A high‐throughput micro‐solid‐phase extraction device based on a 96‐well plate was constructed and applied to the determination of pesticide residues in various apple samples. Butyl methacrylate and ethylene glycol dimethacrylate were copolymerized as a monolithic polymer and placed in the cylindrically shaped stainless‐steel meshes of 96‐micro‐solid‐phase extraction device and used as an extracting unit. Before the micro‐solid‐phase extraction, microwave‐assisted extraction was employed to facilitate the transfer of the pesticide residues from the apple matrix to liquid media. Then, 1 mL of the aquatic samples was transferred into the 96‐well plate and the 96‐micro‐solid‐phase extraction device was applied for the extraction of the selected pesticides. Influential parameters, such as sorbent‐to‐sorbent reproducibility, microwave‐assisted extraction time, ionic strength and micro‐solid‐phase extraction time, were optimized. The limits of quantitation were below 120 μg/kg, which are lower than the maximum residue limits. The developed method was successfully implemented for the extraction and determination of the selected pesticides from 20 different apple samples gathered from local markets. Phosalone was identified and quantified at the concentration level of 147 (±16.4) μg/kg in one of the samples.  相似文献

A rapid, sensitive, and widely applicable method for the simultaneous quantitative analysis of 20 underivatized amino acids in different biological matrices, including serum, plasma, and tissue homogenates, using ultra high performance liquid chromatography with tandem mass spectrometry was developed and validated. Only 4 µL of serum, plasma, or tissue homogenate was extracted with 996 µL of solution (1.7 mM ammonium formate in 85% acetonitrile containing 0.1% formic acid) containing 100 ng/mL phenylalanine‐d5 as an internal standard without any further derivatization step. In addition, the matrix effects were small because a large volume of extraction solution was used. The total run time including reequilibration was 13 min. The results of linearity, accuracy, repeatability, precision, limits of detection, limits of quantification, and sample stability were sufficient to allow the measurement of the amino acids in different biological matrices. We conclude that our method is rapid, sensitive, and widely applicable and represents an improvement over other currently available technologies.  相似文献

As a result of the low water content and high fat matrices in nuts, it is very difficult to simultaneously determine multi‐pesticides in trace levels. Here, a sample pretreatment method was developed in which, microwave‐assisted solvent extraction was firstly used to extract pesticides, and then a two‐step cleanup method was conducted combining freeze‐out with dispersive solid‐phase extraction to remove the lipidic matrix. By this way, 106 pesticides were simultaneously determined in the complicated nut sample by using an ultra‐high pressure liquid chromatography coupled with a tandem mass spectrometer. Average recoveries were 75.3–119.3% with relative standard deviations < 14% at three concentration levels. The limits of detection and quantification were in the ranges of 0.3–3.0 and 1.0–10.0 μg/kg, respectively. Furthermore, the method was successfully applied to the determination of pesticides in 180 commercial nut samples.  相似文献

Detection of pesticide residues in food samples is important for safeguarding food quality and safety. Conventional approaches for detection of pesticides in food samples typically involve labour‐intensive and time‐consuming sample pretreatment and chromatographic separation. In this study, solid phase micro‐extraction fibres were used to rapidly extract and enrich pesticides in honey, a popular agricultural product with complex matrix, and then directly coupled with electrospray ionization mass spectrometry for qualitative and quantitative analysis. Three pesticides, ie, atrazine, benalaxyl, and pirimicarb, were investigated using the technique and their analytical performances were evaluated. The limits of detection and limits of quantitation of all the three pesticides could fulfil the cut‐off values of the international standard. Linear calibration curves were constructed with good R2 coefficients, and the accuracy and precision were in acceptable ranges for all the pesticides. The analysis time is much reduced, with only minimum sample preparation and no chromatographic separation involved. The technique is simple and easy to set up, and can be extended for analysis of other analytes and sample systems.  相似文献

A high‐throughput, rapid, and efficient modified QuEChERS (quick, easy, cheap, effective, rugged, and safe) method with a simple cleanup procedure has been developed for simultaneously determining 227 pesticides in pepper samples by liquid chromatography with tandem mass spectrometry (running time: 10 min). Pesticide residues were extracted/partitioned with an acetonitrile/DisQuE QuEChERS pouch, and the resulting samples were cleaned up with different methods: dispersive solid‐phase extraction with primary secondary amines or multiwalled carbon nanotubes and graphitized carbon solid mini cartridge column. The results indicated that multiwalled carbon nanotubes dispersive sorbents achieved the best recoveries and had less matrix interference. The numbers of pesticides with a recovery in the range of 70–120% were 199 at a spiked level of 40 μg/kg. The correlation coefficients (r2) for 227 pesticides were above 0.99, while the limits of quantitation of pesticides in pepper samples ranged from 0.13 to 13.51 μg/kg (S/N = 10), and the limits of detection ranged from 0.04 to 4.05 μg/kg (S/N = 3). The relative standard deviations of approximately 197 pesticides were below 20% at spiked levels of 40 μg/kg. Based on these results, the proposed method was chosen as the most suitable cleanup procedure for the determination of multiresidue pesticides in pepper samples.  相似文献

In this study, an effective speed‐regulated directly suspended droplet microextraction method was developed to condense pesticide residues from teas through dispersive solid‐phase extraction prior to analysis by gas chromatography with tandem mass spectrometry. The extractant was intentionally dispersed into the sample solution in the form of globules through high‐speed agitation. This procedure increases the contact area between the binary phases and shortens the distribution equilibrium time. The fine globules reassembled by decelerating stirring speed, the extractant could be taken out for gas chromatography with tandem mass spectrometry. Recovery studies were performed under optimized extraction conditions by using matrix blanks fortified with pesticides at three concentrations (10, 50, and 100 µg/kg). Over 87% of the recoveries for the analytes in four tea matrices were acceptable given their recovery ranges of 70–120% and relative standard deviations of ≤20%. The limits of quantification of most pesticides were lower than 10 µg/kg and thus satisfied the requirements for maximum residue levels prescribed by the European Community. A total of 38 tea samples from local markets were analyzed by using the proposed method. Results showed that chlorpyrifos was the most frequently detected pesticide in teas. The method is a potential choice for the routine monitoring of pesticide residues in complex matrices.  相似文献

A modified quick, easy, cheap, effective, rugged, and safe method was established for simultaneous extraction and cleanup of multiple antibiotics in leafy vegetables, and ultra‐high performance liquid chromatography with tandem mass spectrometry was used for analysis. Antibiotics in leafy vegetables were extracted with citric acid/sodium citrate in mixed solvents consisting of acetonitrile/methanol (85:15, v/v) from 10 g of vegetables. Octadecylsilyl and graphitized carbon black were used as dispersant adsorbents. This method was able to effectively extract all of the target antibiotics from leafy vegetables. The average recoveries of 20 antibiotics ranged from 57 to 91%. The limits of detection were 0.33–2.92 μg/kg. The developed method subsequently demonstrated its selectivity, sensitivity, and reliability for detecting multiple antibiotics in 15 samples. Antibiotic residues in vegetables have attracted great concern with respect to human health. It is recommended that standards should be established for antibiotic residues in vegetables to ensure food safety.  相似文献

A multi‐pesticide residue determination method based on a modified QuEChERS (quick, easy, cheap, effective, rugged, and safe) method using multiwalled carbon nanotubes as reversed‐dispersive solid‐phase extraction material was validated in 37 representative pesticides in tobacco. Determination was performed using liquid chromatography with tandem mass spectrometry in multiple reaction monitoring mode. Three major types of tobacco leaf samples, namely, flue‐cured, burley, and oriental tobacco were studied and compared. Three factors (extraction time, external diameter, and amount of extraction material used) that could affect the performance of multi‐walled carbon nanotubes were investigated. Optimization of sample preparation and determination allowed recoveries between 70.8 and 114.8% for all 37 pesticides with < 20.0% relative standard deviations at three spiking levels of 20, 50, and 200 μg/kg. The limits of quantification and limits of detection for the 37 pesticides ranged within 0.46–28.57 and 0.14–8.57 μg/kg at a signal‐to‐noise ratio of 10 and 3, respectively.  相似文献

Abstract

A new analytical method is proposed for determining residues of 70 pesticides of different chemical families at parts per trillion levels in fresh vegetables. For that, only 4 g of the vegetable samples were quickly extracted with 10 ml of ethyl acetate. The method is based on a vanguard/rearguard strategy that reduces the average time required per sample when the method is applied to a high number of vegetable samples in a quality control laboratory. At the beginning, an aliquot of the extract is evaporated and re‐dissolved in a mixture water:acetone (9∶1 v/v). For screening purposes, the pesticides were extracted for only 10 min by direct immersion of a solid‐phase microextraction (SPME) fiber (65 µm polydimethylsiloxane‐divinylbenzene, PDMS‐DVB). The SPME device was automated and on‐line coupled to a gas chromatograph with an ion trap mass spectrometer (GC‐MS) operated in full scan mode for screening in less than 18 min those samples that potentially contain pesticides above 0.01 mg kg?1 (cut off value). After that, only those potentially non‐negative samples were reanalyzed by a sensitive quantifying/confirming method that re‐extract by SPME the pesticides in 55 min of absorption and determine them by GC with tandem MS (MS/MS). The method has been validated following EU guidelines and compared with a conventional extraction method based on the use of higher amounts of organic solvents. The limits of detection (LOD), confirmation (LOC) and quantitation (LOQ) as well as the calibration curves obtained allowed the determination of the target pesticides at concentrations clearly below the maximum residue levels (MRL) stated by EU being possible the determination of parts per trillion of the pesticides in ecological (green) vegetables. The method has been applied to the analysis of real samples and the results compared with those obtained by a conventional extraction method accredited by ENAC (Spanish Accreditation Body). The proposed method was also evaluated participating in a proficiency test with adequate results (z‐score among±2).  相似文献

The AOAC 2007.1 quick, easy, cheap, effective, rugged, and safe official method, together with gas chromatography coupled to a triple quadrupole mass spectrometer was applied for the analysis of 38 multiclass pesticides from dried fruits typically cultivated and exported from Colombia: uchuva (Physalis peruviana), lulo (Solanum quitoense), guanabana (Anona muricata), and pitahaya (Hylocereus undatus). The whole method was validated in terms of matrix‐matched calibration, matrix effect, and recovery using atrazine‐d5 as internal standard, triphenylphosphate for quality control of the injection, and a proper mixture of analytes protectants. Matrix‐matched calibration data were found satisfactory for all pesticides and dried fruits, reporting R2 values above 0.99. Matrix effect values evidenced the existence of such effect in most cases. The applied procedure gave satisfactory recovery percentages (70–120%) and relative standard deviation values (< 20%) for 92% of the 456 combinations pesticide/fruit studied (spiked levels of 25, 200, and 400 µg/kg). Finally, 20 real dried fruit samples were analyzed and residues of tebuconazole were found in two samples of uchuva at a concentration below the lowest calibration level of the method for one of them and at 10.8 ± 1.6 µg/kg for the other, being below or similar to the general maximum residue level established for monitoring purposes in food applications.  相似文献

This work reports the development and application of a multi‐class compound analysis method for the determination of 20 antibiotic residues in compost. Samples were processed by microwave‐enhanced accelerated solvent extraction at 120°C for 7.5 min. Salting‐out homogeneous liquid‐liquid extraction was used to remove water and water‐soluble impurities from the extract before ultra performance convergence chromatography with tandem mass spectrometry analysis. By using the supercritical fluid (carbon dioxide) and organic solvent (methanol) as the mobile phase, the 20 antibiotics and the internal standard were well separated in 8.2 min without obvious matrix effect. Method validation was performed and good trueness (relative error in the range of ±5.0%) and precision (inter‐ and intraday relative standard deviations < 10.8%) were obtained. Method detection and quantitation limits were 0.8–1.9 and 2.7–7.1 ng/g, respectively. Recoveries were assessed at three concentration levels (10, 60, and 400 ng/g) and acceptable mean values (70.4–111.9%) were found. This method has also been used to analyze real samples, and the average concentrations of antibiotics (excepting the concentrations < method quantitation limits) were determined up to 123.6 ng/g. The results showed the method could be helpful for the analysis of multi‐class antibiotics in environmental samples.  相似文献

A novel procedure is put forward based on the combination of the well‐established matrix solid‐phase dispersion and the magnetic and sorption properties of magnetic octadecyl in the presence of n‐octanol and was employed in a proof‐of‐concept sample preparation and determination of several classes of pesticide residues in carrots. The procedure does not require the transfer of blend to cartridge and subsequent packing, nor any co‐sorbent for extract clean up. The hydrophobic magnetic nanoparticles utilized as a sorbent, can be retrieved by n‐octanol under the application of a magnetic field due to hydrophobic interactions. Elution of pesticide residues is then carried out with an organic solvent. A total of 26 pesticides were included in this procedure and the target compounds were analyzed using gas chromatography with mass spectrometry in the selective ion monitoring mode. The average extraction recoveries obtained from carrot samples fortified at three different concentrations (20, 50, and 500 μg/kg) were 77–107%. The estimated limits of quantitation for most target analytes were in the low μg/kg level. The study demonstrates that the proposed extraction procedure is simple and effective, avoiding a clean‐up step for the sample preparation of vegetable.  相似文献

An ion mobility quadrupole time‐of‐flight mass spectrometry‐based pesticide suspect screening methodology was developed and validated covering 20 plant‐derived food matrices deriving from six commodity groups of different complexity according to the actual European Commission document SANTE/11813/2017 applying a QuEChERS sample preparation protocol. The method combines ultra‐performance liquid chromatography, traveling wave ion mobility, and quadrupole time‐of‐flight mass spectrometry. Besides the determination of the physicochemical property collision cross‐section and the establishment of a corresponding scientific suspect screening database comprising 280 pesticides for several pesticides, different protomers, sodium adducts, as well as dimers were identified in ion mobility spectrometry traces. Additionally, collision cross‐section values were included in the validation requirements regarding chromatography and mass spectrometry for the detection of pesticides. A collision cross‐section value window was analyzed within a tolerable error of ±2%. For this cross‐matrix validation, screening detection limits were determined at concentration levels of 0.100 mg/kg (84% of the original pesticide scope), 0.010 mg/kg (56%), and 0.001 mg/kg (21%). By application of ion mobility spectrometry, the compound identification was improved due to independence of commodity of concern and concentration levels of analyte molecules, as false assignments are reduced by application of a collision cross‐section range.  相似文献

Simple and highly efficient sample preparation procedures, namely, dispersive liquid–liquid microextraction and salting‐out liquid–liquid extraction for the analysis of ten Fusarium mycotoxins and metabolites in human urine were compared. Various parameters affecting extraction efficiency were carefully evaluated. Under optimal extraction conditions, salting‐out liquid–liquid extraction showed a better accuracy (84–96%) and precision (<14%) than dispersive liquid–liquid microextraction. Hence, a multibiomarker method based on salting‐out liquid–liquid extraction followed by gas chromatography with tandem mass spectrometry was proposed. Satisfactory results in terms of validation were achieved. The method resulted in low limits of detection and quantitation within the range of 0.12–4 and 0.25–8 μg/L, respectively. The method accuracy and precision were evaluated at three spiking levels (8, 25 and 100 μg/L) and the recoveries were in a range from 70 to 120% with relative standard deviations lower than 15%. Matrix effect was evaluated and matrix‐matched calibrations were used for quantitation purpose. The developed method was applied in 12 human urine samples as a pilot study before and after sample treatment with β‐glucuronidase before the analysis to quantify the mycotoxin conjugates. Total deoxynivalenol (free + conjugated) was found in 83% of samples at an average concentration in positive samples of 31.6 μg/L.  相似文献

A simple and efficient liquid chromatography‐mass spectrometry (LC‐MS) method was developed and validated for simultaneous quantitation of catalpol and harpagide in normal and diabetic rat plasma. Protein precipitation extraction with acetonitrile was carried out using salidroside as the internal standard (IS). The LC separation was performed on an Elite C18 column (150 × 4.6 mm, 5 µm) with the mobile phase consisting of acetonitrile and water within a runtime of 12.0 min. The analytes were detected without endogenous interference in the selected ion monitoring mode with positive electrospray ionization. Calibration curves offered satisfactory linearity (r > 0.99) at linear range of 0.05–50.0 µg/mL for catalpol and 0.025–5.0 µg/mL for harpagide with the lower limits of quantitation of 0.05 and 0.025 µg/mL, respectively. Intra‐ and inter‐day precisions (RSD) were <9.4%, and accuracy (RE) was in the ?6.6 to 4.9% range. The extraction efficiencies of catalpol, harpagide and IS were all >76.5% and the matrix effects of the analytes ranged from 86.5 to 106.0%. The method was successfully applied to the pharmacokinetic study of catalpol and harpagide after oral administration of Zeng‐Ye‐Decoction to normal and diabetic rats, respectively. An easy, effective and sensitive analytical method for the simultaneous determination of a novel fungicide pyrametostrobin and its two metabolites pyrametostrobin‐M1 and pyrametostrobin‐M2 in cucumber and soil was developed using a quick, easy, cheap, effective, rugged, and safe method with high‐performance liquid chromatography and tandem mass spectrometry. The extraction solvent was acetonitrile, and cleanup sorbents were primary secondary amine and graphitized carbon black for cucumber samples and primary secondary amine for soil samples. The three target compounds were successfully separated between 3.2 and 3.9 min using a Waters CORTECS™ C18 column connected to an electrospray ionization source. All the matrix‐matched samples at three fortified levels (10, 100 and 1000  μg/kg) provided satisfactory recoveries in the range of 78.8–93.8% with relative standard deviations below 6.9%. The limits of quantitation for the three compounds were below 0.183 μg/kg. The proposed method was validated by analyzing real samples.  相似文献

A simple and efficient method for the determination of 28 carbamates in high‐fat cheeses is proposed. The methodology is based on a modified quick, easy, cheap, effective, rugged, and safe procedure as sample treatment using a new sorbent (Z‐Sep+) followed by ultra‐high performance liquid chromatography with tandem mass spectrometry determination. The method has been validated in different kinds of cheese (Gorgonzola, Roquefort, and Camembert), achieving recoveries of 70–115%, relative standard deviations lower than 13% and limits of quantification lower than 5.4 μg/kg, below the maximum residue levels tolerated for these compounds by the European legislation. The matrix effect was lower than ±30% for all the studied pesticides. The combination of ultra‐high performance liquid chromatography and tandem mass spectrometry with this modified quick, easy, cheap, effective, rugged, and safe procedure using Z‐Sep+ allowed a high sample throughput and an efficient cleaning of extracts for the control of these residues in cheeses with a high fat content.  相似文献

A new simple and rapid pretreatment method for simultaneous determination of 19 sulfonamides in pork samples was developed through combining the QuEChERS method with dispersive liquid–liquid microextraction followed by ultra‐high performance liquid chromatography with tandem mass spectrometry. The sample preparation involves extraction/partitioning with QuEChERS method followed by dispersive liquid–liquid microextraction using tetrachloroethane as extractive solvent and the acetonitrile extract as dispersive solvent that obtained by QuEChERS. The enriched tetrachloroethane organic phase by dispersive liquid–liquid microextraction was evaporated, reconstituted with 100 μL acetonitrile/water (1:9 v/v) and injected into an ultra‐high performance liquid chromatography with a mobile phase composed of acetonitrile and 0.1% v/v formic acid under gradient elution and separated using a BHE C18 column. Various parameters affecting the extraction efficiency were investigated. Matrix‐matched calibration curves were established. Good linear relationships were obtained for all analytes in a range of 2.0–100 μg/kg and the limits of detection were 0.04–0.49 μg/kg. Average recoveries at three spiking levels were in the range of 78.3–106.1% with relative standard deviations less than 12.7% (n = 6). The developed method was successfully applied to determine sulfonamide residues in pork samples.  相似文献

A high‐performance liquid chromatographic method was developed for the analysis of 3'‐hydroxypterostilbene. This method involves the use of a Luna® C18 column with ultraviolet detection at 325 nm. The mobile phase consisted of acetonitrile, water and formic acid (50:50:0.01, v/v/v) with a flow rate of 0.8 mL/min. The calibration curves were linear over the range 0.5–100.0 µg/mL. The mean extraction efficiency was between 97.40 and 111.16%. The precision of the assay was 0.196–14.39% (RSD%), and within 15% at the limit of quantitation (0.5 µg/mL). The bias of the assay was <16% and within 15% at the limit of quantitation. This assay was successfully applied to pre‐clinical pharmacokinetic samples from rat urine and serum. A syringe‐dispersive solid‐phase extraction method was developed for the determination of seven nitroimidazoles and nine steroids in manure‐based fertilizers by ultra‐high performance liquid chromatography with tandem mass spectrometry. Methanol and acetonitrile were used to extract the sample, and mixed dispersive sorbents dispersed in the syringe were used for purification. The extract was separated with an HSS‐T3 column and detected in positive or negative multiple reaction monitoring mode. Under the optimal conditions, the recoveries of the 16 compounds ranged from 70.3 to 112.3% at the four spiked levels (3, 10, 20, and 50 μg/kg) and the relative standard deviations ranged from 1.0 to 12.4%. The limits of detection and quantification were 0.22–0.86 and 0.73–2.87 μg/kg, respectively. This method is simple, fast, and reliable, and can be used to simultaneously screen and determine nitroimidazoles and steroids in manure‐based fertilizers.  相似文献
