A microscope is described in which singlet molecular oxygen, O2(a1deltag), is produced in a femtoliter focal volume via a nonlinear two-photon photosensitized process, and the 1270 nm phosphorescence from this population of O2(a1deltag) is detected in a photon counting experiment. Although two-photon excitation of a sensitizer is less efficient than excitation by a one-photon process, nonlinear excitation has several distinct advantages with respect to the spatial resolution accessible. Pertinent aspects of this two-photon O2(a1deltag) microscope were characterized using bulk solutions of photosensitizers. These data were compared to those obtained from a single biological cell upon linear one-photon excitation of a sensitizer incorporated in the cell. On the basis of the results obtained, we outline the challenges of using nonlinear optical techniques to create O2(aldeltag) at the single cell level and to then optically detect the O2(aldeltag) thus produced in a time-resolved experiment.  相似文献

Singlet molecular oxygen (a(1)Delta(g)) has been produced and optically detected upon two-photon nonlinear excitation of a sensitizer with a focused laser beam. The experiments were performed using toluene solutions with either a substituted difuranonaphthalene or a substituted distyryl benzene as the sensitizer. The data indicate that the two-photon absorption cross sections of the difuranonaphthalenes are comparatively large and depend significantly on the functional groups attached to the chromophore. The time-resolved 1270 nm phosphorescence signals used to characterize the production of singlet oxygen are limited in much the same way as signals from other two-photon spectroscopic studies (e.g., weak signals that can be masked by scattered radiation). Nevertheless, the two-photon singlet oxygen signals also reflect the unique advantages of this nonlinear optical technique (e.g., depth penetration in the sample afforded by irradiation in a spectral region void of the more dominant one-photon linear transitions and spatial resolution afforded by irradiation with a focused laser beam).  相似文献

Two-photon excitation spectra have been recorded over the large spectral range of 540-1000 nm for five phenylene-vinylene oligomers that differ in the length of the conjugated pi system. The significant changes observed in the two-photon excitation spectra and absorption cross sections as a function of this systematic change in the chromophore are discussed in light of (1) the corresponding one-photon absorption spectra and (2) high-level density functional response theory calculations performed on analogues of these systems. The results obtained illustrate one way to exploit parameters that influence nonlinear optical properties in large organic molecules. Specifically, data are provided to indicate that when the frequency of the laser used in the two-photon experiment is nearly-resonant with an allowed one-photon transition, significant increases in the two-photon absorption cross section can be realized. This phenomenon of the so-called resonance enhancement allows for greater control in obtaining an optimal response when using existing two-photon chromophores, and provides a much-needed guide for the systematic development and efficient use of two-photon singlet oxygen sensitizers.  相似文献

The yield of singlet molecular oxygen, O2(a(1)Delta(g)), produced in a photosensitized process can be very susceptible to environmental perturbations. In the present study, protonation of photosensitizers whose chromophores contain amine functional groups is shown to adversely affect the singlet oxygen yield. Specifically, for bis(amino) phenylene vinylenes dissolved both in water and in toluene, addition of a protic acid to the solution alters properties of the system that, in turn, result in a decrease in the efficiency of singlet oxygen production. In light of previous studies on other molecules where protonation-dependent changes in the yield of photosensitized singlet oxygen production have been ascribed to changes in the quantum yield of the sensitizer triplet state, Phi(T), and to possible changes in the triplet state energy, E(T), our results demonstrate that this photosystem can respond to protonation in other ways. Although protonation-dependent changes in the amount of charge-transfer character in the sensitizer-oxygen complex may influence the singlet oxygen yield, it is likely that other processes also play a role. These include (a) protonation-dependent changes in sensitizer aggregation and (b) nonradiative channels for sensitizer deactivation that are enhanced as a consequence of the reversible protonation/deprotonation of the chromophore. The data obtained, although complicated, are relevant for understanding and ultimately controlling the behavior of photosensitizers in systems with microheterogeneous domains that have appreciable pH gradients. These data are particularly important given the use of such bi-basic chromophores as two-photon singlet oxygen sensitizers, with applications in spatially resolved singlet oxygen experiments (e.g., imaging experiments).  相似文献

We study the porphyrin S1→S0 fluorescence and the photosensitized singlet oxygen 1Δg3Σg phosphorescence, both originating from absorption of photons with energy less than the porphyrin S0→S1 transition energy. By measuring the excitation intensity dependence of fluorescence at lowered sample temperatures, we are able to discriminate between two parallel processes of one-photon hot-band absorption (HBA) and simultaneous two-photon absorption (TPA). When the HBA and TPA contributions are comparable in magnitude, we use this new method to determine absolute TPA cross-section. We also demonstrate for the first time a singlet oxygen photosensitization via HBA in porphyrin.  相似文献

Porphycenes are structural isomers of porphyrins that have many unique properties and features. In the present work, the resonant two-photon absorption of 2,7,12,17-tetraphenylporphycene (TPPo) and its palladium(II) complex (PdTPPo) has been investigated. The data obtained are compared to those from the isomeric compound, meso-tetraphenylporphyrin (TPP). Detection of phosphorescence from singlet molecular oxygen, O2(a(1)Delta(g)), produced upon irradiation of these compounds, was used to obtain two-photon excitation spectra and to quantify two-photon absorption cross sections, delta. In the spectral region of 750-850 nm, the two-photon absorption cross sections at the band maxima for both TPPo and PdTPPo, delta = 2280 and 1750 GM, respectively, are significantly larger than that for TPP. This difference is attributed to the phenomenon of so-called resonance enhancement; for the porphycenes, the two-photon transition is nearly resonant with a comparatively intense one-photon Q-band transition. The results of quantum mechanical calculations using density functional quadratic response theory are in excellent agreement with the experimental data and, as such, demonstrate that comparatively high-level quantum chemical methods can be used to interpret and predict nonlinear optical properties from such large molecular systems. One important point realized through these experiments and calculations is that one must exercise caution when using qualitative molecular-symmetry-derived arguments to predict the expected spectral relationship between allowed one- and two-photon transitions. From a practical perspective, this study establishes that, in comparison to porphyrins and other tetrapyrrolic macrocyclic systems, porphycenes exhibit many desirable attributes for use as sensitizers in two-photon initiated photodynamic therapy.  相似文献

The spectral properties of new fluorene-based photosensitizers for efficient singlet oxygen production are investigated at room temperature and 77 K. Two-photon absorption (2PA) cross-sections of the fluorene derivatives are measured by the open aperture Z-scan method. The quantum yields of singlet oxygen generation under one- and two-photon excitation (phi(delta) and 2PAphi(delta), respectively), are determined by the direct measurement of singlet oxygen luminescence at approximately 1270 nm. The values of phi(delta) are independent of excitation wavelength, ranging from 0.6-0.9. The singlet oxygen quantum yields under two-photon excitation are 2PAphi(delta) approximately 1/2 phi(delta), indicating that the two processes exhibit the same mechanism of singlet oxygen production, independent of the mechanism of photon absorption.  相似文献

Singlet molecular oxygen (a(1)Delta(g)) has been produced and optically detected in time-resolved experiments upon nonlinear two-photon excitation of a photosensitizer dissolved in water. For a given sensitizer, specific functional groups that impart water solubility and that give rise to larger two-photon absorption cross sections are, in many cases, not conducive to the production of singlet oxygen in high yield. This issue involves the competing influence of intramolecular charge transfer that can be pronounced in aqueous systems; more charge transfer in the chromophore facilitates two-photon absorption but decreases the singlet oxygen yield. The excited-state symmetry and molecular reorientation of perylene, 1,7-diazaperylene, and 2,5,8,11-tetra- tert-butylperylene have been studied by different fluorescence depolarization experiments. The first excited electronic singlet state was reached through one-photon excitation (OPE) and two-photon excitation (TPE). A 400 and 800 nm femtosecond laser pulse was used for this purpose, and data were collected by means of the time-correlated single-photon counting technique. It is found that the rotational correlation times for each perylene derivative are very similar in the OPE and TPE depolarization experiments. For the determination of the two-photon absorption tensor, a recently described theoretical model has been applied (Ryderfors et al. J. Phys. Chem. A 2007, 111, 11531). It was found that the two-photon process can be described by a 2 x 2 absorption tensor for which the components are solvent dependent and exhibit mixed vibronic character. In the dipole approximation this is compatible with a parity-forbidden two-photon absorption into the first excited singlet state.  相似文献

Singlet oxygen sensitization by organic molecules is a topic of major interest in the development of both efficient photodynamic therapy (PDT) and aerobic oxidations under complete green chemistry conditions. We report on the design, synthesis, biology, and complete spectroscopic characterization (vis-NIR linear and two-photon absorption spectroscopy, singlet oxygen generation efficiencies for both one- and two-photon excitation, electrochemistry, intrinsic dark toxicity, cellular uptake, and subcellular localization) of three classes of innovative singlet oxygen sensitizers pertaining to the family of symmetric squaraine derivatives originating from pi-excessive heterocycles. The main advantage of pi-extended squaraine photosensitizers over the large number of other known photosensitizers is their exceedingly strong two-photon absorption enabling, together with sizable singlet oxygen sensitization capabilities, for their use at the clinical application relevant wavelength of 806 nm. We finally show encouraging results about the dark toxicity and cellular uptake capabilities of water-soluble squaraine photosensitizers, opening the way for clinical small animal PDT trials.  相似文献

Precise two-photon absorption spectra of the green fluorescent protein (GFP) and the mutants sapphire-GFP (T203I) and enhanced GFP (S65T/F64L), as well as a model compound for the chromophore, 4'-hydroxybenzylidene-2,3-dimethylimidazolinone (HBDI) were measured by multiplex two-photon absorption spectroscopy. The observed TPA bands of the anionic forms of enhanced GFP and HBDI were significantly shifted to the higher energy compared with the lowest-energy bands in one-photon absorption spectra. This result indicated the existence of a hidden electronic excited state in the vicinity of the lowest excited singlet (S1) state of the anionic form of the GFP chromophore, which is the origin of the blue shift of the two-photon absorption spectra as well as two-photon fluorescence excitation spectra.  相似文献

Resonance Raman and resonance hyper-Raman spectra of the "push-pull" conjugated molecule 1-(4'-dihexylaminostyryl)-4-(4"-nitrostyryl)benzene in acetone have been measured at excitation wavelengths from 485 to 356 nm (two-photon wavelengths for the nonlinear spectra), resonant with the first two bands in the linear absorption spectrum. The theory of resonance hyper-Raman scattering intensities is developed and simplified using assumptions appropriate for intramolecular charge-transfer transitions of large molecules in solution. The absorption spectrum and the Raman, hyper-Rayleigh, and hyper-Raman excitation profiles, all in absolute intensity units, are quantitatively simulated to probe the structures and the one- and two-photon transition strengths of the two lowest-energy allowed electronic transitions. All four spectroscopic observables are reasonably well reproduced with a single set of excited-state parameters. The two lowest-energy, one-photon allowed electronic transitions have fairly comparable one-photon and two-photon transition strengths, but the higher-energy transition is largely localized on the nitrophenyl group while the lower-energy transition is more delocalized.  相似文献

The time-of-flight spectra of photofragments produced by a one- and two-photon dissociation of NOCl by 5 eV (248 nm) and 6.4 eV (193 nm) photons from an excimer laser have been measured. The results show that the primary fragments are excited NO and chlorine atoms in the electronic ground state doubl In the case of the one-photon dissociation the NO-fragments are formed highly excited in the X-state. The spectra of excitation for the two different photon energies used in the experiment can be explained by a simple model calculation assuming a direct dissociation from the same intermediate state. The observation of the two-photon dissociation could only be achieved by the analysis of the time-of-flight spectra of NO ions produced by ionizing the primary NO-fragments with the dissociating laser. This method of observation can improve the sensitivity of detection (also for the one-photon process) The results indicate that the NO-fragments from the two-photon dissociation are formed most probably in the Rydberg states of NO.  相似文献

Two-photon-resonant hyper-Raman spectra are reported for three "push-pull" conjugated organic chromophores bearing -NO(2) acceptor groups, two dipolar and one octupolar. The excitation source is an unamplified picosecond mode-locked Ti:sapphire laser tunable from 720 to 950 nm. The linear resonance Raman spectra of the same molecules are measured using excitation from the laser second harmonic. Excitation on resonance with the lowest-lying band in the linear absorption spectrum yields nearly identical resonance Raman and resonance hyper-Raman spectra. However, excitation into a region that appears to contain more than one electronic transition gives rise to different intensity patterns in the linear and nonlinear spectra, indicating that different transitions contribute differently to the one-photon and two-photon oscillator strength. The promise of the hyper-Raman technique for examining electronic transitions that are both one- and two-photon allowed is discussed.  相似文献

The effects of a solvent on the two-photon absorption of microsolvated formaldehyde and liquid water have been studied using hybrid coupled-cluster/molecular mechanics (CC/MM) response theory. Both water and formaldehyde were considered solvated in water, where the solvent water molecules were described within the framework of molecular mechanics. Prior to the CC/MM calculations, molecular dynamics simulations were performed on the water/formaldehyde and water/water aggregates and many configurations were generated. By carrying out CC/MM response calculations on the individual configurations, it was possible to obtain statistically averaged results for both the excitation energies and two-photon absorption cross sections. For liquid water, the comparison between one- and two-photon absorption spectra is in good agreement with the experimental data available in the literature. In particular, the lowest energy transition occurring in the one-photon absorption spectrum of water only occurs with a relatively small strength in the two-photon absorption spectrum. This result is important for the interpretation of two-photon absorption data as these results show that in the absence of selection rules that determine which transitions are forbidden, the spectral profile of the two-photon absorption spectrum can be significantly different from the spectral profile of the one-photon absorption spectrum.  相似文献

Under high-excitation irradiance conditions in one- and two-photon induced fluorescence microscopy, the photostability of fluorescent dyes is of crucial importance for the detection sensitivity of single molecules and for the contrast in fluorescence imaging. Herein, we report on the dependence of photobleaching on the excitation conditions, using the dye Rhodamine 6G as a typical example. The different excitation modes investigated include 1) one-photon excitation into the first-excited singlet state in the range of 500 to 528 nm by continuous wave and picosecond-pulsed lasers and 2) two- and one-photon excitation to higher-excited singlet states at 800 and 350 nm, respectively, by femtosecond pulses. Experimental strategies are presented, which allow resolving the photophysics. From single-molecule trajectories and fluorescence correlation spectroscopy, as well as with a simple theoretical model based on steady-state solutions of molecular rate equation analysis, we determined the underlying photobleaching mechanisms and quantified the photokinetic parameters describing the dependence of the fluorescence signal on the excitation irradiance. The comparison with experimental data and an exact theoretical model show that only minor deviations between the different theoretical approaches can be observed for high-pulsed excitation irradiances. It is shown that fluorescence excitation is in all cases limited by photolysis from higher-excited electronic states. In contrast to picosecond-pulsed excitation, this is extremely severe for both one- and two-photon excitation with femtosecond pulses. Furthermore, the photostability of the higher-excited electronic states is strongly influenced by environmental conditions, such as polarity and temperature.  相似文献

The linear and non-linear photophysical properties of tris-dipicolinate europium and terbium complexes (absorption, emission, lifetime, luminescence induced by two-photon absorption) are studied in the crystalline state as well as in protein derivative crystals and compared to those in solution. Upon laser irradiation at 532 nm, luminescence of terbium is induced by a two-photon antenna effect, whereas luminescence of europium results from one-photon absorption in forbidden f-f transitions. Finally, linear and two-photon microscopy imaging experiments on biological and bio-inspired crystals are performed. These first proof-of-concept experiments open the way for the development of time-resolved non-linear microscopy that should combine the advantages of lanthanide luminescence (long lifetime, sharp emission bands, insensitivity to oxygen) with those of confocal biphotonic excitation (near-IR excitation, 3D resolution and reduced photodamage).  相似文献

Abstract— Triplet-triplet absorption spectra of a series of carotenoid pigments in benzene solution have been determined by pulse radiolysis experiments. The natural lifetimes in deaerated solution have also been measured. They fall in the range 2–10 µ s as found for other carotenoids under similar conditions. Pulsed laser (337 nm) excitation of benzene solutions containing oxygen, carotenoid and a photosensitized molecule (anthracene) showed the generation of absorption spectra of the triplet states. These absorptions decayed by first order kinetics in such a way as to indicate that they were formed in reactions with singlet oxygen, itself generated by interaction with the anthracene triplet state. Bimolecular rate constants for energy transfer from O*2 (1g), to carotenoid have been evaluated.  相似文献
