In 0.01 M 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid (Hepes), pH 7.4 and room temperature, the binding of neodymium to N,N'-ethylenebis[2-(o-hydroxyphenolic)glycine] (EHPG), or N,N'-di(2-hydroxybenzyl)ethylenediamine-N,N' diacetic acid (HBED) had been studied from 210 to 330 nm by means of difference UV spectra. Two peaks at 240 and 292 nm appear in difference UV spectra after neodymium binding to EHPG or HBED. The 1:1 stable complex can be confirmed from spectral titration curves. The molar extinction coefficient of Nd-EHPG and Nd-HBED complexes are Deltaepsilon(Nd-EHPG)=(12.93+/-0.21) x 10(3)cm(-1)M(-1), Deltaepsilon(Nd-HBED)=(14.45+/-0.51) x 10(5)cm(-1)M(-1) at 240 nm, respectively. Using EDTA as a competitor, the conditional equilibrium constants of the complexes are logK(Nd-EHPG)=11.89+/-0.09 and logK(Nd-HBED)=12.19+/-0.15, respectively. At the same conditions, fluorescence measurements show that neodymium binding to EHPG leads to a quenching of the fluorescence of EHPG at near 310 nm. However, there is no obvious fluorescence change of HBED at 318 nm with the binding of neodymium to HBED.  相似文献

The interactions of yttrium with N,N'-ethylenebis[2-(o-hydroxyphenolic)glycine] (EHPG) and N,N'-di(2-hydroxybenzyl)ethylenediamine-N,N'-diacetic acid (HBED) are investigated by using UV difference and fluorescence spectra methods in 0.1M N-2-hydroxyethylpiperazine-N-2-ethanesulfonic acid (Hepes) at pH 7.4. Yttrium binding produces two UV difference peaks near 240 and 294 nm, respectively, that both are the characteristic of phenolic groups binding to yttrium. The molar extinction coefficient of Y-EHPG and Y-HBED are (15.7 +/- 0.40) x 10(3), (15.8 +/- 0.80) x 10(3)cm(-1)M(-1) at 240 nm, respectively. Using EDTA as a competitor the obtained conditional equilibrium constants of the complexes are logK(Y-EHPG) = 15.07 +/- 0.32 and logK(Y-HBED) = 15.18 +/- 0.26, respectively. However, the effects of yttrium binding on the fluorescence intensity of EHPG and HBED are quite different, the former showing a decrease but the latter an increase.  相似文献

The binding of Gd3+ ion to apoovotransferrin (apoOTf) was monitored by means of UV difference spectra in 0.01M Hepes, pH 7.4 at 25 degrees C. Used 2-p-toluidinylnaphthalene-6-sulfonate (TNS) as fluorescence probe the conformational changes of protein were studied while gadolinium ions bound to apoOTf. The results show that Gd3+ binding produces peaks at 244 and 294 nm that is the characteristic of binding at the apoOTf specific metal-binding sites. At 244 nm the molar absorptivity of Gd-apoOTf complex is (1.99+/-0.17)x10(4)cm(-1)M(-1). The apparent binding constants for the complexes of Gd3+ with apoovotransferrin are logK(1)=7.61+/-0.14 and logK(2)=4.96+/-0.26. A very large conformational change of apoovotransferrin appears when Gd3+ is bound to the N-terminal binding site. When Gd3+ is bound to C-terminal binding site there is less conformational change.  相似文献

In the presence of 1.0mM Ca(2+), the interaction between Euplotes octocarinatus centrin (EoCen) and melittin (ME) was studied by means of fluorescence spectra. In 0.1M N-2-hydroxyethylpiperazine-N-2-ethanesulfonic acid (Hepes) and 150mM NaCl at pH 7.4, fluorescence peak of ME was observed at about 353nm indicating that micro-environment of Tryptophan (Trp) residue in ME was hydrophilic. With the addition of 3.2x10(-4)M calcium saturated EoCen (holoEoCen), the peak of ME was blue-shifted to 339nm, which may be resulted from micro-environmental changes of the peptide. At the same time, fluorescence emission of ME was increased significantly suggesting that new complex of ME-holoEoCen was formed under the experimental conditions. Based on the fluorescence titration curves, the 1:1 stoichiometric ratio of holoEoCen to ME was confirmed. In addition, the conditional binding constant of holoEoCen with ME was calculated to be logK(ME-holoEoCen)=6.59+/-0.14.  相似文献

Centrin is a member of the EF-hand superfamily that plays critical role in the centrosome duplication and separation. In the present paper, we characterized properties of metal ions binding to Euplotes octocarinatus centrin (EoCen) by fluorescence spectra and circular dichroism (CD) spectra. Changes of fluorescence spectra and alpha-helix contents of EoCen proved that Tb(3+) and Ca(2+) induced great conformational changes of EoCen resulting in exposing hydrophobic surfaces. At pH 7.4, Ca(2+) (and Tb(3+)) bond with EoCen at the ratio of 4:1. Equilibrium experiment indicated that Ca(2+) and Tb(3+) exhibited different binding capabilities for C- and N-terminal domains of protein. C-terminal domain bond with Ca(2+) or Tb(3+) approximately 100-fold more strongly than N-terminal. Aromatic residue-sensitized Tb(3+) energy transfer suggested that site IV bond to Tb(3+) or Ca(2+) more strongly than site III. Based on fluorescence titration curves, we reckoned the conditional binding constants of EoCen site IV quantitatively to be K(IV)=(1.23+/-0.51)x10(8)M(-1) and K(IV)=(6.82+/-0.33)x10(5)M(-1) with Tb(3+) and Ca(2+), respectively. 铝(Ⅲ)与脱铁伴清蛋白结合的紫外差光谱研究   总被引:2,自引:0,他引:2  
在 pH7.4、 0.1mol· L~(-1)N-2-羟乙基哌嗪- N′-2-乙磺酸( Hepes)及室温条件下,使用紫外吸收差光谱进行了铝(Ⅲ)对脱铁伴清蛋白的滴定。结果表明铝(Ⅲ)与脱铁伴清蛋白结合后其紫外差光谱在 238nm和 291nm处出现吸收峰。在 238nm处铝(Ⅲ)-脱铁伴清蛋白配合物的摩尔吸光系数是 (1.52± 0.04)× 10~4cm~(-1)· mol~(-1)· L。铝(Ⅲ)可占据脱铁伴清蛋白的两个金属离子结合部位,条件稳定常数是 lgK_N=11.21± 0.12,lgKC=9.53± 0.24。 N-端单铁伴清蛋白的紫外差光谱滴定表明,铝 ?优先占据脱铁伴清蛋白的 N端结合部位。  相似文献

The determination of 0.5-4.75 mug of fluoride ion by its catalytic action upon the slow reaction between Methylthymol Blue and zirconium(IV) in aqueous solution is described. Calibration curves obtained after 60 min under optimal conditions are smooth, and yield an effective molar absorptivity of 3.23 x 10(4) 1.mole(-1)mm(-1) at 586 nm. There is considerably less cationic interference than in the alizarin complexan-cerium(III) or lanthanum procedure, but more serious anionic interference is encountered when phosphate, arsenate and, to a lesser extent, sulphate ions are present in the sample solution.  相似文献

The UV absorption spectra of CF(3)CHO, C(2)F(5)CHO, C(3)F(7)CHO, C(4)F(9)CHO, CF(3)CH(2)CHO, and C(6)F(13)CH(2)CHO were recorded over the range 225-400 nm at 249-297 K. C(x)F(2)(x)(+1)CHO and C(x)F(2)(x)(+1)CH(2)CHO have broad absorption features centered at 300-310 and 290-300 nm, respectively. The strength of the absorption increases with the size of the C(x)F(2)(x)(+1) group. There was no discernible (<5%) effect of temperature on the UV spectra. Quantum yields for photolysis at 254 and 308 nm were measured. Quantum yields at 254 nm were 0.79 +/- 0.09 (CF(3)CHO), 0.81 +/- 0.09 (C(2)F(5)CHO), 0.63 +/- 0.09 (C(3)F(7)CHO), 0.60 +/- 0.09 (C(4)F(9)CHO), 0.74 +/- 0.08 (CF(3)CH(2)CHO), and 0.55 +/- 0.09 (C(6)F(13)CH(2)CHO). Quantum yields at 308 nm were 0.17 +/- 0.03 (CF(3)CHO), 0.08 +/- 0.02 (C(4)F(9)CHO), and 0.04 +/- 0.01 (CF(3)CH(2)CHO). The quantum yields decrease with increasing size of the C(x)F(2)(x)(+1) group and with increasing wavelength of the photolysis light. The photolysis quantum yield at 308 nm for CF(3)CHO measured here is a factor of at least 8 greater than that reported previously. Photolysis is probably the dominant atmospheric fate of C(x)F(2)(x)(+1)CHO (x = 1-4) and is an important fate of C(x)F(2)(x)(+1)CH(2)CHO (x = 1 and 6). These results have important ramifications concerning the yield of perfluorocarboxylic acids in the atmospheric oxidation of fluorotelomer alcohols.  相似文献

The UV absorbance and photochemical decomposition kinetics of hydrogen peroxide in borate/boric acid buffers were investigated as a function of pH, total peroxide concentration, and total boron concentration. At higher pH borate/boric acid inhibits the photodecomposition of hydrogen peroxide (molar absorptivity and quantum yield of H(2)O(2) and HO(2) (-), (19.0+/-0.3) M(-1) cm(-1) and 1, and (237+/-7) M(-1) cm(-1) and 0.8+/-0.1, respectively). The results are consistent with the equilibrium formation of the anions monoperoxoborate, K(BOOH)=[H(+)][HOOB(OH)(3) (-)]/([B(OH)(3)][H(2)O(2)]), 2.0 x 10(-8), R. Pizer, C. Tihal, Inorg. Chem. 1987, 26, 3639-3642, and monoperoxodiborate, K(BOOB)=[BOOB(2-)]/([B(OH)(4) (-)][HOOB(OH)(3) (-)]), 1.0+/-0.3 or 4.3+/-0.9, depending upon the conditions, with molar absorptivity, (19+/-1) M(-1) cm(-1) and (86+/-15) M(-1) cm(-1), respectively, and respective quantum yields, 1.1+/-0.1 and 0.04+/-0.04. The low quantum yield of monoperoxodiborate is discussed in terms of the slower diffusion apart of incipient (.)OB(OH)(3) (-) radicals than may be possible for (.)OH radicals, or a possible oxygen-bridged cyclic structure of the monoperoxodiborate.  相似文献

The absorption spectra of five pesticides, namely 2,4-dichloro-phenoxy acetic acid (2,4-D), cymoxanil, fenpropidin, isoproturon and pyrimethanil, have been measured in aqueous solution using a set-up consisting of two parallel absorption cells coupled to a CCD detector. The absolute values of their molar absorptivity coefficients epsilon were determined in the wavelength-range 240-344 nm with a deuterium-lamp at room temperature (298+/-2 K). Using the Beer-Lambert law, values of epsilon were also determined at 253.7 nm with a Hg-Lamp: epsilon = 145+/-14 for 2,4-D, epsilon = 7940+/-920 for cymoxanil, epsilon = 196+/-14 for fenpropidin, epsilon = 7330+/-880 for isoproturon, epsilon = 13200+/-1400 for pyrimethanil (in units of M(-1) cm(-1)). The quoted errors correspond to 2 sigma obtained from the least square fit analysis and the estimated systematic error of 5% due to the uncertainties in aqueous concentrations. For all the studied compounds, the absorbances measured were lower than 2.3 and did not exhibit any deviation from the Beer-Lambert's law. Our experimental data are discussed and compared to UV spectra of similar molecules when such data were available in the literature. Based on their UV spectra and the calculated fractions of these pesticides in the aqueous phase, their direct photolysis under sunlight environment could occur, except may be for fenpropidin, either in water surfaces or in aqueous droplets contained in the atmospheric clouds.  相似文献

Cationic surfactants, such as cetylpyridinium bromide (CPB), sensitize the colour reaction of Nb(V) with 8-hydroxyquinoline-5-sulphonic acid (H(2)L). The formation of a ternary complex of stoichiometry 1:3:3 (Nb-L-CPB) is responsible for the observed enhancement in absorptivity and the quenching in the fluorescence of the Nb-L chelate, when a surfactant is present. The ternary complex exhibits maximum absorption at 383 nm ( = 1.46 +/- 0.01 x 10(4) l.mole(-1).cm(-1)) at pH 5.7, and Beer's law is obeyed up to 6-mug ml Nb concentration. Conditional formation constants of the niobium chelate in the presence and absence of CPB have been determined. On the basis of a detailed spectrophotometric and fluorimetric study the nature of the chromophoric reagent-surfactant interaction and the peculiar features of the sensitization by CPB are discussed.  相似文献

Tranexamic acid is reacted with aqueous alcoholic chloranil, buffered at pH 9, to give a complex with maximum absorption at 346 nm and with an apparent molar absorptivity of 15.7 x 10(3) l.mole(-1).cm(-1). A(max) is linearly related to concentration over the range 2-10 mug ml . When applied to tablets labelled as containing 500 mg each, the mean found was 496 +/- 4 mg. The interaction of chlorobenzylidine with herring sperm DNA has been investigated by fluorescence, absorption, DNA melting experiment and differential scanning calorimetry (DSC). When bound to DNA, chlorobenzylidine shows hypochromism and red shift in absorption spectra, fluorescence quenching and polarization increasing in fluorescence spectra and increasing in DNA melting temperature. These spectral characteristics strongly support intercalation of chlorobenzylidine into herring sperm DNA. Scatchard plots constructed from fluorescence titration data give a binding constant of 3.2 x 10(4) M(-1) and a binding site size of six base pairs per bound drug molecule. The intercalative interaction is exothermic with a van't Hoff enthalpy of -30.6 kJ mol(-1). This result is obtained from DSC experiment. In addition, DeltaG degrees =-28.5 kJ mol(-1), and DeltaS degrees =-7.1 J mol(-1) K(-1). These results show that the binding of chlorobenzylidine to herring sperm DNA is exothermic.  相似文献

In this work, fluorescence quenching technique, Fourier transform infrared (FT-IR) spectroscopy, circular dichroism (CD) spectroscopy and dynamic light scattering (DLS) technique were used to gain the binding information of caffeic acid and human serum albumin (HSA) in AOT/isooctane/water microemulsions. The interaction of HSA with caffeic acid at 296, 303, and 310 K in omega(0) 20 microemulsions was characterized by one binding site with the affinity constant K at (3.23+/-0.01) x 10(4), (3.06+/-0.03) x 10(4) and (2.82+/-0.05) x 10(4)M(-1), respectively. The affinities in microemulsions are much higher than that in buffer solution. The CD spectra and FT-IR spectra with qualitative and quantitative results proved that the protein secondary structure changed in the microemulsions in the absence and presence of caffeic acid compared with the free form of HSA in buffer. The binding process was exothermic and spontaneous, as indicated by the thermodynamic analyses. These data indicated that hydrophobic interaction played a major role in the binding of caffeic acid to HSA in microemulsions and electrostatic interaction can not be excluded. The displacement experiments confirmed that caffeic acid could bind to the site I of HSA, which was in agreement with the result of the molecular modeling study. Furthermore, the DLS data suggested that HSA may locate at the interface of the microemulsion and caffeic acid could interact with them.  相似文献

A simple and sensitive method for spectrophotometric determination of lanthanum has been developed. At pH 9.6, in presence of 50% ethanol, lanthanum reacts with 1-(-2-pyridylazo)-2-naphthol (PAN) to form a red complex which has two absorption maxima, at 545 and 510 nm. The molar absorptivity at 545 nm is 0.55 × 104 liters · mol?1 cm?1. On the other hand, lanthanum reacts with PAN in pure ethanol to form a red complex at 530 nm, with high molar absorptivity (8 × 104 liters · mol?1 cm?1).  相似文献

The interaction between bromocresol purple (BCP) and bovine serum albumin (BSA)/porcine serum albumin (PSA) was investigated both in the absence and presence of bilirubin (BR) using absorption/absorption difference spectroscopy. A significant red shift in the absorption maxima of BCP accompanied by a decrease in absorbance was indicative of BCP binding to albumin. The titration of BSA and PSA with BCP using absorption difference spectroscopy and analysis of results by Benesi-Hildebrand equation yielded the values of association constant, K as 9.9+/-0.9x10(4)Lmol(-1) and 4.1+/-0.3x10(4)Lmol(-1) for BSA and PSA, respectively. The differential extinction coefficient (Deltaepsilon) of 34,484M(-1)cm(-1) at 615nm and 41,870M(-1)cm(-1) at 619nm were estimated for BSA and PSA, respectively. Decrease in (DeltaAbs.)(615nm) of BCP-BSA complex with the increase in ionic strength suggested the role of hydrophobic interactions in the binding phenomenon. A significant blue shift in the absorption maxima and change in (DeltaAbs)(lambdamax) values of BR-albumin complexes upon addition of increasing concentrations of BCP revealed the BR displacing action of BCP on albumin molecule.  相似文献

The properties of 4-amino-4'-methyldiphenylamine as a redox indicator have been studied. The compound acts as a reversible, one-colour indicator, the oxidized form being red-violet, with molar absorptivity 6 x 10(3) l.mole(-1).cm(-1) at lambda(max) = 510 nm. The formal potential is described by the equation E = 0.735-0.059 pH. The two dissociation constants are pK(1) = -0.08 +/- 0.09 and pK(2) = 5.09 +/- 0.02. The interaction of gallium(Ⅲ) with the ligands containing phenolic group(s), such as salicylic acid, 8-hydroxyquinoline, N,N'-bis(2-hydroxybenzyl)ethylenediamine-N,N'diacetic acid (HBED), N,N'-ethylenebis[2-(o- hydroxyphenyl)glycine (EHPG), and ovotransferrin, was studied, respectively, by means of fluorescence in 0.01 mol/L Hepes at pH 7.4 and room temperature. Fluorescence intensity showed an increase when gallium(Ⅲ) was bound to 8-hydroxyquinoline and HBED. In contrast, it was decreased with the interaction of gallium(Ⅲ) with salicylic acid and EHPG. At pH 7.4, there was N…H-O type intramolecular hydrogen bond in the former, and the latter existed O…H-O type intramolecular hydrogen bond. Fluorescence titration of apoovotransferrin with gallium(Ⅲ) displayed that the fluorescence intensity was decreased at the N-terminal binding site, while enhanced at the C-terminal binding site. It can account for the O…H-O type intramolecular hydrogen bonds for the phenolic groups of Tyr92 and Tyr191 residues at the N-terminal binding site. And there are N…H-O type intramolecular hydrogen bonds for Tyr431 and Tyr524 residues at the C-terminal binding site. In addition, under the same conditions, the conditional binding constant of gallium(Ⅲ) with EHPG or HBED determined by fluorescence method is lg KGa-EHPG=19.18 or lg KGa-HBED= 19.08.  相似文献

A new crown hydroxamic acid, 5,14-N,N'-hydroxyphenyl-4,15-dioxo-1,5,14,18-tetraaza hexacosane (NHDTAHA) for the extraction and spectrophotometric determination of lanthanum(III) is described. Lanthanum(III) forms a yellow coloured complex with NHDTAHA, which is extracted with chloroform, having molar absorptivity 7.7 x 10(3) 1 mol(-1) per cm at 372 nm. The system obeys Beer's law in the range 1.2-20 ppm of lanthanum. The extract is directly aspirated for ICP-AES measurements, the limits for estimation are 5-140 ppb of lanthanum. Lanthanum has been determined in monazite sand and standard samples.  相似文献
