Summary The acid and base catalyzed isomerization of some tricyclic 2-pyrazolines with N-Carbamoyl-,N-thiocarbamoyl-and N-phenyl substituents was investigated. Starting fromcis ortrans 3-H, 3a-H diastereomers, equilibrium mixtures ofcis andtrans diastereomers were prepared which were separated and subsequently studied by1H NMR and13C NMR spectroscopy. A mechanism for the isomerization of the pyrazolines is suggested, supported by a deuterium exchange at C-3a.

Zusammenfassung Die Isomerisierung einiger tricyclischer 2-Pyrazoline mit N-Carbamoyl-, N-Thiocarbamoyl-und N-Phenyl-substituenten unter saurer und basischer Katalyse wurde untersucht. Ausgehend von dencis odertrans 3-H,3a-H-Diastereomeren wurdencis- undtrans Gleichgewichtsgemische gewonnen, die getrennt und durch1H- und13C-NMR-Spektroskopie untersucht wurden. Ein Mechanismus für die Isomerisierung von Pyrazolinen wird vorgeschlagen, der durch den Deuteriumaustausch in Position 3a-C unterstützt wird.

The reaction of benzil with methyl alkyl ketones gave three isomeric cyclopentenone derivatives, 2-substituted and cis- and trans-5-substituted 4-hydroxy-3,4-diphenylcyclopent-2-en-1-ones. cis- and trans-2,5-Disubstituted 4-hydroxy-3,4-diphenylcyclopent-2-en-1-ones were formed in analogous reaction of benzil with dialkyl ketones. The structure of the products was confirmed by 1H NMR spectroscopy and molecularmechanics calculations.  相似文献

The stereochemistry of substituted 2-vinyl-2-methyl-1,3-dioxa-2-silacyclohexanes was studied by 1Hand 13C NMR spectroscopy. The configurational and conformational assignment of a mixture of 2-vinyl-2,4-dimethyl-1,3-dioxa-2-silacyclohexane diastereomers with various ratios of the cis and trans forms was made. The molecules of both conformers occur chiefly in the chair conformartion with equatorial location of the methyl group at the C4 atom. The experimental data were confirmed by AM1 and MM+ optimization of the molecular geometry.  相似文献

The paper describes the synthesis of geometrical isomers and diastereomers of Pt(II) bischelates with diastereomeric hydroxy-amino acids threonine (threo-α-amino-β-hydroxybutyric acid CH3C*H(OH)C*H(NH2)COOH=ThrH) and allothreonine (erythro-α-amino-β-hydroxybutyric acid=alloThrH) containing two asymmetric carbon atoms C*: cis-,trans-[Pt(S-Thr)2], cis-,trans-[Pt(RThr)(S-Thr)], cis-,trans-[Pt(R-alloThr)(S-alloThr)] (where R and S are the absolute configurations of the asymmetric carbon atom bonded to the carboxyl group). 195Pt NMR spectroscopy is used to investigate the successive phases of the synthesis of the stereoisomeric Pt(II) complexes with threonine. The synthesized complexes are studied by 1H, 13C, 195Pt NMR spectroscopy, IR spectroscopy, and single crystal XRD.  相似文献

The cis- and trans-4-benzylparaconic acids and their ethyl esters were synthesized with high enantiomeric excess by hydrolysis of the corresponding diastereomeric lactonic esters using α-chymotrypsin. Thus, at low conversion values, cis- and trans-4-benzyl-5-oxo-3-tetrahydrofurancarboxylic acids were separately isolated with 99% ee and 92% ee, respectively. Both ethyl ester diastereomers were also obtained in enantiopure form. The absolute configuration of the trans-lactonic acid was assigned by 1H NMR analysis of its ester derivatives with both enantiomers of 1-(9-anthryl)-2,2,2-trifluoroethanol, while that of the cis-lactonic acid was assigned by means of X-ray analysis of a crystalline derivative. The circular dichroism curves of the products obtained are also reported.  相似文献

The radical-dependent oxidation of unsaturated fatty acids is a fundamental reaction in lipid chemistry, biochemistry, and technology. We report herein the first successful application of 1H–13C HMBC NMR experiment for the identification and quantification of complex and minor (3.9% to 0.85%) components of cis and trans primary hydroperoxide isomers of oxidized oleate and linoleate methyl esters in solution, without the need of laborious isolation of the individual components.  相似文献

The 1H NMR parameters of methyl 3-substituted cis-4-halotetrahydro-2-oxo-3-furancarboxylates are reported, with assignments of the ring protons based on solvent-induced changes in the vicinal trans coupling constants, 3J(H-4, H-5). Preferred conformations, ce with a pseudo-equatorial halogen for the cis isomers and ta with a pseudo-axial halogen for the trans isomers, have been suggested on comparison of the magnitudes of J(trans) and J(gem) in both series. The 3J(13CH3, H-4) values measured for methyl cis-4-bromotetrahydro-3-methyl-3-furancarboxylate, methyl trans-4-bromotetrahydro-3-methyl-3-furancarboxylate and trans-3,4-dibromodihydro-3-methyl-2(3H)-furanone have confirmed the stereochemical assignments.  相似文献

Identification of cis/trans isomers of unsaturated fatty acids cannot usually be achieved by GC-MS (gas chromatography-mass spectrometry) without reference substances. In this study a GC-FTIR-MS system (gas chromatography-Fourier transform-mass spectrometry) was used to identify fatty acid methyl esters (FAMEs) and differentiate between the cis/trans isomers. Besides methyl esters, 2-alkenyl-4,4-dimethyloxazoline derivatives (DMOX), which have been used to locate double bond positions of unsaturated fatty acids, were examined with respect to their suitability for cis/trans differentiation. A combined GC-FTIR-MS system with a wide band (4000–550 cm?1) mercury cadmium telluride (MCT) detector was used in series and parallel to identify 31 reference unsaturated fatty acids, including 7 pairs of cis/trans isomers. Serum samples of healthy persons and commercially available fish oil were analyzed as examples of complex mixtures. Using splitless injection the detection limit for the less sensitive IR detector was 25 ng/μl in case of the weak cis and trans bands. In the FTIR spectra cis/trans isomers were identified by analysis of bands arising from C? H out-of-plane (oop) bending: for both the FAME and DMOX derivatives cis-1,2-disubstituted double bonds give a strong band near 720 cm?1 and the corresponding trans isomers near 967 cm?1. cis Isomers could be identified further by a band at 3012 cm?1. With the combined data of the GC-FTIR-MS system it is now possible to identify polyunsaturated fatty acids with regard to the discrimination of cis/trans isomers.  相似文献

The 1H and 13C NMR spectra of a series of 1,3,2-dioxarsolanes have been obtained at 2.1 T and some at 9.4 T. The chemical shifts and spin-spin coupling constants have been obtained from complete spectral analyses of the 1H and proton-coupled 13C spectra. The spectral data are interpreted on the basis of two rapidly interconverting half-chair conformers with a pseudoaxial substituent at arsenic. Unique assignment of syn/anti or cis/trans geometries have been made from 1H or 13C NMR spectroscopy alone. The syn and trans isomers of the 4-methyl- and 4,5-dimethyl-1,3,2-dioxarsolanes, respectively, appear to be conformationally biased towards the forms with pseudoequatorial methyl groups. The general trends in the geminal and vicinal 1H? 1H and 13C? 1H coupling constants are interpreted in terms of stereospecific, electronegativity and lone-pair effects of the oxygen heteroatoms and conformational factors. The NMR data on the 1,3,2-dioxarsolanes are discussed with reference to related 1,3-dithoxa- and 1,3-dithia- five-membered rings with As, P, S or C at the 2-position.  相似文献

In the effort to create new derivatives of analgesically active spiropiperidines intermediate 1,2,6-trimethyl-4-piperidone was synthesized. The substitution of the skeleton gives rise to configurational as well as conformational isomerism. Despite the symmetry of 1,2,6-trimethyl-4-piperidone two different sets of signals were present in the 1H and 13C NMR spectra. They were supposed to arise from a cis/trans mixture of 1,2,6-trimethyl-4-piperidone. In contrast to this explanation only two signals of the methyl groups and hydrogens at carbon atoms 2 and 6 were observed in the 1H and 13C NMR spectra, normally expecting one for the cis- and two for the trans-isomer. To solve this discrepancy, the kind of isomeric mixture of 1,2,6-trimethyl-4-piperidone leading to the 1H and 13C NMR spectra was examined. Energy differences between chair conformations of both the cis- and the trans-isomer of 1,2,6-trimethyl-4-piperidone and the potential energy surface of the equilibration process of the trans-isomer of 1,2,6-trimethyl-4-piperidone between its chair conformers were determined by quantum chemical calculations. The barrier height of the equilibration process was measured by high and low temperature NMR measurements to confirm the theoretical outcome. The results of all investigations agree nicely and proved a cis-/trans-mixture of 1,2,6-trimethyl-4-piperidone being present at room temperature.  相似文献

A series of ruthenium hydride compounds containing substituted bidentate pyrrole‐imine ligands were synthesized and characterized. Reacting RuHCl(CO)(PPh3)3 with one equivalent of [C4H3NH(2‐CH=NR)] in ethanol in the presence of KOH gave compounds {RuH(CO)(PPh3)2[C4H3N(2‐CH=NR)]} where trans‐Py‐Ru‐H 1, R = CH2CH2C6H9; cis‐Py‐Ru‐H 2, R = Ph‐2‐Me; and cis‐Py‐Ru‐H 3, R = C6H11. Heating trans‐Py‐Ru‐H 1 in toluene at 70°C for 12 hr resulted a thermal conversion of the trans‐Py‐Ru‐H 1 into its cis form, {RuH(CO)(PPh3)2[C4H3N(2‐CH=NCH2CH2C6H9)]} (cis‐Py‐Ru‐H 1) in very high yield. The 1H NMR spectra of trans‐Py‐Ru‐H 1, cis‐Py‐Ru‐H 2, cis‐Py‐Ru‐H 3, and cis‐Py‐Ru‐H 1 all show a typical triplet at ca. δ–11 for the hydride. The trans and cis form indicate the relative positions of pyrrole ring and hydride. The geometries of trans‐Py‐Ru‐H 1, cis‐Py‐Ru‐H 1, and cis‐Py‐Ru‐H 3 are relatively similar showing typical octahedral geometries with two PPh3 fragments arranged in trans positions.  相似文献

1H NMR chemical shifts of 18 methyl substituted cyclohexane-1,2-dicarboxylic anhydrides were determined. In compounds with asymmetric substitution vicinal coupling constants were measured. In the series of trans-anhydrides it can be deduced from the NMR data, that the 6-membered ring has a chair-like conformation. Additionally, in the series of the cis-anhydrides a twisted chair conformation is favoured for the most part.  相似文献

cis‐ and trans‐2‐imino‐1,3‐ and ‐3,1‐perhydrobenzoxazines and the N‐methyl derivatives of the latter were synthesized from the corresponding cyclic 1,3‐amino alcohol with cyanogen bromide. The configurations of the studied compounds were confirmed by 1H and 13C NMR spectra. All trans‐fused compounds exist in biased chair–chair conformations as expected, whereas the cis‐fused 1,3‐benzoxazines attain exclusively the O‐in conformations. The cis‐fused 3,1‐benzoxazines, especially the 1‐methyl‐substituted derivatives, tend to favor the N‐out form, obviously owing to the favorable axial orientation of this N‐methyl. The thioether-group-containing SNS-type pincer complex [({EtSCH2CH2}2NH)RuCl(H)(PPh3)] ( 2 ) exists in three different diastereomers ( 2 a – c ). The molecular structures obtained from single crystal X-Ray diffraction studies of all three isomers reveal a difference in the relative orientation of the respective EtS-groups, while other commonly observed diastereomers as the result of cis-/trans- or fac-/mer-isomerism are not observed. Chemical exchange between the three diastereomers 2 a – c was discovered by phase-sensitive 1H and 31P NOESY NMR spectroscopy, and further quantified by line shape analysis of 1H NMR spectra. The experimentally derived averaged Gibbs energies of activation for the interconversion of the isomers (65–70 kJ/mol) are in good agreement with the results obtained from DFT calculations, which suggest an inversion of the ligating sulfur atoms, although a dissociative pathway for the configurational inversion can be competitive.  相似文献

1H and 13C NMR chemical shifts of cis and trans isonucleoside analogues of purine in which the furanose moiety is substituted by a tetrahydropyran ring were completely assigned using one‐ and two‐dimensional NMR experiments that include NOE, DEPT, COSY and HSQC. The 1H NMR spectra of 35 cyclic and acyclic esters are analysed to give the 1H chemical shifts and couplings. The substituent chemical shifts of the ester group were analysed using three‐bond (γ) effects for near protons and the electric field, magnetic anisotropy and steric effect of the ester group for more distant protons. The electric field is calculated from the partial atomic charges on the O?C = O atoms, and the asymmetric magnetic anisotropy of the carbonyl group acts at the midpoint of the C = O bond. The values of the anisotropies Δχparl and Δχperp were for the aliphatic esters 10.35 and ?18.84 and for the conjugated esters 7.33 and ?15.75 (×10?6 Å3/molecule). The oxygen steric coefficients found were 104.4 (aliphatic C = O), 45.5 (aromatic C = O) and 16.0 (C–O) (×10?6 Å6/molecule). After parameterisation, the overall RMS error for the data set of 280 entries was 0.079 ppm. The strongly coupled 1H NMR spectra of the 2‐methyl, 3‐methyl and 4‐methyl γ‐butyrolactones were analysed and the methyl conformational equilibrium obtained from the observed couplings. The observed versus calculated density functional theory (DFT) ΔG(ax‐eq) was 1.0 (1.01), 0.34 (0.54) and 0.65 (0.71) kcal/mol res. The shielding effect of a methyl cis to a proton in the five‐membered lactone rings is ?0.40 ±0.05 ppm and deshielding trans effect 0.12 ±0.05 ppm, which is common to both five and six membered rings. The cis/trans isomerism in the vinyl esters methyl acrylate, crotonate and methacrylate and methyl furoate was examined using the 1H chemical shifts. The 1H NMR study of 2-alkyl-3-chlorotetrahydropyrans, obtained by reaction of Grignard reagents with a mixture of cis/trans-2,3-dichlorotetrahydropyrans, shows cis/trans configuration of two isomers in which the alkyl substituents are exclusively in the equatorial position. 3-Chloro-2-phenyltetrahydropyran exists in trans (eq-eq) configuration only. The 1H NMR study of cis/trans 2-alkoxy (or aryloxy)-3-chlorotetrahydropyrans, obtained by reaction of alcohols or phenol with 2,3-dichlorotetrahydropyrans, shows the axial position of the alkoxy (or aryloxy) substituent.  相似文献

Resveratrol (3,5,4′‐trihydroxylstilbene), a phytoalexin in response to injury or fungal attack, is found in grapes and other food products. It has been well documented that the compound has beneficial effects as hypolipidemic, anticancer, antiviral, neuroprotective, antiaging, and anti‐inflammatory natural active principle. It was observed that both trans‐ and cis‐resveratrols undergo hydrogen‐deuterium (H/D) exchange at H‐2,6 and H‐4 positions of the A‐ring. The exchange rates were determined by 1H NMR spectroscopy. The results reveal that the exchange rates are configuration and pH dependent. Derivative of 2‐OβD‐glucoside can significantly speed up the H/D exchange reactions for both isomers. The trans‐resveratrol experiences faster H/D exchanging than the cis‐resveratrol. Such isomeric effect is possibly due to the factor that the trans‐resveratrol is in favor of forming larger/super conjugative system and has less spatial interference. Theoretical calculation shows that electronegativity at these positions is in the order of H‐2,6>H‐4, which is in consistent with the exchange rates observed by NMR. The results may be of help in understanding the properties of resveratrol, and in analysis of resveratrol in natural products or body fluids using mass spectroscopy that occasionally requires stable deuterium isotope labeling.  相似文献

The configurations and conformations of cis- and trans-N-methyl- and -N-benzyl-4,5- and -5,6-tetramethylenetetrahydro-1,3-oxazines were determined by 1H and 13C NMR spectroscopy. The cis isomers are conformationally homogeneous, having the hetero atom attached to the cyclohexyl ring, in the axial and equatorial positions, respectively, in the 5,6- and 4,5-tetramethylene compounds, similar to the case of the 2-p-nitrophenyl-substituted analogues investigated previously.  相似文献
