The relationship between peptide structure and electron transfer dissociation (ETD) is important for structural analysis by mass spectrometry. In the present study, the formation, structure and reactivity of the reaction intermediate in the ETD process were examined using a quadrupole ion trap mass spectrometer equipped with an electrospray ionization source. ETD product ions of zwitterionic tryptophan (Trp) and Trp‐containing dipeptides (Trp‐Gly and Gly‐Trp) were detected without reionization using non‐covalent analyte complexes with Ca2+ and 18‐crown‐6 (18C6). In the collision‐induced dissociation, NH3 loss was the main dissociation pathway, and loss related to the dissociation of the carboxyl group was not observed. This indicated that Trp and its dipeptides on Ca2+(18C6) adopted a zwitterionic structure with an NH3+ group and bonded to Ca2+(18C6) through the COO? group. Hydrogen atom loss observed in the ETD spectra indicated that intermolecular electron transfer from a molecular anion to the NH3+ group formed a hypervalent ammonium radical, R‐NH3, as a reaction intermediate, which was unstable and dissociated rapidly through N–H bond cleavage. In addition, N–Cα bond cleavage forming the z1 ion was observed in the ETD spectra of Trp‐GlyCa2+(18C6) and Gly‐TrpCa2+(18C6). This dissociation was induced by transfer of a hydrogen atom in the cluster formed via an N–H bond cleavage of the hypervalent ammonium radical and was in competition with the hydrogen atom loss. The results showed that a hypervalent radical intermediate, forming a delocalized hydrogen atom, contributes to the backbone cleavages of peptides in ETD. Electron transfer and capture mass spectra of a series of doubly charged ions that were phosphorylated pentapeptides of a tryptic type (pS,A,A,A,R) showed conspicuous differences in dissociations of charge-reduced ions. Electron transfer from both gaseous cesium atoms at 100 keV kinetic energies and fluoranthene anion radicals in an ion trap resulted in the loss of a hydrogen atom, ammonia, and backbone cleavages forming complete series of sequence z ions. Elimination of phosphoric acid was negligible. In contrast, capture of low-energy electrons by doubly charged ions in a Penning ion trap induced loss of a hydrogen atom followed by elimination of phosphoric acid as the dominant dissociation channel. Backbone dissociations of charge-reduced ions also occurred but were accompanied by extensive fragmentation of the primary products. z-Ions that were terminated with a deaminated phosphoserine radical competitively eliminated phosphoric acid and H2PO4 radicals. A mechanism is proposed for this novel dissociation on the basis of a computational analysis of reaction pathways and transition states. Electronic structure theory calculations in combination with extensive molecular dynamics mapping of the potential energy surface provided structures for the precursor phosphopeptide dications. Electron attachment produces a multitude of low lying electronic states in charge-reduced ions that determine their reactivity in backbone dissociations and H- atom loss. The predominant loss of H atoms in ECD is explained by a distortion of the Rydberg orbital space by the strong dipolar field of the peptide dication framework. The dipolar field steers the incoming electron to preferentially attach to the positively charged arginine side chain to form guanidinium radicals and trigger their dissociations.  相似文献

We report non‐chiral amino acid residues cis‐ and trans‐1,4‐diaminocyclohexane‐1‐carboxylic acid (cyclo‐ornithine, cO) that exhibit unprecedented stereospecific control of backbone dissociations of singly charged peptide cations and hydrogen‐rich cation radicals produced by electron‐transfer dissociation. Upon collision‐induced dissociation (CID) in the slow heating regime, peptide cations containing trans‐cO residues undergo facile backbone cleavages of amide bonds C‐terminal to trans‐cO. By contrast, peptides with cis‐cO residues undergo dissociations at several amide bonds along the peptide ion backbone. Diastereoisomeric cO‐containing peptides thus provide remarkably distinct tandem mass spectra. The stereospecific effect in CID of the trans‐cO residue is explained by syn‐facially directed proton transfer from the 4‐ammonium group at cO to the C‐terminal amide followed by neighboring group participation in the cleavage of the CO―NH bond, analogous to the aspartic acid and ornithine effects. Backbone dissociations of diastereoisomeric cO‐containing peptide ions generate distinct [bn]+‐type fragment ions that were characterized by CID‐MS3 spectra. Stereospecific control is also reported for electron‐transfer dissociation of cis‐ and trans‐cO containing doubly charged peptide ions. The stereospecific effect upon electron transfer is related to the different conformations of doubly charged peptide ions that affect the electron attachment sites and ensuing N―Cα bond dissociations.  相似文献

Charge tags using basic auxiliary functional groups 6-aminoquinolinylcarboxamido, 4-aminopyrimidyl-1-methylcarboxamido, 2-aminobenzoimidazolyl-1-methylcarboxamido, and the fixed-charge 4-(dimethylamino)pyridyl-1-carboxamido moiety are evaluated as to their properties in electron transfer dissociation mass spectra of arginine C-terminated peptides. The neutral tags have proton affinities that are competitive with those of amino acid residues in peptides. Charge reduction by electron transfer from fluoranthene anion-radicals results in peptide backbone dissociations that improve sequence coverage by providing extensive series of N-terminal c-type fragments without impeding the formation of C-terminal z fragments. Comparison of ETD mass spectra of free and tagged peptides allows one to resolve ambiguities in fragment ion assignment through mass shifts of c ions. Simple chemical procedures are reported for N-terminal tagging of Arg-containing tryptic peptides.  相似文献

The dissociation chemistry of somatostatin‐14 was examined using various tandem mass spectrometry techniques including low‐energy beam‐type and ion trap collision‐induced dissociation (CID) of protonated and deprotonated forms of the peptide, CID of peptide‐gold complexes, and electron transfer dissociation (ETD) of cations. Most of the sequence of somatostatin‐14 is present within a loop defined by the disulfide linkage between Cys‐3 and Cys‐14. The generation of readily interpretable sequence‐related ions from within the loop requires the cleavage of at least one of the bonds of the disulfide linkage and the cleavage of one polypeptide backbone bond. CID of the protonated forms of somatostatin did not appear to give rise to an appreciable degree of dissociation of the disulfide linkage. Sequential fragmentation via multiple alternative pathways tended to generate very complex spectra. CID of the anions proceeded through CH2? S cleavages extensively but relatively few structurally diagnostic ions were generated. The incorporation of Au(I) into the molecule via ion/ion reactions followed by CID gave rise to many structurally relevant dissociation products, particularly for the [M+Au+H]2+ species. The products were generated by a combination of S? S bond cleavage and amide bond cleavage. ETD of the [M+3H]3+ ion generated rich sequence information, as did CID of the electron transfer products that did not fragment directly upon electron transfer. The electron transfer results suggest that both the S? S bond and an N? Cα bond can be cleaved following a single electron transfer reaction. Copyright © 2009 John Wiley & Sons, Ltd.  相似文献

A study has been made of the mass spectral fragmentation upon electron impact of aliphatic C2? C12 chloromethyl esters and all their 66 monochlorinated derivatives. The fragmentation pathways of the parent chloromethyl esters were elucidated with the aid of the 1st FFR metastable ions. A McLafferty rearrangement gives the base peak in the C6? C11 parent esters and in almost all the 4-chloro and ω-chloro isomers. The subsequent loss of HCl gives a very characteristic peak of the chloromethyl esters and their (3-ω)-chloro derivatives at m/z 72, [C3H4O2]+. The 2-chloro isomers have the corresponding chlorine-containing fragment ion at m/z 106/108. The mass spectra of 2-, 3-, 4-, 5- and ω-chloro isomers give the characteristic fragment ions, the mass spectra of the other isomers being very similar.  相似文献

Mass spectra of the three isomeric vinylimidazoles have been compared and the structures of the fragment ions [C3H4N2] and [C5H5N2]+ have been investigated by collisionally activated dissociation mass spectrometry. The greater part of the non-decomposing ions m/z 68 from 2-vinylimidazole and from 2-imidazolecarboxylic acid methyl ester, and a minor part of this ion formed from the free acid, all have the same structure: the imidazole ring system, with hydrogens at both nitrogen atoms but none at C(2). An analogous structure, with an ethyl group at C(2), is proposed for the m/z 93 ion from 2-vinylimidazole.  相似文献

Theoretical calculations have been carried out to investigate the possible dissociation channels of isoprene. We focus on the major fragment ions of C5H7+,C5H5+,C4H5+,C3H6+,C3H5+,C3H4+,C3H3+ and C2H3+, which were observed experimentally from the isoprene dissociative photoionization. The energy calculations were performed with the CBS-QB3 model. All the geometries and energies of the fragments, intermediates and transition states involved in the dissociations channels were determined. Finally, the mechanisms of the dissociation pathways were discussed on the comparison of theoretical and experimental results.  相似文献

The electrospray ionization collisionally activated dissociation (CAD) mass spectra of protonated 2,4,6‐tris(benzylamino)‐1,3,5‐triazine (1) and 2,4,6‐tris(benzyloxy)‐1,3,5‐triazine (6) show abundant product ion of m/z 181 (C14H13+). The likely structure for C14H13+ is α‐[2‐methylphenyl]benzyl cation, indicating that one of the benzyl groups must migrate to another prior to dissociation of the protonated molecule. The collision energy is high for the ‘N’ analog (1) but low for the ‘O’ analog (6) indicating that the fragmentation processes of 1 requires high energy. The other major fragmentations are [M + H‐toluene]+ and [M + H‐benzene]+ for compounds 1 and 6, respectively. The protonated 2,4,6‐tris(4‐methylbenzylamino)‐1,3,5‐triazine (4) exhibits competitive eliminations of p‐xylene and 3,6‐dimethylenecyclohexa‐1,4‐diene. Moreover, protonated 2,4,6‐tris(1‐phenylethylamino)‐1,3,5‐triazine (5) dissociates via three successive losses of styrene. Density functional theory (DFT) calculations indicate that an ion/neutral complex (INC) between benzyl cation and the rest of the molecule is unstable, but the protonated molecules of 1 and 6 rearrange to an intermediate by the migration of a benzyl group to the ring ‘N’. Subsequent shift of a second benzyl group generates an INC for the protonated molecule of 1 and its product ions can be explained from this intermediate. The shift of a second benzyl group to the ring carbon of the first benzyl group followed by an H‐shift from ring carbon to ‘O’ generates the key intermediate for the formation of the ion of m/z 181 from the protonated molecule of 6. The proposed mechanisms are supported by high resolution mass spectrometry data, deuterium‐labeling and CAD experiments combined with DFT calculations. The dissociation behavior of phosphorylated and sulfonated peptide anions was explored using metastable atom-activated dissociation mass spectrometry (MAD-MS) and collision-induced dissociation (CID). A beam of high kinetic energy helium (He) metastable atoms was exposed to isolated phosphorylated and sulfonated peptides in the 3– and 2– charge states. Unlike CID, where phosphate losses are dominant, the major dissociation channels observed using MAD were Cα – C peptide backbone cleavages and neutral losses of CO2, H2O, and [CO2 + H2O] from the charge reduced (oxidized) product ion, consistent with an electron detachment dissociation (EDD) mechanism such as Penning ionization. Regardless of charge state or modification, MAD provides ample backbone cleavages with little modification loss, which allows for unambiguous PTM site determination. The relative abundance of certain fragment ions in MAD is also demonstrated to be somewhat sensitive to the number and location of deprotonation sites, with backbone cleavage somewhat favored adjacent to deprotonated sites like aspartic acid residues. MAD provides a complementary dissociation technique to CID, ECD, ETD, and EDD for peptide sequencing and modification identification. MAD offers the unique ability to analyze highly acidic peptides that contain few to no basic amino acids in either negative or positive ion mode.  相似文献

The extent of isomerization of [C9H10] ions, with lifetimes of approximately 10?11 and 10?6 s has been investigated using field ionization, collisionally activated dissociation and charge stripping techniques. The [C9H10] ions which were investigated included the molecular ions of α-methylstyrene, β-methylstyrene, o-methylstyrene, m-methylstyrene, p-methylstyrene, indan, cyclopropylbenzene, allylbenzene and the product of water loss from 3-phenylpropanol. The field ionization spectra of all the C9H10 hydrocarbons were different indicating that isomerization to a common ion structure had not occurred to a measurable extent for ions with lifetimes of approximately 10?11 s. Collisionally activated dissociation and charge stripping results indicated that most of the [C9H10] ions continued to maintain unique ion structures (or mixtures of structures) at ion lifetimes of 10?6 s. Possible exceptions are the [C9H10] ions from allylbenzene and cyclopropylbenzene which gave indistinguishable collisionally activated dissociation and charge stripping spectra.  相似文献

Our previous study showed that selenamide reagents such as ebselen and N-(phenylseleno)phthalimide (NPSP) can be used for selective and rapid derivatization of protein/peptide thiols in high conversion yield. This paper reports the systematic investigation of MS/MS dissociation behaviors of selenamide-derivatized peptide ions upon collision induced dissociation (CID) and electron transfer dissociation (ETD). In the positive ion mode, derivatized peptide ions exhibit tag-dependent CID dissociation pathways. For instance, ebselen-derivatized peptide ions preferentially undergo Se–S bond cleavage upon CID to produce a characteristic fragment ion, the protonated ebselen (m/z 276), which allows selective identification of thiol peptides from protein digest as well as selective detection of thiol proteins from protein mixture using precursor ion scan (PIS). In contrast, NPSP-derivatized peptide ions retain their phenylselenenyl tags during CID, which is useful in sequencing peptides and locating cysteine residues. In the negative ion CID mode, both types of tags are preferentially lost via the Se–S cleavage, analogous to the S–S bond cleavage during CID of disulfide-containing peptide anions. In consideration of the convenience in preparing selenamide-derivatized peptides and the similarity of Se–S of the tag to the S–S bond, we also examined ETD of the derivatized peptide ions to probe the mechanism for electron-based ion dissociation. Interestingly, facile cleavage of Se–S bond occurs to the peptide ions carrying either protons or alkali metal ions, while backbone cleavage to form c/z ions is severely inhibited. These results are in agreement with the Utah-Washington mechanism proposed for depicting electron-based ion dissociation processes.  相似文献

Dissociative electron ionization (70eV) of selenophene (C4H4Se) generates m/z 106 ions of composition [H2, C2, 80Se]+? and m/z 105 ions of [H, C2, 80Se]+. From tandem mass spectrometric experiments, Density Functional Theory (DFT) and ab initio calculations, it is concluded that these ions have the structure of selenoketene H2C?C?Se+? (1a+? )and selenoketyl HC?C?Se+ (2a+) ions respectively. The calculations predict that selenoketene ion 1a+? is separated by high energy barriers from its isomers selenirene (H e)+? 1b+?, ethyne selenol (HCCSeH)+? 1c+?, (CCHSeH)+? 1d+? and (CCSeH2)+? 1e+?. The selenoketyl ion 2a+ is separated by high barriers from its isomers (CCHSe)+ 2b+, and (CCSeH)+ 2c+. Neutralization‐reionization mass spectra (NRMS) of these structurally characterized ions confirmed that the corresponding neutral analogues, selenoketene H2CCSe 1a and selenoketyl radical HCCSe 2a? are stable in the rarefied gas phase. The relative, dissociation, and isomerization energies for selenoketene and selenoketyl ions and neutrals studied at B3LYP/6–31G(d,p) and G2/G2(MP2) levels are used to support and interpret the experimental results. The gas phase heats of formation of several organosulfur cations were determined from thiirane, thietane and tetrahydrothiophene precursor molecules by photoionization mass spectrometry. Heats of formation at 0 K and 298 K are reported for the following ions: [H2CS], [H3CS]+, [C2H3S]+, [C2H4S], [C3H5S]+, [C3H6S], [C4H7S]+ and [C4H8S]. The [C4H7S]+ (m/z 87), [C2H4S] (m/z 60), [C2H3S]+ (m/z 59), [C4H7]+ (m/z 55), [C4H6] (m/z 54) and [CH2S] (m/z 46) ions are produced from metastable tetrahydrothiophene ions at photon energies between 10.2 and 10.7 eV. Decay rates of internal energy selected parent ions to the m/z 60, 59, 55 and 54 fragments were measured by threshold photoelectron-photoion coincidence, the results of which are compared to statistical theory (RRKM/QET) calculations. The [C2H4S] ion from tetrahydrothiophene is found to have the thioacetaldehyde structure. From the measured [C2H4S] onset a ΔH = 50±8 kJ mol?1 was calculated for the thioacetaldehyde molecule.  相似文献

Some newly synthesized 10B nido‐carborane derivatives, i.e., 7,8‐dicarba‐nido‐undecaborane monoanions ([7‐Me‐8‐R‐C2B9H10]K+, R = H, butyl, hexyl, octyl and decyl), have been fully characterised and examined by electrospray ionization and Fourier transform ion cyclotron resonance mass spectrometry with liquid chromatographic separation (LC/ESI‐FTICR‐MS). These boron‐containing compounds exhibit abundant molecular ions ([M]?) at m/z 140.22631 [CB9H14]?, m/z 196.28883 [CB9H22]?, m/z 224.32032 [CB9H26]?, m/z 252.35133 [CB9H30]? and m/z 280.38354 [CB9H34]? at the normal tube lens voltage setting of ?90 V, which was an instrumental parameter value selected in the tuning operation. Additional [M–nH2]? (n = 1?4) ions were observed in the mass spectra when higher tube lens voltages were applied, i.e., ?140 V. High‐resolution FTICR‐MS data revealed the accurate masses of fragment ions, bearing either an even or an odd number of electrons. Collision‐induced dissociation of the [M–nH2]? ions (n = 0–4) in the quadrupole linear ion trap (LTQ) analyzer confirmed the loss of hydrogen molecules from the molecular ions. It is suggested that the loss of H2 molecules from the alkyl chain is a consequence of the stabilization effect of the nido‐carborane charged polyhedral skeleton. The unimolecular dissociations of C5 epoxides ions mono- or disubstituted at C1 give exclusive loss of CH3 and exclusive formation of methoxy vinyl carbenium ion, both in the source and in the 2nd field-free region. In the case of the 1,2-disubstituted ion in the 2nd field-free region the loss of ethene is the only pathway, while a competition occurs for the trisubstituted ion leading to [C3H6O]+˙ and [C4H7O]+˙ ions, the structure of which are demonstrated. The first step of the different mechanisms is the cleavage of the heterocyclic C? C bond.  相似文献

The potential energy profiles for the mutual conversion of the isomeric molecular ions [C5H6O]+? of 2‐methylfuran, 3‐methylfuran and 4H‐pyran and the fragmentations that lead to [C5H5O]+ ions were obtained from calculations at the B3LYP/6‐311G + + (3df,3pd)//B3LYP/6‐31G(d,p) level of theory. The various competing unimolecular processes were characterized by their RRKM microcanonical rate coefficients, k(E), using the sets of reactant and transition state frequencies and the kinetic barriers obtained from the density functional method. In either a high‐ or a low‐energy regime, the pyrylium ion [C5H5O]+ is generated directly from the 4H‐pyran molecular ion by a simple cleavage. In contrast, in the metastable kinetic window, the molecular ions of methylfurans irreversibly isomerize to a mixture of interconverting structures before dissociation, which includes the 2H‐ and 3H‐pyran ions. The hydrogen atoms attached to saturated carbons of the pyran rings are very stabilizing at the position 2, but they are very labile at position 3 and can be shifted to adjacent positions. Once 4H‐pyran ion has been formed, the C? H bond cleavage begins before any hydrogen shift occurs. According to our calculation, there would not be complete H scrambling preceding the dissociation of the molecular ions [C5H6O]+?. On the other hand, as the internal energy of the 2‐methylfuran molecular ion increases, H? loss can become more important. These results agree with the available experimental data. Using mass spectrometric technique, the effect of geometrical isomerism on the first and higher appearance energy values for C4H3 +, C4H7 + and C3H,3 + ions obtained from cis-2-butene andtrans-2-butene is reported. The structure in the ionization efficiency curves (studied for 9 eV above threshold) for the same ions obtained from the two isomers is reported and compared. It is believed that at threshold C4H7 + fragment is formed from the two isomers as methallyl ion. For C3H3 + fragment formed from the cw-isomer at threshold the proposed structure is the propargyl ion with ΔHf equal to 279-4 kcal/mole while for that ion obtained fromtransisomer the proposed structure is the allenyl ion with ΔHf equal to 296.6 kcal/mole.  相似文献

Upon collision‐induced activation, gaseous sodium adducts generated by electrospray ionization of disodium salts of 1,2‐ 1,3‐, and 1,4‐benzene dicarboxylic acids (m/z 233) undergo an unprecedented expulsion of CO2 by a rearrangement process to produce an ion of m/z 189 in which all three sodium atoms are retained. When isolated in a collision cell of a tandem‐in‐space mass spectrometer, and subjected to collision‐induced dissociation (CID), only the m/z 189 ions derived from the meta and para isomers underwent a further CO2 loss to produce a peak at m/z 145 for a sodiated arene of formula (Na3C6H4)+. This previously unreported m/z 145 ion, which is useful to differentiate meta and para benzene dicarboxylates from their ortho isomer, is in fact the sodium adduct of phenelenedisodium. Moreover, the m/z 189 ion from all three isomers readily expelled a sodium radical to produce a peak at m/z 166 for a radical cation [(?C6H4CO2Na2)+], which then eliminated CO2 to produce a peak at m/z 122 for the distonic cation (?C6H4Na2)+.
