Several derivatives of the new pyrimido[4′,5′:3,4]pyrazolo[1,2‐b]phthalazine‐4,7,12‐trione ring system have been prepared by the reaction of 3‐amino‐1‐aryl‐5,10‐dioxo‐5,10‐dihydro‐1H‐pyrazolo[1,2‐b]phthalazine‐2‐carbonitriles with aliphatic carboxylic acids in the presence of phosphoryl chloride (POCl3). The synthesized compounds were characterized on the basis of IR, 1H NMR, and 13C NMR spectral and microanalytical data.  相似文献

Following the principles of green chemistry, cardanol derivatives have been used as renewable, low‐cost, and available natural starting materials to construct a variety of protected and unprotected amino acids derivatives. The reaction of cardanol derivatives with different phthaloylamino acids including glycine, alanine, phenylalanine, and valine in the presence N,N'‐ dicyclohexylcarbodiimide (DCC ) as coupling reagent afforded high yields of the target compounds. Deprotection of phthaloylamino acids derivatives was achieved by heating with hydrazine hydrate. The chemical structures of all products were confirmed by spectral data (IR , MS , 1H NMR , 13C NMR ) and elemental analyses. Antibacterial evaluation of the synthesized products was performed, which exhibited potent to weak activity in comparison with a standard drug.  相似文献

Some new 1,2,4‐triazolo‐, 1,3,4‐oxadiazolo‐, 1,3,4‐thiadiazol‐, and pyrazolo‐2,4,6‐trimethylphenyl‐1(2H)‐oxo‐phthalazine derivatives were synthesized and identified by IR, 1H NMR, 13C NMR, MS and elemental analysis. A new series of phthalazine derivatives was synthesized from the reaction of 1‐chloro‐4‐(4‐phenoxyphenyl)phthalazine as a reactive starting material with different carbon, nitrogen, oxygen, and sulfur nucleophiles. The structural formulae of all products were confirmed and characterized by elemental analyses and spectral data (IR, 1H NMR, 13C NMR, and mass spectra). Some of the synthesized derivatives were screened for their antitumor activity against four human tumor cell lines using MTT assay. In comparison with doxorubicin as standard drug, compounds 1 , 20 , and 25 showed the most potent cytotoxic effect. In addition, investigation of antioxidant activity revealed that hydrazinylphthalazine 20 has the highest activity.  相似文献

Substituted biaryl compounds 9, 14–16 were synthesized through Pd(0)-catalyzed cross coupling reactions between boronic acids or tin derivatives and aryl halides. N-Amination, and subsequent ring closure resulted in the new angularly-fused pyrido[1,2-b]pyridazinium systems 1–4 . The use of silver oxide as a cocatalyst in the couplings of tin derivatives was essential in order to obtain rapid reaction and better yields. Structures were determined by 1H nmr and 13C nmr spectra.  相似文献

A novel series of (Z)‐1‐(arylhydrazono)‐1‐(benzothiazol‐2′‐ylthio)propan‐2‐ones have been prepared and their utility as building blocks in the synthesis of novel derivatives of Pyridazin‐6(1H)‐ones, pyridazin‐6(1H)‐imines, and phthalazine incorporating a benzothiazole residue are investigated. All of the title compounds were confirmed by elemental analysis, IR, 1H NMR, 13C NMR, NOE, and mass spectrometry. The synthesized compounds are screened for their antibacterial as well as antifungal activity. All the tested compounds showed high to moderate activity against the selected microorganisms. J. Phthalazines have received considerable attention for their wide antimicrobial activity. Regiospecific nucleophilic attack of 4‐benzylphthalazin‐1‐ol by the 1‐oxo rather than the aza group on different alkyl halides gave novel phthalazine heterocyclic derivatives. Moreover, a variety of nucleosides bonded to electron‐withdrawing groups were synthesized using 4‐benzylphthalazine‐1‐ol. The density functional theory has been used to investigate the electronic structure of the synthesized compounds. All of the synthesized derivatives showed remarkable activity when tested against Gram‐positive and Gram‐negative bacteria, Aspergillus niger, and Candida albicans. The reactivity of these nucleosides was expected to arise from their bonding with the lone pair of N‐atom of the macromolecules of bacteria. These bonding were expected to inhibit the enzyme by forming highly stable complex with lower highest occupied molecular orbital energy. The structures of these synthesized derivatives were established by Fourier transform infrared, 1H‐NMR, and 13C‐NMR spectroscopic evidence.  相似文献

Cyclic anhydrides of di- and tetracarboxylic acids react with hexamethyldisilazane to give the corresponding N-trimethylsilylimides in 90-96% yields. However, cyclic anhydrides of succinic, 4-nitrophthalic, and tetrachlorophthalic acids react differently, giving acyclic trimethylsilyl esters of mono-N-trimethylsilylamides of these acids in 90-93% yields. The 1H NMR, IR, and mass spectra of the compounds synthesized were studied.  相似文献

A series of new symmetrical tetrazole-based carbazole derivatives starting from the initially generated 3,6-diformyl-N-alkylcarbazoles were successfully synthesized through a one-pot Ugi-azide reaction in moderate to high yields. Simplicity, easily accessible chemicals, mild reaction conditions, and fast separation of the products with the formation of bistetrazole-based carbazole derivatives in one step are some advantages of this method. The structure of the products was characterized and confirmed by using spectroscopic techniques such as 1H NMR, 13C NMR, FT-IR, and MS spectroscopy.  相似文献

Novel sulfur‐containing biphenol monomers were prepared in high yields by the reaction of 4‐mercaptophenol with chloropyridazine or chlorophthalazine compounds. High‐molecular‐weight poly(arylene ether)s were synthesized by a nucleophilic substitution reaction between these sulfur‐containing monomers and activated difluoro aromatic compounds. The inherent viscosities of these polymers ranged from 0.34 to 0.93 dL/g. The poly(pyridazine)s exhibited glass‐transition temperatures greater than 165 °C. The poly(phthalazine)s showed higher glass‐transition temperatures than the poly(pyridazine)s. A polymer synthesized from a bisphthalazinebiphenol and bis(4‐fluorophenyl)sulfone had the highest glass‐transition temperature (240 °C). The thermal stabilities of the poly(pyridazine)s and poly(phthalazine)s showed similar patterns of decomposition, with no significant weight loss below 390 °C. The poly(phthalazine)s were soluble in chlorinated solvents such as chloroform, and the poly(pyridazine)s were soluble in dipolar aprotic solvents such as N,N′‐dimethylacetamide. Twenty-two novel N-nitro-N′-substituted phenyl-N-(2,6-dibromo-4-fluorophenyl)urea derivatives were designed and synthesized via a simple and convenient BTC ‘one-pot’ procedure using DMAP as the catalyst. The structures of all newly synthesized compounds were confirmed by IR, 1H NMR, and elemental analysis, and a part has been identified by 13C NMR. The preliminary bioassay indicates that the target compounds possesses moderate herbicidal activity against Sorghum sudanense. However, some of the title compounds presented high plant growth regulating activity against rape.  相似文献

Novel asymmetric macrocyclic Schiff bases were synthesized by the condensation of N,N′-bis(2-aminophenyl)-3,4-diphenylthiophene-2,5-dicarboxamide (1) with diformyl derivatives of phenol, furan, difurans, pyridine, pyrrole, and dipyrroles. The reaction proceeds in high yields and without by-products in methanol in the presence of inorganic and organic acids (proton-template condensation). In the case of monocyclic diformyl derivatives and di(5-formylfuran-2-yl) sulfide, the reaction occurs in 1,4-dioxane (templateless synthesis). A series of pyrrolo[2, 1-a]phthalazine derivatives has been prepared including examples carrying a variety of O- and N-substituents at position-6. A number of the products and related tricyclic structures show mild anti-hypertensive activity including 6-allyloxy-1, 2-dichloro-3-phenylpyrrolo[2, 1-a]phthalazine.  相似文献

Propargyloxycarbonyl group is used as a protecting group for the hydroxyl groups of serine, threonine and tyrosine. The propargyloxycarbonyl derivatives of these hydroxy amino acids are stable to acidic and basic reagents commonly employed in peptide synthesis. The deprotection of the O-Poc derivatives using tetrathiomolybdate does not affect commonly used protecting groups such as N-Boc, N-Cbz, N-Fmoc, methyl and benzyl esters. The di-and tripeptides synthesized using O-Poc derivatives of serine, threonine and tyrosine are stable, isolable compounds and give the hydroxy peptides in good yields when treated with tetrathiomolybdate.  相似文献

A new series of ruthenium(II) N-heterocyclic carbene complexes [RuL1,2,3(p-cymene)Cl2] (3a–c) (where L is a N-heterocyclic carbene), have been synthesized via transmetalation. The new ruthenium(II)-NHC complexes were applied to transfer hydrogenation of acetophenone derivatives and aldehydes using 2-propanol as a hydrogen source and KOH as a co-catalyst. The results show that the corresponding alcohols could be obtained in good yield with high catalyst activity (up to 100%) under mild conditions. [RuL1(p-cymene)Cl2] (3a) is much more active than the other complexes in transfer hydrogenation. Reactions, catalyzed by 3a–c, showed the highest reaction rates and yields of alcohol when the substrates bear more electron-withdrawing substituents. All new compounds were characterized by IR, elemental analysis, LC–MS (ESI), and NMR spectroscopy.  相似文献

One-pot four-component reactions of phenacyl bromides, parabanic or thioparabanic acids, thiophenols, and triphenylphosphine in the presence of triethylamine afforded new derivatives of hydantoin or thiohydantoin in good to high yields (65%-90%). Their antibacterial activities were evaluated against two Gram-positive bacteria (Staphylococcus aureus and Bacillus subtilis) and two Gram-negative bacteria (Escherichia coli and Pseudomonas aeruginosa). Among the synthesized compounds, the obtained products from 2-hydroxythiophenol exhibited higher antibacterial activity than those obtained from 2-aminothiophenol. Compound 9l including N,N′-diphenyl thiohydantoin moiety showed the highest antibacterial activity (26.0 ± 01.4) against B subtilis, in comparison with other synthesized samples. The antioxidant activities of the synthesized hydantoins and thiohydantoins were investigated by DPPH radical-scavenging based on Blois method. The results showed that all the compounds have high DPPH inhibition potency (77.4%-83.9%) that it could be due to existence of heteroatoms with lone pair electrons and exchangeable protons on their NH2 and OH groups.  相似文献

Thirty-three alkyl and aryl isothiocyanates, as well as isothiocyanate derivatives from esters of coded amino acids and from esters of unnatural amino acids (6-aminocaproic, 4-(aminomethyl)benzoic, and tranexamic acids), were synthesized with satisfactory or very good yields (25–97%). Synthesis was performed in a “one-pot”, two-step procedure, in the presence of organic base (Et3N, DBU or NMM), and carbon disulfide via dithiocarbamates, with 4-(4,6-dimethoxy-1,3,5-triazin-2-yl)-4-methylmorpholinium toluene-4-sulfonate (DMT/NMM/TsO) as a desulfurization reagent. For the synthesis of aliphatic and aromatic isothiocyanates, reactions were carried out in a microwave reactor, and selected alkyl isothiocyanates were also synthesized in aqueous medium with high yields (72–96%). Isothiocyanate derivatives of L- and D-amino acid methyl esters were synthesized, under conditions without microwave radiation assistance, with low racemization (er 99 > 1), and their absolute configuration was confirmed by circular dichroism. Isothiocyanate derivatives of natural and unnatural amino acids were evaluated for antibacterial activity on E. coli and S. aureus bacterial strains, where the most active was ITC 9e.  相似文献

A new convenient synthesis of N‐carboxyanhydrides (NCAs) of α‐amino acids was achieved by selective cyclization of urethane derivatives of α‐amino acids. The urethanes were readily synthesized via N‐carbamoylation of α‐amino acids by bis(4‐nitrophenyl)carbonate quantitatively. These urethanes having 4‐nitrophenoxy moiety were tolerant to air and moisture to allow their facile purification and storage. When the obtained urethanes were heated in 2‐butanone at 60 °C, they underwent the selective cyclization via intramolecular nucleophilic attack of the carboxyl moiety to the urethane moiety with releasing 4‐nitrophenol, leading to the successful formation of the corresponding NCAs. Addition of carboxylic acids remarkably stabilized the formed NCAs during the reaction, allowing their isolation in high yields. © 2009 Wiley Periodicals, Inc. New 6‐chloro‐ and 6,7‐dichloro‐4‐(4‐aminophenyl)‐2‐(N‐alkylcarbamoyl)‐1,2‐dihydrophthalazines ( 12,18 ) were synthesized starting from 6‐chloro‐ and 6,7‐dichloro‐4‐(4‐nitrophenyl)phthalazine ( 8 ). Routes to 1‐unsubstituted ( 12 ) and 1‐methyl ( 18 ) derivatives are different. Key intermediates 8 were prepared from 4‐chloro‐ and 3,4‐dichlorotoluene via ortho‐formylbenzophenone derivatives 7 .  相似文献

The mono-N-amino salts of 3-phenylcinnoline, phthalazine, 1-phenylphthalazine, 4-phenylquinazoline, 2-phenylquinoxaline, 1,5- and 1,8-naphthyridines are in high yields prepared by direct N-amination of the parent heterocycles with O-mesitylenesulfonylhydroxylamine. With the one expection of 3-phenylcinnoline, the site of N-amination was determined by mass and nmr spectral techniques. The results indicate that the N-amination occurs preferentially at the least sterically hindered nitrogen atom.  相似文献
