The first example of a catalytic asymmetric thia hetero-Diels-Alder reaction, in which a chiral Lewis acid activates the thiocarbonyl dienophile, is described. The reactions catalyzed by copper(II)-bis(oxazolines) complexes of three dithioesters with simple dienes (such as cyclopentadiene or 2,3-dimethyl-1,3-butadiene) afford optically active dihydrothiopyrans with ees up to 82%. Some aspects of the reaction were investigated by computational means.  相似文献

Diels-Alder reactions of the acrylate of (S)-ethyl lactate with cyclopentadiene proceed with diastereoface-selectivity of up to 85:15 (non-catalyzed) and 93:7 (TiCl4-promoted). Depending on the Lewis acid, products of inverse configuration are obtained. In conjunction with facile product analysis by LC, effective means for suppression of polymerization of the diene, and virtually epimerization-free ester hydrolysis these findings constitute a method for large scale practical applications of the asymmetric Diels-Alder reaction.  相似文献

The reaction of cyclopentadiene and various bidentate dienophiles in the presence of a catalytic amount of Pt(II)- or Pd(II)-BINAP complex proceeds with excellent endo/exo selectivity as well as endo enantioselectivity (up to 99% ee).  相似文献

Enantiomerically pure 2-p-tolylsulfinylmaleates 1, 2 and 3 have been readily prepared by Knoevenagel reaction between (S)-menthyl p-toluenesulfinate and glyoxylic acid. Their asymmetric Diels-Alder reactions with cyclopentadiene have been studied under a wide range of uncatalyzed and catalyzed conditions and the stereochemical results have been explained by assuming a steric control approach, in term of S-cis or S-trans favoured conformations. Uncatalyzed Diels-Alder reactions of 1 and some Lewis acid catalyzed Diels-Alder reactions of 2 show high facial and endo selectivities. The facial selectivity of dienophile 2 highly depends on the Lewis acid, whereas reactivity of 1 and 3 is very sensitive to the solvent. These sulfinylmaleates 1, 2 and 3 act as synthetic equivalents of chiral acetylenedicarboxylates in Diels-Alder reactions after basic elimination of the sulfinylic moiety in the resulting adducts.  相似文献

Three possible reaction schemes for the cycloaddition reaction between methyleneketene and cyclopentadiene were studied by DFT (density functional theory) and ab initio calculations. All of these cycloaddition reactions are exothermic, concerted but nonsynchronous processes. The computed activation energies indicate that the norbornene product yielded from a 1,2-addition of methyleneketene with cyclopentadiene (reaction (1)) is the primary product. The performance of various computational methodologies, MP2, MP4, and CCSD(T), in conjunction with a wide array of basis sets, 6-31G(d), 6-311+G(d,p), aug-cc-pVDZ, and aug-cc-pVTZ, in obtaining reliable activation and reaction energies of the reactions under investigation has been critically analyzed.  相似文献

Carbocyclic nucleosides are of considerable interest for the development of new therapeutic agents. A key reaction in the preparation of many such nucleoside analogues is dihydroxylation of appropriately substituted cyclopentenes. Although often considered a routine reaction, in this paper, we report the dramatic influence of substituents on the facial selectivity of dihydroxylations. The substituted cyclopentene substrates are derived from acylnitroso cycloaddition reactions of cyclopentadiene, followed by N-O reduction and efficient enzymatic resolution. The results are directly utilized in a very efficient asymmetric synthesis of an antiviral carbocyclic nucleoside, noraristeromycin 5. Extensions toward the synthesis of carbocyclic sinefungin 7 document the importance of realizing the substituent dependence of the dihydroxylation reaction.  相似文献

利用半经验分子轨道理论AM1方法,研究了烯酮及取代烯酮与环戊二烯环加成反应机理。采用Berny梯度法优化得到各反应的过渡态和中间体,并进行了振动分析确认。计算结果表明,该环加成反应是按照协同的非同步途径进行的,经过一个四元环发生扭曲的过渡态,并有部分电荷从环戊二烯迁移到烯酮或取代烯酮上,前线轨道分析表明反应机理为“2×[1+1]”机理;而氯甲基取代的烯酮与环戊二烯的环加成反应是按照分步途径发生的。计算结果可以很好地说明实验所观察到的立体选择性,并根据烯酮上取代基的电子效应和位阻效应对反应机理的影响进行了分析。  相似文献

The reaction between methyl N-acetyl-,β-didehydroalaninate and cyclopentadiene in the presence of several chiral Lewis acids is studied and the results obtained are compared with those described for the reactions of the same diene with chiral N-acetyl-,β-didehydroalaninates. In the presence of the titanium complex 24d methyl (1R,2R,4R) 2-acetamido-5-norbornen-2-carboxylate is preferably obtained. Thus, the reaction between methyl N-acetyl-(,β-didehydroalaninate and cyclopentadiene is a good method for the synthesis of (1S, 2R, 4R) 2-aminonorbomane-2-carboxylic acid.  相似文献

An organocatalyst formed from a binaphthyl‐substituted diamine and trifluoromethanesulfonic acid exhibited unprecedented levels of exo selectivity in the Diels–Alder reaction of α,β‐unsaturated aldehydes with cyclopentadiene. A novel axially chiral diamine was also designed as an organocatalyst for an asymmetric variant of this reaction, in which the desired cycloadducts were formed with high diastereo‐ and enantioselectivities. The highest diastereoselectivity observed was greater than 20:1 in favor of the exo cycloadduct in the asymmetric Diels–Alder reaction of crotonaldehyde with cyclopentadiene.  相似文献

The rare earth(III) salt catalysed asymmetric Diels–Alder reaction of cyclopentadiene with a chiral dienophile in supercritical carbon dioxide (scCO2) proceeded rapidly to give the adduct with a higher diastereoselectivity than that in dichloromethane; Yb(ClO4)3 gave the endo adduct with value up to 77% de at 40°C, 8 MPa. The chiral rare earth diketonate catalyzed hetero Diels–Alder reaction of the Danishefsky's diene with benzaldehyde gave a higher yield and an enantioselectivity in scCO2 than that in dichloromethane. Scandium/pybox 8a complex catalysed asymmetric Diels–Alder reaction of 3-crotonoyl-2-oxazolidinone with cyclopentadiene in the presence of MS4A proceeded smoothly in scCO2 to give the endo adduct 10 in a good yield with up to 88% ee.  相似文献

The review discusses available data on asymmetric Diels-Alder reactions of cyclopentadiene, which were published in the past decade. Both noncatalytic and catalytic (in the presence of achiral and chiral catalysts) versions of these reactions are considered. Effects of various factors on the chemical and optical yields, stereoselectivity, and optical purity of the Diels-Alder adducts are analyzed. The complex [Ru(Cp)(R,R-BIPHOP-F)(acetone)][SbF(6)], (R,R)-1 a, was used as catalyst for asymmetric Diels-Alder reactions between dienes (cyclopentadiene, methylcyclopentadiene, isoprene, 2,3-dimethylbutadiene) and alpha,beta-unsaturated ketones (methyl vinyl ketone (MVK), ethyl vinyl ketone, divinyl ketone, alpha-bromovinyl methyl ketone and alpha-chlorovinyl methyl ketone). The cycloaddition products were obtained in yields of 50-90 % and with enantioselectivities up to 96 % ee. Ethyl vinyl ketone, divinyl ketone and the halogenated vinyl ketones worked best and their reactions with acyclic dienes consistently provided products with >90 % ee. alpha-Chlorovinyl methyl ketone performed better than alpha-bromovinyl methyl ketone. The reaction also provided a [4.3.1]bicyclic ring system in 95 % ee through an intramolecular cycloaddition reaction. Crystal structure determinations of [Ru(Cp)((S,S)-BIPHOP-F)(mvk)][SbF(6)], (S,S)-1 b, and [Ru(Cp)((R,R)-Me(4)BIPHOP-F)(acrolein)][SbF(6)], (R,R)-2 b, provided the basis for a rationalization of the asymmetric induction.  相似文献

A two-step general approach to a new chiral auxiliary starting from levoglucosenone is reported. The compound is obtained by a [4+2] cycloaddition reaction with anthracene followed by a diastereoselective reduction of the C-2 keto function in high overall yield. The auxiliary has been used as chiral template in an asymmetric Diels-Alder reaction of the corresponding acrylic ester derivative with cyclopentadiene and shown to be efficient for asymmetric induction.  相似文献

A new Ti(IV) complex obtained from the C2-symmetric amino diol (1R,5R)-3-aza-3-benzyl-1,5-diphenyl pentan-1,5-diol, (1R,5R)-1, is used effectively as a Lewis acid promoter in asymmetric Diels–Alder reactions. Using various Evans' oxazolidinones as dienophiles and cyclopentadiene as the diene high yields of the adducts with moderate enantioselectivity, under different reaction conditions are achieved. The effects of solvent, temperature and ligand on the enantioselectivity of the Diels–Alder products are reported. Molecular modelling studies provide an understanding of the diastereofacial selectivity of the Diels–Alder reactions. Asymmetric carbonyl-ene reactions between various glyoxylate esters and α-methyl styrene are also described.  相似文献

A B3LYP/6-31G* study has been carried out for the reactions of methacrolein with cyclopentadiene, parent nitrone, 1-pyrroline-1-oxide, and (Z)-C,N-diphenylnitrone, in which the coordination of a Lewis acid (borane) and the solvent polarity (dichloromethane) have been taken into account. Calculated activation parameters, regioselectivities (for 1,3-dipolar cycloaddition reactions), and endo/exo stereoselectivities show good agreement with available experimental data. Gas-phase calculations show a varied behavior of the s-cis/s-trans TS stability for noncatalyzed reactions (from the systematic s-cis preference for the cyclopentadiene reaction to the systematic s-trans predilection encountered in the diphenylnitrone cycloaddition). BH3 coordination leads to a preferential stabilization of s-trans TSs in the reactions of cyclopentadiene (exo approach) and diphenylnitrone but a larger stabilization of s-cis structures in the processes involving the parent nitrone or 1-pyrroline-1-oxide. Additionally, a rather systematic preferential stabilization of s-trans structures is induced by solvent polarity in most reactions. As a consequence, an s-trans preference is predicted in solution for both thermal and catalyzed types of reactions in most approaches. Such a conclusion is consistent with some experimental results suggesting a preference for a particular conformation of the methacrolein-Lewis acid complexes.  相似文献

The synthesis of a chiral alcohol derived from levoglucosenone has been studied. The alcohol was employed as chiral template in an asymmetric Diels-Alder reaction of the corresponding acrylic ester derivative with cyclopentadiene, and was shown to be an efficient asymmetric inductor. The oxidation reaction detected during the hydrogenation of a secondary allylic alcohol intermediate with Pd/C was also investigated.  相似文献

Thus, in the presence of (S)‐TMS‐Segphos, the reactions took place under very mild conditions to afford metallocene‐fused siloles in good to excellent yields and with ee values of up to 97 %. During this study it was observed that the steric hindrance of chiral ligands had a profound influence on the reactivity and enantioselectivity of the reaction, and might hold the key to accomplishing conventionally challenging asymmetric C?
