In this study, C18-silica monoliths were synthesized as a porous layer in open tubular capillary columns, to be cut later into microcartridges for the analysis of neuropeptides by on-line solid-phase extraction capillary electrophoresis with UV and MS detection (SPE-CE-UV and SPE-CE-MS). First, several types of C18-silica monolithic (MtC18) microcartridges were used to analyse standard solutions of five neuropeptides (i.e. dynorphin A (1–7), substance P (7–11), endomorphin 1, methionine enkephalin and [Ala]-methionine enkephalin). The MtC18 sorbents were especially selective against endomorphin 1 and substance P (7–11)). The best results in terms of sensitivity and inter-microcartridge reproducibility were achieved with the microcartridges obtained from a 10-cm open tubular capillary column with a thin monolithic coating with large through-pores (1–5 μm). Run-to-run repeatability, microcartridge durability, linearity ranges and LODs were studied by MtC18-SPE-CE-MS. As expected due to their greater selectivity, the best LOD enhancement was obtained for End1 and SP (7–11) (50 times with regard to CE-MS). Finally, the suitability of the methodology for analysing biological fluids was tested with plasma samples spiked with End1 and SP (7–11). Results obtained were promising because both neuropeptides could be detected at 0.05 μg mL−1, which was almost the same concentration level as for the standard solutions (0.01 μg mL−1).  相似文献

A conducting copolymer of pyrrole and phenol was electrochemically synthesized on steel frits as a sorbent. The applicability of the frit was assessed for the solid-phase extraction of trace amounts of polycyclic aromatic hydrocarbons (PAHs) in aqueous samples followed by HPLC–UV. The coating produced was very adherent and the scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDX) and FTIR spectrum for the coated frit were studied. The effects of various parameters on the efficiency of the solid-phase extraction process, such as the sample loading rate, elution solvent type, salt effect, volume and flow rate of sample and elution solvent were investigated. Under the optimal conditions, the calibration curves were obtained in the range of 0.1–500 ng mL−1 (r2 > 0.98) and the LODs (S/N = 3) were obtained in the range of 0.01–0.08 ng mL−1. Relative standard deviations (RSDs) for intra- and inter-day precision were 2.7–10.2% and 3.6–11.4%, respectively. The recoveries (8 and 40 ng mL−1) ranged from 79% to 115%. The simplicity of experimental procedure, short sample analysis, high extraction efficiency, and the use of low-cost adsorbent show the potential of this method for routine analysis of PAHs in real samples.  相似文献

A simple, sensitive, and specific method with gas chromatography-mass spectrometry was developed for simultaneous extraction and derivatization of amphetamines (APs) and 3,4-methylenedioxyamphetamines (MDAs) in human urine by using a monolithic silica spin column. All the procedures, such as sample loading, washing, and elution were performed by centrifugation. APs and MDAs in urine were adsorbed on the monolithic silica and derivatized with propyl chloroformate in the column. Methamphetamine-d5 was used as an internal standard. The linear ranges were 0.01-5.0 μg mL−1 for methamphetamine (MA) and 3,4-methylenedioxymethamphetamine (MDMA) and 0.02-5.0 μg mL−1 for amphetamine (AP) and 3,4-methylenedioxyamphetamine (MDA) (coefficient of correlation ≧0.995). The recovery of APs and MDAs in urine was 84-94%, and the relative standard deviation of the intra- and interday reproducibility for urine samples containing 0.1, 1.0, and 4.0 μg mL−1 of APs and MDAs ranged from 1.4% to 13.6%. The lowest detection limit (signal-to-noise ratio ≧ 3) in urine was 5 ng mL−1 for MA and MDMA and 10 ng mL−1 for AP and MDA. The proposed method can be used to perform simultaneous extraction and derivatization on spin columns that have been loaded with a small quantity of solvent by using centrifugation.  相似文献

Rohypnol (flunitrazepam) has been successfully determined in coffee by high performance liquid chromatography dual electrode detection (LC-DED) in the dual reductive mode. Initial studies were performed to optimise the chromatographic conditions and these were found to be 50% acetonitrile, 50% 50 mM pH 2.0 phosphate buffer at a flow rate of 0.75 mL min−1, employing a Hypersil C18, 5 μm, 250 mm × 4.6 mm column. Cyclic voltammetric studies were made to ascertain the redox behaviour of Rohypnol at a glassy carbon electrode over the pH range 2–12. Hydrodynamic voltammetry was used to optimise the applied potential at the generator and detector cells; these were identified to be −2.4 V and +0.8 V for the redox mode and −2.4 V and −0.1 V for the dual reductive mode respectively. A linear range of 0.5–100 μg mL−1, with a detection limit of 20 ng mL−1 was obtained for the dual reductive mode. Further studies were then performed to identify the optimum conditions required for the LC-DED determination of Rohypnol in beverage samples. A convenient and rapid method for the determination of Rohypnol in beverage samples was developed using a simple sample pre-treatment procedure. A recovery of 95.5% was achieved for a sample of white coffee fortified at 9.6 μg mL−1 Rohypnol.  相似文献

A novel bimodal porous N-(2-aminoethyl)-3-aminopropyltrimethoxysilane (AAPTS)-silica monolithic capillary was prepared by sol–gel technology, and used as capillary microextraction (CME) column for aluminum fractionation by electrothermal vaporization inductively coupled plasma mass spectrometry (ETV)–ICP–MS with the use of polytetrafluoroethylene (PTFE) slurry as fluorinating agent. The extraction behaviors of different Al species were studied and it was found that in the pH range of 4–7, labile monomeric Al (free Al3+, Al–OH and Al–F) could be retained quantitatively on the monolithic capillary, while non-labile monomeric Al (Al–Cit and Al–EDTA) passed through the capillary directly. The labile monomeric Al retained on monolithic capillary was eluted with 10 μL 1 mol L− 1 HCl and the elution was introduced into the ETV for fluorination assisted ETV–ICP–MS determination. The total monomeric Al fraction was also determined by AAPTS-silica monolithic CME–fluorination-assisted electrothermal vaporization (FETV)–ICP–MS after the sample solution was adjusted to pH 8.8. Non-labile monomeric Al was obtained by subtracting labile monomeric Al from the total monomeric Al. Under the optimized conditions, the relative standard deviation (R.S.D) was 6.2% (C = 1 μg L− 1, = 7; sample volume, 5 mL), and the limit of detection was 1.6 ng L− 1 for Al with an enrichment factor of 436 fold and a sampling frequency of 9 h− 1. The prepared AAPTS-silica monolithic capillary showed an excellent pH tolerance and solvent stability and could be used for more than 250 times without decreasing adsorption efficiency. The developed method was applied to the fraction of Al in rainwater and fruit juice, and the results demonstrated that the established system had advantages over the existing 8-hydroxyquinoline (8-HQ) chelating system for Al fractionation such as wider pH range, higher tolerance of interference and better regeneration.  相似文献

Hemimicelles of tetradecanoate chemisorbed onto magnetic nanoparticles (MNPs) are here proposed as a sorbent for the single-step extraction and cleanup of bisphenol A (BPA) in soft drinks. The purpose of this work was to develop a simple, rapid and low-cost sample treatment suitable to assess the human exposure to BPA from this type of high consumption food. The nanoparticles were easily coated by mixing commercially available magnetite of 20–30 nm mean particle diameter with tetradecanoate at 85 °C for 30 min. The extraction/cleanup procedure involved stirring the samples (3 mL) with 200 mg of tetradecanoate-coated MNPs for 20 min, isolating the sorbent with a Nd–Fe–B magnet and eluting BPA with methanol. The extraction efficiency was not influenced by salt concentrations up to 1 M and pH values over the range 4–9. No cleanup of the extracts was needed, and the method proved matrix-independent. The extracts were analyzed by liquid chromatography, electrospray ionization tandem mass spectrometry. Quantitation was performed by internal standard calibration using BPA-13C12. The limit of quantitation obtained for the method, 0.03 ng mL−1, was below the usual range of concentrations reported for BPA in soft drinks (0.1–3.4 ng mL−1). The proposed method was successfully applied to the determination of BPA in different samples acquired from various supermarkets in southern Spain; the concentrations found ranged from 0.066 to 1.08 ng mL−1. Recoveries from samples spiked with 0.33 ng mL−1 of BPA ranged from 91% to 105% with relative standard deviations from 3% to 8%.  相似文献

Reaction of formaldehyde with amino acids followed by oxidation with hydrogen peroxide to produce a fluorophore Norharman product is well known and was used for the spectrofluorimetric determination of l-tryptophan (Trp). This study aimed to use graphene oxide (GO) to enhance the selectivity and sensitivity of Trp in presence of other amino acids and possible interfering compounds. Different parameters such as pH, temperature, incubation time, and concentrations of formaldehyde, H2O2 and GO were studied to optimize the condition of determination. Experimental data showed that the maximum fluorescence intensity was achieved in pH 7.0–9.0 phosphate buffer mixed with 7–10% (v/v) formaldehyde and 1–2% (v/v) H2O2 as oxidizing agent at 60 ?C for 1 h. On the basis of calibration curve of various concentrations of Trp in the presence of 20 μg mL−1 GO, the lower limit of detection (LOD) of Trp was determined as 0.092 nmol mL−1 and the lower limit of quantification (LOQ) was 0.3 nmol mL−1. The selectivity of Trp in presence of other amino acids and possible interfering compounds were studied with and without GO. The data obtained after inner filter effect corrections revealed that the selectivity of Trp in presence of amino acids and other possible interfering agents was improved in the range of 76–96%, compared with that in absence of GO. The enhancement of selectivity in the presence of GO indicates that the Trp and other amino acid and possible interfering compounds were adsorbed by GO, and the selective uptaking of Trp-by the reaction with formaldehyde followed by oxidation with H2O2 at 60 ?C with high selectivity and sensitivity was achieved successfully.  相似文献

An on-line solid-phase extraction (SPE)-HPLC method was developed for simultaneous screening of nicardipine and amlodipine in human plasma. A short monolithic poly(glycidyl methacrylate-co-ethylene glycol dimethacrylate) [p(GMA-EDMA)]-based weak cation-exchange (WCX) column was prepared and employed as the selective extraction sorbent, which exhibited good permeability and biocompatibility. During the on-line SPE protocol, high-abundance proteins (human serum albumin, immunoglobulin G, immunoglobulin A and transferrin) and most matrixes in plasma were fast removed while nicardipine and amlodipine were effectively trapped on this monolithic column. Furthermore, the monolithic WCX sorbent could be continuously reused more than 300 times without obvious changes in analytes extraction and proteins cleanup. The proposed method was linear over a range of 0.5-50.0 ng mL−1 for both analytes with a linear regression coefficient greater than 0.998, and the limit of detection (LOD) for each analyte was 0.2 ng mL−1. Validation assays also demonstrated acceptable precision and adequate recovery for simultaneous quantitative screening of nicardipine and amlodipine in human plasma. Real plasma samples from hypertensive patients receiving a dosing of 5 mg antagonists were examined by using the proposed method. Results indicated that the on-line SPE-HPLC method could be applied for simultaneously monitoring of nicardipine and amlodipine in clinical plasma samples.  相似文献

Mutagenic and carcinogenic heterocyclic amines (HCAs) are formed during heating of various proteinaceous foods, but human exposure to HCAs has not yet been elucidated in detail. To assess long-term exposure to HCAs, we developed a simple and sensitive method for measuring HCAs in hair by automated on-line in-tube solid-phase microextraction (SPME) coupled with liquid chromatography–tandem mass spectrometry (LC–MS/MS). Using a Zorbax Eclipse XDB-C8 column, 16 HCAs were analyzed within 15 min. The optimum in-tube SPME conditions were 20 draw/eject cycles of 40 μL sample at a flow rate of 200 μL min−1 using a Supel-Q PLOT capillary column as an extraction device. The extracted HCAs were easily desorbed from the column by passage of the mobile phase, with no carryover observed. This in-tube SPME LC–MS/MS method showed good linearity for HCAs in the range of 10–2000 pg mL−1, with correlation coefficients above 0.9989 (n = 18), using stable isotope-labeled HCA internal standards. The detection limits (S/N = 3) of 14 HCAs except for MeAαC and Glu-P-1 were 0.10–0.79 pg mL−1. This method was successfully utilized to analyze 14 HCAs in hair samples without any interference peaks, with quantitative limits (S/N = 10) of about 0.17–1.32 pg mg−1 hair. Using this method, we evaluated the exposure to HCAs in cigarette smoke and the suitability of using hair HCAs as exposure biomarkers.  相似文献

The effect of a feed ratio of methyltrimethoxysilane (MTMS) to tetramethoxysilane (TMOS) was studied to improve the performance of a hybrid monolithic silica capillary column with 100-μm i.d. in HPLC in a range MTMS/TMOS (v/v) = 10/90–25/75. The domain size was also varied by adjusting the amount of PEG to control permeability (K = 2.8 × 10−14–6.9 × 10−14 m2). Evaluation of the performance for those capillary columns following octadecylsilylation proved an increase in retention factor (k) and a decrease in steric selectivity α(triphenylene/ortho-terphenyl) with the increase in MTMS content in the feed. The effect of the feed ratio was also observed in porosity and hydrophobic property of the C18 stationary phase from the results of size exclusion chromatography (SEC) and reversed phase characterization. The monolithic silica capillary columns prepared under new preparation conditions were able to produce a plate height of 4.6–6.0 μm for hexylbenzene in a mobile phase acetonitrile/water = 80/20 at a linear velocity of 2 mm/s. Consequently, it was possible to prepare hybrid monolithic silica capillary columns with higher performance than those reported previously while maintaining the retention factors in a similar range by reducing the MTMS/TMOS ratio and increasing the total silane concentration in feed.  相似文献

The application of sulfur microparticles as efficient adsorbents for the solid-phase extraction (SPE) and determination of trace amounts of 10 polycyclic aromatic hydrocarbons (PAHs) was investigated in sea water and wastewater samples using high performance liquid chromatography coupled with an ultraviolet detector (HPLC–UV). Parameters influencing the preconcentration of PAHs such as the amount of sulfur, solution flow rate and volume, elution solvent, type and concentration of organic modifier, and salt effect were examined. The results showed that at a flow rate of 10 mL min−1 for the sample solutions (100 mL), the PAHs could be adsorbed on the sulfur microparticles and then eluted by 2.0 mL of acetonitrile. For HPLC–UV analysis of extracted PAHs, the calibration curves were linear in the range of 0.05–80.0 μg L−1; the coefficients of determinations (r2) were between 0.9934 and 0.9995. The relative standard deviations (RSDs) for eight replicates at two concentration levels (0.5 and 4.0 μg L−1) of PAHs were lower than 7.3%, under optimized conditions. The limits of detection (LODs, <!-- no-mfc -->S/N<!-- /no-mfc --> = 3) of the proposed method for the studied PAHs were 0.007–0.048 μg L−1. The recoveries of spiked PAHs (0.5 and 4 μg L−1) in the wastewater and sea water samples ranged from 78% to 108%. The simplicity of experimental procedure, high extraction efficiency, short sample analysis, and using of low cost sorbent demonstrate the potential of this approach for routine trace PAH analysis in water and wastewater samples.  相似文献

Camptothecin (CPT) and its derivative have been revealed to possess special anti-cancer activity, extraction methods are necessary for trace determination of CPTs in complex samples. In this work, we prepared a high efficient boronic acid-based polymer monolithic layer for microextraction of CPTs. A disposable membrane filter-based extraction device was developed, and boronic acid groups were co-polymerized into a polyporous polymer skeleton and served as the monolithic sorbent. The prepared poly(4-VB-MA-TRIM) showed good stability and great extraction efficiency toward four CPTs. After optimization of extraction conditions, poly(4-VB-MA-TRIM)-based solid-phase microextraction was coupled HPLC for determination of CPTs in biological samples. The method exhibited low limits of detection of 0.05–0.2 ng mL−1, which is significantly more sensitive than reported HPLC methods. The method also showed wide linear range (0.1–100 and 0.5–200 ng mL−1), good linearity (R2 ≥ 0.9981) and good reproducibility (RSD ≤3.76%). The method has been applied in plasma samples, with good selectivity and good recoveries ranging from 85.1 to 104.7%.  相似文献

The bisphenol A (BPA) imprinted polymer microspheres were prepared by simple Pickering emulsion polymerization. Compared to traditional bulk polymerization, both high yields of polymer and good control of particle sizes were achieved. The characterization results of scanning electron microscopy and nitrogen adsorption–desorption measurements showed that the obtained molecularly imprinted polymer microsphere (MIPMS) particles possessed regular spherical shape, narrow diameter distribution (30–60 μm), a specific surface area (SBET) of 281.26 m2 g−1 and a total pore volume (Vt) of 0.459 cm3 g−1. Good specific adsorption capacity for BPA was obtained in the sorption experiment and good class selectivity for BPA and its seven structural analogs (bisphenol F, bisphenol B, bisphenol E, bisphenol AF, bisphenol S, bisphenol AP and bisphenol Z) was demonstrated by the chromatographic evaluation experiment. The MIPMS as solid-phase extraction (SPE) packing material was then evaluated for extraction and clean-up of these bisphenols (BPs) from human urine samples. An accurate and sensitive analytical method based on the MIPMS-SPE coupled with HPLC-DAD has been successfully established for simultaneous determination of eight BPs from human urine samples with detection limits of 1.2–2.2 ng mL−1. The recoveries of BPs for urine samples at two spiking levels (100 and 500 ng mL−1 for each BP) were in the range of 81.3–106.7% with RSD values below 8.3%.  相似文献

An on-line method, based on coupling dynamic ultrasonic extraction (DUE), continuously sampling the suspension of sample and solvent, high performance liquid chromatographic separation with diode array detection, has been developed for the determination of the flavonoids, including baicalin, baicalein and wogonin, from the root of Scutellaria baicalensis Georgi. Variables influencing the DUE were evaluated by orthogonal test. The extraction yields of baicalin, baicalein and wogonin in the roots of S. baicalensis Georgi obtained from five different cultivated areas are 73.8–131.5 μg mg−1 (RSD ≤ 6.24%), 6.8–15.9 μg mg−1 (RSD ≤ 5.36%) and 4.4–14.3 μg mg−1 (RSD ≤ 5.30%), respectively. The limits of detection for baicalin, baicalein and wogonin are 0.30, 0.37 and 0.41 μg mL−1, respectively. Linearity is from 0.55 to 109 μg mL−1 for baicalin, from 0.51 to 105 μg mL−1 for baicalein and from 0.53 to 102 μg mL−1 for wogonin. Compared with off-line continuous flow-DUE, the proposed method would be more convenient for the determination of the analytes and the rapid optimization of the extraction process. The extraction yields of flavonoids obtained by the proposed method are comparable with those obtained by dynamic microwave assisted extraction, static ultrasonic extraction and reflux extraction. The result indicated that the proposed method is suitable to determine the active components in Chinese herbal medicine.  相似文献

An immunoaffinity (IA) sorbent with antibody fragments was prepared for the analysis of opioid peptides by on-line immunoaffinity solid-phase extraction capillary electrophoresis–mass spectrometry (IA-SPE-CE–MS). The antibody fragmentation was evaluated by MALDI-TOF-MS. Fab′ fragments obtained from a polyclonal IgG antibody against Endomorphins 1 and 2 (End1 and End2) were covalently attached to succinimidyl silica particles to prepare the IA sorbent. An IA-SPE-CE–MS methodology was established analyzing standard solutions of End1 and End2 and acceptable repeatability, linearity ranges and LODs (0.5 and 5 ng mL−1, respectively) were obtained. The LOD of End1 was slightly better than that previously obtained using an IA sorbent with intact antibodies (1 ng mL−1). In human plasma samples, End1 and End2 could be detected at 1 and 50 ng mL−1, respectively, which meant an improvement of 100 and 2-fold with regard to the LODs using an IA sorbent with intact antibodies (100 ng mL−1).  相似文献

A method based on amino-modified silica-coated magnetic nanoparticles (ASMNPs) and polymerase chain reaction (PCR) was developed to rapidly and sensitively detect foodborne pathogens in raw milk. After optimizing parameters such as pH, temperature, and time, a trace amount of genomic DNA of pathogens could be extracted directly from complex matrices such as raw milk using ASMNPs. The magnetically separated complexes of genomic DNA and ASMNPs were directly subjected to single PCR (S-PCR) or multiplex PCR (M-PCR) to detect single or multiple pathogens from raw milk samples. Salmonella Enteritidis (Gram-negative) and Listeria monocytogenes (Gram-positive) were used as model organisms to artificially contaminate raw milk samples. After magnetic separation and S-PCR, the detection sensitivities were 8 CFU mL−1 and 13 CFU mL−1 respectively for these two types of pathogens. Furthermore, this method was successfully used to detect multiple pathogens (S. Enteritidis and L. monocytogenes) from artificially contaminated raw milk using M-PCR at sensitivities of 15 CFU mL−1 and 25 CFU mL−1, respectively. This method has great potential to rapidly and sensitively detect pathogens in raw milk or other complex food matrices.  相似文献

A novel solid-phase microextraction (SPME) fiber coated with multiwalled carbon nanotubes (MWCNTs)/Nafion was developed and applied for the extraction of polar aromatic compounds (PACs) in natural water samples. The characteristics and the application of this fiber were investigated. Electron microscope photographs indicated that the MWCNTs/Nafion coating with average thickness of 12.5 μm was homogeneous and porous. The MWCNTs/Nafion coated fiber exhibited higher extraction efficiency towards polar aromatic compounds compared to an 85 μm commercial PA fiber. SPME experimental conditions, such as fiber coating, extraction time, stirring rate, desorption temperature and desorption time, were optimized in order to improve the extraction efficiency. The calibration curves were linear from 0.01 to 10 μg mL−1 for five PACs studied except p-nitroaniline (from 0.005 to 10 μg mL−1) and m-cresol (from 0.001 to 10 μg mL−1), and detection limits were within the range of 0.03–0.57 ng mL−1. Single fiber and fiber-to-fiber reproducibility were less than 7.5 (n = 7) and 10.0% (n = 5), respectively. The recovery of the PACs spiked in natural water samples at 1 μg mL−1 ranged from 83.3 to 106.0%.  相似文献

Mequitazine has been found to be extractable from human plasma samples using MonoTip C18 tips, inside which C18-bonded monolithic silica gel was fixed. Human plasma (0.1 mL) containing mequitazine and cyproheptadine as an internal standard (IS) was mixed with 0.4 mL of distilled water and 25 μL of 1 M potassium phosphate buffer (pH 8.0). After centrifugation of the mixture, the supernatant fraction was extracted to the C18 phase of the tip by 25 repeated aspirating/dispensing cycles using a manual micropipettor. The analytes retained on the C18 phase were then eluted with methanol by five repeated aspirating/dispensing cycles. Without evaporation and reconstitution, the eluate was injected into a gas chromatograph injector and detected by a mass spectrometer with selected ion monitoring in the positive-ion electron impact mode. The separation of mequitazine and the IS from each other and from impurities was generally satisfactory using a DB-1MS capillary column (30 m × 0.32 mm i.d., film thickness 0.25 μm). The recoveries of mequitazine and the IS spiked into plasma were more than 90.0%. The regression equation for mequitazine showed excellent linearity in the range of 0.2-200 ng 0.1 mL−1, and the detection limit was 0.05 ng 0.1 mL−1of plasma. The intra-day and inter-day coefficients of variation for mequitazine in human plasma were not greater than 8.16 and 9.24%, respectively. Accuracy for the drug was in the range of 90.0-97.4%. The data obtained from determination of mequitazine in human plasma after oral administration of the drug are also presented.  相似文献

The ability and efficiency of micro precolumns made of C30 particles, monolithic silica C18 stationary phase and quartz wool coated with C30, which act as novel solid phase absorbing materials, for the on-line enrichment of aqueous polycyclic aromatic hydrocarbons (PAHs) in microcolumn liquid chromatography (LC) was investigated. The enrichment unit was designed in such a way that micro precolumns were directly connected to a 6-port micro injection valve via fused-silica tubing (0.05 mm I.D.) in order to minimize band broadening of the samples, and the enrichment efficiency of the three materials was tested using 14 PAHs, which are selected by the US Environmental Protection Agency (US EPA), as the analytes. The separation of PAHs was evaluated by using laboratory-made C30 or ODS capillary columns and the results were compared. There were no significant differences showed from the separation of PAHs in terms of peak signal between the C30 and ODS capillary columns, but the C30 capillary column was chosen for the following experiment due to its ability to produce better repeatability than the ODS column. By using the three kinds of precolumn materials, results showed that the precolumn packed with C30 particles as well as the capillary monolithic C18 precolumns (0.1 or 0.2 mm I.D.) provided better recovery than those of the quartz wool's. As long as the recovery and separation of the PAHs were concerned, 0.1 mm I.D. monolithic C18 precolumn showed the best results and the R.S.D.s (N = 7) for the retention time, peak area and peak height were between 0.70-1.5, 2.3-5.8 and 2.4-6.6%, respectively. Large volume injection up to 0.5 mL, i.e. 2500-fold enrichment, was possible and no negative effect on the separation profile was found. The LOD (S/N = 3) were between 0.10 and 4.6 pg mL−1, while the LOQ (S/N = 10) were in the range of 0.32-15 pg mL−1, which showed that the system is comparable to many major analytical techniques and is sensitive enough for the trace analysis of PAHs in environmental samples. The system was then applied to the determination of trace PAHs present in soil sample which was randomly taken from a nearby highway.  相似文献

An instrumental setup including on-line solid phased extraction coupled to capillary liquid chromatography-electrospray ionization-mass spectrometry (SPE-capLC-ESI-MS) has been constructed to improve the sensitivity for quantification of fluoxetine hydrochloride in human plasma. Prior to injection, 0.5 mL of plasma spiked with metronidazole (internal standard) was mixed with ammonium formate buffer for effective chloroform liquid-liquid extraction. The method was validated in the range 5-60 ng mL−1 fluoxetine, yielding a correlation coefficient of 0.999 (r2). The within-assay and between-assay precisions were between (8.5 and 11%) and (6.6 and 7.5%), respectively. The method was used to determine the amount of fluoxetine in a healthy male 14 h after an intake of one capsule of the antidepressant and anorectic Flutin®, which contains 20 mg fluoxetine per each capsule. Fluoxetine was detected, and the concentration was calculated to 9.0 ng mL−1 plasma. In the preliminary experiments, conventional LC-UV instrumentation was employed. However, it was found that employing a capillary column with an inner diameter of (0.3 mm I.D. × 50 mm, Zorbax C18) increased the sensitivity by a factor of ∼100, when injecting the same mass of analyte. Incorporating an easily automated C18 reversed phase column switching system with SPE (1.0 mm I.D. × 5.0 mm, 5 μm) made it possible to inject up to 100 μL of solution, and the total analysis time was 5.5 min.  相似文献
