One of the major disadvantages of the current cancer therapy is the suppression of the immune system. Brazilian flora is considered one of the most diverse in the world and many plants were found to contain active constituents that can be valuable sources of new drugs. The plant Indigofera suffruticosa was studied to determine its potential to stimulate the immune system and also to be effective against tumour cells. We investigated the effects of the alkaloidal fraction and the pure alkaloid indigo obtained from I. suffruticosa on macrophage activation by measuring nitric oxide (NO) and tumour necrosis factor-α (TNF-α) production. Cytotoxic activity was also evaluated against the two tumour murine cells lines, LM2 (breast adenocarcinoma) and LP07 (lung adenocarcinoma). The alkaloidal fraction induced a high NO production and a moderated TNF-α release. The pure indigo demonstrated an elevated NO and TNF-α production. The fraction and the pure compound also exhibited cytotoxic activity against both adenocarcinoma cell lines and indigo showed the strongest cytotoxic activity with IC?? value of 0.89?μg mL?1 against LM2 and 1.44?μg?mL?1 against LP07. Our results presented the immunostimulatory and cytotoxic activity of I. suffruticosa, enhancing macrophage function and therefore contributing to the host defence against tumours.  相似文献

This study investigates the in vitro antioxidant and antimicrobial activities of eight extracts obtained from the dried barks of Commiphora berryi and Commiphora caudata (Burseraceae). The radical scavenging activity was assessed by 1,1-diphenyl-2-picryl hydrazyl (DPPH) and nitric oxide assays. The methanolic extracts of C. berryi and C. caudata showed significant DPPH radical scavenging activity, with IC?? values of 26.92 and 21.16?μg?mL?1, respectively, and low radical scavenging activity against the nitric oxide assay. The antimicrobial activity of the plants was tested against the Gram-positive and Gram-negative bacteria. The ethyl acetate, chloroform and petroleum ether extracts of C. berryi showed good antibacterial activity against Pseudomonas aeruginosa, with a minimum inhibitory concentration (MIC) of 0.26?mg?mL?1, whereas the ethyl acetate and methanol extracts of C. caudata showed moderate antimicrobial activity with an MIC of more than 2.0?mg?mL?1 against P. aeruginosa compared to the petroleum ether and chloroform extracts, which showed an MIC of 1.1?mg?mL?1. The methanolic extracts of C. berryi and C. caudata also showed moderate cytotoxic activity against a human mammary carcinoma cell line (MCF-7), with values IC?? of 82.6 and 88.4?μg?mL?1, respectively.  相似文献

Maesilea quadrifolia Linn. (Marsileaceae) is a leafy vegetable well known in India. The current study aims to explore the phytochemical profile of M. quadrifolia and investigate its anti-cholinesterase potential. The methanol extract of the plant was subjected to qualitative and quantitative phytochemical screening (total alkaloidal content, saponin content and phenol content) and its anti-cholinesterase potential was tested by TLC bioautography and other screening methods using acytylcholinesterase (AChE) and butyrylcholinesterase (BChE). The study revealed that the extract contains various classes of phytoconstituents including steroids, saponins, alkaloids and other polyphenols. Total alkaloid, phenolic and saponin contents were found to be 19.3?mg?g?1 and 158.5?±?1.02?mg?g?1 as gallic acid equivalents and 2.63?mg?g?1 of the extract, respectively. The TLC bioautography method exhibited the inhibition of both enzymes. In a microtiter plate assay, the IC?? values of the extract for AChE and BChE were found to be 51.89?±?0.24?μg?mL?1 and 109.43?±?2.82?μg?mL?1, respectively. These findings suggest that M. quadrifolia is a potential lead as an AChE and BChE inhibitor, which may be useful in the management of Alzheimer's disease.  相似文献

Two new norditerpenoid alkaloids, named scutebarbatines M-N (1 and 2), were isolated from the whole plant of Scutellaria barbata D. Don. Their structures were established on the basis of detailed spectral analyses. In vitro, two new compounds showed significant cytotoxic activities against three human cancer lines (HONE-1 nasopharyngeal, KB oral epidermoid carcinoma and HT29 colorectal carcinoma cells), and gave IC?? values in the range of 3.5-6.3?μM.  相似文献

A wide range of proteinaceous inhibitors are present in plants to protect themselves from hydrolytic enzymes. In this study, turmerin, a water-soluble peptide in turmeric rhizomes, was evaluated for its inhibitory potential against glucosidase and its antioxidant (AO) capacity. Turmerin inhibited α-amylase and α-glucosidase activities with IC?? values 31 and 192?μg?mL?1, respectively. Under the experimental conditions, those values for a standard glucosidase inhibitor, acarbose, were 81 and 296?μg?mL?1, respectively. The AO capacity of turmerin was evaluated using in vitro assay systems. Turmerin showed good DPPH (IC???=?29?μg?mL?1) and superoxide (IC???=?48?μg?mL?1) and moderate ABTS (IC???=?83?μg?mL?1) radical scavenging and Fe(II) chelation (IC???=?101?μg?mL?1) capacities. The inhibitory potential showed by turmerin against enzymes linked to type 2 diabetes, as well as its moderate AO capacity, could rationalise the traditional usage of turmeric rhizome preparations against diabetes.  相似文献

Aizoaceae is a large succulent family characterized by many psychoactive species. Aizoon canariense L., a wild neglected plant traditionally used in gastrointestinal ailments, has been the subject of a limited number of phytochemical and biological studies. Therefore, herein, we investigated the in vitro cytotoxic, antimicrobial, and anticholinesteraseactivity of the aerial parts of A. canariense L. and analyzed the phytochemical compositions of the lipoidal and alkaloidal fractions. Petroleum ether extract showed the presence of behenic and tricosylic acid, while an in-depth investigation of the alkaloidal fraction revealed the identification of new adenine based alkaloids (1–5), which were isolated and identified for the first time from Aizoon canariense L. Their structures were elucidated based on extensive spectroscopic analyses. The alkaloidal extract showed a powerful cytotoxic effect (IC50 14–28 μg/mL), with the best effect against colon carcinoma, followed by liver and breast carcinomas. The alkaloidal extract also had a potent effect against Candida albicans and Escherichia coli, with minimum inhibitory concentrations (MIC) values of 312.5 and 625 µg/mL. The in vitro anticholinesterase activity was potent, with IC50 < 200 ng/mL for the tested extracts compared with 27.29 ± 0.49 ng/mL for tacrine.  相似文献

The methanol and fractionated extracts (hexane, ethyl acetate and water) of Alpinia mutica (Zingiberaceae) rhizomes were investigated for their cytotoxic effect against six human carcinoma cell lines, namely KB, MCF7, A549, Caski, HCT116, HT29 and non-human fibroblast cell line (MRC 5) using an in vitro cytotoxicity assay. The ethyl acetate extract possessed high inhibitory effect against KB, MCF7 and Caski cells (IC?? values of 9.4, 19.7 and 19.8 μg/mL, respectively). Flavokawin B (1), 5,6-dehydrokawain (2), pinostrobin chalcone (3) and alpinetin (4), isolated from the active ethyl acetate extract were also evaluated for their cytotoxic activity. Of these, pinostrobin chalcone (3) and alpinetin (4) were isolated from this plant for the first time. Pinostrobin chalcone (3) displayed very remarkable cytotoxic activity against the tested human cancer cells, such as KB, MCF7 and Caski cells (IC?? values of 6.2, 7.3 and 7.7 μg/mL, respectively). The antioxidant and antiproliferative activities of the essential oils from Laurus nobilis leaves and seeds in relation to their composition were analysed. The most abundant components of the leaf essential oil were 1,8-cineole, 1-p-menthen-8-ethyl acetate, linalool and sabinene, while the seed oil was characterised by β-ocimene, 1,8-cineole, α-pinene and β-pinene as main constituents. Both seed and leaf essential oils exhibited a scavenging effect on the DPPH radical, with IC?? values of 66.1 and 53.5?μg?mL?1, respectively. The leaf essential oil showed the strongest antioxidant activity in the β-carotene/linoleic acid system, with an IC?? value of 35.6?μg?mL?1 after 30?min of incubation. Both leaf and seed oils inhibited proliferation of the K562 tumour cell line with IC?? values of 95 and 75?μg?mL?1, respectively. The L. nobilis leaf oil showed a percentage of erythroide differentiation of 15% at a concentration of 10?μg?mL?1. A value of 12% was found for the seed essential oil at a concentration of 50?μg?mL?1. When the oils were added to a suboptimal concentration of the commercial drug, cytosine arabinoside, a clear synergic effect was observed.  相似文献

The antiproliferative activities of six medicinal plant extracts from Burkina Faso were evaluated in order to justify their traditional use for the treatment of cancer. The SOS chromotest method was used in vitro on Escherichia coli PQ37 to evaluate the mutagenic effect of the plant extracts. The DPPH method was used to evaluate the antioxidant activity of each plant. The antiproliferative activity was evaluated by MTS method on normal cells (Vero and MCR5) and cancer cells (KB) in contact with the extracts for 72?h. The results showed that the studied plants are not genotoxic. Lantana ukambensis and Acacia macrostachya induced a very significant antiproliferative effect against cancer cells with 94% and 95%, respectively. They also developed a strong antioxidant activity. The IC?? values were 5.96?±?0.40?μg?mL?1 for L. ukambensis and 4.30?±?0.26?μg?mL?1 for A. macrostachya. These two plants are therefore potential sources for isolating new antioxidant and anticancer molecules.  相似文献

The petroleum ether and chloroform extracts of the root of Piper chaba showed antimicrobial, antileishmanial and cytotoxic activities. Further bioactivity-guided fractionation led to the isolation of Bornyl piperate (1), piperlonguminine (2) and piperine (3). This is the first report of isolation of compounds (1) and (2) from P. chaba. It was observed that the isolated compounds (1 and 2) showed potent antifungal activity when compared with standard drug Nystatin, and significant cytotoxic activity with the IC?? values of 0.76 and 0.83?μg?mL?1, respectively. These compounds were also found to have weak antibacterial and antileishmanial activities. This is the first report about the antileishmanial activity of Piper isolates.  相似文献

The methanolic extract of Verbascum thapsus was evaluated for its antiviral activity against the pseudorabies virus strain RC/79 (PrV), and also for its cytotoxic activity on Vero cells. The extract showed CC?? values of 1100?μg?mL?1 and 1426?μg?mL?1 by NRU and MTT assays, respectively. The 50% inhibitory concentration of the extract for PrV plaque formation was determined at 35?μg?mL?1, and selectivity indices were 31.4 (NRU) and 40.7 (MTT). When cells were pre-treated with the extract prior to virus infection, the inhibition in plaque formation was 70%. PrV was highly inhibited when it was incubated with plant extract or when the extract was added during the adsorption phase (99%). However, no inhibitory effect was observed when the extract was added to the cells after the adsorption period. Thus, these results suggest that the methanolic extract of Verbascum thapsus may contain bioactive compound(s) that affect PrV mostly in the adsorption phase.  相似文献

This study investigates the effect of Artocarpus altilis leaf extracts on angiotensin-converting enzyme (ACE) activity. Among the extracts tested, hot ethanol extract exhibited a potent ACE-inhibitory activity with an IC?? value of 54.08?±?0.29?μg?mL?1 followed by cold ethyl acetate extract (IC?? of 85.44?±?0.85?μg?mL?1). In contrast, the hot aqueous extracts showed minimum inhibition with the IC?? value of 765.52?±?11.97?μg?mL?1 at the maximum concentration tested. Further, the phytochemical analysis indicated the varied distribution of tannins, phenolics, glycosides, saponins, steroids, terpenoids and anthraquinones in cold and hot leaf extracts. The correlation between the phytochemical analysis and ACE-inhibitory activity suggests that the high content of glycosidic and phenolic compounds could be involved in exerting ACE-inhibitory activity. In conclusion, this study supports the utilisation of A. altilis leaf in the folk medicine for the better treatment of hypertension. Further studies on isolation and characterisation of specific ACE-inhibitory molecule(s) from ethyl acetate, ethanol and methanol extracts of A. altilis leaf would be highly interesting.  相似文献

Artemisia sacrorum Ledeb. was extracted by 95% ethanol and water, respectively. By partitioning the 95% ethanol extract successively with different solvents and separating the water extract by macroporous resin, nine separate parts were obtained. According to the results of in vitro experiments, the CH?Cl? (dichloromethane) fraction showed the most pronounced cytotoxic activity against HepG2, HT-29 and MCF-7 cells, with EC?? values 122.35, 49.76 and 28.51?μg?mL?1, respectively, at 48?h. Following this, the compounds of the CH?Cl? fraction were separated and identified. Ten compounds were isolated from A. sacrorum Ledeb. and identified by spectral analysis. Antioxidant activity of several classes of lichen metabolites were assessed in the in vitro superoxide radical (SOR), nitric oxide radical and 2,2-diphenyl-1-picrylhydrazil radical scavenging assays. The despsides sekikaic acid and lecanoric acid showed promising antioxidant activity in SOR assay with IC?? values of 82.0?±?0.3?μmol and 91.5?±?2.1?μmol, respectively, while the depsidone lobaric acid exhibited an IC?? value of 97.9?±?1.6?μmol, all relative to the standard, propyl gallate (IC???=?106.0?±?1.7?μmol). The chemical composition of the essential oil of Citrus medica L. cv. Diamante peel obtained by hydrodistillation, cold-pressing and supercritical carbon dioxide extraction techniques was determined by GC/MS analysis. Forty-six components were fully characterised. Limonene and γ-terpinene were the major components of the oils obtained by hydrodistillation (HD) and cold-pressing (CP), while citropten was the major constituent in the oil obtained by supercritical carbon dioxide extraction (SFE). Acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) inhibitory activities were evaluated. The essential oil obtained by hydrodistillation exerted the highest inhibitory activity against BChE (IC?? value of 154.6 μg mL?1) and AChE (IC?? value of 171.3 μg mL?1. Interestingly, the oil obtained by cold-pressing exhibited a selective inhibitory activity against AChE. The essential oils have also been evaluated for the inhibition of NO production in LPS induced RAW 264.7 macrophages. The oil obtained by hydrodistillation exerted a significant inhibition of NO production with an IC?? value of 17 μg mL?1 (IC?? of positive control 53 μg mL?1).  相似文献

A series of novel 1,3,4-oxadiazole derivatives based on benzisoselenazolone has been prepared and tested for antiproliferative activity in vitro against the cells of human cancer cell lines: SSMC-7721 (human liver cancer cell), MCF-7 (human breast cancer cell) and A549 (human lung cancer cell). All the compounds obtained exhibited antiproliferative activity and showed selective cytotoxicity against different cancer cells. Compounds 7d and 7i showed significant antiproliferative activities against MCF-7 cells, with IC50 values of 1.07 and 1.76?μM respectively. Compound 7d were found to be the most potent compound against SSMC-7721 cells, with IC50 values 4.46?μM.  相似文献

The phytochemical investigation of an alkaloidal extract of Holarrhena pubescens roots led to the isolation and identification of a new pregnene-type alkaloid, mokluangin D (1), together with nine known steroidal alkaloids (2–10). The structure of the new metabolite was determined on the basis of spectroscopic analyses including 1D- and 2D-NMR spectroscopy and mass spectrometry. Compounds 3 and 4 showed potent antimalarial activity against Plasmodium falciparum K1 stain with IC50 values of 1.2 and 2.0 μM, respectively, and showed weak cytotoxic activity against the NCI-H187 cell line with IC50 values of 27.7 and 30.6 μM, respectively. The substituent groups at C-3 and the carbonyl group at C-18 are important for the activity against the P. falciparum K1 stain.  相似文献

2,9-Dimethoxymethylboldine (2), 2,9-dimethoxymethyl-3-bromoboldine (3) and 2,9-dimethoxymethyl-3-diphenylphosphinylboldine (4) have been synthesized in an effort to find compounds with potential pharmacological applications. The cytotoxic activities of the natural precursor 1 and these three derivatives have been measured as IC?? inhibitory growth. The diphenylphosphinyl derivative 4 showed a significant cytotoxic activity on two breast cancer cell lines, namely MCF-7 and MDA-MB-231, with IC?? values of 55.5 and 62.7 [μM], respectively. These results suggest that the kind of structural modifications introduced to synthesize compound 4 represent a promising way to enhance the cytotoxic activity of boldine derivatives.  相似文献

Diospyros peregrina is an edible seasonal fruit found in coastal West Bengal, India. The fruits have been reported to possess a significant antioxidant activity. In this study, the aim was to isolate the lead compound responsible for the above-mentioned activity. The aqueous extract of D. peregrina fruit was subjected to dereplication coupled with an in?vitro 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging assay. The n-butanol fraction of the aqueous extract of D. peregrina fruit exhibited significant antioxidant activity (IC(50), 131.10?μg?mL(-1)) as compared with the parent extract (IC(50), 285.15?μg?mL(-1)). The n-butanol fraction was subjected to silica gel column chromatographic separation coupled with a chemo-autographic study of column eluents, employing ethanolic DPPH as a spraying reagent. Two bioactive flavonoid glycosides, namely luteoline-4'-methyl-ether-7-O-glucoside and quercetin-3-O-(glucosyl)-glucoside, were identified to exhibit IC(50) values of 74.04 and 65.78?μg?mL(-1), respectively in the DPPH assay.  相似文献
