The construction of complex polycyclic terpenoid products in an efficient and step‐economical manner using multicomponent and tandem processes is highly valuable. Herein, we report a tandem cyclization sequence that initiates with a multicomponent double Diels–Alder reaction of cross‐conjugated diynones, followed by a Nazarov cyclization to efficiently produce [6‐5‐6] tricyclic products with excellent regio‐ and diastereoselectivity. This methodology generates five new carbon–carbon bonds, three rings, quaternary or vicinal quaternary carbons, and stereogenic centers in a one‐pot reaction.  相似文献

Primary amine‐catalyzed direct conversion of α,α‐disubstituted aldehydes into 3‐pyrrolines with a quaternary stereocenter is reported. The one‐pot enantioselective sequence is based on a α‐amination, an aza‐Michael addition of hydrazine, an aldol condensation dehydratation and proceeds with good yields and excellent levels of enantioselectivity. Synthetically attractive applications including the formation of aziridinopyrrolidine or epoxypyrrolidine derivatives with good yields and selectivities are also described.  相似文献

An asymmetric organocatalytic one‐pot strategy for the construction of spirooctahydroacridine‐3,3′‐oxindole scaffolds has been successfully developed by means of a domino Michael/Povarov reaction sequence. The one‐pot protocol affords the chiral spirocyclohexaneoxindoles bearing an octahydroacridine motif with five stereocenters in good to high yields (up to 89 % yield) with excellent to perfect diastereoselectivities (up to >20:1 d.r.) and enantioselectivities (up to >99 % ee). This highly efficient one‐pot domino procedure will allow diversity‐oriented syntheses of this intriguing class of compounds with potential biological activities.  相似文献

The highly enantioselective organocatalytic addition of ethyl nitroacetate to isatin‐derived N‐Boc ketimines (Boc=tert‐butoxycarbonyl), followed by the removal of the nitro group, is described. The scalable reaction sequence leads to the title compounds as important intermediates of pyrroloindoline alkaloids and related drugs in excellent yields and enantioselectivities. The synthesis of the hexahydrofurano[2,3‐b]indole skeleton, the spirocarbamate oxindole unit, and the formal synthesis of AG‐041R have been carried out to demonstrate the synthetic utility of this protocol.  相似文献

In this article, the total syntheses of antimalarial compound decursivine and its biologically inactive sibling serotobenine are presented. The biomimetic synthesis of (±)‐serotobenine was investigated first, but failed. During the subsequent investigation of other synthetic routes, we discovered a new cascade Witkop photocyclization/elimination/addition sequence, which enabled the expedient synthesis of not only racemic decursivine and serotobenine, but also enantiopure (+)‐ and (?)‐decursivine and a variety of their analogues. The present syntheses represent the shortest pathway for the total synthesis of decursivine and serotobenine to date. Moreover, the newly developed cascade sequence for the total synthesis of decursivine does not need any protecting steps. The scope and the reaction mechanism of the cascade sequence were also studied. A rational mechanism for the cascade sequence is proposed, which is consistent with the previous studies and our current experimental results.  相似文献

Gold catalysts have been applied in cascade‐type reactions for the synthesis of different nitrogen‐based compounds. The reactions likely proceed by a new gold‐catalyzed cascade intermolecular α‐amidoalkylation/intramolecular carbocyclization cascade process by unifying both the σ‐ and π‐Lewis acid properties of the gold salts. In the first part of this report we show that the σ‐Lewis acidity of gold(I) and gold(III) could be exploited to efficiently catalyze the nucleophilic substitution of various alkoxy‐ and acetoxylactams. The reaction was found to be applicable to a wide range of cyclic N‐acyliminium ion precursors and various nucleophiles, including allyltrimethylsilane, silyl enol ethers, arenes, and active methylene derivatives. As a logical progression of this study, a combined hard/soft binary catalytic gold system was then used to implement an unprecedented tandem intermolecular Friedel–Crafts amidoalkylation/intramolecular hydroarylation sequence allowing an expedient access to new, complex, fused polyheterocyclic structures from trivial materials.  相似文献

A cobalt‐catalyzed chelation‐assisted tandem C?H activation/C?C cleavage/C?H cyclization of aromatic amides with alkylidenecyclopropanes is reported. This process allows the sequential formation of two C?C bonds, which is in sharp contrast to previous reports on using rhodium catalysts for the formation of C?N bonds. Here the inexpensive catalyst system exhibits good functional‐group compatibility and relatively broad substrate scope. The desired products can be easily transformed into polycyclic lactones with m‐CPBA. Mechanistic studies revealed that the tandem reaction proceeds through a C?H cobaltation, β‐carbon elimination, and intramolecular C?H cobaltation sequence.  相似文献

Acid‐catalyzed tandem reactions with auto‐tandem catalysis are effective for simplifying organic synthesis. However, some of the reported reactions were established based on the use of well‐designed substrate with complex structure. In some cases, owing to the existence of a big gap between each catalytic cycle, it is hard to bind all the individual reaction steps to be a peaceful sequence. To enrich the diversity and also to strengthen the practical usefulness of the methodology developed by auto‐tandem catalysis, an additive‐like component was added to induce acid‐acid‐catalyzed tandem reaction. During the reaction, the additive‐like component acted either as an activator to increase the reactivity of the starting material or a hided reagent to enable successful transformation of the intermediate. Many novel tandem reactions were established in a one‐pot manner with the aid of this strategy. Importantly, this strategy not only allows the use of simple and commercially available chemicals as substrates, but also possesses multiple merits, such as simplifying operation, lowering waste generation and enhancing synthetic efficiency and atom‐economy. A summarization of the additive‐like component‐induced auto‐tandem catalysis with an acid catalyst was given in this review, in which many acid‐acid‐catalyzed tandem reactions were discussed. The reported additive‐like components were classified as three types: oxidative type, reductive type and neutral type depending on their mechanisms in assisting the establishment of acid‐acid‐catalyzed tandem reactions. Many examples were collected and analyzed from the viewpoints of simplifying the synthesis and manifesting their superior and distinct functionalities of the additives. A perspective of this concept was also given at the end of this review.  相似文献

An allele‐specific voltammetric genoassay for the detection of allele‐specific toll‐like receptor‐2 gene arg753gln polymorphism (TLR‐2) from polymerase chain reaction (PCR) amplified real samples was described in this study. Meldola blue (MDB), an intercalator molecule, was used as hybridization label. The wild‐type and mutant type oligonucleotide probes were immobilized onto disposable graphite electrode surfaces by covalent attachment method. The extent of hybridization between probe and target sequences was determined by using differential pulse voltammetry (DPV). As a result of the interaction between MDB and DNA at electrode surface, the MDB signal observed from probe sequence before hybridization and after hybridization with MM sequence is lower than that observed after hybridization with complementary sequence. The differences between the MDB reduction peaks obtained from probe modified, hybrid modified and MM modified electrode were used to detect TLR‐2 from PCR amplified real samples. The discrimination of homozygous and heterozygous alleles was also established by comparing the peak currents of MDB reduction signals. Numerous factors affecting the target hybridization and indicator binding reactions are optimized to maximize the sensitivity.  相似文献

A ruthenium hydride/Brønsted acid‐catalyzed tandem sequence is reported for the synthesis of 1,3,4,9‐tetrahydropyrano[3,4‐b]indoles (THPIs) and related oxacyclic scaffolds. The process was designed on the premise that readily available allylic ethers would undergo sequential isomerization, first to enol ethers (Ru catalysis), then to oxocarbenium ions (Brønsted acid catalysis) amenable to endo cyclization with tethered nucleophiles. This methodology provides not only an attractive alternative to the traditional oxa‐Pictet–Spengler reaction for the synthesis of THPIs, but also convenient access to THPI congeners and other important oxacycles such as acetals.  相似文献

A new and convenient method for the preparation of 3,4‐dihydroquinazolines 5 with aryl and Ac groups at C(2) and N(3), respectively, has been developed. The key sequence is the formation of aza‐phosphorane intermediates by the reaction of N‐[2‐(azidomethyl)phenyl]benzamides 1 with Ph3P, followed by intramolecular aza‐Wittig reaction and 3‐acetylation, which can be conducted in one‐pot.  相似文献

The efficient synthesis of a sequence‐defined decamer, its characterization, and its straightforward dimerization through self‐metathesis are described. For this purpose, a monoprotected AB monomer was designed and used to synthesize a decamer bearing ten different and selectable side chains by iterative Passerini three‐component reaction (P‐3CR) and subsequent deprotection. The highly efficient procedure provided excellent yields and allows for the multigram‐scale synthesis of such perfectly defined macromolecules. An olefin was introduced at the end of the synthesis, allowing the self‐metathesis reaction of the resulting decamer to provide a sequence‐defined 20‐mer with a molecular weight of 7046.40 g mol?1. The obtained oligomers were carefully characterized by NMR and IR spectroscopy, GPC and GPC coupled to ESI‐MS, and mass spectrometry (FAB and orbitrap ESI‐MS).  相似文献

Our goal was to compare two popular analytical techniques used nowadays in proteomic investigations for proteins/peptides sequencing and identification, a widely used nanoLC‐MS/MS approach applied in the bottom‐up proteomics and electron transfer dissociation/proton transfer reaction fragmentation preferably used when top‐down strategy is applied. Comparison was carried out with the aid of the ESI‐quadrupole ion‐trap instrument using the following criteria: total time of analysis including sample preparation, sequence coverage, Mascot scoring, capability to detect modifications, quality of the results as a function of protein molecular weight and sample consumption. An efficient method for Knoevenagel condensation of arenecarbaldehydes with active methylene compounds such as barbituric acid and Meldrum's acid in the presence of gel‐entrapped base catalysts is reported. The method has been extended to the one‐pot synthesis of arylmethylene‐bis[3‐hydroxycyclohex‐2‐en‐1‐one] derivatives from dimedone (=5,5‐dimethylcyclohexane‐1,3‐dione) and arenecarbaldehydes by using domino Knoevenagel/Michael reaction sequence.  相似文献

The first asymmetric total synthesis of the antifeedant terpenoids (+)‐norleucosceptroid A, (?)‐norleucosceptroid B, and (?)‐leucosceptroid K has been accomplished. This highly concise synthetic route was guided by our efforts to develop a platform for the collective synthesis of a whole family of antifeedant natural products. The synthesis features a Hauser–Kraus‐type annulation followed by an unprecedented, highly efficient intramolecular dilactol aldol‐type condensation reaction to produce the 5,6,5 skeleton. The developed synthetic route proceeds for norleucosceptroid A and B in 16 steps (longest linear sequence) from known compounds.  相似文献

We applied a facile LIF dual‐channel monitoring system recently developed and reported by our group to the polymerase chain reaction/ligase detection reaction/CGE method for detecting low‐abundance point mutations present in a wild‐type sequence‐dominated population. Mutation discrimination limits and signaling fidelity of the analytical system were evaluated using three mutant variations in codon 12 of the K‐ras oncogene that have high diagnostic value for colorectal cancer. We demonstrated the high sensitivity of the present method by detecting rare mutations present among an excess of wild‐type alleles (one mutation among ~100 normal sequences). This method also simultaneously interrogated the allelic compositions of the test samples with high specificity through spectral discrimination of the dye‐tagged ligase detection reaction products using the dual‐channel monitoring system.  相似文献

A reaction sequence of 2‐(diphenylsilyl)phenylation by Negishi coupling and intramolecular sila‐Friedel–Crafts reaction has been explored for the synthesis of mono‐triphenylsilane‐fused porphyrins 5 M and 6 M (M= Ni, Zn) and bis‐triphenylsilane‐fused porphyrins 7 M and 8 Ni . A triply linked triphenylsilane‐fused NiII porphyrin, 13 Ni , was synthesized in a stepwise manner involving the above reaction sequence and a final Pd‐catalyzed C?H activating arylative cyclization. The silicon atom in 13 Ni takes a distorted planarized structure with an almost perpendicular Si‐phenyl group, causing an electronic effect due to effective σ*–π* interaction.  相似文献

A simple and straightforward approach was developed to construct 5H‐benzo[b]carbazole derivatives by iron catalysis in a cascade sequence. The notable features of this work include an atom‐economical cascade sequence, unprecedented 1,4‐sulfonyl migration, tolerance of a variety of functional groups, good yields, and an economical catalytic system.  相似文献
