An efficient synthesis of 1,2,3‐triaroylindolizines has been developed via CuBr2‐promoted reaction of three molecules of aromatic methyl ketones and one molecule of pyridine derivative. A wide range of methyl aryl ketones and methyl heteroaryl ketones took part in the reaction and generate 1,2,3‐triaroylindolizines in good yields. This protocol also features such advantages as mild reaction conditions and high atom economy and step economy.  相似文献

The condensation of α,β‐unsaturated ketones with substituted o‐aminothiophenols, obtained by reductive cleavage of the corresponding disulfides in the presence of triphenylphosphine, is an effective method for the synthesis of 2,4‐diaryl‐2,3‐dihydro‐1,5‐benzothiazepines under neutral conditions.  相似文献

The treatment of α‐bromoalkyl aryl ketones and 2‐(propan‐2‐ylidene)hydrazine carbothioamide afforded 4‐aryl‐2‐(2‐(propan‐2‐ylidene)hydrazinyl)thiazoles via a Hantzsch‐thiazole synthesis, which reacted with 4‐aryl‐2,4‐diketoesters via a sequential Knorr‐pyrazole reaction to deliver a variety of aryl‐substituted ethyl 1‐(thiazol‐2‐yl)‐1H‐pyrazole‐3‐carboxylates in a one‐pot fashion with moderate to high yields. The key intermediates 4‐aryl‐2,4‐diketoesters, existing as its enolic lithium salt, were synthesized in situ by a high‐yield tert‐BuOLi‐mediated Claisen condensation of alkylphenones and diethyl oxalate. This class of elegant molecule comprises aryl groups on the two different heterocyclic cores, and the configurations of two representative molecules were determined by single crystal X‐ray crystallography.  相似文献

An efficient chemoselective general procedure for the synthesis of γ‐substituted β,γ‐unsaturated α‐ketomethylthioesters from α,β‐unsaturated ketones has been achieved through an unprecedented PPh3?HBr‐DMSO mediated oxidative bromination and Kornblum oxidation sequence. The newly developed reagent system serves admirably for the synthesis of α‐bromoenals from enals. Furthermore, AuCl3‐catalyzed efficient access to 3(2H)‐furanones from the above intermediates under extremely mild conditions are described.  相似文献

γ‐Ketophosphine chalcogenides, precursors for plethora of novel functionalized phosphine chalcogenides and phosphines, are synthesized by chemo‐ and regioselective addition of secondary phosphine chalcogenides to β,γ‐ethylenic ketones under catalyst‐ and solvent‐free conditions (80–100°C, 8–70 h) in excellent yields. The straightforward superbase‐catalyzed synthesis of starting β,γ‐ethylenic ketones from ketones and acetylenes insures the expedient access to the target γ‐ketophosphine chalcogenides.  相似文献

The synthesis of α‐substituted carbonyl compounds is of great importance due to their ubiquity in both natural and man‐made biologically active compounds. The field of hypervalent iodine chemistry has been a great contributor to access these molecules. For example, the α‐oxidation of carbonyl compounds has been one of the most investigated iodine(III)‐mediated stereoselective transformations. Yet, it is also the transformation that has met the most challenge in terms of achieving high stereoselectivities. The different mechanistic pathways of the iodine(III)‐mediated α‐tosyloxylation of ketones have been investigated. The calculations suggest an unprecedented iodine(III)‐promoted enolization process. Indications that iodonium intermediates could serve as proficient Lewis acids are reported. This concept could have broad impact and foster new developments in the field of hypervalent iodine chemistry.  相似文献

Inspired by the biogenetic origin of goniomitine, new synthetic bio‐inspired fragmentation strategies for the synthesis of functionalized 2‐quinolinones and indolones have been developed. Remarkable synthetic efficiency was achieved by telescoping several transformations into one‐pot reactions, allowing for the direct coupling of 2‐alkynyl‐anilines and diazo ketones. The synthetic utility was demonstrated by the 5‐step asymmetric total synthesis of (−)‐goniomitine from 2‐ethyl‐cyclopentanone.  相似文献

A [1,4]‐S‐ to O‐silyl migration has been exploited to chemoselectively transform esters into ketones by using organolithium reagents, allowing multicomponent synthesis of α‐thioketones. Mechanistic studies reveal that this migration proceeds in an intramolecular manner and is more favorable than the corresponding [1,5]‐S‐ to O‐ and [1,3]‐C‐ to O‐silyl migrations. The resulting α‐thioketones are valuable building blocks for the synthesis of cyclic or multifunctionalized organosulfur compounds.  相似文献

Chiral α‐amino ketones are excellent nucleophiles for stereoselective palladium‐catalyzed allylic alkylations. Both chiral as well as achiral allylic substrates can be applied, while the stereochemical outcome of the reaction is controlled by the chiral ketone enolate. The substituted amino ketones formed can be reduced stereoselectively, and up to five consecutive stereogenic centers can be obtained. This approach can be used for the synthesis of highly substituted piperidine derivatives.  相似文献

Late‐stage synthesis of α,β‐unsaturated aryl ketones remains an unmet challenge in organic synthesis. Reported herein is a photocatalytic non‐chain‐radical aroyl chlorination of alkenes by a 1,3‐chlorine atom shift to form β‐chloroketones as masked enones that liberate the desired enones upon workup. This strategy suppresses side reactions of the enone products. The reaction tolerates a wide array of functional groups and complex molecules including derivatives of peptides, sugars, natural products, nucleosides, and marketed drugs. Notably, addition of 2,6‐di‐tert‐butyl‐4‐methyl‐pyridine enhances the quantum yield and efficiency of the cross‐coupling reaction. Experimental and computational studies suggest a mechanism involving PCET, formation and reaction of an α‐chloro‐α‐hydroxy benzyl radical, and 1,3‐chlorine atom shift.  相似文献

A versatile synthesis of 3‐cyano‐2‐pyridones via a one‐pot, four‐component condensation of ethyl cyanoacetate, ketones, aldehydes, and ammonium acetate under very mild conditions has been developed. The development of efficient methods for stereocontrolled synthesis of polyol derivatives has been of continuing interest for the synthetic community. We describe herein tandem olefin cross‐metathesis/hemiacetalization/intramolecular oxa‐Michael addition of allylic/homoallylic alcohols, α,β‐unsaturated ketones, and aldehydes, which enabled the synthesis of syn‐1,2‐ and syn‐1,3‐diol derivatives in a step‐economical manner. A series of differentially protected polyol derivatives could be obtained in subsequent transformations via chemoselective/regioselective cleavage of the acetal moiety of the tandem reaction products.  相似文献

Zirconium(IV) chloride catalyzed efficient one-pot synthesis of β-amino/β-acetamido carbonyl compounds at room temperature is described. In the presence of ZrCl4, the three-component Mannich-type reaction via a variety of in situ generated aldimines, with various ketones, aromatic aldehydes and aromatic amines in ethanol, led to the formation of β-amino carbonyl compounds and the four-component Mannich-type reaction of aromatic aldehydes with various ketones, acetonitrile and acetyl chloride resulted in the corresponding β-acetamido carbonyl compounds in high to excellent yields. This methodology has also been applied towards the synthesis of dimeric β-amino/β-acetamido carbonyl compounds.  相似文献

An efficient synthesis of (±)‐2‐monosubstituted and (±)‐2,2‐disubstituted 2,3‐dihydro‐4(1H)‐quinazolinones has been developed using a dissolving metal reduction‐condensative cyclization strategy. Treatment of 2‐nitrobenzamide and an aldehyde or ketone with iron powder in refluxing acetic acid affords high yields of the title compounds. More complex ring systems are available by incorporating additional reactive functionality γ to the carbonyl of the aldehyde or ketone substrate. The scope and limitations of the process along with optimized procedural details are presented. A catalytic enantioselective method for the synthesis of 1,4‐keto‐alkenylboronate esters by a rhodium‐catalyzed conjugate addition pathway is disclosed. A variety of novel, bench‐stable alkenyl gem‐diboronate esters are synthesized. These easily accessible reagents react smoothly with a collection of cyclic α,β‐unsaturated ketones, generating a new C?C bond and stereocenter. Products are isolated in up to 99 % yield with greater than 20:1 E/Z and greater than 99:1 e.r. Mechanistic studies show the site‐selectivity of transmetalation and reactivity is ligand dependent. The utility of the approach is highlighted by gram‐scale synthesis of enantioenriched cyclic 1,4‐diketones, and stereoselective transformations of the products by hydrogenation, allylation, and isomerization.  相似文献

An expeditious preparation of the 6‐exo‐hydroxybicyclo[2.2.2]octan‐2‐one ethylene dithioacetal 2b , a key intermediate in the synthesis of (+)‐13‐stemarene ( 4 ) and (+)‐18‐deoxystemarin ( 5 ) is described. Compound 2b was obtained as the major product by equilibrating the endo rich mixture of 6‐hydroxybicyclo[2.2.2]octan‐2‐one ethylene dithioacetals 2 with TsOH in benzene at reflux, easily available from the corresponding hydroxy ketones 9 . The model experiments which preceeded the above transformation, not previously described in the literature, are also presented.  相似文献

Reported herein is the first example of 2‐allylazaarenes in asymmetric catalysis. Highly γ‐selective allylation was demonstrated for activated ketones, including isatins and trifluoromethyl ketones. In the presence of either an amino‐acid‐based tertiary amine or quaternary ammonium salt catalyst, two series of tertiary hydroxy‐containing moieties were installed at the remote δ‐position of azaarenes in good chemical yields, excellent enantioselectivities, and E /Z ratios. The success of current γ‐selective reactions should provide inspiration for expansion to other allylazaarene derivatives and would open up new paradigms for the synthesis of chiral γ‐ and/or δ‐functionalized azaarenes.  相似文献
