As a new detection model, the reversible fluorescence “turn-off-on” sensor based on quantum dots (QDs) has already been successfully employed in the detections of many biochemical materials, especially in the researches on the interactions between anticancer drugs. The previous studies, however, mainly focused on simple-structured oligonucleotides and Calf thymus DNA. G-quadruplex, an important target for anti-cancer drug with special secondary structure, has been stimulating increasing research interests. In this paper, we report a new detection method based on the fluorescence “turn-off-on” model with water-soluble ZnCdSe QDs as the fluorescent probe, to analyze the interactions between anticancer drug (N-methyl-4-pyridyl) porphyrin (TMPyP) and nucleic acid, especially the G-quadruplex. The fluorescence of QDs can be quenched by TMPyP via photo-induced electron transfer and fluorescence resonance energy transfer, while on the other hand, the combination between TMPyP and G-quadruplex releases QDs from their quenchers and thus recovers the fluorescence. Most importantly, the fluorescence “turn-off-on” model has been employed, for the first time, to analyze the impacts of special factors on the interaction between TMPyP and G-quadruplex. The excellent selectivity of the system has been verified in the studies of the interactions between TMPyP and different DNAs (double-stranded DNA, single-stranded G-quadruplex, and different types of G-quadruplexes) in Na+ or K+-containing buffer.  相似文献

An ultrasensitive and simple method for the determination of melamine was developed based on the fluorescence quenching of thioglycolic acid (TGA) capped CdTe quantum dots (QDs) at pH 11.0. In strong alkaline aqueous solution, the selectivity of the method has been greatly improved due to most heavy metal ions show no interference as they are in the precipitation form or in their anion form. Furthermore, CdTe quantum dots have higher quantum yields at higher pH. The method has a wider concentration range and lower detection limit. The influence factors on the determination of melamine were investigated and the optimum conditions were determined. Under optimum conditions, the fluorescence intensity change of TGA coated CdTe quantum dots was linearly proportional to melamine over a concentration range from 1.0 × 10−11 to 1.0 × 10−5 mol L−1 with a correlation coefficient of 0.9943 and a detection limit of 5 × 10−12 mol L−1. The mechanism of fluorescence quenching of the QDs has been proposed based on the infrared spectroscopy information and electrophoresis experiments in presence of melamine under alkaline condition. The proposed method was employed to detect trace melamine in milk powder and pet feeds with satisfactory results.  相似文献

We present a facile fabrication of layer-by-layer (LbL) microarrays of quantum dots (QDs) and acetylcholinesterase enzyme (AChE). The resulting arrays had several unique properties, such as low cost, high integration and excellent flexibility and time–saving. The presence of organophosphorous pesticides (OPs) can inhibit the AChE activity and thus changes the fluorescent intensity of QDs/AChE microscopic dot arrays. Therefore, the QDs/AChE microscopic dot arrays were used for the sensitive visual detection of OPs. Linear calibration for parathion and paraoxon was obtained in the range of 5–100 μg L−1 under the optimized conditions with the limit of detection (LOD) of 10 μg L−1. The arrays have been successfully used for detection of OPs in fruits and water real samples. The new array was validated by comparison with conventional high performance liquid chromatography-mass spectrometry (HPLC-MS).  相似文献

CdS quantum dots (QDs) modified with l-cysteine has been prepared by one step. They are water-soluble and biocompatible. To improve CdS QDs stability and interaction between silver ion and functionalized CdS QDs in aqueous solution, some amounts of fresh l-cysteine were added to functionalized CdS solution. Based on the characteristic fluorescence enhancement of CdS QDs at 545 nm by silver ions in the presence of some amounts of fresh l-cysteine, simultaneously, a gradual red shift of fluorescence emission bands of CdS QDs from 545 to 558 nm was observed. A simple, rapid, sensitive and specific detection method for silver ion was proposed. Under optimum conditions, the fluorescence intensity of CdS QDs is linearly proportional to silver concentration from 2.0 × 10−8 to 1.0 × 10−6 mol/L with a detection limit of 5.0 × 10−9 mol/L. In comparison with single organic fluorophores, functionalized CdS quantum dots are brighter, more stable against photobleaching, and don’t suffer from blinking. Furthermore, owing to the fluorescence enhancement effect of CdS QDs by silver ion, the proposed method showed lower detection blank and higher sensitivity. Possible fluorescence enhancement mechanism was also studied.  相似文献

Health or environmental issue caused by abnormal level of metal ions like Zn2+ or Cd2+ is a worldwide concern. Developing an inexpensive and facile detection method for Zn2+ and Cd2+ is in urgent demand. Due to their super optical properties, fluorescent quantum dots (QDs) have been developed as a promising alternative for organic dyes in fluorescence analysis. In this study, a CdTe QDs-based sensitive and selective probe for Zn2+ and Cd2+ in aqueous media was reported. The proposed probe worked in fluorescence “turn-on” mode. The initial bright fluorescence of CdTe QDs was effectively quenched by sulfur anions (S2−). The presence of Zn2+ (or Cd2+) can “turn-on” the weak fluorescence of QDs quenched by S2− due to the formation of ZnS (or CdS) passivation shell. Under optimal conditions, a good linear relationship between the fluorescence response and concentration of Zn2+ (or Cd2+) could be obtained in the range from 1.6 to 35 μM (1.3–25 μM for Cd2+). The limit of detection (LOD) for Zn2+ and Cd2+ were found to be 1.2 and 0.5 μM, respectively. Furthermore, the present probe exhibited a high selectivity for Zn2+ and Cd2+ over other metal ions and was successfully used in the detection of Zn2+ or Cd2+ in real water samples.  相似文献

This paper described an investigation of a novel eco-friendly fluorescence sensor for Hg2+ ions based on N-acetyl-l-cysteine (NAC)-capped ZnS quantum dots (QDs) in aqueous solution. By using safe and low-cost materials, ZnS QDs modified by NAC were easily synthesized in aqueous medium via a one-step method. The quantitative detection of Hg2+ ions was developed based on fluorescence quenching of ZnS QDs with high sensitivity and selectivity. Under optimal conditions, its response was linearly proportional to the concentration of Hg2+ ions in a range from 0 to 2.4 × 10−6 mol L−1 with a detection limit of 5.0 × 10−9 mol L−1. Most of common physiologically relevant cations and anions did not interfere with the detection of Hg2+. The proposed method was applied to the trace determination of Hg2+ ions in water samples. The possible quenching mechanism was also examined by fluorescence and UV-vis absorption spectra.  相似文献

Based on CdTe/CdS quantum dots (CdTe/CdS QDs) fluorescence (FL) reversible control, a new and sensitive FL sensor for determination of anthraquinone (AQ) anticancer drugs (adriamycin and daunorubicin) and herring sperm DNA (hsDNA) was developed. Under the experimental conditions, FL of CdTe/CdS QDs can be effectively quenched by AQ anticancer drugs due to the binding of AQ anticancer drugs on the surface of CdTe/CdS QDs and photoinduced electron transfer (PET) process from CdTe/CdS QDs to AQ anticancer drugs. Addition of hsDNA afterwards brought the restoration of CdTe/CdS QDs FL intensity, as AQ anticancer drugs peeled off from the surface of CdTe/CdS QDs and embedded into hsDNA double helix structure. The liner ranges and the detection limits of FL quenching methods for two AQ anticancer drugs were 0.33-9 μg mL−1 and 0.09 μg mL−1 for ADM and 0.15-9 μg mL−1 and 0.04 μg mL−1 for DNR, respectively. The restored FL intensity was proportional to concentration of hsDNA in the range of 1.38-28 μg mL−1and the detection limit for hsDNA was 0.41 μg mL−1. It was applied to the determination of AQ anticancer drugs in human serum and urine samples with satisfactory results. The reaction mechanism of CdTe/CdS QDs FL reversible control was studied.  相似文献

We have developed a novel method for the determination of iodate based on the carboxymethyl cellulose-capped CdS quantum dots (QDs). Factors affecting the iodate detection were investigated, and the optimum conditions were determined. Under the optimum conditions, the relative fluorescence intensity of CdS quantum dots was linearly proportional to IO3 over a concentration range from 1.0 × 10−8 to 1.0 × 10−5 mol L−1 with a correlation coefficient of 0.9987 and a detection limit of 6.0 nmol L−1. Iodide, being oxidized by bromine to form iodate, was detected indirectly. The method was successfully applied to the determination of iodate and total amount of iodine in table salt samples. We have developed an analytical method to detect adenosine-5′-triphosphate (ATP) and alkaline phosphatase (ALP) based on the generation of CdS quantum dots (QDs). We demonstrated that Cd2+ cation reacts with S2− anion to generate fluorescent CdS QDs in the presence of some certain amount of ATP. With increase in the ATP concentration, the fluorescence intensity of CdS QDs was also enhanced. ATP can be converted into adenosine by the dephosphorylation of ALP, so that the generation of CdS QDs would be inhibited in the presence of ALP. Therefore, this novel analysis system could be applied to assay ATP and ALP based on the growth of fluorescent CdS QDs.  相似文献

This work developed a novel method to greatly enhance the electrochemiluminescence (ECL) of CdS quantum dots (QDs). The ECL amplification was achieved by the assembly of QDs on poly (diallyldimethylammonium chloride)-functionalized carbon nanospheres (PFCNSs), and successfully employed for sensitive ECL biosensing of oxidase substrates. The carbon nanospheres were prepared by a “green” method, and the high loading of QDs on carbon nanospheres led to a 4-times increased ECL intensity with dissolved O2 as the coreactant. Using xanthine oxidase (XOD) as a model, an ECL biosensor was fabricated by immobilizing the enzyme on the mixing membrane of PFCNSs and QDs. The ECL biosensor showed a fast response to hypoxanthine with a linear concentration range from 2.5 × 10−8 to 1.4 × 10−5 M. The limit of detection was 5 nM at a signal-to-noise ratio of 3. The assay results of hypoxanthine in fish samples were in a good agreement with the reference values by amperometric technique. This facile approach to prepare the PFCNSs/QDs system for ECL biosensing could be of promising application in bioanalysis and electronic device.  相似文献

We have demonstrated the design of a new type fluorescence assay based on the inner filter effect (IFE) of gold nanoparticles (AuNPs) on the fluorescence of quantum dots (QDs). With a high extinction coefficient, AuNPs are expected to be capable of functioning as powerful absorbers. QDs with tunable emission wavelength are ideal fluorophores because the emission spectra of the rationally synthesized QDs can perfectly overlap with the absorption band of the absorber. Aminothiols are chosen as the model analytes, and the IFE-based fluorescent method for detection of aminothiols was suggested. Under the optimum conditions, the response is linearly proportional to the concentration of cysteine in the range of 0.05-0.9 μg mL−1. The present IFE-based fluorescent strategy could be also used to detect glutathione and homocysteine. The linear concentration ranges were 0.05-1.0 μg mL−1 for glutathione and 0.01-1.0 μg mL−1 for homocysteine.  相似文献

In this work, a novel homogeneous and signal “off–on” aptamer based fluorescence assay was successfully developed to detect chloramphenicol (CAP) residues in food based on the fluorescence resonance energy transfer (FRET). The vesicle nanotracer was prepared through labeling single stranded DNA binding protein (SSB) on limposome-CdSe/ZnS quantum dot (SSB/L-QD) complexes. It was worth mentioning that the signal tracer (SSB/L-QD) with vesicle shape, which was fabricated being encapsulated with a number of quantum dots and SSB. The nanotracer has excellent signal amplification effects. The vesicle composite probe was formed by combining aptamer labeled nano-gold (Au-Apt) and SSB/L-QD. Which based on SSB's specific affinity towards aptamer. This probe can't emit fluoresce which is in “off” state because the signal from SSB/L-QD as donor can be quenched by the Au-aptas acceptor. When CAP was added in the composite probe solution, the aptamer on the Au-Apt can be preferentially bounded with CAP then release from the composite probe, which can turn the “off” signal of SSB/L-QD tracer into “on” state. The assay indicates excellent linear response to CAP from 0.001 nM to 10 nM and detection limit down to 0.3 pM. The vesicle probes with size of 88 nm have strong signal amplification. Because a larger number of QDs can be labeled inside the double phosphorus lipid membrane. Besides, it was employed to detect CAP residues in the milk samples with results being agreed well with those from ELISA, verifying its accuracy and reliability.  相似文献

A sequential injection analysis system for determination of arsenic based on hydride generation and fluorescence quenching of mercaptoacetic acid capped cadmium sulfide quantum dots (CdS-MAA QDs) is described. The generated arsine diffused across the PTFE membrane in a gas-diffusion unit and subsequently interacted with CdS-MAA QDs. The parameters affecting the arsine generation and the fluorescence quenching of QDs were studied. Under the optimum conditions, it was observed that a increase in the concentration of As(III) corresponded well to a decrease in fluorescence intensity according to the Stern-Volmer relationship. The extent of quenching was dependent on the concentration of arsenic in the range of 0.08-3.20 mmol L−1, with the detection limit of 0.07 mg L−1. The precision (%RSD) from eight replicates of the determination of As(III) 1.0 mg L−1 was found to be 1.4%. The proposed method was applied to the determination of arsenic in ground water samples with satisfactory recoveries.  相似文献

A novel immunosensor based on graphite screen-printed electrodes (SPEs) modified with bismuth citrate was developed for the voltammetric determination of C-reactive protein (CRP) in human serum using quantum dots (QDs) labels. The sandwich-type immunoassay involved physisorption of CRP capture antibody on the surface of the sensor, sequential immunoreactions with CRP and biotinylated CRP reporter antibody and finally reaction with streptavidin-conjugated PbS QDs. The quantification of the target protein was performed with acidic dissolution of the PbS QDs and anodic stripping voltammetric detection of the Pb(II) released. Detection was performed at bismuth nanodomains formed on the sensor surface during the electrolytic preconcentration step, as bismuth citrate was reduced to metallic bismuth simultaneously with the deposition of Pb on the surface of the immunosensor. Under optimal conditions, the response was linear over the range 0.2–100 ng mL−1 CRP and the limit of detection was 0.05 ng mL−1 CRP. Since the modified SPE serves as both the biorecognition element and the QDs reader, the analytical procedure is simplified, the drawbacks of existing electroplated immunosensors are minimized while the proposed disposable sensing platform provides convenient, low-cost and ultrasensitive detection of proteins and wider scope for mass-production.  相似文献

A novel fluorescent probe for Cu2+ determination based on the fluorescence quenching of glyphosate (Glyp)-functionalized quantum dots (QDs) was firstly reported. Glyp had been used to modify the surface of QDs to form Glyp-functionalized QDs following the capping of thioglycolic acid on the core–shell CdTe/CdS QDs. Under the optimal conditions, the response was linearly proportional to the concentration of Cu2+ between 2.4 × 10−2 μg mL−1 and 28 μg mL−1, with a detection limit of 1.3 × 10−3 μg mL−1 (3δ). The Glyp-functionalized QDs fluorescent probe offers good sensitivity and selectivity for detecting Cu2+. The fluorescent probe was successfully used for the determination of Cu2+ in environmental samples. The mechanism of reaction was also discussed.  相似文献

The importance of cysteine (Cys) in biological systems has stimulated a great deal of efforts in the development of analytical methods for the determination of this amino acid. In this work, a novel fluorescent probe for Cys based on citrate (Cit)-capped CdS quantum dots (QDs) is reported. The Cit-capped CdS QDs fluorescent probe offers good sensitivity and selectivity for detecting Cys. A good linear relationship was obtained from 1.0 × 10−8 mol L−1 to 5.0 × 10−5 mol L−1 for Cys. The detection limit was calculated as 5.4 × 10−9 mol L−1. The proposed method was applied to detect Cys in human urine samples, which showed satisfactory results. This assay is based on both the lability of Cit and the strong affinity of thiols to the surface of CdS QDs. The addition of Cys improved the passivation of the surface traps of CdS QDs and enhanced the fluorescence intensity.  相似文献

Luminescent surface-modified CdSe semiconductor quantum dots (QDs), with nanoparticle (NP) size distribution in the order of 2-7 nm, have been synthesized for optical determination of cyanide ions. The nanoparticles have been functionalised with tert-butyl-N-(2-mercaptoethyl)-carbamate (BMC) groups and exhibit a strong fluorescent emission at about 580 nm with rather long fluorescence lifetimes (several hundred nanoseconds) in aerated methanolic solution. The observed luminescence emitted by the synthesized nanocrystals was tremendously increased by photo-activation under sunlight exposure. The functionalised QDs turned out to exhibit excellent long-term stability when stored in the dark (no significant changes in QDs luminescence emission intensity was observed even after two months from synthesis). The functionalisation of the NPs with carbamate ligand allowed a highly sensitive determination of free cyanide via analyte-induced changes in the photoluminescence (fluorescence quenching of intensity at 580 nm and lifetime changes) of the modified quantum dots (excited at 400 nm). A detection limit of 1.1 × 10−7 M (2.9 μg l−1) of cyanide ions was obtained, while the interfering effect of other inorganic anions (including NO3, Cl or SCN) was negligible even at 200-fold level concentrations in excess of cyanide.  相似文献

The present study described a novel fluorescence enzyme-linked immunosorbent assay (ELISA) used to detect ochratoxin A (OTA) by using the glucose oxidase (GOx)-mediated fluorescence quenching of mercaptopropionic acid-capped CdTe quantum dots (MPA-QDs), in which GOx was used as an alternative to horseradish peroxidase (HRP) for the oxidization of glucose into hydrogen peroxide (H2O2) and gluconic acid. The MPA-QDs were used as a fluorescent signal output, whose fluorescence variation was extremely sensitive to the presence of H2O2 or hydrogen ions in the solution. Under the optimized conditions, the proposed fluorescence ELISA demonstrated a good linear detection of OTA in corn extract from 2.4 pg mL−1 to 625 pg mL−1 with a limit of detection of 2.2 pg mL−1, which was approximately 15-fold lower than that of conventional HRP-based ELISA. Our developed fluorescence immunoassay was also similar to HRP-based ELISA in terms of selectivity, accuracy, and reproducibility. In summary, this study was the first to use the GOx-mediated fluorescence quenching of QDs in immunoassay to detect OTA, offering a new possibility for the analysis of other mycotoxins and biomolecules.  相似文献

In this study, thioglycolic acid capped-CdTe quantum dots (QDs) were modified by polyethylenimine (PEI), and then combined with fluorescein isothiocyanate (FITC) to fabricate FITC–CdTe conjugates. The self-assembly of FITC, CdTe and PEI was ascribed to electrostatic interactions in aqueous solution. The resulting conjugates were developed toward two routes. In route one, ratiometric photoluminescence (PL) intensity of conjugates (IFITC/IQDs) was almost linear toward pH from 5.3 to 8.7, and a ratiometric PL sensor of pH was favorable obtained. In route two, firstly added S2− induced remarkable quenching of QDs PL peak (at the “OFF” state), which was restored due to following addition of Cd2+ (at the “ON” state). In the conjugates, successive introduction of S2− and Cd2+ hardly influenced on FITC PL peaks. According to this PL “OFF-ON” mode, a ratiometric PL method for the detection of Cd2+ was achieved. Experimental results confirmed that the IFITC/IQDs exhibited near linear proportion toward Cd2+ concentration in the range from 0.1 to 15 μM, and the limit of detection was 12 nM. Interferential experiments adequately testified that the proposed sensors of pH and Cd2+ were practicable in real samples and complex systems. In comparison with conventional analytical techniques, the ratiometric PL method was simple, rapid, economic and highly selective.  相似文献
