One-step membrane-based competitive colloidal gold-based immunoassays in immunochromatographic formats for the rapid detection of diethylstilbestrol (DES) were developed. Nitro-cellulose membrane strip was separately coated with goat anti-rabbit IgG (control line) and DES hapten-ovalubumin conjugate (test line). Anti-DES polyclonal antibody labeled with colloidal gold particles was first incubated with DES. A positive reaction as a result of the remaining antibody-gold conjugate combining with antigen coated on the membrane was obvious by visual detection, with detection limits for immunochromatographic of 0.5 microg/kg for detecting DES standard solution, and the limit of detection was 5 microg/kg for detecting the DES spiked in swine pork and liver. The assay time for test was less than 5 min, suitable for rapid testing on-site.  相似文献

A competitive colloidal gold-based immunoassay in lateral-flow format for the rapid detection of medroxyprogesterone acetate (MPA) in biological materials was developed. A nitro-cellulose membrane strip was separately coated with goat anti-rabbit IgG (control line) and MPA hapten-OVA conjugate (test line). Anti-MPA polyclonal antibody labelled with colloidal gold particles was first incubated with MPA. The limit of detection for lateral flow was 5?ng?g?1 for detecting an MPA standard solution, and the limit of detection was 10?ng?mL?1 for detecting the MPA spiked in pig urine and 10?ng?g?1 for spiked in pig liver. The results were confirmed by high-performance liquid chromatography tandem mass spectrometry (HPLC/MS/MS) and indicated that there was a good agreement between both methods (R 2?=?0.976). An immunochromatographic assay (ICA) based on competitive antigen-coated format using colloidal gold as the label was developed for the detection of the organophosphorus insecticide chlorpyrifos. The ICA test strip consisted of a membrane with a detection zone, a sample pad and an absorbent pad. The membrane was separately coated with chlorpyrifos Hapten-OVA conjugate (test line) and anti-mouse IgG (control line). Based on the fact that the competition is between the migrating analyte in the sample and the analyte hapten immobilized on the test strip for the binding sites of the antibody-colloidal gold (Ab-CG) conjugate migrating on the test strip, this study suggests that the relative migration speed between the two migrating substances is a critically important factor for the sensitive detection by competitive ICA. This criterion was utilized for the confirmation of appropriateness of a nitrocellulose (NC) membrane for chlorpyrifos ICA. The detection limit of the ICA for chlorpyrifos standard and chlorpyrifos spiked into agricultural samples were 10 and 50 ng mL(-1), respectively. The assay time for the ICA test was less than 10 min, suitable for rapid on-site testing of chlorpyrifos.  相似文献

An analytical system of immunochromatographic assay based on gold nanoparticles was developed for the detection of 7-aminoclonazepam (7-ACLZ) in human urine. The qualitative assay was based on the competitive immunoassay using anti-7-ACLZ polyclonal antibody (PcAb) and a detector reagent that contains colloidal gold particles coated with anti-7-ACLZ PcAb. Nitrocellulose membrane was separately immobilised with goat anti-rabbit IgG (control line) and 7-ACLZ-OVA conjugate (test line). The sensitivity of the strip was tested for detecting 7-ACLZ spiked in urine and each specimen was independently measured by liquid chromatography tandem mass spectrometry. Good correlation was showed by the recovery results. The limit of detection for the strip test in urine was 100 ng mL?1. The assay can be applied to the rapid detection of 7-ACLZ with the short testing time.  相似文献

Gel-based and membrane-based flow-through immunoassay formats were investigated for rapid ochratoxin A (OTA) detection in red wine. The flow-through set-up consisted of an antibody containing gel or membrane placed at the bottom of a standard solid-phase extraction column (i.e. the flow-through column), combined with a clean-up column. Different clean-up methods were studied for red wine clarification and purification. The optimal method consisted of passing wine, diluted with an aqueous solution containing 1% polyethylene glycol (PEG 6000) and 5% sodium hydrogencarbonate, through strong anion exchange (SAX) silica. An immunoassay for OTA detection in red wine was optimized and a cut-off level at 2 μg L−1 according to EU legislation was achieved with both formats. A more significant colour difference between blank and spiked samples was observed for the gel-based assay making this superior to the membrane-based assay. The proposed rapid gel-based test was compared with a standard immunoaffinity column - high-performance liquid chromatography - fluorescent detection (IAC-HPLC-FLD) method and a good correlation of the results was obtained for naturally contaminated wine samples.  相似文献

A flow injection (FI) system combined with two photochemical processes is developed for the sensitive and rapid determination of carbaryl. It is based on the on-line photo-conversion of carbaryl into methylamine which subsequently reacts with Ru(bpy)33+ generated through the on-line photo-oxidation of Ru(bpy)32+ with peroxydisulphate. The linear concentration range of application was 0.04-4.0 μg ml−1 of carbaryl, with an R.S.D. of 1.2% (for a level of 0.50 μg ml−1) and a detection limit of 0.012 μg ml−1. The sample throughput was 200 injections per hour. The applicability of the method was demonstrated by determining carbaryl in commercial formulations, water, soil, grain and blood serum.  相似文献

The screen-printed four-electrode system was used as the amperometric transducer for determination of phenols and pesticides using immobilised tyrosinase, peroxidase, acetylcholinesterase and butyrylcholinesterase. Acetylthiocholine chloride was chosen as substrate for cholinesterases to measure inhibition by pesticides, hydrogen peroxide served as co-substrate for peroxidase to measure phenols. The compatibility of hydrolases and oxidoreductases working in the same array was studied. The detection of p-cresol, catechol and phenol as well as of pesticides including carbaryl, heptenophos and fenitrothion was carried out in flow-through and steady state arrangements. In addition, the effects of heavy metals (Cu2+, Cd2+, Fe3+), fluoride (NaF), benzene and dimethylsulphoxide on cholinesterase activities were evaluated. It was demonstrated that electrodes modified with hydrolases and oxidoreductases can function in the same array.The achieved R.S.D. values obtained for the flow system were below 4% for the same sensor and less than 10% within a group of five sensors. For the steady state system, R.S.D.s were approximately twice higher. One assay was completed in less than 6 min. The limit of detection for catechol using tyrosinase was equal to 0.35 and 1.7 μM in the flow and steady state systems, respectively. On the contrary, lower limits of detection for pesticides were achieved in the steady state system—carbaryl 26 nM, heptenophos 14 nM and fenitrothion 0.58 μM.  相似文献

A simple and rapid spectrophotometry for the determination of carbaryl based on acid-induced anionic surfactant micelle-mediated extraction (acid-induced cloud-point extraction) coupled to derivatization with 2-naphthylamine-1-sulfonic acid (ANSA) reagent has been demonstrated. Sodium dodecyl sulfate (SDS) and concentrated HCl were used as extractants at room temperature. ANSA derivatization was directly reacted with carbaryl without alkali hydrolysis. The optimum conditions for both acid-induced extraction and derivatization were investigated. The proposed method shows good analytical features with low detection limit (50 µg L− 1) as well as linearity covered a wide range up to 7.0 mg L− 1, good precision with the relative standard deviation of 2.3% (n = 10), and high recoveries in the samples (> 85%). The high risks of carbaryl contamination in the local vegetable samples studied were observed with the level of 2.2 g kg− 1. The results of the proposed method were comparable to those obtained from HPLC (t-test, = 0.05).  相似文献

A quartz crystal microbalance (QCM) immunosensor was developed for the determination of the insecticide carbaryl and 3,5,6-trichloro-2-pyridinol (TCP), the main metabolite of the insecticide chlorpyrifos and of the herbicide triclopyr. The detection was based on a competitive conjugate-immobilized immunoassay format using monoclonal antibodies (MAbs). Hapten conjugates were covalently immobilized, via thioctic acid self-assembled monolayer (SAM), onto the gold electrode sensitive surface of the quartz crystal. This covalent immobilization allowed the reusability of the modified electrode surface for at least one hundred and fifty assays without significant loss of sensitivity. The piezoimmunosensor showed detection limits (analyte concentrations producing 10% inhibition of the maximum signal) of 11 and 7 μg l−1 for carbaryl and TCP, respectively. The sensitivity attained (I50 value) was around 30 μg l−1 for both compounds. Linear working ranges were 15-53 μg l−1 for carbaryl and 13-83 μg l−1 for TCP. Each complete assay cycle took 20 min. The good sensitivity, specificity, and reusability achieved, together with the short response time, allowed the application of this immunosensor to the determination of carbaryl and TCP in fruits and vegetables at European regulatory levels, with high precision and accuracy.  相似文献

Mercury ions (Hg(II)) are considered highly toxic and hazardous element even at low levels. The contamination of Hg(II) is a global problem. To develop selective and sensitive technique for the detection of Hg(II) has attracted considerable attention. In this study, a multi-component chemically reactive detection conjugate for determination of Hg(II) has been synthesized and a competitive format assay was proposed. In the technique, the chemically reactive capture conjugate was coated on the plate. The reactive detection conjugate was then captured by the capture conjugate. TMB solution was added and catalyzed by HRP molecules immobilized on AuNPs. Finally, the developed enzymatic signal was measured at 450 nm. The linear range of the assay was 0.35–350 ppb with a detection limit of 0.1 ppb. The average recoveries of Hg(II) from mineral water, tap water and lake water were 100.03%, 103.13% and 102.03%, respectively. All coefficients of variation (CVs) were less than 10%. The results are closely correlated with those from inductively coupled plasma mass spectrometry (ICP-MS), which indicated that the developed technique is a reliable method for and sensitive detection of Hg(II) in water samples.  相似文献

A one-step immunochromatographic assay (ICA) was developed for the detection of seven kinds of cephems in milk. Polyclonal antibodies (PcAb) with group-specific to cephems were raised in rabbits after immunization with cephalexin-keyhole limpet hemocyanin (KLH) conjugate. The specificity of anti-sera was determined by indirect competitive enzyme-linked immunosorbent assay (icELISA), and the 50% inhibitions (IC50) of cephalexin and cefadroxil were obtained at 1.5 ng mL−1; IC50 of cefatiofur, cefapirin, cefazolin, cefalothin and cefotaxine were 4, 3.7, 3.2, 4.5 and 5 ng mL−1, respectively. The PcAb against cephems were conjugated to colloidal gold particles as the detection reagent for ICA strips to test for cephems. This method achieved semi-quantitative detection of cephems in <5 min, with high sensitivity to cephalexin and cefadroxil (both 0.5 ng mL−1). At the same time, cefatiofur, cefapirin, cefazolin, cefalothin and cefotaxine were detected at <100 ng mL−1 in spiked processed-milk samples. This method was compared with an enzyme-linked immunosorbent assay by testing 40 milk samples, and the positive samples were validated by a high-performance liquid chromatographic method, with an agreement rate of 100% for both comparisons. In conclusion, the method was rapid and accurate for the multi-residue detection of cephems in milk.  相似文献

In this work, a simple, rapid and sensitive sample pretreatment technique, dispersive liquid-liquid microextraction (DLLME) coupled with high performance liquid chromatography-fluorescence detection (HPLC-FLD), has been developed to determine carbamate (carbaryl) and organophosphorus (triazophos) pesticide residues in water and fruit juice samples. Parameters, affecting the DLLME performance such as the kind and volume of extraction and dispersive solvents, extraction time and salt concentration, were studied and optimized. Under the optimum extraction conditions (extraction solvent: tetrachloroethane, 15.0 μL; dispersive solvent: acetonitrile, 1.0 mL; no addition of salt and extraction time below 5 s), the performance of the proposed method was evaluated. The enrichment factors for the carbaryl and triazophos were 87.3 and 275.6, respectively. The linearity was obtained in the concentration range of 0.1-1000 ng mL−1 with correlation coefficients from 0.9991 to 0.9999. The limits of detection (LODs), based on signal-to-noise ratio (S/N) of 3, ranged from 12.3 to 16.0 pg mL−1. The relative standard deviations (RSDs, for 10 ng mL−1 of carbaryl and 20 ng mL−1 of triazophos) varied from 1.38% to 2.74% (n = 6). The environmental water (at the fortified level of 1.0 ng mL−1) and fruit juice samples (at the fortified level of 1.0 and 5.0 ng mL−1) were successfully analyzed by the proposed method, and the relative recoveries of them were in the range of 80.4-114.2%, 89.8-117.9% and 86.3-105.3%, respectively.  相似文献

The electrochemical detection of carbaryl at low potentials, in order to avoid matrix interferences, is an important challenge. This study describes the development, electrochemical characterization and utilization of a glassy carbon (GC) electrode modified with multi-wall carbon nanotubes (MWCNT) plus cobalt phthalocyanine (CoPc) for the quantitative determination of carbaryl in natural waters. The surface morphology was examined by scanning electron microscopy, enhanced sensitivity was observed with respect to bare glassy carbon and electrocatalytic effects reduced the oxidation potential to +0.80 V vs. SCE in acetate buffer solution at pH 4.0. Electrochemical impedance spectroscopy was used to estimate the rate constant of the oxidation process and square-wave voltammetry to investigate the effect of electrolyte pH. Square-wave voltammetry in acetate buffer solution at pH 4.0, allowed the development of a method to determine carbaryl, without any previous step of extraction, clean-up, or derivatization, in the range of 0.33-6.61 μmol L−1, with a detection limit of 5.46 ± 0.02 nmol L−1 (1.09 ± 0.02 μg L−1) in water. Natural water samples spiked with carbaryl and without any purification step were successfully analyzed by the standard addition method using the GC/MWCNT/CoPc film electrode.  相似文献

A direct competitive chemiluminescent immunosensor system that exploits the competition between chloramphenicol (CAP) as an analyte and CAP-horseradish peroxidase conjugate as a tracer for binding to an anti-CAP antibody on a solid support was devised by installing a flow-through cell which was connected to an injector and a peristaltic pump inside a dark box, followed by positioning a photomultiplier tube as light detector in front of it. The anti-CAP antibody was immobilized onto positively charged Biodyne B membrane pieces by a dipping procedure. The operating conditions for the immunosensor were selected with respect to substrate composition (0.25, 13.3 and 0.66 mM for luminol, H2O2 and p-iodophenol, respectively), injection volume of the substrate solution (200 μL) and the concentrations of antibody for immobilization (0.10 mg mL−1) and tracer (0.030 mg mL−1). At these conditions, sensor response according to analyte concentration was well fitted to a linear equation when plotted in semi-logarithmic scale, with the limit of detection for CAP of 10−8 M. By using the immunosensor, CAP measurement in the model samples prepared from five food materials was conducted.  相似文献

A portable surface plasmon resonance (SPR) optical biosensor device is described as a direct immunosensing system to determine organic pollutants in natural water samples. Monitoring of organochlorine (DDT), organophosphorus (chlorpyrifos) and carbamate (carbaryl) compounds within the concentration levels stipulated by the European legislation, can be accomplished using this immunosensor. The lowest limit of detection (LOD) was obtained for DDT, at 20 ng L−1, whilst 50 ng L−1 and 0.9 μg L−1, were achieved for chlorpyrifos and carbaryl, respectively. Matrix effects were evaluated for the carbaryl immunoassay in different water types with detection limits within the range of carbaryl standard curves in distilled water (0.9-1.4 μg L−1). The covalent immobilization of the analyte derivative through an alkanethiol self-assembled monolayer (SAM) allowed the reusability of the sensor surface during more than 250 regeneration cycles. The quality of the regeneration was proved over a 1-month period of continuous working. The analysis time for a complete assay cycle, including regeneration, comprises 24 min. Our portable SPR-sensor system is already a market product, commercialized by the company SENSIA, SL. The size and electronic configuration of the device allow its portability and utilization on real contaminated locations.  相似文献

Two patterns of signal amplification lateral flow immunoassay (LFIA), which used anti-mouse secondary antibody-linked gold nanoparticle (AuNP) for dual AuNP-LFIA were developed. Escherichia coli O157:H7 was selected as the model analyte. In the signal amplification direct LFIA method, anti-mouse secondary antibody-linked AuNP (anti-mouse-Ab-AuNP) was mixed with sample solution in an ELISA well, after which it was added to LFIA, which already contained anti-E. coli O157:H7 monoclonal antibody-AuNP (anti-E. coli O157:H7-mAb-AuNP) dispersed in the conjugate pad. Polyclonal antibody was the test line, and anti-mouse secondary antibody was the control line in nitrocellulose (NC) membrane. In the signal amplification indirect LFIA method, anti-mouse-Ab-AuNP was mixed with sample solution and anti-E. coli O157:H7-mAb-AuNP complex in ELISA well, creating a dual AuNP complex. This complex was added to LFIA, which had a polyclonal antibody as the test line and secondary antibody as the control line in NC membrane. The detection sensitivity of both LFIAs improved 100-fold and reached 1.14 × 103 CFU mL−1. The 28 nm and 45 nm AuNPs were demonstrated to be the optimal dual AuNP pairs. Signal amplification LFIA was perfectly applied to the detection of milk samples with E. coli O157:H7 via naked eye observation.  相似文献

A polyclonal antibody against ochratoxin A (OTA) was produced from rabbits immunized with the OTA–BSA conjugate. A competitive direct enzyme-linked immunosorbent assay (cdELISA) and a membrane-base colloidal gold immunoassay in flow-through format were developed for the rapid detection of OTA in various food matrices. In the cdELISA, the concentration causing 50% inhibition was 0.07 ng mL−1, and the effects of different chemical conditions (ionic strength, pH value, and organic solvent) were studied. The sensitivity of the assay was higher than those previously reported. A simple, rapid, and efficient extraction method was developed and 74–110% recoveries of spiked samples were obtained. Fifty percent methanol extracts of some food samples such as barley, wheat, oat, corn, rice, and raisins could be analyzed directly by immunoassay after dilution in PBS; grape juice and beer samples could be analyzed directly after dilution with PBS; for coffee samples, a more complex method was used to remove the matrix effect effectively. Membrane-based colloidal gold immunoassays had a visual detection limit of 1.0 ng mL−1 for OTA with a detection time of less than 10 min. For the validation of the cdELISA and membrane-based colloidal gold immunoassay, samples were analyzed by high-performance liquid chromatography. The correlation between data obtained using the microwell assay and HPLC was good (R 2 = 0.984). The developed immunoassay methods are suitable for the rapid quantitative or qualitative determination of OTA in food samples.  相似文献

In the present work, enzyme-linked immunosorbent assays (ELISAs) with chemiluminescent detection for the determination of carbofuran, carbaryl and methiocarb were developed and the analytical parameters of these assays were compared with those of ELISAs with colorimetric detection. Both were conjugate-coated formats based on identical monoclonal antibodies and homologous protein conjugates. In comparison with colorimetric ELISA, the ability of the chemiluminescent reagents to detect lower concentrations of horseradish peroxidase allowed to decrease the optimal antibody and conjugate concentrations and to reach better analytical parameters. The experimental comparison of the analytical performance of the ELISAs was carried out by analysing extracts of apple-strawberry baby food and simply diluted fruit juices, both spiked at different concentration levels with the above mentioned pesticides. Recovery values for both ELISAs were around 100% and no matrix effects were observed when fruit juices were diluted 1:20 or more. Results obtained by ELISAs correlated well, both in terms of accuracy and precision, with those obtained by a liquid chromatography-electrospray mass spectrometry (LC/ESI/MS/MS) analysis, used as reference method to validate the immunoassays results. The limits of detection reached by using the chemiluminescent assay were 0.03, 0.007 and 0.004 ng ml−1 for carbofuran, carbaryl and methiocarb, respectively.  相似文献

A pretreatment-free immunochromatographic assay for detection of chloramphenicol (CAP) in milk was developed. The assay is based on competition between CAP molecules in the sample and immobilized CAP-protein conjugate for binding to monoclonal anti-CAP antibodies conjugated with colloidal gold particles (average diameter 30 nm). The assay is carried out in the course of sample flowing along test strip with immobilized reactants, and its results can be detected by the naked eye or by a photometric device. Effect of the concentration of immunoreactants on assay characteristics was studied. The assay protocol with maximal sensitivity and reliability was optimized using measured values of brightness of lines. Detection limit for CAP is 10 ng mL−1. Assay duration is 10 min, and it can be carried out at room temperature without any additional devices and reactants. The developed test strip has been applied to CAP detection in dairy products.  相似文献
