The synthesis and reactions of methyl 2-[3-(trifluoromethyl)phenyl]-4H-furo[3,2-b]pyrrole-5-carboxylate (1a) are described. Upon reaction with methyl iodide, benzyl chloride, or acetic anhydride, this compound gave N-substituted products 1b-d. By hydrolysis of compounds 1a-c, the corresponding acids 2a-c were formed, or by reaction with hydrazine-hydrate, the corresponding carbohydrazides 3a-c were formed. By heating 2-[3-(trifluoromethyl)phenly]-4H-furo[3,2-b]pyrrole-5-carboxylic acid (2a) in acetic anhydride, 4-acetyl-2-[3-(trifluoromethyl)phenyl]furo[3,2-b]pyrrole (4) was formed. By hydrolysis of 4, 2-[3-(trifluoromethyl)phenyl]-4H-furo[3,2-b]pyrrole (5a) was formed, and reactions with methyl iodide or benzyl chloride gave N-substituted products 5b-c. The reaction of 4 with dimethyl butynedioate gave substituted benzo[b]furan 6. Compound 3a reacted with triethyl orthoesters giving 7a-c, which afforded with phosphorus (V) sulphide the corresponding thiones 8a-c. The thiones 8a-c reacted with hydrazine hydrate to form hydrazine derivatives 9a-c. The reaction of triethyl orthoformiate with compounds 9a-c led to furo[2′,3′: 4,5]pyrrolo[1,2-d][1,2,4]triazolo[3,4-f][1,2,4]triazines 10a-c. Hydrazones 11a-c were formed from 3a-c and 5-[3-(trifluoromethyl)phenyl]furan-2-carboxaldehyde. The effect of microwave irradiation on some condensation reactions was compared with “classical” conditions. The results showed that microwave irradiation shortens the reaction time while affording comparable yields.  相似文献

Summary The absorption bands of the C=O Stretching vibrations of a series of thirty-nine substitutedZ-3-methylene phthalides (1a-s, 2a-h, 3a-f, 4a-c, and5a-c) were measured in CHCl3 and CCl4. The two-levelFermi resonance effect on the infrared spectra of the above compounds was investigated after deconvolution and band separation. The wave numbers of the unperturbed fundamental C=O stretching vibrations exhibit excellent linear correlations withHammet's constants of substituents and13C NMR chemical shifts of the C=O group.

Zusammenfassung Die Absorptionsbanden der C=O-Streckschwingung von 39 substituiertenZ-3-Methylenphthaliden (1a-s, 2a-h, 3a-f, 4a-c und5a-c) wurden in CHCl3 und CCl4 vermessen. Durch Dekonvolution und Bandentrennung konnte der EinflußFermi-Resonanz auf die Infrarotspektren der obengenannten Verbindungen untersucht werden. Die Wellenzahlen der ungestörten C=O-Streckschwingungen ergeben ausgezeichnete lineare Korrelationen mit denHammetschen Substituentenkonstanten und den13C-NMR-Verschiebungen der Carbonylgruppe.

The reactivity of 3-amino-2,4-dicyano-methyl-(1d) and-ethyl-crotonate (1e), resp., a new type of codimer from malononitrile and cyanoacetate, was investigated. Condensation reactions with aromatic aldehydes (2a-g), with heterocyclic aldehydes (2j-l), nitrosobenzenes (3a-c) ando-hydroxybenz-and-naphtaldehydes (4a-f), resp., are described, the differences in reactivity, compared with dimer malononitrile and dimeric forms of cyanoacetate (1a-c) are negligible. Condensation compounds of1a with cycloalkanones (5a-c) are able to cyclise with sulfur to cycloalkanonothieno-pyridines (6a-b), while the cycloalkano-derivatives of1d gave no ring closure.

Condensation of 4-bromomethylquinoline derivatives1 a-1 c with glycine and thioglycolic acid gave the corresponding quinolylmethylglycine and quinolylmethylthioacetic acid derivatives2 a-2 c and2 d-2 f, respectively. Cyclization of2 a-2 f was affected either by polyphosphoric acid or concentrated sulphuric acid to give3 a-3 f. Chlorination of2 a-2 f and3 a-3 f were also accomplished.

Methyl 4-oxiranylmethyl-4H-furo[3,2-b]pyrrole-5-carboxylates 2a-c and methyl 1-oxiranylmethyl-1H-benzo[4,5]furo[3,2-b]pyrrole-2-carboxylate (2d) were prepared by reaction of the appropriate starting compounds 1a-d with excess chloromethyloxirane. The compounds 2a-d undergo oxirane ring opening by heterocyclic amines (morpholine, pyrrolidine, piperidine or 4-methylpiperazine) giving N-2-hydroxy-3-heteroaminopropyl-substituted compounds 3a-f or substituted 4,5-dihydrofuro[2',3':4,5]pyrrolo[2,1-c][1,4]oxazin-8-ones 4a-e.  相似文献

The quinolinetrionesTMCH, PCH, MCH and phenalene trione (PT) do not react with aryl isocyanates toSchiff bases5a, but react by ring contraction to yield the spiro-imidazolidines2 a-d,3 a-e,4 a-b and5 a-cbd. The reactions ofo-tosylaminobenzaldehyde (1) withp-aminobenzenesulfonamides (2a-c) yielded 13-(p-RNHSO2C6H4)-6,12-epimino-5,11-ditosyl-5,6,11,12-tetrahydrodibenzo[b f]-1,5-diazocines (3a-c) (R=H (a); 2-thiazolyl (b); or 2,6-dimethoxy-4-pyrimidinyl 2,6-dimethoxy-4-pyrimidinyl (c). The Friedel-Crafts acetylation of 5-hydroxyindole derivativesla-c regioselectively produced 6-acetyl-5-hydroxy-indole derivatives2a-c which were condensed with cinnamic acid using POC13 and dry pyridine to afford 5-cinnamoyloxy-6-acetyl-indoles3a-c. These upon Baker-Venkataraman transformation furnished 6-cinnamoylacetyl-5-hydroxyindoles4a-c which were cyclised separately with AcOH/HCl and Ac2O/AcONa to 4H-pyrano-(2,3-f)indol-4-one derivatives5a-c and6a-c respectively.  相似文献

Five groups of 4-substituted phenyl 4?-(2?- (or 3″-) substituted-4?-alkoxyphenylazo) benzoates (Ina-c to Vna-c) were investigated in which, within each group, the length of the terminal alkoxy group varies between 8 and 16 carbons, while the other terminal substituent, X, is a polar group that alternatively changed between the electron-donating CH3O and the electron-withdrawing Br group, in addition to the un-substituted analogue (X = H). The lateral substituent (Y) in the five groups IV varies, respectively, between H, 3-CH3, 2-CH3, 3-F and 2-F. Their mesophase stabilities were determined by differential scanning calorimetry and phases identified by polarised light microscopy. The two newly prepared groups of compounds (IVna-c and Vna-c) are structurally characterised by infrared, 1H-NMR, mass spectroscopy, thermogravimetric and elemental analyses. Binary phase diagrams were constructed for each pair of isomers from groups IV and V bearing the same wing substituents but the lateral F is attached to different positions (2? or 3″).  相似文献

Abstract

A simple and eco-compatible synthesis of podands as 4a-i is performed using amines (3a-c ), CS2, and dichlordiamides (DCDs) (2a, c ) in the absence of a catalyst in water. Three reacting DCDs (2a-c ) were obtained in the reaction of aromatic diamines (1a, c ) with chloroacetyl chloride. Dynamic NMR spectroscopic data of three series of podands (4a-c , 4d-f , and 4g-i ) are discussed and their free energies of activations are calculated (ΔG c s) at coalescence temperatures. The ΔG c of these podands were attributed to conformational isomerization in the range of 14.7–17 kcal/mol due to rotation and resonance effects about the thioamide C?N bond.

Abstract

3-Aryloxymethyl-4-phenyl-5-mercapto-s-triazoles (Ia-c) have been synthesized and reacted with N-chloroacetyl derivatives of aromatic and/or heterocyclic amines to yield 5-(N-aryl/heterocyclyl)-carboxamidomethyl thio-s-triazole derivatives 2a-o and 3a-o respectively. Reaction of Ia-u with ethyl chloroacetate gave the corresponding esters 4a-c which were reacted with hydrazine hydrate to give hydrazides 5a-c. Condensation of 5a-c with aromatic aldehydes gave Schiff s bases 6a-u with on cycloaddition reaction with thioglycolic acid yielded 4-thiazolidinones 7a-g. Some of these compounds were screened in vitro for their antibacterial activities.  相似文献

Summary Thep-benzoquinones5e-j react with diazomethane (after dehydrogenation) to 6-anilino-2e, dimethylamino-2h, phenylthio-2i, and methylthioindazolquinone2j. Methylation with dimethylsulfate of these as well as of the already known indazolquinones 6-toluidino-2f, 6-methylanilino-2g, 5-tert.butyl-2m, 5,6-methyl- (isomer mixture)2l,n, and benz-indazolquinone2o yield the 1-and 2-N-methyl derivatives3 and4. The structure of the 2-methyl derivatives is established by reaction of the corresponding benzoquinones with 3-methylsydnon in the case of4e,4g. UV/VIS-, IR- and1H-NMR-spectroscopy (in CDCl3 andDMSO) were used for structure determination. Comparison of the UV/VIS-spectra of2–4 shows that the indazolquinones are existing as 2 H-4,7-diones. For the structure elucidation of the 1- rs. 2-methylderivatives (whichh can be attributed to the methylation products) NMR-spectroscopy is well suited even without knowing the second isomer (solvent-effect). The course of the reaction of quinones with diazomethane and of the methylation reactions of the indazolquinones is discussed.

Cyclic α-oxo-α-(1,2,4-triazolyl)ketene S,S-acetals 1a-e react with hydrazine affording the substituted pyrazolyl-1,2,4-triazoles 3a-e. With hydroxylamine hydrochloride under basic conditions the substituted isoxazolyl-1,2,4-triazoles 5a-c are obtained.  相似文献

以豆腐果苷为原料, 与盐酸羟胺缩合反应生成4-β-D-吡喃阿洛糖苷-苯甲醛肟(2), 2与次氯酸叔丁酯发生取代反应生成4-β-D-吡喃阿洛糖苷-α-氯苯甲醛肟(3); 再将3与Schiff碱通过1,3-偶极环加成生成一系列3-(4-β-D-吡喃阿洛糖苷-苯基)-4-芳基-5-芳基-1,2,4-噁二唑啉(5a5h). 35a5h共9个化合物均未见文献报道, 其结构经1H NMR, IR和MS (HRMS)加以确认, 并对5a5h进行了药理活性筛选. 结果表明, 部分化合物具有良好的镇静活性. 其中, 化合物5g (200, 100 mg•kg-1)和5h (200, 100 mg•kg-1)与豆腐果苷相比较具有更强的活性.  相似文献

Identical reaction products, viz. 2,3,5,6-tetrafluoro-4-perfluoroalkylthiopyridines 3a-c, were obtained on thermal decomposition of Xe(II) bisperfluoroalkylcarboxylates in the presence of mercaptotetrafluoropyridine and bis(2,3,5,6-tetrafluoro-4-pyridyl) disulfide. Compounds 3a-c readily interact with N-, O-, and S-containing nucleophiles, though with the first two types of reactant only with substitution of fluorine atoms in position 2 of the pyridine ring. In the latter case products are obtained both with retention of the perfluoroalkylthio group on interaction with sodium N,N-dimethyldithiocarbamate, and with total fission of that group in the case of alkali metal methanethiolate and toluenethiolate.  相似文献

Addition of amines to dibenzylidene sulfamide1 a yields the correspondingSchiff bases3 a, b and monobenzylidene sulfamide2. Reaction of several dibenzylidene sulfamides1 with various lithium-amides gives N1-substituted N2-benzylsulfamoyl-benzamidines7 a-k via an intramolecular hydride transfer reaction, whereas by treatment of1 a with sodium amide 2-benzyl-3,5-diphenyl-3,4-dihydro-2H-1,2,4,6-thiatriazine-1,1-dioxide8 is obtained, which on hydrolysis yields9 and11. Alkylation products are described, the isomeric products12a/13a and12b/13b are isolated, their structures are confirmed by synthesis, IR and NMR-spectra.

Condensation of 3,5-dinitro-4-chloro-6-methoxy-2-methylquinoline (1) with benzylamine, ethanolamine and/or thioglycolic acid afforded the quinoline derivatives4 a-c. Cyclization of4 a and4 b with alkali and condensation of1 with glycine in sodium carbonate solution furnish 2H-imidazo[4,5-c]quinoline derivatives5 a-c, respectively. Treatment of5 b with benzaldehyde in presence of zinc chloride gave the styryl derivative6. 1 reacted with sodium azide to give the azido derivative4 d, which upon treatment with phenylhydrazine or sodium borohydride yielded the 4-amino derivative4 3. Moreover,1 was treated with phenylhydrazine to give4 f, which cyclized in 10% sodium hydroxide solution to the corresponding v-triazolo[4,5-c]quinoline 3-oxide derivative7. When however4 f was treated with dilute hydrochloric acid, the corresponding phenylpyrazolo[3,4,5-de]quinoline derivative8 was obtained.

Zusammenfassung Kondensation von 3,5-Dinitro-4-chlor-6-methoxy-2-methylchinolin (1) mit Benzylamin, Ethanolamin und/oder Thioglycolsäure ergab die Chinolinderivate4 a-c. Cyclisierung von4 a und4 b mit Alkali und Kondensation von1 mit Glycin in Natriumcarbonatlösung lieferte 2H-imidazo[4,5-c]chinolin-Derivate5 a-c. Behandlung von5 b mit Benzaldehyd in Gegenwart von Zinkchlorid ergab das Styrylderivat6. 1 wurde mit Natriumazid zum Azidoderivat4 d umgesetzt, das mit Phenylhydrazin oder Natriumborhydrid zum 4-Aminoderivat4 e weiterreagierte.1 ergab mit Phenylhydrazin4 f, das in 10% NaOH-Lösung zum entsprechenden Triazolo[4,5-c]chinolin-Derivat7 cyclisierte. Aus4 f wurde mit verdünnter Salzsäure das Phenylpyrazolo[3,4,5-de]chinolin8 erhalten.
