Comb‐like amphiphilic poly(poly((lactic acid‐co‐glycolic acid)‐block‐poly(ethylene glycol)) methacrylate (poly((PLGA‐b‐PEG)MA)) copolymers were synthesized by radical polymerization. (PLGA‐b‐PEG)MA macromonomer was prepared by ring‐opening bulk polymerization of DL ‐lactide and glycolide using purified poly(ethylene glycol) monomethacrylate (PEGMA) as an initiator. (PLGA‐b‐PEG)MA macromonomer was copolymerized with PEGMA and/or acrylic acid (AA) by radical polymerization to produce comb‐like amphiphilic block copolymers. The molecular weight and chemical structure were investigated by GPC and 1H NMR. Poly((PLGA‐b‐PEG)MA) copolymer aqueous solutions showed gel–sol transition behavior with increasing temperature, and gel‐to‐sol transition temperature decreased as the compositions of the hydrophilic PEGMA and AA increased. The gel‐to‐sol transition temperature of the terpolymers of the poly((PLGA‐b‐PEG)MA‐co‐PEGMA‐co‐AA) also decreased when the pH was increased. The effective micelle diameter obtained from dynamic light scattering increased with increasing temperature and with increasing pH. The critical micelle concentration increased as the composition of the hydrophilic monomer component, PEGMA and AA, were increased. The spherical shape of the hyperbranched polymers in aqueous environment was observed by atomic force microscopy. © 2008 Wiley Periodicals, Inc. A novel visible light responsive random copolymer consisting of hydrophobic azobenzene‐containing acrylate units and hydrophilic acrylic acid units has been prepared. The azobenzene molecule bearing methoxy groups at all four ortho positions is readily synthesized by one‐step conversion of diazotization. The as‐prepared polymer can self‐assemble into nanoparticles in water due to its amphiphilic nature. The tetra‐o‐methoxy‐substituted azobenzene‐functionalized polymer can exhibit the trans‐to‐cis photoswitching under the irradiation with green light of 520 nm and the cis‐to‐trans photoswitching under the irradiation with blue light of 420 nm in both solution and aggregate state. The morphologies of the self‐assembled nanoparticles are revealed by TEM and DLS. The controlled release of loaded molecules from the nanoparticles can be realized by adjusting pH value since the copolymer possesses pH responsive acrylic acid groups. The fluorescence of loaded Nile Red in the nanoparticles can be tuned upon the visible light irradiation. The reversible photoswitching of the azobenzene‐functionalized polymer under visible light may endow the polymer with wide applications without using ultraviolet light at all. © 2015 Wiley Periodicals, Inc. This review summarizes recent advances in the design and synthesis of amino‐acid‐based block copolymers by reversible addition–fragmentation chain transfer (RAFT) polymerization of amino‐acid‐bearing monomers. We will mainly focus on stimuli‐responsive block copolymers, such as pH‐, thermo‐, and dual‐stimuli‐responsive block copolymers, and self‐assembled block copolymers, including amphiphilic and double‐hydrophilic block copolymers having tunable chiroptical properties. We will also highlight recent results in RAFT synthesis of amino‐acid‐based copolymers having various properties, such as catalytic and optoelectronic properties, cross‐linked block copolymer micelles, unimolecular micelles, and organic–inorganic hybrids.  相似文献

We report on the preparation of reduction‐responsive amphiphilic block copolymers containing pendent p‐nitrobenzyl carbamate (pNBC)‐caged primary amine moieties by reversible addition–fragmentation chain transfer (RAFT) radical polymerization using a poly(ethylene glycol)‐based macro‐RAFT agent. The block copolymers self‐assembled to form micelles or vesicles in water, depending on the length of hydrophobic block. Triggered by a chemical reductant, sodium dithionite, the pNBC moieties decomposed through a cascade 1,6‐elimination and decarboxylation reactions to liberate primary amine groups of the linkages, resulting in the disruption of the assemblies. The reduction sensitivity of assemblies was affected by the length of hydrophobic block and the structure of amino acid‐derived linkers. Using hydrophobic dye Nile red (NR) as a model drug, the polymeric assemblies were used as nanocarriers to evaluate the potential for drug delivery. The NR‐loaded nanoparticles demonstrated a reduction‐triggered release profile. Moreover, the liberation of amine groups converted the reduction‐responsive polymer into a pH‐sensitive polymer with which an accelerated release of NR was observed by simultaneous application of reduction and pH triggers. Multistimuli‐responsive precise morphological control over self‐assembled polymers is of great importance for applications in nanoscience as drug delivery system. A novel pH, photoresponsive, and cyclodextrin‐responsive block copolymer were developed to investigate the reversible morphological transition from micelles to vesicles. The azobenzene‐containing block copolymer poly(ethylene oxide)‐b‐poly(2‐(diethylamino)ethyl methacrylate‐co‐6‐(4‐phenylazo phenoxy)hexyl methacrylate) [PEO‐b‐P(DEAEMA‐co‐PPHMA)] was synthesized by atom transfer radical polymerization. This system can self‐assemble into vesicles in aqueous solution at pH 8. On adjusting the solution pH to 3, there was a transition from vesicles to micelles. The same behavior, that is, transition from vesicles to micelles was also realizable on addition of β‐cyclodextrin (β‐CD) to the PEO‐b‐P(DEAEMA‐co‐PPHMA) solution at pH 8. Furthermore, after β‐CD was added, alternating irradiation of the solution with UV and visible light can also induce the reversible micelle‐to‐vesicle transition because of the photoinduced trans‐to‐cis isomerization of azobenzene units. A novel amphiphilic copolymer was synthesized from poly (ethylene glycol) methyl ether methacrylate (PEGMA950), methyl methacrylate (MMA) and acryloyl‐β‐cyclodextrin (acryloyl‐β‐CD) using the composites of (NH4)2S2O8/NaHSO3 as the oxidation–reduction initiators. The successful fabrication of poly(PEGMA‐co‐MMA‐co‐acryloyl‐β‐CD) copolymers was confirmed by Fourier transform infrared spectrometer (FTIR), 1H‐nuclear magnetic resonance (1H NMR) spectra. The amphiphilic copolymer could self‐assemble into nanoparticles (NPs), and their morphology and particle size distribution were characterized with transmission electron microscopy (TEM), atomic force microscope (AFM) and dynamic light scattering (DLS) methods. Ibuprofen (IBU) was encapsulated in the novel NPs, and the release profiles of IBU were investigated. FTIR and 1H NMR spectra illustrated that the poly(PEGMA‐co‐MMA‐co‐acryloyl‐β‐CD) copolymers were synthesized without any residual monomers and initiators. TEM and AFM photographs suggested that the obtained NPs were spherical, and the DLS results indicated that the diameter of blank NPs was 157.3 ± 32.7 nm. Ethyl cellulose graft poly(poly(ethylene glycol) methyl ether methacrylate) (EC‐g‐P(PEGMA)) amphiphilic copolymers were synthesized via atom transfer radical polymerization (ATRP) and characterized by FTIR, 1H NMR, and gel permeation chromatography. Reaction kinetics analysis indicated that the graft copolymerization is living and controllable. The self‐assembly and thermosensitive property of the obtained EC‐g‐P(PEGMA) amphiphilic copolymers in water were investigated by dynamic light scattering, transmission electron microscopy, and transmittance. It was found that the EC‐g‐P(PEGMA) amphiphilic copolymers can self‐assemble into spherical micelles in water. The size of the micelles increases with the increase of the side chain length. The spherical micelles show thermosensitive properties with a lower critical solution temperature around 65 °C, which almost independent on the graft density and the length of the side chains. The obtained EC‐g‐P(PEGMA) graft copolymers have both the unique properties of poly(ethylene glycol) and cellulose, which may have the potential applications in biomedicine and biotechnology. © 2008 Wiley Periodicals, Inc. Redox‐responsive core cross‐linked (CCL) micelles of poly(ethylene oxide)‐b‐poly(furfuryl methacrylate) (PEO‐b‐PFMA) block copolymers were prepared by the Diels‐Alder click‐type reaction. First, the PEO‐b‐PFMA amphiphilic block copolymer was synthesized by the reversible addition‐fragmentation chain transfer polymerization. The hydrophobic blocks of PFMA were employed to encapsulate the doxorubicin (DOX) drug, and they were cross‐linked using dithiobismaleimidoethane at 60 °C without any catalyst. Under physiological circumstance, the CCL micelles demonstrated the enhanced structural stability of the micelles, whereas dissociation of the micelles took place rapidly through the breaking of disulfide bonds in the cross‐linking linkages under reduction environment. The core‐cross‐linked micelles showed fine spherical distribution with hydrodynamic diameter of 68 ± 2.9  nm. The in vitro drug release profiles presented a slight release of DOX at pH 7.4, while a significant release of DOX was observed at pH 5.0 in the presence of 1,4‐dithiothreitol. MTT assays demonstrated that the block copolymer did not have any practically cytotoxicity against the normal HEK293 cell line while DOX‐loaded CCL micelles exhibited a high antitumor activity towards HepG2 cells. © 2016 Wiley Periodicals, Inc. Photo‐responsive block copolymer mPEG‐b‐poly(Tyr)‐g‐NB was prepared by introduction of o‐nitrobenzyl ester group into the side chain of amphiphilic poly(ethylene glycol)‐b‐poly(α‐hydroxy acids) (mPEG‐b‐poly(Tyr)) containing pendent alkynyl group via copper‐catalyzed azide‐alkyne cycloaddition reaction. The amphiphilic mPEG‐b‐poly(Tyr) was synthesized via the ring‐opening polymerization of O‐carboxyanhydrides, with monomethoxy poly(ethylene glycol) (mPEG) as macroinitiator. The molecular structure, self‐assembly, and photo‐controlled release of the obtained mPEG‐b‐poly(Tyr)‐g‐NB were thoroughly investigated. mPEG‐b‐poly(Tyr)‐g‐NB could self‐assemble into spherical micelles in water and showed disassembly under UV light irradiation, which was demonstrated by means of UV‐vis spectroscopy, scan electron microscopes, and dynamic light scattering measurement. Fluorescence emission measurements demonstrated that Nile red, encapsulated by micelles, can be released upon UV irradiation. Self‐assembled thermo‐ and pH‐responsive poly(acrylic acid)‐b‐poly(N‐isopropylacrylamide) (PAA‐b‐PNIPAM) micelles for entrapment and release of doxorubicin (DOX) was described. Block copolymer PAA‐b‐PNIPAM associated into core‐shell micelles in aqueous solution with collapsed PNIPAM block or protonated PAA block as the core on changing temperature or pH. Complexation of DOX with PAA‐b‐PNIPAM triggered by the electrostatic interaction and release of DOX from the complexes due to the changing of pH or temperature were studied. Complex micelles incorporated with DOX exhibited pH‐responsive and thermoresponsive drug release profile. The release of DOX from micelles was suppressed at pH 7.2 and accelerated at pH 4.0 due to the protonation of carboxyl groups. Furthermore, the cumulative release of DOX from complex micelles was enhanced around LCST ascribed to the structure deformation of the micelles. © 2008 Wiley Periodicals, Inc. A double‐responsive amphiphilic random copolymer (P(OEtOxA)‐ran‐PNBA) composed of thermoresponsive poly(oligo(2‐ethyl‐2‐oxazoline)acrylate) (P(OEtOxA)) segments and photocleavable poly(2‐nitrobenzyl acrylate) (PNBA) segments is synthesized via combination of cationic ring‐opening polymerization (CROP) and reversible addition‐fragmentation chain transfer (RAFT) polymerization techniques. The P(OEtOxA)‐ran‐PNBA copolymer exhibits lower critical solution (LCST)‐type soluble‐to‐turbid phase transition in water with tunable cloud point (Tcp) with respect to chain length of P(OEtOxA) segment present. The photocleavage of PNBA segments by UV irradiation transforms amphiphilic P(OEtOxA)‐ran‐PNBA to fully hydrophilic P(OEtOxA)‐ran‐poly(acrylic acid) resulting in the appreciable increase of Tcp of copolymer in aqueous solution. Owing to the amphiphilic nature, the P(OEtOxA)‐ran‐PNBA copolymer molecules self‐assemble into well‐dispersed spherical micelles in water. There is a disruption of the copolymer micelles with UV light irradiation as well as shrinkage of micellar size with increasing temperature above the LCST of copolymer in solution. Finally, the encapsulation of hydrophobic guest molecule (nile red) into P(OEtOxA)‐ran‐PNBA copolymer micelles and thermo‐ and photo‐triggered release of nile red are demonstrated. © 2017 Wiley Periodicals, Inc. In this article, a light and pH dual‐sensitive block copolymer PEG‐b‐poly(MPC‐Azo/DEA) was facilely prepared for the first time by azide‐alkyne click chemistry between amphiphilic block copolymer bearing pendant alkynyl group poly(ethylene glycol)‐poly(5‐methyl‐5‐propargylxycarbonyl‐1,3‐dioxane‐2‐one) (PEG‐b‐poly(MPC)) and two azide‐containing compounds azobenzene derivative (Azo‐N3) and 2‐azido‐1‐ethyl‐diethylamine (DEA‐N3). Light response of the polymeric nanoparticles benefits from the azobenzene segments and pH responsiveness is attributed to DEA moieties. The prepared copolymer could self‐assemble into spherical micelle particles. The morphological changes of these particles in response to dual stimuli were investigated by UV/vis spectroscopy, dynamic light scattering (DLS), and transmission electron microscopy (TEM). Nile Red (NR) was utilized as probe, and fluorescence spectroscopy was served as an evidence for the enhanced release of cargos from polymeric nanoparticles under combined stimulation. Anticancer drug, DOX was loaded into the nanoparticles and the loaded‐DOX could be released from these nanoparticles under dual stimuli. MTT assays further demonstrated that PEG‐b‐poly(MPC) and PEG‐b‐poly(MPC‐Azo/DEA) were of biocompatibility and low toxicity against HepG2 cells as well as SMCC‐7721 cells. More importantly, the prepared DOX‐loaded nanoparticles exhibited good anticancer ability for the two cells. The self‐assembly behavior of poly(isoprene‐b‐acrylic acid) and poly(styrene‐b‐2‐vinylpyridine) amphiphilic block copolymers, as well as a poly(styrene‐r‐2‐vinylpyridine) amphiphilic random copolymer was investigated in slightly selective organic solvents (tetrahydrofuran and toluene) in the presence of Ag and Au ions and subsequently Ag, Au metal nanoparticles, by means of dynamic light scattering. In the range of concentrations studied the copolymers exist in the form of micelles with cores composed of acrylic acid and 2‐vinylpyridine segments in equilibrium with unimers. The addition of metal ions and their subsequent transformation to metal nanoparticles shifts the equilibrium in favor of the micelles. The concentration of the inorganic components has also a considerable effect on the size of the polymeric aggregates. A similar behavior is observed for the random copolymer. Attenuated total reflectance Fourier transform infrared spectroscopy (ATR‐FTIR), UV‐visible spectroscopy, and transmission electron microscopy (TEM) give valuable additional information on the nature of the interactions between the polymeric and inorganic components, as well as on the characteristics of the metal nanoparticles and the hybrid micelles formed in each case. The presented results have a direct relation to the synthesis of metal nanoparticles under confinement by utilization of copolymer nanoreactors and appropriate solution conditions. © 2008 Wiley Periodicals, Inc. A series of amphiphilic temperature‐responsive star‐shaped poly(D,L‐lactic‐co‐glycolic acid)‐b‐methoxy poly(ethylene glycol) (PLGA‐mPEG) block copolymers with different arm numbers were synthesized via the arm‐first method. Gel permeation chromatography data confirmed that star‐shaped PLGA‐mPEG copolymers had narrow polydispersity index, indicating the successful formation of star‐shaped block copolymers. Indirectly, the 1H NMR spectra in two kinds of solvents and dye solubilization method had confirmed the formation of core‐shell micelles. Further, core‐shell micelles with sizes of about 30–50 nm were directly observed by transmission electron microscopy. Subsequently, the micellar sizes and distributions as a function of concentrations and temperature were measured. At various copolymer concentrations, individual micelles with size of 20–40 nm and grouped micelles with size of 600–700 nm were found. Micellar mechanism of star‐shaped block copolymers in aqueous solution was simultaneously discussed. In addition, sol–gel transition of star‐shaped block copolymers in water was also investigated via the inverting test method. The critical gel temperature (CGT) and critical gel concentration (CGC) values of two‐arm, three‐arm and four‐arm copolymer solutions were markedly higher than ones of one‐arm copolymer. Moreover, the same CGC values of copolymer solution with different molecular weight and the same arm composition were ~15 wt %, and CGT values increased from ~38 to ~47°C with increasing arm numbers. A series of methoxypolyethylene glycol‐terminated self‐fluorescent polyurethane multi‐block copolymers with excellent pH‐responsivity, self‐fluorescence, and biocompatibility are designed and synthesized. In our design, 1, 4‐bis (hydroxyethyl) piperazine is chosen as a pH‐responsive segment which can donate or accept protons in response to the change of environmental pH, and fluorescein isothiocyanate is used as a fluorescent dye conjugated into the micelles to offer self‐fluorescence. The chemical structure of the polyurethane multi‐block copolymers is characterized by nuclear magnetic resonance and Fourier transform infrared spectroscopy. The results of the acid‐based titration, the fluorescence spectrometry, and the ultraviolet visible spectroscopy indicate that the polyurethane multi‐block copolymers own an excellent pH‐buffering capacity responded to the change of pH values and the favorable self‐fluorescence property in an aqueous solution. And the ultraviolet absorption peaks of samples are strengthened with increasing of pH values, indicating that methoxypolyethylene glycol‐terminated self‐fluorescent polyurethane multi‐block copolymer can be a pH‐dependent fluorescent probe in a broad pH range. In addition, the in vitro cytotoxicity test showed that the polyurethane multi‐block copolymer has low cytotoxicity and good biocompatibility, which make it a promising nanoplatform for molecular imaging, diagnosis, and treatment of disease.  相似文献

Photoresponsive amphiphilic diblock poly(carbonate)s mPEG113‐b‐PMNCn with pendent o‐nitrobenzyl ester group were synthesized through ring‐opening polymerization (ROP) using 1,8‐diazabi‐cyclo[5.4.0]undec‐7‐ene (DBU) as catalyst and monomethoxy poly(ethylene glycol) (mPEG) as macroinitiator. In aqueous solution, the copolymers can self‐assemble to spherical micelles with a PC core and a PEG shell. The critical micelle concentration (CMC), size, and morphology of the micelles were demonstrated by means of fluorescence spectroscopy, transmission electron microscopes (TEM), and dynamic light scattering (DLS). Under UV light irradiation, the amphiphilic copolymer micelles disassembled because of the photocleavage of o‐NB ester, and the light‐controlled release behaviors of payload Nile red were further proved. Summary: Diblock terpolymers that consist of homopolymer and statistical copolymer (polyampholyte) building blocks are synthesized by group transfer polymerization. Two types of block tepolymers are explored in aqueous media: the amphiphilic poly{[(diethylamino)ethyl methacrylate]‐co‐(methacrylic acid)}‐block‐poly(methyl methacrylate) and the double hydrophilic poly[oligo(ethylene glycol) methacrylate]‐block‐poly{[(diethylamino)ethyl methacrylate]‐co‐(methacrylic acid)}. The first terpolymer self‐assembles in aqueous media to form responsive micelles that change their corona charge sign upon switching pH. The second terpolymer exhibits a multi‐responsive behavior. It forms neutral, positive, or negative micelles depending on a combination of different environmental conditions such as temperature, pH, and ionic strength.

Glucose responsive block copolymer featuring boronic acid as a glucose responsive moiety and glycine are reported. The first block is polymerized through reversible addition–fragmentation chain transfer (RAFT) polymerization and the resulting poly(N‐acryloylmorpholine)113 (PAcM) is employed as a macro‐chain transfer agent for chain extension with pentafluorophenyl acrylate (PFPA) yielding a well‐defined PAcM113block‐poly(pentafluorophenyl acrylate)84 (PPFPA). The PPFPA block is then reacted with functional (3‐aminomethyl) phenyl boronic acid and glycine via post‐polymerization modification and the structure of the block copolymer is confirmed by proton nuclear magnetic resonance (NMR), 19F NMR, Fourier transform infrared, and gel permeation chromatography. By copolymerizing glycine into the polymer backbone, the relative pKa of the block copolymer is significantly lowered. The block copolymer can self‐assemble into core–shell micelles in aqueous solution and disassemble in response to glucose at the physiological pH. Furthermore, the encapsulation and release of Nile red (NR) as a hydrophobic model drug is studied under the physiological pH. The influence of the glucose concentration on the NR release from the polymeric micelles is demonstrated. These results suggested that the glucose‐responsive poly[(AcM)113b‐(3‐(aminomethyl)phenylboronic acid hydrochloride(‐co‐Gly)84] block copolymer has potential applications as a glucose‐responsive polymer for insulin delivery. © 2018 Wiley Periodicals, Inc. Well‐defined amphiphilic block copolymers were prepared by ring opening metathesis polymerization and their stimuli responsive behavior of formed micelles in aqueous solution was investigated. The hydrophobic core of the micelles consists of either a poly[5,6‐bis(ethoxymethyl)bicyclo[2.2.1]hept‐2‐ene]‐block with a glass transition Tg at room temperature or a poly[endo,exo[2.2.1]bicyclohept‐5‐ene‐2,3‐diylbis (phenylmethanone)] with a Tg of 143 °C. For the polyelectrolyte shell, the precursor block poly[endo,exo[2.2.1]bicyclohept‐5‐ene‐2,3‐dicarboxyclic tert‐butylester] was transformed into the free acidic block by cleavage of the tert‐butyl groups using trifluoroacetic acid. Micellar solutions were prepared by dialysis, dissolving the copolymers in dimethyl sulfoxide which was subsequently replaced by water. All polymers form micelles with radii between 10 and 20 nm at a pH‐value below 5, where the carboxylic acid groups are in the protonated state. The block copolymer micelles show a strong increase of the hydrodynamic radius with increasing pH‐value, due to the repulsion among the formed carboxylate anions resulting in a stretching of the polymer chains. Self‐immolative polymers (SIPs) undergo depolymerization in response to the cleavage of stimuli‐responsive end‐caps from their termini. Some classes of SIPs, including polycarbamates, have depolymerization rates that depend on environmental factors such as solvent and pH. In previous work, hydrophobic SIPs have been incorporated into amphiphilic block copolymers and used to prepare nanoassemblies. However, stimuli‐responsive hydrophilic blocks have not previously been incorporated. In this work, we synthesized amphiphilic copolymers composed of a hydrophobic polycarbamate SIP block and a hydrophilic poly(2‐(dimethylamino)ethyl methacrylate) (PDMAEMA) block connected by a UV light‐responsive linker end‐cap. It was hypothesized that after assembly of the block copolymers into nanoparticles, chain collapse of the PDMAEMA above its lower critical solution temperature (LCST) might change the environment of the SIP block, thereby altering its depolymerization rate. Self‐assembly of the block copolymers was performed, and the depolymerization of the resulting assemblies was studied by fluorescence spectroscopy, dynamic light scattering, and NMR spectroscopy. At 20 °C, the system exhibited a selective response to the UV light. At 65 °C, above the LCST of PDMAEMA, the systems underwent more rapid depolymerization, suggesting that the increase in rate arising from the higher temperature dominated over environmental effects arising from chain collapse. © 2018 Wiley Periodicals, Inc.
