICs in different pH and ionic strength environment were studied by transmission electron microscopy. The formed biocompatible micelles have potential applications as biomedical and stimulus‐responsive material. © 2010 Wiley Periodicals, Inc. Multiarm star triblock terpolymers were obtained by using two different click reactions sequentially: Cu(I) catalyzed azide–alkyne and Diels–Alder. The synthetic strategy is described as follows: (poly(methyl methacrylate))n‐(polystyrene)m‐poly(divinyl benzene)) ((PMMA)n‐(PS)m‐polyDVB) multiarm star diblock copolymer was first obtained from an azide–alkyne click reaction of (alkyne‐PS)m‐polyDVB multiarm star polymer with α‐anthracene‐ω‐azide PMMA (anth‐PMMA‐N3), followed by a Diels–Alder click reaction of the anthracene groups at the star periphery with α‐maleimide poly (tert‐butyl acrylate) (PtBA‐MI) or α‐maleimide poly(ethylene glycol) (PEG‐MI) leading to target (PtBA)k‐(PMMA)n‐(PS)m‐polyDVB and (PEG)p‐(PMMA)n‐(PS)m‐polyDVB multiarm star triblock terpolymers. The hydrodynamic diameter of individual multiarm star triblock terpolymers were measured by dynamic light scattering (DLS) to be ~24–27 nm in consistent with the atomic force microscopy (AFM) images on silicon substrates. © 2010 Wiley Periodicals, Inc. An amphiphilic block copolymer, poly(ethylene glycol)‐block‐poly(L ‐lactide‐co‐2‐methyl‐2‐benzoxycarbonyl‐propylene carbonate) [PEG‐b‐P(LA‐co‐MBC)], was synthesized in bulk by the ring‐opening polymerization of L ‐lactide with 2‐methyl‐2‐benzoxycarbonyl‐propylene carbonate (MBC) in the presence of poly(ethylene glycol) as a macroinitiator with diethyl zinc as a catalyst. The subsequent catalytic hydrogenation of PEG‐b‐P(LA‐co‐MBC) with palladium hydroxide on activated charcoal (20%) as a catalyst was carried out to obtain the corresponding linear copolymer poly(ethyleneglycol)‐block‐poly(L ‐lactide‐co‐2‐methyl‐2‐carboxyl‐propylenecarbonate) [PEG‐b‐P(LA‐co‐MCC)] with pendant carboxyl groups. DSC analysis indicated that the glass‐transition temperature (Tg) of PEG‐b‐P(LA‐co‐MBC) decreased with increasing MBC content in the copolymer, and Tg of PEG‐b‐P(LA‐co‐MCC) was higher than that of the corresponding PEG‐b‐P(LA‐co‐MBC). The in vitro degradation rate of PEG‐b‐P(LA‐co‐MCC) in the presence of proteinase K was faster than that of PEG‐b‐P(LA‐co‐MBC), and the cytotoxicity of PEG‐b‐P(LA‐co‐MCC) to chondrocytes from human fetal arthrosis was lower than that of poly(L ‐lactide). © 2005 Wiley Periodicals, Inc. The synthesis of a new cyclic carbonate monomer containing an allyl group was reported and its biodegradable amphiphilic block copolymer, poly(ethylene glycol)‐block‐poly(L ‐lactide‐co‐5‐methyl‐5‐allyloxycarbonyl‐propylene carbonate) [PEG‐b‐P(LA‐co‐MAC)] was synthesized by ring‐opening polymerization (ROP) of L ‐lactide (LA) and 5‐methyl‐5‐allyloxycarbonyl‐1,3‐dioxan‐2‐one (MAC) in the presence of poly (ethylene glycol) as a macroinitiator, with diethyl zinc as a catalyst. 13C NMR and 1H NMR were used for microstructure identification of the copolymers. The copolymer could form micelles in aqueous solution. The core of the micelles is built of the hydrophobic P(LA‐co‐MAC) chains, whereas the shell is set up by the hydrophilic PEG blocks. The micelles exhibited a homogeneous spherical morphology and unimodal size distribution. By using the cyclic carbonate monomer containing allyl side‐groups, crosslinking of the PEG‐b‐P(LA‐co‐MAC) inner core was possible. The adhesion and spreading of ECV‐304 cells on the copolymer were better than that on PLA films. Therefore, this biodegradable amphiphilic block copolymer is expected to be used as a biomaterial for drug delivery and tissue engineering. © 2007 Wiley Periodicals, Inc. This paper presents a new approach to improving the physical stability of biodegradable poly‐(ethylene glycol)‐block‐poly[(DL ‐lactic acid)‐co‐(glycolic acid)] (PEG‐PLGA) micelles. A hydroxyl‐terminated PEG monomethacrylate (PEGmer) macroinitiator was used to prepare a methacrylate‐end‐capped PEG‐PLGA diblock copolymer by the ring‐opening polymerization of D ,L ‐lactide and glycolide. The surface‐exposed methacrylate groups in the shell layer of the micelles can be polymerized with N‐vinyl‐2‐pyrrolidone. The resulting micelles show substantially enhanced stability.  相似文献

The double click reactions (Cu catalyzed Huisgen and Diels–Alder reactions) were used as a new strategy for the preparation of well‐defined heterograft copolymers in one‐pot technique. The synthetic strategy to the various stages of this work is outlined: (i) preparing random copolymers of styrene (St) and p‐chloromethylstyrene (CMS) (which is a functionalizable monomer) via nitroxide mediated radical polymerization (NMP); (ii) attachment of anthracene functionality to the preformed copolymer by the o‐etherification procedure and then conversion of the remaining ? Heteroarm H‐shaped terpolymers, (polystyrene)(poly(methyl methacrylate))‐ poly(tert‐butyl acrylate)‐(polystyrene)(poly(methyl methacrylate)), (PS)(PMMA)‐PtBA‐(PMMA)(PS), and, (PS)(PMMA)‐poly(ethylene glycol)(PEG)‐(PMMA)(PS), through click reaction strategy between PS‐PMMA copolymer (as side chains) with an alkyne functional group at the junction point and diazide end‐functionalized PtBA or PEG (as a main chain). A diblock copolymer poly(ethylene glycol)-block-polystyrene or PEG-b-PS with an olefinic double bond at the PEG and PS junction has been prepared by modular synthesis via “click” chemistry. This involved the synthesis of PS by atom transfer radical polymerization and the nucleophilic substitution of the terminal bromide group with azide to yield azide-terminated PS. PEG with an alkynyl terminal group was prepared from reacting carboxyl-end-functionalized PEG with 4-hydroxybut-2-enyl prop-2′-ynyl succinate, which contained an alkynyl group as well as an olefin group. The PS and PEG polymers were linked via the 1,3-dipolar cycloaddition of the end azide and alkyne groups. The obtained copolymer was characterized by 1H NMR spectroscopy and size exclusion chromatography (SEC). SEC analysis indicated that the diblock copolymer produced could be readily cleaved by ozonolysis to regenerate the constituent homopolymers.  相似文献

A poly(ethylene glycol)‐b‐poly(L ‐lysine) diblock copolymer (PEG‐b‐PLL) was synthesized. Micellization of this hydrophilic copolymer due to the block‐specific threading of α‐cyclodextrin (α‐CD) molecules onto the polyethylene glycol (PEG) block yielded supramolecular‐structured nanoparticles, which undergoes pH‐inducible gelation in aqueous media. The pH‐inducible gelation of supramolecular micelle in water appeared to be completely reversible upon pH changes. The synergetic effect of selective complexation between PEG block and α‐CD and the pH‐inducible hydrophobic interaction between PLL blocks at pH 10 was believed to be the driving force for the formation of the supramolecular hydrogel. 1H NMR and wide angle X‐ray diffraction (WAXD) were employed to confirm the inclusion complexation between α‐CD and PEG block. Meanwhile, the morphology of the micellized nanoparticles was investigated by transmission electron microscopy (TEM). The thermal stability of inclusion complexes (ICs) was investigated and the rheologic experiment was conducted to reveal the micelle‐gel transition. Such pH‐induced reversible micelle‐gel transition of the supramolecular aggregates may find applications in several fields, for example as advanced biomedical material possessing stimulus‐responsiveness. © 2008 Wiley Periodicals, Inc. Double hydrophilic diblock copolymer, poly(N,N‐dimethylacrylamide)‐b‐poly(N‐isopropylacrylamide‐co‐3‐azidopropylacrylamide) (PDMA‐b‐P(NIPAM‐co‐AzPAM), containing azide moieties in one of the blocks was synthesized via consecutive reversible addition‐fragmentation chain transfer polymerization. The obtained diblock copolymer molecularly dissolves in aqueous solution at room temperature, and can further supramolecularly self‐assemble into core‐shell nanoparticles consisting of thermoresponsive P(NIPAM‐co‐AzPAM) cores and water‐soluble PDMA coronas above the lower critical solution temperature of P(NIPAM‐co‐AzPAM) block. As the micelle cores contain reactive azide residues, core crosslinking can be facilely achieved upon addition of difunctional propargyl ether via click chemistry. In an alternate approach in which the PDMA‐b‐P(NIPAM‐co‐AzPAM) diblock copolymer was dissolved in a common organic solvent (DMF), the core‐crosslinked (CCL) micelles can be fabricated via “click” crosslinking upon addition of propargyl ether and subsequent dialysis against water. CCL micelles prepared by the latter approach typically possess larger sizes and broader size distributions, compared with that obtained by the former one. In both cases, the obtained (CCL) micelles possess thermoresponsive cores, and the swelling/shrinking of which can be finely tuned with temperature, rendering them as excellent candidates as intelligent drug nanocarriers. Because of the high efficiency and quite mild conditions of click reactions, we expect that this strategy can be generalized for the structural fixation of other self‐assembled nanostructures. © 2007 Wiley Periodicals, Inc. In this article, a light and pH dual‐sensitive block copolymer PEG‐b‐poly(MPC‐Azo/DEA) was facilely prepared for the first time by azide‐alkyne click chemistry between amphiphilic block copolymer bearing pendant alkynyl group poly(ethylene glycol)‐poly(5‐methyl‐5‐propargylxycarbonyl‐1,3‐dioxane‐2‐one) (PEG‐b‐poly(MPC)) and two azide‐containing compounds azobenzene derivative (Azo‐N3) and 2‐azido‐1‐ethyl‐diethylamine (DEA‐N3). Light response of the polymeric nanoparticles benefits from the azobenzene segments and pH responsiveness is attributed to DEA moieties. The prepared copolymer could self‐assemble into spherical micelle particles. The morphological changes of these particles in response to dual stimuli were investigated by UV/vis spectroscopy, dynamic light scattering (DLS), and transmission electron microscopy (TEM). Nile Red (NR) was utilized as probe, and fluorescence spectroscopy was served as an evidence for the enhanced release of cargos from polymeric nanoparticles under combined stimulation. Anticancer drug, DOX was loaded into the nanoparticles and the loaded‐DOX could be released from these nanoparticles under dual stimuli. MTT assays further demonstrated that PEG‐b‐poly(MPC) and PEG‐b‐poly(MPC‐Azo/DEA) were of biocompatibility and low toxicity against HepG2 cells as well as SMCC‐7721 cells. More importantly, the prepared DOX‐loaded nanoparticles exhibited good anticancer ability for the two cells. The synthesized light and pH dual‐sensitive biodegradable polymeric nanoparticles were expected to be platforms for precisely controlled release of encapsulated molecules. © 2017 Wiley Periodicals, Inc. Biodegradable, amphiphilic, diblock poly(ε‐caprolactone)‐block‐poly(ethylene glycol) (PCL‐b‐PEG), triblock poly(ε‐caprolactone)‐block‐poly(ethylene glycol)‐block‐poly(ε‐caprolactone) (PCL‐b‐PEG‐b‐PCL), and star shaped copolymers were synthesized by ring opening polymerization of ε‐caprolactone in the presence of poly(ethylene glycol) methyl ether or poly(ethylene glycol) or star poly(ethylene glycol) and potassium hexamethyldisilazide as a catalyst. Polymerizations were carried out in toluene at room temperature to yield monomodal polymers of controlled molecular weight. The chemical structure of the copolymers was investigated by 1H and 13C NMR. The formation of block copolymers was confirmed by 13C NMR and DSC investigations. The effects of copolymer composition and molecular structure on the physical properties were investigated by GPC and DSC. For the same PCL chain length, the materials obtained in the case of linear copolymers are viscous whereas in the case of star copolymer solid materials are obtained with low Tg and Tm temperatures. © 2007 Wiley Periodicals, Inc. Copper‐catalyzed azide‐alkyne cycloaddition (CuAAC) was used to prepare glycosylated polyethylene (PE)–poly(ethylene glycol) (PEG) amphiphilic block copolymers. The synthetic approach involves preparation of alkyne‐terminated PE‐b‐PEG followed by CuAAC reaction with different azide functionalized sugars. The alkyne‐terminated PE‐b‐PEG was prepared by etherification reaction between hydroxyl‐terminated PE‐b‐PEG (Mn ~ 875 g mol?1) and propargyl bromide and azidoethyl glycosides were prepared by glycosylation of 2‐azidoethanol. Atmospheric pressure solids analysis probe‐mass spectrometry was used as a novel solid state characterization tool to determine the outcome of the CuAAC click reaction and end‐capping of PE‐b‐PEG by the azidoethyl glycoside group. The aqueous solution self‐assembly behavior of these amphiphilic glycosylated polymers was explored by TEM and dye solubilization studies. Carbohydrate‐bearing spherical aggregates with the ability to solubilize a hydrophobic dye were observed. The potential of these amphiphilic glycosylated polymers to self‐assemble via electro‐formation into giant carbohydrate‐bearing polymersomes was also investigated using confocal fluorescence microscopy. A hetero‐arm star polymer, poly(ethylene glycol)‐poly(N‐isopropylacrylamide)‐poly(L‐lysine) (PEG‐PNIPAM‐PLys), was synthesized by “clicking” the azide group at the junction of PEG‐b‐PNIPAM diblock copolymer with the alkyne end‐group of poly(L‐lysine) (PLys) homopolymer via 1,3‐dipolar cycloaddition. The resultant polymer was characterized by gel permeation chromatography, proton nuclear magnetic resonance, and Fourier transform infrared spectroscopes. Surprisingly, the PNIPAM arm of this hetero‐arm star polymer nearly lose its thermal responsibility. It is found that stable polyelectrolyte complex micelles are formed when mixing the synthesized polymer with poly(acrylic acid) (PAA) in water. The resultant polyelectrolyte complex micelles are core‐shell spheres with the ion‐bonded PLys/PAA chains as core and the PEG and PNIPAM chains as shell. The PNIPAM shell is, as expected, thermally responsive. However, its lower critical solution temperature is shifted to 37.5 °C, presumably because of the existence of hydrophilic components in the micelles. Such star‐like PEG‐PNIPAM‐PLys polymer with different functional arms as well as its complexation with anionic polymers provides an excellent and well‐defined model for the design of nonviral vectors to deliver DNA, RNA, and anionic molecular medicines. © 2009 Wiley Periodicals, Inc. Well‐defined azobenzene‐containing side‐chain liquid crystalline diblock copolymers composed of poly[6‐(4‐methoxy‐azobenzene‐4′‐oxy) hexyl methacrylate] (PMMAZO) and poly(γ‐benzyl‐L ‐glutamate) (PBLG) were synthesized by click reaction from alkyne‐ and azide‐functionalized homopolymers. The alkyne‐terminated PMMAZO homopolymers were synthesized by copper‐mediated atom transfer radical polymerization with a bromine‐containing alkyne bifunctional initiator, and the azido‐terminated PBLG homopolymers were synthesized by ring‐opening polymerization of γ‐benzyl‐L ‐glutamate‐N‐carboxyanhydride in DMF at room temperature using an amine‐containing azide initiator. The thermotropic phase behavior of PMMAZO‐b‐PBLG diblock copolymers in bulk were investigated using differential scanning calorimetry and polarized light microscopy. The PMMAZO‐b‐PBLG diblock copolymers exhibited a smectic phase and a nematic phase when the weight fraction of PMMAZO block was more than 50%. Photoisomerization behavior of PMMAZO‐b‐PBLG diblock copolymers and the corresponding PMMAZO homopolymers in solid film and in solution were investigated using UV–vis. In solution, trans–cis isomerization of diblock copolymers was slower than that of the corresponding PMMAZO homopolymers. Hetero‐arm star ABC‐type terpolymers, poly(methyl methacrylate)‐polystyrene‐poly(tert‐butyl acrylate) (PMMA‐PS‐PtBA) and PMMA‐PS‐poly(ethylene glycol) (PEG), were prepared by using “Click” chemistry strategy. For this, first, PMMA‐b‐PS with alkyne functional group at the junction point was obtained from successive atom transfer radical polymerization (ATRP) and nitroxide‐mediated radical polymerization (NMP) routes. Furthermore, PtBA obtained from ATRP of tBA and commercially available monohydroxyl PEG were efficiently converted to the azide end‐functionalized polymers. As a second step, the alkyne and azide functional polymers were reacted to give the hetero‐arm star polymers in the presence of CuBr/N,N,N′,N″,N″‐pentamethyldiethylenetriamine ( PMDETA) in DMF at room temperature for 24 h.
