Malaria is a serious disease caused by Plasmodium parasites that infect red blood cells (RBCs). This paper presents the continuous separation of malaria-infected RBCs (iRBCs) from normal blood cells. The proposed method employed the discrete dielectrophoresis (DEP) in a microfluidic device with interdigitated electrodes. Our aim is to treat a sample having high concentration of cells to realize high throughput and to prevent the clogging of the microchannel with the use of the discrete DEP. The discrete DEP force for deflecting cells in the device was controlled by adjusting the magnitude, frequency, and duty cycle of the applied voltage. The effectiveness of the proposed method was demonstrated by separating the malaria-infected cells in samples having a cell concentration of 106 cells/µl. From experimental results, we determined the enrichment that is needed to enhance the detection in the case of low parasitemia. The enrichment of the infected cells at the device output was 3000 times as high as that of the input containing 1 infected cell to 106 normal cells. Therefore, the proposed method is highly effective and can significantly facilitate the detection of the infected cells for the identification of Malaria patients.  相似文献

We describe the design, microfabrication, and testing of a microfluidic device for the separation of cancer cells based on dielectrophoresis. Cancer cells, specifically green fluorescent protein‐labeled MDA‐MB‐231, are successfully separated from a heterogeneous mixture of the same and normal blood cells. MDA‐MB‐231 cancer cells are separated with an accuracy that enables precise detection and counting of circulating tumor cells present among normal blood cells. The separation is performed using a set of planar interdigitated transducer electrodes that are deposited on the surface of a glass wafer and slightly protrude into the separation microchannel at one side. The device includes two parts, namely, a glass wafer and polydimethylsiloxane element. The device is fabricated using standard microfabrication techniques. All experiments are conducted with low conductivity sucrose‐dextrose isotonic medium. The variation in response between MDA‐MB‐231 cancer cells and normal cells to a certain band of alternating‐current frequencies is used for continuous separation of cells. The fabrication of the microfluidic device, preparation of cells and medium, and flow conditions are detailed. The proposed microdevice can be used to detect and separate malignant cells from heterogeneous mixture of cells for the purpose of early screening for cancer.  相似文献

We developed a low-cost polymer-film spiral inertial microfluidic device for the effective size-dependent separation of malignant tumor cells. The device was fabricated in polymer films by rapid laser cutting and chemical bonding. After fabricating the prototype device, the separation performance of our device was evaluated using particles and cells. The effects of operational flow rate, cell diameter, and cell concentration on the separation performance were explored. Our device successfully separated tumor cells from polydisperse white blood cells according to their different migration modes and lateral positions. Then, the separation of rare cells was carried out using the high-concentration lysed blood spiked with 200 tumor cells. Experimental results showed that 83.90% of the tumor cells could be recovered, while 99.87% of white blood cells could be removed. We successfully employed our device for processing clinical pleural effusion samples from patients with advanced metastatic breast cancer. Malignant tumor cells with an average purity of 2.37% could be effectively enriched, improving downstream diagnostic accuracy. Our device offers the advantages of label-free operation, low cost, and fast fabrication, thus being a potential tool for effective cell separation.  相似文献

A novel alcohol dehydration membrane with a three layer structure has been prepared. The top layer is a thin dense film of chitosan (CS), and the support layer is made of microporous polyacrylonitrile (PAN). Between the dense and microporous layer, there is an intermolecular cross-linking layer. This novel composite membrane has a high separation factor of more than 8000 and a good permeation rate of 0.26 kg/m2 h for the pervaporation of 90 wt% ethanol aqueous solution at 60°C, 0.8 kg/m2 h flux for a n-PrOH/water system and around 1 kg/m2 h flux for an i-PrOH/water system using 80 wt% alcohol concentration at 60°C. The separation factor for both cases is more than 105. The separation performance varies with feed composition, operating temperature and conditions of membrane preparation. The results show that the separation factor and flux of this membrane increase with raising the operating temperature. At the same time, the crosslinking layer improves durability of the composite membrane, and the pervaporation performance can be adjusted by changing the structure of the cross-linking layer. The cross section of the composite membrane has been examined by SEM.  相似文献

A microfluidic device for continuous, real time blood plasma separation is introduced. The principle of the blood plasma separation from blood cells is supported by the Zweifach-Fung effect and was experimentally demonstrated using simple microchannels. The blood plasma separation device is composed of a blood inlet, a bifurcating region which leads to a purified plasma outlet, and a concentrated blood cell outlet. It was designed to separate blood plasma from an initial blood sample of up to 45% inlet hematocrit (volume percentage of cells). The microfluidic network was designed using an analogous electrical circuit, as well as analytical and numerical studies. The functionality of this device was demonstrated using defibrinated sheep blood. During 30 minutes of continuous blood infusion through the device, all the erythrocytes (red blood cells) traveled through the device toward the concentrated blood outlet while only the plasma was separated at the bifurcating regions and flowed towards the plasma outlet. The device has been operated continuously without any clogging or hemolysis of cells. The experimentally determined plasma selectivity with respect to blood hematocrit level was almost 100% regardless of the inlet hematocrit. The total plasma separation volume percent varied from 15% to 25% with increasing inlet hematocrit. Due to the device's simple structure and control mechanism, this microdevice is expected to be used for highly efficient continuous, real time cell-free blood plasma separation from blood samples for use in lab on a chip applications.  相似文献

A miniaturized multichamber device was constructed for solution isoelectric focusing (IEF) separation of complex peptide mixtures. The system, based on immobilized pH gels, consisted of 96 minichambers ( approximately 75 nuL each) arranged in eight rows. Neighboring chambers in a given row were separated by short glass tubes (4 mm inner diameter, 3 mm long), within which Immobiline gels of specific pH values were polymerized. During focusing, the device was sandwiched between two supporting blocks incorporating the reservoirs for anolyte and catholyte. In principle, multiple samples could be simultaneously fractionated, each separated into 12 fractions of various pI ranges. A variety of standard peptide mixtures and tryptic digests of proteins were separated by IEF using this device, and the fractions were characterized by mass spectrometry. For a codigested nine-protein mixture, both the total number of peptides identified and the average sequence coverage were similar to the results of ion-exchange chromatography (IEC), according to matrix assisted laser/desorption/ionization--time of flight (MALDI-TOF) data. The IEF separation provided concentrated and desalted fractions, suitable for an additional separation liquid chromatography, capillary electrophoresis (LC, CE) or mass spectrometry (MS) detection without additional sample cleanup. High loading capacity was achieved for the miniaturized multichamber IEF device. Importantly, a linear correlation was found between the experimentally determined and calculated pI values of peptides.  相似文献

A new technique for the separation of99mTc from low specific activity99Mo is reported. A separation based on the principle of precipitation of99Mo as calcium molybdate has been investigated. On precipitating99MoO 4 2– from alkaline solution as calcium molybdate under controlled conditions, the99mTcO 4 is found to remain quantitatively in the supernatant solution with little carry-over of99Mo. This calcium molybdate (99Mo) could be redissolved and reprecipitated at regular intervals, yielding99mTc quantitatively in aqueous neutral solutions. Calcium molybdate precipitates containing up to 1.5 GBq of99Mo and 130–180 mg of molybdenum were prepared and evaluated. The performance in terms of repeated99mTc separation gave yields of 75–93% with acceptable readionuclidic and radiochemical purity.  相似文献

A monolithic column was prepared using l-phenylalanine as template and a covalent approach through the formation of Schiff base with o-phthalaldehyde (OPA). OPA, allylmercaptan, l-phenylalanine, and triethylamine were stirred at first, then methacrylic acid, 2-vinylpyridine, ethyleneglycol dimethacrylate, α,α-azobisisobutyronitrile, and 1-propanol were added to the reaction mixture. The resulting material was introduced into a capillary column. Following thermal polymerization, the template was then extracted with a mixture of HCl and methanol. The column was employed for the capillary electrochromatographic separation of oligopeptides. A capillary column of 75 (50) cm × 75 μm ID with a mobile phase of phosphate buffer (pH 7.0, 40 mM)/methanol (5%, v/v), an applied voltage of +15 kV, and detection at 214 nm, could baseline separate angiotensin I, angiotensin II, [Sar1, Thr8] angiotensin, oxytocin, vasopressin, tocinoic acid, β-casomorphin bovine, β-casomorphin human, and FMRF amide within 20 min. The separation behavior of the templated polymer was also compared with that of the non-templated polymer. As a result, it can be concluded that the electrochromatographic separation of this set of peptides was mediated by a combination of electrophoretic migration and chromatographic retention involving hydrophobic, hydrogen bonding, electrostatic as well as the Schiff base formation with OPA in the cavity of the templated polymer.  相似文献

A novel straightforward approach to selective separation for flavonoid compounds was reported. The solid phase material was prepared by copolymerization using allyl-bromide-modified chitosan as macromonomer, and ethylene glycol dimethacrylate as cross-linker. The material was evaluated by chromatographic analysis; it exhibited high selectivity separation for quercetin and its structural analogues using different mobile phases. The material could directly trap a specific class of compounds including quercetin and kaempferol from the hydrolyzate of Ginkgo biloba extract. These results demonstrated the possibility of direct extraction of certain constituents from herb using this material.  相似文献

A novel fiber optical device for ultraviolet disinfection of water   总被引:2,自引:0,他引:2  
Since there are several problems in traditional UV disinfection techniques, a highly efficient, reliable and economical method, using quartz optical fibers to deliver UV light is proposed. The principle of the experimental setup is that ultraviolet rays are gathered by a reflector and converge on a light point, the diameter of approximately 5mm. In this way UV light can be transferred into water to kill the bacteria in the water. This paper presents preliminary results on water disinfection using this new UV disinfection setup. Its suitability for application could be shown in experiments with E. coli (ATCC8099) as test microorganisms. We have optimized the distribution of the optical fibers in the water in bench-scale study. This result can provide guidance for pilot-scale and field-scale study of this new technique. The results show that the new technique had a good performance under different conditions as follows: (a) turbidity level=10.2 NTU, (b) ferric ion concentration=0.3 mg/L, and (c) humic acid concentration=5 mg/L. The new technique provides a promising approach to disinfection treatment of drinking water.  相似文献

We report here a low-cost, rapid-prototyping, and beehive-like multilayer polymer microfluidic device for ultrahigh-throughput blood plasma separation. To understand the device physics and optimize the device structure, the effect of cross-sectional dimension and operational parameter on particle focusing behavior was explored using a single spiral microchannel device. Then, the blood plasma separation performance of the determined channel structure was validated using the blood samples with different hematocrits (HCTs). It was found that a high separation efficiency of 99% could be achieved using the blood sample with an HCT of 0.5% at a high throughput of 1 mL/min. Finally, a multilayer microfluidic device with a novel beehive-like multiplexing channel arrangement was developed for ultrahigh-throughput blood plasma separation. The prototype device could be fabricated within ∼1 hour utilizing the laser cutting and thermal lamination methods. The total processing throughput could reach up to 72 mL/min for 0.5% HCT sample with a plasma separation ratio close to 90%. Our device may hold potentials for the ultrahigh-throughput separation of blood plasma from large volume blood samples for downstream disease diagnosis.  相似文献

A sample dispenser with electronic control of delivered volume has been designed and evaluated. With this device, liquid samples are dispensed in the form of uniform droplets less than 4 nl in volume; total volumes as small as 40 nl can be delivered with 1.5% precision. Important features of the microdroplet formation process have been studied and the effect of sample solution concentration and viscosity have been examined. The dispenser is expected to find application in several areas, including electrothermal atomization atomic absorption spectrometry, automated titrimetry, and high-precision, low-volume solution sampling.  相似文献

Herein we present a novel way to create arrays of different proteins or lipid vesicles using a crossed microfluidic device. The concept relies on the combination of I) a designated two-step surface chemistry, which allows activation for subsequent binding events, and II) crossing microfluidic channels for the local functionalization by separated laminar streams. Besides its simplicity and cost efficiency, this concept has the advantage that it keeps the proteins in a hydrated environment throughout the experiment. We have demonstrated the feasibility of such a device to create a chessboard pattern of different fluorescently labeled lipid vesicles, which offers the possibility to contain biomolecules, drugs or membrane proteins.  相似文献

This work assesses the use of modified natural clinoptilolite as an adsorptive material for separation and preconcentration of trace amounts of zirconium ions. A simple, rapid and economical method was developed for the preconcentration of trace amounts of zirconium in aqueous medium using 1-(2-pyridylazo)-2-naphthol as a complexing agent. Effect of sample pH, flow rate of sample and elution solutions, breakthrough volume and interference of several ions were studied. Determination of zirconium was made by ICP-AES technique. The sorption was quantitative in the pH range from 3.0 to 4.0, whereas quantitative desorption occurred instantaneously with 2 mol L?1 hydrochloric acid. Linearity was maintained between 0.05 and 9.0 μg mL?1. Relative standard deviations range from ±0.9% to ±2.3% (n?=?5). The detection limit was 0.1 ng mL?1. Because of good recovery (>97%), this method is suitable for preconcentration and determination of zirconium in effluents containing trace amount of zirconium.  相似文献

Two new types of potential liquid‐crystalline azo compounds were synthesized in the form of racemic mixtures and as the individual S enantiomers. Both materials consisting of two substituted aromatic rings in the molecular core and one chiral center at the aliphatic moiety showed mesomorphic behavior. The separation of the R and S enantiomers by chiral high‐performance liquid chromatography was unsuccessful when the azo compounds were in their natural E state. However, the irradiation of the compounds in the solution by UV light led to an almost quantitative transition to their Z forms. The chromatographic behavior of the compounds in their Z forms was significantly different, and partial separation of the individual enantiomers on chiral polysaccharide‐based stationary phases was obtained. Thus, the proposed procedure represents a novel approach to the enantioseparation of chiral diazenes.  相似文献

The simplified amperometric detection scheme demonstrated is based on the amperometric working and electrophoretic ground electrodes only. The latter serves as counter and pseudo-reference as well. It is shown via the successful determination of neurotransmitters, ascorbic acid and phenols on gold or platinum working electrodes that this approach is feasible for detection on a channel based electrophoretic separation device. Also presented is the detection of carbohydrates and amino acids with copper electrodes. The results were found to be similar to those obtained with conventional capillary systems with amperometric detection, albeit at much reduced analysis times.  相似文献

The purpose of this study was to extract impurities from compounds using a simple separatory bottle to purify target compounds with a foam column and allow for the further characterization of impurities. Charged dyes were used as target compounds due to the ease of detection of dyes and isolated impurities. Foaming agents were used in a glass bottle with a modified cap to separate a target impurity using an appropriately charged ligand. By passing N2 gas through the solution, the surfactants sodium dodecyl sulfate and cetylpyridinium chloride generated foams that separated the dyes, Methylene blue and Orange G, respectively, from a solution containing both dyes. Sodium dodecyl sulfate condensed Methylene blue from the solution with high purity while cetylpyridinium chloride condensed Orange G with less purity. A range of concentrations (0.01–0.5 mmol/L) of dyes were used for separation. The condensability (volume and/or concentration) of the target compound increased as its concentration decreased. This novel separation method is a simple, rapid, inexpensive, and effective way to prepare samples and allows for the characterization of these impurities using sensitive analytical detection techniques.  相似文献

This paper presents theoretical analysis, design, simulation, fabrication and test of a microfluidic device ('Micro-hydrocyclone') for separation of micron and submicron size solid particles from liquid in a particle liquid mixture. A theoretical analysis of the micro-hydrocyclone is performed to understand the physics and develop suitable design models. The structure of the proposed device is designed based on the Bradley model, as it offers lower cut-size thus making it suitable for microfluidics applications. The operational parameters are derived from the dimensional group model. The particle separation process inside the micro-hydrocyclone is simulated by solving fluid flows using Navier-Stokes equations and particle dynamics using a Lagrangian approach in a Eulerian fluid. The influence of inlet velocity and density on separation efficiency is investigated. The device is fabricated with SU-8 photoresist on a PMMA substrate using a combination of photolithography and micro-milling. Experiments are performed to demonstrate particle-liquid separation using polystyrene microbeads suspended in PBS as the feed sample. The influence of inlet velocity and particle size on particle separation efficiency is measured and compared with that obtained from simulations and a good match was found. The proposed device can be easily integrated with micro-environments thus it is suitable for lab-on-chip and microsystems development. The device may have applications in chemical analysis, materials research, point-of-care, blood sample preparation and other biomedical applications.  相似文献
