An efficient system for the production of (R)-hydroxyalkanoicacids (RHAs) was developed in natural polyhydroxyalkanoate (PHA)-producing bacteria and recombinant Escherichia coli. Acidic alcoholysis of purified PHA and in vivo depolymerization of PHA accumulated in the cells allowed the production of RHAs. In recombinant E. coli, RHA production was achieved by removing CoA from (R)-3-hydroxyacyl-CoA and by in vivo depolymerization of PHA. When the recombinant E. coli harboring the Ralstonia eutropha PHA biosynthesis genes and the depolymerase gene was cultured in a complex or a chemically defined medium containing glucose, (R)-3-hydroxybutyric acid (R3HB) was produced as monomers and dimers. R3HB dimers could be efficiently converted to monomers by mild alkaline heat treatment. A stable recombinant E. coli strain in which the R. eutropha PHA biosynthesis genes were integrated into the chromosome disrupting the pta gene was constructed and examined for the production of R3HB. When the R. eutropha intracellular depolymerase gene was expressed by using a stable plasmid containing the hok/sok locus of plasmid R1, R3HB could be efficiently produced.  相似文献

Plastic wastes constitute a worldwide environmental problem, and the demand for biodegradable plastics has become high. One of the most important characteristics of microbial polyesters is that they are thermoplastic with environmentally degradable properties. In this study, pUC 19/PHA was cloned and transformed into three different Escherichia coli strains. Among the three strains that were successfully expressed in the production of polyhydroxyalkanoates (PHA), E. coli HMS174 had the highest yield in the production of poly-(3-hydroxybutyrate-co-3-hydroxyvalerate) (P[HB-HV]). The cell dry weight and PHA content of recombinant HMS174 reached as high as 10.27 g/L and 43% (w/w), respectively, in fed-batch fermentor culture. The copolymer of PHA, P(HB-HV), was found in the cells, and the biopolymers accumulated were identified and analyzed by gas chromatography, proton nuclear magnetic resonance spectroscopy, and differential scanning calorimetry. We demonstrated clearly that the E. coli host for PHA production has to be carefully selected to obtain a high yield. The results obtained indicated that a superior E. coli with high PHA production can be constructed with a desirable ratio of P(HB-HV), which has potential applications in industry and medicine.  相似文献

Poly(3-hydroxybutyrate) [P(3HB)], a polymer belonging to the polyhydroxyalkanoate (PHA) family, is accumulated by numerous bacteria as carbon and energy storage material. The mobilization of accumulated P(3HB) is associated with increased stress and starvation tolerance. However, the potential function of accumulated copolymer such as poly(3-hydroxybutyrate-co-3-hydroxyvalerate) [P(3HB-co-3HV)] remained unknown. In this study, Delftia acidovorans DS 17 was used to evaluate the contributions of P(3HB) and P(3HB-co-3HV) granules during simulated exogenous carbon deprivation on cell survival by transferring cells with PHAs to carbon-free mineral salt medium supplemented with 1 % (w/v) nitrogen source. By mobilizing the intracellular P(3HB) and P(3HB-co-3HV) at 11 and 40 mol% 3HV compositions, the cells survived starvation. Surprisingly, D. acidovorans containing P(3HB-co-94 mol% 3HV) also survived although the mobilization was not as effective. Similarly, recombinant Escherichia coli pGEM-T::phbCAB Cn (harboring the PHA biosynthesis genes of Cupriavidus necator) containing P(3HB) granules had a higher viable cell counts compared to those without P(3HB) granules but without any P(3HB) mobilization when exposed to oxidative stress by photoactivated titanium dioxide. This study provided strong evidence that enhancement of stress tolerance in PHA producers can be achieved without mobilization of the previously accumulated granules. Instead, PHA biosynthesis may improve bacterial survival via multiple mechanisms.  相似文献

The thermogenic curves of metabolism of the four strains of Escherichia coli pUC19cab/JM109, pUC19cab/XL-IBlue, JM109 and XL-IBlue were determined using the LKB-2277 BioActivity Monitor and the ampoule method at 37°C. The pUC19cab/JM109 and pUC19cab/XL-IBlue are recombinant E. coli strains bearing the same foreign plasmid pUC19cab, which confers the ability to produce polyhydroxyalkanoates (PHAs). The yield of PHAs of pUC19cab/XL-IBlue was higher than that of pUC19cab/JM109. XL-IBlue and JM109 were the host bacteria. The heat flow of these strains was XL-Blue≈JM109>pUC19cab/JM109>pUC19cab/XL-IBlue. These results indicate an obvious interrelation between the PHAs production and the heat flow rate of E. coli strains.  相似文献

Polyhydroxyalkanoates (PHAs) are polyesters of hydroxyalkanoates synthesized by numerous bacteria as intracellular carbon and energy storage compounds and accumulated as granules in the cytoplasm of cells. In this work, we constructed two recombinant plasmids, pBE2C1, and pBE2C1AB, containing one or two PHA synthse, genes, respectively. The two plasmids were inserted into Bacillus subtilis DB104 to generate modified strains, B. subtilis/pBE2C1 and B. subtilis/pBE2C1AB. The two recombinants strains were subjected to fermentation and showed PHA accumulation, the first reported example of mcl-PHA production in B. subtilis. Gas Chromatography analysis identified the compound produced by B. subtilis/pBE2C1 to be a hydroxydecanoate-co-hydroxydodecanoate (HD-co-HDD) polymer whereas that produced by B. subtilis/pBE2C1AB was a hydroxybutyrate-co-hydroxyde-canoate-co-hydroxydodecanoate (HB-HD-HDD) polymer.  相似文献

Poly(hydroxyalkanoate)s (PHAs) are a class of microbially synthesized polyesters that combine biological properties, such as biocompatibility and biodegradability, and non-bioproperties such as thermoprocessability, piezoelectricity, and nonlinear optical activity. PHA monomer structures and their contents strongly affect the PHA properties. Using metabolic engineering approaches, PHA structures and contents can be manipulated to achieve controllable monomer and PHA cellular contents. This paper focuses on metabolic engineering methods to produce PHA consisting of 3-hydroxybutyrate (3HB) and medium-chain-length 3-hydroxyalkanoates (3HA) in recombinant microbial systems. This type of copolyester has mechanical and thermal properties similar to conventional plastics such as poly(propylene) and poly(ethylene terephthalate) (PET). In addition, pathways containing engineered PHA synthases have proven to be useful for enhanced PHA production with adjustable PHA monomers and contents. The applications of PHA as implant biomaterials are briefly discussed here. In the very near term, metabolic engineering will help solve many problems in promoting PHA as a new type of plastic material for many applications.  相似文献

This article provides an overview of biopolymers, classed according to their chemical structures, function and occurrence, the principles of biosynthesis and metabolism in organisms. It will then focus on polyhydroxyalkanoates (PHA) for which technical applications in several areas are currently considered. PHAs represent a complex class of bacterial polyesters consisting of various hydroxyalkanoic acids that are synthesized by bacteria as storage compounds for energy and carbon if a carbon source is present in excess. Poly(3‐hydroxybutyrate), poly(3HB), is just one example. Most other PHAs are only synthesized if pathways exist which mediate between central intermediates of the metabolism or special precursor substrates on one side and coenzyme A thioesters of hydroxyalkanoic acids, which are the substrates of the PHA synthase catalyzing the polymerization, on the other side. During the last decade, basic and applied research have revealed much knowledge about the biochemical and molecular basis of the enzymatic processes for the synthesis of PHAs in microorganisms. The combination of detailed physiological studies, utilization of the overwhelming information provided by the numerous genome sequencing projects, application of recombinant DNA technology, engineering of metabolic pathways or enzymes and molecular breeding techniques applied to plants have provided new perspectives to produce these technically interesting biopolymers by novel or significantly improved biotechnological processes or by agriculture. Some examples for successful in vivo and in vitro engineering of pathways suitable for the synthesis and biotechnological production of PHAs consisting of medium‐chain‐length 3‐hydroxyalkanoic acids and short‐chain‐length hydroxyalkanoic acids will be provided.  相似文献

The first objective of this study was the measurement of physical properties of P(3HB-co-3HV) copolymers with different (hydroxybutyrate) HB to (hydroxyvalerate) HV ratios produced by Bacillus cereus (TRY2) isolated from activated sludge. The 3HV PHBV copolymers were 0.05, 22.6, 39.2, 54.1, and 69.1 mol%, respectively. The second objective was to study possible wastewater treatment and production of PHAs at the same time by B. cereus (TRY2) and Pseudomonas spp. (TOB17) (both were isolated from activated sludge), recombinant Bacillus DH5α, and a combination of the above three bacteria. The results were satisfactory; the maximum COD and TOC of the sewage sludge reduced were 53.5% and 67.5%, respectively.  相似文献

This study investigates the biosynthesis and characterization of P(3HB-co-3HV-co-3HHx) terpolymer from mixtures of palm kernel oil and 3HV-precursors by using recombinant Cupriavidus necator PHB4/pBBREE32d13. Sodium valerate and propionate have been evaluated for the generation of 3HV monomers. The feeding time of these precursors was a crucial factor that significantly affected the 3HV molar fractions, which ranged from 2 to 60 mol%. Sodium valerate was generally the better precursor in initiating the accumulation of 3HV monomers while maintaining high cell dry weight (7.9 g/L) and good PHA accumulation (79 wt%). However, the 3HHx molar fractions in the terpolymers at 72 h were consistent at about 2-7 mol%. P(3HB-co-3HV) copolymers have been known to exhibit approximately the same degree of crystallinity throughout a wide range of 3HV composition. Interestingly, in this study, terpolymers containing 58 mol% 3HB, 39 mol% 3HV and 3 mol% 3HHx showed elastomeric behavior. This study demonstrates the suitability of palm kernel oil as the main carbon source and both sodium propionate and sodium valerate as 3HV-precursors for the synthesis of novel compositions of P(3HB-co-3HV-co-3HHx) terpolymers with interesting properties.  相似文献

This study describes the biosynthesis and thermal degradation of medium-chain-length polyhydroxyalkanoate (PHA), focusing on 2-alkenoic acids as a recyclable carbon source. Using metabolically engineered Escherichia coli, PHA consisting of 3-hydroxydecanoate (3HD) was synthesized from 2-decenoic acid. Solvent cast film of poly(3HD) [P(3HD)] was transparent and showed thermal property similar to that of polycaprolactone. In addition, the use of various 2-alkenoic acids (C6-C12) resulted in production of PHAs with over 95 mol% of the corresponding single monomer units. The pyrolysis product of P(3HD) was dominantly 2-decenoic acid used for the P(3HD) biosynthesis. This demonstrates the feasibility of PHA recycling via 2-alkenoic acids, which act as pyrolysis products and raw materials for PHA biosynthesis.  相似文献

This study investigated the relationship of growth conditions, host strains and molecular weights of poly[(R)-3-hydroxybutyrate] [P(3HB)] synthesized by genetically engineered Escherichia coli. Various PHA synthases belonging to types I-IV enzymes were expressed in E. coli JM109 under the same experimental conditions, and the molecular weights of the polymers were characterized by gel permeation chromatography. The results demonstrate that P(3HB) polymers have varied molecular weights and polydispersities dependent on the characteristics of the individual PHA synthase employed. P(3HB) with high number-average molecular weights (Mn) [(1.5-4.0) × 106] and narrow polydispersities (1.6-1.8) were synthesized by PHA synthases from Ralstonia eutropha (type I), Delftia acidovorans (type I) and Allochromatium vinosum (type III). Contrary to these, P(3HB) with relatively low Mn [(0.17-0.79) × 106] and broad polydispersities (2.2-9.0) were synthesized by PHA synthases from Aeromonas caviae (type I), Pseudomonas sp. 61-3 (type II) and Bacillus sp. INT005 (type IV). Furthermore, the molecular weights of P(3HB) synthesized under various culture conditions, in various hosts of E. coli and by mutants of PHA synthase were characterized. It was found that, in addition to culture pH [Kusaka et al. Appl Microbiol Biotechnol 1997;47:140], other variances such as culture temperature, host strain and use of mutants are effective in changing polymer molecular weight.  相似文献

Enzymatic degradation of poly(3-hydroxybutyrate-co-3-hydroxyalkanoates) (PHBA) biopolyester consisting of 3-hydroxybutyrate (HB) and 15 mol% medium-chain-length 3-hydroxyalkanoates (HA) was studied using a polyhydroxyalkanoates (PHA) depolymerase produced by Ralstonia pickettii T1. It was found that PHBA films did not lose their weight after 25 h of depolymerase treatment. In contrast, three commercially available PHAs including poly-3-hydroxybutyrate (PHB), poly(3-hydroxybutyrate-19 mol% 3-hydroxyvalerate) (PHBV) and poly(3-hydroxybutyrate-19 mol% 3-hydroxyhexanoate) (PHBHHx) lost 75%, 94% and 39% of their original weights. Slow degradation of PHBA was also confirmed by the absence of HA monomers, dimers or trimers as degradation products in their depolymerase solution compared with abundance of degradation products released by the other three PHAs under the same condition. Surface erosion of PHBA was only observed after 48 h of enzymatic treatment compared with those of PHB, PHBV and PHBHHx which already had obvious surface changes after 7.5 h of same treatment. Although the crystallinities of PHB, PHBV, PHBHHx and PHBA were in the order PHB > PHBV > PHBHHx > PHBA valued at 55.8%, 47.8%, 45.9% and 40.9%, respectively, the order of degradability was PHBV > PHB > PHBHHx > PHBA. It can be proposed that PHA enzymatic degradation using this depolymerase was structure related: longer side-chain PHA including PHBHHx and PHBA was less favorable for the depolymerase degradation, longer the side chain, less the biodegradation.  相似文献

Morphologic and physiologic changes taking place in carbon-limited submerged cultures of Aspergillus nidulans ΔflbA and fadA G203R strains were studied. Loss-of-function mutation of the flbA gene resulted in an altered germination with unusually thick germination tubes, “fluffy” pellet morphology, as well as a reduced fragmentation rate of hyphae during autolysis. In the fadA G203R mutant strain, conidiophores formed in the stationary phase of growth, and the size of pellets shrank considerably. There were no significant differences in the generation of reactive oxygen species (ROS) and in the specific catalase and superoxide dismutase activities by the tested mutants and the appropriate parental strains. Therefore, the participation of ROS or antioxidative enzymes in FadA/FlbA signaling pathways seems to be unlikely in submerged cultures. On the other hand, earlier increases in the extracellular protease and ammonia production were recorded with the ΔflbA strain, whereas the protease and ammonia production of the fadA G203R mutant lagged behind those of the wild-type strains. Similar changes in the time courses of the induction of γ-glutamyltranspeptidase and the degradation of glutathione were observed. These results suggest that FadA/FlbA signaling may be involved in the mobilization of protein and peptide reserves as energy sources during carbon starvation.  相似文献

Polyhydroxyalkanoates (PHAs) are polymers of hydroxyalkanoate, which are accumulated by many bacteria as food storage material under excess carbon source and limited nitrogen source. In our study, Enterobacter cloacae SU-1 isolated from the rhizospheric soil of Arachis hypogea was allowed to grow as batch culture in minimal media containing either glucose or lactose, and the pattern of PHA accumulation by E. cloacae SU-1 was studied. E. cloacae SU-1 was found to accumulate 94% of PHA/dry weight of the organism in 8 g/l lactose-containing medium. When the monomeric units of PHA of E. cloacae SU-1 was analyzed by gas chromatography, it was also found to accumulate medium chain length PHA 3-hydroxyoctanoate (3HO)/3-hydroxyhexanoate (3HH) in the presence of glucose and lactose, but the ratio of these monomers differed as 11:1 and 6:1, respectively.  相似文献

Cloning and expression of L-asparaginase gene in Escherichia coli   总被引:1,自引:0,他引:1  
The L-asparaginase (ASN) from Escherichia coli AS1.357 was cloned as a DNA fragment generated using polymerase chain reaction technology and primers derived from conserved regions of published ASN gene sequences. Recombinant plasmid pASN containing ASN gene and expression vector pBV220 was transformed in different E. coli host strains. The activity and expression level of ASN in the engineering strains could reach 228 IU/mL of culture fluid and about 50% of the total soluble cell protein respectively, more than 40-fold the enzyme activity of the wild strain. The recombinant plasmid in E. coli AS1.357 remained stable after 72h of cultivation and 5h of heat induction without selective pressure. The ASN gene of E. coli AS1.357 was sequenced and had high homology compared to the reported data.  相似文献

Large scale availability of bacterial polyhydroxyalkanoates (PHAs) is still limited to a few types of short-chain-length PHAs, namely poly(3-hydroxybutyrate) (PHB) and its copolymer Biopol™, consisting of 3-hydroxybutyrate and 3-hydroxyvalerate repeating units. In order to increase the number of available medium-chain-length PHA (mcl-PHA) copolymers a flexible high-cell-density fed-batch process was developed. Continuous process monitoring and substrate control were achieved by coupling on-line gaschromatography (on-line GC) to a software-based Proportional Integral (PI) substrate controller. System development time and continuous system upgrading were considerably shortened by using LABView™, a powerful graphical programming environment. The control of octanoic acid and 10-undecenoic acid at 1.5 and 0.5 gL−1 respectively, enabled the production of high levels of biomass (30 gL−1) and mcl-PHA (10.5 gL−1) by avoiding substrate limitations or toxicities. The resulting mcl-PHA was an amorphous copolyester consisting of 37 mol% unsaturated monomers. The present system represents a valuable tool for the production of tailor-made mcl-PHAs, where the desired monomer composition is determined by the ratio of added cosubstrates.  相似文献

Poly(3-hydroxybutyrate-co-3-hydroxyhexanoate) (PHBHHx), a copolyester consisting of 3-hydroxybutyrate (3HB) and 3-hydroxyhexanoate (3HHx), can be synthesized by Aeromonas hydrophila strain 4AK4 using long chain fatty acids as the carbon source. The wild type A. hydrophila 4AK4 accumulated PHBHHx consisting of 12-15 mol% 3HHx regardless of growth conditions. When phbA, phbB and vgb genes encoding β-ketothiolase, acetoacetyl-CoA reductase and vitreoscilla hemoglobin, respectively, were introduced together into A. hydrophila 4AK4, the recombinant strain grew to over 20 g/L cell dry weight (CDW) after 48 h of shake flask cultivation in co-substrates of dodecanoate and gluconate (weight ratio 1:1), and the CDW contained 50% PHBHHx consisting of 9 mol% 3HHx. Under similar conditions, the wild type strain produced only 12 g/L CDW containing 32% PHBHHx with 15 mol% 3HHx. In comparison, recombinant harboring phbA and phbB produced 35% PHBHHx with 9 mol% 3HHx in 15 g/L CDW under the same conditions. The obvious differences in terms of the cell growth and PHBHHx production were attributed to the expression of vgb in A. hydrophila 4AK4, which was clearly observed in carbon monoxide difference spectra. The expression of vgb in the recombinant not only improved cell growth and PHBHHx accumulation, but also increased the plasmid stability during cell growth, especially under low dissolved oxygen tension in fermentors. PHBHHx production could be further increased to over 60% of the CDW by the over expression of phaC and phaJ from Aeromonas caviae encoding PHBHHx synthase and (R)-specific enoyl-CoA hydratase, respectively. Over expression of phaC, phaJ and phaP, alone or in various combinations, also increased the 3HHx content of PHBHHx from 14-34%. The above results showed that A. hydrophila was amenable to genetic manipulation, and that these modifications could be exploited to produce compounds with different properties for commercial and research applications.  相似文献

Summary : Haloferax mediterranei was investigated for the production of two different high-performance polyhydroxyalkanoates (PHAs). A copolyester containing 6 mol-% 3-hydroxyvalerate (3HV) was produced from whey sugars as sole carbon source. The maximum specific growth rate (µmax.) and the maximum specific PHA production rate (qp max.) were determined with 0.10 1/h and 0.15 1/h, respectively. The cells contained 72.8 wt.-% of P-(3HB-co-6%-3HV) which featured low melting points between 150 and 160 °C and narrow molecular mass distribution (polydispersity PDI = 1.5). Further, a PHA terpolyester with an increased 3HV fraction as well as 4-hydroxybutyrate (4HB) building blocks was accumulated by feeding of whey sugars plus 3HV - and 4HB precursors. Kinetic analysis of the process reveals a µmax. of 0.14 1/h and a qp max. of 0.23 1/h, respectively. The final percentage of P-(3HB-co-21.8%-3HV-co-5.1%-4HB) in biomass amounted to 87.5 wt.-%. Also this material showed a narrow molecular mass distribution (PDI = 1.5) and a high difference between the two melting endotherms of the material (between 140 and 150 °C) and the onset of decomposition at 236 °C. The accomplished work provides viable strategies to obtain different high-quality PHAs which might be potential candidates for application in the medical and pharmaceutical field.  相似文献

Polyhydroxyalkanoate (PHA) copolymers consisting of (R)-3-hydroxybutyrate (3HB) and medium-chain-length (R)-3-hydroxyalkanoate (3HA), P(3HB-co-3HA), are usually solved in chloroform. However, we found that some of the P(3HB-co-3HA) aged for more than 1 month under ambient conditions were not solved in chloroform, but instead swelled when the 3HA fraction was over 14 mol%. On the basis of differential scanning calorimetry and wide-angle x-ray diffraction analyses, we predicted that swellable P(3HB-co-3HA) contained numerous P(3HB) microcrystals, which may form physical crosslinks between adjacent PHA polymer chains.  相似文献

Unlike polyhydroxyalkanoates (PHAs) copolymers, the controlled and efficient synthesis of PHA terpolymers from triglycerides and fatty acids are yet to be established. This study demonstrates the production of P(3HB-co-3HV-co-3HHx) terpolymer with a wide range of 3HV monomer compositions from mixtures of crude palm kernel oil and 3HV precursors using a mutant Cupriavidus necator PHB4 transformant harboring the PHA synthase gene (phaC) of a locally isolated Chromobacterium sp. USM2. The PHA synthase of Chromobacterium has an unusually high affinity towards 3HV monomer. P(3HB-co-3HV-co-3HHx) terpolymers with 3HV monomer composition ranging from 2 to 91 mol% were produced. Generation of 3HHx monomers was affected by the concentration and feeding time of 3HV precursor. P(3HB-co-24 mol% 3HV-co-7 mol% 3HHx) exhibited mechanical properties similar to that of common low-density polyethylene. P(3HB-co-3HV-co-3HHx) terpolymers with a wide range of 3HV molar fraction had been successfully synthesized by adding lower concentrations of 3HV precursors and using a PHA synthase with high affinity towards 3HV monomer.  相似文献
