DNA hypermethylation of CpG islands plays an important role in gene regulation during cancer development. Many techniques have been developed to detect global DNA methylation in cancer cells compared to normal tissues. This knowledge helps us to better understand cancer progression and also aids in the development of new biomarker for early cancer detection. New prognostic tools for monitoring drug efficacy during cancer treatment can also be developed. In this review, we will examine the different techniques that have been used to study DNA methylation, as well as the emerging high resolution, high throughput techniques for identification of methylated regions to defining cancer related genes in the cancer methylome.  相似文献

In Western countries, breast cancer tends to occur in older postmenopausal women. However, in Asian countries, the proportion of younger premenopausal breast cancer patients is increasing. Increasing evidence suggests that the gut microbiota plays a critical role in breast cancer. However, studies on the gut microbiota in the context of breast cancer have mainly focused on postmenopausal breast cancer. Little is known about the gut microbiota in the context of premenopausal breast cancer. This study aimed to comprehensively explore the gut microbial profiles, diagnostic value, and functional pathways in premenopausal breast cancer patients. Here, we analyzed 267 breast cancer patients with different menopausal statuses and age-matched female controls. The α-diversity was significantly reduced in premenopausal breast cancer patients, and the β-diversity differed significantly between breast cancer patients and controls. By performing multiple analyses and classification, 14 microbial markers were identified in the different menopausal statuses of breast cancer. Bacteroides fragilis was specifically found in young women of premenopausal statuses and Klebsiella pneumoniae in older women of postmenopausal statuses. In addition, menopausal-specific microbial markers could exhibit excellent discriminatory ability in distinguishing breast cancer patients from controls. Finally, the functional pathways differed between breast cancer patients and controls. Our findings provide the first evidence that the gut microbiota in premenopausal breast cancer patients differs from that in postmenopausal breast cancer patients and shed light on menopausal-specific microbial markers for diagnosis and investigation, ultimately providing a noninvasive approach for breast cancer detection and a novel strategy for preventing premenopausal breast cancer.Subject terms: Bacterial genetics, Breast cancer  相似文献

Clinically, different approaches are adopted worldwide for the treatment of cancer, which still ranks second among all causes of death. Immunotherapy for cancer treatment has been the focus of attention in recent years, aiming for an eventual antitumoral effect through the immune system response to cancer cells both prophylactically and therapeutically. The application of nanoparticulate delivery systems for cancer immunotherapy, which is defined as the use of immune system features in cancer treatment, is currently the focus of research. Nanomedicines and nanoparticulate macromolecule delivery for cancer therapy is believed to facilitate selective cytotoxicity based on passive or active targeting to tumors resulting in improved therapeutic efficacy and reduced side effects. Today, with more than 55 different nanomedicines in the market, it is possible to provide more effective cancer diagnosis and treatment by using nanotechnology. Cancer immunotherapy uses the body’s immune system to respond to cancer cells; however, this may lead to increased immune response and immunogenicity. Selectivity and targeting to cancer cells and tumors may lead the way to safer immunotherapy and nanotechnology-based delivery approaches that can help achieve the desired success in cancer treatment.  相似文献

Exosomes play a vital role in cell–cell communication within the cancer microenvironment. Exosomal long noncoding RNAs (lncRNAs) are important regulators in cancer development and are involved in multiple processes, including cancer cell proliferation, angiogenesis, metastasis, drug resistance, and immunomodulation. Changes in the levels of exosomal lncRNAs often appear with the occurrence and development of cancer. Therefore, exosomal lncRNAs can be used as biomarkers for cancer diagnosis and prognosis. Exosomal lncRNAs can also indicate the treatment response of patients receiving chemotherapy. Moreover, exosomal lncRNAs are potential therapeutic targets for cancer treatment. In this review, we summarize the role of exosomal lncRNAs in cancer biology as well as in clinical management. A more comprehensive and in-depth understanding of the role of exosomal lncRNAs in cancer may help us better understand the mechanism of cancer development and clinically manage cancer patients.Subject terms: Cancer, Molecular biology  相似文献

提出了一种新的基于傅里叶变换红外光谱(Fourier Transform Infrared Spectroscopy, FTIR)的小波特征提取与支持向量机(SVM)分类方法以提高FTIR对早期肺癌的诊断准确率. 对肺正常组织、早期肺癌及进展期肺癌组织的FTIR, 利用连续小波(CW)多分辨率分析法提取9个特征量, 支持向量机把其分为正常组与非正常组(包括早期肺癌和进展期肺癌), 对正常组织、早期肺癌和进展期肺癌的识别, 多项式核函数和径向基函数的识别准确率最高. 多项式核函数对正常组织、早期肺癌和进展期肺癌的识别准确率分别为100%, 95%及100%; 径向基函数分别为100%, 95%和100%. 实验结果表明FTIR-CW-SVM模式分类方法对正常肺癌组织、早期肺癌及进展肺癌的识别具有较好的可行性.  相似文献

目的研究并探讨人乳头状瘤病毒(human papillomavirus,简称HPV)在宫颈炎、宫颈癌前病变、宫颈癌中的检测价值及临床意义。方法选取菏泽市妇幼保健院2014年1月—2016年12月期间收治的120例慢性宫颈炎患者、90例宫颈癌前病变患者、60例宫颈癌患者作为研究对象,分别设置为宫颈炎组、癌前病变组、宫颈癌组,并将癌前病变组分为CIN Ⅰ级、CIN Ⅱ级、CIN Ⅲ级,所有患者均接受HPV检测,比较其HPV阳性率、HPV DNA负荷载量、HPV感染持续时间,计算HPV感染持续时间、HPV DNA负荷载量与宫颈癌发生的相关性。结果宫颈炎组、癌前病变组、宫颈癌组的HPV阳性率、HPV DNA负荷载量、HPV感染持续时间比较,差异均具有统计学意义(P0.05),从高至低依次为宫颈癌组、癌前病变组、宫颈炎组,且不同分级宫颈癌前病变患者的HPV DNA负荷载量、HPV感染持续时间均存在显著差异(P0.05);经相关性分析,HPV感染持续时间、HPV DNA负荷载量均与宫颈癌的发生密切相关,呈正相关。结论HPV感染可能参与到宫颈癌前病变、宫颈癌的发生、发展中,开展HPV检测可对宫颈癌前病变、宫颈癌进行筛查,有利于宫颈癌的防治。  相似文献

报道了湛江市胃癌患者发中微量元素的变化信息。胃癌患者与正常人相比,Zn、Zn/Cu、Mg、Ca显著降低(P<0.05);胃癌与胃溃疡患者相比,Zn、Zn/Cu、Mg均呈显著降低(P<0.05)。因此,发中微量元素化学信息的变化,可作为本地区胃癌的辅助诊断和胃癌与胃溃疡的区别辅助诊断的依据之一。  相似文献

Delivering alternative strategies to deal with cancer is a huge milestone in research. Cancer cells can give a senescence response as consequence of cellular stress or external stimuli such as the use of chemotherapies, which could end up eventually in cancer relapse. Controlling cellular senescence will surely open new cancer treatment approaches. Cancer senescence induction can be used as an added timeframe to look for alternative treatments and senolytic drugs to avoid cancer relapse destroying the senescent cells (SnCs). Within cancer senescence research, metal complexes are underdeveloped in comparison with that of organic molecules or nanoparticles. Herein, we highlight the scarce investigation performed with metal complexes in the field of senescence and how a great input on them could be a huge step towards the search of alternative cancer treatments.  相似文献

Accumulating evidence in recent years indicates that DNA methylation (5-methyl-2′-deoxycytidine, 5-mdC) and hydroxymethylation (5-hydroxymethyl-2′-deoxycytidine, 5-hmdC) have been implicated in various biological processes, and the aberrations of these DNA cytosine modifications is tightly associated with cancer. N6-methyl-2′-deoxyadenosine (m6dA), as a newly discovered epigenetic modification in genome of mammals, has been demonstrated to play vital regulatory roles in tumorigenesis. However, the content information of m6dA in human tumor tissues is still limited and pan-cancer analysis of these DNA epigenetic modifications is lacked. Herein, we developed a sensitive and robust stable isotope-diluted hydrophilic interaction liquid chromatography-tandem mass spectrometry (HILIC-MS/MS) method for accurate quantification of m6dA, 5-mdC and 5-hmdC in genomic DNA from 82 pairs of human tumor tissues and matched tumor-adjacent normal tissues. The types of tumors included esophagus cancer, lung cancer, breast cancer, liver cancer, pancreatic cancer, gastric cancer, stromal tumor and colorectal cancer. Compared to the normal tissues, we revealed the level of m6dA was increased in tumor tissues of esophagus cancer, lung cancer and liver cancer, whereas the level of m6dA was diminished in tumor tissues of pancreatic cancer and gastric cancer; while the contents of 5-mdC and 5-hmdC exhibited significant decrease in tumor tissues of most types of cancer. It is worth noting that we revealed, for the first time, the content of genomic m6dA in pancreatic cancer, stromal tumor and colorectal cancer. The significant changes of these DNA epigenetic modifications indicate they may serve as indicators of cancers. In addition, this study will benefit for better understanding of the regulatory roles of these DNA epigenetic modifications in cancers.  相似文献

The incidence rates of urinary bladder cancer continue to rise yearly, and thus new therapeutic approaches and early diagnostic markers for bladder cancer are urgently needed. Thus, identifying the key mediators and molecular mechanisms responsible for the survival of bladder cancer has valuable implications for the development of therapy. In this study, the role of BLT2, a receptor for leukotriene B((4)) (LTB((4))) and 12(S)-hydroxyeicosatetraenoic acid (HETE), in the survival of bladder cancer 253J-BV cells was investigated. We found that the expression of BLT2 is highly elevated in bladder cancer cells. Also, we observed that blockade of BLT2 with an antagonist or BLT2 siRNA resulted in cell cycle arrest and apoptotic cell death, suggesting a role of BLT2 in the survival of human bladder cancer 253J-BV cells. Further experiments aimed at elucidating the mechanism by which BLT2 mediates survival revealed that enhanced level of reactive oxygen species (ROS) are generated via a BLT2-dependent up-regulation of NADPH oxidase members NOX1 and NOX4. Additionally, we observed that inhibition of ROS generation by either NOX1/4 siRNAs or treatment with an ROS-scavenging agent results in apoptotic cell death in 253J-BV bladder cancer cells. These results demonstrated that a 'BLT2-NOX1/4-ROS' cascade plays a role in the survival of this aggressive bladder cancer cells, thus pointing to BLT2 as a potential target for anti-bladder cancer therapy.  相似文献

采用ICP-AES法测定了肺癌、肝癌、胃癌患者头发中24种无机元素的含量,与对照组比较,探讨对癌症实验诊断有意义的指标。结果表明:肺癌、肝癌、胃癌患者头发中Al、Cu、Co、Ni、Sr、Zn、Mn、Mo等元素含量与对照组存在显著差异(P〈0.05)。因此采用多元素谱作为上述癌症辅助诊断指标相对于采用单元素值或元素比值作为诊断标准更具参考价值。  相似文献

Poor outcome of pancreatic cancer necessitates development of an early diagnostic method to reduce mortality. No reliable early diagnostic test for pancreatic cancer detection has been developed and validated to date. In the current study, metabolic profiling of plasma samples from selected cancer patients and noncancerous controls was performed to seek novel metabolic biomarkers of pancreatic cancer. A comprehensive mass spectrometry based analytical platform established at the Metabolomics Core of the UC Davis Genome Center allowed detection of multiple compounds previously unreported in plasma from pancreatic cancer patients. It was found that selective amino acids, bile acids, and polar lipids were detected with increased or decreased levels in pancreatic cancer samples compared to controls. These findings on blood plasma levels of the relevant metabolites might be very useful clinical parameters for early diagnosis of pancreatic cancer. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

Blood serum from people suffering from cancer and healthy subjects was subjected to a comparative study by X-ray diffraction. The diffraction patterns were referred to that of pure water. The patterns obtained for blood serum of healthy subjects were similar to that of pure water, while those of cancer patients (two kinds of cancer changes) were different. The former similarity is accounted for by the dominant interaction of water molecules in healthy blood serum with non-polar side chains of amino acids, stabilising the serum structure. In samples from cancer patients the structure of water in blood serum is destroyed because of enhanced interactions of water with polar molecules of conformationally changed proteins. This observation suggests X-ray examination of blood serum from cancer patients and healthy subjects, indicating X-ray diffraction as a diagnostic tool for the occurrence of cancer. The results of this work have shown that the presence of protein affected by cancer has a destructive effect on the structure of water in human serum. The results reported confirm the earlier finding relating cancer changes to optical circular birefringence effects.  相似文献

用实时荧光定量PCR方法检测56对人卵巢癌组织及对应的癌旁组织中iASPP(Inhibitor of ASPP fami-ly)mRNA表达水平,应用受试者工作特征曲线(Receiver operating characteristic curve,ROC曲线)分析癌组织与癌旁组织中iASPP mRNA表达水平,探索iASPP在卵巢癌发生中的作用。从细胞水平进一步研究其作用机制,用siRNA干扰的方法使iASPP表达降低后用Hoechst 33342和CCK-8分别检测iASPP沉默后对人类卵巢癌细胞株OVCA420的影响。结果显示,卵巢癌组织中iASPP表达较癌旁组织明显增高;iASPP沉默后,细胞凋亡增多,细胞增殖水平降低。据此推断,iASPP mRNA水平对卵巢癌的临床诊断具有一定价值,可作为卵巢癌治疗的一个重要靶点。  相似文献

Breast cancer persists as a diffuse source of cancer despite persistent detection and treatment. Flavonoids, a type of polyphenol, appear to be a productive option in the treatment of breast cancer, because of their capacity to regulate the tumor related functions of class of compounds. Plant polyphenols are flavonoids that appear to exhibit properties which are beneficial for breast cancer therapy. Numerous epidemiologic studies have been performed on the dynamic effect of plant polyphenols in the prevention of breast cancer. There are also subclasses of flavonoids that have antioxidant and anticarcinogenic activity. These can regulate the scavenging activity of reactive oxygen species (ROS) which help in cell cycle arrest and suppress the uncontrolled division of cancer cells. Numerous studies have also been performed at the population level, one of which reported a connection between cancer risk and intake of dietary flavonoids. Breast cancer appears to show intertumoral heterogeneity with estrogen receptor positive and negative cells. This review describes breast cancer, its various factors, and the function of flavonoids in the prevention and treatment of breast cancer, namely, how flavonoids and their subtypes are used in treatment. This review proposes that cancer risk can be reduced, and that cancer can be even cured by improving dietary intake. A large number of studies also suggested that the intake of fruit and vegetables is associated with reduced breast cancer and paper also includes the role and the use of nanodelivery of flavonoids in the healing of breast cancer. In addition, the therapeutic potential of orally administered phyto-bioactive compounds (PBCs) is narrowed because of poor stability and oral bioavailability of compounds in the gastrointestinal tract (GIT), and solubility also affects bioavailability. In recent years, creative nanotechnology-based approaches have been advised to enhance the activity of PBCs. Nanotechnology also offers the potential to become aware of disease at earlier stages, such as the detection of hidden or unconcealed metastasis colonies in patients diagnosed with lung, colon, prostate, ovarian, and breast cancer. However, nanoformulation-related effects and safety must not be overlooked. This review gives a brief discussion of nanoformulations and the effect of nanotechnology on herbal drugs.  相似文献

Basal-type breast cancers are among the most aggressive and deadly breast cancer subtypes, displaying a high metastatic ability associated with mesenchymal features. However, the molecular mechanisms underlying the maintenance of mesenchymal phenotypes of basal-type breast cancer cells remain obscure. Here, we report that KRAS is a critical regulator for the maintenance of mesenchymal features in basal-type breast cancer cells. KRAS is preferentially activated in basal-type breast cancer cells as compared with luminal type. By loss and gain of KRAS, we found that KRAS is necessary and sufficient for the maintenance of mesenchymal phenotypes and metastatic ability through SLUG expression. Taken together, this study demonstrates that KRAS is a critical regulator for the metastatic behavior associated with mesenchymal features of breast cancer cells, implicating a novel therapeutic target for basal-type breast cancer.  相似文献

Bladder cancer is the most common malignant tumor in the urinary system,with high morbidity,mortality and recurrence after surgery.However,current bladder cancer urine diagnosis methods are limited by the low accuracy and specificity due to the low abundance of bladder cancer biomarkers in the urine with complex biological environments.Herein,we present a high stability indium gallium zinc oxide field effect transistor(IGZO-FET) biosensor for efficient identification of bladder cancer biomarkers from human urine samples.The recognition molecular functionalized IGZO-FET biosensor exhibits stable electronic and sensing performance due to the large-area fabrication of IGZO thin-film FET.Owing to the excellent electrical performance of IGZO-FET,the IGZO-FET biosensor exhibits high sensitivity and extremely low detection limit(2.7 amol/L) towards bladder cancer biomarkers.The IGZO-FET biosensor is also able to directly detect bladder tumor biomarker in human urine with high sensitivity and specificity,and could differentiate bladder cancer patients' urine samples from healthy donors effectively.These results indicate that our designed high-performance biosensor shows great potential in the application of portable digital bladder cancer diagnosis devices.  相似文献

Ultraviolet (UV) exposure has an array of damaging effects and is the main cause of skin cancer in humans. Nonmelanoma skin cancer (NMSC), including basal cell carcinoma and squamous cell carcinoma, is the most common type of cancer. Incidence of NMSC has increased due to greater UV radiation, increased life expectancy and other changes in lifestyle; the annual cost of skin cancer treatment in the United States has increased concurrently to around eight billion dollars. Because of these trends, novel approaches to skin cancer prevention have become an important area of research to decrease skin cancer morbidity and defray the costs associated with treatment. Chemoprevention aims to prevent or delay the development of skin cancer through the use of phytochemicals. Use of phytochemicals as chemopreventive agents has gained attention due to their low toxicity and anticarcinogenic properties. Phytochemicals also exhibit antioxidant, anti‐inflammatory and antiproliferative effects which support their use as chemopreventive agents, particularly for skin cancer. Preclinical and human studies have shown that phytochemicals decrease UV‐induced skin damage and photocarcinogenesis. In this review article, we discuss the selected phytochemicals that may prevent or delay UV‐induced carcinogenesis and highlight their potential use for skin protection.  相似文献

Breast tumors were the first tumors of epithelial origin shown to follow the cancer stem cell model. The model proposes that cancer stem cells are uniquely endowed with tumorigenic capacity and that their aberrant differentiation yields non-tumorigenic progeny, which constitute the bulk of the tumor cell population. Breast cancer stem cells resist therapies and seed metastases; thus, they account for breast cancer recurrence. Hence, targeting these cells is essential to achieve durable breast cancer remissions. We identified compounds including selective antagonists of multiple serotonergic system pathway components required for serotonin biosynthesis, transport, activity via multiple 5-HT receptors (5-HTRs), and catabolism that reduce the viability of breast cancer stem cells of both mouse and human origin using multiple orthologous assays. The molecular targets of the selective antagonists are expressed in breast tumors and breast cancer cell lines, which also produce serotonin, implying that it plays a required functional role in these cells. The selective antagonists act synergistically with chemotherapy to shrink mouse mammary tumors and human breast tumor xenografts primarily by inducing programmed tumor cell death. We hypothesize those serotonergic proteins of diverse activity function by common signaling pathways to maintain cancer stem cell viability. Here, we summarize our recent findings and the relevant literature regarding the role of serotonin in breast cancer.  相似文献
