Highly accelerated and safe nitration of phenolic compounds has been found to be feasible with a mixture of calcium nitrate and acetic acid as an efficient nitration agent under brief microwave irradiation. This method is compatible with the green chemistry approach because calcium salts—the inorganic byproducts—can be useful agrochemicals rather than waste chemicals. Commercially available 15N-labeled calcium nitrate is convenient for the preparation of 15N-labeled compounds for metabolic studies and mass spectrometry. This safe nitration method can be included in laboratory experiments for high school and college students.  相似文献

A solid-phase route for synthesis of 15N-labeled acylpolyamines is described. Utilizing alkylation at benzylic N-atom as a key step, 15N-atoms are incorporated by stepwise construction of the polyamine framework on the solid support. A 15N-labeled nitroxide was incarcerated into an octa acid nanocapsule, which was confirmed by 1H NMR and EPR spectroscopy. Electron paramagnetic interaction between the 15N-labeled incarcerated nitroxide and a 14N-labeled free nitroxide in the external aqueous solution was observed by EPR spectroscopy. The observation of spin-spin interaction, through the walls of the cancer and is reflected in the simultaneous line-broadening of both the 15N-labeled and 14N-labeled nitroxides. The computer-assisted analysis of the EPR data further provides direct information on the motion and the polarity of both the incarcerated paramagnetic nitroxide and the nitroxides in the external bulk aqueous phase. We also show how communication between an incarcerated guest and molecules in the bulk solvent can be enhanced or inhibited by supramolecular factors such as Coulombic attraction or repulsion between a charged guest@host complex (incarcerated 15N nitroxide) and charged molecules in the aqueous phase.  相似文献

In situ 1H nuclear magnetic resonance spectroscopy was used to investigate the processes that occur during the synthesis of 1,3,6,8-tetraazatricyclo[4.3.1.13,8]undecane (TATU). NMR analysis showed a reaction mixture containing more than one compound. The production of these intermediates and collateral products was rationally supported by a careful 1H NMR monitoring study. We characterized 1,3,5-triazabicyclo[3.2.1]octane (TABO, 4) and 3-(2-aminoethyl)-1,3,5-triazabicyclo[3.2.1]octane (AETABO, 7) by 1H and 13C NMR in D2O solution inside the NMR sample tube, as an intermediate and collateral product of the reaction, respectively. Further, a reaction of 1,3,6,8-tetraazatricyclo[4.4.1.13,8]dodecane (TATD) with 15N-labeled ammonium chloride was carried out. The 15N NMR and GC-MS experiments indicated that 15N was incorporated into TATU, TABO, and urotropine.  相似文献

Per-15N-labeled microcystins were prepared for use as surrogates for accurate liquid chromatography–mass spectrometry analysis. Two strains of Microcystis aeruginosa were cultured in 15NO3-containing TS-15 medium. To change from the incorporation of 14N to 15N into all cell components, cells of Microcystis aeruginosa were precultured in Na15NO3-containing medium for more than 6 months. After mass cultivation of the strains, cells of each strain were harvested and lyophilized. Microcystin variants were extracted from the lyophilized cells and per-15N-labeled microcystin variants were purified using high-performance liquid chromatography and high-performance thin-layer chromatography. The structures of per-15N-labeled microcystin variants were confirmed by their mass spectrometry spectra and NMR spectra. When per-15N-labeled microcystins were used as surrogates for quantitative analysis of these toxins in cyanobacterial cells, excellent accuracy (98–106%) was obtained, with the m/z of M+, [M+1]+, and [M+2]+ of both microcystins and the per-15N-labeled microcystins as surrogates being completely separated. In conclusion, per-15N-labeled microcystins are excellent surrogates for microcystin analysis using liquid chromatography–mass spectrometry.  相似文献

The molecular structures of two aminophosphonium salts (bromide and tetrafluoroborate) have been determined by X-ray analysis. They have similar conformations and hydrogen bond (HB) networks: the N–H acid proton is bonded to the anion and, in the case of the fluoroborate, to the oxygen atom of the phosphine oxide, forming a pseudo six-membered ring closed by a weak N–HO intramolecular hydrogen bond (IMHB). These compounds have been studied by multinuclear NMR in solution, including the 15N-labeled derivatives, to determine a complete set of coupling constants. A coupling of 1.5 Hz between the 15N and the 31P nuclei, separated by three bonds, was observed experimentally for the bromide in CDCl3 solution, which appears to be a classical 3 J N-P across the covalent bonds and not a 3h J N-P across the IMHB.  相似文献

Lithocholic acid N-(2-aminoethyl)amide (1) and deoxycholic acid N-(2-aminoethyl)amide(2) have been prepared and characterized by1H, 13C and 15N NMR. The accurate molecular masses of 1 and 2 have been determined by ESI MS. The formation of the Cd2+-complexes (1+Cd and 2+Cd) in CD3OD solution have been detected by 1H,13C, 15N and 113Cd NMR. The 13C NMR chemical shift assignments of 1 and 2 and their Cd2+-complexes are based on DEPT-135 and z-GS 1H,13C HMQC experiments as well as comparison with the assignments of the related structures. The 15N NMR chemical shiftassignments of the ligands and theirCd2+-complexes are based on z-GS1H,15N HMBC experiments. 13C NMR chemical shift differences between 1and its 1:1 Cd2+-complex based on ab initiocalculations at Hartree-Fock SCI-PCM level using3-21G(d) basis set are in agreement with theexperimental shift changes observed onCd2+-complexation.  相似文献

Dimethyl dihydro-1,2,4,5-tetrazine-3,6-dicarboxylate can exist either in 1,2- or 1,4-dihydro tautomeric forms. The 15N NMR spectra of dimethyl dihydro-1,2,4,5-tetrazine-3,6-dicarboxylate were measured at the 15N natural abundance level as well as in 15N doubly labelled selectively and in 15N completely labelled compounds (20% 15N). The J(15N,15N) value was determined in 15N completely labelled compounds (20% 15N) using 1D 15N INADEQUATE and was found to be 12.2 ± 0.2 Hz in deuteriochloroform, acetonitrile-d3, DMSO-d6 and CD3OH. Very similar 15N chemical shifts and 1J(15N,1H) values were also observed in all the solvents. This indicates that compound 1 exists completely in the 1,4-dihydro tautomeric form (i.e., as dimethyl 1,4-dihydro-1,2,4,5-tetrazine-3,6-dicarboxylate) in all the solvents tested.  相似文献

ABSTRACT

Compound independent calibration (CIC) can be used to measure stable isotopically labeled molecules and their metabolites. This study was designed to demonstrate that gas chromatography coupled to atomic emission detection and CIC are useful analytical tools for the specific and quantitative determination of 13C and 15N labeled caffeine and its metabolites. These labeled compounds can be measured by monitoring either 13C or 15N atomic emission lines and using only one calibration function from one of the analytes. This method can be helpful when it is difficult or impossible to obtain an authentic reference material, which is often the case in metabolic studies.  相似文献

An analytical method for the determination of three polyamines (putrescine, spermidine, and spermine) and five acetylpolyamines [N1-acetylspermidine (N1AcSpd), N8-acetylspermidine (N8AcSpd), N1-acetylspermine, N1,N8-diacetylspermidine, and N1,N12-diacetylspermine] involved in the polyamine catabolic pathway has been developed using a hybrid tandem mass spectrometer. Heptafluorobutyryl (HFB) derivatives of these compounds and respective internal standards labeled with stable isotopes were analyzed simultaneously by TOF MS, based on peak areas appearing at appropriate m/z values. The isomers, N1AcSpd and N8AcSpd were determined from their fragment ions, the acetylamidopropyl and acetylamidobutyl groups, respectively, using MS/MS with 13C2-N1AcSpd and 13C2-N8AcSpd which have the 13C2-acetyl group as an internal standard. The TOF MS method was successfully applied to measure the activity of enzymes involved in polyamine catabolic pathways, namely N1-acetylpolyamine oxidase (APAO), spermine oxidase (SMO), and spermidine/spermine N1-acetyltransferase (SSAT). The following natural substrates and products labeled with stable isotopes considering the application to biological samples were identified; for APAO, [4,9,12-15N3]-N1-acetylspermine and [1,4,8-15N3]spermidine (15N3-Spd), respectively; for SMO, [1,4,8,12-15N4]spermine and 15N3-Spd, respectively; and for SSAT, 15N3-Spd and [1,4,8-15N3]-N1-acetylspermidine, respectively.  相似文献

The mechanism proposed earlier for explanation of the furoxan ring formation in the nitrosation of dipotassium salts of 1-hydroxyimino-2,2-dinitro-1-R-ethanes with NaNO2/AcOH was confirmed experimentally by determining the ionization constants of the dinitromethyl and oxime fragments in the starting dipotassium salt and by examining 1H, 13C, 14N, and 15N NMR and mass spectra of isomeric 3(4)-nitro-4(3)-R-furoxans with the 15N(5) and 15N(2) ring atoms, respectively, and 3,4-diarylfuroxan with both 15N-labeled ring atoms. A comparative analysis of the IR and Raman spectra of the 15N-labeled furoxan derivatives obtained and their unlabeled analogs allowed refinement of the literature data on interpretation of the vibrational spectra of furoxan derivatives.  相似文献

A method was developed for the synthesis of mixed-metal heterospin compounds with the direct coordination of the nitroxide fragment based on the replacement of acetonitrile molecules in the heterotrinuclear complex [Co2Gd(NO3)Piv6(CH3CN)2] with nitroxide molecules. The molecular and crystal structure of the heterospin mixed-ligand heterotrinuclear CoII, GdIII, CoII complex [Co2Gd(NO3)Piv6(NIT-Me)2], where NIT-Me is stable nitronyl nitroxide, was established. The magnetic properties of this complex were investigated in the temperature range of 2–300 K. The coordination of nitroxide groups to CoII ions is responsible for strong exchange interactions between the unpaired electrons in the exchange clusters {>-·O-CoII}, resulting in the virtually complete spin coupling between each coordinated >N-·O group and one of the unpaired electrons of each CoII ion at temperatures below 200 K. The structures of substitutedN-aminonitropyrazoles and 1- and 2-amino-4-nitro-1,2,3-triazoles as well as the site of protonation of 1-aminopyrazole were determined based on the1H,13C, and15N (14N) NMR spectra. The13C NMR spectra were recorded under conditions of13C-{1H,14N} triple resonance. Effects of substituents in the pyrazole ring on the13C and14N chemical shifts were studied. Asmarines are marine alkaloids, with a unique tetrahydro[1,4]diazepino-[1,2,3-g,h]purine (THDAP) structure and interesting biological properties. Three synthetic approaches were employed for the preparation of the THDAP system. Several N-9, of the purine, protecting groups were investigated. 15N-Chemical shifts measured from 15NH HMBC experiments for several compounds, that demonstrate the influence of various structural features on the 15N-resonances are reported.  相似文献

The 1H, 13C and 15N NMR spectroscopic properties of six thallium tris-(pyrazol-1-yl)borates, including a tetrakis derivative, were determined. The results in solution were necessary to understand those, more complicated, in the solid state. A collection of 205Tl-15N and 205Tl-13C couplings was measured in the latter state. Among those, a very large coupling constant (between 194 and 282 Hz) has been measured on the carbon at the position 4 of the pyrazole ring in several compounds and particularly for the cyclobutyl derivative [Tl(TpCbu)]. It has been assigned to a direct interaction of the 4-C-H ? Tl type and related to the X-ray structures, when available.  相似文献

A rapid method for the extraction and monitoring of nanogram level of Pb2+, Cu2+ and Zn2+ ions using uniform silanized mesopor (SBA-15) functionalized with diethylenetriamine groups and flame atomic absorption spectrometry (FAAS) is presented. The preconcentration factor of the method is 100 and detection limit of the technique is 5.5?ng?mL?1 and 1.4?ng?mL?1 and 0.1?ng?mL?1 for Pb2+, Cu2+ and Zn2+ respectively. The time and the optimum amount of the sorbent, pH and minimum amount of acid for stripping of ions from functionalized SBA-15 were tested. The maximum capacity the functionalized SBA-15 was found to be 183.0 (±1.9) µg, 156.0 (±1.5) µg and 80.0 (±1.6) µg of Pb2+, Cu2+ and Zn2+/mg functionalized SBA-15, respectively.  相似文献

The dibenzo[3n]crown-n were synthesised from1,2-bis(o-hydroxyphenoxy)ethane obtained from 1,2-bis(o-formylphenoxy)ethane via Bayer-Willigeroxidations with H2O2/CH3COOH in good yields. The cyclic condensation of 1,2-bis(o-hydroxyphenoxy)ethanewith dichlorides, and ditosylates of polyethylene glycols in DMF/Me2CO3 gave the macrocyclesdibenzo[15]crown-5, dibenzo[18]crown-6, dibenzo[21]crown-7 anddibenzo[24]crown-8. The structures were identified using IR, mass, 1H and 13C NMR spectroscopy. Therecognition of the molecules for the cations, Li+, Na+, K+, Rb+ and Zn2+were conducted quantitatively with steady state fluorescencespectroscopy. The 1:1 association constants in acetonitrileshowed a good relation of the appropriate size of the macrocyclic ether towards the fitting cationradii. Namely, dibenzo[15]crown-5 was the best for Li+ binding and more than 100 times better thanNa+ and K+. Dibenzo[21]crown-7 was excellent for Rb+ binding while K+ is 100 timesless preferred. The largest crown ether studied, dibenzo[24]crown-8, exhibited the order of binding power,Rb+ > K+ > Na+. Zn2+ displayed, however, a marked binding with only dibenzo[18]crown-6.p>  相似文献
