The aldol reaction of tetrahydro-4H-thiopyranone with 1,4-dioxa-8-thiaspiro[4.5]decane-6-carboxaldehyde (I) gives four possible diastereomeric adducts (II). Aldol reactions of I with each of the diastereomers of II and their corresponding methoxymethyl ethers III via the Ti enolates were investigated. Using racemic reactants, reactions with II proceeded with high levels of mutual kinetic enantioselection (MKE) and double stereodifferentiation (DS) to give one of the eight possible bisaldol adducts. Similar reactions of III proceeded with low levels of MKE and DS and gave two bisaldol adducts, one from each of the possible combinations of enantiomeric reactants. By extension, 11 of the 20 possible diastereomers of the bisaldol adduct could be obtained selectively. These adducts are useful for polypropionate synthesis.  相似文献

[reaction: see text] Reported herein is a strategy employing a Mukaiyama reaction in tandem with a hydrogen transfer reaction for the elaboration of 2,3-anti-3,4-anti and 2,3-anti-3,4-syn propionate motifs. The mode of complexation is controlled through monodentate or chelate pathways for the Mukaiyama reaction to give access to either syn or anti aldol products, precursors of the free-radical reduction reaction. Boron Lewis acid is used to control the free-radical reaction through the exocyclic pathway.  相似文献

Reactions of 4-Bromo-1,6-methano[10]annulene-3-carbaldehyde: Syntheses of 3,4-Heteroanellated 1,6-Methano[10]annulenes Similarly to β-chlorovinyl ketones, the title compound 1 reacts with dinucleophiles to give the 3,4-heteroanellated 1,6-methano[10]annulenes 2–4, 6 , and 12–16 , containing thiophene, pyrazole, pyrimidine, and thiazepine rings (Schemes 2 and 3). On reaction of 1 with tert-butylhydrazin, an unusual Michael-addition-dehydrogenation sequence, resulting in 2,3-anellation of a pyrazole ring (→ 5 ), is observed. Synthetic as well as some mechanistic aspects are briefly discussed.  相似文献

New pyrrolo[3,4-c]isoxazole derivatives were synthesized from the key intermediates 4-cyanopyrrolidin-3-ones in two steps. Pyrrolo[2,3-d][1,2,3]triazoles and triazolo[4,5-c]pyridazine were obtained from 2-arylhydrazono-4-cyano-1-(4′-methoxyphenyl)-3-oxopyrrolidines by refluxing with phenylhydrazine in either ethanol or glacial acetic acid. Cyclization of thioglycolic acids derivatives 3a-d gave isoindolo[1,2-b]thieno[2,3(3,2 or 3,4)-e][1,3]-thiazocines 4a-d . Isoindolo[2,1-a]thieno[2,3(3,2 or 3,4)-f][1,4] or [1,5]diazocines 10b or 11a-c were synthesized from Beckmann or Schmidt rearrangement of the ketones 7a-c .  相似文献

4-Aryl-1-(4-oxo-3,4-dihydrothieno[2,3-d]pyrimidin-2-yl)thiosemicarbazides react with dimethyl acetylenedicarboxylate in methanol to give the corresponding methyl {3-aryl-4-oxo-2-[(4-oxo-3,4-dihydro-thieno[2,3-d]pyrimidin-2-yl)hydrazono]-1,3-thiazolidin-5-ylidene}acetates, whereas in dioxane methyl 5-aryl-amino-2-methoxycarbonylmethyl-3-(4-oxo-3,4-dihydrothieno[2,3-d]pyrimidin-2-yl)-2,3-dihydro-1,3,4-thiadiazole-2-carboxylates are mainly formed.  相似文献

Reactions of aliphatic 2,2,3,3-tetracyanocyclopropyl ketones with aqueous ammonia afforded 4-alkyl- 4-amino-2-oxo-3-azabicyclo[3.1.0]hexane-1,6,6-tricarbonitriles with conservation of the three-membered ring. Reactions of the same compounds with primary amines were accompanied by opening of the cyclopropane ring, and they led to the formation of 5-amino-6,6a-dimethyl-2-oxo-1,2,6,6a-tetrahydropyrrolo[2,3-b]-pyrrole-3,4-dicarbonitriles as a result of successive heterocyclizations.  相似文献

A convenient and efficient three-step route toward both symmetrically and asymmetrically functionalized 4H-cyclopenta[2,1-b:3,4-b']dithiophenes has been developed. Using this method a broad collection of functionalized bridged bithiophenes can smoothly be accessed. Starting from 3-bromo-2,2'-bithiophene, prepared by Kumada coupling of 2-thienylmagnesium bromide with 2,3-dibromothiophene under Pd(dppf)Cl(2) catalysis, lithiation and subsequent reaction with dialkyl ketones afforded (a)symmetrically dialkylated tertiary alcohol derivatives. By means of final Friedel-Crafts dehydration cyclization in sulfuric acid medium, these derivatives were converted to 4,4-dialkyl-4H-cyclopenta[2,1-b:3,4-b']dithiophenes. Upon replacement of the dialkyl ketone reagent by ethyl levulinate, an ester-functionalized 4H-cyclopenta[2,1-b:3,4-b']dithiophene was prepared, representing an attractive precursor for variously functionalized cyclopentadithiophene compounds.  相似文献

-Isonitroso ketones, derivatives of tetrahydrobenzofurazan and furoxan, react with aldehydes (acetaldehyde and propionaldehyde) and morpholine to form derivatives of tetrahydropyrrolo[2,3-e]-2,1,3-benzoxadiazole 6-oxide. Treatment of the latter with hydrazine hydrate gave derivatives of 4,5-dihydro-1,2,5-oxadiazolo[3,4-f]cinnoline which are readily dehydrogenated with tetrachloro-benzoquinone to derivatives of 1,2,5-oxadiazolo[3,4-f]cinnoline. Reduction of tetrahydropyrrolo[2,3-e]-2,1,3-benzoxadiazole 6-oxides with sodium borohydride gave derivatives of N-hydroxyhexahydro-pyrrolo[2,3-e]-2,1,3-benzoxazole.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 10, 1558–1563. The o-diamine, 3,4-diamino-1,2,5-thiadiazole ( 2 ), was synthesized from 3,4-dichloro-1,2,5-thiadiazole ( 3 ) hy three methods. Aqueous glyoxal cyclized 2 into [1,2,5]thiadiazolo[3,4–6]-pyrazine ( 14 ). 3,4-Dichloro-1,2,5-thiadiazole 1,1-dioxide ( 18 ) reaeted with 2 to give 1,3-dihydro-bis[1,2,5]thiadiazolo[3,4-b:3′,4′-e]pyrazine 2,2-dioxide ( 19 ). The reaction of 2 with selenium oxyehloride led to [1,2,5]selenadiazolo[3,4-c] [1,2,5]thiadiazole ( 12 ). Ring closure of 2,3-diaminoquinoxaline ( 4 ) with thionyl chloride or selenium oxychloride gave [1,2,5]thiadiazolo-[3,4-b]quinoxaline ( 21 ) and [1,2,5]selenadiazolo[3,4-b]quinoxaline ( 22 ), respectively. Sulfurous acid reduced 21 to the 4,9-dihydro derivative 23 , which was reoxidized to 21 with chloranil. Aqueous hase hydrolyzed 21 to 4 via the hydrated intermediate 24 . Aqueous glyoxal cyclized 4 to the covalent hydrate of pyrazino[2,3-b]quinoxaline ( 26 ), 27 , which was dehydrated to 26 . Compound 26 underwent rapid addition of two alcohols in a process analogous to covalent hydration.  相似文献

Meglumine, a bio-based chemical, was demonstrated to be a highly efficient and reusable catalyst for the synthesis of a series of pyranopyrazole derivatives via a one-pot, four-component reaction of carbonyl compound or isatin, hydrazine hydrate, malononitrile, and β-keto ester in EtOH–H2O. The catalyst was found to work extremely for aldehydes, ketones or isatins at room temperature to give the corresponding dihydropyrano[2,3-c]pyrazole or spiro[indoline-3,4′-pyrano[2,3-c]pyrazole] derivatives in high yields. The salient features of this new methodology are broad substrate scope, room temperature reaction conditions, short reaction times, high yields, easy work-up process, reusability of catalyst, and the absence of hazardous organic solvents.  相似文献

Thiophene-3,4-dicarbaldehyde 1 reacts in the presence of 2-mercaptoethanol to yield N-aryl-5,6-dihydro-4-oxo-4H-thieno[3,4-c]pyrroles 2 and N-aryl-4-arylimino-5,6-dihydro-4H-thieno[3,4-c]pyrroles 3 , while thiophene 2,3-dicarbaldehyde 4 reacts with aromatic amines to give N-aryl-5,6-dihydro-6-oxo-4H-thieno[2,3-c]pyrroles 5 in good yields. 5‐Amino‐3,4‐diphenylthieno[2,3‐c]pyridazine‐6‐carbonitrile 2a was reacted with propylene diamine to give tetrahydropyrimidinyl derivative 3 . The latter compound ( 3 ) underwent certain cyclocondensation reactions to produce pyrimidothienopyridazines 4–6 . Also, the reactions of 5‐amino‐3,4‐diphenylthieno[2,3‐c]pyridazine‐6‐carboxamide with heterocyclic aldehydes and/or cycloalkanones were carried out and their products were identified. Moreover, some novel pyridazinothienopyrimidobenzimidazoles 14–17 were synthesized.  相似文献

Michael addition of 1,5‐diaryl‐2,3‐dioxopyrrolidine derivatives with α‐cyanocinnamonitriles and ethyl α‐cyanocinnamates afforded 4H‐pyrano[2,3‐c]pyrrole derivatives in the presence of sodium ethoxide. Under the same reaction condition, the ylidenes of 1,5‐diaryl‐2,3‐dioxopyrrolidine were reacted with malononitrile or ethyl cyanoacetate to give isoindole derivatives; however, pyrrolo[3,4‐b]pyridine derivatives were formed when cyanoacetamide was used. Moreover, pyrrolo[3,4‐d]pyrimidine derivatives were synthesized by treating 4‐benzylidene‐1,5‐diphenyl‐2,3‐dioxopyrrolidine with urea and/or thiourea under basic conditions. The structures of all the new synthesized compounds were confirmed by elemental analysis, IR and NMR spectra.  相似文献

The synthesis of two new heterocycles is described: pyrido-[2,3-d]-.s-triazolo[ 3,4-f] pyrimidine and pyrido[3,2-d]-.s-triayzolo-[3,4-f] pyrimidine. 4-[I'-Pyrazolyl]pyrido[2,3-d]pyrimidines and 4-[1′-pyrazoly1] pyrido[ 3,2-d] pyrimidine are obtained by the action of 4-hydrazinopyrido[2,3-d]pyrimidine and 4-hydrazinopyrido-[3,2-d]pyrimidine with several β-diketones.  相似文献

Reaction of 2,2,3,3-tetracyanocyclopropyl ketones with water solution of sodium hydroxide after neutralization with sulfuric acid leads to the formation of 4-amino-1-hydroxy-3,6-dioxo-2,3,5,6-tetrahydro-1Hpyrrolo[3,4-c]pyridine-7-carbonitriles. Pivaloyltetracyanocyclopropane reacts in another way and is converted into sodium 6a-tert-butyl-3,4-dicyano-5-oxo-1,5,6,6a-tetrahydropyrrolo[2,3-b]pyrrol-2-olate. 1-Benzoyl-1-methylcyclopropane-2,2,3,3-tetracarbonitrile reacts with the sodium hydroxide with the retention of the threemembered ring and the formation of 11-methyl-4-phenyl-3,5,9-triazatetracyclo[5.3.1.01,7.04,11]undecane-2,6,8,10-tetraone.  相似文献

6‐(2‐Thienyl)‐4‐(trifluoromethyl)‐1H‐pyrazolo[3,4‐b]pyridine‐3‐amine reacted with different active methylene compounds to afford pyridopyrazolopyrimidine derivatives. On the other hand, it reacted with some halo compounds to give the imidazo[1′,2′:1,5]pyrazolo[3,4‐b]pyridine derivatives. Also, it diazotized to give the corresponding diazonium chloride that is coupled with several active methylene compounds to give the corresponding triazine derivatives. Furthermore, compound 3‐amino‐6‐(2(thienyl)‐4‐(trifluoromethyl)thieno[2,3‐b]pyridine‐2‐carbohydrazide reacted with some β‐dicarbonyl compounds and some sulfur‐containing compounds to afford the corresponding pyrazolyl oxadiazolylthieno[2,3‐b]pyridine derivatives.  相似文献

A one‐pot and convenient synthesis of multisubstituted pyrazolo[3,4‐b]pyridines in moderate to high yields has been achieved by a two‐step reaction: diazo‐transfer of 5‐azido‐1‐phenylpyrazole‐4‐carboxaldehydes to ketones in ethanolic KOH to give 5‐amino‐1‐phenylpyrazole‐4‐carboxaldehyde and subsequent Friedlaender reaction of 5‐amino‐1‐phenylpyrazole‐4‐carboxaldehyde with ketones.  相似文献

Two different convenient methods have been developed for the synthesis of condensed thieno[2,3-b]pyridines. The intramolecular cyclization of 3-amino-2-hydrazinocarbonyl-7,7-dimethyl-7,8-dihydo-5H-pyrano[3,4-e]-thieno[2,3-b]pyridine was carried out to give 7,7-dimethyl-2-oxo-1,2,6,7-tetrahydro-9H-pyrano[3′,4′-e]-imidazo[4″,5″∶2,3]thieno[5,4-b]pyridine, which is the first representative of a new heterocyclic system.  相似文献
