Abstract Relative to their L5178Y-S counterparts, L5178Y-R cells have an impaired capacity to form patches in DNA after exposure to UVC radiation. The photolysis of 5'-bromodeoxyuridine (BrdUrd) incorporated into DNA was used to estimate the number of 'repair patches'formed in response to a 254 nm UV (UVC) exposure. L5178Y-S cells, typical of rodent cell lines, formed a small number of patches in exposed DNA (1-2 patches per 1 times 108 dalton during a 6 h recovery after an exposure of 20 J/m2). In contrast, DNA extracted from L5178Y-R cells exposed to UVC and subsequently incubated with BrdUrd for 6 h showed no evidence of BrdUrd incorporation indicating no capacity to form sites of repair (fewer than 0.5 sites of BrdUrd incorporation per 1 times 108 dalton). Moreover, in L5178Y-R cells high fluences of UVC caused an extensive DNA degradation. Such degradation was not observed in L5178Y-S cells during the 24-h post-exposure period. These results are consistent with the notion that L5178Y-R cells have a reduced capacity to repair DNA damage induced by UVC radiation.  相似文献

Abstract— The mechanism of inactivating action of high-power picosecond laser UV radiation (λ= 266 nm) on the λ and φX174 bacteriophages and the pBR 322 plasmid has been studied. It has been shown that at UV radiation intensities from 1011 to 1013 W/m2, inactivation of viruses and bacterial plasmids occurs mainly on account of single-strand breaks in the DNA chain unlike the case of less powerful UV radiation where the inactivation is associated with the formation of pyrimidine dimers.  相似文献

Abstract— A fluorescent colloid of chlorophyll a , of which some qualitative properties were noted by Krasnovsky and Brin(11), has been quantitatively characterized. The colloid is formed in neutral PO4 buffer containing 0 1 to 8.0% Tween 20, and is stable in darkness. The extinction coefficient is 7.8 × 104 1. mole cm-1 at the red absorption peak (668 mμ), the yield of fluorescence is ˜ 0.25, and the yield of photoautooxidative bleaching is ˜ 2 times 10-4. The colloid sensitizes the autooxidation of paratoluenediamine with a yield of ˜0.01 to ˜0.3 depending on light intensity and substrate concentration. The yield is independent of detergent and chlorophyll concentrations. In all respects—except the dependence of yield on illumination—the colloid appears to be physically and photochemically equivalent to dissolved chlorophyll, as known in dilute solutions in organic solvents. The light dependence—the yield is inversely proportional to the cube root of absorbed intensity—could be due to a bimolecular back reaction of a chlorophyll or substrate derivative.  相似文献

Abstract— The rate constant k5/ > for physical quenching of singlet oxygen O21;) by the sensitizer in dye-sensitized photooxygenations is determined in the case of chlorophylls a and b (7.3 times 108, 4.2 times 108 M-1 s-1 respectively), pheophytins a and b (7.4 times 107, 3.0 times 107 M-1 s_1 respectively), tetraphenylporphyrin (4.4 times 107 M-1 s_1), magnesium tetraphenylporphyrin (5.0 times 108 M-1 s_1), zinc tetraphenylporphyrin (1.5 times 108 M-1 s_l) and protoporphyrin IX-dimethylester (9.1 times 107 M -1 s_1) in benzene. These sensitizers show a linear correlation between log ksO , and their half-wave oxidation potentials and the value of the slope is similar to that observed for various compounds such as phenols. It is concluded that (i) the interaction between chlorophylls and related compounds with singlet oxygen may involve an exciplex as for phenols, and (ii) physical quenching may be envisaged as a spin-orbit-induced intersystem crossing within the exciplex.  相似文献

Monodelphis domestica was further characterized as a model for photobiological studies by measuring the excision repair capabilities of this mammal's cells both in vivo and in vitro. Excision repair capability of the established marsupial cell line, Pt K2 ( Potorous tridactylus ), was also determined. In animals held in the dark, we observed that ˜50% of the dimers were removed by 12 and 15 h after irradiation with 400 J m−2 and 600 J m−2, respectively, from an FS-40 sunlamp (280–400 nm). Cells from primary cultures of M. domestica excised ˜50% of the dimers by 24 h after irradiating with 50 J m−2 and 36 h after exposure to 100 J m−2 with no loss of dimers observed 24 h following a fluence of 300 J m−2. Pt K2 cells were observed to have removed -50% of the dimers at -12 h after 50 J m−2 with only -10% of the dimers removed at 24 h following 300 J m−2. The observed loss of pyrimidine dimers from epidermal DNA of UV-irradiated animals and from fibroblasts in culture, held in the dark, suggests that these marsupial cells are capable of DNA excision repair.  相似文献

Abstract— Nanosecond laser flash photolysis and pulse radiolysis have been used to generate and characterise the triplet state, and semioxidised and semireduced radicals of haematoporphyrin, and three 0 -acyl compounds derived from it (the monoacetate, the diacetate and the disuccinate).
After 347 nm irradiation in water containing 2% Triton X-100, haematoporphyrin forms the triplet state (φT= 0.92) and photoionises monophotonically (φI= 0.03). For the O -acyl derivatives, φT approaches unity and photoionisation is reduced. In acetone the triplet yield of all four compounds are close to unity. The difference and corrected spectra for the triplet species are presented and decay rates ( k 1˜104s-1) and oxygen quenching constants ( k Q˜1.5times109 M -1s-1) for the triplet state have been measured. The difference and corrected spectra for the semi-reduced species in methanol and semi-oxidised species in aqueous Triton X-100 are presented.
The photophysical characteristics in fluid solution of haematoporphyrin and its 0 -acyl derivatives are rather similar to those previously recorded for other photosensitising porphyrins.  相似文献

Abstract— From time-resolved measurements of the decay of singlet molecular oxygen phosphorescence at 1270 nm in D2O, direct estimates have been gained for the rate constants of the singlet oxygen reactions with a group of sulphur compounds in the pD range 5 to 13. In the case of most of the thiols, the results are consistent with singlet oxygen reacting exclusively with the thiolate anions. At the normal physiological pH 7, the apparent rate constants (in units of M-1 s-1) were 8.9 times 106 (cysteine), 2.5 times 106 (N-acetyl cysteine), 2.9 times 106 (glutathione), 3.0 times 105 (2-mercaptoethanol), 2.3 times 107 (ergothioneine) and 2.7 times 106 (2-mercaptopropionyl glycine). For methionine the rate constant, 1.4 times 107, was independent of pD in the range studied. These sulphur compounds, in particular N-acetyl cysteine and ergothioneine, or related compounds, might be considered as possible candidates for protection against skin photosensitivity side effects associated with the photodynamic therapy of solid tumours and as observed in the disease erythropoietic protoporphyria.  相似文献

Abstract— The triplet state of vitamin D3 in benzene has been characterised in terms of its absorption spectrum, Λmax 315 nm, its lifetime, 300 ns, its rate constant for reaction with oxygen, 4.2 times 109mol−1 s−1 and the efficiency with which it sensitizes the formation of singlet oxygen, 25%. There is a large difference in the electronic excitation energies of the spectroscopic and relaxed triplets, ˜ 237 kJ mol−1 and147–168 kJ mol−1, respectively. It has been shown that, in the endothermic situation, the vitamin D3 molecule is a 'nonvertical'acceptor of triplet energy. This is in accord with the non-planar character of its acyclic conjugated Il-system.  相似文献

Abstract— Flash photolysis at 450 nm has been used to study the quenching of the excited triplet state of lumiflavin and the transient species formed in subsequent reactions in deaerated phosphate buffer (pH 6.9).
The effect of the presence of ferricyanide on the life time of triplet lumiflavin has been studied. The results suggest an energy transfer reaction without concurrent electron transfer reactions. The rate constant for the process was 2.8 times 109 M -1 s-1. The analogous reaction with ferrocyanide could not be observed because of the efficient electron transfer reaction (δG = -20.6 kcal mol-1) leading to the formation of the semireduced lumiflavin and ferricyanide. The rate constant for this reaction was 3.3 times 109 M -1 s-1. The semireduced lumiflavin radical was found to disappear in a second order reaction with a rate constant of 1.7 times 109 M -1 s-1. It was found to react with ferricyanide with a rate constant of 0.7 times 109 M -1 s-1.
A model for the various photochemical and photophysical processes involved in the decay and quenching of the lumiflavin triplet state is suggested and discussed.  相似文献

Abstract— Transient absorption spectra produced by laser flash photolysis of an aqueous solution of 8-methoxypsoralen (8-MOP) have been studied. The biphotonic production of hydrated electrons and of the radical ions, 8-MOP + and 8-MOP- is reported. The hydrated electron was found to react with ground state 8-MOP with k ˜ 3 × 1010 M -1 s-1. In order to obtain a true triplet-triplet absorption spectrum. contributions from the radical ions were subtracted from the overall transient absorption. In addition, contributions from e-aq to the transient spectrum were removed by using N2O, low laser intensity to minimize photoionization or by measuring the transient O.D. after the electron has decayed. These three methods each produced the same triplet-triplet spectrum which differs in the red region from previously reported spectra.  相似文献

Abstract— The kinetics of photooxidation of triplets of metalloporphyrin compounds to their corresponding radical cations was investigated. Zn-tetraphenyl porphyrin (ZnTPP) and Mg-tetraphenylpor-phyrin (MgTPP) triplets were oxidized by europium salt with rate constants of 4.8 × 105M-1s-1 and 2.1 × 106M-1s-1, respectively. The high rate constant of oxidation of MgTPP triplet might be related to the ground state oxidation potential, being 0.54 V (SCE) for the Mg complex and 0.71 (SCE) for the Zn complex.
The rate constant of oxidation of ZnTPP excited singlet is in the order of diffusion control, i.e. ˜ 1010M -1 s-1. Excitation of ferric, cupric, cobaltic, and vanadyl tetraphenylporphyrin did not result in a long-lived triplet state that would allow oxidation studies using flash photolysis.  相似文献

Abstract— Radiolytic formation and peroxidation of fatty acid radicals have been investigated by pulse radiolysis techniques in oleate, linoleate, linolenate and arachidonate systems. A strong absorption band at 280 nm associated with conjugated radicals, Rconj, formed in polyunsaturated fatty acid moieties has been used as a probe for kinetic processes occurring at doubly allylic sites in the hydrocarbon chain. Formation of Rconj by O- has been found to be more efficient than the less selective OH radical. Peroxidation of Rconj is shown to be somewhat slower, ( k R+ O2˜ 3 × 108 M -1 s-1), than O2 reactions with radicals in oleate ( k R+ O2= 1 × 109 M -1 s-1). Peroxy radicals generated in these reactions disappear slowly by essentially second order processes (2 k RO1˜ 107 M -1 s-1). The superoxide radical, O-2, shows little if any reactivity towards 0.01 M linolenate or arachidonate over periods of 20 s.  相似文献

Abstract— The colony-forming ability of Chinese hamster cells (V-79) and HeLa cells has been measured after near-ultraviolet (UV) irradiation, predominantly at 365 nm. To avoid the production of toxic photoproducts, cells were irradiated in an inorganic buffer rather than in tissue culture medium. Under these circumstances near-UV lethality was strongly oxygen-dependent. Both cell lines were approximately 104 times more sensitive to 254 nm irradiation than to 365 nm radiation when irradiated aerobically. Pretreatment with 6 times 105 Jm-2 365 nm radiation sensitised the HeLa, but not the V-79 cell line to subsequent X-irradiation. Pretreatment of cells with 17 Jm-2 254 nm radiation, a dose calculated to produce twenty times more pyrimidine dimers than the 365 nm dose, produced only slight sensitisa-tion to X-rays. It is suggested that the sensitisation to X-rays seen in the HeLa cells after 365 nm treatment is not the result of lesions induced in DNA by the near-UV radiation, but may reflect the disruption of DNA-repair systems.  相似文献

Abstract— We measured excision repair of ultraviolet radiation (UVR)-induced pyrimidine dimers in DNA of the corneal epithelium of the marsupial, Monodelphis domestica , using damage-specific nucleases from Micrococcus luteus in conjunction with agarose gel electrophoresis. We observed that 100 J -2 of UVR from aFS–40 sunlamp(280–400 nm) induced an average of 2.2 ± 0.2 times 10-2 endonuclease-sensitive sites per kilobase (ESS/kb) (pyrimidine dimers) and that ∼ 50% of the dimers were repaired within 12 h after exposure. We also determined that an exposure of 400 J m-2 was needed to induce comparable numbers of pyrimidine dimers (2.5 times 10-2) in the DNA of skin of M. domestica in vivo . In addition, we found that 50% of the dimers were also removed from the epidermal cells of M. domestica within 12 h after exposure. A dose of 100 J m-2 was necessary to induce similar levels of pyrimidine dimers (2.0 ± 0.2 times 10-2) in the DNA of the cultured marsupial cell line Pt K2 ( Potorous tridactylus ).  相似文献

Pretreatment of human cells with near UV radiation (UVA) in fluences exceeding 5 × 104 Jm−2 caused a decrease in the amount of the unscheduled DNA synthesis induced by far UV radiation (UVC). The DNA repair synthesis, as measured by the incorporation of [3H] -thymidine, is reduced by nearly a factor of 2 for a UVA radiation exposure of 1.5 × 105 Jm−2. Since solar UVA fluence rate is rather independent of latitude, this figure corresponds to a UVA exposure time of 50-60 min from noon sunlight in the summer time.  相似文献

Abstract— A Xenon-chloride excimer laser emitting energy at 308 nm was used to induce single-strand breaks (SSBs, frank breaks plus alkali-labile lesions as assayed by alkaline sucrose sedimentation techniques) in purified DNA from Bacillus subtilis . A fluence response study and a peak pulse intensity study were performed. At a pulse energy of 0.1 mJ/pulse, the radiation induced SSBs in a linear fashion (91 SSB/108 Da per MJ/m2) to a maximum exprimental fluence of 1.28 MJ/m2. The pulse intensity study showed that there were no significant changes in DNA breakage (105 SSB/108 Da) between 2.93 times 109 and 5.86 times 1011 W/m2 (0.11 and 22.0 mJ/pulse) at a constant total fluence of 1.1 MJ/m2 (27000 mJ dose). This study has verified and extended previous work by quantifying the yield of SSBs induced in DNA by this laser radiation.  相似文献

Abstract—Reaction rate constants for the reaction of singlet oxygen with a series of 24 sulfides in chloroform have been measured by inhibition of the self-sensitized photooxidation of rubrene. The reaction rate constant is sensitive to steric effects, decreasing as the carbons α- to sulfur become more highly substituted. Addition of a methyl group to each of the carbons α- to sulfur decreases the rate constant by about a factor of 10. From a series of p - and m -substituted thioanisoles, a ρ of -1.67 ± 0.09 was found. A much better correlation was found with σ than with σ+ indicating there is no resonance interaction with the reaction center. Typical rate constants are: di- n -butyl sulfide, 2.3 × 107 M -1 s-1; CBZ-L-methionine methyl ester, 1.4 × 107; di-s-butyl sulfide, 1.8 × 106; di- t -butyl sulfide, 1.3 × 105; and thioanisole, 2.3 × 106.  相似文献

Abstract— Using the technique of flash photolysis-electron spin resonance, we have shown, by means of a kinetic analysis, that phenazine methosulfate (PMS) interacts with reaction-center preparations from the blue-green mutant R26 of Rhodopseudomonas spheroides. At intermediate concentrations of PMS, biphasic decay kinetics of the P870+ ESR signal are observed demonstrating that the PMS radical interacts with reaction centers by a specific binding mechanism. With PMS bound to reaction centers, the P870+ ESR signal decays in ˜ 1 ms; whereas, in unbound reaction centers the decay is ˜ 120 ms. A model is proposed involving the interaction of PMS on the donor side of P870.  相似文献

Abstract— While studying the photoreduction of some dyes (D) by reducing agents (R), it was observed that the quantum yield of the photoreduction increases considerably upon addition of a third substance (C), whereas it is very small when the dye is photoreduced by C alone (catalytic effect), (see Table 1).
The system thionine (D), allylthiourea (R), and azulene (C) was investigated in detail using both flash photolysis and continuous illumination. On photolysis, thionine is converted into its photo-reduced form, leucothionine. Azulene reacts with the basic form of the thionine triplet 3 TH + to produce the semithionine radical. In the system thionine and azulene, most of these radicals revert back to thionine. When ATU (˜ 102- M ) is added to thionine and azulene (3 × 10-4 M ), the semithionine radicals are reduced to leucothionine; the quantum yield of this reduction is considerably higher than in the system thionine and allylthiourea. Flash experiments demonstrate that allylthiourea does not react with the semithionine radicals.
At very high ATU concentrations (≥ 10-1 M ), however, the primary reaction is between thionine triplet and allylthiourea; under these conditions the quantum yield is not influenced by azulene (3 × 10-4 M ).  相似文献

Abstract. Using single picosecond laser pulses at 610 nm, the fluorescence yield (φ) of spinach chloroplasts as a function of intensity ( I ) (1012-1016 photons/pulse/cm2) was studied in the range of 21–300 K. The quantum yield decreases with increasing intensity and the φ vs I curves are identical at the emission maxima of 685 and 735 nm. This result is interpreted in terms of singlet exciton-exciton annihilation on the level of the light-harvesting pigments which occurs before energy is transferred to the Photosystem I pigments which emit at 735 nm.
The yield φ is decreased by factors of 12 and 43 at 300 and 21 K, respectively. The shapes of the φ vs I curves are not well accounted for in terms of a model which is based on a Poisson distribution of photon hits in separate photosynthetic units, but can be satisfactorily described using a one-parameter fit and an exciton-exciton annihilation model. The bimolecular annihilation rate constant is found to be γ= (5–15) times 10-9cm3s-1 and to exhibit only a minor temperature dependence. Lower bound values of the singlet exciton diffusion coefficient (≥ 10-3cm2s-1), diffusion length (≥ 2 times 10-6cm) and Förster energy transfer rates (≥ 3 ≥ 1010s-1) are estimated from γ using the appropriate theoretical relationships.  相似文献
