We present the access to [5-19F, 5-13C]-uridine and -cytidine phosphoramidites for the production of site-specifically modified RNAs up to 65 nucleotides (nts). The amidites were used to introduce [5-19F, 5-13C]-pyrimidine labels into five RNAs—the 30 nt human immunodeficiency virus trans activation response (HIV TAR) 2 RNA, the 61 nt human hepatitis B virus ϵ (hHBV ϵ) RNA, the 49 nt SAM VI riboswitch aptamer domain from B. angulatum, the 29 nt apical stem loop of the pre-microRNA (miRNA) 21 and the 59 nt full length pre-miRNA 21. The main stimulus to introduce the aromatic 19F–13C-spin topology into RNA comes from a work of Boeszoermenyi et al., in which the dipole-dipole interaction and the chemical shift anisotropy relaxation mechanisms cancel each other leading to advantageous TROSY properties shown for aromatic protein sidechains. This aromatic 13C–19F labeling scheme is now transferred to RNA. We provide a protocol for the resonance assignment by solid phase synthesis based on diluted [5-19F, 5-13C]/[5-19F] pyrimidine labeling. For the 61 nt hHBV ϵ we find a beneficial 19F–13C TROSY enhancement, which should be even more pronounced in larger RNAs and will facilitate the NMR studies of larger RNAs. The [19F, 13C]-labeling of the SAM VI aptamer domain and the pre-miRNA 21 further opens the possibility to use the biorthogonal stable isotope reporter nuclei in in vivo NMR to observe ligand binding and microRNA processing in a biological relevant setting.  相似文献

Pseudo‐meta and pseudo‐para diamino‐octafluoro[2.2]paracyclophanes are challenging to separate either by chromatography or recrystallization, but through the use of a mixture of the two isomers, the 1H, 19F, and 13C NMR spectra of these compounds have been fully and unambiguously assigned using 1H COSY, 1H‐19F HOESY, 1H‐13C HSQC, 1H‐13C HMBC, and 19F‐13C HSQC techniques. This permits the easy identification of either of the individual isomers. In addition, the 13C spectrum of the pseudo‐ortho analogue is reported and assigned for the first time. The gem shift effect in this series of bridge‐fluorinated paracyclophanes serves to deshield 1H resonances and shield 13C. Copyright © 2012 John Wiley & Sons, Ltd.  相似文献

The conformation of [bis‐(N,N′‐difluoroboryl)]‐3,3′‐diethyl‐4,4′,8,8′,9,9′,10,10′‐octamethyl‐2,2′‐bidipyrrin (1) in solution was studied by analyzing the 13C? 19F and 19F? 19F through‐space spin–spin couplings. The 1H and 13C NMR spectra were assigned on the basis of nuclear Overhauser effect spectroscopy (NOESY), heteronuclear single‐quantum correlation (HSQC), and heteronuclear multiple‐bond correlation (HMBC) experiments. The 19F spectrum of 1 was compared with that of 2‐ethyl‐1,3,5,6,7‐pentamethyl‐4,4‐difluoro‐4‐bor‐3a,4a‐diaza‐s‐indacen (2). The 19F? 19F through‐space spin? spin coupling in 1 was thus assigned and the coupling constant was obtained by simulating the coupling patterns. The obtained conformation of 1 was compared with those of the known complexes [bis‐(N,N′‐difluoroboryl)]‐3,3′,8,8′,9,9′‐hexaethyl‐4,4′,10,10′‐tetramethyl‐6,6′‐(4‐methylphenyl)‐2,2′‐bidipyrrin (3)and [bis‐(N,N′‐difluoroboryl)]‐9,9′‐diethyl‐4,4′,8,8′,10,10′‐hexamethyl‐3,3′‐bis(methoxycarbonylethyl)‐2,2′‐bidipyrrin (4). The conformational dynamics of 1, 3, and 4 was surveyed by observing the temperature dependence of the through‐space coupling constants between 253 and 333 K. The 13C? 19F and 19F? 19F through‐space spin–spin couplings thus confirm similar conformations of different BisBODIPYs in solution in contrast to earlier findings in the solid state. 2′‐O‐[(4‐Trifluoromethyl‐triazol‐1‐yl)methyl] reporter groups have been incorporated into guanosine‐rich RNA models (including a known bistable Qd/Hp RNA and two G‐rich regions of mRNA of human prion protein, PrP) and applied for the 19F NMR spectroscopic characterization of plausible G‐quadruplex/hairpin (Qd/Hp) transitions in these RNA structures. For the synthesis of the CF3‐labeled RNAs, phosphoramidite building blocks of 2′‐O‐[(4‐CF3‐triazol‐1‐yl)methyl] nucleosides (cytidine, adenosine, and guanosine) were prepared and used as an integral part of the standard solid‐phase RNA synthesis. The obtained 19F NMR spectra supported the usual characterization data (obtained by UV‐ and CD‐melting profiles and by 1H NMR spectra of the imino regions) and additionally gave more detailed information on the Qd/Hp transitions. The molar fractions of the secondary structural species (Qd, Hp) upon thermal denaturation and under varying ionic conditions could be determined from the intensities and shifts of the 19F NMR signals. For a well‐behaved Qd/Hp transition, thermodynamic parameters could be extracted.  相似文献

The 13C? 13C spin coupling constants have been determined in substituted [1-13C]tetrahydronaphthylenes, [5-13C]hexahydrobenzanthracenes and [5-13C]benzanthracene. In addition, the 13C? 13C spin coupling constants for 7-hydroxy[7-13C]benzopyrene, trans-7,8-dihydro[7-13C]benzopyrene-7,8-diol and trans-7,8-dihydro[10-13C]benzopyrene-7,8-diol are reported, together with the one-bond carbon-carbon coupling constants between C-4 and C-5 in selected 4,5-disubstituted benzopyrenes. Values for the directly bonded coupling constants and long-range coupling constants are similar to those reported previously for other aromatic and aliphatic systems. Substituent effects on carbon-carbon coupling are compared for similarly substituted cyclic and acyclic systems.  相似文献

NMR relaxation measurement of perfluorocarbons (PFCs), such as perfluorotributylamine (FTBA), is a convenient method for the determination of oxygen concentrations in tissues and tumors. Previous relaxation studies of FTBA used different 19F NMR assignments causing some confusion. Fluorine‐detected 19F, 13C HMQC and HMBC and selectively 19F‐decoupled 13C NMR provided unequivocal 19F and 13C assignments for FTBA and perfluoropentanoic acid (FPA). Based on those assignments, 13C spin–lattice relaxation time constants (T1) and effective correlation times for FTBA and FPA are reported and discussed. As part of our ongoing research for molecular structures binding to the translocator protein (TSPO 18 kDa), we investigated the preparation of a number of new 2,4,5‐trisubstituted pyridines as novel building blocks. In particular, 5‐amino‐2‐halo‐4‐phenoxypyridines ( 11 , 12 , 13 ) were designed as key intermediates for the synthesis of the recently developed TSPO ligand 6‐F‐PBR28 and its fluorine‐18‐labeled version for positron emission tomography, 6‐[18F]F‐PBR28. We hereby report the chemical preparation as well as the 1H and 13C NMR spectroscopic data of polysubstituted pyridines 2 , 3 , 4 , 5 , 6 , 7 , 7b , 8 , 9 , 10 , 11 , 12 , 13 , 14 . The latter demonstrates dramatic changes in electron density repartition of the aromatic ring upon substitution.  相似文献

We report through‐space (TS) 19F–19F coupling for ortho‐fluoro‐substituted Z ‐azobenzenes. The magnitude of the TS‐coupling constant (TSJFF) ranged from 2.2–5.9 Hz. Using empirical formulas reported in the literature, these coupling constants correspond to non‐bonded F–F distances (dFF) of 3.0–3.5 Å. These non‐bonded distances are significantly smaller than those determined by X‐ray crystallography or density functional theory, which argues that simple models of 19F–19F TS spin–spin coupling solely based dFF are not applicable. 1H, 13C and 19F data are reported for both the E and Z isomers of ten fluorinated azobenzenes. Density functional theory [B3YLP/6‐311++G(d,p)] was used to calculate 19F chemical shifts, and the calculated values deviated 0.3–10.0 ppm compared with experimental values. We have accomplished the synthesis of 13C‐labeled tautomycin at the C18, C19, C21, and C22 positions starting from 100 % [13C]triethylphosphonoacetate for the purpose of elucidating the dynamics and conformation of the C17–C26 moiety. NMR spectroscopy of 13C‐labeled tautomycin revealed strong binding with protein phosphatase type 1 and new features in the 13C NMR spectrum, such as the very small three‐bond coupling constants (2J).  相似文献

We report the 1H NMR and 13C NMR chemical shifts and J(H,H), J(H,F) and J(C,F) coupling constants of 13 2,4‐diamino‐10‐methylpyrimido[4,5‐b]‐5‐quinolone derivatives, some of them with moderate activity against Plasmodium falciparum in vitro. Nearest‐neighbor chain packing in a homogeneous blend of carbonate 13C‐labeled bisphenol A polycarbonate and CF3‐labeled bisphenol A polycarbonate has been characterized using a shifted‐pulse version of magic‐angle spinning 13C{19F} rotational‐echo double‐resonance (REDOR) NMR. Complementary NMR experiments have also been performed on a polycarbonate homopolymer containing the same 13C and 19F labels. In the blend, the 13C observed spin was at high concentration, and the 19F dephasing or probe spin was at low concentration. In this situation, an analysis in terms of a distribution of isolated heteronuclear pairs of spins is valid. A comparison of the results for the blend and homopolymer defines the NMR conditions under which higher concentrations of probe labels can be used and a simple analysis of the REDOR results is still valid. The nearest neighbors of a CF3 on one chain generally include a carbonate group on an adjacent chain. A direct interpretation of the REDOR total dephasing for the polycarbonate blend indicates that at least 75% of carbonate‐carbon 13C ··· F3 nearest neighbors are separated by a narrow distribution of distances 4.7 ± 0.3 Å. In addition, analysis of the variations in REDOR spinning‐sideband dephasing shows that most of the 13C ··· F3 dipolar vectors have a preferred orientation relative to the polycarbonate mainchain axis. This combination of distance and orientational constraints is interpreted in terms of local order in the packing of the carbonate group of one polycarbonate chain relative to the isopropylidene moiety in a neighboring chain. © 2006 Wiley Periodicals, Inc. Molecules labeled with fluorine‐18 are used as radiotracers for positron emission tomography. An important challenge is the labeling of arenes not amenable to aromatic nucleophilic substitution (SNAr) with [18F]F?. In the ideal case, the 18F fluorination of these substrates would be performed through reaction of [18F]KF with shelf‐stable readily available precursors using a broadly applicable method suitable for automation. Herein, we describe the realization of these requirements with the production of 18F arenes from pinacol‐derived aryl boronic esters (arylBPin) upon treatment with [18F]KF/K222 and [Cu(OTf)2(py)4] (OTf=trifluoromethanesulfonate, py=pyridine). This method tolerates electron‐poor and electron‐rich arenes and various functional groups, and allows access to 6‐[18F]fluoro‐L ‐DOPA, 6‐[18F]fluoro‐m‐tyrosine, and the translocator protein (TSPO) PET ligand [18F]DAA1106.  相似文献

The origin of broadening of 13C(carborane) NMR signals of 1,2‐, 1,7‐ and 1,12‐dicarba‐closo‐dodecaboranes(12) and several diphenylsilyl derivatives has been examined in detail and could be traced only partially to unresolved 13C–11B spin‐spin coupling. Other contributions to the line widths arise from 13C–1H dipole‐dipole interactions and, in particular, from isotope‐induced chemical shifts 1Δ10/11B(13C), observed here for carboranes for the first time. In the case of 1‐diphenylsilyl‐1,2‐dicarba‐closo‐dodecaborane(12), the coupling constant 1J(13C,13C) = 9.3 Hz was measured in natural abundance of 13C. The small value of this coupling constant and its negative sign is predicted by calculations based on optimised structures [B3LYP/6‐311+G(d,p) level of theory] of the parent carboranes and 1‐silyl‐1,2‐dicarba‐closo‐dodecaborane(12) as a model compound [calcd. 1J(13C,13C) = –10.5 Hz]. Calculated coupling constants 1J(13C,11B) are small (<7 Hz), in contrast to published assumptions, and of either sign, whereas 1J(11B,11B) are all positive and range up to 15 Hz.  相似文献

Some monomer model compounds of lignin have been selectively 2H and 13C labelled: vanillin, ethyl ferulate, coniferyl alcohol and ethyl hydrogen malonate. Deuterium isotope effects on the 13C chemical shifts in [formyl-2H]vanillin, [5-2H]vanillin and [α,α,5-2H3]coniferyl alcohol made the unambiguous assignment of the aromatic 13C signals possible. Absolute 1,2,3J(CC) values have been determined on 13C spectra of [formyl-13C]vanillin, and of ethyl ferulate and coniferyl alcohol in which the vinylic C-γ and C-β carbons were 13C enriched. It has been possible to measure 4J(C?O, C-4) in vanillin and 4J(C-γ, C-4) in ethyl ferulate. The determination of 1,2,3,4J (CH) absolute values was done by means of gated decoupled 13C spectra of the non-labelled compounds. When second order effects made the use of this technique impossible we determined certain J(CH) values and their signs either by analysing the 1H NMR spectra of 13C labelled coniferyl alcohol [2J(C-β, H-γ), 2J(C-β, H-α), 2J(C-γ, H-β), 3J(C-γ, H-α)] or by a double irradiation experiment on the 250 MHz 1H NMR spectrum of ethyl [β-13C] ferulate [for 2J(C-β, H-γ)].  相似文献

By using 13C MAS NMR spectroscopy (MAS=magic angle spinning), the conversion of selectively 13C‐labeled n‐butane on zeolite H‐ZSM‐5 at 430–470 K has been demonstrated to proceed through two pathways: 1) scrambling of the selective 13C‐label in the n‐butane molecule, and 2) oligomerization–cracking and conjunct polymerization. The latter processes (2) produce isobutane and propane simultaneously with alkyl‐substituted cyclopentenyl cations and condensed aromatic compounds. In situ 13C MAS NMR and complementary ex situ GC–MS data provided evidence for a monomolecular mechanism of the 13C‐label scrambling, whereas both isobutane and propane are formed through intermolecular pathways. According to 13C MAS NMR kinetic measurements, both pathways proceed with nearly the same activation energies (Ea=75 kJ mol?1 for the scrambling and 71 kJ mol?1 for isobutane and propane formation). This can be rationalized by considering the intermolecular hydride transfer between a primarily initiated carbenium ion and n‐butane as being the rate‐determining stage of the n‐butane conversion on zeolite H‐ZSM‐5.  相似文献
