A room temperature ionic liquid (RTIL) modified carbon paste electrode was constructed based on the substitute of paraffin with 1-butyl-3-methyl-imidazolium hexafluorophosphate (BMIMPF6) as binder for carbon paste. Direct electrochemistry and electrocatalytic behaviors of hemoglobin (Hb) entrapped in the sodium alginate (SA) hydrogel film on the surface of this carbon ionic liquid electrode (CILE) were investigated. The presence of IL in the CILE increased the electron transfer rate and provided a biocompatible interface. Hb remained its bioactivity on the surface of CILE and the SA/Hb modified electrode showed a pair of well-defined, quasi-reversible cyclic voltammetric peaks with the apparent standard potential (E0′) at about −0.344 V (vs. SCE) in pH 7.0 Britton–Robinson (B–R) buffer solution, which was attributed to the Hb Fe(III)/Fe(II) redox couple. UV–Vis absorption spectra indicated that heme microenvironment of Hb in SA film was similar to its native status. Hb showed a thin-layer electrochemical behavior in the SA film with the direct electron transfer achieved on CILE without the help of electron mediator. Electrochemical investigation indicated that Hb took place one proton with one electron electrode process and the average surface coverage of Hb in the SA film was 3.2 × 10−10 mol/cm2. The immobilized Hb showed excellent electrocatalytic responses to the reduction of H2O2 and nitrite.  相似文献

Direct electrochemical response of Myoglobin (Myb) at the basal plane graphite (BPG) electrode was observed when a room temperature ionic liquid (RTIL), 1-(2-hydroxyethyl)-3-methyl imidazolium tetrafluoroborate ([HEMIm][BF4]), was used as the supporting electrolyte. In a 0.17 M [HEMIm]BF4 aqueous solution, a couple of well-defined redox peaks of Myb could be obtained, whose anodic and cathodic peak potentials were at −0.158 and −0.224 V (vs. Ag/AgCl), respectively. Both anodic and cathodic peak currents increased linearly with the potential scan rate. Compared with the supporting electrolyte of phosphate buffer, [HEMIm][BF4] played an obvious promotion for the direct electron transfer between Myb and the BPG electrode. Further investigation suggested that Myb was adsorbed tightly on the surface of the BPG electrode in the presence of [HEMIm][BF4] to form a stable, approximate monolayer Myb film. Myb adsorbed on the BPG electrode surface showed a remarkable electrocatalytic activity for the reduction of oxygen in a [HEMIm][BF4] aqueous solution. Based on these, a third-generation biosensor could be constructed to directly detect the concentration of oxygen in aqueous solution with a limit of detection of 2.3 × 10−8 M.  相似文献

A robust and effective composite film based on gold nanoparticles (GNPs)/room temperature ionic liquid (RTIL)/multi-wall carbon nanotubes (MWNTs) modified glassy carbon (GC) electrode was prepared by a layer-by-layer self-assembly technique. Cytochrome c (Cyt c) was successfully immobilized on the RTIL-nanohybrid film modified GC electrode by electrostatic adsorption. Direct electrochemistry and electrocatalysis of Cyt c were investigated. The results suggested that Cyt c could be tightly adsorbed on the modified electrode. A pair of well-defined quasi-reversible redox peaks of Cyt c was obtained in 0.10 M, pH 7.0 phosphate buffer solution (PBS). RTIL-nanohybrid film showed an obvious promotion for the direct electron transfer between Cyt c and the underlying electrode. The immobilized Cyt c exhibited an excellent electrocatalytic activity towards the reduction of H2O2. The catalysis currents increased linearly to the H2O2 concentration in a wide range of 5.0 × 10−5– 1.15 × 10−3 M. Based on the multilayer film, the third-generation biosensor could be constructed for the determination of H2O2.  相似文献

A protein-based electrochemical sensor for hydrogen peroxide (H2O2) was developed by an easy and effective film fabrication method where spinach ferredoxin (Fdx) containing [2Fe–2S] metal center was cross linked with 11-mercaptoundecanoic acid (MUA) on a gold (Au) surface. The surface morphology of Fdx molecules on Au electrodes was investigated by atomic force microscopy (AFM). Electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV) were employed to study the electrochemical behavior of adsorbed Fdx on Au. The interfacial properties of the modified electrode were evaluated in the presence of Fe(CN)63?/4? redox couple as a probe. From CV, a pair of well-defined and quasi-reversible redox peaks of Fdx was obtained in 10 mM, pH 7.0 Tris–HCl buffer solution at ?170 and ?120 mV respectively. One electron reduction of the [2Fe-2S]2+ cluster occurs at one of the iron atoms to give the reduced [2Fe-2S]+. The formal reduction potential of Fdx ca. ?150 mV (vs. Ag/AgCl electrode) at pH 7.0. The electron-transfer rate constant, ks, for electron transfer between the Au electrode and Fdx was estimated to be 0.12 s?1. From the electrochemical experiments, it is observed that Fdx/MUA/Au promoted direct electron transfer between Fdx and electrode and it catalyzes the reduction of H2O2. The Fdx/MUA/Au electrode displays a linear increase in amperometric current for increasing concentration of H2O2.The sensor calibration plot was linear with r2 = 0.998 with sensitivity approximately 68.24 μAm M?1 cm?2. Further, the effect of nitrite on the developed sensor was examined which does not interfere with the detection of H2O2. Finally, the addition of H2O2 on MUA/Au electrode was observed which has no effect on amperometric current.  相似文献

Direct electrochemistry of hemoglobin (Hb) has been achieved by its direct immobilization on carbon ionic liquid electrode (CILE). CILE was immersed in a solution containing Hb and ionic liquid, octylpyridinium chloride ([OcPy][Cl]), to directly immobilize Hb on CILE. Cyclic voltammetry of modified electrode exhibited quasi-reversible peaks corresponding to Hb. The oxidation and reduction peak potentials of immobilized Hb in acetate buffer solution, pH 5.0 and at a scan rate of 0.1 V s−1 were obtained at about –150 mV and –290 mV, respectively. The average surface coverage of the electroactive Hb adsorbed on the electrode surface was calculated as 8.4 × 10−11 mol cm−2. Hb retained its bioactivity on modified electrode and showed excellent electrocatalytic activity towards oxygen, hydrogen peroxide and nitrite. Hydrogen peroxide can be determined in the range of 1.0 × 10−4–5.0 × 10−3 M.  相似文献

TiO2 nanocrystals had been modified on the surface of the glassy carbon electrode (GCE) with the help of Nafion. The electrochemiluminescence (ECL) behavior of the TiO2/Nafion GCE in aqueous solution was investigated. A possible mechanism about the ECL of TiO2 had also been proposed. The ECL intensity was linear with the dissolved oxygen concentration in the range of 0.30–10.00 mg/L with a detection limit of 0.12 mg/L. The developed method can be applied to detect the dissolved oxygen concentration or biochemical oxygen demand (BOD).  相似文献

A robust and effective composite film combined the benefits of Nafion, room temperature ionic liquid (RTIL) and multi‐wall carbon nanotubes (MWNTs) was prepared. Hemoglobin (Hb) was successfully immobilized on glassy carbon electrode surface by entrapping in the composite film. Direct electrochemistry and electrocatalysis of immobilized Hb were investigated in detail. A pair of well‐defined and quasi‐reversible redox peaks of Hb was obtained in 0.10 mol·L?1 pH 7.0 phosphate buffer solution (PBS), indicating that the Nafion‐RTIL‐MWNTs film showed an obvious promotion for the direct electron transfer between Hb and the underlying electrode. The immobilized Hb exhibited an excellent electrocatalytic activity towards the reduction of H2O2. The catalysis current was linear to H2O2 concentration in the range of 2.0×10?6 to 2.5×10?4 mol·L?1, with a detection limit of 8.0×10?7 mol·L?1 (S/N=3). The apparent Michaelis‐Menten constant (Kmapp) was calculated to be 0.34 mmol·L?1. Moreover, the modified electrode displayed a good stability and reproducibility. Based on the composite film, a third‐generation reagentless biosensor could be constructed for the determination of H2O2.  相似文献

This work points out that electrogeneration of silica gel (SG) films on glassy carbon electrodes (GCEs) can be applied to immobilize biomolecules – hemoglobin (Hb) or glucose oxidase (GOD) or both of them in mixture – without preventing their activity. These proteins were physically entrapped in the sol–gel material in the course of the electro-assisted deposition process applied to form the thin films onto the electrode surface. SG films were prepared from a precursor solution by applying a suitable cathodic potential likely to induce a local pH increase at the electrode/solution interface, accelerating thereby polycondensation of the silica precursors with concomitant film formation. Successful immobilization of proteins was checked by various physico-chemical techniques. Both Hb and GOD were found to undergo direct electron transfer, as demonstrated by cyclic voltammetry. GCE–SG–Hb gave rise to well-defined peaks at potentials Ec = −0.29 V and Ea = −0.17 V in acetate buffer, corresponding to the FeIII/FeII redox system of heme group of the protein, while GCE–SG–GOD was characterized by the typical signals of FAD group at Ec = −0.41 V and Ea = −0.33 V in phosphate buffer. These two redox processes were also evidenced on a single voltammogram when both Hb and GOD were present together in the same SG film. Hb entrapped in the silica thin film displayed an electrocatalytic behavior towards O2 and H2O2 in solution, respectively in the mM and μM concentration ranges. Immobilized GOD kept its biocatalytic properties towards glucose. Combined use of these two proteins in mixture has proven to be promising for detection of glucose in solution via the electrochemical monitoring of oxygen consumption (decrease of the oxygen electrocatalytic signal).  相似文献

The electrochemical reduction of molecular oxygen (O2) has been performed at gold electrodes modified with a submonolayer of a self-assembly (sub-SAM/Au) of a thiol compound (typically cysteine (CYST)) in O2-saturated 0.5 M KOH. At bare gold electrode O2 reduction reaction proceeds irreversibly, while this reaction is totally hindered at gold electrodes with a compact structure of CYST over its surface. The partial reductive desorption of the compact CYST monolayer was achieved by controlling the potential of the CYST/Au electrode, leading to the formation of a submonolayer coverage of the thiol compound over the Au electrode surface (sub-SAM/Au), at which the CYST molecules selectively block the Au(1 0 0) and Au(1 1 0) fractions (the so-called rough domains) of the polycrystalline Au while the Au(1 1 1) component (the so-called smooth domains) remains bare (i.e., uncovered with CYST). This sub-SAM/Au electrode extraordinarily exhibits a quasi-reversible two-electron reduction of molecular oxygen (O2) in alkaline medium with a peak separation (ΔEp) between the cathodic and anodic peak potentials (Epc,Epa) of about 60 mV. The ratio of the anodic current to the cathodic one is close to unity. The formal potential (Eo) of this reaction is found to equal −150 mV vs. Ag/AgCl/KCl(sat.).  相似文献

The electrochemical reduction of CO2 is strongly influenced by both the applied potential and the surface adsorption status of the catalyst. In this work a gas diffusion electrode (GDE) coated with Pd nanoparticles/carbon black (Pd/XC72) was used to study the electrochemical reduction of CO2. Cyclic voltammetric (CV) analysis of Pd/XC72 between 1.5 V and − 0.6 V (vs. RHE) shows the formation of intermediates and the blocking of hydrogen absorption on the Pd nanoparticles (NPs) under a CO2 atmosphere. The relationships between the Faradaic efficiency/current density and the applied potential reveal that the onset potential of CO formation is around − 0.4 V. Moreover, the presence of adsorbed CO was confirmed through CV analysis of Pd/XC72 under CO2 and CO/He atmospheres. This demonstrates that H atoms and CO intermediates co-adsorb on the surface of the Pd NPs at an applied potential of around − 0.4 V. When the applied potential is more negative than − 0.6 V, adsorption of CO intermediates on the surface of the Pd NPs becomes dominant.  相似文献

Multilayer film of laccase, poly-l-lysine (PLL) and multi-walled carbon nanotubes (MWNTs) were prepared by a layer-by-layer self-assembly technique. The results of the UV–vis spectroscopy and scanning electron microscopy studies demonstrated a uniform growth of the multilayer. The catalytic behavior of the modified electrode was investigated. The (MWNTs/PLL/laccase)n multilayer modified electrode catalyzed four-electron reduction of O2 to water, without any mediator. The possible application of the laccase-catalyzed O2 reduction at the (MWNTs/PLL/laccase)n multilayer modified ITO electrode was illustrated by constructing a glucose/O2 biofuel cell with the (MWNTs/thionine/AuNPs)8 GDH film modified ITO electrode as a bioanode and the (MWNTs/PLL/laccase)15 film modified ITO electrode as a biocathode. The open-circuit voltage reached to 700 mV, and the maximum power density achieved 329 μW cm−2 at 470 mV of the cell voltage.  相似文献

Hemoglobin modified electrode was successfully fabricated to realize direct electrochemistry by immobilizing of Hemoglobin (Hb) in bimodal mesoporous silica (BMS) and chitosan (CS) inorganic–organic hybrid film. Here, BMS acted as a support to immobilize Hb due to its large pores and CS acted as a binder to increase film adherence and stabilizer to prevent the leakage of Hb. The resulting electrode (Hb/BMS/CS) gave a well-defined, reversible redox couple for HbFe(III)/Fe(II) with a formal potential of about −0.32 V (vs. Ag/AgCl) in pH 7.0 phosphate buffer solution. Hb/BMS/CS electrode showed a better electrocatalytial performance to H2O2 with wider linear detection range, lower detection limit, and higher sensitivity than that at electrode without BMS. The improved electrocatalytic performance for Hb/BMS/CS electrode was possibly contributed to BMS bimodal structure, whose large pores with 10–40 nm provide favorable conditions for protein immobilization and small pores with 2–3 nm avoid the mass-transfer limitations. In addition, UV–Vis and FTIR spectra indicated that Hb well maintained its native structure in the hybrid film.  相似文献

The direct electron transfer between hemoglobin (Hb) and the underlying glassy carbon electrode (GCE) can be readily achieved via a high biocompatible composite system based on biopolymer chitosan (CHT) and inorganic CaCO3 nanoparticles (nano-CaCO3). Cyclic voltammetry of Hb-CHT/nano-CaCO3/GCE showed a pair of stable and quasi-reversible peaks for HbFe(III)/Fe(II) redox couple in pH 7.0 buffer. The electrochemical reaction of Hb immobilized in CHT/nano-CaCO3 composite matrix exhibited a surface-controlled process accompanied by electron and proton transfer. The electron transfer rate constant was estimated to be 1.8 s−1. This modified electrode showed a high thermal stability up to 60 °C. The apparent Michaelis–Menten constant was calculated to be 7.5 × 10−4 M, indicating a high catalytic activity of the immobilized Hb toward H2O2. The interaction between Hb and this nano-hybrid material was also investigated using FT-IR and UV–vis spectroscopy, indicating that Hb retained its native structure in this hybrid matrix.  相似文献

Myoglobin (Myb) was successfully immobilized on a room temperature ionic liquid (RTIL), 1‐ethyl‐3‐methyl imidazolium tetrafluoroborate ([EMIM][BF4]) modified basal plane graphite (BPG) electrode. The electrochemical behavior of Myb on RTIL modified BPG electrode was explored and the results from cyclic voltammetry (CV) showed a well‐defined and quasi‐reversible CV peaks with a formal potential of ?0.379 V (versus Ag/AgCl) in a phosphate buffer solution (pH 7.0). RTIL shows an obvious promotion for the direct electron‐transfer between Myb and BPG electrode. Myb adsorbed on electrode surface exhibits an obvious electrocatalytic activity for the reduction of nitric oxide (NO). The catalytic current is corresponding linearly to the NO concentration in the range of 7.0×10?7 to 7.0×10?6 M with a limit of detection of 2.0×10?7 M (three times the ratio of signal to noise, S/N=3).  相似文献

The role of surface oxygen groups on the kinetics of the V(II) oxidation reaction was studied on modified glassy carbon (GC) electrodes by cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The reaction was found to be sensitive to the presence of oxygen groups on the electrode surface. Higher O/C ratios determined by X-ray photoelectron spectroscopy (XPS) corresponded to higher reactivities and lower charge transfer resistances measured in a 1 M V(II) electrolyte. The stability of an oxidised GC surface was also investigated in a 1 M V(II) electrolyte by potential holding and cycling experiments. It was found that after holding and cycling to successively more negative potentials up to − 0.8 V/RHE, the electrode surface lost its initial reactivity.  相似文献

An interesting mode of reactivity of MnO2 nanoparticles modified electrode in the presence of H2O2 is reported. The MnO2 nanoparticles modified electrodes show a bi-direction electrocatalytic ability toward the reduction/oxidation of H2O2. Based on this property, a choline biosensor was fabricated via a direct and facile electrochemical deposition of a biocomposite that was made of chitosan hydrogel, choline oxidase (ChOx) and MnO2 nanoparticles onto a glassy carbon (GC) electrode. The biocomposite is homogeneous and easily prepared and provides a shelter for the enzyme to retain its bioactivity. The results of square wave voltammetry showed that the electrocatalytic reduction currents increased linearly with the increase of choline chloride concentration in the range of 1.0 × 10−5 –2.1 × 10−3 M and no obvious interference from ascorbic acid and uric acid was observed. Good reproducibility and stability were obtained. A possible reaction mechanism was proposed.  相似文献

In this paper, a novel biosensor was prepared by immobilizing glucose oxidase (GOx) on carbon nanotube-gold-titania nanocomposites (CNT/Au/TiO2) modified glassy carbon electrode (GCE). SEM was initially used to investigate the surface morphology of CNT/Au/TiO2 nanocomposites modified GCE, indicating the formation of the nano-porous structure which could readily facilitate the attachment of GOx on the electrode surface. Cyclic voltammogram (CV) and electrochemical impedance spectrum (EIS) were further utilized to explore relevant electrochemical activity on CNT]Au/TiO2 nanocomposites modified GCE. The observations demonstrated that the immobilized GOx could efficiently execute its bioelectrocatalytic activity for the oxidation of glucose. The biosensor exhibited a wider linearity range from 0.1 mmol L-1 to 8 mmol L^-1 glucose with a detection limit of 0.077 mmol L^- 1.  相似文献

The composite film based on Nafion and hydrophobic room-temperature ionic liquid (RTIL) 1-butyl-3-methyl-imidazolium hexafluorophosphate ([bmim] PF6) was explored. Here, Nafion was used as a binder to form Nafion-ionic liquids composite film and help [bmim] PF6 effectively adhered on glassy carbon (GC) electrode. X-ray photoelectron spectroscopy (XPS), cyclic voltammtery (CV) and electrochemical impedance spectroscopy (EIS) were used to characterize this composite film, showing that the composite film can effectively adhere on the GC electrode surface through Nafion interacting with [bmim] PF6 and GC electrode. Meanwhile, doping [bmim] PF6 in Nafion can also effectively reduce the electron transfer resistance of Nafion. The composite film can be readily used as an immobilization matrix to entrap horseradish peroxidase (HRP). A pair of well-defined redox peaks of HRP was obtained at the HRP/Nafion-[bmim] PF6 composite film-modified GC electrode through direct electron transfer between the protein and the underlying electrode. HRP can still retain its biological activity and enhance electrochemical reduction towards O2 and H2O2. It is expected that this composite film may find more potential applications in biosensors and biocatalysis.  相似文献

4-Amino-2,2,6,6-tetramethyl-1-piperridine N-oxyl (4-amino-TEMPO), an electroactive nitroxide radical, was attached to the surface of graphene oxide (GO) and electrochemically reduced graphene oxide (ERGO) modified glassy carbon electrode by a simple, rapid and green electrografting method. The electroactive interfaces were analyzed by X-ray photoelectron spectroscopy (XPS) and cyclic voltammetry (CV). The calculated surface coverage for 4-amino-TEMPO is up to 1.55 × 10 9 mol·cm 2. The modified electroactive interface exhibited excellent electrocatalytic activity towards the electro-oxidation of reduced glutathione (GSH) and hydrogen peroxide (H2O2).  相似文献

The voltammetric (CV and DPV) behavior of multi-walled carbon nanotube/Nafion composite coupled with a glassy carbon electrode was investigated for the determination of 2,4-dichlorophenol. The structural and morphological evaluation by XRD and FESEM revealed that the acid treated MWCNT retained their morphology without any structural change. The existence of the possible functional groups was investigated by FTIR and Raman spectroscopy. Compared to bare GCE, a significantly reduced interfacial charge transfer resistance was noticed for MWCNT/Nafion/GCE by electrochemical impedance spectroscopy (EIS). The use of Nafion not only contributed to the non-covalent functionalization of MWCNT, but also protected the electrode surface against the polymerization of phenoxy radicals forming a passivating film. For MWCNT/Nafion/GCE, the combination of anti-passivating ability and excellent catalytic properties resulted in the rapid and direct electrochemical determination of 2,4-DCP. Under optimal experimental conditions, the DPV responses for MWCNT/Nafion/GCE is linear over the 1–150 μmol/L range with a detection limit (S/N = 3) of 0.01 μmol/L. The presence of many interfering species had no influence on the signals of 2,4-DCP. The proposed sensor was successfully tested for the determination of 2,4-DCP in tap water samples and the recovery was in the range of 99.0–102.5%.  相似文献
