The orientation of the cycloaddition of diazomethane on unsaturated branchedchain sugars has been studied. For 3-C-cyanomethylidene-3-deoxy-1,2-O-isopropylidene-α-D-glycero-tetrofuranose the orientation was ‘normal’ and did not depend on the configuration at the double bond. The same situation prevailed with derivatives of 3-deoxy-1,2:5,6-di-O-isopropylidene-3-C-methylidene-α-D-xylo-hexofuranose. For the 3-C-acylmethylidene- and the 3-C-cyanomethylidene-3-deoxy-1,2:5,6-di-O-isopropylidene-α-D-ribo-hexofuranoses, the trans-(H–C(3′)–C(2))-isomer gave the ‘normal’ cycloadduct whereas the cis-isomer gave predominantly the αabnormal spiro-pyrazoline. This observation represents the first instance where the regioselectivity of a cycloaddition reaction is affected by the geometrical isomerism of the dipolarophile. The most probable explanation of the phenomenon is the conformational perturbation about the C(4)--C(5) bond of the unsaturated sugars induced by a change in the configuration at C(3). The consequence of that ‘conformational transmission’ of a difference in configuration at C(3) is that the steric crowding on the cis- than in the trans-isomer. Several novel examples of a new series of C-glycosylidenic derivatives, the spiro-pyrazolines, are described.  相似文献

The stereoisomers (3 and 4) of O-ethyl O-phenyl O-(1-methyl-2-ethoxycarbonyl) vinyl phosphorothionate have been synthesized by the reaction of optically active O-ethyl O-phenyl phosphorothiochloride 2 with ethyl acetoacetate under different conditions. 3 (100% Z-isomer, determined by 1H NMR) was synthesized by the reaction of 2 with ethyl sodio-acetoacetate in the mixed solvent of 1:3 toluene-dioxane at 50°C. 4 (>95% E-isomer) was obtained by the reaction of 2 with ethyl acetoacetate in presence of t-BuOK in DMSO at 15°C. 100% E-isomer 4 was separated from crude 4(>95% E-isomer) by column chromatography on silica gel (petroleum ether-ether 6:1). By this reaction either Z- or E-isomers were formed with inversion of the configuration at phosphorus atom. Thus, six stereoisomers of 3 and 4 which were prepared from 2 (RS, S, R) by the above method namely (RS)-Z, (R)-Z, (S)-Z and (RS)-E, (R)-E, (S)-E.  相似文献

Reactions of stabilized ylides with aldehydosugars: influence of some factors, particularly the structure of the aldehydosugar, on the ratio of the geometrical isomers formed Aldehydosugars bearing no cis-substituent on the C-atom β to the carbonyl group ( 2, 3, 6, 7 ) or bearing a substituent without an electron lone-pair ( 5 ) gave, upon treatment with acetylmethylidenetriphenylphosphorane, exclusively the E-isomer, this representing the classical behaviour of aldehydes. On the other hand, aldehydosugars having a lone-pair bearing substituent ( 1, 4, 8, 9 ) gave a mixture of E- and Z-isomers. In the case of most of the aldehydosugars of the latter type (giving some Z-isomer) a partial epimerization of the C-atom α to the carbonyl group took place when the solvent of the Wittig reaction was HCONMe2. It is probable that the presence of an electron lone-pair in the adequate position inhibits the reversion of the kinetic erythro-betaine, allowing the formation of the Z-isomer and, in particular solvent conditions, a cycloelimination leading to the epimeric aldehydosugar which ultimately reacts to give the epimeric E-enone.  相似文献

Reactions of Some 3-Deoxy-3-trifluoroacetamido-D -ribofuranosyl Halides with Mercuric Cyanide The expected 2,5-anhydro-D -allononitrile 11 was obtained when the 2-O-benzoyl-3-deoxy-3-trifluoroacetamido-D -ribofuranosyl bromide reacted with mercuric cyanide, whereas the isomeric 1,2-O (1-cyanoethylidene) derivative was formed from the 2-O-acetylated halide; no reaction occurred when the halide was 2-O-p-nitrobenzoylated.  相似文献

Novel types of acetylenic sugars The coupling, following Cadiot's procedure, of a 6-bromo-5,6-dideoxy-1,2-O-isopropylidène-3-O-methyl-α-D -xylo-hex-5-yno-1, 4-furanose (1) with phenylacetylene, 2-propyn-1-ol or terminal acetylenic sugars gave with excellent yields the expected diynes (an enediyne when the terminal acetylene was the 3,5, 6-trideoxy-1,2-O-isopropylidene-α-D -glycero-hex-3-en-5-yno-1,4-furanose 7 ). The chloro analogue 8 of 1 on treatment with lithium thiophenate gave the corresponding phenylthio-acetylenic sugar 9 . An acetylene was also formed by reacting the gem-difluoro-olefinic sugar 10 with butyllithium whereas the same olefinic sugar and its 3-O-benzyl analogue 11 gave only a gem-fluoro-arylthio-olefinic sugar (13–15) as a mixture of the Z and E isomers (Z/E > 4) when treated with the conjugate base of an arylmercaptan.  相似文献

Reaction of 2-formyl-2-(2,3-O-isopropylidene-5-O-trityl-D-ribofuranosyl)acetonitrile (VII) with semicarbazide hydrochloride followed by sodium ethoxide treatment afforded an α,β-mixture of 3-amino-2N-carbamoyl-4-(2,3-O-isopropylidene-5-O-trityl-D-ribofuranosyl)pyrazole (IX). Conversion of IX to 4-oxo-8-(2,3-O-isopropylidene-5-O-trityl-D-ribofuranosyl)-3H-pyrazolo[1,5-a]-1,3,5-triazine (XIII) was achieved by treatment of IX with ethylorthoformate. The β-isomer IXb gave only the β-isomer XIIIb, and the α-isomer IXa was converted exclusively into the α-isomer XIIIa. Upon deprotection with 3% n-butanolic hydrogen chloride, both IXa and IXb gave the same mixture of the α- and β-isomers of 3-amino-2N-carbamoyl-4-(D-ribosyl)pyrazole, which were separated by chromatography. The syntheses of the hitherto unknown compounds, 3-amino-2N-carbamoylpyrazole (IVa) and its 4-methyl analog (IVb) are also reported. Experimental details of the synthesis of 3-amino-4-(2,3-O-isopropylidene-5-O-trityl-β-D-ribofuranosyl)pyrazole (XIIb), an important intermediate for “purine-like” C-nucleosides, are also described.  相似文献

Search for Antiviral Nucleosides Analogues In contrast to the reported impossibility of obtaining the expected products when 1-(2,3-O-isopropylidene-1,5-dialdoribofurannos-1-yl)uracile ( 2 ) was treated with stabilized ylides, we prepared in good to excellent yields the modified nucleosides 4–9 by reaction of 2 respectively with acetylmethylidene-, benzoylmethylidene-, ethoxymethylidene-, methoxymethylidene-, succinimidylidene- or N-phenylsuccinimidylidene-triphenylphosphorane. Compounds 4–10 were tested against the herpes virus type 1 (HF) and influenza virus A2/Scotland. The modified nucleoside 9 showed a limited but significative activity against the herpes virus.  相似文献

Identification and synthesis of new depsides isolated from oakmoss (Evernia Prunastri (L .) ACH .) Lecanoric acid 5 and 4 new depsides have been isolated from oakmoss (Evernia Prunastri (L .) ACH .) extracts by means of silicagel column chromatography. Various spectral methods including 13C-NMR. were used in determining the structure of 4,2″-O-methylgyrophoric acid ( 1 ), lecanoric acid ( 5 ), 2′-O-methylevernic acid ( 2 ), 3′-methylevernic acid ( 3 ) and methyl 3′-methyllecanorate ( 4 ).  相似文献

Thermal decomposition of azido-1,2-dithienylethenes 1 gave thienyl 4H-thieno[3,2-b]pyrroles 3 . For the 3,4-disubstituted thiophene derivatives 2 , the same reaction led to the amino-1,2-dithienylethenes 4 . In contrast, only azido-1,2-dithienylethanes 7 led to thienyl-5,6-dihydro-4H-thieno[3,4-b]pyrroles 8 . The structure of the obtained derivatives was established on the basis of 1H nmr, ir, and mass spectral data.  相似文献

Phytochemistry of genus Gentiana. XXII. Identification of new O-glucosides of mangiferin in Gentiana asclepiadea L. Two new O-glucosides of mangiferin (mangiferin-7-O-β-D -glucoside ( 1 ) and mangiferin-6-O-β-D -glucoside ( 2 )) have been isolated from the leaves of Gentiana asclepiadea L . This is the first full structure elucidation of naturally occurring O-glucosides of C-glucosylxanthones. The known C-glucosylflavone, saponarin ( 5 ) was also identified.  相似文献

C -Glycosylic derivatives XXXII. Synthesis of spiro-C -glycosylidenic derivatives via nucleophilic cyclization. On treatment with compounds bearing two nucleophilic groups as ethylenediamine, o-phenylenediamine or their monooxa or monothia analogues, 1,2:5, 6-di-O-isopropylidene-α-D -ribo-hexofuranos-3-ulose gave with excellent yields the corresponding spiro-C-glycosylidenic derivative; for example, when using o-phenylenediamine, a spirobenzimidazoline ( 5 ) was obtained. The latter compound underwent, on oxidation, a ring expansion to a morpholinobenzimidazole ( 8 ). Spirobenzodiazepines, spirobenzooxazepines and spirobenzothiazepines were formed when applying the same type of cyclization reaction to 3-C-acetylmethylene-3-deoxy-1,2:5,6-di-O-isopropylidene-α-D -ribo- and α-D -xylo-hexofuranoses.  相似文献

Some sugar phosphates, phosphonates and phosphine oxides, Preliminary communication Some new phosphates of 1, 2-O-isopropylidene-α-D-ribofuranose have been prepared. Reactions of 3-C-cyanomethylidene-3-deoxy-1, 2-O-isopropylidene-5-O-trityl-α-D -erythro-pentofuranose (9) have been studied. On treatment with phosphorus nucleophiles, 9 led to phosphorus bearing branched-chain sugar derivatives. Branched-chain cyanosugars as 15 and 16 prepared by cis-vic-dihydroxylation of 9 constitute interesting potential precursors of new types of cyclic sugar phosphates.  相似文献

The reaction of the trimethylsilyl derivative of 4,6-dichloroimidazo[4,5-c]pyridine with 2,3,5-tri-O-benzoyl- D -ribofuranosyl bromide gave four nucleosides-the α- and β-anomers of the 1-isomer and the α- and β-anomers of the 3-isomer (3.9:2.7:1.5:1). In contrast, the fusion reaction of 4,6-dichloroimidazo[4,5-c ]pyridine with 1,2,3,5-tetra-O-acetyl-β- D -ribofuranose gave a high yield of the 1-β-isomer, which was converted to the known 3-deazaadenosine (4-amino-l-β- D -ribofuranosylimidazo[4,5-c]pyridine).  相似文献

S-Methylation of 6-S-benzyl-6-deoxy-1,2-O-isopropylidene-3-O-methyl-α-D-xylo-6-thiohexofuranos-5-ulose ( 1 ) gave the expected sulfonium salt 2 which on alcaline treatment yielded the stable sulfur ylide 3 . This compound constitutes an useful synthetic intermediate in carbohydrate chemistry. On heating in 1,2-dimethoxyethane, it underwent a Stevens rearrangement which led to an extension of the carbon chain of the sugar and, reacted with Michael acceptors, it gave cyclopropanation reactions.  相似文献

Reaction of 2-t-butylthiobenzaldehyde with N-(2-dialkylaminoethyl)-cyanacetamides followed by heating of the resulting 3-(2-t-butylthiophenyl)-2-cyanoacrylamides in polyphosphoric acid leads to the formation of 3-carboxamido-2-imino-2H-benzo[b][l]thiopyrann intermediates which afford thiocoumarins on hydrolysis. A re-examination of the reaction of Fischer's base on tetracyanoethylene Depending on the order of addition, Fischer's base 5 (1,3,3-trimethyl-2-methylidene-indoline) reacts 1:1 with tetracyanoethylene to give either the tricyanovinylation product 6 or the spiro compound 7 . A skeletal rearrangement of a zwitterionic intermediate can explain the formation of the spiro compound. The latter undergoes a thermal isomerization yielding by ring expansion the tetrahydroquinoléine 8 . On reaction with LiAlH4 or CH3ONa 7 and 8 lead both to triazatetracycles. A comparatie kinetic study of nucleophilic substitution of some condensted 2-chloropyridines by piperidine was carried out and an “autocatalytic effect” was observed in the case of 1-methyl-4-chloro-5-azaindole. The rate constants of such substitutions were determined for 1-methyl-4-chloro-5-azaindole, 4-chlorofuro[3,2-c]pyridine, 1-phenyl- and 1-benzy;(1H)pyrazolo[4,3-c]pyridines, 4-chloro-6,7-dimethylpyrrolo[2,3-d]pyrimidine, and a classification of reactivity of these compounds, as compared to 2-chloropyridine, was established.  相似文献

Under basic conditions. [1]benzopyrano[2,3-b]quioxakin-12-one leads to 3-(i-hydroxybenzoy1)-1H-quinoxalin-2-one. This ketone reacts with hydroxylamine and phenylhydrazine to give the expected derivatives or those of [1]benzopyrano92,3-b]quinoxalin-12-one. The reduction of [1]benzopyrano[2,3-b]quinoxalin-12-one was unsucessful by chemical means. However, electrochemical reduction leads to a dihydropyrazine nucleus.  相似文献

A Novel Example of Unsaturated Branched-chain Sugar Nucleoside: 3′-Deoxy-3′-methylidene-adenosine Starting from 5-O-benzoyl-3-C-methylidene-3-deoxy-1,2-O-isopropylidene-α-D -erythro-pentofuranose ( 11 ) the title compound 8 has been prepared. Its α-anomer ( 9 ) and the acyclic sugar nucleoside 10 have been obtained as by-products. Adenosine deaminase slowly deaminated 8, 9 being not affected. Compound 8 exhibited no antiviral activity, whereas one of its saturated analogues ( 13 ) inhibited the multiplication of the herpes-1 (HF) virus.  相似文献

Conformational analysis using 1H n.m.r. data (δ, 3J and NOE) has been carried out on several derivatives of 2′,3′,-O-isopropylideneadenosine bearing various substituents at positions C-5′, C-8 and N-6.
