A direct and efficient approach to synthesize imidazo[1,2-α]pyridines through three-component one-pot reaction of 2-aminopyridine,aldehyde and terminal alkyne catalyzed by Cu-nanoparticles under solvent-free conditions has been developed.This method provides a rapid access to substituted imidazo[1,2-α]pyridines with good yields(up to 85%).  相似文献

4-Methylthiopyrido[1,2-α]pyrimidin-2-one and 2-hydroxypyrido[1,2-α]pyrimidine-4-thione derivatives were synthesized by the addition ofN-(4-R-pyrid-2-yl)acetoacetamides (R = H, Me) to CS2 under phase-transfer conditions followed by the alkylation of the reaction products with Mel. The molecular structure of 3-acetyl-4-methylthiopyrido[1,2-α]pyrimidin-2-one is established by X-ray analysis.  相似文献

Some novel pyrazolo[1,5-α]pyrimidines 5a-f, 10a,b, 1,2,4-triazolo[1.5-α]-pyrimidine (12), and pyrimido[1,2-α]benzimidazole 18 could be synthesized by reacting 3-(4-chlorophenyl)-2-(N,N-dimethylamino)methylene-3-oxopropanenitrile (2) with 5-amino-3- and/or 4-substituted-1H-pyrazoles 3a-f and 9a,b, 3-amino-1,2,4-triazole (11) and 2-aminobenzimidazole (14), respectively. The reaction of 2 with 1H-benzimidazol-2-ylacetonitrile (19) afforded the pyrido[1,2-α]benzimidazole 20. On the other hand, the reaction of 2 with guanidine, hydrazine, and phenyl hydrazine afforded the pyrimidine 24, and the pyrazoles 27, 28, respectively. However, the reaction of 2 with hydroxyl amine did not afford the isoxazole 32.  相似文献

The reaction of various anthranilic acid derivatives or their esters with 4-oxotetrahydrothiophene-3-carbonitrile 2, 2-oxocyclopentanecarbonitrile (9, n = 1) or 2-oxocyclohexanecarbonitrile (9, n = 2) in ethanol under reflux conditions giving rise the formation of single products isolated in each case after simple filtration. The products were characterized as pyrazolo[1,5-a]quinazolin-5-ones 4 instead of the expected pyrazol-3-amines 3. These cascade condensation–intramolecular acylation processes generated in one-step reactions from simple starting materials novel heterocyclic scaffolds ready for further functionalization. The present synthetic protocol provides acceptable yields of new tetracyclic products in high purity.  相似文献

Structure containing the CS bond exist widely in nature, drugs and chemical materials. Here, a novel sulfenylation protocol employing an aryl or alkyl halide and odorless and cheap S as reagents was developed, generating regioselective alkyl-S- and Ar-S-substituted imidazol[1,2-α]pyridine derivatives in good yields under relatively environmentally friendly and mild conditions. This protocol enriches current thioether-producing methods, making up for the shortcomings of previous sulfenylation methods which can only make MeS- and ArS-substituted imidazol[1,2-α]pyridine derivatives.  相似文献

An efficient catalyst-free synthesis of 3′-benzoyl-4′,5′-diphenyl-2H-spiro[acenaphthylene-1,2′-pyrrolidin]-2-one derivatives via one-pot 1,3-dipolar cycloaddition of acenaphthenequinone, arylmethyl amines, and chalcones with high regioselectivity is described. The structure of the cycloadducts were characterized by infrared, high-resolution mass spectrometry (electrospray ionization), 1H NMR, and 13C NMR spectra, and the structure of 4a was confirmed using x-ray single-crystal structure analysis.  相似文献

An efficient, four-component reaction of isatin, 1,3-indanedione, ethyl acetoacetate, and ammonium acetate in ethanol/water (9:1) system furnished spiro[4H-indeno[1,2-b]pyridine-4,3′-[3H]indoles] at room temperature. Merits of the method are mild reaction conditions, simple workup procedure, and ambient temperature. The synthesized compounds exhibit excellent fluorescence properties.  相似文献

3-Phenyl-5-methyl(phenyl)-1H,3H-pyrimido[4,5:4,5]pyrimido[1,2-a]benzimi-dazole-2,4-diones were obtained by the cyclization of N-phenyl-N-[3-ethoxycar-bonyl-4-methyl(phenyl)pyrimido [1,2-a]benzimidazol-2-yl]ureas by the action of potassium carbonate. 2-Methyl-5-phenyl-1H-pyrimido[4,5:4,5]pyrimido[1,2-a]benzimidazol-4-one was obtained by the reaction of N-3-ethoxycarbonyl-4-phenylpyrimido[1,2-a]benzimidazol-2-yl]-N,N-dimethylacetamidine with ammonium acetate in ethanol. The synthesized compounds are members of a new heterocyclic system. 2-Aminocyclopenta[d]pyrimidines 3a–c were achieved via a one-pot, three-component reactions of cyclopentanone 1, aromatic aldehyde and guanidine hydrochloride(1:2:1 molar ratio). Also, cyclization of 2,5-bis-(arylmethylidene)cyclopentanones 2 with guanidine hydrochloride(1:1 molar ratio) in methanol in the presence of sodium methoxide afforded cyclopenta-[d]pyrimidines 3. Compound 3c has been shown to be a useful building block for the synthesis of some novel pyrimido[1,2-a]pyrimidines 5, 7and 12. The structures of the newly synthesized compounds were confirmed on the basis of analytical and spectral data.  相似文献

N-Substituted imidazo[4,5-e]benzo[1,2-c;3,4-c’]difuroxans were synthesized by phase-transfer catalyzed alkylation of imidazo[4,5-e]benzo[1,2-c;3,4-c’]difuroxan salts with various alkylating agents. Acid hydrolysis of N-(ethoxycarbonylmethyl)imidazo[4,5-e]benzo-[1,2-c;3,4-c’]difuroxans gives the corresponding carboxylic acids. Structures of the synthesized compounds were confirmed by NMR spectroscopy, high resolution mass spectrometry, IR spectroscopy, and elemental anslysis.

New series of pyrazolo[1,5-α]pyrimidine derivatives 7a-i,11a-c and Schiff bases 13a-c were synthesized and screened for their in vitro antitumor activity against three human carcinoma cell lines,namely colorectal carcinoma(HCT116),prostate adenocarcinoma(PC-3) and liver carcinoma(HepG-2) using MTT cytotoxicity assay at 100 μg/mL.Some of the tested compounds displayed good anticancer activities against HCT-116 and PC-3 cells.Whereas,compounds 7d and 11 a showed better antitumor activity than the rest of the compounds against both cell lines.A structure-activity relationship(SAR) has been discussed and structures of the newly synthesized compounds were confirmed by different spectral data(MS,IR,~1H NMR and ~(13)C NMR) and elemental analysis.  相似文献

The synthesis of the protected glycopeptide Z-Thr(tBu)-Ala-Thr-[α-D-Gal(Bzl)4 ]-Thr(tBu)-NHNH-Boc was performed in solution by a stepwise coupling strategy. Fmoc-Thr[(α+β)-D- Gal(Bzl)4]-0Su, obtained from the reducing sugar and the protected amino acid by the trifluoromethanesulfonic anhydride procedure, was reacted with H-Thr(tBu)-NHNH-Boc. The two diastereoisomers of the resulting glycosylated dipeptide derivative were purified and separated by column chromatography. The anomeric configuration of the 0-glycosidic linkage was assigned by 1H-NMR-spectroscopy. The α-anomeric glycodipeptide, obtained in a larger amount, was selectively deblocked at the amino function and acylated with Z-Thr(tBu)-Ala-OH in the presence of either IIDQ or EEDQ, to afford the desired protected glycotetrapeptide.  相似文献
