An accurate, simple and sensitive method based on reversed-phase high-performance liquid chromatography with UV detection has been developed for determination of tiopronin (TP) in human plasma. TP in plasma was reacted with p-bromophenacyl bromide (p-BPB) to give the TP-p-BPB adduct and this derivative was then extracted from the plasma on a silica gel cartridge. Potential interfering compounds were removed by washing with water, and the TP-p-BPB adduct was then eluted with acetonitrile. The organic phase obtained was evaporated to complete dryness under a stream of nitrogen. The residue was dissolved in acetonitrile and this solution was injected on to a reversed-phase ODS HPLC column. The mobile phase was usually the ternary mixture acetonitrile–water–trifluoroacetic acid, 40:59.88:0.12 (v/v). The retention times of TP-p-BPB and the internal standard adduct were 14.4 and 17.9 min, respectively. No interfering peaks were encountered in several blank plasma samples examined. The limit of detection for TP was 12 ng mL?1. Extraction recovery exceeded 70%. The calibration plot for the TP derivative was linear in the range 40?4000 ng mL?1, regression coefficient 0.9989, and the coefficient of the variation of the points of the calibration plot was below 10%. The method was validated appropriately and successfully applied to the determination of TP in human plasma.  相似文献

Spectrophotometric and high performance liquid chromatographic (HPLC) methods have been developed for the determination of cis-platin and carboplatin based on the pre-column derivatization of platinum(II) with 2-acetylpyridine-4-phenyl-3-thiosemicarbazone. The complex was extracted in chloroform with molar absorptivity of 2.2 × 104 L mol−1 cm−1 at 380 nm. The complex eluted from a Phenomenex C-18 (150 mm × 4.6 mm i.d.) column with methanol:water:acetonitrile:tetrabutyl ammonium bromide (1 mM) (44:30:25:1, v/v/v/v) with a flow rate of 1 ml/min and UV detection at 260 nm. Ruthenium(IV) and selenium(IV) also separated completely. The linear calibration curve was with 0.5-12.5 μg/ml and detection limit of 10 ng/ml platinum(II).The analysis of cis-platin and carboplatin injections by spectrophotometric and HPLC methods indicated relative standard deviation (R.S.D.) of 0.66-2.1%. The method was used for the determinations of cis-platin in serum and urine of cancer patients after chemotherapy and platinum contents were found 148-444 and 50-90 ng/ml with R.S.D. of 0.3-3.0 and 0.6-2.4% for the serum and urine, respectively. The recovery of platinum(II) from serum was 97% with R.S.D. 2.2%.  相似文献

Kynurenine (KYN), a tryptophan metabolite, is a crucial compound for modulating neurotransmission because it can be metabolized in vivo into both quinolinic acid and kynurenic acid, which are the agonist and antagonist, respectively, of N-methyl-d-aspartate receptor. For the highly sensitive detection of KYN by high-performance liquid chromatography (HPLC), a fluorescence derivatization of KYN with a benzofurazan-type fluorogenic reagent, 4-N,N-dimethylaminosulfonyl-7-fluoro-2,1,3-benzoxadiazole (DBD-F) was investigated in the present study. KYN was derivatized with DBD-F (DBD-KYN) at 60 °C for 30 min, and separated on an octadecylsilica column with a gradient elution of the mobile phase, which consists of 0.1% formic acid in acetonitrile/methanol/water. DBD-KYN was detected fluorimetrically at 553 nm with an excitation wavelength of 431 nm. The limits of detection and quantification were approximately 0.30 pmol [signal-to-noise ratio (S/N) 3] and 1.0 pmol (S/N, 10) on column, respectively. Plasma KYN levels were successfully determined using 10 μL of rat plasma with satisfactory precision and accuracy. Intra- and inter-day precisions and accuracies were 1.7-6.8%, and −10 to 9.6%, respectively. KYN levels in plasma of male Sprague-Dawley rats (7 weeks old) were approximately 2.4 ± 0.32 μmol L−1 (n = 4). The proposed HPLC method was applied to determine KYN levels in the plasma of ketamine-treated rats—the animal model of schizophrenia.  相似文献

A facile, sensitive and universal method was established for analysis of biogenic amines using micellar electrokinetic chromatography coupled with chemiluminescent (CL) detection. It was found that diperiodatocuprate (III) (K5[Cu(HIO6)2], DPC), a transition metal chelate at unstable high oxidation state, could effectively enhance the reaction between luminol-type compound and hydrogen peroxide, to produce very strong CL signal. In addition, triethylamine was found to be able to effectively improve the yield of the derivatization reaction between biogenic amines and a luminol-type derivatization reagent, N-(4-aminobutyl)-N-ethylisoluminol (ABEI). Based on these facts, three biogenic amines were pre-column derivatized with ABEI, and post-column detected using high sensitive luminol-hydrogen peroxide-DPC CL system. Since the background was quite low, and the signal was quite strong, a considerable improved sensitivity was obtained. The presented method had been successfully applied to simultaneously analyze glycine, proline and phenylalanine with the detection limits (S/N = 3) of 0.030 μmol L−1, 0.23 μmol L−1 and 0.21 μmol L−1, respectively. To evaluate its potential application value, glycine in saliva and urine samples was detected using this method, and satisfied results were obtained. This approach can be further extended to detection of many other compounds such as peptides and drugs by using luminol-type derivatization reagent.  相似文献

Silica gel-bound amines phase modified with p-dimethylaminobenzaldehyde (p-DMABD) was prepared based on chemical immobilization technique. The product (SG-p-DMABD) was used as an adsorbent for the solid-phase extraction (SPE) Cr(III), Cu(II), Ni(II), Pb(II) and Zn(II) prior to their determination by inductively coupled plasma optical emission spectrometry (ICP-OES). The uptake behaviors of SG-p-DMABD for extracting these metal ions were studied using batch and column procedures. For the batch method, the optimum pH range for Cr(III) and Ni(II) extraction was ≥ 3, for Cu(II), Pb(II) and Zn(II) extraction it was ≥ 4. For simultaneous enrichment and determination of all the metals on the newly designed adsorbent, the pH value if 4.0 was selected. All the metal ions can be desorbed with 2.0 mL of 0.5 mol L− 1 of HCl. The results indicate that SG-p-DMABD has rapid adsorption kinetics using the batch method. The adsorption capacity for these metal ions is in the range of 0.40-1.15 mmol g− 1, with a high enrichment factor of 125. The presence of commonly coexisting ions does not affect the sorption capacities. The detection limits of the method were found to be 1.10, 0.69, 0.99, 1.10 and 6.50 μg L− 1 for Cr(III), Cu(II), Ni(II), Pb(II) and Zn(II), respectively. The relative standard deviation (RSD) of the method under optimum conditions was 5.0% (n = 8) for all metal ions. The method was applied to the preconcentration of Cr(III), Cu(II), Ni(II), Pb(II) and Zn(II) from the certified reference material (GBW 08301, river sediment) and water samples with satisfactory results.  相似文献

Two procedures are proposed in this work for the determination of methanol impurities in o,o-dimethyldithiophosphoric acid (DMDTPA). To avoid possible interferences from the main component, DMDTPA was precipitated in the form of insoluble lead complex. Free Pb(II) ions were eliminated with sulfuric acid and methanol was oxidized to formaldehyde with potassium permanganate in methanesulfonic acid medium. Finally, the excess of oxidizing agent was neutralized with saturated sodium oxalate. The above pretreatment procedure was identical for spectrophotometric assay and for chromatographic determination. In the first case, the solution obtained was treated with Nash reagent to form 3,5-diacetyl-1,4-dihydrolutidine (λmax = 415 nm). In the calibration range 0.1-1.0% (methanol in DMDTPA), the analytical figures of merit were: R2 = 0.9993, quantification limit 0.02% methanol in DMDTPA coefficient of variance (n = 5) for 0.1% and 0.4% methanol respectively 6.7% and 2.4%. Recoveries obtained in the sample fortified with 0.1, 0.2, 0.4% of methanol (in DMDTPA) were in the range 99-105%. For chromatographic procedure, formaldehyde was derivatized with 2,4-dinitrophenylhydrazine and separation was achieved on Luna C18(2) column using the isocratic elution with acetonitrile-water (70:30, v/v) and spectrophotometric detection at 360 nm. In the calibration range 0.05-0.25% (methanol in DMDTPA), R2 was always higher than 0.999, the quantification limit was 0.004% and the recoveries in these same fortified samples in the range 98-101%. No statistically significant differences were observed between the results obtained in the analysis of technical grade DMDTPA by the two procedures (ANOVA, p < 0.05)  相似文献

A new simple, sensitive and selective liquid chromatography coupled with mass spectrometry (LC/MS) method for quantification of captopril after precolumn derivatization with p-bromo-phenacyl-bromide in human plasma was validated. Plasma samples were analysed on a monolithic column (Cromolith Performance-RP 18e, 100 mm × 4.6 mm I.D., 3 μm) under isocratic conditions using a mobile phase of a 40:60 (v/v) mixture of acetonitrile and 0.1% (v/v) formic acid in water. The flow rate was 1 mL/min at the column temperature of 30 °C. In these chromatographic conditions, the retention time was 4.4 min for captopril derivative. The detection of the analyte was in MRM mode using an ion trap mass spectrometer with electrospray positive ionisation. The monitored ions were 216, 253, 255, 268, 270 m/z derived from 415 m/z for derivatized captopril. The sample preparation was very simple and consisted in plasma protein precipitation from 0.2 mL plasma using 0.3 mL methanol after the derivatization reaction was completed. Calibration curves were generated over the range of 10-3000 ng/mL with values for coefficient of correlation greater than 0.993 and by using a weighted (1/y2) quadratic regression. The values for precision (CV %) and accuracy (relative error %) at quantification limit were less than 9.9% and 3.9%, for within- and between-run, respectively. The mean recovery of the analyte was 99%. Derivatized samples demonstrated good short-term, long-term, post-preparative and freeze-thaw stability. This is the first reported LC-MS/MS method for analysis of captopril in human plasma that uses protein precipitation as sample processing procedure. The method is very simple and allows obtaining a very good recovery of the analyte. The validated LC-MS/MS method has been applied to a pharmacokinetic study of 50 mg captopril tablets on healthy volunteers.  相似文献

A trace element preconcentration procedure is described utilizing a minicolumn of yeast (Yamadazyma spartinae) immobilized TiO2 nanoparticles for determination of Cr, Cu, Fe, Mn, Ni and Zn from water samples by inductively coupled plasma atomic emission spectrometry. The elements were quantitatively retained on the column between pH 6 and 8. Elution was made with 5% (v/v) HNO3 solution. Recoveries ranged from 98 ± 2 (Cr) to 100 ± 4 (Zn) for preconcentration of 50 mL multielement solution (50 μg L−1). The column made up of 100 mg sorbent (yeast immobilized TiO2 NP) offers a capacity to preconcentrate up to 500 mL of sample solution to achieve an enrichment factor of 250 with 2 mL of 5% (v/v) HNO3 eluent. The detection limits obtained from preconcentration of 50 mL blank solutions (5%, v/v, HNO3, n = 11) were 0.17, 0.45, 0.25, 0.15, 0.33 and 0.10 μg L−1 for Cr, Cu, Fe, Mn, Ni and Zn, respectively. Relative standard deviation (RSD) for five replicate analyses was better than 5%. The retention of the elements was not affected from up to 500 μg L−1 Na+ and K+ (as chlorides), 100 μg L−1 Ca2+ (as nitrate) and 50 μg L−1 Mg2+ (as sulfate). The method was validated by analysis of freshwater standard reference material (SRM 1643e) and applied to the determination of the elements from tap water and lake water samples.  相似文献

A high performance liquid chromatographic method (HPLC) for the simultaneous determination of 4-nonylphenol, bisphenol A, 17α-ethinylestradiol and three endogenic estrogens including 17α-estradiol, 17β-estradiol, estriol in urine sample, based on precolumn derivatization with p-nitrobenzoyl chloride, is presented in this paper. The estrogens mentioned above in urine were firstly hydrolyzed with 0.6 mol/l HCl, and then enriched and cleaned-up by ENV-18 C18 solid phase extraction (SPE) column. The estrogens on column were eluted with dichloromethane, and the eluent was evaporated to dryness under gentle nitrogen flow. The residue was allowed to react with p-nitrobenzoyl chloride at 25 °C for 30 min. Separation was performed on a C18 column with gradient elution using acetonitrile and water as mobile phase. A fluorescence detection system was used to detect the fluorescent derivatization products. The detection limit of the method was 2.7 μg/l for bisphenol A and 17β-estradiol, 2.9 μg/l for 4-nonylphenol, 4.6 μg/l for 17α-estradiol and 17α-ethinylestradiol and 8.3 μg/l for estriol, respectively. The relative standard deviations (R.S.D.) ranged from 1.29 to 4.52% and the recoveries ranged from 85.5 to 99.9%. The method was applied to the determination of those six estrogens mentioned above in human urine samples collected from 20 healthy volunteers (aged 21-29). Bisphenol A (BPA) and 4-nonylphenol (NP) were detected with average contents of 1.22 ± 1.38 mg/l and 0.38 ± 0.77 mg/l in 10 male urine samples and 1.29 ± 1.22 mg/l and 0.05 ± 0.05 mg/l in 10 female urine samples, respectively. 17α-ethinylestradiol (α-EE2) was also detected with average contents of 0.13 ± 0.41 mg/l and 0.06 ± 0.15 mg/l in male and female urine samples, respectively.  相似文献

There has been considerable public interest and a growing number of scientific studies linking certain phenolic compounds in grapes and wines, particularly trans-resveratrol (trans-3,5,4′-trihydroxystilbene, TRA), to human health benefits. Typical TRA concentrations in wine are very low. It is a polar compound with very low volatility, which makes it difficult to extract and to separate on a gas chromatography (GC) column without derivatization. In this study, a new method for trace analysis of TRA was developed using solid-phase microextraction (SPME) with on-fiber silylation derivatization. Multidimensional GC equipped with a heartcut valve and cryogenic focusing was coupled with a mass-selective detector and used for improved separations and analysis. The effects of SPME fiber selection, extraction time, temperature, and desorption time were investigated. The derivatization conditions, time/temperature and the volume of derivatization reagent were also optimized. The calibration curve was linear over the concentration range from 10 ng L−1 to 5 mg L−1, with a correlation coefficient of 0.9996. The average recovery of TRA in red wine was 83.6 ± 5.6%. The method detection limit (MDL) for TRA in ethanol:water (12.5:87.5, v/v) solution in this study was 7.08 ng L−1 whereas the MDL for TRA in pure water was 2.85 ng L−1. The new method was used to test the TRA content in six selected Iowa red wine samples. Measured concentrations varied from 12.72 to 851.9 μg L−1.  相似文献

The solid phase microextraction (SPME) technique with on-fiber derivatization was evaluated for the analysis of alkylphenols (APs), including 4-tert-octylphenol (4-t-OP), technical nonylphenol isomers (t-NPs) and 4-nonylphenol (4-NP), in water. The 85 μm polyacrylate (PA) fiber was used and a two-step sample preparation procedure was established. In the first step, water sample of 2 mL was placed in a 4 mL PTFE-capped glass vial. Headspace extraction of APs in water was then performed under 65 °C for 30 min with 800 rpm magnetic stirring and the addition of 5% of sodium chloride. In the second step, the SPME fiber was placed in another 4 mL vial, which contained 100 μL of N-tert-butyl-dimethylsilyl-N-methyltrifluoroacetamide (MTBSTFA) with 1% tert-butyl-dimethylchlorosilane (TBDMCS). Headspace extraction of MTBSTFA and on-fiber derivatization with APs were performed at 45 °C for 10 min. Gas chromatography/mass spectrometry (GC/MS) was used for the analysis of derivatives formed on-fiber. The adsorption-time profiles were also examined. The precision, accuracy and method detection limits (MDLs) for the analysis of all the APs were evaluated with spiked water samples, including detergent water, chlorinated tap water, and lake water. The relative standard deviations were all less than 10% and the accuracies were 100 ± 15%. With 2 mL of water sample, MDLs were in the range of 1.58-3.85 ng L−1. Compared with other techniques, the study described here provided a simple, fast and reliable method for the analysis of APs in water.  相似文献

Estrone (E1), estradiols (α/β-E2), and estriol (E3) are four major metabolically active estrogens exerting strong biological activities at very low circulating concentrations. This paper reports a sensitive and efficient method with automated, on-line clean-up and detection to determine trace estrogens in a small volume of serum samples using liquid chromatography–electrospray ionization–tandem mass spectrometry directly, without off-line liquid–liquid or solid-phase extraction pretreatments. Serum aliquots (charcoal stripped fetal bovine serum, 100 μL) were spiked with four estrogen standards and their corresponding isotope-labeled internal standards, then bulk derivatized with 2-fluoro-1-methyl-pyridium p-toluenesulfonate (2-FMP) to establish the calibration curves and perform method validation. Calibration was established in the concentration ranges of 5–1000 pg mL−1, and demonstrated good linearity of R2 from 0.9944 to 0.9997 for the four derivatized estrogens. The lower detection limits obtained were 3–7 pg mL−1. Good accuracy and precision in the range of 86–112% and 2.3–11.9%, respectively, were observed for the quality control (QC) samples at low, medium, and high concentration levels. The stability tests showed that the derivatized serum samples were stable 8 h after derivatization at room temperature and at least to 48 h if stored at −20 °C. The method was applied to measure trace estrogens in real human and bovine serum samples, and three of four estrogen compounds studied were observed and quantified.  相似文献

p-Nitrophenyl organophosphates (OPs) including paraoxon, parathion and methyl parathion, etc, are highly poisonous OPs, for which sensitive and rapid detection method is most needed. In this work, an ultrasensitive electrochemical sensor for the determination of p-nitrophenyl OPs was developed based on ordered mesoporous carbons (OMCs) modified glassy carbon electrode (GCE) (OMCs/GCE). The electrochemical behavior and reaction mechanism of p-nitrophenyl OPs at OMCs/GCE was elaborated by taking paraoxon as an example. Experimental conditions such as buffer pH, preconcentration potential and time were optimized. By using differential pulse voltammetry, the current response of the sensor at −0.085 V was linear with concentration within 0.01–1.00 μM and 1.00–20 μM paraoxon. Similar linear ranges of 0.015–0.5 μM and 0.5–10 μM were found for parathion, and 0.01–0.5 μM and 0.5–10 μM for methyl parathion. The low limits of detection were evaluated to be 1.9 nM for paraoxon, 3.4 nM for parathion and 2.1 nM for methyl parathion (S/N = 3). Common interfering species had no interference to the detection of p-nitrophenyl OPs. The sensor can be applicable to real samples measurement. Therefore, a simple, sensitive, reproducible and cost-effective electrochemical sensor was proposed for the fast direct determination of trace p-nitrophenyl OPs at low potential without deoxygenization.  相似文献

In this study, pKa values were determined using the dependence of the retention factor on the pH of the mobile phase for three ionizable substances, namely, enalapril, lercanidipine and ramipril (IS). The effect of the mobile phase composition on the ionization constant was studied by measuring the pKa at different methanol-water mixtures, ranging between 50 and 65% (v/v), using LC-DAD method. Two simple, accurate, precise and fully validated analytical methods for the simultaneous determination of enalapril and lercanidipine in combined dosage forms have been developed. Separation was performed on an X-Terra RP-18 column (250 mm × 4.60 mm ID × 5 μm) at 40 °C with the mobile phase of methanol-water 55:45 (v/v) adjusted to pH 2.7 with 15 mM orthophosphoric acid. Isocratic elution was performed in less than 12 min with a flow rate of 1.2 mL min−1. Good sensitivity for the analytes was observed with DAD detection. The LC method allowed quantitation over the 0.50-20.00 μg mL−1 range for enalapril and lercanidipine. The second method depends on first derivative of the ratio-spectra by measurements of the amplitudes at 219.7 nm for enalapril and 233.0 nm for lercanidipine. Calibration graphs were established for 1-20 μg mL−1 for enalapril and 1-16 μg mL−1 lercanidipine, using first derivative of the ratio spectrophotometric method. Both methods have been extensively validated. These methods allow a number of cost and time saving benefits. The described methods can be readily utilized for analysis of pharmaceutical formulations. The methods have been applied, without any interference from excipients, for the simultaneous determination of these compounds in tablets. There was no significant difference between the performance of the proposed methods regarding the mean values and standard deviations.  相似文献

An ion-pairing reversed-phase liquid chromatography–mass spectrometry (IP-RP-LC–MS) was developed for the determination of nucleotides, nucleosides and their transformation products in Cordyceps. Perfluorinated carboxylic acid, namely pentadecafluorooctanoic acid (PDFOA, 0.25 mM), was used as volatile ion-paring agent and a reversed-phase column (Agilent ZORBAX SB-Aq column) was used for the separation of three nucleotides namely uridine-5′-monophosphate (UMP, 0.638–10.200 μg/mL), adenosine-5′-monophosphate (AMP, 0.24–7.80 μg/mL) and guanosine-5′-monophosphate (GMP, 0.42–13.50 μg/mL), seven nucleosides including adenosine (0.55–8.85 μg/mL), guanosine (0.42–6.75 μg/mL), uridine (0.33–10.50 μg/mL), inosine (0.21–6.60 μg/mL), cytidine (0.48–15.30 μg/mL), thymidine (0.20–6.30 μg/mL) and cordycepin (0.09–1.50 μg/mL), as well as six nucleobases, adenine (0.22–6.90 μg/mL), guanine (0.26–4.20 μg/mL), uracil (0.38–12.15 μg/mL), hypoxanthine (0.13–4.20 μg/mL), cytosine (0.39–12.45 μg/mL) and thymine (0.26–8.25 μg/mL) with 5-chlorocytosine arabinoside as the internal standard. The overall LODs and LOQs were between 0.01–0.16 μg/mL and 0.04–0.41 μg/mL for the 16 analytes, respectively. The contents of 16 investigated compounds in natural and cultured Cordyceps were also determined and compared after validation of the developed IP-RP-LC-MS method. The transformations of nucleotides and nucleosides in Cordyceps were evaluated based on the quantification of the investigated compounds in three extracts, including boiling water extraction (BWE), 24 h ambient temperature water immersion (ATWE) and 56 h ATWE extracts. Two transformation pathways including UMP → uridine → uracil and GMP → guanosine → guanine were proposed in both natural Cordyceps sinensis and cultured Cordyceps militaris. The pathway of AMP → adenosine → inosine → hypoxanthine was proposed in natural C. sinensis, while AMP → adenosine → adenine in cultured C. militaris. However, the transformation of nucleotides and nucleosides was not found in commercial cultured C. sinensis.  相似文献

The paper presents a new method based on simultaneous derivatization and air-assisted liquid–liquid microextraction (AALLME) for the extraction and preconcentration of some aliphatic amines prior to gas chromatography-flame ionization detection (GC-FID). Primary aliphatic amines are derivatized and extracted simultaneously by a fast reaction with butylchloroformate (derivatization agent/extraction solvent) under mild conditions. The mixture of butylchloroformate and aqueous sample solution is rapidly sucked into a 10-mL glass syringe and then is injected into a test tube with conical bottom and the procedure is repeated seven times. After centrifuging the resulted cloudy solution, the derivatized analytes in the sedimented phase are determined by GC-FID. The influence of main factors on the efficiency of derivatization/extraction procedure is studied. Under the optimal conditions, the enrichment factors (EFs) for aliphatic amines are obtained in the range of 248–360 and limits of detection (LODs) are between 0.30 and 2.6 μg L−1. The obtained extraction recoveries ranged from 50 to 72% and the relative standard deviation (RSD) was less than 4.8% for intra-day (n = 6) and inter-days (n = 4) precision. The method is successfully applied to determine some aliphatic amines in environmental water samples.  相似文献

A sensitive and selective method was developed for the determination of N-acetyl-p-aminophenol (APAP) and p-aminophenol (PAP) using poly(3,4-ethylenedioxythiophene) (PEDOT)-modified glassy carbon electrode (GCE). Cyclic voltammetry and differential pulse voltammetry were used to investigate the electrochemical reaction of APAP and PAP at the modified electrode. Both APAP and PAP showed quasireversible redox reactions with formal potentials of 367 mV and 101 mV (vs. Ag/AgCl), respectively, in phosphate buffer solution of pH 7.0. The significant peak potential difference (266 mV) between APAP and PAP enabled the simultaneous determination both species based on differential pulse voltammetry. The voltammetric responses gave linear ranges of 1.0 × 10−6-1.0 × 10−4 mol L−1 and 4.0 × 10−6-3.2 × 10−4 mol L−1, with detection limits of 4.0 × 10−7 mol L−1 and 1.2 × 10−6 mol L−1 for APAP and PAP, respectively. The method was successfully applied for the determination of APAP and PAP in pharmaceutical formulations and biological samples.  相似文献

The chromatographic fingerprints of industrial o-toluic acid, m-toluic acid and p-toluic acid have been established by HPLC-UV detection according to their impurity groups. HPLC separation of all relative substances involved in the groups was developed on a Kromasil C18 column by using methanol-water-NH4Ac-HAc buffer (100 mM, pH 4.70) 15/65/20 (v/v/v) as the mobile phase at a flow rate of 1.5 mL/min, and detection was operated by UV adsorption at a wavelength of 254 nm. The ultraviolet spectra corresponding to each chromatographic peak were also recorded for further identification of all components. Whether the limits of relative impurities residues in a toluic acid product are qualified or not can be intuitively estimated by analyzing its chromatogram with comparison to the fingerprint. This protocol has successfully provided some Chinese manufacturers with a simple and feasible method for quality control of toluic acids for industrial use.  相似文献

A microwave-assisted extraction used to extract trace triorganotin from aquatic organisms and a sensitive analytical method for the determination of ultratrace triorganotin (namely trimethyltin, triethyltin, tripropyltin and tributyltin) with capillary electrophoresis-inductively coupled plasma mass spectrometry were firstly described in this study. The extraction method is simple, effective and can be used to extract trace triorganotin in aquatic organisms within several min. The analytical method has a much lower detection limit of 0.2-0.7 ng Sn/mL for triorganotin compounds, and can be used to determine trace triorganotin in aquatic organisms directly without any derivatization and preconcentration. Using above methods, we have successfully determined trimethyltin, triethyltin, tripropyltin and tributyltin in dried Mya arenaria Linnaeus and Corbicula fluminea within 17 min with a recovery of 93-104% and a RSD (relative standard deviation, n = 6) of 2-5%. Our results showed that dried M. arenaria Linnaeus contained an extremely high tributyltin of 5.1 μg Sn/g dried weight, indicating that it may be a good biomarker for the organotin pollution in ocean.  相似文献

Hydrophobic interaction chromatography coupled online with chemical vapour atomic fluorescence spectrometry (HIC-CVGAFS) has been optimized for the analysis of thiolic proteins in denaturing conditions. Proteins are pre-column simultaneously denatured and derivatized in phosphate buffer solution containing 8.0 mol dm−3 urea and p-hydroxymercurybenzoate (PHMB) and the derivatized denatured proteins are separated on a silica HIC Eichrom Propyl column in the presence of 8.0 M urea in the mobile phase. Post-column online reaction of derivatized denatured proteins with bromine, generated in situ by KBr/KBrO3 in HCl medium, allowed the fast conversion of the uncomplexed PHMB and of the PHMB bound to proteins to inorganic mercury also in presence of urea. Hg2+, present in solution as Hg2+-urea complex, is selectively detected by AFS in a Ar/H2 miniaturized flame after sodium borohydride reduction to Hg. Under optimized conditions, online bromine treatment gives a 100±2% recovery of both free and protein-complexed PHMB. Denatured glyceraldehyde-3-phosphate dehydrogenase, aldolase, lactate dehydrogenase, trioso phosphate isomerase and β-lactoglobulin have been examined. As the sensitivity and limit of detection of proteins in the HIC-CVGAFS apparatus depends on number of SH groups reacting with PHMB, the denaturation process, which increases the number of PHMB-reactive thiolic groups in proteins, improves the analytical performances of the described system in protein analysis. The detection limit for the denatured proteins examined was found in the range of 10−10-10−12 mol dm−3, depending on the considered protein, with linear calibration curves spanning over four decades of concentration.  相似文献
