Here, we describe a new approach for electrochemiluminescence (ECL) assay with Ru(bpy)32+-encapsulated silica nanoparticle (SiO2@Ru) as labels. A water-in-oil (W/O) microemulsion method was employed for one-pot synthesis of SiO2@Ru nanoparticles. The as-synthesized SiO2@Ru nanoparticles have a narrow size distribution, which allows reproducible loading of Ru(bpy)32+ inside the silica shell and of α-fetoprotein antibody (anti-AFP), a model antibody, on the silica surface with glutaraldehyde as linkage. The silica shell effectively prevents leakage of Ru(bpy)32+ into the aqueous solution due to strong electrostatic interaction between the positively charged Ru(bpy)32+ and the negatively charged surface of silica. The porous structure of silica shell allowed the ion to move easily through the pore to exchange energy/electrons with the entrapped Ru(bpy)32+. The as-synthesized SiO2@Ru can be used as a label for ultrasensitive detection of biomarkers through a sandwiched immunoassay process. The calibration range of AFP concentration was 0.05-30 ng mL−1 with linear relation from 0.05 to 20 ng mL−1 and a detection limit of 0.035 ng mL−1 at 3σ. The resulting immunosensors possess high sensitivity and good analytical performance.  相似文献

A new method for solid-phase extraction and preconcentration of trace amounts Hg(II) from environmental samples was developed by using sodium dodecyle sulphate-coated magnetite nanoparticles (SDS-coated Fe3O4 NPs) as a new extractant. The procedure is based on the adsorption of the analyte, as mercury-Michler's thioketone [Hg2(TMK)4]2+ complex on the negatively charged surface of the SDS-coated Fe3O4 NPs and then elution of the preconcentrated mercury from the surface of the SDS-coated Fe3O4 NPs prior to its determination by flow injection inductively coupled plasma-optical emission spectrometry. The effects of pH, TMK concentration, SDS and Fe3O4 NPs amounts, eluent type, sample volume and interfering ions on the recovery of the analyte were investigated. Under optimized conditions, the calibration curve was linear in the range of 0.2-100 ng mL−1 with r2 = 0.9994 (n = 8). The limit of detection for Hg(II) determination was 0.04 ng mL−1. Also, relative standard deviation (R.S.D.) for the determination of 2 and 50 ng mL−1 of Hg(II) was 5.2 and 4.7% (n = 6), respectively. Due to the quantitative extraction of Hg(II) from 1000 mL of the sample solution an enhancement factor as large as 1230-fold can be obtained. The proposed method has been validated using a certified reference materials, and also the method has been applied successfully for the determination of Hg(II) in aqueous samples.  相似文献

A simple dispersive solid-phase micro-extraction method based on CoFe2O4 nano-particles (NPs) functionalized with 8-hydroxyquinoline (8-HQ) with the aid of sodium dodecyl sulfate (SDS) was developed for separation of Al(III) ions from aqueous solutions. Al(III) ions are separated at pH 7 via complex formation with 8-HQ using the functionalized CoFe2O4 nano-particles sol solution as a dispersed solid-phase extractor. The separated analyte is directly quantified by a spectrofluorometric method at 370 nm excitation and 506 nm emission wavelengths. A comparison of the fluorescence of Al(III)–8-HQ complex in bulk solution and that of Al(III) ion interacted with 8-HQ/SDS/CoFe2O4 NPs revealed a nearly 5-fold improvement in intensity. The experimental factors influencing the separation and in situ monitoring of the analyte were optimized. Under these conditions, the calibration graph was linear in the range of 0.1–300 ng mL−1 with a correlation coefficient of 0.9986. The limit of detection and limit of quantification were 0.03 ng mL−1 and 0.10 ng mL−1, respectively. The inter-day and intra-day relative standard deviations for six replicate determinations of 150 ng mL−1 Al(III) ion were 2.8% and 1.7%, respectively. The method was successfully applied to direct determine Al(III) ion in various human serum and water samples.  相似文献

Aptamer-silver decahedral nanoparticles (Ag10NPs-aptamer) based detection was developed for protein. Ag10NPs were synthesized by photochemical method. The advantage of Ag10NPs was its tolerance of NaCl which facilitates the functionalization of silver nanoparticles with all kinds of ssDNA. Attaching aptamers to Ag10NPs could be achieved within 2 h, much faster than traditional methods. Human platelet-derived growth factor-BB (PDGF-BB) was used as a model protein to test the binding capacity of aptamers attached on Ag10NPs. Our data showed that the aptamer-Ag10NPs conjugates were successful in detecting human PDGF-BB. Furthermore, we developed an aptamer-Ag10NPs conjugates-based colorimetric sensor to detect PDGF-BB. The results showed a linear relationship between PDGF-BB concentrations (5 ng mL−1–200 ng mL−1) and ΔOD with excellent detection specificity in serum. Therefore, the sensor based on aptamer-Ag10NPs conjugates was highly effective and sensitive and had great promise for further development and applications.  相似文献

Enzyme-linked immunosorbent assay (ELISA) methods based on natural enzyme-labeled probes have been applied in the immunoassays, but most have some inevitable limitations (e.g. harsh preparation, purification and storage) and are unsuitable for routine use. Herein we synthesized a new class of irregular-shaped platinum nanoparticles (ISPtNP) with a mean length of 7.0 nm and a narrowing width from 2.0 to 5.0 nm along the longitudinal axes, which were utilized as peroxidase-like mimics for the development of colorimetric immunoassays. Compared with bioactive horseradish peroxidase (HRP), the synthesized ISPtNP exhibited a low Km value (~0.12 mM) and a high Kcat value (~2.27 × 104 s−1) for 3,3′,5,5′-tetramethylbenzidine (TMB) with strong thermal stability and pH tolerance. The catalytic mechanism of the ISPtNP toward TMB/H2O2 was for the first time discussed and deliberated in this work. Based on a sandwich-type assay format, two types of colorimetric immunoassay protocols were designed and developed for the detection of rabbit IgG (RIgG, as a model) by using the synthesized ISPtNP and conventional HRP as the labeling of detection antibodies, respectively. Similar detection limits (LODs) of 2.5 ng mL−1 vs. 1.0 ng mL−1 were obtained toward RIgG with the ISPtNP labeling compared to HRP format. Intra- and inter-assay coefficients of variation were less than 13%. Importantly, the ISPtNP-based assay system could be suitable for use in a mass production of miniaturized lab-on-a-chip devices and open new opportunities for protein diagnostics and biosecurity.  相似文献

In this paper a novel solid phase extraction method to determine Co(II) and Ni(II) using silica gel-polyethylene glycol (Silica-PEG) as a new adsorbent is described. The method is based on the adsorption of cobalt and nickel ions in alkaline media on polyethylene glycol-silica gel in a mini-column, elution with nitric acid and determination by flame atomic absorption spectrometry. The adsorption conditions such as NaOH concentration, sample volume and amount of adsorbent were optimized in order to achieve highest sensitivity. The calibration graph was linear in the range of 0.5-200.0 ng mL−1 for Co(II) and 2.0-100.0 ng mL−1 for Ni(II) in the initial solution. The limit of detection based on 3Sb was 0.37 ng mL−1 for Co(II) and 0.71 ng mL−1 for Ni(II). The relative standard deviations (R.S.D.) for ten replicate measurements of 40 ng mL−1 of Co(II), and Ni(II) were 3.24 and 3.13%, respectively. The method was applied to determine Co(II) and Ni(II) in black tea, rice flour, sesame seeds, tap water and river water samples.  相似文献

Polyclonal antibody (PAb) with broad-specificity for O,O-diethyl organophosphorus pesticides (OPs) against a generic hapten, 4-(diethoxyphosphorothioyloxy)benzoic acid, was produced. The obtained PAb showed high sensitivity to seven commonly used O,O-diethyl OPs in a competitive indirect enzyme-linked immunosorbent assay (ciELISA) using a heterologous coating antigen, 4-(3-(diethoxyphosphorothioyloxy)phenylamino)-4-oxobutanoic acid. The 50% inhibition value (IC50) was 348 ng mL−1 for parathion, 13 ng mL−1 for coumaphos, 22 ng mL−1 for quinalphos, 35 ng mL−1 for triazophos, 751 ng mL−1 for phorate, 850 ng mL−1 for dichlofenthion, and 1301 ng mL−1 for phoxim. The limit of detection (LOD) met the ideal detection criteria of all the seven OP residues. A quantitative structure-activity relationship (QSAR) model was constructed to study the mechanism of antibody recognition using multiple linear regression analysis. The results indicated that the frontier-orbital energies (energy of the highest occupied molecular orbital, EHOMO, and energy of the lowest unoccupied molecular orbital, ELUMO) and hydrophobicity (log of the octanol/water partition coefficient, log P) were mainly responsible for the antibody recognition. The linear equation was log(IC50) = −63.274EHOMO + 15.985ELUMO + 0.556 log P − 25.015, with a determination coefficient (r2) of 0.908.  相似文献

A novel label-free amperometric immunosensor is proposed for the ultrasensitive detection of zearalenone (ZEN) based on mesoporous carbon (MC) and trimetallic nanorattles (core/shell particles with movable cores encapsulated in the shells). The nanorattles are composed of special Au-core and imperfect AgPt-shell structure (Au@AgPt). The Au@AgPt nanorattles are loaded onto the MC by physical adsorption. The structure of the Au@AgPt nanorattles was characterized by using scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Energy dispersive X-ray spectroscopy (EDS) confirmed the composition of the synthesized nanorattles. Compared with monometallic and bimetallic nanoparticles (NPs), Au@AgPt nanorattles show a higher electron transfer rate due to the synergistic effect of the Au, Ag and Pt NPs. MC further improves the sensitivity of the immunosensor because of its extraordinarily large specific surface area, suitable pore arrangement and outstanding conductivity. The large specific surface area of MC and MC@Au@AgPt were characterized by the BET method. ZEN antibodies are immobilized onto the nanorattles via Ag–NH2 bonds and Pt–NH2 bonds. Cyclic voltammetry and square wave voltammetry were used to characterize the recognizability of ZEN. Under optimum experimental conditions, the proposed immunosensor exhibited a low detection limit (1.7 pg mL−1), a wide linear range (from 0.005 to 15 ng mL−1) as well as good stability, reproducibility and selectivity. An immunoaffinity (IA) sorbent with antibody fragments was prepared for the analysis of opioid peptides by on-line immunoaffinity solid-phase extraction capillary electrophoresis–mass spectrometry (IA-SPE-CE–MS). The antibody fragmentation was evaluated by MALDI-TOF-MS. Fab′ fragments obtained from a polyclonal IgG antibody against Endomorphins 1 and 2 (End1 and End2) were covalently attached to succinimidyl silica particles to prepare the IA sorbent. An IA-SPE-CE–MS methodology was established analyzing standard solutions of End1 and End2 and acceptable repeatability, linearity ranges and LODs (0.5 and 5 ng mL−1, respectively) were obtained. The LOD of End1 was slightly better than that previously obtained using an IA sorbent with intact antibodies (1 ng mL−1). In human plasma samples, End1 and End2 could be detected at 1 and 50 ng mL−1, respectively, which meant an improvement of 100 and 2-fold with regard to the LODs using an IA sorbent with intact antibodies (100 ng mL−1).  相似文献

A method based on cloud point extraction was developed to determine four kinds of estrogens: estriol (E3), estradiol (E2), estrone (E1), and progesterone (P) in water by high performance liquid chromatography separation and ultraviolet detection (HPLC-UV). The non-ionic surfactant Triton X-114 was chosen as extractant solvent. The parameters affecting extraction efficiency, such as concentrations of Triton X-114 and Na2SO4, equilibration temperature, equilibration time and centrifugation time were evaluated and optimized. Under the optimum conditions, preconcentration factors of 99 for E3, 73 for E2, 152 for E1 and 86 for P were obtained for 10 mL water sample. The detection of limitation was 0.23 ng mL−1 for E3, 0.32 ng mL−1 for E2, 0.25 ng mL−1 for E1 and 5.0 ng mL−1 for P. The proposed method was successfully applied to the determination of trace amount of estrogens in wastewater treatment plant (WWTP) effluent water and exposure water with 10 ng mL−1 E2 for toxicological study in our lab. For the case of WWTP effluent water samples, no estrogen was found. The accuracy of the proposed method was tested by recovery measurements of spiked samples and good recoveries of 81.2-99.5% were obtained.  相似文献

In this work, we reported a scanometric assay system based on the aptamer-functionalized silver nanoparticles (apt-AgNPs) for detection of platelet-derived growth factor-BB (PDGF-BB) protein. The aptamer and ssDNA were bound with silver nanoparticles by self-assembly of sulfhydryl group at 5′ end to form the apt-AgNPs probe. The apt-AgNPs probe can catalyze the reduction of metallic ions in color agent to generate metal deposition that can be captured both by human eyes and a flatbed scanner. Two different color agents, silver enhancer solution and color agent 1 (10 mM HAuCl4 + 2 mM hydroquinone) were used to develop silver and gold shell on the surface of AgNPs separately. The results demonstrated that the formation of Ag core–Au shell structure had some advantages especially in the low concentrations. The apt-AgNPs probe coupled with color agent 1 showed remarkable superiority in both sensitivity and detection limit compared to the apt-AuNPs system. The apt-AgNPs system also produced a wider linear range from 1.56 ng mL−1 to 100 ng mL−1 for PDGF-BB with the detection limit lower than 1.56 ng mL−1. The present strategy was applied to the determination of PDGF-BB in 10% serum, and the results showed that it had good specificity in complex biological media.  相似文献

A simple extraction method for the analysis of PGE2 and PGF in gonad samples from Atlantic cod and further quantification by using liquid chromatography–tandem mass spectrometry is proposed. The evaluation of the best solvent extraction conditions and the analytical performance parameters are reported. The method was highly selective for both prostaglandins and the calibration curves, based on the internal standard method, were linear between 5 and 1000 ng mL−1 for PGE2 and PGF, with limits of detection of 1 ng mL−1 and 1.5 ng mL−1 and recovery values of 99.999 ± 0.002 and 99.967 ± 0.023 respectively. The homogenization of samples using liquid nitrogen combined with the developed extraction protocol can be implemented in different types of biological tissues.  相似文献

A new method has been developed for the determination of gold based on separation and preconcentration with a microcolumn packed with nanometer TiO2 immobilized on silica gel (immobilized nanometer TiO2) prior to its determination by flame atomic absorption spectrometry. The optimum experimental parameters for preconcentration of gold, such as pH of the sample, sample flow rate and volume, eluent and interfering ions, have been investigated. Gold could be quantitatively retained by immobilized nanometer TiO2 in the pH range of 8-10, then eluted completely with 0.1 mol L−1 HNO3. The detection limit of this method for Au was 0.21 ng mL−1 with an enrichment factor of 50, and the relative standard deviation (R.S.D.) was 1.8% at the 100 ng mL−1 Au level. The method has been applied for the determination of trace amounts of Au in geological and water samples with satisfactory results.  相似文献

A new method was developed for determination of micro amounts of nucleic acids based on near-infrared (near-IR) fluorescence recovery, employing a two-reagent system which is composed of an anionic tetracarboxy aluminum phthalocyanine (AlC4Pc) and a cationic tetra-N-hexadecylpyridiniumyl porphyrin (TC16PyP). The fluorescence of the AlC4Pc, with the maximum emission wavelength at 701 nm, could be quenched by TC16PyP at its proper concentration, but recovered by adding nucleic acids. Under optimal conditions, the recovered fluorescence is proportional to the concentration of nucleic acids. The calibration graphs are linear over the range of 1-200 ng mL−1 for fish sperm DNA (FS DNA) and 2-400 ng mL−1 for calf thymus DNA (CT DNA). The corresponding detection limits are 0.59 ng mL−1 for FS DNA and 0.82 ng mL−1 for CT DNA, respectively. Four synthetic and three real nucleic acid samples were determined with satisfactory results.  相似文献

A simple, sensitive, and specific method with gas chromatography-mass spectrometry was developed for simultaneous extraction and derivatization of amphetamines (APs) and 3,4-methylenedioxyamphetamines (MDAs) in human urine by using a monolithic silica spin column. All the procedures, such as sample loading, washing, and elution were performed by centrifugation. APs and MDAs in urine were adsorbed on the monolithic silica and derivatized with propyl chloroformate in the column. Methamphetamine-d5 was used as an internal standard. The linear ranges were 0.01-5.0 μg mL−1 for methamphetamine (MA) and 3,4-methylenedioxymethamphetamine (MDMA) and 0.02-5.0 μg mL−1 for amphetamine (AP) and 3,4-methylenedioxyamphetamine (MDA) (coefficient of correlation ≧0.995). The recovery of APs and MDAs in urine was 84-94%, and the relative standard deviation of the intra- and interday reproducibility for urine samples containing 0.1, 1.0, and 4.0 μg mL−1 of APs and MDAs ranged from 1.4% to 13.6%. The lowest detection limit (signal-to-noise ratio ≧ 3) in urine was 5 ng mL−1 for MA and MDMA and 10 ng mL−1 for AP and MDA. The proposed method can be used to perform simultaneous extraction and derivatization on spin columns that have been loaded with a small quantity of solvent by using centrifugation.  相似文献

Nanogold exhibits strong catalytic effect on the slow reaction between glucose and Fehling reagent at 70 °C. The production of Cu2O particles have two stronger resonance scattering (RS) peaks at 390 nm and 505 nm. The catalytic effect of nanogold-labeled goat anti-human IgG (AuIgG) on the reaction was investigated with the RS technique. Coupled the immunoreaction and the immunonanogold catalytic reaction and centrifugal technique, a highly sensitive and selective RS method was developed for the detection of immunoglobulin G (IgG) as a model. With the concentration of IgG increased, the RS intensity at 505 nm decreased. The decreased intensity at 505 nm ΔI505 nm was proportional to IgG concentration in the range of 0.13-53.3 ng mL−1, with a detection limit of 0.04 ng mL−1 IgG. This new immunonanogold-catalytic Cu2O-particle RS bioassay was applied to the determination of IgG in serum sample, with high sensitivity, good selectivity, and low cost.  相似文献

An ultrasensitive electrochemiluminescence (ECL) immunosensor based on CdSe quantum dots (QDs) has been designed for the detection of clenbuterol. The immunosensor was fabricated by layer by layer and characterized with atomic force microscopic images (AFM) and electrochemical impedance spectra (EIS). In oxygen-saturated pH = 9.0 Tris-HCl buffer, a strong ECL emission of QDs could be observed during the cathodic process due to the H2O2 product from electrochemical reduction of dissolved oxygen. Upon the formation of immunocomplex, the second antibody labeled with horseradish peroxidase was simply immobilized on the electrode surface. The ECL emission decreased since steric hindrance of the immunocomplex slowed down the electron-transfer speed of dissolved oxygen, and also could be greatly amplified by an enzymatic cycle to consume the self-produced coreactant. Using clenbuterol as model analyte, the ECL intensity was determined by the concentration of competitive immunoassay of clenbuterol with a wide calibration in the range of 0.05 ng mL−1 to 1000 ng mL−1, and a low detection limit was 0.02 ng mL−1. The immunosensor shows good stability and fabrication reproducibility. It was applied to detecting practical samples with the satisfactory results. This immunosensing strategy opens a new avenue for detection of residue and application of QDs in ECL biosensing.  相似文献

CeO2-deposited mesoporous silica nanoparticles were synthesized as a probe to determine carcinoembryonic antigen (CEA) in serum by inductively coupled plasma-mass spectrometry (ICP-MS). The prepared mesoporous nanoparticles were modified and tagged to the target for sandwich-type immunoassay. Fe3O4 magnetic nanoparticles (MNPs) were also synthesized and immobilized with antibody to extract the target biomarker. The calibration curve of the synthesized CeO2-deposited silica nanoparticles, which was plotted by the signal ratio of 140Ce/57Fe measured by ICP-MS vs. the concentration of CEA, showed excellent linearity and sensitivity owing to the signal amplification and low spectral interference. Under optimal conditions, the sandwich-type analytical method was applied to determine CEA in serum spiked in the range of 0.001–5 ng mL−1 and showed a limit of detection of 0.36 ng mL−1. Since the deposited CeO2 in the mesoporous silica layer can be substituted by other metal compounds, various kinds of metal-deposited nanoparticles can be prepared as probe materials for multiplex detection in bioanalysis.  相似文献

We proposed an electrochemical DNA sensor by using peroxidase-like magnetic ZnFe2O4–graphene quantum dots (ZnFe2O4/GQDs) nanohybrid as a mimic enzymatic label. Aminated graphene and Pd nanowires were successively modified on glassy carbon electrode, which improved the electronic transfer rate as well as increased the amount of immobilized capture ssDNA (S1). The nanohybrid ZnFe2O4/GQDs was prepared by assembling the GQDs on the surface of ZnFe2O4 through a photo-Fenton reaction, which was not only used as a mimic enzyme but also as a carrier to label complementary ssDNA (S3). By synergistically integrating highly catalytically activity of nano-sized GQDs and ZnFe2O4, the nanohybrid possessed highly-efficient peroxidase-like catalytic activity which could produce a large current toward the reduction of H2O2 for signal amplification. Thionine was used as an excellent electron mediator. Compared with traditional enzyme labels, the mimic enzyme ZnFe2O4/GQDs exhibited many advantages such as environment friendly and better stability. Under the optimal conditions, the approach provided a wide linear range from 10−16 to 5 × 10−9 M and low detection limit of 6.2 × 10−17 M. The remarkable high catalytic capability could allow the nanohybrid to replace conventional peroxidase-based assay systems. The new, robust and convenient assay systems can be widely utilized for the identification of other target molecules.  相似文献

The effect of dimensions (length and external diameter) of multi-walled carbon nanotubes (MWCNTs) on its preconcentration efficiency towards some metal ions (Pb2+, Cd2+, Cu2+, Zn2+ and MnO4) from environmental waters prior to their analysis by flame atomic absorption spectroscopy (FAAS) was investigated. MWCNTs (as-received from the manufacturer) of various external diameters and lengths were involved. Other variables optimized included effects of pH of water sample, composition and volume of eluent, mass of the MWCNTs, breakthrough volume and coexisting ions. Maximum recovery of metal ions was obtained at pH 9 where it was thought that precipitation of metals as their hydroxides played the major factor in metals uptake by MWCNT. It was suggested that the use of appropriate dimensions of MWCNTs may support the trapping process of the precipitated metal hydroxides by MWCNTs. It was found that long MWCNT of length 5-15 μm and external diameter 10-30 nm gave the highest enrichment efficiency towards almost all the targeted metal ions. It could be used for preconcentration of MnO4, Cu2+, Zn2+ and Pb2+ with almost full recovery; but not for Cd2+ due to its low recovery. The optimized solid phase extraction (SPE) procedure was capable of determining metal ions in the linear range 20-100 ng mL−1 (except for Zn2+ from 20 to 150 ng mL−1). Detection limits were 0.709 ng mL−1 for MnO4, 0.278 ng mL−1 for Pb2+, 0.465 ng mL−1 for Cu2+, 0.867 ng mL−1 for Zn2+. Application of the optimized SPE procedure to environmental waters (tap water, reservoir water and stream water) gave spike recoveries of the metals in the range of 81-100%.  相似文献
