建立了高效毛细管电泳-电喷雾飞行时间质谱联用(HPCE-ESI-TOF/MS)快速定性分析黄连中生物碱类化合物的分析方法. 使用未涂层石英毛细管, 以50 mmol/L乙酸铵-0.5%甲醇溶液(用氨水调至pH=7.2)作为运行缓冲液, 分离电压为25 kV; 鞘液组成为50%甲醇-49.5%水-0.5%乙酸, 鞘液流速为4 μL/min; 质谱选用正离子模式, 碰撞电压(Fragmentor)为100 V. 结果表明, 通过各色谱峰紫外光谱和质谱测得精确分子量结果, 结合文献, 对黄连中7种生物碱进行了鉴定. 表明本方法简便、快速, 是黄连中生物碱类化合物快速分离、鉴别的有效方法.  相似文献

The current widely utilized polymer or C8, C18 end-capped material-based sorbents for solid-phase extraction could not capture alkaloids well only based on “like dissolves like” principle. In this paper, a layer-by-layer functionalized porous Zinc sulfide nanospheres-based solid-phase extraction (SPE) combined with liquid chromatography time-of-flight/mass spectrometry (LC-TOF/MS) and gas chromatography-mass spectrometry (GC-MS) was developed for the specific enrichment and identification of alkaloids from complex matrixes, Crinum asiaticum var. sinicum crude extracts. The functionalized porous Zinc sulfide nanospheres were prepared by the amidation reaction of poly-(acrylic acid) (PAA) homopolymer with amino groups onto the porous ZnS nanospheres. Tandem LC-TOF/MS spectrometry presented that the almost all of the twenty-three main peaks in elution fraction from the SPE could be inferred as alkaloids with ion of mass according to the nitrogen rule and hit formula with Peak View1.2@software from AB SCIEX, and seven alkaloids including two new found chemical entities were directly identified from their GC-MS spectra and retention indices. We believe that this SPE protocol can also be utilized in the future to selectively enrich alkaloids from extracts of other plant species.  相似文献

A new method based on accelerated solvent extraction (ASE) followed by a reliable high-performance liquid chromatography-diode array detector (HPLC-DAD) and positive ion electrospray-time of flight mass spectrometry (ESI-TOF/MS) analysis has been developed for the characterization and quantification of four major saponins in extracts of the seeds of Aesculus chinensis Bunge (Semen Aesculi). The saponins escin Ia, escin Ib, isoescin Ia and isoescin Ib were extracted from seeds of A. chinesis Bunge via ASE, and the operational parameters of ASE were optimized, such as extraction solvent, extraction temperature, static extraction time and extraction cycles. The optimized procedure employed 70% MeOH as extraction solvent, 120 °C of extraction temperature, 7 min of static extraction time, 60% flush volume and the extraction recoveries of the four compounds were nearly to 100% for two cycles. The HPLC conditions are as follows: SinoChrom ODS BP C18 (4.6 mm × 200 mm, 5 μm) column, acetonitrile and 0.10% phosphoric acid solution as mobile phase, flow rate is 1.0 mL min−1, detection length of UV is 203 nm, injection volume is 10 μL. The results indicated that the developed HPLC method is simple, sensitive and reliable for the determination of four major saponins in seeds of A. chinesis Bunge with a good linearity (r2 > 0.9994), precision (relative standard deviation (R.S.D.) <1.5%) and the recovery ranges of 95.2-97.3%. The limits of detection (LOD) of the four compounds were in the range of 0.40-0.75 μg mL−1. This assay can be readily utilized as a quality control method for Semen Aesculi and other related medicinal plants.  相似文献

Tussilago farfara (Kuan Donghua) is an important Chinese herbal medicine which has been shown to contain many bioactive compounds and widely used to relieve cough and resolve phlegm. However, besides therapeutic bioactive compounds, this herb has been found to contain toxic pyrrolizidine alkaloids (PAs), mainly senkirkine and traces of senecionine. In this report, conditions for microwave-assisted extraction (MAE) and pressurized hot water extraction (PHWE) were optimized for the extraction of the PAs. The results were compared against heating under reflux. It was found that the binary mixture of MeOH:H2O (1:1) acidified using HCl to pH 2-3 was the optimal solvent for the extraction of the PAs in the plant materials. Liquid chromatography (LC) with ultra-violet (UV) detection and electrospray ionization mass spectrometry (ESI-MS) in the positive mode was used for the determination and quantitation of senkirkine and senecionine in the botanical extract. The proposed extraction methods with LC/MS allow for the rapid detection of the major and the minor alkaloids in T. farfara in the presence of co-eluting peaks. With LC/MS, the quantitative analysis of PAs in the extract was done using internal standard calibration and the precision was found to vary from 0.6% to 5.4% on different days. The limits of detection (LODs) and limits of quantitation (LOQs) for MAE and PHWE were found to vary from 0.26 μg/g to 1.04 μg/g and 1.32 μg/g to 5.29 μg/g, respectively. The method precision of MAE and PHWE were found to vary from 3.7% to 10.4% on different days. The results showed that major and minor alkaloids extracted using MAE and PHWE were comparable to that by heating under reflux. Our data also showed that significant ion suppression was not observed in the analysis of senkirkine and senecionine in the botanical extracts with co-eluting peaks.  相似文献

The objective of this work is to investigate the influence of process parameters on the pressurized liquid extraction (PLE) of Ilex paraguariensis leaves. A factorial 26−2 experimental design was employed using responses as the extraction yield and the chromatographic profile of the extracts. The extraction time, polarity of solvent, amount of sample, numbers of PLE cycles, flushing volume and extraction temperature were selected as independent variables (factors). Results obtained indicated that the solvent polarity was the most significant variable in the study, while the amount of sample and extraction temperature also showed significant effect. The other variables did not present significant influence in the yield of extraction. GC/MS analysis of the extract enabled the identification of saturated hydrocarbons, fatty acids, fatty acid methyl esters, phytosterols and theobromine in the extracts. Quantitative analysis of four compounds presented in the extracts (caffeine, phytol, vitamin E and squalene) was performed by the GC/MS in the SIM mode.  相似文献

采用加速溶剂萃取结合超高效液相色谱-串联质谱(UPLC-MS/MS)技术,建立了一种快速提取和测定棉花中8种脱叶剂(噻苯隆、脱叶磷、甲基苯噻隆、脱落酸、氟酮唑草酯、敌草隆、百草枯、嘧草硫醚)的分析方法。样品经提取、浓缩,乙腈-水溶液(1:9, v/v)溶解,采用Acquity UPLC® HSS T3柱(50 mm×2.1 mm, 1.8 μ m)分离,以乙腈-0.05%(v/v)甲酸水溶液为流动相,梯度洗脱,电喷雾正离子模式多反应监测,外标法定量。该方法在0.01~0.3 mg/L范围内线性关系良好(r >0.99),在添加含量水平为0.1、0.5、1.0 mg/kg时,平均回收率范围为(84.18±8.04)%~(95.99±6.76)%,相对标准偏差(RSD)为7.04%~10.60%,方法检出限(LOD)为0.8~29 μ g/kg,方法定量限(LOQ)为2.5~96 μ g/kg。该方法操作简便、快捷、灵敏、准确,适合棉花中8种脱叶剂的确证和定量测定。  相似文献

The extraction and identification of bioactive compounds from herbs is of great interest. In this study, accelerated solvent extraction (ASE) technique was used to analyze the secondary volatile profiles in Nigella sativa seeds obtained from two different origins, Egypt and Bangladesh. The main extraction parameters, including extraction temperature, pressure and static extraction time, were investigated and optimized. Identification and quantification of the major constituents in nonpolar extracts (hexane) were achieved by means of GC‐FID/GC‐MS analysis with external standards. The two seeds showed a similar variety of chemical composition; however, the secondary volatiles profile of Bangladesh seed was higher than that of the Egyptian seed. A total of 25 compounds were identified from the ASE extract under the following optimum extraction conditions: 100°C, 1500 psi and 5 min, for extraction temperature, pressure and static time, respectively. By optimizing the extraction and analytical conditions, a reliable and precise HPLC method coupled with photodiode array detection (HPLC–DAD) has been developed for the identification and quantification of three major aconitine-type alkaloids (aconitine, mesaconitine, hypaconitine) in the roots of Aconitum carmichaelii Debeaux. The qualitative analysis of the plant material was carried out by LC-APCI-MSn. By means of this method, 26 lipo-alkaloids were also identified from the roots of A. carmichaelii. The effect of processing on aconitine-type alkaloids, lipo-alkaloids and pure aconitine was studied. As part of our investigation, two lipo-alkaloids, 14-benzoylaconine-8-palmitate and 14-benzoylaconine-8-linoleate were produced semisynthetically. The COX-1, COX-2 and LTB4 formation inhibitory activity of aconite root extracts and different types of diterpene alkaloids and the toxicity of lipo-alkaloids were also investigated.  相似文献

Ultra-performance liquid chromatography-quadrupole-time-of-flight mass spectrometry (UPLC-Q-TOF-MS) is an effective technique for analysis of complex samples with offering rapid, efficient separation in combination with accurate mass measurement and tandem mass spectrometry (MS/MS). This paper exploits this technique to identify the alkaloids in corydalis yanhusuo, an important antalgic Traditional Chinese Medicine (TCM). The mass spectral fragmentation behavior of one tertiary alkaloid and two quaternary alkaloids was studied in detail. Low-abundance product ions of tertiary and quaternary alkaloids were investigated and compared between each other. Sixteen alkaloids were screened out by using a systematic screening method developed in our laboratory; structures of eight therein were identified by characteristic UV absorption spectrum and positive ion mode of Q-TOF-MS/MS; and two of them were discovered for the first time in corydalis yanhusuo to our knowledge. This research demonstrates the potential of UPLC-Q-TOF-MS in structural characterization and identification of components in traditional Chinese herbal medicines.  相似文献

The total content of polyphenols and flavonoids determined in the same plant and their corresponding antioxidant activities may vary widely, depending on the extraction conditions applied. This study was conducted to optimise the extraction conditions of phenolics and flavonoids from the mistletoe plant. Various extraction methods, i.e. ultrasound-assisted extraction technology, maceration, maceration with stirring, accelerated solvent extraction (ASE), and extraction under reflux were evaluated for their percentage extraction of polyphenols (TPC) and flavonoids (TFC) from Viscum album subsp. abietis. In addition, the anti-radical activity of extracts was analysed using the 2,2-diphenyl-1-picrylhydrazyl method. The effects of temperature, solvent type, and concentration on the phenolic extraction efficiency and antioxidant activity were studied using chemometric and statistical methods. The results showed that the extracts of V. album subsp. abietis contained large amounts of polyphenols and flavonoids (up to 57.673 mg g?1 and 9.955 mg g?1 of dry extract, respectively) and exhibited potent antioxidant activity, hence representing promising sources of powerful antioxidants. Due to its high extraction efficiency and considerable saving of time and solvent, ASE was more effective than the other extraction techniques. Extracts prepared with water-polar solvent mixtures displayed the highest TPC, TFC, and antioxidant activity, while organic polar solvents were the least efficient extractants.  相似文献

Liquid chromatography coupled with electrospray ionization quadrupole time-of-flight tandem mass spectrometry (LC/ESI-QTOF-MS/MS) was performed to study the fragmentation behaviors of steroidal alkaloids from Fritillaria species, the antitussive and expectorant herbs widely used in traditional Chinese medicine. We propose, herein, a strategy that combining key diagnostic fragment ions and the relative abundances and amounts of major fragment ions (the ions exceeding 10% in abundance) to distinguish different sub-classes of Fritillaria alkaloids (FAs). It was found that hydrogen rearrangement and induction effects result in ring cleavage of the basic skeletons occurred in the MS/MS process and produced characteristic fragment ions, which are useful for structural elucidation. This method was finally used to investigate the primary steroidal alkaloids in the extracts of eight major Fritillaria species. As a result, 41 steroidal alkaloids (29 cevanine type, 1 jervine type, 6 veratramine type and 5 secosolanidine type alkaloids) were selectively identified in these Fritillaria species. Twenty-six compounds were unambiguously identified by comparing with the reference compounds and 15 compounds were tentatively identified or deduced according to their MS/MS data. Logical fragmentation pathways for different types of FAs have been proposed and are useful for the identification of these types of steroidal alkaloids in natural products especially when there are no reference compounds available.  相似文献

Rhizoma coptidis, a broadly used traditional Chinese medicine, derives from the dried rhizomes of Coptis chinensis Franch, Coptis deltoidea C.Y. Cheng et Hsiao and Coptis teeta Wall. Quantitative determination of protoberberine alkaloids in R. coptidis is critical for controlling its quality. In this study, a rapid, simple and accurate quantitative 1H NMR (qNMR) method was developed for simultaneous determination of berberine, jatrorrhizine, epiberberine, coptisine, palmatine and columbamine in R. coptidis from the three species. Method validation was performed in terms of selectivity, precision, repeatability, stability, accuracy, robustness and linearity. The average recoveries obtained were in the range of 96.9–102.4% for all the six alkaloids. In addition, the qNMR data were analyzed with analysis of variance (ANOVA), hierarchical clustering analysis (HCA) and principal component analysis (PCA), and the results showed that the contents of the active alkaloids have significant difference among the three species. Compared with the conventional HPLC approach, the proposed qNMR method was demonstrated to be a powerful tool for quantifying the six alkaloids due to its unique advantages of high robustness, rapid analysis time and no need of standard compounds for calibration curves preparation. These findings indicate that this method has potential as a reliable method for quality evaluation of herb medicines, especially for protoberberine alkaloid-containing ones.  相似文献

Alkaloids of the Lycopodiaceae family are of great interest to researchers due to their numerous properties and wide applications in medicine. They play a very important role mainly due to their potent antioxidant, antidepressant effects and a reversible ability to inhibit acetylcholinesterase (AChE) enzyme activity. This property is of immense importance due to the growing problem of an increasing number of patients with neurodegenerative diseases in developed countries and a lack of effective and efficient treatment for them. Numerous studies have shown that Lycopodiaceae alkaloids are a rich source of AChE inhibitors. In the obtaining of new therapeutic phytochemicals from plant material, the extraction process and its efficiency is crucial. Therefore, the aim of this work was to optimize the conditions of modern PLE to obtain bioactive alkaloids from two Lycopodium species: L. clavatum L. and L. annotinum L. Five different solvents of different polarity were used for prepared plant extracts in order to compare the alkaloid content in and thereby effectiveness of the entire extraction. PLE parameters were used based on multiple studies conducted that gave the highest alkaloids recovery. Crude extracts were purified using solid-phase extraction (SPE) on Oasis HLB cartridge and examined by HPLC/ESI-QTOF–MS of the highly abundant alkaloids. To the best of our knowledge, this is the first time such high recoveries have been obtained for known Lycopodiaceae alkaloids. The best extraction results of alkaloid-lycopodine were detected in the dichloromethane extract from L. clavatum, where the yield exceeded 45%. The high recovery of annotinine above 40% presented in L. annotinum was noticed in dichloromethane and ethyl acetate extracts. Moreover, chromatograms were obtained with all isolated alkaloids and the best separation and quality of the bands in methanolic extracts. Interestingly, no alkaloid amounts were detected in cyclohexane extracts belonging to the non-polar solvent. These results could be helpful for understanding and optimizing the best conditions for isolating potent AChE inhibitors.  相似文献

Peanut stems and leaves have been used traditionally as both herbal medicines and special food in Asia. In this study, the main functional compounds of peanut stems and leaves extracts were identified using UPLC separation coupled to high resolution mass spectrometry (QTOF‐MS), and a traditional medicine library. Three different extraction solvents (ethyl acetate, petroleum ether and n‐butanol) were evaluated to prepare the extracts of peanut stems and leaves. A total of 283 chemical compounds were identified in peanut stems and leaves extracts, of which 207 compounds are tentatively new identifications in Genus Arachis. The integration of data acquisition and processing with the traditional medicine library provides a simple, efficient process to effectively facilitate the identification of chemical ingredients in complex natural product extracts. The integrated workflow for separation, detection and identification of functional compounds in natural products using UPLC/QTOF‐MS greatly improves productivity for development of traditional herbal medicines. Modern extraction technique-pressurized liquid extraction (PLE) was optimised for extraction of lycorine and galanthamine (Amaryllidaceae alkaloids) from Narcissus jonquilla ‘Pipit’. Crude extracts were purified on Oasis MCX cartridges, and the alkaloids eluted with 80-100% recoveries using methanol-10% ammonia solution (3:1, v/v). Quantitative results were obtained by both HPTLC-densitometry on silica gel plates and RP-HPLC with diode array (DAD) on XTerra C18 stationary phase. Both methods were fully validated in terms of specificity, precision (including intra- and inter-day measurements), LOD and LOQ values, correlation of UV spectra and linearity of calibration curves. The methods were also well correlated each other with correlation coefficients (r) 0.98823 and 0.99081, respectively, for the mean values of galanthamine and lycorine.Among the investigated solvents methanol and 1% tartaric acid methanolic solution at default conditions (120 °C, p = 60 bar, time: 10 min, one static cycle) permit the highest yields of the total sum of the alkaloids, whereas for toluene the lowest amounts were measured. Lycorine to galanthamine mean ratios were dependant on the type of solvent used, and in toluene galanthamine and related alkaloids were preferably extracted.In temperature experiments for galanthamine, the levels of this compound increased from the temperature of 20 till 150 °C in the investigated solvent systems, then decreased with slight increase from the temperature of 175 to 200 °C in 1% tartaric acid methanolic solution. When lycorine was analysed, similar trends were observed, however the maximum of the concentration was measured at a temperature about 125 °C. The ratios of the mean values of these two compounds differed in temperature-dependant experiments in both solvent systems.Further more, two TLC with bioautography approaches were used in screening for anticholinesterese properties of the extracts. No qualitative differences were found among the different solvent extracts, and AChE inhibition was correlated with galanthamine and related compounds.In conclusion, optimised PLE was much more effective than previously applied hot-solvent extraction, microwave-assisted extraction (MAE) or ultrasound-assisted extraction (USAE).  相似文献

建立了加速溶剂萃取-QuEChERS-超高效液相色谱-串联质谱测定药食同源性食品中16种真菌毒素的方法。样品经过加速溶剂萃取后用QuEChERS方法净化,液相色谱分离,在正、负离子同时扫描和多反应离子监测模式下检测,黄曲霉毒素B1和伏马毒素B1采用内标法定量,其余毒素采用基质外标法定量。在较宽的线性范围内,16种目标化合物的线性相关系数(r2)均大于0.99。该方法的检出限为0.008~0.3 μg/kg,定量限为0.03~1.0 μg/kg,在3个不同添加水平下的加标回收率为70.8%~118%,RSD为2.5%~10.2%。采用建立的方法分别对市面上销售的30个批次的山银花、葛根和沙棘产品进行检测,部分产品检出不同含量的真菌毒素。该方法快速、灵敏,适用于药食同源性食品中多种真菌毒素的同时检测。  相似文献

Terminalia sericea is used throughout Africa for the treatment of a variety of conditions and has been identified as a potential commercial plant. The study was aimed at establishing a high-performance thin layer chromatography (HPTLC) chemical fingerprint for T. sericea root bark as a reference for quality control and exploring chemical variation within the species using HPTLC metabo3lomics. Forty-two root bark samples were collected from ten populations in South Africa and extracted with dichloromethane: methanol (1:1). An HPTLC method was optimized to resolve the major compounds from other sample components. Dichloromethane: ethyl acetate: methanol: formic acid (90:10:30:1) was used as the developing solvent and the plates were visualized using 10% sulfuric acid in methanol as derivatizing agent. The concentrations of three major bioactive compounds, sericic acid, sericoside and resveratrol-3-O-β-rutinoside, in the extracts were determined using a validated ultra-performance liquid chromatography-photodiode array (UPLC-PDA) detection method. The rTLC software (written in the R-programming language) was used to select the most informative retardation factor (Rf) ranges from the images of the analysed sample extracts. Further chemometric models, including principal component analysis (PCA) and hierarchical cluster analysis (HCA), were constructed using the web-based high throughput metabolomic software. The rTLC chemometric models were compared with the models previously obtained from ultra-performance liquid chromatography coupled with mass spectrometry (UPLC-MS). A characteristic fingerprint containing clear bands for the three bioactive compounds was established. All three bioactive compounds were present in all the samples, although their corresponding band intensities varied. The intensities correlated with the UPLC-PDA results, in that samples containing a high concentration of a particular compound, displayed a more intense band. Chemometric analysis using HCA revealed two chemotypes, and the subsequent construction of a loadings plot indicated that sericic acid and sericoside were responsible for the chemotypic variation; with sericoside concentrated in Chemotype 1, while sericic acid was more abundant in Chemotype 2. A characteristic chemical fingerprint with clearly distinguishable features was established for T. sericea root bark that can be used for species authentication, and to select samples with high concentrations of a particular marker compound(s). Different chemotypes, potentially differing in their therapeutic potency towards a particular target, could be distinguished. The models revealed the three analytes as biomarkers, corresponding to results reported for UPLC-MS profiling and thereby indicating that HPTLC is a suitable technique for the quality control of T. sericea root bark.  相似文献

In the present study, the qualitative and quantitative analysis of alkaloids in the stem and root of Sinomenium acutum (S. acutum) is presented by gas chromatography-mass spectrometry (GC–MS) and non-aqueous capillary electrophoresis (NACE). The extract of alkaloids in S. acutum was examined by GC–MS and the major alkaloids were identified. Sinomenine (SIN) was found as the principal alkaloid in the extracts (about 84.38%). The quantitation of SIN was then accomplished by GC–MS and NACE with diode array detection. NACE was selected in order to use a running buffer fully compatible with samples in organic solvent. Optimum separation was achieved with a fused-silica capillary column and a running buffer containing 80 mM ammonium acetate, 2.0% acetic acid and 20% (v/v) acetonitrile in methanol medium. The applied voltage was 22 kV. The different selectivity displayed by these techniques allowed different separation profiles that could be useful in phytochemical characterization of the sample. The GC–MS and NACE methods were successfully validated and applied for the quantitation of SIN in S. acutum.  相似文献

Determination of 16 priority pesticides and transformation products specified in the EU Baby Food Directive 2003/13/EC has been compared using high-performance liquid chromatography (HPLC) and ultra-performance liquid chromatography (UPLC) coupled to tandem quadrupole mass spectrometry (MS/MS). Prior to analysis, co-extractives were removed from acetonitrile extracts using dispersive solid-phase extraction (SPE) with primary secondary amine (50 mg). Extracts spiked with pesticides at 1 microg kg(-1) yielded average recoveries in the range 85-119%, with relative standard deviations less than 17%. The HPLC-MS/MS and UPLC-MS/MS multi-residue methods developed are simple, rapid and suitable for the quantification and confirmation of the 16 priority pesticides in fruit-, potato- and cereal-based baby food at 1 microg kg(-1). The major advantages of UPLC, using 1.7 microm particles, over HPLC are the speed of analysis, the narrower peaks (giving increased signal-to-noise ratio) and improved confirmation for the targeted pesticides in the analyses of baby foods.  相似文献

A fast, sensitive and selective procedure employing a combination of microwave-assisted extraction (MAE) and solid phase extraction (SPE) was applied prior to liquid chromatographic identification and quantification of phenolic compounds in plant materials. MAE has been tested and optimized for the isolation of phenolic acids (gallic, protocatechuic, p-hydroxybenzoic, chlorogenic, vanilic, caffeic, syringic, p-coumaric, ferulic, sinapic, benzoic, m-coumaric, o-coumaric, rosmarinic, cinnamic acids) and 3,4-dihydroxybenzaldehyde, syringaldehyde, p-hydroxybenzaldehyde, and vanillin in various plants. The effects of experimental conditions on MAE efficiency, such as solvent composition, temperature, extraction time, have been studied. The extraction efficiencies were compared with those obtained by computer-controlled, two-step Soxhlet-like extractions. Plant extracts were purified and phenolic compounds were pre-concentrated using SPE on polymeric RP-105 SPE sorbent prior to HPLC analysis. Chromatographic separation was carried out on a Hypersil BDS C18 column using a mobile phase consisted of 0.3% (v/v) acetic acid in water (solvent A) and methanol (solvent B) at flow rate 0.6 ml min−1 and column temperature 30 °C with gradient elution.  相似文献
