A novel organic gel film modified electrode was simply and conveniently fabricated by casting LixMoOy and polypropylene carbonate (PPC) onto the surface of a gold electrode. The cyclic voltammetry and amperometry studies demonstrated that the LixMoOy film modified electrode has a high stability and a good electrocatalytic activity for the reduction of iodate. In amperometry, a good linear relationship between the steady current and the concentration of iodate was obtained in the range from 3×10–7 to 1×10–4 mol L–1 with a correlation coefficient of 0.9997 and a detection limit of 1×10–7 mol L–1.  相似文献

Group-theoretical and quantum-chemical investigations of the spectrum of low-lying excited states have been performed by the ROHF and FCI-RAS (Full CI in Restricted Active Space) methods for 3d metal endofullerenes (MEFs) M@C60 (M =Mn, Cr, and Fe) in different charged states. The major purpose of this study is quantum-chemical verification of the anomalous (“non-Bethe’s”) character of splitting of the d N atomic states in an electrostatic field with icosahedral symmetry, predicted previously within the theory of integral invariants theory. The interrelation between the integral invariants theory and the quantumchemical methods applied in this work is considered in detail. Our calculations suggest that the d N atomic states in the icosahedral field generated by fullerene C60 (I h ) on a metal atom (ion) remain non-split for different charged states of the metal and C60. Reasons for this phenomenon and other possible approaches to verification of the prediction are discussed. It is demonstrated that the d N states of the encapsulated metal are split in icosahedral 3d MEFs only under very strong compression of these structures.  相似文献

The polymerization of o-phenylenediamine (OPD) on l-tyrosine (Tyr) functionalized glassy carbon electrode (GCE) and its electro-catalytic oxidation towards ascorbic acid (AA) had been studied in this report. l-Tyrosine was first covalently grafted on GCE surface via electrochemical oxidation, which was followed by the electrochemical polymerization of OPD on the l-tyrosine functionalized GCE. Then, the poly(o-phenylenediamine)/l-tyrosine composite film modified GCE (POPD-Tyr/GCE) was obtained. X-ray photo-electron spectroscopy (XPS), field emission scanning electron microscope (SEM), and electrochemical techniques have been used to characterize the grafting of l-tyrosine and the polymerization and morphology of OPD film on GCE surface. Due to the doping of the carboxylic functionalities in l-tyrosine molecules, the POPD film showed good redox activity in neutral medium, and thus, the POPD-Tyr/GCE exhibited excellent electrocatalytic response to AA in 0.1 mol l−1 phosphate buffer solution (PBS, pH 6.8). The anode peak potential of AA shifted from 0.58 V at GCE to 0.35 V at POPD-Tyr/GCE with a greatly enhanced current response. A linear calibration graph was obtained over the AA concentration range of 2.5 × 10−4–1.5 × 10–3 mol l−1 with a correlation coefficient of 0.9998. The detection limit (3δ) for AA was 9.2 × 10−5 mol l−1. The modified electrode showed good stability and reproducibility and had been used for the determination of AA content in vitamin C tablet with satisfactory results.  相似文献

We have developed new DNA extraction and purification procedures for investigation of mycorrhized seedlings and canned truffles. Use of these procedures on approximately 100 mg initial material enabled good sample representation. For mycorrhized seedlings, Taq polymerase inhibitors were discarded irrespective of tree species. In routine analysis we systematically used consensus primers ITS1/ITS4 to check the absence of Taq polymerase inhibitors and the presence of fungus DNA. Positive response with ITS validates other positive or negative PCR results. Absence of amplification with ITS prevents validation of other results. For canned truffles, DNA harvested from ascocarps sterilized for one and a half hours at 115°C was amplified with specific primers. We have developed consensus primers, named R12/F12, to check for the presence of amplifiable fungus DNA and the absence of Taq polymerase inhibitors. Here also, positive response with consensus R12/F12 validates other positive or negative PCR results. We have developed one primer pair specific for T. brumale and another specific for T. melanosporum. We can then characterize these two taxa, which enables the use of truffle or truffled French designations. We can also characterize T. indicum, the Asiatic black truffle that might fraudulently be sold as T. melanosporum and T. brumale. These three specific primer pairs were used independently of DNA extraction from tree seedlings or canned truffles. Our process is specific, sensitive, convenient, and quick.J.P. Douet and D. The regioselective reaction of N1-benzyl-N2-(4-nitrophenyl)ethanediamide with dialkyl acetylenedicarboxylates or alkyl propiolates in the presence of triphenylphosphine leads to dialkyl 4-benzylamino-1-(4-nitrophenyl)-5-oxo-2,5-dihydro-1H-pyrrole-2,3-dicarboxylates or alkyl 4-benzylamino-1-(4-nitrophenyl)-2-oxo-5-pyrrolidinecarboxylates in good yields.  相似文献

The electrochemical properties of n-sulfonatothiacalyx[4]arene (H4XNa4) complexes with [Co(dipy)3]3+ and Fe3+ ions were studied by means of cyclic voltammetry in aqueous solution at pH 2.5. The observed single-electron reduction of [Co(dipy)3]3+ bound extraspherically to the upper rim and Fe3+ ion bound intraspherically to the lower rim of n-sulfonatothiacalyx[4]arene in binary [Co(dipy)3]3+ · H3X5?, H3X5? · Fe3+, and ternary [Co(dipy)3]3+ · H3X5? · Fe3+ heterometal complexes was more difficult than in the free state. The reversible single-electron transfer to the metal ion results in lower binding energy ([Co(dipy)3]3+, ΔΔG 0 = 3.9 kJ/mol) or in full fast dissociation of the complex (Fe3+). The ternary complex in the solution forms the aggregates, in which inner encapsulated Fe(III) and Co(III) ions are not reduced on the electrode. Their quantitative reduction takes place by the relay mechanism of intra- and intermolecular electron transfer through electrochemically generated [Co(dipy)3]2+ outer ions.  相似文献

A simple membrane-strip-based biosensor assay has been combined with a nucleic acid sequence-based amplification (NASBA) reaction for rapid (4 h) detection of a small number (ten) of viable B. anthracis spores. The biosensor is based on identification of a unique mRNA sequence from one of the anthrax toxin genes, the protective antigen (pag), encoded on the toxin plasmid, pXO1, and thus provides high specificity toward B. anthracis. Previously, the anthrax toxins activator (atxA) mRNA had been used in our laboratory for the development of a biosensor for the detection of a single B. anthracis spore within 12 h. Changing the target sequence to the pag mRNA provided the ability to shorten the overall assay time significantly. The vaccine strain of B. anthracis (Sterne strain) was used in all experiments. A 500-L sample containing as few as ten spores was mixed with 500 L growth medium and incubated for 30 min for spore germination and mRNA production. Thus, only spores that are viable were detected. Subsequently, RNA was extracted from lysed cells, selectively amplified using NASBA, and rapidly identified by the biosensor. While the biosensor assay requires only 15 min assay time, the overall process takes 4 h for detection of ten viable B. anthracis spores, and is shortened significantly if more spores are present. The biosensor is based on an oligonucleotide sandwich-hybridization assay format. It uses a membrane flow-through system with an immobilized DNA probe that hybridizes with the target sequence. Signal amplification is provided when the target sequence hybridizes to a second DNA probe that has been coupled to liposomes encapsulating the dye sulforhodamine B. The amount of liposomes captured in the detection zone can be read visually or quantified with a hand-held reflectometer. The biosensor can detect as little as 1 fmol target mRNA (1 nmol L–1). Specificity analysis revealed no cross-reactivity with 11 organisms tested, among them closely related species such as B. cereus, B. megaterium, B. subtilis, B. thuringiensis, Lactococcus lactis, Lactobacillus plantarum, and Chlostridium butyricum. Also, no false positive signals were obtained from nonviable spores. We suggest that this inexpensive biosensor is a viable option for rapid, on-site analysis providing highly specific data on the presence of viable B. anthracis spores.  相似文献

Summary. Interactions of the DNA bases adenine (A), guanine (G), cytosine (C), and thymine (T) with various metal ions (M) of groups IA and IIA of the periodic table of the elements were studied at the HF, MP2, and DFT levels of theory. The structures and thermodynamic stabilities of these species were studied at the gas phase. The calculations uphold that there exist two active sites in G and one in A, C, and T. The calculations also show that the O2 atom in T is a more active site for metal ion bindings than that in C. The stability energies for G … M complexes are larger than those for A … M complexes and the stability energies for T … M complexes are larger than those for C … M complexes. As z/r ratio for the metal ion increases, the interaction energy for the complex increases systematically. Thermodynamic quantities such as ΔH, ΔG, ΔS, and ln K were determined for each complexation reaction, [Base+M n+ →(Base … M) n+]. A, G, and C complexation reactions except for C … Rb+ are exothermic. The situation is quite different for T complexation reactions and all except for T … Be2+ and T … Mg2+ are endothermic.  相似文献

Efficient synthesis of an epothilone molecules fragment (15R)-C13-C21 was carried out from D-mannitol through its conversion into methyl 2,3-O-cyclohexylidene-D-glycerate followed by the cyclopropanation of the ester group with ethylmagnesium bromide in the presence of titanium(IV) isopropoxide and the transformation of the obtained cyclopropanol into the corresponding 2-substituted allyl bromide. The coupling of the latter catalyzed by copper with 2-methylthiazolyl-4-magnesium bromide, the shift of a double carbon-carbon bond in the product obtained into the position, conjugated with the thiazole ring, and the common transformation of the protected 1,2-diol function afforded the target compound in 15% yield.  相似文献

This study concerns the removal of the 137Cs+ and 60Co2+ β+γ-radioactive ions in Azolla caroliniana Willd. water fern. The living fern and two different types of biosorbent prepared from Azolla caroliniana were tested to remove the above-mentioned radioactive ions from dilute solutions, in the absence and in the presence of the ionic competition. Effective 137Cs+ and 60Co2+ ions removal from low radioactive wastewaters was demonstrated. The time dependent K d (t) values were calculated from the absorption data. These results indicate that removal process achieved equilibrium in about 120 min and that it involves two steps: rapid and slow absorption; the active process (metabolic bioaccumulation on the living fern) was responsible for above one half of the total removal process. A thin layer radiochromatography study leads to the conclusion that the biochemical components in which 137Cs+ and 60Co2+ place themselves are of a polysaccharide and lipoid fractions.  相似文献

Abstract  A series of variously substituted N-methylpiperidin-4-one-O-benzyloximes were synthesized by three different methods. Among them, the direct conversion of 2,6-diarylpiperidin-4-ones into the corresponding oxime ethers (method A) was proved to be better than the other two methods in the sense of good yield, convenience, easy work-up and quick reaction time. All the synthesized compounds are characterized by IR, Mass and NMR (1H NMR, 13C NMR, 1H-1H COSY, 1H-13C COSY and HMBC) spectral studies. The conformational preference of the synthesized oxime ethers with/without alkyl and aryl substituents at C-3/C-5 and C-2/C-6 is discussed using the spectral data. The observed chemical shifts and coupling constants suggest that the synthesized oxime ethers adopt chair conformation with equatorial orientation of all the substituents, whereas 1-methyl-3-isopropyl-2,6-diphenylpiperidin-4-one-O-benzyloxime also exists in boat conformation. Based on the NMR data, the effects of oximination on ring carbons and their associated protons and alkyl substituents are discussed. In addition, the effect of NMe group on the 2,6-diarylpiperidin-4-one-O-benzyloximes was also studied. Graphical abstract     相似文献

Abstract  In this work, we report on the development of a DNA-based piezoelectric biosensor specific for the detection of an amplicon of the aflD gene of Aspergillus flavus and A. parasiticus. Expression of this gene is consistently correlated with a strain’s ability to produce aflatoxins that are considered very potent liver carcinogens in various animal species and humans. The DNA biosensor has been characterized with synthetic oligonucleotides and amplicons. Moreover, it has been applied to the analysis of real samples consisting of amplicons of DNA extracted from flours and feed contaminated with A. flavus and A. parasiticus. We have developed an aptamer-based electrochemical sensor for detection of Botulinum neurotoxin, where steric hindrance is applied to achieve specific signal amplification via conformational change of the aptamer. The incubation time and potassium concentration of the reaction buffer were found to be key parameters affecting the sensitivity of detection of the recognition of Botulinum neurotoxin by the aptamer. Under optimized experimental conditions, a high signal-to-noise ratio was obtained within 24 h with a limit of detection (LOD) of 40 pg/ml by two standard deviation cutoffs above the noise level.  相似文献

The structure of galactomannans isolated from seeds of G. delavayi and G. aquatica was studied by 1H and 13C NMR spectroscopy. Electrochemical measurements including cyclic voltammetry, differential capacity, and chronocoulometry have been used to characterize the adsorption behaviors of the zwitterionic surfactant N-dodecyl-N,N-dimethyl-3-ammonio-1-propanesulfonate (DDAPS) on the Au(111) electrode surface. The thermodynamics of the ideally polarized electrode have been employed to determine the Gibbs excess and the Gibbs energy of adsorption. The results show that the adsorption of DDAPS has a multistate character. The first two states are observed at potentials close to zero charge. At low bulk DDAPS concentrations, it corresponds to the formation of a film of nearly flat adsorbed molecules. At higher concentrations it is converted into a hemimicellar state. The second state is formed at negative potentials and charge densities close to 0 C cm–2. It corresponds to a film of tilted molecules oriented with the hydrocarbon tail towards the metal and the polar head toward the solution.Dedicated to Professor Zbigniew Galus on the occasion of his 70th birthday and in recognition of his many contributions to electrochemistry.  相似文献

A novel uranyl ion-selective bulk optode membrane, incorporating tri-n-octylphosphine oxide for cation recognition and a lipophilic chromoionophore dibenzoylmethane, has been prepared. The PVC membrane composition was optimized to result in the widest working concentration range. The response range of the proposed optode is 4.1×10–6 to 2.0×10–4 mol L–1 UO22+. The probe works at pH 4.0. Ion interference is low and selectivity, reproducibility, and stability are good.  相似文献

Phase transitions of poly(N-isopropylacrylamide-co-acrylic acid) (PiPA-AA) and poly(N,N- diethylacrylamide-co-acrylic acid) (PdEA-AA) in water have been investigated by means of turbidimetry, Fourier transform infrared (FTIR) spectroscopy and differential scanning calorimetry (DSC). The phase transition temperatures (Tp) of these copolymers increase with the degree of ionization () of the acrylic acid (AA) units, which in turn is dependent on the pH of the solutions. Apparent values of pKa for the AA units, determined from the pH dependencies of Tp, are 4.7 and 5.4 for PiPA-AA and PdEA-AA, respectively. Differences between Tp for PiPA-AA and Tp for PiPA homopolymer (Tp) are +1.5 and –0.2 °C/mol% of AA at =1 and 0, respectively. The values of Tp for PdEA-AA are +2.6 (ionic) and –0.5 (nonionic)°C/mol%, indicating that the incorporated AA units have a larger effect on PdEA than on PiPA. DSC measurements performed with each of these copolymers at different pH values show a linear relationship between Tp and the enthalpy of transition (H). IR measurements of PiPA-AA show that the profiles of IR bands from both iPA and AA units exhibit critical changes at Tp of the copolymer. Heating the solution above Tp leads to shifts of the amide II, C–H stretch, and C–H bend bands from the iPA units toward lower wavenumbers, as well as a shift of the amide I band from the iPA units toward higher wavenumbers. A decrease in the intensity of the symmetric C=O stretch IR band from carboxylate anions (1560 cm–1), and an increase in the intensity of the C=O stretch band from COOH groups (1705 cm–1) suggest that a partial protonation of the carboxylate groups (COO+H+COOH) takes place upon the phase transition.  相似文献

A new flavonoid, kaempferol-3,4′-di-O-α-L-rhamnopyranoside (1), and three known flavonoids (2–4) were isolated from the aerial parts of T. communis L. The structure of the new compound was elucidated on the basis of spectroscopic data. Compounds 1 and 2 showed significant antioxidant activity (IC50 187.151 ± 0.821 μM, and 92.079±0.513 μM, respectively), whereas compounds 3 and 4 showed moderate activity in DPPH free radical scavenging assays. The special projective linear groups PSL(2ℓ + 1) or L 2(2ℓ + 1) of order 2ℓ(2ℓ + 1)(ℓ + 1) can be used to study atomic shells of electrons with angular momentum quantum number ℓ corresponding to the atomic p, d, f, and g shells for ℓ = 1, 2, 3, 4, respectively. For the atomic g shell the group L 2(9) is isomorphic with the alternating group A 6 on six objects of order 360 or the symmetry group of the 5-dimensional simplex, a 5-dimensional analogue of the tetrahedron with 6 vertices and 15 edges. This leads to the subgroup chain SO(9) ⊃ SO(5) ⊃ L 2(9) for the atomic g shell analogous to the subgroup chain SO(7) ⊃ G 2L 2(7) ≈7 O for the atomic f shell. In the L 2(9) group only the representations of spherical harmonics or sums thereof, Γ(Y), with dimensions dim Γ(Y) or dim Γ(Y) ± 1 divisible by 9 are found to be individually reducible to irreducible representations (irreps) or sums of irreps of L 2(9). This leads to term groupings such as S, PD, G, PF, DH, L, PK, DI, FH, M, FI, PO, DN, HK, R, etc., of increasing total dimension for the irreps of SO(9) for various g n configurations in the atomic g shell.  相似文献
