The aim of this study is to validate isotope-dilution electrospray ionization tandem mass spectrometry (ESI-MS-MS) method with a dual-loop cleanup device for simultaneous quantitation of two benzene metabolites, trans, trans-muconic acid (ttMA) and S-phenylmercapturic acid (SPMA), in human urine. In this study, a pooled blank urine matrix from rural residents was adopted for validation of the analytical method. The calibration curve, detection limit, recovery, precision, accuracy and the stability of sample storage for the system have been characterized. Calibration plots of ttMA and SPMA standards spiked into two kinds of urine matrixes over a wide concentration range, 1/32-8-fold biological exposure indices (BEIs) values, showed good linearity (R > 0.9992). The detection limits in pooled urine matrix for ttMA and SPMA were 1.27 and 0.042 μg g−1 creatinine, respectively. For both of ttMA and SPMA, the intra- and inter-day precision values were considered acceptable well below 25% at the various spiked concentrations. The intra- and inter-day apparent recovery values were also considered acceptable (apparent recovery >90%). The ttMA accuracy was estimated by urinary standard reference material (SRM). The accuracy reported in terms of relative error (RE) was 5.0 ± 2.0% (n = 3). The stability of sample storage at 4 or −20 °C were assessed. Urinary ttMA and SPMA were found to be stable for at least 8 weeks when stored at 4 or −20 °C. In addition, urine samples from different benzene exposure groups were collected and measured in this system. Without tedious manual sample preparation procedure, the analytical system was able to quantify simultaneously ttMA and SPMA in less than 20 min.  相似文献

A highly sensitive and selective solid-phase spectrophotometric method for the determination of sub-μg l−1 level nickel(II) is described. Nickel(II) was sorbed on a styrene-divinylbenzene-type resin Amberlite XAD-4 as a Ni(II)-o-carboxylphenyldiazoaminoazobenzene (o-CDAA) complex. At pH 9.0, resin phase absorbances at 588 and 800 nm were measured directly with an apparent molar absorptivity of 2.95×107 g mol−1 cm−1. The linear range of the determination was 1.2-41 μg g−1 resin. The detection limit and the quantification limit were found to be 0.24 and 0.76 μg g−1 resin, respectively. The relative standard deviation of 10 replicate determinations of 1.0 μg nickel(II) in 100 ml sample was of 1.5%. The tolerance limit of coexistent ions was also investigated. Most of them are in tolerable amount. For practical analyses, 1 ml acetylacetone used can eliminate the interferences caused by Cu and Fe. The procedure was validated by analysis a certified water reference material (GBW 08618 Beijing, China) and a tomato leaf certified reference material (GBW 08402 Beijing, China) with the results in agreement with the certified values. The method was applied to the determination of nickel(II) in water and vegetable samples with satisfactory results.  相似文献

A hollow fiber renewal liquid membrane (HFRLM) extraction method to determine cadmium (II) in water samples using Flame Atomic Absorption Spectrometry (FAAS) was developed. Ammonium O,O-diethyl dithiophosphate (DDTP) was used to complex cadmium (II) in an acid medium to obtain a neutral hydrophobic complex (ML2). The organic solvent introduced to the sample extracts this complex from the aqueous solution and carries it over the poly(dimethylsiloxane) (PDMS) membrane, that had their walls previously filled with the same organic solvent. The organic solvent is solubilized inside the PDMS membrane, leading to a homogeneous phase. The complex strips the lumen of the membrane where, at higher pH, the complex Cd-DDTP is broken down and cadmium (II) is released into the stripping phase. EDTA was used to complex the cadmium (II), helping to trap the analyte in the stripping phase. A multivariate procedure was used to optimize the studied variables. The optimized variables were: sample (donor phase) pH 3.25, DDTP concentration 0.05% (m/v), stripping (acceptor phase) pH 8.75, EDTA concentration 1.5 × 10−2 mol L−1, extraction temperature 40 °C, extraction time 40 min, a solvent mixture N-butyl acetate and hexane (60/40%, v/v) with a volume of 100 μL, and addition of ammonium sulfate to saturate the sample. The sample volume used was 20 mL and the stripping volume was 165 μL. The analyte enrichment factor was 120, limit of detection (LOD) 1.3 μg L−1, relative standard deviation (RSD) 5.5% and the working linear range 2-30 μg L−1.  相似文献

A simple, sensitive, and specific method with gas chromatography-mass spectrometry was developed for simultaneous extraction and derivatization of amphetamines (APs) and 3,4-methylenedioxyamphetamines (MDAs) in human urine by using a monolithic silica spin column. All the procedures, such as sample loading, washing, and elution were performed by centrifugation. APs and MDAs in urine were adsorbed on the monolithic silica and derivatized with propyl chloroformate in the column. Methamphetamine-d5 was used as an internal standard. The linear ranges were 0.01-5.0 μg mL−1 for methamphetamine (MA) and 3,4-methylenedioxymethamphetamine (MDMA) and 0.02-5.0 μg mL−1 for amphetamine (AP) and 3,4-methylenedioxyamphetamine (MDA) (coefficient of correlation ≧0.995). The recovery of APs and MDAs in urine was 84-94%, and the relative standard deviation of the intra- and interday reproducibility for urine samples containing 0.1, 1.0, and 4.0 μg mL−1 of APs and MDAs ranged from 1.4% to 13.6%. The lowest detection limit (signal-to-noise ratio ≧ 3) in urine was 5 ng mL−1 for MA and MDMA and 10 ng mL−1 for AP and MDA. The proposed method can be used to perform simultaneous extraction and derivatization on spin columns that have been loaded with a small quantity of solvent by using centrifugation.  相似文献

New, simple, rapid and precise HPLC-PDA method has been developed and validated for quantification of biomarker myrsinoic acid B in stem bark extracts of Rapanea ferruginea Mez. The method employs a Phenomenex C18 column (250 mm × 4.6 mm I.D., 5 μm) with acetonitrile:methanol:water (pH 2.6 with phosphoric acid) at 48:30:22 as mobile phase, at a flow rate of 0.7 mL min−1 and photo diode array (PDA) detection at 270 nm. The validation data show that the method is specific, accurate, precise and robust. The method was linear, over a range of 5-100.0 μg mL−1, with a limit of detection of 0.369 μg mL−1 and limit of quantification of 1.233 μg mL−1. The method has also shown consistent recoveries (average of 101.3% and 0.12% RSD) of the biomarker, with low intra and inter-day relative standard deviation (1.26% and 1.62%, respectively). The evaluated hydroethanolic extract and dry extract presented MAB values of 63.53 and 36.07 mg g−1, respectively.  相似文献

An on-line flow injection spectrofluorimetric method for the direct determination of aluminium in water samples is described. The method is based on the reaction of aluminium with N-o-vanillidine-2-amino-p-cresol (OVAC) in acidic medium at pH 4.0 to form a water-soluble complex. The excitation and emission wavelengths were 423.0 and 553.0 nm, respectively, at which the OVAC-Al complex gave the maximum fluorescence intensity at pH 4.0 in a 50% methanol-50% water medium at 50 °C. An interference from fluoride ions was minimised by the addition of Be2+. Other ions were found not to interfere at the concentrations likely to be found in natural waters. The proposed methods were validated in terms of linearity, repeatability, detection limit, accuracy and selectivity. Under these conditions, the calibration was linear up to 1000 μg L−1 (r = 0.999). The limit of detection (3σ) for the determination of Al(III) was 0.057 μg L−1 and the precision for multiple determinations of 3 ng mL−1 Al(III) prepared in ultra-pure water was found to be 0.62% (n = 10).The Schiff base ligand could be used to determine ultra-trace aluminium from natural waters. Analysis of environmental certified reference materials showed good agreement with the certified values. The procedure was found to be equally applicable to both freshwater and saline solutions, including seawater.  相似文献

A microwave-assisted extraction used to extract trace triorganotin from aquatic organisms and a sensitive analytical method for the determination of ultratrace triorganotin (namely trimethyltin, triethyltin, tripropyltin and tributyltin) with capillary electrophoresis-inductively coupled plasma mass spectrometry were firstly described in this study. The extraction method is simple, effective and can be used to extract trace triorganotin in aquatic organisms within several min. The analytical method has a much lower detection limit of 0.2-0.7 ng Sn/mL for triorganotin compounds, and can be used to determine trace triorganotin in aquatic organisms directly without any derivatization and preconcentration. Using above methods, we have successfully determined trimethyltin, triethyltin, tripropyltin and tributyltin in dried Mya arenaria Linnaeus and Corbicula fluminea within 17 min with a recovery of 93-104% and a RSD (relative standard deviation, n = 6) of 2-5%. Our results showed that dried M. arenaria Linnaeus contained an extremely high tributyltin of 5.1 μg Sn/g dried weight, indicating that it may be a good biomarker for the organotin pollution in ocean.  相似文献

This work reports the preparation of molecularly imprinted polymer (MIP) particles for selective extraction and determination of selenium ions from aqueous media. Polymerization was achieved in a glass tube containing SeO2, o-phenylenediamine, 2-vinylpyridine (VP), ethyleneglycoldimethacrylate (EDMA), 2,2′-azobisisobutyronitrile (AIBN). The polymer block obtained was ground and sieved (55-75 μm) and the Se-o-phenylenediamine complex was removed from polymer particles by leaching with 2 M of HCl, which leaves a cavity in the polymer particles. The polymer particles both prior to and after leaching have been characterized by IR and thermogravimetric (TG) studies. The effect of different parameters, such as pH, extraction time, type and least amount of eluent for elution of complex from polymer were evaluated. Extraction efficiencies >99% were obtained by elution of the polymers with 15 mL of methanol-acetonitrile mixture (1:2, v/v). The limit of detection of the proposed method followed by hydride generation atomic absorption spectroscopy (HG-AAS) was found to be 3.3 μg L−1 and a dynamic linear range (DLR) of 10-200 μg L−1 was obtained. The relative standard deviations (R.S.D.s) at 30.0 μg L−1 of Se were below than 8.1%. The influence of various cationic interferences on percent recovery of complex was studied. The method was applied to the recovery and determination of selenium in different real samples.  相似文献

Four flavonoids (rutin, hyperoside, quercitrin and quercetin) in Houttuynia cordata Thunb. and Saururus chinensis (Lour.) Bail. were determined by capillary electrophoresis with wall-jet amperometric detection. The working electrode was a 500 μm diameter carbon disc electrode and the detection potential was +0.95 V (versus Ag/AgCl). Effects of several important factors, such as the running buffer and its corresponding pH and concentration, separation voltage, injection time were investigated to acquire the optimum conditions for separation of these four flavonoids. Baseline separation for the four flavonoids was obtained within 21 min in a 60 cm length capillary at a separation voltage of 15 kV with a 60 mmoL/L Na2B4O7-120 mmoL/L NaH2PO4 buffer (pH 8.8) as running buffer. The relationship between peak currents and analyte concentrations was linear over about two orders of magnitude with detection limits (defined as S/N = 3) ranging from 0.02 to 0.05 μg/mL for all analytes. This method was applied for the determination of the above four flavonoids in H. cordata Thunb. and S. chinensis (Lour.) Bail. with simple extraction procedures, and the assay results were satisfactory.  相似文献

A screening method was developed to discriminate among water samples contaminated or uncontaminated with N-nitrosamines in order to reduce the use of expensive instruments such as chromatographs. The system is based on the preconcentration of the analytes onto a sorbent column, elution and derivatization to form nitrite, then formation of a coloured product (Griess reaction) and photometric detection. The limit of detection achieved for 100 ml of sample volume was 0.2 μg/l and the sample frequency 3 h−1. The reliability of the proposed method of the N-nitrosamines was established at five concentrations (between 0.5 and 3 times the limit of detection). For a level concentration of 0.6 μg/l (three times the limit of detection), the percentage of false negatives is 0%. The method was applied to the screening of several water samples (river, pond, well, tap and waste) with a positive response only for waste water samples.  相似文献

Paeonol is the active component in the traditional Chinese medicines (TCMs), such as Cynanchum paniculatum, which has been used to treat many diseases, such as eczema. In this work, a simple, rapid and sensitive method was developed for the determination of paeonol in rabbit plasma, which was based on headspace solid-phase microextraction (HS-SPME) followed by gas chromatography-mass spectrometry (GC-MS). The extraction parameters of fiber coating, sample temperature, extraction time, stirring rate and ion strength were systemically optimized; the method linearity, detection limit and precision were also investigated. It was shown that the proposed method provided a good linearity (0.02-20 μg mL−1, R2 > 0.990), low detection limit (2.0 ng mL−1) and good precision (R.S.D. value less than 8%). Finally, GC/MS following HS-SPME was applied to fast determination of paeonol in rabbit plasma at different time point after oral demonstration of Cynanchum paniculatum essential oil. The experimental results suggest that the proposed method provided an alternative and novel approach to the pharmacokinetics study of paeonol in the TCMs.  相似文献

In this study a new method for determination of cadmium in alcohol fuel using Moringa oleifera seeds as a biosorbent in an on-line preconcentration system coupled to flame atomic absorption spectrometry (FAAS) was developed. Flow and chemical variables of the proposed system were optimized through multivariate designs. The limit of detection for cadmium was 5.50 μg L−1 and the precision was below 2.3% (35.0 μg L−1, n = 9). The analytical curve was linear from 5 to 150 μg L−1, with a correlation coefficient of 0.9993. The developed method was successfully applied to spiked alcohol fuel, and accuracy was assessed through recovery tests, with recovery ranging from 97.50 to 100%.  相似文献

The cell-sorption of paramagnetic ions of Mn2+ and Cr3+ onto a Chlorella vulgaris(C. vulgaris) cell-immobilized micro-column was significantly improved in the presence of an external magnetic field generated in a finite solenoid, by placing the micro-column in the center of the solenoid in a sequential injection system. Magnetic field creates an opposite drift velocity on the hydrated paramagnetic ions against the flow of the sample zone, retards the moving velocity of the metal ions and provides extra contacting time with the cells on the micro-column and offers more chances for the paramagnetic ions to interact with the various functional groups or binding sites on the cell surface, which significantly facilitates cell-sorption of the paramagnetic ions. The sorption efficiencies of Mn2+ and Cr3+ at the 20 μg L−1 level were improved from 45 to 80% and 60 to 90%, respectively, in a magnetic field of 240 mT.The system was applied for the separation/preconcentration of ultra-trace level of manganese. The presence of an external magnetic field significantly alleviated the interfering effects from coexisting metal ions. Within a liner range of 0.025-0.5 μg L−1 and a sampling volume of 500 μL, an enrichment factor of 21.2, a limit of detection of 0.008 μg L−1, along with a sampling frequency of 20 h−1 was attained, achieving a precision of 2.1% R.S.D. (0.2 μg L−1). Manganese contents in a certified reference material of riverine water and a snow water were analyzed.  相似文献

A simple and sensitive method for determination of the O-demethylation activity of rat, dog, minipig, and human liver micrsomes toward paeonol using ultra-performance liquid chromatography with mass detection (UPLC-MS) has been developed. The method uses chemically synthesized O-demethylated metabolite of paeonol (2,4-dihydroxyacetophenone, DHA) as a standard for method validation. Validation was done with respect to specificity, linearity, detection limit, recovery, stability, precision and accuracy. The chromatographic separation was achieved on a UPLC BEH C18 column (50 mm × 2.1 mm i.d., 1.7 μm), with phase of acetonitrile-water (ratio 30:70). Selective ion reaction (SIR) monitor was specific for paeonol, DHA and I.S. The method was specific since there were no interference peaks from the reaction matrix. The calibration curve for DHA was linear from 0.5-100 μM with r2 = 0.9999. The newly developed method has good precision and accuracy. The method was successfully used to determine the kinetics of DHA activities toward paeonol in liver microsomes from different species. Dog liver microsomes (DLMs) were the most active in paeonol O-demethylation (709.7 pmol/min/mg protein) followed by rat liver microsomes (RLMs) (579.6 pmol/min/mg protein), HLMs (569.3 pmol/min/mg protein), and then minipig liver microsomes (PLMs) (417.3 pmol/min/mg protein). The developed method was appropriated for rapid screening paeonol O-demethylation activity in liver microsomes from different species.  相似文献

A new capillary electrophoresis (CE) method for the determination of quinolizidine alkaloids in Sophora medicinal plants was developed. A total of seven alkaloid components (cytisine, sophocarpine, matrine, lehmannine, sophoranol, oxymatrine and oxysophocarpine) were separated within 15 min. The running buffer was a 50 mM phosphate buffer containing 1%HP-β-CD and 3.3% isopropanol. The linear calibration ranges were 5.50-88.0 μg ml−1 for cytisine and lehmannine, 5.00-88.0 μg ml−1 for sophocarpine and sophoranol, 5.60-89.6 μg ml−1 for matrine and oxysophocarpine, and 24.0-384 μg ml−1 for oxymatrine. The recoveries of the seven alkaloids were 96.0-102.9% with relative standard deviations from 1.50 to 3.00% (n = 5). The method was successfully applied to different Sophora medicinal plants including Sophora flavescens, Sophora tonkinensis and Sophora alopecuroides.  相似文献

A new approach of flow injection chemiluminescent immunoassay (FICLIA) with synergistic enhancement was developed. The synergistic action was significant in the chemiluminescence (CL) system of luminol-H2O2-HRP with two enhancers, para-phenylphenol (PPP) and sodium tetraphenylborate (NaTPB). The relative CL intensity was 128.84% stronger (compared to NaTPB alone) and 122.66% stronger (compared to PPP alone) using both enhancers than any one alone (both P<0.001). While the present approach was applied to the determination of rabbit IgG as a model analyte, satisfactory results were obtained in the dynamic range of 2-60 μg l−1 with a detection limit of 0.68 fmol per injection and a precision of 4.7-9.3%, and the recoveries were 92.5-99.4%. The results of rabbit IgG determination accorded with those of radio immunoassay (RIA). The lifetime of the immuno-column was considerably long: one column was applied to more than 200 determinations in 1 month without significant loss of activity.  相似文献

Molybdate was examined as a complex-forming additive to the CE background electrolytes (BGE) to affect the selectivity of separation of polyhydric phenols such as flavonoids (apigenin, hyperoside, luteolin, quercetin and rutin) and hydroxyphenylcarboxylic acids (ferulic, caffeic, p-coumaric and chlorogenic acid). Effects of the buffer concentrations and pH and the influence of molybdate concentration on the migration times of the analytes were investigated. In contrast to borate (which is a buffering and complex-forming agent generally used in CE at pH ≥9) molybdate forms more stable complexes with aromatic o-dihydroxy compounds and hence the complex-formation effect is observed at considerably lower pH. Model mixtures of cinnamic acid, ferulic acid, caffeic acid and 3-hydroxycinnamic acid were separated with 25 mM morpholinoethanesulfonic acid of pH 5.4 (adjusted with Tris) containing 0.15 mM sodium molybdate as the BGE (25 kV, silica capillary effective length 45 cm × 0.1 mm I.D., UV-vis detection at 280 nm). With 25 mM 2-hydroxy-3-[4-(2-hydroxyethyl)-1-piperazinyl]propanesulphonic acid/Tris of pH* 7.4 containing 2 mM sodium molybdate in aqueous 25% (v/v) methanol as the BGE mixtures of all the above mentioned flavonoids, p-coumaric acid and chlorogenic acid could be separated (the same capillary as above, UV-vis detection at 263 nm). The calibration curves (analyte peak area versus concentration) were rectilinear (r > 0.998) for ≈8-35 μg/ml of an analyte (with 1-nitroso-2-naphthol as internal standard). The limit of quantification values ranged between 1.1 mg l−1 for p-coumaric acid and 2.8 mg l−1 for quercetin. The CE method was employed for the assay of flavonoids in medicinal plant extracts. The R.S.D. values ranged between 0.9 and 4.7% (n = 3) when determining luteolin (0.08%) and apigenin (0.92%) in dry Matricaria recutita flowers and rutin (1.03%) and hyperoside (0.82%) in dry Hypericum perforatum haulm. This paper describes an analytical method for the determination of reductive sulphur (S(IV), S(-II)) in glass. The glass sample is dissolved in hydrofluoric/hydrochloric acid mixture and the sulphur is separated via distillation in an apparatus made of polyfluoralkoxyethylene (PFA). The distilled hydrogen sulphide is trapped in buffered boric acid-zinc acetate solution and subsequently determined after conversion to an ethylene blue dye. The range of the method lies within a range of 2-1200 μg g−1 reductive sulphur. The quantification limit for reductive sulphur is 2 μg g−1.Different analysed glass types show either no detectable reductive sulphur or up to 30% of the total sulphur content reductive sulphur. The inter-laboratory standard deviation shown by a round robin test performed is excellent (±4 μg g−1; average 59 μg g−1). Sources of error of the methodology are discussed.  相似文献

A simple and sensitive liquid chromatographic method is described for the analysis of γ-amino-n-butyric acid (GABA) in human urine. GABA is increased in the urine of cancer patients and could be used as a biomarker in the diagnosis and treatment of related patients. The method is based on derivatizing GABA with a fluorescent reagent (naproxen acyl chloride) for transforming the non-chromophoric GABA to a derivative with chromophoric and fluorophoric properties. The resulting derivative is highly responsive to a fluorimetric detector (λex = 230 nm, λem = 350 nm). The lower quantitation of the method is attainable at 100 nM GABA with a detection limit about 10 nM (S/N = 3 with 20 μL injected). Application of the method to the analysis of GABA in the urine of patients with ovarian and uterine cancer was demonstrated.  相似文献

A method using an immunoaffinity column (IAC) and liquid chromatography-electrospray ionization mass spectrometry (LC/MS) for on-line detecting the presence of MA in the effluent was developed for the quantitative and enantiomeric determination of (S)-(+)-methamphetamine (d-MA) in urine. The IAC was made in our laboratory and utilized in the LC/MS to simultaneously extract and separate enantiomers of MA from urine samples. An aqueous ammonium acetate buffer was used as the mobile phase. Urine samples were spiked with racemic deuterated methamphetamine (MA-d14) as internal standard (IS), filtered through a membrane, and injected into the LC/MS without any further pre-treatment. Protonated molecular ion of MA and MA-d14 (m/z 150 and 164) were isolated and further fragmented, the respective product ions, m/z 119 and 130, were collected for quantitative determination. This is an improvement of our previous method (A.C. Lua, Tsong-Yung Chou, J. Chromatogr. A 967 (2002) 191). In the previous method, MA was separated with HPLC, the efflux was fractionated and each fraction was either determined with an immunoassay or GC/MS. Monitoring of MA in the efflux is tedious and time consuming. Urine samples spiked with different concentrations of d-MA were measured by this method. A linear relationship exists in the 150-1050 ng/mL range, and the detection limit (defined as signal-to-noise ratio 3) of d-MA was determined to be 18 ng/mL. The linearity of the method for d-MA can be described by the equation (Y = 1.415 × 10−3X + 0.034, correlation coefficient: r2 = 0.999). Within run, accuracy and precision (n = 6, relative error: −7.2 to +4.0% and relative standard deviation: 3.8-9.3%) of the method are fairly good.  相似文献
