Abstract Phototropism is a common property of plants, but it is not known if different species use the same photoreceptor for their response. We have determined fluence-response relations for phototropism in response to brief, broad-band blue irradiation for four plant species grown under red light (Amaranthus paniculatus, Linum usitatissimum, Vigna radiata and Medicago sativa) and compared these to ones previously obtained for Pisum sativum and Zea mays, grown under similar conditions. Curves for all species showed a bell-shaped dependence on fluence, a characteristic of first positive curvature as originally defined for the Avena coleoptile, and had a similar optimal fluence, near 3 H.mol m?2. We have obtained an action spectrum in the blue and UV spectral regions for first positive phototropism of the hypocotyl of alfalfa grown under red light. Fluence-response curves at wavelengths between 300 and 500 nm were nearly identical in shape and magnitude; whereas below 300 nm, their slopes and maximum curvatures were reduced. The action spectrum showed that activity rose sharply at wavelengths below 500 nm, peaked at 450 nm with shoulders on either side of that peak, and had lesser peaks at 380 and, in the far ultraviolet, at 280 nm. This action spectrum was very similar to ones in the literature (obtained between 350 and 500 nm) for first and second positive phototropism of oat coleoptiles. We conclude that the same photoreceptor mediates phototropism in oat and alfalfa.  相似文献

Abstract— The action spectrum for cell killing by UV radiation in human lens epithelial (HLE) cells is not known. Here we report the action spectrum in the 297–365 nm region in cultured HLE cells with an extended lifespan (HLE B-3 cells) and define their usefulness as a model system for photobiological studies. Cells were irradiated with monochromatic radiation at 297, 302, 313, 325, 334 and 365 nm. Cell survival was determined using a clonogenic assay. Analysis of survival curves showed that radiation at 297 nm was six times more effective in cell killing than 302 nm radiation; 297 nm radiation was more than 260, 590, 1400 and 3000 times as effective in cell killing as 313, 325, 334 and 365 nm radiation, respectively. The action spectrum was similar in shape to that for other human epithelial cell lines and rabbit lens epithelial cells. The effect of UV radiation on crystallin synthesis was also determined at different wavelengths. To determine whether exposure to UV radiation affects the synthesis of β-crystallin, cells were exposed to sublethal fluences of UV radiation at 302 and 313 nm, labeled with [35S]methionine and the newly synthesized βY-crystallin was analyzed by immunoprecipitation and western blotting using an antibody to β-crystallin. The results show a decrease in crystallin synthesis in HLE cells irradiated at 302 and 313 nm at fluences causing low cytotoxicity. The effect of radiation on membrane perturbation was determined by measuring enhancement of synthesis of prostaglandin E2 (PGE2). Synthesis of PGE2 occurs at all UV wavelengths tested in the 297–365 nm region. The slope of the PGE2 response curves was higher than that of cell killing curves in cultured HLE cells. These data show that cultured HLE cells with extended lifespan are a suitable system for investigating photobiological responses of cells to UV radiation.  相似文献

Abstract An action spectrum was obtained for photoreactivation (PR) of morphological abnormality arising from ultraviolet (UV)-irradiation of sea urchin sperm. The wavelength dependence of PR was measured by the restoration of the formation of normal pluteus larvae after the exposure of fertilized eggs to various fluences of monochromatic PR light (313 to 500 nm). The PR action spectrum showed a maximum around 365 nm and a secondary peak somewhere above 400 nm. High PR activity beyond 400 nm wavelengths may reflect an advantageous or adaptational ability to cope with harmful effects of solar UV radiation.  相似文献

Abstract— Photogeotropic equilibrium angles were measured for Phycomyces blakesleeanus wild type firstly by means of dichromatic fluence rate response curves using simultaneous irradiation with near threshold 450 nm reference light (constant at 1.2 × 10?8 W m?2) and variable fluence rates of test light (498–630 nm) from the same side. These curves showed minima for test light fluence rates that were close to the photogeotropic threshold for these wavelengths. Secondly, the time course of this inhibitory effect was studied with both the inductive reference 450 nm light (2 × 10?-7 W m?2) and the test light (606 or 450 nm) given as light pulses of 2 s duration (2 s light/48 s dark periods for 6 h). The dark period between the onset of the inductive reference light and test light pulses was varied between 0 and 48 s. No inhibitory effects were observed for simultaneous pulses; however, inhibitory effects were demonstrated for delay times of 2 s and 20 s for 606 nm as well as 450 nm test light. If the test light pulses were given immediately before the inductive reference light, only 606 nm test light was effective in producing a significant inhibitory effect. The results are discussed with regard to a multichromophoric photoreceptor system and to the wavelength dependence of the effects observed. The data and conclusions favor a photoreceptor system with at least two separate chromophoric absorptions of the blue light receptor type, one acting positively, the other acting inhibitorily, and at least one other photoreceptor of presumably minor influence.  相似文献

Phototactic responses of newly hatched silkworm larvae to monochromatic lights and their mixtures were measured to determine wavelength-dependent properties. Each fluence rate-response curve for a monochromatic light was composed of a log-linearly increasing part, a plateau and a decreasing part. Curves were classified into two groups according to the slopes of the linear parts: responses to UV-blue light (364-482 nm) and responses to green-red light (513-681 nm). Different plateau values were found for both groups. The action spectrum had its maximum at 557 nm and showed another large value at about 364 nm, and its minimum was at 447 nm. When green-orange lights (540-577 nm) were added to 557 nm, the silkworms were attracted more by the mixed lights than by the 557 nm component light only. Mixed lights of UV (364 nm) and blue (447 nm) attracted the silkworms more than either component. The magnitude of the response decreased remarkably when green light was mixed with U V or blue light. We conclude that the silkworm possesses colour vision and responds differently to green-red lights (≥ 513 nm) from UV-blue lights (≲ 482 nm).  相似文献

Abstract— The repair proficient human lymphoblastoid line (TK6) has been employed to construcr an action spectrum for the lethal action of ultraviolet (UV) radiation in the range254–434 nm and to examine possible interactions between longer (334, 365 and 405 nm) and shorter wavelength (254 and 313 nm) radiations. The action spectrum follows a DNA absorption spectrum fairly closely out to 360 nm. As in previously determined lethal action spectra for procaryotic and eucaryotic cell populations, there is a broad shoulder in the334–405 nm region which could reflect the existence of either (a) a non-DNA chromophore or (b) a unique photochemical reaction in the DNA over this region. Pre-treatment with radiation at 334 or 365 nm causes either a slight sensitivity to (low fluences) or protection from (higher fluences) subsequent exposure to radiation at a shorter wavelength (254 or 313 nm). Pre-irradiation at a visible wavelength (405 nm) at all fluence levels employed sensitizes the populations to treatment with 254 or 313 nm radiations. These interactions will influence the lethal outcome of cellular exposure to broad-band radiation sources.  相似文献

To determine the action spectrum for photoinduction of the ultraviolet (UV)-absorbing mycosporine-like amino acid shinorine, specimens of the marine red alga Chondrus crispus were irradiated with monochromatic light of various wavelengths using the Okazaki large spectrograph at the National Institute for Basic Biology, Okazaki, Japan. Fluence response curves were determined for the wavelengths between 280 and 750 nm, by irradiating the algae with monochromatic light for 10 h, followed by 4 h of 25 micromol m(-2) s(-1) photosynthetically active radiation and 10 h darkness. Samples were taken after the second exposure interval. A linear correlation between fluence rate and accumulated shinorine concentration was detected for wavelengths between 350 and 490 nm in the fluence rate range of 20-30 micromol m(-2) s(-1), whereas there was no induction above 490 nm. Below 350 nm a decline in shinorine concentration could be observed at fluence rates above 30 micromol m(-2) s(-1), probably due to an inhibition of photosynthetic activity and a subsequent impairment of shinorine biosynthesis. The constructed action spectrum indicated that the photoreceptors mediating shinorine photoinduction might be an unidentified UV-A-type photoreceptor with absorption peaks at 320, 340 and 400 nm.  相似文献

Abstract— The initial photochemical process leading to photoavoidance by plasmodia of an albino strain of Physarum Plasmodium was studied. Superoxide (O), detected as superoxide dismutase (SOD)-inhibitable electron spin resonance (ESR) signal of a spin trap (tBN), was formed upon irradiation. The amount of O formed increased linearl) with log fluence rate above the threshold. The photoavoidance to radiation at wavelengths between200–800 nm also showed the similar linear relationship in log fluence rate-response curves. Thresholds for photoavoidance and O generation agreed with each other and the action spectra showed peaks at about 260, 370, and 460 nm. Thus, active oxygen generated by photosensitization seems to trigger the UV and blue light photoavoidance.  相似文献

Abstract— DNA-protein crosslinking was induced in cultured human P3 teratocarcinoma cells by irradiation with monochromatic radiation with wavelengths in the range254–434 nm (far-UV, near-UV, and blue light). Wavelength 545 nm green light did not induce these crosslinks, using the method of alkaline elution of the DNA from membrane filters. The action spectrum for the formation of DNA-protein crosslinks revealed two maxima, one in the far-UV spectrum that closely coincided with the relative spectrum of DNA at 254 and 290 nm, and one in the visible light spectrum at 405 nm, which has no counterpart in the DNA spectrum. The primary events for the formation of DNA-protein crosslinks by such long-wavelength radiation probably involve photosensitizers. This dual mechanism for DNA-protein crosslink formation is in strong contrast to the single mechanism for pyrimidine dimer formation in DNA, which apparently has no component in the visible light spectrum.  相似文献

The light-dependent utilization of nitrate by the green alga Monoraphidium braunii, coming from nocturnal dark periods, shows an action spectrum of flavin type with two main bands: one in the blue, peaking at 450 and 480 nm, and the other in the near-UV region with a maximum at 365 nm. Other results indicate that cells growing on nitrate as the only nitrogen source resynthesize nitrate reductase daily, which implies the nocturnal loss of this enzyme. The biosynthesis of nitrate reductase at the beginning of the light periods can proceed under red light. In addition, blue or near-UV light is required for the activation of the previously formed nitrate reductase.  相似文献

Using 290-nm light, which excites only a UV-B photoreceptor, and 385- and 660-nm light, which activate only phytochrome, the fluence rate-response curves of monochromatic irradiations for anthocyanin synthesis in the first internodes of broom sorghum (Sorghum bicolor Moench, cv. Acme Broomcorn) were analyzed. Although the two photoreceptors absorbed light independently, they multiplicatively increased the action of each other. Accordingly, when the fluence rates of both wavelengths were changed together, the resulting slopes of the fluence rate-response curves of double-log plots were steep compared with the slopes obtained with the respective monochromatic irradiations. The slopes of fluence rate-response curves for monochromatic irradiations at 325 to 345 nm were steeper than those at other wavelengths. This difference was shown to be due to the multiplicative actions of both photoreceptors.  相似文献

Abstract. Action spectra were determined for hyphal aggregation in Pleurotus ostreatus at wavelengths between 360 and 600 nm. The action spectrum for a 50% response had two maxima, one at 370 nm in the near-UV and the other a broad peak at 440–450 nm in the blue. Both were approximately of the same magnitude. A minimum was present at 400 nm and wavelengths greater than 530 nm invoked no response. Action spectra for a range of responses, 10–90%, were also determined which showed gradual changes in the peaks in the blue region. It was concluded that hyphal aggregation in P. ostreatus is under the control of a cryptochrome-like photoreceptor system.  相似文献

Two new poly(ortho‐diaminophenylene) derivatives containing fluorene and/or quinoxaline moieties per repeat unit in the main chain were synthesized via Suzuki coupling reaction followed by reduction process. The synthesized polymers were characterized and explored as colorimetric and fluorometric anion‐sensing materials. The polymers in dilute tetrahydrofuran (THF) solution emitted green light (about 530 nm) in their precursor benzothiadiazole forms and blue to green light (477–523 nm) in their reduced forms. The color of polymer solution was dramatically altered upon addition of fluoride anion without noticeable absorption change in UV–vis spectrum. The fluorescence was ratiometrically quenched with a linear relationship between fluorescence intensity and fluoride anion concentration implying static quenching mechanism could be applied judging from the maintenance of constant fluorescence lifetime with variable fluoride anion concentration. © 2007 Wiley Periodicals, Inc. –Quantitative comparison of the decay rate for absorbance photochanges in chromatophores of R. rubrum at the major wavelengths of peaks and troughs (280, 365, 385, 433, 605, 763, 790, 810, 850, 865, 890 nm) reveal no major differences under a variety of sample conditions. In addition, the decay kinetics of EPR phtochange at the two environmental potentials used in this study are identical with the absorbance ph;otochange decay. Decay curves for fresh chromatophores, aged chromatophores, and fresh chromatophores at high ionic strength display a variety of half times and curve shapes. All of these data however, may be fit (within 10 per cent) by the equation x=αe8.2t+βe0.18t by merely varying the values of α and β. This is interpreted as meaning that any single trapping site may exist in one of two major forms each of which decays following a first order or pseudo-first order reaction. Although the change in decay pattern upon aging is not reversible, that due to high ionic strength is. The time dependencies for two light emission phenomena have been measured as well as their response to the oxidation state of molecules at the trapping site. A major component of long-lived luminescence follows pseudo-second order kinetics with a half time of 38 msec. A very long lived luminescence shows an increse with time which is nearly proportional to the decay of absorbance photochanges.  相似文献

Abstract. A photoreceptor system, “mycochrome”, is involved in a blue and near UV reversible photo-reaction which in turn plays an important role in the photocontrol of conidial development in Alternaria tomato, Botrytis cinerea and Helminthosporium oryzae. Conidial development was controlled by alternating exposures to blue and near UV light with the final response being determined by the final light received. When the final light was near UV a conidium developed; when it was blue, conidiation was inhibited and a “sterile” conidiophore was formed. The effects of the two lights were alternatively reversible. Blue and near UV reversible photoreaction was also found in a light-minus-dark difference spectrum of an intracellular particulate fraction isolated from dark-grown mycelia of A. tomato, and two pigments, PB (blue absorbing type) and PNUV (near UV absorbing type), were contained in that photoreaction. PB was tightly bound to the particulate fraction and PNUV was either soluble in the cytoplasm or very loosely bound to the particulate fraction. Blue and near UV reversible photoreaction occurred only under the coexistence of PB and PNUV. That is, a dip in the near UV region and peak absorbance in the blue region was induced by irradiation with near UV light; however, both conditions were partially dissipated with a subsequent irradiation of blue light. These absorbance changes were revers-ibly repeated by alternating doses of blue and near UV light. When molecular oxygen was introduced after exposure to near UV light, peak absorbance in the blue region rapidly disappeared, and these absorbance changes were repeatedly reversible. Similar results were obtained by treatment with sodium hydrosulfite and molecular oxygen. Thus, it was concluded that the mycochrome system suggested in photo-conidiation was consistent with certain active substances showing blue and near UV reversible absorbance changes and caused by photooxidation reduction reaction.  相似文献

Abstract— The induction of single-strand breaks (alkali-labile bonds plus frank breaks) in the DNA of Bacillus subtilis irradiated in vivo by monochromatic UV light at wavelengths from 254 to 434 nm was measured. The spectrum consists of a major far-UV (below 320 nm) component and a minor near-UV shoulder. A mutant deficient in DNA polymerase I accumulates breaks caused by near-UV (above 320 nm) wavelengths faster than the wild-type strain proficient in polymerase I. Measurable breaks in extracted DNA are induced at a higher frequency than those induced in vivo. Anoxia, glycerol, and diazobicyclo (2.2.2.) octane inhibit break formation in extracted DNA. Alkali-labile bonds induced by 365-nm UV radiation are largely (78%) covalent bond chain breaks, the remainder consists of true alkali-labile bonds, probably apurinic and apyrimidinic sites.  相似文献

A simplified procedure was developed to purify the photoactive yellow protein (PYP) from Ecrorhiorhodospira halophila. Specific antibodies were used to follow the distribution of PYP through the separate purification steps. Low temperature absorbance and fluorescence characteristics of this photoactive protein were investigated. The absorbance spectrum of PYP in 67% (vol/vol) glycerol peaked at 449 and 447 nm, at room-and liquid nitrogen temperatures, respectively. It sharpened significantly upon cooling to 77 K and displayed fine-structure on the blue side of its absorbance maximum, with a spacing of 25 nm. At room temperature PYP fluoresced with a quantum yield of approximately 3.5 times 10?-3 an emission maximum of 495 nm. Maximal excitation occurred at 457 nm, 10 nm red-shifted with respect to the absorbance maximum. At -low temperature the excitation maximum remained unaltered but maximal emission shifted significantly to the blue (to 482 nm). The quantum yield of fluorescence increased to 0.07 at this temperature. Illumination of PYP at low temperature with light from the visible part of the spectrum of electromagnetic radiation induced pronounced changes in its absorbance and fluorescence characteristics. At least two new stable intermediates were formed: one highly fluorescent, with an excitation maximum at 430 nm; additionally, a non-fluorescent red-shifted intermediate with an absorbance maximum at 490 nm. The amount formed of these two intermediates depended strongly on the wavelength of actinic illumination. In combination, these data underline the spectroscopic similarities between PYP and the retinal-containing chromoproteins that are present in Halobacterium halobium.  相似文献

The sensitivity of positive phototactic orientation of cells of the ciliated protozoan Ophryoglena flava has been measured for white light, broad-band blue and red light, and narrow-band monochromatic light, using a laboratory-developed computer aided system. The white-light fluence rate-response curve shows that there is no negative phototaxis in the fluence rate range investigated (0-15 W/m2) and no adaptation phenomena; it is very well fitted by a hyperbolic function; the fluence rate curves under broad band blue and red light (full width at half maximum, FWHM= 100 nm) can be fitted by the same model. The saturation level is, within experimental errors, the same for the three curves, indicating that there are no chromaticity effects and that if there is more than one photoreceptor pigment, they act independently of each other. The fluence rate-response curves determined under narrow band monochromatic light (FWHM = 10 nm) can also be fitted by the same model and show, within experimental errors, the same saturation level. An action spectrum for positive phototaxis at 10-nm intervals has been calculated from fluence rate-response curves: it shows three maxima, at 420, 540 and 590 nm. This action spectrum is significantly different from the ones for photomotile responses in Blepharisma japonicum, Stentor coeruleus and Chlamydodon mnemosyne, whereas it resembles the ones of Paramecium bursaria and Fabrea salina.  相似文献

The effect of monochromatic UVB (280–315 nm) irradiation on the photosynthetic activity of the marine green alga Dunaliella salina was investigated by monitoring the rate of oxygen exchange. Samples were irradiated with narrow bands centered at different UV wavelengths (281, 290, 300, 310 and 322 nm). In a first set of experiments the samples were preirradiated keeping constant the irradiance and varying exposure time. By increasing UVB fluence, a wavelength-dependent decrease in the rate of oxygen production was observed. In a second set of experiments oxygen exchange was monitored simultaneously to UVB irradiation. The decline in the rate of oxygen production was sharper at short wavelengths and faster with increasing UVB photon flux density. The photon fluence action spectra derived from the two sets of experiments are in good agreement and indicate a decrease in the UVB-inhibiting effectiveness of two orders of magnitude from about 0.2 (mmol m-2)-1 at 281 nm to about 0.003 (mmol m-2)-1 at 322 nm. The photon flux density action spectrum derived from the second set of experiments indicates a smoother decrease from about 0.2 (pmol s-l m-2)-1 at 281 nm to about 0.05 (pmol s-l m-z)-l at 310 nm.  相似文献
