Gold nanorods (GNRs) were synthesized by a seed–mediated growth approach followed by TEOS polymerization leading to the formation of silica layer surrounding the gold nanorod core. TEM images showed that the silica-coated gold nanorods (GNRs@SiO2) were dispersed with an average aspect ratio of 3.1 for the GNRs cores and a uniform thickness of the silica shell. The core/shell nanocomposites were further used as efficient supports for the immobilization of hemoglobin (Hb) to fabricate a novel biosensor. The immobilized Hb showed an enhanced electron transfer for its heme Fe(III) to Fe(II) redox couple. This biosensor showed an excellent bioelectrocatalytic activity towards H2O2 with a linear range from 8.0 × 10−7 to 6.1 × 10−5 M, and the detection limit was 6.0 × 10−8 M at 3σ. The apparent Michaelis–Menten constant of the immobilized hemoglobin was calculated to be 0.13 mM.  相似文献

A robust and effective composite film based on gold nanoparticles (GNPs)/room temperature ionic liquid (RTIL)/multi-wall carbon nanotubes (MWNTs) modified glassy carbon (GC) electrode was prepared by a layer-by-layer self-assembly technique. Cytochrome c (Cyt c) was successfully immobilized on the RTIL-nanohybrid film modified GC electrode by electrostatic adsorption. Direct electrochemistry and electrocatalysis of Cyt c were investigated. The results suggested that Cyt c could be tightly adsorbed on the modified electrode. A pair of well-defined quasi-reversible redox peaks of Cyt c was obtained in 0.10 M, pH 7.0 phosphate buffer solution (PBS). RTIL-nanohybrid film showed an obvious promotion for the direct electron transfer between Cyt c and the underlying electrode. The immobilized Cyt c exhibited an excellent electrocatalytic activity towards the reduction of H2O2. The catalysis currents increased linearly to the H2O2 concentration in a wide range of 5.0 × 10−5– 1.15 × 10−3 M. Based on the multilayer film, the third-generation biosensor could be constructed for the determination of H2O2.  相似文献

A new polymeric membrane electrode has been constructed for the determination of phenylpropanolamine hydrochloride. The electrode was prepared by solubilizing the phenylpropanolamine-phosphomolybdate ion associate into a polyvinyl chloride matrix plasticized by dibutylphthalate as a solvent mediator. The electrode showed near-Nernstian response over the concentration range of 1 × 10?5–1 × 10?2 M with low detection limit of 6.3 × 10?6 M. The electrode displays a good selectivity for phenylpropanolamine with respect to a number of common inorganic and organic species. The electrode was successfully applied to the potentiometric determination of phenylpropanolamine ion in its pure state and its pharmaceutical preparation in batch and flow injection conditions.  相似文献

A novel strategy based on the Ugi multicomponent reaction was employed for immobilizing horseradish peroxidase on sodium alginate-coated gold electrode. The electrode was employed for constructing an amperometric biosensor device using 1 mM hydroquinone as electrochemical mediator. The electrode showed linear response (poised at −300 mV vs Ag/AgCl) toward H2O2 concentration between 70 μM and 8.8 mM at pH 7.0. The biosensor reached 95% of steady-state current in about 12 s and its sensitivity was 33.8 mA/M cm2. The electrode retained full initial activity after 30 days of storage at 4 °C in 50 mM sodium phosphate buffer, pH 7.0.  相似文献

An interesting mode of reactivity of MnO2 nanoparticles modified electrode in the presence of H2O2 is reported. The MnO2 nanoparticles modified electrodes show a bi-direction electrocatalytic ability toward the reduction/oxidation of H2O2. Based on this property, a choline biosensor was fabricated via a direct and facile electrochemical deposition of a biocomposite that was made of chitosan hydrogel, choline oxidase (ChOx) and MnO2 nanoparticles onto a glassy carbon (GC) electrode. The biocomposite is homogeneous and easily prepared and provides a shelter for the enzyme to retain its bioactivity. The results of square wave voltammetry showed that the electrocatalytic reduction currents increased linearly with the increase of choline chloride concentration in the range of 1.0 × 10−5 –2.1 × 10−3 M and no obvious interference from ascorbic acid and uric acid was observed. Good reproducibility and stability were obtained. A possible reaction mechanism was proposed.  相似文献

In this work, we have prepared nano-material modified carbon paste electrode (CPE) for the sensing of an antidepressant, buzepide methiodide (BZP) by incorporating TiO2 nanoparticles in carbon paste matrix. Electrochemical studies indicated that the TiO2 nanoparticles efficiently increased the electron transfer kinetics between drug and the electrode. Compared with the nonmodified CPE, the TiO2-modified CPE greatly enhances the oxidation signal of BZP with negative shift in peak potential. Based on this, we have proposed a sensitive, rapid and convenient electrochemical method for the determination of BZP. Under the optimized conditions, the oxidation peak current of BZP is found to be proportional to its concentration in the range of 5 × 10−8 to 5 × 10−5 M with a detection limit of 8.2 × 10−9 M. Finally, this sensing method was successfully applied for the determination of BZP in human blood serum and urine samples with good recoveries.  相似文献

A novel electrogenerated chemiluminescence aptamer-based (ECL-AB) biosensor for the determination of a small molecule drug is designed employing cocaine-binding aptamer as molecular recognition element for cocaine as a model analyte and ruthenium complex served as an ECL label. A 5′-terminal cocaine-binding aptamer with the ECL label at 3′-terminal of the aptamer was utilized as an ECL probe. The ECL-AB biosensors were fabricated by immobilizing the ECL probe onto a gold electrode surface via thiol-Au interactions. An enhanced ECL signal is generated upon recognition of the target cocaine, attributed to a change in the conformation of the ECL probe from random coil-like configuration on the probe-modified film to three-way junction structure, in close proximity to the sensor interface. The integrated ECL intensity versus the concentration of cocaine was linear in the range from 5.0 × 10−9 to 3.0 × 10−7 M. The detection limit was 1.0 × 10−9 M. This work demonstrates that the combination of a highly binding aptamer to analyte with a highly sensitive ECL technique to design ECL-AB biosensor is a great promising approach for the determination of small molecule drugs.  相似文献

Direct electrochemical response of Myoglobin (Myb) at the basal plane graphite (BPG) electrode was observed when a room temperature ionic liquid (RTIL), 1-(2-hydroxyethyl)-3-methyl imidazolium tetrafluoroborate ([HEMIm][BF4]), was used as the supporting electrolyte. In a 0.17 M [HEMIm]BF4 aqueous solution, a couple of well-defined redox peaks of Myb could be obtained, whose anodic and cathodic peak potentials were at −0.158 and −0.224 V (vs. Ag/AgCl), respectively. Both anodic and cathodic peak currents increased linearly with the potential scan rate. Compared with the supporting electrolyte of phosphate buffer, [HEMIm][BF4] played an obvious promotion for the direct electron transfer between Myb and the BPG electrode. Further investigation suggested that Myb was adsorbed tightly on the surface of the BPG electrode in the presence of [HEMIm][BF4] to form a stable, approximate monolayer Myb film. Myb adsorbed on the BPG electrode surface showed a remarkable electrocatalytic activity for the reduction of oxygen in a [HEMIm][BF4] aqueous solution. Based on these, a third-generation biosensor could be constructed to directly detect the concentration of oxygen in aqueous solution with a limit of detection of 2.3 × 10−8 M.  相似文献

This work deals with a novel preparation method of bilirubin oxidase/2,2′-azinobis-3-ethylbenzothiazoline-6-sulfonic acid electrode. The enzyme and its mediator were adsorbed on carbon Vulcan XC-72R before their immobilization into a Nafion® matrix. Promising results were obtained when this biocathode was associated with Au70Pt30 nanoparticles as anode in a single concentric glucose/O2 biofuel cell (BFC). The latter BFC delivered at 37 °C a power density of 90 μW cm?2 for a cell voltage of 0.4 V in phosphate buffer (pH 7.4) containing 0.01 M glucose. Moreover, the electrical performances were increased with the concentration of glucose by generating up to 190 μW cm?2 for a cell voltage of 0.52 V when the concentration of the renewable fuel reached 0.7 M.  相似文献

The direct electron transfer between hemoglobin (Hb) and the underlying glassy carbon electrode (GCE) can be readily achieved via a high biocompatible composite system based on biopolymer chitosan (CHT) and inorganic CaCO3 nanoparticles (nano-CaCO3). Cyclic voltammetry of Hb-CHT/nano-CaCO3/GCE showed a pair of stable and quasi-reversible peaks for HbFe(III)/Fe(II) redox couple in pH 7.0 buffer. The electrochemical reaction of Hb immobilized in CHT/nano-CaCO3 composite matrix exhibited a surface-controlled process accompanied by electron and proton transfer. The electron transfer rate constant was estimated to be 1.8 s−1. This modified electrode showed a high thermal stability up to 60 °C. The apparent Michaelis–Menten constant was calculated to be 7.5 × 10−4 M, indicating a high catalytic activity of the immobilized Hb toward H2O2. The interaction between Hb and this nano-hybrid material was also investigated using FT-IR and UV–vis spectroscopy, indicating that Hb retained its native structure in this hybrid matrix.  相似文献

A highly sensitive method was investigated for the simultaneous determination of acetaminophen (AC), dopamine (DA), and ascorbic acid (AA) using a PbS nanoparticles Schiff base-modified carbon paste electrode (PSNSB/CPE). Differential pulse voltammetry peak currents of AC, DA and AA increased linearly with their concentrations within the ranges of 3.30 × 10−8–1.58 × 10−4 M, 5.0 × 10−8–1.2 × 10−4 M and 2.50 × 10−6–1.05 × 10−3 M, respectively, and the detection limits for AC, DA and AA were 5.36 × 10−9, 2.45 × 10−9 and 1.86 × 10−8 M, respectively. The peak potentials recorded in a phosphate buffer solution (PBS) of pH 4.6 were 0.672, 0.390, and 0.168 V (vs Ag/AgCl) for AC, DA and AA, respectively. The modified electrode was used for the determination of AC, DA, and AA simultaneously in real and synthetic samples.  相似文献

The present work is dedicated to making the best of vertically-aligned TiO2 nanotubes (TNTs) array to serve as a prospectively ideal “vessel” for protein immobilization and biosensor applications. The TNTs fabricated by electrochemical anodizing possess the advantageous of perpendicular alignment and tailored tubular architecture, as well as the good biocompatibility and hydrophilicity. But the electron-transfer resistance of the as-grown (AG-) TNTs is too large for the direct electron transfer and electrochemical biosensing. A simple strategy on controllable electrochemical reduction treatment of TNTs is adopted on it, leading TNTs in situ self-doped with Ti(III), which makes the Ti(III)–TNTs much better conductivity while the tubular and crystal structure of TNTs array still well maintained. Results show that the TNTs can be used as a super vessel for rapid and substantive immobilization of hemoglobin (Hb), with a large surface electroactive Hb coverage (Γ*) of 1.5 × 10?9 mol cm?2. The enhanced direct electron transfer of Hb is commendably observed on the Ti(III)–TNTs/Hb biosensor with a couple of well-defined redox peaks compared with the AG-TNTs/Hb. The biosensor further exhibits fast response, high sensitivity and stability for the amperometric biosensing of H2O2 with the detection limit of 1.5 × 10?6 M, and the apparent Michaelis–Menten constant of 1.02 mM.  相似文献

MgO polyhedral nanocages and nanocrystals, synthesized by non-catalytic simple thermal evaporation process, were used to fabricate high-sensitive amperometric glucose biosensor which showed a high and reproducible sensitivity of 31.6 μA μM?1 cm?2 with a response time less than 5 s, linear dynamic range from 1.0 to 9.0 μM and correlation coefficient of R = 0.9993. The detection limit of fabricated biosensor (based on S/N ratio = 3) was estimated to be 68.3 ± 0.02 nM. To the best of our knowledge, this is the first report which demonstrates the use of MgO nanostructures for the fabrication of glucose biosensor; hence, this work opens a new way to utilize MgO nanostructures as an efficient electron mediator to fabricate efficient glucose biosensors.  相似文献

A new type of amperometric hydrogen peroxide biosensor was fabricated by entrapping horseradish peroxidase (HRP) in the organic–inorganic hybrid material composed of zirconia–chitosan sol–gel and Au nanoparticles (ZrO2–CS–AuNPs). The sensitivity of the biosensor was enhanced by a flowerlike polymer–copper nanostructure composite (pPA–FCu) which was prepared from co-electrodeposition of CuSO4 solution and 2,6-pyridinediamine solution. Several techniques, including UV–vis absorption spectroscopy, scanning electron microscopy, cyclic voltammetry, chronoamperometry and electrochemical impedance spectroscopy were employed to characterize the assembly process and performance of the biosensor. The results showed that this pPA–FCu nanostructure not only had excellent redox electrochemical activity, but also had good catalytic efficiency for hydrogen peroxide. Also the ZrO2–CS–AuNPs had good film forming ability, high stability and good retention of bioactivity of the immobilized enzyme. The resulting biosensors showed a linear range from 7.80 × 10?7 to 3.7 × 10?3 mol L?1, with a detection limit of 3.2 × 10?7 mol L?1 (S/N = 3) under optimized experimental conditions. The apparent Michaelis–Menten constant was determined to be 0.32 mM, showing good affinity. In addition, the biosensor which exhibits good analytical performance, acceptable stability and good selectivity, has potential for practical applications.  相似文献

We describe the preparation of novel poly(thionine)-Au materials, where the poly(thionine)-Au nano-network and nanowires have been synthesized in aqueous solution via the polymerization of thionine using HAuCl4 as the oxidant in a single reaction setup. The synthesis process does not require templates, nor does it require large amounts of organic solvents or electrochemical methods. The morphology of the nanocomposites can be controlled by varying the thionine/HAuCl4 ratio. The resulting poly(thionine)-Au network was used to fabricate a novel non-enzyme hydrogen peroxide (H2O2) biosensor. In pH 7.0 phosphate buffer, almost interference-free determination of H2O2 was realized at − 0.1 V versus Ag/AgCl with a linear of 1 × 10 4 to 5 × 10 2 M, a correlation coefficient of 0.998 and a response time of < 2 s. The developed biosensor showed a detection limit of 0.2 μM (S/N = 3) with very good stability, reproducibility and high selectivity.  相似文献

In this communication, a hydrogen peroxide (H2O2) sensor based on self-assembled Prussian Blue (PB) modified electrode was reported. Thin film of PB was deposited on the electrode by self-assembly process including multiple sequential adsorption of ferric ions and hexacyanoferrate ions. The as-prepared PB modified electrode displayed sufficient stability for practical sensing application. At an applied potential of ?0.05 V vs. Ag/AgCl (sat. KCl), PB modified electrode with 30 layers exhibited a linear dependence on H2O2 concentration in the range of 1 × 10?6–4 × 10?4 M (r = 0.9998) with a sensitivity of 625 mA M?1 cm?2. It was found that the sensitivity of H2O2 sensors could be well controlled by adjusting the number of deposition cycles for PB preparation. This work demonstrates the feasibility of self-assembled PB modified electrode in sensing application, and provides an effective approach to control the sensitivity of PB-based amperometric biosensors.  相似文献

Using porous cuprous oxide (Cu2O) microcubes, a simple non-enzymatic amperometric sensor for the detection of H2O2 and glucose has been fabricated. Cyclic voltammetry (CV) revealed that porous Cu2O microcubes exhibited a direct electrocatalytic activity for the reduction of H2O2 in phosphate buffer solution and the oxidation of glucose in an alkaline medium. The non-enzymatic amperometric sensor used in the detection of H2O2 with detection limit of 1.5 × 10?6 M over wide linear detection ranges up to 1.5 mM and with a high sensitivity of 50.6 μA/mM. This non-enzymatic voltammetric sensor was further utilized in detection of glucose with a detection limit of 8.0 × 10?7 M, a linear detection range up to 500 μM and with a sensitivity of ?70.8 μA/mM.  相似文献

New biocomposite materials, based on the incorporation of DNA doped p-aminobenzensulfonic acid, was fabricated by electrochemical method. A carbon fiber microelectrode modified by this thin film was fabricated for selective determination of uric acid (UA) in the presence of a larger amount of ascorbic acid (AA). It was found that the voltammetric oxidation peak separation between UA and AA is about 260 mV at the modified electrode. A linear response of the peak current versus the concentration was found in the range of 8 × 10−7–6 × 10−4 M with correlation coefficient of 0.9991 and the detection limit was 5 × 10−7 M (s/n = 3) at the 5 × 10−4 M AA. The presence of high concentration AA did not interference the determination. The electropolymerized film was characterized by SEM techniques. The modified electrode shows good sensitivity, selectivity and stability.  相似文献

A new PVC membrane based strontium(II) ion-selective electrode has been constructed using acetophenone semicarbazone as a neutral carrier. The sensor exhibits a Nerstian response for strontium(II) ion over a wide concentration range 1.0 × 10−2–1.0 × 10−7 M with the slope of 29.4 mV/per decade. The limit of detection was 2.7 × 10−8 M. It was relatively fast response time (<10 s for concentration ⩾1.0 × 10−3 and <15 s for concentration of ⩾1.0 × 10−6 M) and can be used for 8 months without any considerable divergence in potentials. The proposed sensor revealed relatively good selectivity and high sensitivity for strontium(II) over a mono, di, trivalent cation and can be used in a pH range of 2.5–10.5. It was also successfully used as an indicator electrode in potentiometer titration and in the analysis of concentration in various real samples.  相似文献

Binding of copper to three peptide fragments of prion (Cu2+ binding sites: 60–91, 92–96 and 180–193 amino acid residues) was investigated by anodic stripping voltammetry to determine the stoichiometries of Cu2+-prion peptide interactions. The method relies on the synthesis of N-terminally acetylated/C-terminally amidated peptide fragments of prion by solid-phase synthesis and direct monitoring of the oxidation current of copper in the absence and presence of each prion fragment. Titration curves of Cu2+ with Ac-PHGGGWGQ-NH2, Ac-GGGTH-NH2 and Ac-VNITKQHTVTTTT-NH2 were obtained in concentrations ranging from 8.52 × 10?7 to 5.08 × 10?6, 3.95 × 10?7 to 1.94 × 10?6 and 7.82 × 10?8 to 4.51 × 10?7 M, respectively. The acquired data were used to calculate the stoichiometries (one peptide per Cu2+ ion for all the three studied systems) and apparent dissociation constants (Kd = 4.37 × 10?8–3.50 × 10?10 M) for the three complexes.  相似文献
