In the case of poly(lactic acid) stereocopolymers, it has been shown that the hydrolytic degradation of derived devices depends very much on whether zinc lactate or tin octoate was used to polymerize lactides. In contrast, no effect was found in the case of nanoparticles derived from poly(dl-lactic acid)-block-poly(ethylene glycol) copolymers obtained by anionic polymerization of dl-lactide initiated by the sodium salt of monomethoxypoly(ethylene glycol) or by coordination-insertion polymerization of dl-lactide initiated by monomethoxypoly(ethylene glycol) in the presence of tin octoate as catalyst. To understand the difference of behaviour, in vitro hydrolytic degradation of thick plates made of the same copolymers but under different conditions was investigated. Changes were monitored by 1H Nuclear Magnetic Resonance, Size Exclusion Chromatography, Electrospray Mass Spectrometry and Capillary Zone Electrophoresis. It is shown that chain cleavage occurred from the very beginning of degradation and that plates disintegrated after 13 weeks. In all cases, degradation proceeded faster inside than at the surface, in contrast to what was observed for nanoparticles. Tin-type copolymer plates degraded more slowly than sodium macroalcoholate-type ones and were sensitive to purification conditions, in contrast to the latter.  相似文献

A novel biocompatible BAB amphiphilic triblock copolymers which consist of poly(ethylene glycol) (PEG) and poly(ε-benzyloxycarbonyl l-Lysine) (PLL(Z)) were synthesized by anion ring opening polymerization of N-carboxyanhydride of ε-benzyloxycarbonyl-l-Lysine (l-Lys(z)-NCA) using α-amino-ω-amino-poly(ethylene glycol) as initiator in DMF. The block copolymers were characterized by IR, 1H-NMR, GPC, DSC. The results showed that the block copolymers were of narrow molecular distribution and well defined structure. The self-assembled behaviors of block copolymers in aqueous medium were investigated. The effects of various factors, such as cosolvents, initial concentration, temperature, annealing time and times of frozen-thaw cycle etc., on the aggregate morphologies were investigated by transmission electron microscopy (TEM). Different morphological aggregates such as sphere, rod and vesicle etc. could be obtained at controlled conditions, at the same time, a novel helical aggregate was observed. These regular nanometer structures have potential applications in biomedicine due to the biocompatibility of two blocks.  相似文献

Two synthetic ways were experimented to prepare new architectures of block copolymers made of poly(ethylene glycol) (PEG) and poly(methylthiirane). The coupling of both blocks conveniently end-capped as well as anionic polymerization of methylthiirane initiated by PEG-thiols gave readily the copolymers. Their characterization by 1H NMR, SEC and IR confirmed the expected structures.  相似文献

Amphiphilic block copolymers, methoxy poly(ethylene glycol)-b-poly(valerolactone) (mPEG-b-PVL), were synthesized via ring opening polymerization of δ-valerolactone in the presence of methoxy poly(ethylene glycol) (mPEG). The copolymers form micelle-like nanoparticles by their amphiphilic characteristics and their structures were examined by Nuclear Magnetic Resonance (NMR). The sizes of nanoparticles ranged from 60 to 120 nm as measured by dynamic light scattering detection, and were larger with higher molecular weight of the copolymers. The Critical Micelle Concentration (CMC) of these nanoparticles in water decreased with increasing molecular weight of hydrophobic segment. Stability analysis showed that the micellar solutions maintain their sizes at 37 °C for six weeks without aggregation or dissociation. The lyophilization method was better than the evaporation method when camptothecin (CPT) was incorporated to the micelles. The former method yielded higher CPT loading efficiency and lower aggregation. The loading efficiency of CPT could be more than 96% and a steady release rate of CPT was kept for twenty six days. Moreover, the mPEG-b-PVL polymeric micelles offered good protection of CPT lactone form at 37 °C for sixteen days. The copolymers showed no cytotoxicity towards L929 mouse muscular cells when incubated for one day. Taken together, the mPEG-b-PVL copolymer has potential to be used for the delivery of CPT or other similar drugs.  相似文献

Poly(ethylene oxide)-b-poly(styrene) block copolymers were prepared in the form of latex particles by emulsion polymerization of styrene with poly(ethylene glycol)–azo-initiators as well as with the redox initiation system poly(ethylene glycol)/Ce4+. The emulsion polymerization can be carried out in the absence of additional stabilizers if the chain length of the poly(ethylene glycol) is greater than 40. The latex particles as well as the copolymers were characterized by capillary hydrodynamic fractionation, 13C-nuclear magnetic resonance (NMR) spectroscopy and Fourier transform infrared spectroscopy. By 13C-NMR spectroscopy a side reaction of the primary radicals arising from the azo-initiator was found which can contribute to the low efficiency of azo-initiators in emulsion polymerization.  相似文献

Biodegradable and amphiphilic diblock copolymers [polylactide-block-poly(ethylene glycol)] and triblock copolymers [polylactide-block-poly(ethylene glycol)-block-polylactide] were synthesized by the anionic ring-opening polymerization of lactides in the presence of poly(ethylene glycol) methyl ether or poly(ethylene glycol) and potassium hexamethyldisilazide as a catalyst. The polymerization in toluene at room temperature was very fast, yielding copolymers of controlled molecular weights and tailored molecular architectures. The chemical structure of the copolymers was investigated with 1H and 13C NMR. The formation of block copolymers was confirmed by 13C NMR and differential scanning calorimetry investigations. The monomodal profile of the molecular weight distribution by gel permeation chromatography provided further evidence of block copolymer formation as well as the absence of cyclic species. Additional confirmation of the block copolymers was obtained by the substitution of 2-butanol for poly(ethylene glycol); butyl groups were clearly identified by 1H NMR as polymer chain end groups. The conditions of synthesis of statistical poly(ethylene succinate-co-terephthalate) copolymers (2GTS) and high molecular weight poly(ethylene succinate) (PES) with good hydrolytic and optical parameters, designed for the production of biodegradable products and resins, are presented in this article. Copolymers were prepared by melt polycondensation of bis-(β-hydroxyethylene terephthalate) (BHET) and succinic acid (SA) with excess of ethylene glycol (2G) in the presence of a novel titanium/silicate catalyst (C-94) and catalytic grade of germanium dioxide (GeO2) as cocatalyst. The chemical structure and physical properties of those materials were characterized by 1H NMR, FT-IR, dynamical-mechanical thermal analyses (DMTA), differential scanning calorimetry (DSC), solution viscosity and spectroscopic methods. The hydrolytic degradation was performed in a water solution with variable pH, also in garden soil and in compost. The highest hydrolytic degradation rate was observed for pH 4 and for compost. Better hydrolytic degradation values in compost medium were observed for copolyester prepared in the presence of GeO2 as polycondensation cocatalyst. The copolyester with 40 mol% of aliphatic units was chosen for industrial syntheses which were performed in ELANA and subsequently the processing parameters and compatibility with potato starch of this polyester were checked by BIOP Biopolymer Technologies AG.  相似文献

A set of amphiphilic poly(ethylene glycol)-b-poly(ethylene brassylate) (PEG-b-PEB) copolymers based on the PEB hydrophobic block was first synthesized by ring-opening polymerization of ethylene brassylate with an organic catalyst. The EB/PEGmolar ratios and reaction times were adjusted to achieve different chain lengths of PEB. Block copolymers that were characterized by 1H NMR and GPC could selfassemble into multimorphological aggregates in aqueous solution, which were characterized by DLS and TEM. The hydrophobic doxorubicin (DOX) was chosen as a drug model and successfully encapsulated into the nanoparticles. The release kinetics of DOX were investigated.  相似文献

Well-defined ABC block copolymers consisting of poly(ethylene oxide) monomethylene ether (MPEO) as A block, poly(styrene) (PS) as B block and poly(γ-benzyl-l-glutamate) (PBLG) as C block were synthesized by the combination of atom transfer radical polymerization (ATRP) and click reactions. The bromine-terminated diblock copolymer poly(ethylene oxide) monomethylene ether-block-poly(styrene) (MPEO-PS-Br) was prepared by ATRP of styrene initiated with macro-initiator MPEO-Br, which was prepared from the esterification of MPEO and 2-bromoisobutyryl bromide, and converted into the azido-terminated diblock copolymer MPEO-PS-N3 by simple nucleophilic substitutions in DMF in the presence of sodium azide. Propargyl-terminated PBLGs were synthesized by ring-opening polymerization of γ-benzyl-l-glutamate-N-carboxyanhydride in DMF at room temperature using propargyl amine as an initiator. ABC triblock copolymers MPEO-PS-PBLG with a wide range of number-average molecular weights from 1.55 to 3.75 × 104 and a narrow polydispersity from 1.07 to 1.10 were synthesized via the click reaction of MPEO-PS-N3 and the propargyl-terminated PBLG in the presence of CuBr and 1,1,4,7,7-pentamethyldiethylenetriamine (PMDETA) catalyst system. The structures of these ABC block copolymers and corresponding precursors were characterized by NMR, IR and GPC. The results showed that click reaction was efficient. Therefore, a facile approach was offered to synthesize ABC triblock copolymers composed of crystallizable polymer MPEO, conventional vinylic polymer PS and rod-like α-helix polypeptide PBLG.  相似文献

Well-defined amphiphilic block copolymers composed of hydrophilic and hydrophobic blocks linked through an acid-labile acetal bond were synthesized directly by RAFT polymerization using a new poly(ethylene glycol) (PEG) macroRAFT agent modified with an acid-labile group at its R-terminal. The new macroRAFT agent was used for polymerization of poly(t-butyl methacrylate) (PtBMA) or poly(cholesterol-methacrylate) (PCMA) to synthesize well-defined block copolymers with a PEG block sheddable under acidic conditions. The chain extension polymerization kinetics showed known traits of RAFT polymerization. The molecular weight distributions of the copolymers prepared using the new macroRAFT agent remained below 1.2 during the polymerizations and the molecular weight of the copolymers was linearly proportional to monomer conversions. The acid-catalyzed hydrolysis behavior of the PEG-macroRAFT agent and the PEG-b-PtBMA (Mn = 13,600 by GPC, PDI = 1.10) was studied by GPC, 1H NMR and UV–vis spectroscopy. The half-life of acid-hydrolysis was 70 min at pH 2.2 and 92 h at pH 4.0. The potential use of the pH-labile shedding behavior of the copolymers was demonstrated by conjugating a thiol-modified siRNA to ω-pyridyldisulfide modified PEG-b-PCMA. The resultant PEG-b-PCMA-b-siRNA triblock modular polymer released PCMA-b-siRNA segment in acidic and siRNA segment in reductive conditions, as confirmed by polyacrylamide gel electrophoresis.  相似文献

Poly(ϵ-caprolactone)–poly(ethylene glycol)–poly(ϵ-caprolactone) triblock copolymers (PECL) covering a wide range of poly(ethylene glycol) (PEG) lengths were synthesized with alkali metal alkoxide derivatives of poly(ethylene glycol). The effects of various factors, such as amount of the initiator, reaction time and temperature, polarity of solvent, length of PEG segment, and counterion on the polymerization were investigated. The copolymers were characterized by 1H-NMR, IR, GPC, and DSC. It was found that THF system is superior to toluene system. The conversion of the monomer increased with increase of the initiator concentration. High molecular weight of the copolymer and high conversion of the monomer was obtained at below 30°C within 5 min. The polymerization process was studied by GPC and the coexistence of propagation and transesterification reaction was found, which leaded to relatively broad molecular weight distribution of the copolymers. © 1997 John Wiley & Sons, Inc.  相似文献

Core‐shell structured nanoparticles of poly(ethylene glycol) (PEG)/polypeptide/poly(D ,L ‐lactide) (PLA) copolymers were prepared and their properties were investigated. The copolymers had a poly(L ‐serine) or poly(L ‐phenylalanine) block as a linker between a hydrophilic PEG and a hydrophobic PLA unit. They formed core‐shell structured nanoparticles, where the polypeptide block resided at the interface between a hydrophilic PEG shell and a hydrophobic PLA core. In the synthesis, poly(ethylene glycol)‐b‐poly(L ‐serine) (PEG‐PSER) was prepared by ring opening polymerization of N‐carboxyanhydride of O‐(tert‐butyl)‐L ‐serine and subsequent removal of tert‐butyl groups. Poly(ethylene glycol)‐b‐poly(L ‐phenylalanine) (PEG‐PPA) was obtained by ring opening polymerization of N‐carboxyanhydride of L ‐phenylalanine. Methoxy‐poly(ethylene glycol)‐amine with a MW of 5000 was used as an initiator for both polymerizations. The polymerization of D ,L ‐lactide by initiation with PEG‐PSER and PEG‐PPA produced a comb‐like copolymer, poly(ethylene glycol)‐b‐[poly(L ‐serine)‐g‐poly(D ,L ‐lactide)] (PEG‐PSER‐PLA) and a linear copolymer, poly(ethylene glycol)‐b‐poly(L ‐phenylalanine)‐b‐poly(D ,L ‐lactide) (PEG‐PPA‐PLA), respectively. The nanoparticles obtained from PEG‐PPA‐PLA showed a negative zeta potential value of ?16.6 mV, while those of PEG‐PSER‐PLA exhibited a positive value of about 19.3 mV. In pH 7.0 phosphate buffer solution at 36 °C, the nanoparticles of PEG/polypeptide/PLA copolymers showed much better stability than those of a linear PEG‐PLA copolymer having a comparable molecular weight. © 2011 Wiley Periodicals, Inc. 利用L 谷氨酸和苯甲醇反应制备了L 谷氨酸 苄酯 ,然后将其与三聚光气反应制备了N 羧基 L 谷氨酸 环内酸酐 (NCA) .以聚乙二醇单甲醚 (MPEG)为原料 ,制备了端氨基聚乙二醇单甲醚 (MPEG NH2 ) ,并以此作为引发剂 ,引发NCA开环聚合 ,合成了不同分子量的聚L 谷氨酸 苄酯 聚乙二醇单甲醚 (PBGM )嵌段共聚物 .利用IR、1 H NMR、DSC、GPC等方法对共聚物结构进行了表征 .结果表明 ,MPEG NH2 引发NCA开环聚合得到的是嵌段共聚物 ,通过1 H NMR谱得到共聚物组成及数均分子量 ;随着共聚物中MPEG含量的增高 ,聚L 谷氨酸 苄酯的亲水性有所改善  相似文献

Block and graft copolymers with poly(N-isopropylacrylamide) and poly[(N-acetylimino)ethylene] (PNAI) sequences were synthesized via PNAI derivatives (macroinitiators or macromers). The polymerization yields for block copolymers synthesized in ethanol, using the PNAI macroinitiator, were low (<10%), except where photochemical polymerization was applied. By contrast, for the copolymerizations of N-isopropylacrylamide with the PNAI macromers, performed in alcoholic solution, quite high polymerization yields, around 80-90%, were reached. 1H-NMR and IR spectral and differential scanning calorimeter thermal data confirmed the copolymer formation. Thermosensitivity of the copolymers was investigated by means of turbidimetric technique as a function of their nature, average molecular weight and composition. It was found that the length of the chain of the PNAI macromer and the content in hydrophilic PNAI units of the resulted copolymer affected this behavior.  相似文献

The amphiphilic π-shaped copolymers with narrow molecular weight distribution (Mw/Mn = 1.04-1.09) based on polystyrene (PSt) and poly(ethylene glycol) have been synthesized successfully. The reversible addition-fragmentation transfer (RAFT) polymerization of St in the presence of dibenzyl trithiocarbonate and N,N′-azobis(isobutyronitrile) (AIBN) yielded macro RAFT agent PSt-SC(S)S-PSt, subsequent reaction with excess maleic anhydride (MAh) at 80 °C in tetrahydrofuran afforded the PSt-MAh-SC(S)S-MAh-PSt. It was used as RAFT agent in the RAFT polymerization of St, and finally the amphiphilic π-shaped copolymers were obtained by the reaction of MAh with hydroxyl-terminated poly(ethylene glycol methyl ether) at 90 °C for 48 h. Their structures were confirmed by FT-IR and 1H NMR spectra, and their molecular weight and molecular weight distribution were measured by gel permeation chromatography.  相似文献

Monomethoxy poly(ethylene glycol)‐b‐poly(ε‐caprolactone)(MPEG‐b‐PCL) diblock copolymers were synthesized via a ring‐opening polymerization. The polymeric nanoparticles prepared by precipitation/solvent evaporation exhibit a core–shell structure, characterized by dynamic light scattering (DLS), nuclear magnetic resonance (NMR), and atomic force microscopy (AFM). The hydrolytic degradation of those nanoparticles was studied by DLS, NMR, and gel permeation chromatography (GPC). It was found that the molecular weight of PCL block in a copolymer significantly affects the stability of nanoparticles in aqueous solution and nanoparticles with shorter PCL block length degraded faster. The degradation behaviors could be divided into two stages with slow degradation at the interface region via swelling effect and fast degradation at inner core via caves and channels formed by cleavage of ester bonds. Biodegradable and nontoxic alternating multiblock copolymers based on poly (p-dioxanone) (PPDO) and poly (ethylene glycol) (PEG) were synthesized by the coupling reaction of two bifunctional prepolymers, a dihydroxyl-terminated PPDO and dicarboxylated PEG. The prepolymers and the resulting PPDO/PEG multiblock copolymers were characterized by various analytical techniques such as FT-IR, 1H NMR, GPC, DSC and TG. At high concentration levels above critical gelation concentration (CGC), the aqueous solution of copolymers formed a gel. Temperature-sensitive gel to sol transition behaviors were investigated by the test tube inverting method. Dynamic light scattering (DLS) was used to investigate the micelle of copolymers, whose association probably caused the gelation of the system. Therefore, this novel copolymer has a great potential in injectable drug-delivery system for long-term delivery of drugs.  相似文献

Star-shaped poly(ethylene glycol) (PEG)-poly(?-caprolactone) (?-PCL) block copolymers were synthesized via a ring-opening polymerization. Nanoparticles prepared by the precipitation/solvent evaporation technique exhibit a core-shell structure. The hydrolytic degradation of 3-arm PEG-PCL copolymeric nanoparticles was studied by dynamic light scattering (DLS), nuclear magnetic resonance (NMR) and gel permeation chromatography (GPC). It was found that copolymers with shorter PCL block length degraded faster. The sizes of nanoparticles fluctuated during the initial degradation period, and then increased slightly before finally dropping off. The degradation mainly occurred at CL-CL linkages firstly then at the EO-CL linkages. The CL/EO molar ratios and the molecular weights of copolymers decreased as degradation time and a zero-order degradation behavior was observed.  相似文献

Novel biodegradable poly(ester anhydride) block copolymers based on ε‐caprolactone (ε‐CL) and adipic anhydride (AA) were prepared by sequential polymerization. ε‐CL was first initiated by potassium poly(ethylene glycol)ate and polymerized into active chains (PCL‐O?K+), which were then used to initiate the ring‐opening polymerization of AA. The effects of the AA feed ratio, solvent polarity, monomer concentration, and temperature on sequential polymerization were investigated. The copolymers, obtained under different conditions, were characterized by Fourier transform infrared, 1H NMR, gel permeation chromatography (GPC), and differential scanning calorimetry (DSC). The GPC results showed that the weight‐average molecular weights of the block copolymers were approximately 6.0 × 104. Biodegradable, amphiphilic, diblock poly(ε‐caprolactone)‐block‐poly(ethylene glycol) (PCL‐b‐PEG), triblock poly(ε‐caprolactone)‐block‐poly(ethylene glycol)‐block‐poly(ε‐caprolactone) (PCL‐b‐PEG‐b‐PCL), and star shaped copolymers were synthesized by ring opening polymerization of ε‐caprolactone in the presence of poly(ethylene glycol) methyl ether or poly(ethylene glycol) or star poly(ethylene glycol) and potassium hexamethyldisilazide as a catalyst. Polymerizations were carried out in toluene at room temperature to yield monomodal polymers of controlled molecular weight. The chemical structure of the copolymers was investigated by 1H and 13C NMR. The formation of block copolymers was confirmed by 13C NMR and DSC investigations. The effects of copolymer composition and molecular structure on the physical properties were investigated by GPC and DSC. For the same PCL chain length, the materials obtained in the case of linear copolymers are viscous whereas in the case of star copolymer solid materials are obtained with low Tg and Tm temperatures. © 2007 Wiley Periodicals, Inc.
