Macroporous cellulose beads (MCB) used as anion exchangers were successfully prepared from cellulose solution in ionic liquid by double emulsification followed by cross-linking and modification with diethylaminoethyl. The pore structure and properties of the MCB were investigated and the results were compared with homogeneous cellulose beads (HCB). The MCB in size of about 71 μm is characterized by two sets of pores, i.e., diffusion pores (10–20 nm) and macropores (800–2000 nm), determined by mercury porosimeter. In addition, the bed permeability and effective porosity for BSA of MCB-packed column are 58% and 25% higher than those of HCB-packed column, respectively. The adsorption properties of MCB were evaluated, and compared with HCB and commercial absorbent (Sepharose 6 Fast Flow, CSFF). It is found that the pore diffusivity of BSA in MCB is over 7.9 times higher than HCB, and 6.7 times higher than CSFF, respectively. While the equilibrium adsorption capacity (qm) of BSA on MCB is obviously lower than that on HCB and CSFF, the dynamic binding capacity (DBC) on MCB at 10% breakthrough reaches 47.7 mg/mL, higher than HCB (40.3 mg/mL) and CSFF (46.2 mg/mL) at flow rate of 360 cm/h. In addition, the MCB-packed column showed better column efficiency over the HCB packed one. Therefore, we demonstrated that the MCB possessed more advantages than other ones, like HCB and CSFF, and was expected as an ideal material for fast chromatography.  相似文献

Agarose gels were fabricated by water-in-oil emulsification with the addition of CaCO3 granules at 8–16 wt%. Thus agarose beads of different superporosities were produced after dissolving the solid porogen. The superporous agarose (SA) and homogeneous agarose gels were double cross-linked and modified with diethylaminoethyl chloride to produce anion exchangers. We have proposed to use a superporous replica (porous titania microspheres) to examine the superporous structure and pore size distribution of the soft gel. The replica was prepared with the agarose gel entrapping CaCO3 granules by a sol–gel-templating method. It was found that the superpores created by CaCO3 granules were uniformly distributed and ranged from 0.95 μm to 1.33 μm. The physical properties of the gels were significantly affected by the porogen content. Importantly, by increasing the solid porogen to 12 wt%, the bed permeability and effective porosity increased about 48% and 33%, respectively. Further increase in the porogen to 16 wt% led to a decrease of the mechanical strength. With increasing superpores in the beads, the dynamic adsorption capacity of the packed columns increased obviously at 305–916 cm/h. Besides, the column efficiency changed less with increasing flow velocity up to 1200 cm/h. It was concluded that the use of 12 wt% CaCO3 granules in agarose solution was beneficial for the fabrication of the SA gel with good mechanical stability and promising performance for protein chromatography.  相似文献

Silica particles have been used as supports for the preparation of three different propazine-imprinted polymer formats. First format refers to grafting of thin films of molecularly imprinted polymers (MIPs) using an immobilised iniferter-type initiator (inif-MIP). The other two new formats were obtained by complete filling of the silica pores with the appropriate polymerisation mixture leading to a silica-MIP composite material (c-MIP) followed by the dissolution of the silica matrix resulting in spherical MIP beads (dis-MIP). These techniques offer a mean of fine-tuning the particle morphology of the resulting MIP particles leading to enhanced capacity in chromatographic applications. Porous silica (specific surface area S = 380 m2 g−1, particle size ps = 10 μm, pore volume Vp = 1.083 ml g−1 and pore diameter dp = 10.5 nm), methacrylic acid and ethylenglycol dimethacrylate were used for the preparation of the materials. All the MIP formats imprinted with propazine have been characterised by elemental analysis, FT-IR spectroscopy, nitrogen adsorption and scanning electron microscopy. Further, the materials were assessed as stationary phases in HPLC. Capacity factors, imprinting factors and theoretical plate numbers were calculated for propazine and other related triazines in order to compare the chromatographic properties of the three different stationary phases. For the inif-MIPs the column efficiency depended strongly on the amount of grafted polymer. Thus, only the polymers grafted as thin films of ca. 1.3 nm average thickness show imprinting effects and the highest column efficiency giving plate numbers (N) of 1600 m−1 for the imprinted propazine. The performance of the c-MIP stationary phase decreases as result of the complete pore filling after polymerisation and increases again after the removal of the silica matrix due to a better mass transfer in the porous mirror-image resulting polymer. From this study can be concluded that the inif-MIP shows the best efficiency for use as stationary phase in HPLC for the separation of triazinic herbicides.  相似文献

A pyrimethanil-imprinted polymer (P1) was prepared by iniferter-mediated photografting a mixture of methacrylic acid and ethylene dimethacrylate onto homemade near-monodispersed chloromethylated polydivinylbenzene beads. The chromatographic behaviour of a column packed with these imprinted beads was compared with another column packed with irregular particles obtained by grinding a bulk pyrimethanil-imprinted polymer (P2). The comparison was made using the kinetic model of non-linear chromatography, studying the elution of the template and of two related substances, cyprodinil and mepanipyrim. Extension of the region of linearity, capacity factors for the template and the related substances, column selectivity, binding site heterogeneity, apparent affinity constant (K) and lumped kinetic association (ka) and dissociation rate constant (kd) were studied during a large interval of solute concentration, ranging between 1 and 2000 μg/ml. From the experimental results obtained, in the linearity region of solute concentration column selectivity and binding site heterogeneity remained essentially the same for the two columns, while column capacity (at 20 μg/ml, P1 = 23.1, P2 = 11.5), K (at 20 μg/ml, P1 = 8.3 × 106 M−1, P2 = 2.5 × 106 M−1) and ka (at 20 μg/ml, P1 = 3.5 μM−1 s−1, P2 = 0.47 μM−1 s−1) significantly increased and kd (at 20 μg/ml, P1 = 0.42 s−1, P2 = 0.67 s−1) decreased for the column packed with the imprinted beads. These results are consistent with an influence of the polymerisation method on the morphology of the resulting polymer and not on the molecular recognition properties due to the molecular imprinting process.  相似文献

A constant development of dye‐affinity chromatography to replace more traditional techniques is verified, with the aim of increasing specificity in the purification of biomolecules. The establishment of a new dye‐affinity chromatographic support imposes their complete characterization, namely with relation to the binding capacity for proteins, in order to evaluate its applicability on global purification processes. Following previous studies, the adsorption of lysozyme onto a thiacarbocyanine dye immobilized on beaded cellulose was investigated. The effect of different parameters, such as temperature, ionic strength, pH, protein concentration and flow rate, on the dynamic binding capacity of the support to retain lysozyme was also studied. Increasing the temperature and the lysozyme concentration had a positive effect on the dynamic binding capacity (DBC), whereas increasing the ionic strength and the flow rate resulted in the opposite. It was also discovered that the pH used had an important impact on the lysozyme binding onto the immobilized dye. The maximum DBC value obtained for lysozyme was 8.6 mg/mL, which was achieved at 30°C and pH 9 with a protein concentration of 0.5 mg/mL and a flow rate of 0.05 mL/min. The dissociation constant (Kd) obtained was 2.61 ± 0.36 × 10–5 m , proving the affinity interaction between the thiacarbocyanine dye ligand and the lysozyme. An IgG-specific camelid antibody matrix (BAC, Naarden, The Netherlands), developed from an immune phage display library, was characterized regarding engineering properties including mass transfer characteristics. Uptake kinetics and equilibrium binding capacity were determined by a finite bath method. Adsorption kinetic parameters were also determined using a real time biosensor. Slightly different properties to conventional Staphylococcal protein A affinity media were shown; especially a 2–2.5 times lower maximal binding capacity with a value of 26 mg/ml polyclonal IgG was obtained. Mass transfer could be described by using a film and pore diffusion model (De = 5 × 10−8 cm2/s). Determined engineering parameters were used to predict breakthrough behaviour in column mode considering film and pore resistances. The dynamic binding capacity at 10% breakthrough did not change when residence time was at least 6 min.  相似文献

[Mg1−x Alx(OH)2][(NO3)x, nH2O] Layered Double Hydroxide (LDH) sorbents with variable Mg/Al molar (R=(1−x)/x) ratios were investigated for adsorption of azo dye, orange II (OII) at various pH and temperature conditions. Mg2AlNO3 displays the highest adsorption capacity with 3.611 mmol of OII per gram of Mg2AlNO3 at 40 °C. Adsorption isotherms have been fitted using the Langmuir model and free energy of adsorption (ΔG°), enthalpy (ΔH°) and entropy (ΔS°) were calculated. The experimental values for ΔG° in temperature range between 10 and 40 °C were found to be negative indicating that a spontaneous process occurred. Positive calculated enthalpy values, characteristic of an endothermic process were found. Characterization of solids (PXRD, FTIR, UV-vis, TGA/DTA, adsorption isotherm BET analysis, SEM and Zetametry) before and after adsorption showed that adsorption proceeds in two steps. First, adsorption occurs at the LDH surface, followed by intercalation via anion exchange.  相似文献

The suitability of the use of macroporous adsorbent Amberlite XAD7HP in expanded bed adsorption processes for the isolation of flavonoids from crude extracts of Ginkgo biloba L. has been assessed. The expansion and hydrodynamic properties of expanded beds were investigated and analyzed. The bed expansion as a function of operational fluid velocity was measured and correlated with the Richardson–Zaki equation. Theoretical predictions of the correlation parameters (the terminal settling velocity ut and exponent n) were improved by modifying equations in the literature. Residence time distributions (RTDs) were studied using acetone as a tracer. Three measures of liquid phase dispersion (the height equivalent of theoretical plate, Bodenstein number and axial distribution coefficient) were investigated and compared to values previously obtained with commercial EBA adsorbents developed for protein purification. A suitable bed expansion ratio was found to be 1.25 times the settled bed height, which occurred at a corresponding flow velocity of 183 cm/h. For an initial settled bed height of 42 cm, the mean residence time of liquid in the expanded bed was around 28 min. Under these flow conditions, the axial mixing coefficient Dax was 7.54 × 10−6 m2/s and the Bodenstein number was 28; the number of theoretical plates (N) was 19 and the height equivalent of a theoretical plate (HETP) was 2.77 cm. Rutin trihydrate was used as a model flavonoid for the characterization of the adsorption properties of Amberlite XAD7HP. Adsorption was observed to reach equilibrium within 3 h with 70% of the adsorption capacity being achieved within 30 min. The estimated maximum equilibrium adsorption capacity for rutin was estimated to be 43.0 mg/(g resin) when the results were fitted to Langmuir isotherms. The adsorption performance was not seriously impaired by the physical presence of G. biloba leaf powders. Assessment of the kinetics of the adsorption of rutin revealed that the rate constant for adsorption was only reduced by 15% in the presence of leaf powders at a concentration of 50 mg/mL. The results demonstrated that Amberlite XAD7HP should be suitable for expanded bed adsorption of flavonoids from crude extracts of G. biloba L.  相似文献

Functionalized monolithic columns were prepared via ring-opening metathesis polymerization (ROMP) within silanized fused silica capillaries with an internal diameter of 200 μm by in situ grafting. This procedure is conducted in two steps, the first of which is the formation of the basic monolithic structure by polymerization of norborn-2-ene (NBE) and 1,4,4a,5,8,8a-hexahydro-1,4,5,8-exo,endo-dimethanonaphthalene (DMN-H6) in a porogenic system (toluene and 2-propanol) using RuCl2(PCy3)2(CHPh) as ROMP initiator. In the second step the still active initiator sites located on the surface of the structure-forming microglobules were used as receptor groups for the attachment (“grafting”) of functional groups onto the monolithic backbone by flushing the monolith with 7-oxanorborn-2-ene-5,6-carboxylic anhydride (ONDCA). Functionalization conditions were first defined that did not damage the backbone of low polymer content (20%) monoliths allowing high-throughput chromatographic separations. Variation of the functionalization conditions was then shown to provide a means of controlling the degree of functionalization and resulting ion-exchange capacity. The maximum level of in situ ONDCA grafting was obtained by a 3 h polymerization in toluene at 40 °C. The weak cation-exchange monoliths obtained provided good separation of a standard peptide mixture comprising four synthetic peptides designed specifically for the evaluation of cation-exchange columns. An equivalent separation was also achieved using the lowest capacity column studied, indicative of a high degree of robustness of the functionalization procedure. As well as demonstrably bearing ionic functional groups enabling analyte separation in the cation-exchange mode, the columns exhibited additional hydrophobic characteristics which influenced the separation process. The functionalized monoliths thus represent useful tools for mixed-mode separations.  相似文献

A new Dynamic Extractions Maxi-counter-current chromatography (CCC) centrifuge with a column volume of 18-l has been installed in the Advanced Bioprocessing Centre at Brunel. This instrument has four times the capacity of the 4.6-l Maxi-CCC centrifuge which has been operating robustly for 3 years. Tests using the model sample system benzyl alcohol and p-cresol with a heptane:ethyl acetate:methanol:water (HEMWat) phase system (1.4:0.1:0.5:1.0) show that resolution is almost double with this new high capacity device. Commissioning tests with a mixture of caffeine, KD = 0.21; ferulic acid, KD = 0.82; umbelliferone, KD = 1.2 and vanillin, KD = 1.49 using a HEMWat phase system of 1:1.5:1:1.5 on the 9-l column show that resolutions equivalent to analytical instruments will be possible using the full 18-l capacity. They also show that predictable scale-up from simple test tube tests is feasible with knowledge of the stationary phase retention for the planned process scale run.  相似文献

High-quality free-standing poly(dibenzo-18-crown-6) (PDBC) films with a conductivity of 4.1 × 10−2 S cm−1 and good thermal stability were synthesized electrochemically on stainless steel electrode by direct anodic oxidation of dibenzo-18-crown-6 (DBC) in pure boron trifluoride diethyl etherate (BFEE). In this medium, the oxidation potential onset of DBC was measured to be only 0.98 V vs. SCE, which was much lower than that in acetonitrile + 0.1 mol L−1 Bu4NBF4 (1.45 V vs. SCE). PDBC films obtained from this medium showed good redox activity and stability in BFEE. The structural characterization of PDBC was performed using UV-vis, FTIR spectroscopy. The results of quantum chemistry calculations of DBC monomer and FTIR spectroscopy of PDBC films indicated that the polymerization mainly occurred at C(4) and C(5) positions). Fluorescent spectral studies indicated that PDBC was a blue light emitter. To the best of our knowledge, this is the first report on the electrodeposition of free-standing PDBC films.  相似文献

In this study, a comparative investigation was performed of HPLC Ascentis® (2.7 μm particles) columns based on fused-core particle technology and Acquity® (1.7 μm particles) columns requiring UPLC instruments, in comparison with Chromolith™ RP-18e columns. The study was carried out on mother and vegetal tinctures of Passiflora incarnata L. on one single or two coupled columns. The fundamental attributions of the chromatographic profiles are evaluated using a chemometric procedure, based on the AutoCovariance Function (ACVF). Different chromatographic systems are compared in terms of their separation parameters, i.e., number of total chemical components (mtot), separation efficiency (σ), peak capacity (nc), overlap degree of peaks and peak purity. The obtained results show the improvements achieved by HPLC columns with narrow size particles in terms of total analysis time and chromatographic efficiency: comparable performance are achieved by Ascentis® (2.7 μm particle) column and Acquity® (1.7 μm particle) column requiring UPLC instruments. The ACVF plot is proposed as a simplified tool describing the chromatographic fingerprint to be used for evaluating and comparing chemical composition of plant extracts by using the parameters D% – relative abundance of the deterministic component – and cEACF – similarity index computed on ACVF.  相似文献

Binary diffusion coefficients D12 and retention factors k for dibenzo-24-crown-8 and 15-crown-5 ethers at 308.18 and 313.20 K, and vitamin K1 at 313.20 K were measured in supercritical carbon dioxide by the chromatographic impulse response technique, and the effects of molecular shapes on the D12 values were studied. At 313.2 K and 11.0 MPa the D12 value of 15-crown-5 was higher than that predicted from the D12 correlation with molecular weight MW over the range from 32 of methanol to 1138 of trinervonin reported in our previous studies, while the D12 value of dibenzo-24-crown-8, disk shape molecule with MW = 448.5, was almost in agreement with that of vitamin K1, long chain molecule with MW = 450.7, and with those predicted from the correlation.  相似文献

For BSA and β-lactoglobulin adsorption to hydrophobic interaction chromatography (HIC) stationary phases leads to conformational changes. In order to study the enthalpy (ΔHads), entropy (ΔSads), free energy (ΔGads) and heat capacity (Δcp,ads) changes associated with adsorption we evaluated chromatographic data by the non-linear van’t Hoff model. Additionally, we performed isothermal titration calorimetry (ITC) experiments. van’t Hoff analysis revealed that a temperature raise from 278 to 308 K increasingly favoured adsorption seen by a decrease of ΔGads from −12.9 to −20.5 kJ/mol for BSA and from −6.6 to −13.2 kJ/mol for β-lactoglobulin. Δcp,ads values were positive at 1.2 m (NH4)2SO4 and negative at 0.7 m (NH4)2SO4. Positive Δcp,ads values imply hydration of apolar groups and protein unfolding. These results further corroborate conformational changes upon adsorption and their dependence on mobile phase (NH4)2SO4 concentration. ITC measurements showed that ΔHads is dependent on surface coverage already at very low loadings. Discrepancies between ΔHads determined by van’t Hoff analysis and ITC were observed. We explain this with protein conformational changes upon adsorption which are not accounted for by van’t Hoff analysis.  相似文献

Highly ordered 2D-hexagonal mesoporous titanium silicate Ti-SBA-15 materials (space group p6mm) have been synthesized hydrothermally in acidic medium employing amphiphilic tri-block copolymer, Pluronic F127 as structure directing agent. Samples are characterized by powder X-ray diffraction, transmission electron microscopy, scanning electron microscopy, FT IR spectroscopy, UV-visible diffuse reflectance measurements, N2 adsorption/desorption and TG-DTA analysis. XRD and TEM results suggested the presence of highly ordered mesophase with hexagonal pore arrangements. BET surface area for Ti-SBA-15 (924 m2 g−1) is considerably higher than the pure silica SBA-15 (611 m2 g−1) prepared following the same synthetic route. UV-visible and FT-IR studies suggested the incorporation of mostly tetrahedral titanium (IV) species, along with some six-coordinated sites in the silicate network. This material shows very good H2 adsorption capacity at higher pressure and excellent catalytic activity in the photocatalytic degradation of ecologically abundant dye methylene blue.  相似文献

In this work, biomimetic technique was proposed for the first time to prepare chromatographic packings (2HAp-ZM) for protein separation. By mimicking the mineralization procedures in vivo, this type of matrix with hydroxyapatite (HAp) coating and zirconia-magnesia (ZM) composite core was fabricated. Systematic characterizations, including scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), X-ray diffraction (XRD) and specific surface area analysis, were carried out to investigate the properties of the material. Results showed that the surface of 2HAp-ZM was composed of bead-like, amorphous or nanocrystalline HAp. The specific surface area, total pore volume, and average pore diameter of the resultant material were 25 m2/g, 0.09 cm3/g, and 14 nm, respectively. Furthermore, 2HAp-ZM exhibited good mechanical stability through repeated testing and its application as stationary phases for protein separation was then studied. Five model proteins (bovine serum albumin, trypsin, lysozyme, ribonuclease A, and cytochrome c) were successfully separated on 2HAp-ZM. Finally, the mass recovery of protein and the dynamic loading capacity were studied; the results were calculated to be no less than 93% and 80 μg/mL of blank column volume, respectively.  相似文献

The bisphenol A (BPA) imprinted polymer microspheres were prepared by simple Pickering emulsion polymerization. Compared to traditional bulk polymerization, both high yields of polymer and good control of particle sizes were achieved. The characterization results of scanning electron microscopy and nitrogen adsorption–desorption measurements showed that the obtained molecularly imprinted polymer microsphere (MIPMS) particles possessed regular spherical shape, narrow diameter distribution (30–60 μm), a specific surface area (SBET) of 281.26 m2 g−1 and a total pore volume (Vt) of 0.459 cm3 g−1. Good specific adsorption capacity for BPA was obtained in the sorption experiment and good class selectivity for BPA and its seven structural analogs (bisphenol F, bisphenol B, bisphenol E, bisphenol AF, bisphenol S, bisphenol AP and bisphenol Z) was demonstrated by the chromatographic evaluation experiment. The MIPMS as solid-phase extraction (SPE) packing material was then evaluated for extraction and clean-up of these bisphenols (BPs) from human urine samples. An accurate and sensitive analytical method based on the MIPMS-SPE coupled with HPLC-DAD has been successfully established for simultaneous determination of eight BPs from human urine samples with detection limits of 1.2–2.2 ng mL−1. The recoveries of BPs for urine samples at two spiking levels (100 and 500 ng mL−1 for each BP) were in the range of 81.3–106.7% with RSD values below 8.3%.  相似文献

Chitosan (CS) and poly(acrylic acid) (PAA) were crosslinked by an ionic gelation method to form super absorbent polymers (SAPs). CS and PAA form amide bonds between the amino and carboxyl groups. The CS-PAA copolymers were synthetically engineered by varying the feed ratios of the prepolymer units. The copolymer materials possess tunable sorption and mucoadhesive properties with a backbone structure resembling proteinaceous materials. The sorption properties of the copolymers toward methylene blue (MB) in aqueous solution were studied using UV-Vis spectrophotometry at ambient pH and 295 K. The copolymers showed markedly varied interactions with MB, from physisorption- to chemisorption-like behavior, in accordance with their composition, surface area, and pore structure characteristics. The sorption isotherms were evaluated with the Sips model to provide estimates of the sorption properties. The sorbent surface area (271 and 943 m2/g) and the sorption capacity (Qm = 1.03 and 3.59 mmol/g) were estimated for the CS-PAA copolymer/MB systems in aqueous solution.  相似文献

Mesoporous carbons containing cobalt nanoparticles are synthesized by tri-or quad-constituent self assembly of Pluronic F127, phenol-formaldehyde oligomer (resol), cobalt acetylacetonate (acac), and optionally tetraethyl orthosilicate (TEOS, optional). Upon pyrolysis in N2 atmosphere, the resol provides sufficient carbon yield to maintain the ordered structure, while decomposition of the Co(acac) yields cobalt nanoparticles. To provide increased surface area, the dispersed silicate from condensation of TEOS can be etched after carbonization to yield micropores, Without silica templated micropores, the surface area decreases as the cobalt content increases, but there is a concurrent increase in the volume-average pore diameter (BHJ) and a dramatic increase in the adsorption capacity of methylene green with the equilibrium adsorption capacity from 2 to 90 mg/g with increasing Co content. Moreover, the surface area and pore size of mesoporous composites can be dramatically increased by addition of TEOS and subsequent etching. These composites exhibit extremely high adsorption capacity up to 1151 mg/g, which also increases with increases in the Co content. Additionally, the inclusion of cobalt nanoparticles provides magnetic separation from aqueous suspension. The in situ synthesis of the Co nanoparticles yields to a carbon shell that can partially protect the Co from leaching in acidic media; after 96 h in 2 M HCl, the powders remain magnetic.  相似文献

A novel chelating resin containing S, N and O atoms (PSME-EDA) was synthesized by using poly(2-hydroxyethylmercaptomethylstyrene) (PSME) and diethanolamine (EDA) as materials. Its structure was characterized by elemental analysis, Fourier transform-infrared spectra (FTIR), scanning electron microscopy (SEM) and X-ray diffraction (XRD). The adsorption of the resin for Hg2+ was investigated. The saturated adsorption capacity of PSME-EDA for Hg2+ could reach to about 1.1 mmol/g at 25 °C when the initial Hg2+ concentration was 0.02 mol/l. Some factors affecting the adsorption such as temperature, reaction time and ion concentration were also studied. The results showed that adsorption was controlled by liquid film diffusion. The increasing of temperature was beneficial to adsorption. The Langmuir model was better than the Freundlich model to describe the isothermal process. The values of ΔG, ΔH, and ΔS calculated at 25 °C were −7.99 kJ mol−1, 22.5 kJ mol−1 and 34.4 J mol−1 K−1, respectively. The adsorption mechanism of PSME-EDA resin for Hg(II) was confirmed by X-ray photoelectron spectroscopy (XPS).  相似文献
