Supercritical fluid chromatography with tandem mass spectrometry was used to comprehensively profile polyene phosphatidyl choline (PPC) extracted from soybean. We achieved an efficient chromatographic analysis using a BEH‐2EP column (3 × 100 mm2, 1.7 μm) with a mobile phase consisting of CO2 and a cosolvent in gradient combination at a flow rate of 1.0 mL/min. The cosolvent consisted of methanol, acetonitrile, and water (containing 10 mM ammonium acetate and 0.2% formic acid). The total single‐run time was 7 min. We used this method to accurately detect ten different phospholipids (PLs) during extraction. The limits of quantification for phosphatidyl choline, lyso‐phosphatidylcholine (LPC), phosphatidic acid (PA), sphingomyelin, phosphatidyl glycerol, phosphatidyl inositol (PI), cholesterol, cardiolipin, phosphatidyl serine, and phosphatidyl ethanolamine (PE) were 20.6, 19.52, 1.21, 2.38, 0.50, 2.28, 54.3, 0.60, 0.65, and 4.85 ng/mL, respectively. However, adopting the high‐performance liquid chromatography with evaporative light scattering detection method issued by the China Food and Drug Administration, only PA, LPC, PE, PI, and PPC could be analyzed accurately, and the limits of quantification were 33.89, 60.5, 30.3, 10.9, and 61.79 μg/mL, respectively. The total single‐run time was at the least 20 min. Consequently, the supercritical fluid chromatography with tandem mass spectrometry method was more suitable for the analysis of related PLs.  相似文献

The first supercritical fluid chromatography method for the determination of five major coumarins (dihydrosamidin, visnadin, samidin, khellin, and visnagin) in Ammi visnaga fruits is described. Their baseline separation was possible in less than 5 min by using a UPC2 HSS C18 SB column with 1.8 μm particle size and a mobile phase comprising CO2, methanol, acetonitrile, and diethylamine. The type of stationary phase used was of particular relevance because, except for the selected one, the others did not resolve the two structural isomers dihydrosamidin and visnadin. Method validation confirmed that the procedure is linear (R2 ≥ 0.9996) in a concentration range from 6 to 480 μg/mL, it is accurate (recovery rates: 97.2–103.6%) and precise (intraday deviation ≤ 6.6%, intraday deviation ≤ 1.7%); injecting 1 μL of standard solution, the determined limit of detection was below 1.9 μg/mL for all compounds. The analysis of different A. visnaga samples revealed their similar compositions, and khellin (0.75–1.01%) and visnagin (0.18–0.46%) were the dominant coumarins. Visnadin and dihydrosamidin, the individual quantification of which is described for the first time, were present at concentrations below 0.14%.  相似文献

The standard molar enthalpy of combustion of cholesterol was measured at constant volume. According to value of Δr U mθ(−14358.4±20.65 kJ mol−1), Δr H mθ(−14385.7 kJ mol−1) of combustion reaction and Δf H mθ(2812.9 kJ mol−1) of cholesterol were obtained from the reaction equation. The enthalpy of combustion reaction of cholesterol was also estimated by the average bond enthalpies. By design of a thermo-chemical recycle, the enthalpy of combustion of cholesterol were calculated between 283.15∼373.15 K. Besides, molar enthalpy and entropy of fusion of cholesterol was obtained by DSC technique.  相似文献

Solid dispersion technology was used to improve the bioavailability of probucol due to its low hydrophilicity and high lipophilicity. In this study, a highly rapid and sensitive supercritical fluid chromatography with tandem mass spectrometry method was optimized and validated for the determination of probucol in beagle dog plasma with diazepam as an internal standard. The analyte and internal standard were extracted by acetone and then separated on a polar 2‐ethylpyridine phase column (100 mm × 3 mm, 1.7 μm) at a flow rate of 1.0 mL/min using CO2 (≥99.99%) and methanol (95:5, v/v) as the mobile phase. The mass transition ion‐pair was m/z 515.6→236.2 and 285.2→193.1 for probucol and internal standard, respectively. Excellent linearity was observed over the concentration range of 5–5000 ng/mL (r2 ≥ 0.9999) with a lower limit of quantification of 5 ng/mL. The intra‐ and inter‐day accuracy and precision for all quality control samples were within ±15%. The proposed method was accurate, rapid and reproducible, which was successfully applied to a bioavailabilty evaluation of probucol solid dispersion tablets.  相似文献

Summary Methods have been developed for determination of semivolatile organic pollutants in air. Extraction efficiencies for test compounds are >83%, and rsd’s are <3.4% (n=3). Atmospheric samples were collected from a variety of sites in Lanzhou, China by an adsorptive enrichment method. Over 50 organic pollutants were extracted by SFE with CO2 modified with methanol. Extracts were subjected to GC and GC-MS analysis. The results showed that one of the most serious polluted regions was an oil refinery, in which total concentration of pollutants amounted to 285.75 μg·m−3. The relationships between the total concentration of pollutants and sampling time and weather were also investigated. Results showed that pollution on sunny days is more serious than that on rainy days, while pollution is most serious during the evening.  相似文献

A high‐throughput, rapid, sensitive, environmentally friendly, and economical supercritical fluid chromatography with triple quadruple mass spectrometry method was established and validated for the first time to determine a cerebral stroke treatment drug named 3‐n‐butylphthalide in dog plasma. Plasma samples were prepared by protein precipitation with methanol and the analytes were eluted on an ACQUITY UPC2TM HSS‐C18 SB column (3 × 100 mm, 1.8 μm) maintained at 50°C. The mobile phase comprised supercritical carbon dioxide/methanol (90:10, v/v) at a flow rate of 1.5 mL/min, the compensation solvent was methanol at a flow rate of 0.2 mL/min and the total run time was 1.5 min per sample. The detection was carried out on a tandem mass spectrometer with an electrospray ionization source. Calibration curves were linear over the concentration range of 1.02–1021.00 ng/mL (r2 ≥ 0.993) with the lower limit of quantification of 1.02 ng/mL. The intra‐ and inter‐day precision values were below 15% and the accuracy was from 97.90 to 103.70% at all quality control levels. The method was suitable for a pharmacokinetic study of 3‐n‐butylphthalide in beagle dogs.  相似文献

Summary Packed columns containing microparticles provide high column efficiency per unit time and strong retention characteristics compared with open tubular columns, and they are favored for fast separations. Nonporous particles eliminate the contribution of solute mass transfer resistance in the intraparticle void volume characteristic of porous particles, and they should be more suitable for fast separations. In this paper, the evaluation of nonporous silica particles of sizes ranging from 5 to 25 μm in packed capillary columns for fast supercritical fluid chromatography (SFC) using neat CO2 is reported. These particles were first deactivated using polymethyl-hydrosiloxanes and then encapsulated with a methylphenylpolysiloxane stationary phase. The retention factors, column efficiencies, column efficiencies per unit time, separation resolution, and separation resolution per unit time for fast SFC were determined for various length capillaries packed with various sizes of polymerencapsulated nonporous particles. It was found that 15 μm nonporous particles provided the highest column efficiency per unit time and resolution per unit time for fast packed capillary SFC. Under certain conditions, separations were completed in less than 1 min. Several thermally labile silylation reagent samples were separated in times less than 5 min. Two chromatographic techniques are described for the separation and quantitative determination of azadirachtin A and B, salannin, and nimbin present in the crude extract of neem seed kernels and commercial neem based formulations. The high performance thin-layer chromatography (HPTLC) separation of markers was carried out on Merck TLC aluminium sheets of silica gel 60 F254 using ethylacetate-benzene (7.0: 3.0, v/v) as mobile phase. The other technique was based on extended length packed column supercritical fluid chromatographic (PC-SFC) separation of the markers using Cyano column (250 mm × 4.6 mm I.D, S-5.0 μ) and Kromasil 100 NH2 column (250 mm × 4.6 mm I.D, 5.0 μ) connected in series. The detection was carried out using photodiode array detector at 338 K using methanol-modified carbon dioxide (10%) as the mobile phase at flow rate of 2.0 mL min−1. The current study assesses the effect of extending column length during PC-SFC experiment to obtain maximum resolution between a number of unknown components and known markers present in neem seed extracts. Both the chromatographic methods were validated in terms of precision, robustness, recovery, limits of detection and quantitation. The analysis of variance (ANOVA) and Student’s t-test were applied to correlate the results of quantitative determination of markers by means of HPTLC and PC-SFC method.  相似文献

Summary A mini extractor of 85 L void volume and a micro extractor of 3–4 L void volume have been coupled directly with a packed column SFC and used under sub- and supercritical conditions. The mini extractor is suitable for holding adsorbates which can be on-line extracted and the extract chromatographed (direct SFE-SFC). The micro extractor can be used for direct sample introduction of liquid and solid materials under SF conditions. Thus any solvent interference with the sample and the chromatographic conditions is excluded. Standard samples of wood tar residue, engine oil, and metal organic compounds have been tested.  相似文献

Capillary SFC was applied to the analysis of Some plant volatile oils. The results were compared with those obtained by capillary GC. For thyme oil, SFC without derivatization gave about the same percentage composition of the main compounds as CGC with silylation, thus eliminating one working step. For a complex mixture of peppermint oil or basil oil, SFC seems to give more reliable quantification than CGC, especially for oxygenated compounds. However, the separation efficiency of CGC for monoterpene hydrocarbons was, as expected, much better than that of SFC.  相似文献

A sensitive and specific liquid chromatographic method with electrospray ionization mass spectrometry (LC–ESI-MS) has been developed and validated for identification and quantification of mitiglinide in human urine. A simple liquid–liquid extraction procedure was followed by separation on a C18 column with gradient elution, and detection using a single-quadrupole mass spectrometer in selected-ion-monitoring (SIM) mode. The method was tested using six different batches of urine. Linearity was established for the mitiglinide concentrations in the range 0.005–1.0 μg mL−1, with a coefficient of determination (r) of 0.9998 and good back-calculated accuracy and precision. Intra- and inter-day precision (as RSD, %) was below 10% and accuracy for mitiglinide ranged from 85 to 115%. The lower limit of quantification was reproducible at 0.002 μg mL−1 for 500 μL urine. The proposed method enables unambiguous identification and quantification of mitiglinide in pre-clinical and clinical studies.  相似文献

Summary Peak compression of a dihydropyridine drug, clevidipine, is obtained in both analytical and semi-preparative scale supercritical fluid chromatography, resulting in extremely high apparent efficiencies. The observed effect, when utilising a carbon dioxide/2-propanol mobile phase with a bare silica stationary phase, is achieved when the retention of the clevidipine peak is controlled to coalesce with a system peak, generated as a result of having water in the sample. Apparent efficiencies of 350,000 and 170,000 plates meter−1 were obtained when 0.25 and 0.5 mg, respectively, are directly injected to a 200×4.6 mm ID 5 μm Hypersil silica packed column. The effect was extended to a semi-preparative system where apparent efficiencies in the region of 2,000,000 plates meter−1 were observed when 0.3 mg of a clevidipine sample containing 80% water was injected to a 250×10 mm I.D. column containing 5-μm Hypersil silica particles.  相似文献

Summary A method for the determination of the food mutagen 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP) in human hair has been developed and validated. Hair samples (200 mg) were dissolved in NaOH (1 M) and PhIP was isolated by successive solid-phase extraction on a polystyrene-divinylbenzene column and on a silica-based mixed-mode column with C8 and-SO3 functional groups. Quantification was performed by gas chromatography-electron-impact ionization high-resolution mass spectrometry in selected-ion-monitoring mode. The method was validated for determination of PhIP in the concentration range 0.5–25 ng g−1 hair with [2H3]PhIP as internal standard. The limit of quantification was 0.26 ng g−1 hair. Within-day and between-day precision were in the ranges 1–27% and 2–15% relative standard deviation, respectively. The hair sample used for method validation was found to contain 0.26 ng PhIP g−1 hair.  相似文献

A combined method involving electrochemical oxidation of iodide to iodate at a platinum electrode followed by extraction in CCl4 of ionic associates of iodine-iodide complexes with brilliant green, formed in excess of iodide, was developed for the spectrophotometric quantification of iodide. The slope of the calibration curve yields a molar extinction coefficient of ɛ = 3·105 L mol−1cm−1. This method can be used for the quantification of iodide in the concentration range of 3·10−7 − 3·10−6 mol L−1 with a detection limit of 5·10−8 mol L−1. The interfering effect of other ions on the determination of the iodide concentration was also investigated. The method was successfully applied for the determination of iodide in real samples of NaCl and spring water. Relative standard deviation is 1–2%.  相似文献

An HPLC method was developed and validated for the determination of the cephalosporins cefotaxime and cephalexine in skimmed bovine milk. The analytical column, Kromasil C18 (250 mm × 4.0 mm, 5 μm) was operated at ambient temperature. Mobile phase consisted of CH3OH-acetate buffer (pH = 4.0) and it was delivered isocratically at a flow rate of 1.0 mL · min−1. Total analysis time was less than 5 min. Caffeine was used as internal standard (5 ng · μL−1). UV detection was performed at 265 nm. Method validation was performed by means of intra-day (n = 5) and inter-day accuracy and precision (n = 8), sensitivity and linearity. Limits of detection (LOD) and limits of quantification (LOQ) were 0.1 and 0.3 ng · μL−1, respectively. The method was applied to the analysis of a veterinary drug (CEPOREX) containing cephalexine. The results were quite accurate with the relative error varying from −8.0 to −3.5%. Solid-phase extraction was applied to remove all matrix interference from milk samples. High extraction recoveries (average 84–121%) were achieved by using Abselut NEXUS cartridges with acetonitrile as eluent and a rinsing step with water and n-butanol. A pre-concentration step was necessary in a 1/10 level to reach the EU MRL concentration level (100 μg · kg−1). RSD values were less than 7% for both cephalosporins. Correspondence: Ioannis N. A method for the identification and quantification of underivatised steryl sulphates in invertebrates by liquid chromatography (LC) coupled with tandem mass spectrometry (MS) involving a single cleanup step has been developed. Negative electrospray ionisation and positive and negative atmospheric-pressure chemical ionisation (APCI) spectra of steryl sulphate showed pseudomolecular ions ([M+H–H2SO4]+or [M–H]). Collision-induced dissociation (CID) was efficient only in positive APCI. LC-MS in negative APCI was least susceptible to interference and possible differences in response factors. The detection limits (signal-to-noise ratio of 3) based on cholest-5-enyl-3-sulphate in positive and negative APCI modes are 3.66 and 0.73 pmol μL−1, respectively. Calibration plots and response factors for cholest-5-enyl-3-sulphate relative to the internal standard, cholecalciferyl-3-sulphate, in both positive and negative polarities, were linear in the concentration range from 1.22 to 16.4 pmol μL−1 with good coefficients of determination (R 2>0.98). It is suggested that the structure elucidation of steryl sulphates is best achieved in CID positive APCI mode, whereas their quantification should be carried out using negative APCI.  相似文献

A cheap, simple and rapid sample preparation method has been developed for quantification of ulifloxacin, the active metabolite of prulifloxacin in human plasma, by HPLC with fluorescence detection using lemefloxacin as the internal standard. One-step protein precipitation with 10% perchloric acid (2:1, v/v) on a 200 μL sample was used. The separation was performed at 30 °C on a C18 column using an eluent of acetonitrile-0.5% triethylamine buffer. The compounds were monitored at λ ex of 280 nm, λ em of 425 nm. The calibration curve for ulifloxacin in human plasma was linear over the range 0.01–1.00 μg mL−1. The lower limit of quantification is 0.01 μg mL−1. The intra- and inter-day precision ranged from 3.0 to 6.7%, respectively. The method had been used for clinical pharmacokinetic studies of prulifloxacin formulation product after oral administration to healthy volunteers. Nowadays, supercritical fluid technology (SFT) has been an interesting scientific subject in disparate industrial-based activities such as drug delivery, chromatography, and purification. In this technology, solubility plays an incontrovertible role. Therefore, achieving more knowledge about the development of promising numerical/computational methods of solubility prediction to validate the experimental data may be advantageous for increasing the quality of research and therefore, the efficacy of novel drugs. Decitabine with the chemical formula C8H12N4O4 is a chemotherapeutic agent applied for the treatment of disparate bone-marrow-related malignancies such as acute myeloid leukemia (AML) by preventing DNA methyltransferase and activation of silent genes. This study aims to predict the optimum value of decitabine solubility in CO2SCF by employing different machine learning-based mathematical models. In this investigation, we used AdaBoost (Adaptive Boosting) to boost three base models such as Linear Regression (LR), Decision Tree (DT), and GRNN. We used a dataset that has 32 sample points to make solubility models. One of the two input features is P (bar) and the other is T (k). ADA-DT (Adaboost Algorithm-Decision Tree), ADA-LR (Adaboost Algorithm-Linear Regresion), and ADA-GRNN (Generative Regression Neural Network) models showed MAE of 6.54 × 10?5, 4.66 × 10?5, and 8.35 × 10?5, respectively. Also, in terms of R-squared score, these models have 0.986, 0.983, and 0.911 scores, respectively. ADA-LR was selected as the primary model according to numerical and visual analysis. Finally, the optimal values are (P = 400 bar, T = 3.38 K × 102, Y = 1.064 × 10?3 mol fraction) using this model.  相似文献

A L18 (21 × 37) OAD was employed to investigate the effects of SFC parameters on the asymmetric factor of a basic compound on C18 columns. The effect of active sites of silanol group on SFC could be much more significant than that of HPLC due to the low viscosity and high diffusivity of the supercritical fluid. Three Waters C18 columns with different silanol activities were employed to examine the effect of the SFC parameters on the asymmetric factor of 5‐(α‐phenylethyl)‐semioxamazide. The OAD data were evaluated by ANOVA to determine significant parameters of SFC on the asymmetric factor. The results of ANOVA indicate that the silanol activity is the most dominant factor affecting the asymmetric factor. The influence of other parameters on the asymmetric factor follows the order of: modifier type > modifier concentration > additive type > pressure > temperature ~ additive concentration > injected sample concentration.  相似文献

A simple multi-residue analysis method for the quantitative determination of eprinomectin, abamectin, doramectin and ivermectin in bovine tissues was developed. The tissue sample was extracted with acetonitrile, followed by clean-up on a C18 solid phase extraction cartridge. The eluate was derivatised before being analyzed by HPLC coupled to a fluorescence detector. The method was validated using bovine liver and muscle fortified with the drugs at 0, 5, 10 and 50 ng g−1. The mean recoveries of the four drugs were 70.31–87.11% in liver and 79.57–93.65% in muscle, with relative standard deviations below 17.84% in liver and 14.68% in muscle. The limits of detection were between 0.5 and 1.0 ng g−1 and the limits of quantification were 1–2 ng g−1 in bovine tissues for the four drugs.  相似文献
