Abstract— The set of final products of thymine conversion induced by high-intensity UV irradiation (λ= 266nm, intensity 1024-5 × 1029 photons·s−1·m−2, pulse duration 10ns) of the dilute aqueous solution to the first approximation is similar to that formed with ionizing irradiation (γ-irradiation of aqueous solution or autoradiolysis of a solid 2-[14C]-thymine). The data obtained suggest that high-intensity UV-induced photochemical conversion of thymine involves photoionization and/or photodissociation. These processes pass through the higher excited state(s) populated as a result of the second photon absorption by excited (most probably in the T1 triplet state) thymine molecules.  相似文献

Abstract— A broad-band UVA source that emits primarily350–400 nm radiation and no measurable radiation below 340 nm was used to test toxicity and mutagenicity at the thymidine kinase locus in L5178Y, subclone 3.7.2C (TK+/-) mouse lymphoma cells. Cells were exposed to a fluence of 0 to 80 × 104 J/m2. The relationship between UVA fluence and survival was found to have a shoulder region followed by an exponential decrease in survival at higher fluence levels. An exposure-dependent increase in mutation was observed with increasing fluences from 0 to about 60 × 104 J/m2. An approximately 3- to 4-fold increase in mutations (trifluorothymidine resistance) over unexposed, control cells was seen at a fluence that resulted in 90% cell killing. We conclude that UVA radiation is a mutagen in the L5178Y mouse lymphoma cells used in this study.  相似文献

Abstract— Luminol-enhanced chemiluminescence in suspensions of human polymorphonuclear leucocytes stimulated with the chemotactic oligopeptide formyl-methionyl-leucyl-phenylalanine was increased by exposure of the cells to long wavelength UV radiation (mainly320–400 nm). Leucocytes treated with 0.3 × 104 J m-2 of UV responded with doubled peak values, and 4 × 104 J m-2 lead to a five-fold increase in peak chemiluminescence, as compared with non-exposed cells. Supernatants isolated from irradiated leucocytes contained increased amounts of both myeloperoxidase and lactate dehydrogenase and showed higher chemiluminescence values, when compared with supernatants from sham-irradiated cells. The results suggest that leucocyte degranulation contributes to the inflammatory properties of long wavelength UV.  相似文献

Abstract— In 1, 1, 2-trichlorotrifluoroethane solution biliverdin physically quenches singlet oxygen at a rate of 8 × l0sM-1s-1 and reacts chemically at 6 × 10 5M-1s-1 to give a red product. Abstract Relative to their L5178Y-S counterparts, L5178Y-R cells have an impaired capacity to form patches in DNA after exposure to UVC radiation. The photolysis of 5'-bromodeoxyuridine (BrdUrd) incorporated into DNA was used to estimate the number of 'repair patches'formed in response to a 254 nm UV (UVC) exposure. L5178Y-S cells, typical of rodent cell lines, formed a small number of patches in exposed DNA (1-2 patches per 1 times 108 dalton during a 6 h recovery after an exposure of 20 J/m2). In contrast, DNA extracted from L5178Y-R cells exposed to UVC and subsequently incubated with BrdUrd for 6 h showed no evidence of BrdUrd incorporation indicating no capacity to form sites of repair (fewer than 0.5 sites of BrdUrd incorporation per 1 times 108 dalton). Moreover, in L5178Y-R cells high fluences of UVC caused an extensive DNA degradation. Such degradation was not observed in L5178Y-S cells during the 24-h post-exposure period. These results are consistent with the notion that L5178Y-R cells have a reduced capacity to repair DNA damage induced by UVC radiation.  相似文献

Abstract— The mechanism of inactivating action of high-power picosecond laser UV radiation (λ= 266 nm) on the λ and φX174 bacteriophages and the pBR 322 plasmid has been studied. It has been shown that at UV radiation intensities from 1011 to 1013 W/m2, inactivation of viruses and bacterial plasmids occurs mainly on account of single-strand breaks in the DNA chain unlike the case of less powerful UV radiation where the inactivation is associated with the formation of pyrimidine dimers.  相似文献

Abstract— Kinetic data are reported for the thermal decay of colored transients formed by U.V. irradiation of aqueous solutions of 2-nitrotoluene. The transients display an acid-base equilibrium with a pK value of 3.7. The decay is catalyzed by acids and the following rate constants in liter sec-l mole-1 were evaluated for the base form of the transient reacting with an acid at 30.0°C: 3.5 × 10-3 (H2O), 2.6×103 (CH3COOH), 4.7×104 (+NH3CH2COOH) and 4.2×105(H+).  相似文献

Abstract— The levels of 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxodGuo) in purified calf thymus DNA and HeLa cells were measured following exposure to either UVC, UVB or UVA wavelengths. This DNA damage was quantitated using HPLC coupled with an electrochemical detector. The 8-oxodGuo was induced in purified DNA in a linear dose-dependent fashion by each portion of the UV spectrum at yields of 100, 0.46 and 0.16 8-oxodGuo per 105 2'-deoxyguanosine (dGuo) per kJ/m2 for UVC, UVB and UVA, respectively. However, the amount of 8-oxodGuo in HeLa cells irradiated with these UV sources decreased to approximately 2.0, 0.013 and 0.0034 8-oxodGuo per 105 dGuo per kJ/m2, respectively. In contrast, the levels of cyclobutyl pyrimidine dimers were similar in both irradiated DNA and cells. Therefore, 8-oxodGuo is induced in cells exposed to wavelengths throughout the UV spectrum although it appears that protective precesses exist within cells that reduce the UV-induced formation of this oxidative DNA damage. Cell survival was also measured and the number of dimers or 8-oxodGuo per genome per lethal event determined. These calculations are consistent with the conclusion that dimers play a major role in cell lethality for UVC- or UVB-irradiated cells but only a minor role in cells exposed to UVA wavelengths. In addition, it was found that the relative yield of 8-oxodGuo to dimers increased nearly 1000-fold in both UVA-irra-diated cells and DNA compared with cells subjected to either UVC or UVB. These results are supportive of the hypothesis that 8-oxodGuo, and possible other forms of oxidative damage, play an important role in the induction of biological effects caused by wavelengths in the UVA portion of the solar spectrum.  相似文献

Abstract The excited state properties of the chalcone isomers of malvidin 3,5-diglucoside (malvin) in acidic aqueous solution (0 < pH < 4) were investigated using steady-state and time-resolved fluorescence spectroscopy. The two chalcone isomers of malvin were first isolated by high-performance liquid chromatography and then characterized by UV/visible absorption and fluorescence spectroscopy. The results were supported by molecular orbital calculations. The rate constants for photodeprotonation ( k 1= 1 × 109 s−l) and protonation ( k −1= 1.3 × 1010 L mol−1 s−l) were determined, both from the multiexponential fluorescence decays and the fluorescence intensities measured at the emission wavelengths of the neutral and ionized chalcone forms. The results here obtained are relevant for the understanding of the photoreactivity of anthocyanins in acidic medium.  相似文献

Abstract— The effects of repeated UV exposure on the skin of the European crested newt, Triturus cristatus carnifex , have been investigated. The animals were irradiated 3 times per week with a Westing-house FS40T12 fluorescent sun lamp (wavelength spectrum 275–350 nm). Two groups of animals received the same total fluence of 1.3 × 105 J/m2 in single fluences of either 1570 J/m2 (group A) or 9430 J/m2 (group C), and one group received a total fluence of 2.6 × 105 J/m2 in single fluences of 4710 J/m2 (group B). All the animals were killed 7 months after the first UV exposure, but at different intervals after the last exposure. Striking epidermal hyperplasia was found in the newts irradiated at the lower fluence rate (group A). In the animals given the higher total fluence (group B), the most prominent skin changes were dermal fibrosis and irregular thinning and thickening of the epidermis. No significant skin changes were found in group C., in which if there had been UV lesions, they had been repaired during the 5 month interval between the last irradiation and the killing of the animals. No skin tumors developed in any experimental group.  相似文献

Abstract— The spectra and molar absorbances of the HO2 and O2- free radicals have been redetermined in aqueous formate solutions by pulse and stopped-flow radiolysis as well as by 60Co gamma-ray studies. The extinction coefficients at the corresponding maxima and 23°C are 225= 1400 ± 80 M -1 cm-1 and 225= 2350 ± 120 M -1 cm-1 respectively. Reevaluation of earlier published rate data in terms of the new extinction coefficients yielded the following rate constants for the spontaneous decay of HO2 and O2-: K Ho2+HO2= (8.60 ± 0.62) × 105 M -1 s-1; K Ho2+O2-= (1.02 ± 0.49) × 108 M -1 s-1; K Ho2+O2- < 0.35 M -1 s-1. For the equilibrium HO2→ O2-+ H+ the dissociation constant is K Ho2= (2.05 ± 0.39) × 10-5 M or p K HO2= 4.69 ± 0.08. G (O2-) has been evaluated as a function of formate concentration.  相似文献

Abstract— The Kubelka-Munk theory for diffuse reflectance has been applied to a quantitative study of photochromism in the crystalline state. For three systems investigated it was found possible to assign first order rate constants to the thermal relaxation process and estimate the pre-exponential factor A and the activation energy Ea in Arrhenius equation. For the fading of the red photocolored form, Λmax=490 mμ, of benzaldehyde phenylhydrazone A = 1.4×108 min-1 and Ea= 15.7 kcal mole-1. For the fading of the blue photocolored form, Λmax=590 mμ, of 2–(2,4-dinitrobenzyl)pyridine A= 5×1014 min-1 Ea =23.3 kcal mole-1, Cinnamaldehyde semicarbazone showing 'reversed phototropy' has a photoactivated state, Λmax=400 mμ, which in dark is transformed into a strongly absorbing yellow species, Λmax= 430 mμ with A = 14 × 1010 min-1 and Ea= 18.7 kcal mole-1.  相似文献

Abstract— Longwave ultraviolet radiation(UVA–320–400nm) is known to induce inflammation, pigmentation and tumor production in mammalian skin. The mechanisms by which such radiation induces these biologic phenomena are poorly defined. In an effort to broaden our knowledge in this area, we examined the effect of UVA on DNA biosynthesis in Hartley strain albino guinea pig skin. The animals were irradiated with selected doses of solar simulated UVA, and DNA was assayed by [3H]thymidine incorporation into epidermal DNA by autoradiography. These studies revealed that UVA inhibited DNA synthesis in a dose dependent manner between 40 and 80 J cm-2 at 3 h post-irradiation. This inhibition was followed by a stimulation of synthesis at 5 h and a second inhibition/ stimulation at 8 and 24 h, respectively. Although the mechanism of alteration is undefined, our data suggest that UVA has profound effects on DNA biosynthesis in mammalian epidermis.  相似文献

Abstract— The dose response for tumor induction in albino rat skin by single exposures of UV radiation has been characterized. The shaved dorsal skin of 202 animals was exposed to either of two sources: one emitting a broad spectrum of wavelengths from 275 to 375 nm, and the other emitting at 254 nm. Skin tumors began to appear within 10 weeks of exposure and continued to appear for 70 weeks. The highest tumor yield was 5.5 tumors per rat and occurred when the rats were exposed to 13.0 times 104 J/m2 of the 275–375 nm UV. The 275–375 nm UV was about eight times as effective as the 254 nm UV for the induction of tumors throughout the exposure range from 0.8 times 104 to 26.0 times 104J/m2. Tissue destruction and hair follicle damage was found at the highest exposure to 275–375 nm UV but at none of the exposures to 254 nm UV. Repeated weekly exposures to 275–375 nm UV proved less effective than an equivalent single exposure for inducing tumors, even though the multiple exposures caused more severe skin damage. The transmission of the UV through excised samples of rat epidermis indicated that the exposure to the basal cell layer was about 3% of the surface exposure at 254 nm and about 15% of the surface exposure between 275 and 320 nm. The dependence of tumor yield on UV exposure was linear for 254 nm UV but was more complex for the 275–375 nm UV. For the latter more tumors were produced per unit exposure at lower exposures than at higher exposures.  相似文献

Abstract The effect of chlorpromazine (CPZ) and UVA on lysosomes of cultured normal human fibroblasts has been investigated. Acid phosphatase (ACPase) activity in 12 000 g pellet of cells treated with CPZ (10 μg/m l ) and UVA (6 × 104 J/m2) was found to be decreased as compared with non-treated, CPZ or UVA treated control cells. This decrease, however, was not accompanied by a concomitant increase in ACPase activity in the 12 000 g supernatant. The addition of Triton X-100 to cells pretreated with CPZ + UVA resulted in only a moderate increase in ACPase activity of the 12 000 g supernatant. ACPase activity of the cells incubated in media containing preirradiated CPZ was also found to he decreased. These results indicate that CPZ + UVA directly inactivate lysosomal enzymes, possibly without affecting the membrane.  相似文献

Abstract— Electrochromism of oriented all- trans -β-apo-8'-carotenoic acid is studied in thin capacitors. The linear electrochromism is very strong, in contrast to that of symmetrical carotenoids. It is proportional to the first derivative of the absorption spectrum. The quadratic electrochromism can be described as a superposition of fractions proportional to the first and second derivatives of the absorption spectrum. The permanent dipole moment difference between the ground state and the excited state of the carotenoic acid molecule is Δμ= 3.6 × 10-29 C·m (±20%) (10.7 Debyes). The polarizability difference parallel to the long axis of the molecule is Δα|| = 1.17 × 10-37 C·m2·V-1 (±20%) (1050 Å3). Furthermore, the relative permittivity of the solid carotenoic ethyl ester is r= 3.5 ± 0.2.
Δμ is due to the polarizing force of the carboxylic group. This force is equivalent to a mean local electric field of F t≅3 × 106V/cm. Such a "local field" may also be exerted on a symmetrical carotenoid in the membrane of photosynthesis, e.g. by asymmetrical complex formation with a polarizing molecule. To obtain an effective permanent field of F p≅ 2 × 106V/cm across the membrane, as postulated in photosynthesis, a local field of F l≅ 5.5 × 105 V/cm would be sufficient. F p is shown to be directed from inside to outside of the thylakoid. To realize this, e.g. a positive polar (i.e. electron-attracting) complex partner of the carotenoid, located more to the inside of the thylakoid, can be postulated.  相似文献

Abstract— The colony-forming ability of Chinese hamster cells (V-79) and HeLa cells has been measured after near-ultraviolet (UV) irradiation, predominantly at 365 nm. To avoid the production of toxic photoproducts, cells were irradiated in an inorganic buffer rather than in tissue culture medium. Under these circumstances near-UV lethality was strongly oxygen-dependent. Both cell lines were approximately 104 times more sensitive to 254 nm irradiation than to 365 nm radiation when irradiated aerobically. Pretreatment with 6 times 105 Jm-2 365 nm radiation sensitised the HeLa, but not the V-79 cell line to subsequent X-irradiation. Pretreatment of cells with 17 Jm-2 254 nm radiation, a dose calculated to produce twenty times more pyrimidine dimers than the 365 nm dose, produced only slight sensitisa-tion to X-rays. It is suggested that the sensitisation to X-rays seen in the HeLa cells after 365 nm treatment is not the result of lesions induced in DNA by the near-UV radiation, but may reflect the disruption of DNA-repair systems.  相似文献
