The reaction of 2,4‐pentanedione ( 1 ) with (R)‐(—)‐2‐phenylglycine methyl ester ( 2 ), (R)‐(—)‐2‐phenylglycinol ( 3 ) and the proteinogenic amino acids (2S,3R)‐(—)‐2‐amino‐3‐hydroxybutyric acid (L ‐threonine) ( 4 ) and (R)‐(—)‐2‐amino‐3‐mercaptopropionic acid (L ‐cysteine) ( 5 ) methyl esters was investigated. The corresponding enamines 6 , 7 and 8 were isolated and characterized spectroscopically whereas 9 , which is unstable, was transformed in situ into 13 . Treatment of 7 , 8 and 9 with boron trifluoride etherate afforded the new [1,4]oxazepines 10 , 11 and [1,4]thiazepine ( 12 ) as their BF3O? salts. The structures of the enamines and their corresponding seven‐membered heterocycles were assessed by 1D and 2D NMR spectroscopy. Phosphorylation of suitable piperidine precursors yielded a series of novel decalin‐type O,N,P‐heterocycles. The title compounds, P(3)‐axially and P(3)‐equatorially X‐substituted, cis‐ and trans‐configurated 2,4‐dioxa‐7‐aza‐, 2,4‐dioxa‐8‐aza‐, and 2,4‐dioxa‐9‐aza‐3‐phosphabicyclo[4.4.0]decane 3‐oxides (X=Cl, F, 4‐nitrophenoxy, and 2,4‐dinitrophenoxy), are configuratively fixed and conformationally constrained P‐analogues of acetylcholine and as such represent acetylcholine (7‐aza and 9‐aza isomers) or γ‐homo‐acetylcholine mimetics (8‐aza isomers). Being irreversible inhibitors of acetylcholinesterase (AChE), the compounds are considered to be suitable probes for the investigation of the stereochemical course of the inhibition reaction by 31P‐NMR spectroscopy. Moreover, the design of these mimetics will enable studies of molecular interactions with AChE, in particular, the recognition conformation of acetylcholine.  相似文献

Sixteen different P(III) and P(V) heterocycles derived from 2‐(2‐hydroxyphenyl)‐1H‐benzimidazole ( 1 ) are reported. In these heterocycles the phosphorus atom is part of a six‐membered unsaturated ring. They were mainly studied by multinuclear NMR. The X‐ray diffraction of 3,4‐ benzimidazole‐5,6‐benzo‐2‐dimethylamino‐2‐seleno‐ 1,3,2‐oxazaphosphorinane is reported. Phosphoranes derived from 1 and 3,5‐di‐tert‐butylcatechol, and bearing Cl, NMe2, or phenyl as substituent at phosphorus are presented. © 2004 Wiley Periodicals, Inc. A new series of 1,2,4‐triazole was designed, synthesized, and characterized as remarkable antimicrobial and antioxidant agents. These heterocycles have been prepared from the cyclization reactions of Schiff bases 3 ( a‐k ) with phenylhydrazine by refluxing under the alkaline medium. The Schiff bases in turn were realized in good yields from the condensation reactions of N‐phenylurea with different aromatic aldehydes. The structures of the intermediates 3 ( a‐k ) and final heterocycles 4 ( a‐k ) have been fully characterized through their spectral parameters.  相似文献

New phosphorus‐containing, five‐membered P,P,P and P,N,P heterocycles were synthesized and fully characterized. The P,P,P heterocycles, 1,2,3‐triphospholanes, can be synthesized by two different facile pathways, whereas the P,N,P compound, a 1‐aza‐2,5‐diphospholane, can only be obtained with silylamine.  相似文献

Reactivity of the title P‐heterocycles ( 1‐14 ) in the photoinduced fragmentation‐related phosphinylation of methanol was found to be influenced by the extent of ring strain and the UV absorption at 254 nm. The 7‐phosphanorbornene oxides ( 7‐14 ) are universal precursors due to their ring strain, no matter if they are UV‐active or not at 254 nm. The easily available 2,5‐dihydro‐1H‐phosphole oxides can be applied only in case of 1‐phenyl substitution that enhances the absorption at 254 nm. The ring strain of representative P‐heterocycles ( 5‐8 ) was evaluated by HF/6‐31G* and B3LYP/6‐31+G* calculations. UV spectra of compounds 5‐8 were interpreted by ZINDO/S and MNDO‐d calculations. The new precursors ( 11‐14 ) made possible the extension of the phosphinylations.  相似文献

Energetic compounds that incorporate multiple nitrogen‐rich heterocycles are of great interest for high‐density energetic materials. A facile synthetic strategy to combine an oxy bridge and furazan groups, as well as tetrazole‐ols, into a molecule ( 5 ) was found. Some energetic salts based on 5 were prepared by neutralization. All of the compounds were fully characterized. Additionally, the structure of 7 has been elucidated by single‐crystal XRD analysis. Physicochemical and energetic properties were also studied; these show that these newly designed energetic salts exhibit good thermal stabilities. Hydroxylammonium salt ( 6 ) has a detonation performance and sensitivities comparable with those of 1,3,5‐trinitroperhydro‐1,3,5‐triazine (RDX).  相似文献

A tandem reductive amination‐SNAr reaction has been developed for the synthesis of 6‐nitro‐1,2,3,4‐tetrahydroquinolines. Treatment of 4‐(2‐fluoro‐5‐nitrophenyl)‐2‐butanone or 3‐(2‐fluoro‐5‐nitrophenyl)‐propanal with primary amines and sodium cyanoborohydride in methanol at room temperature provided good to excellent yields of the substituted tetrahydroquinolines. The reaction proceeded best with the ketone substrate using primary amines that were unbranched at the α‐carbon. The aldehyde also produced the target heterocycles, but these were accompanied by 10‐15% of the uncyclized side chain reductive amination products.  相似文献

Eight heterocycles have been prepared in a one‐pot reaction manner based on the Hantzsch dihydropyridine synthesis. The synthesis afforded seven dihydropyridines (DHP) and one unexpected 1,3‐oxazin‐6‐one. Their structures were confirmed based on NMR spectroscopy and mass spectrometry. The obtained products have been evaluated for their cytotoxicity against eight cancer cell lines and one normal cell line. Two halogenated DHPs ( 7 and 8 ) displayed cytotoxicity toward all the nine tested cancer cell lines with IC50 values from 4.10 to 58.90 μm, while others showed selective activities. DHPs ( 7 and 8 ) bearing a Me group at C(2) and C(6) as well as a halogenated substituent at C(4′) were more antiproliferative than the others.  相似文献

A new family of phosphorus heterocycles, namely 8‐alkylcarbamato‐16H‐dinaphtho‐[2,1‐d: 1′,2′‐g] 1,3,2‐dioxaphosphocin 8‐oxides ( 4a–j ) has been obtained by reaction of bis(2‐hydroxy‐1‐naphthyl)methane ( 3 ) with a series of dichlorophosphosphinyl carbamates ( 2a–j ) in dry toluene in the presence of triethylamine at 40–45°C. The intermediates 2a–j were obtained by the addition of alcohols/thiol to isocyanatophosphonic dichloride ( 1 ) at −10°C in dry toluene. The structures of the title compounds were confirmed by the elemental analyses, IR, 1H, 13C, and 31P NMR spectra. The FAB mass spectrum of one member of the family is discussed. A series of novel heterocycles 1‐aryl‐ or alkyl‐substituted‐4‐arylazamethylene‐6‐arylpyrazolo[5,4‐d]‐1,3‐oxazines were synthesized from the acylation of (5‐amino‐1‐substituted‐pyrazol‐4‐yl)‐N‐carboxamide in 63‐89% isolated yields at room temperature within 12 hours. The structure was confirmed by X‐ray crystal analysis.  相似文献

The first rhodium(I)‐catalyzed enantioselective intermolecular C –H activation of various saturated aza‐heterocycles including tetrahydroquinolines, piperidines, piperazines, azetidines, pyrrolidines, and azepanes is presented. The combination of a rhodium(I) precatalyst and a chiral monodentate phosphonite ligand is shown to be a powerful catalytic system to access a variety of important enantio‐enriched heterocycles from simple starting materials. Notably, the C –H activation of tetrahydroquinolines is especially challenging due to the adjacent C −H bond. This redox‐neutral methodology provides a new synthetic route to α‐N‐arylated heterocycles with high chemoselectivity and enantioselectivity up to 97 % ee.  相似文献

To evaluate the effect of substituents on biological activities of electron‐rich N‐containing heterocycles, the variably 2‐substituted 5,6‐dihydro‐5‐oxo‐4H‐1,3,4‐oxadiazine‐4‐propanenitriles 26 – 33 were synthesized and evaluated for antibacterial, antifungal, and enzyme‐inhibition activities. The target compounds were obtained from alkyl 4‐ or 3‐hydroxy benzoates 1 and 2 , respectively, and from methyl indoleacetate 3 . The phenolic OH group of benzoates 1 and 2 were substituted with p‐toluenesulfonyl (→ 4 and 5 ), benzoyl (→ 6 and 7 ), and benzyl groups (→ 8 and 9 ) and then converted to 5,6‐dihydro‐5‐oxo‐4H‐1,3,4‐oxadiazine‐4‐propanenitriles. To establish structure‐activity relationships (SAR), a pharmacological screening of the intervening intermediates was also conducted, which revealed that the intermediate hydrazide 11 possesses significant antimicrobial and MAO‐A inhibiting properties and intermediates 12, 24, 28 , and 29 appreciable antifungal activities. The first stable 1‐phospha‐2‐boraacenaphthene 1 was synthesized by the reduction of 1‐dimesitylboryl‐8‐dichlorophosphinonaphthalene ( 2 a ) with elemental magnesium, and it was fully characterized. The chalcogenation reaction of 1 with elemental sulfur or selenium afforded the unique heterocycles, 2‐thia‐ and 2‐selena‐1‐phospha‐3‐boraphenalenes 9 S and 9 Se , respectively, through the insertion of the chalcogen atom into a P?B bond of 1 . Further chalcogenation of 9 afforded the corresponding phosphine chalcogenides. These newly obtained chalcogenated compounds have been characterized. The unique dynamic behavior of 2‐chalcogena‐1‐phospha‐3‐boraphenalene‐1‐chalcogenides 10 in solution has also been described.  相似文献

A new asymmetric synthesis of chiral 1,4‐dioxanes and other oxa‐heterocycles has been developed by means of organocatalytic enantioselective desymmetrization of oxetanes. This mild process proceeds with exceedingly high efficiency and enantioselectivity to establish the quaternary stereocenters. This method complements the existing, yet limited, strategies for the synthesis of these oxa‐heterocycles.  相似文献

Reported herein is the divergent syntheses of [5,5] and [6,5] spiro‐heterocycles under Lewis‐acid‐assisted palladium catalysis. In particular, an unprecedented switch from alkoxide‐π‐allyl to dienolate reactivity was achieved by the use of palladium‐titanium relay catalysis, and represents umpolung reactivity of vinylethylene carbonates. This method uses a simple procedure and commercially available catalysts, and delivers both classes of spiro‐heterocycles, bearing three contiguous stereocenters, in high yield and uniformly excellent diastereoselectivity.  相似文献

Nucleophilic trifluoromethylation of α‐imino ketones 2 , derived from arylglyoxal, with RuppertPrakash reagent (CF3SiMe3) offers a convenient access to the corresponding O‐silylated β‐imino‐α‐(trifluoromethyl) alcohols. In a ‘one‐pot’ procedure, by treatment with NaBH4, these products smoothly undergo reduction and desilylation yielding the expected β‐amino‐α‐(trifluoromethyl) alcohols 4 . The latter were used as starting materials for the synthesis of diverse trifluoromethylated heterocycles, including aziridines 5 , 1,3‐oxazolidines 8 , 1,3‐oxazolidin‐2‐ones 9 , 1,3,2‐oxazaphospholidine 2‐oxides 10 , 1,2,3‐oxathiazolidine 2‐oxides 11 , and morpholine‐2,3‐diones 12 . An optically active 5‐(trifluoromethyl)‐substituted 1,3‐oxazolidin‐2‐one 9g was also obtained.  相似文献

A general catalytic hydrogen transfer‐mediated α‐functionalization of 1,8‐naphthyridines is reported for the first time that benefits from a hydrogen transfer‐mediated activation mode for non‐activated pyridyl cores. The pyridyl α‐site selectively couples with the C8‐site of various tetrahydroquinolines (THQs) to afford novel α‐functionalized tetrahydro 1,8‐naphthyridines, a class of synthetically useful building blocks and potential candidates for the discovery of therapeutic and bio‐active products. The utilization of THQs as inactive hydrogen donors (HDs) appears to be a key strategy to overcome the over‐hydrogenation barrier and address the chemoselectivity issue. The developed chemistry features operational simplicity, readily available catalyst and good functional group tolerance, and offers a significant basis for further development of new protocols to directly transform or functionalize inert N‐heterocycles.  相似文献
