A simple, selective and sensitive heavy atom-induced room temperature phosphorimetric method (HAI-RTP) is described for the determination of naproxen (NAP) in pharmaceutical preparations. The phosphorescence signals are a consequence of intermolecular protection when analytes are, exclusively, in presence of a heavy atom salt and sodium sulfite as an oxygen scavenger to minimize RTP quenching. These variables selection constitute the basis of a HAI-RTP method for the determination of naproxen (detection limit 17.6 ng ml(-1); 1.71% relative standard deviation at 250 ng ml(-1)). The method has been applied satisfactorily to the analysis of pharmaceutical preparations.  相似文献

A new method for the determination of ochratoxin A (OTA) in beer has been developed. The new method has been compared with a reference method currently accepted as AOAC official first action. The limits of detection and quantification of the proposed method were 0.0008 and 0.0025 ng/ml, respectively, while they were 0.0025 and 0.0075 ng/ml, respectively, in the AOAC method used as reference. The recovery levels in the 0.025-0.40 ng OTA/ml spiking range for the proposed and the reference methods were 80.6-87.6% and 78.2-83.8%, respectively. The relative standard deviations of recoveries were 2.6-7.5% for the proposed method and 0.7-6.1% for the reference method. Passing and Bablok regression analysis of recovery data obtained by the proposed method versus data obtained by the reference method on an OTA-spiked beer sample showed good correlation (r2 = 0.9993), while the slope and intercept were 1.049 and -0.0013, respectively. The advantage of the proposed method is the low cost of the materials used in sample preparation because expensive immunoaffinity columns are not needed to clean-up samples while it maintains or even increases the good performance of the reference method. The proposed method was applied to 69 beer samples from different geographic origins (national and imported) but purchased in the Spanish market. They were found to be contaminated with OTA in the range from 0.008 to 0.498 ng/ml (average: 0.070 ng/ml). Five samples surpassed the limit recommended by the European Union (0.2 ng OTA/g).  相似文献

A method for the determination of stale-flavor carbonyl compounds including E-2-octenal, E-2-nonenal, E,Z-2,6-nonadienal and E,E-2,4-decadienal in beer was developed using stir bar sorptive extraction (SBSE) with in-situ derivatization followed by thermal desorption-GC-MS analysis. The derivatization conditions with O-(2,3,4,5,6-pentafluorobenzyl)hydroxylamine and the SBSE conditions--sampling mode, salt addition, sample volume, polydimethylsiloxane volume (sample/polydimethylsiloxane phase ratio) and extraction time--were examined. The method showed good linearity over the concentration range from 0.1 to 10 ng ml(-1) for all analytes and the correlation coefficients were higher than 0.9993. The limits of detection ranged from 0.021 to 0.032 ng ml(-1) for all analytes. The recoveries (98-101%) and precision (RSD 2.4-7.3%) of the method were examined by analyzing beer samples fortified at the 0.5-ng ml(-1) level. The method was successfully applied to low-level concentration samples.  相似文献

A high-performance method of capillary micellar electrokinetic chromatography (CMEKC) with amperometric detection (AD), using a newly designed pre-aligned electrochemical cell, has been developed for the separation and determination of L-tryptophan (Trp) and its eight metabolites including 3-hydroxy-L-kynurenine (3-HK), 5-hydroxy-L-tryptophan (5-HTP), L-kynurenine (KN), 5-hydroxyindole-3-acetic acid (5-HIAA), xanthurenic acid (XA), indole-3-pyruvic acid (IPA), 5-hydroxytryptamine (5-HT), and tryptamine (Tryp). A carbon disk electrode was used as the working electrode and the optimal detection potential was 0.85 V (versus Ag/AgCl). At 24 kV of applied voltage, the nine compounds were completely separated, within 23 min, in a 10 mol/L Na(2)HPO(4)-NaOH buffer (pH 11.0) containing 40 mmol/L sodium dodecyl sulfate (SDS) and 3% methanol (v/v). A good linear relationship was obtained for all analytes in this paper and the detection limits of 3-HK, 5-HTP, KN, Trp, 5-HIAA, XA, IPA, 5-HT, and Tryp were 7.42, 5.18, 34.6, 3.99, 15.1, 12.7, 260, 6.72, and 8.01 nmol/L, respectively. This method has been applied to analyze the metabolism of Trp in rabbit urine.  相似文献

Indole, skatole, indole-3-acetic acid, indole-3-propionic acid, tryptamine, and free tryptophan have been extracted from rat fecal samples and quantitated by thin-layer chromatography. Additional tryptophan metabolites have been recovered in yields of 87-97% from supplemented fecal samples. Detection limits for all the compounds studied were in the 0.1 mug/g feces range. Quantitation was possible in the mug/g range with an estimated accuracy of plus or minus 10%.  相似文献

Two enkephalin-containing peptides, peptide E and dynorphin(1-24), were synthesized by conventional solution methods employing a new Trp derivative, Nin-(2,4,6-triisopropylphenylsulfonyl)tryptophan [Trp(Tps)]. All protecting groups employed including the Tps group were removed by treatment with 1 M trifluoromethanesulfonic acid (TFMSA)-thioanisole in trifluoroacetic acid (TFA) at the final steps of these syntheses. Subsequent purifications by Sephadex G-25 chromatography, CM-Biogel A ion exchange chromatography, and reversed-phase high-performance liquid chromatography afforded highly purified samples. Both synthetic peptide E and dynorphin(1-24) exhibited high in vitro opioid activity. The usefulness of this new tryptophan derivative for practical peptide synthesis was established through these syntheses of complex Trp-containing peptides.  相似文献

A novel micro-injector has been fabricated for capillary electrophoresis (CE). It was successfully used for the determination of some indole derivatives for example melatonin (MT), serotonin (5-HT), tryptophan (Trp), and 5-hydroxy-tryptophane (5-HTrp) in the rat pineal gland by capillary electrophoresis with electrochemical detection (CE-EC). CE was performed in 0.20 mol L(-1) phosphate buffer (pH 8.0). The compounds investigated can be well separated and detected within 15 min. The working electrode used was a 300-microm diameter carbon electrode positioned opposite the outlet of the capillary. The relationship between peak current and analyte concentration was highly linear in the range from 0.10 to 500 micromol L(-1); detection limits (S/N = 3) were 0.03-0.13 micromol L(-1). The proposed method has been successfully used to analyze real biological samples.  相似文献

Two direct HPLC analytical methods for the screening of the major indole alkaloids of Catharanthus roseus hairy roots and their iridoid precursors have been developed. Photodiode array and fluorescence detection were performed. The separation was achieved on a reversed-phase C18 column. The first method allowed the separation of catharanthine, serpentine, tabersonine, vindoline, vinblastine, and vincristine in 20 min. Ajmalicine, tryptophan, tryptamine and secologanine were separated using the second method in 13 min. The identification of the compounds was based on the retention time and the comparison of UV spectra with those of authentic standards. A simplified alkaloid extraction method was developed in order to accelerate sample preparation. The assays were successfully used to quantify major compounds of the secondary metabolism of hairy root cultures of C. roseus, thus providing a reliable tool for rapid screening of C. roseus secondary metabolite samples. In these cultures, ajmalicine, serpentine, catharanthine, tabersonine, and tryptamine were detected, but tryptophan, vindoline, vinblastine and vincristine were not.  相似文献

A new sample treatment for liquid chromatographic analysis of ochratoxin A (OTA) in beer is proposed. Degassed beer is mixed with lead hydroxyacetate, which precipitates some bulk components but does not remove OTA. The precipitate is separated and the acidified liquid is extracted with chloroform. The solvent is evaporated and the residue is dissolved in mobile phase (acetonitrile-water, 40:60, v/v; acidified at pH 3.0 with phosphoric acid) and separated by liquid chromatography using fluorescence detection. The limit of detection was 0.005 ng/ml. The average recovery rate and the average RSD of recovery in the spiking level range 0.01-0.5 ng/ml were 95.5% and about 5%, respectively. The method is cheaper that other alternative ones using immunoaffinity columns or other solid-phase extraction cleanup:The separation was optimised with regard to composition and flow of the mobile phase and no interference from the matrix was found. The method was applied to 88 samples of beer (domestic and imported) marketed in Spain. OTA was detected in 82.9% of them. The range for positive samples was 0.007-0.204 ng of OTA/ml.  相似文献

A fast screening method consisting of off-line solid-phase microextraction coupled to HPLC and fluorescence detection, suitable for the analysis of several bisphenol derivatives and their degradation products in aqueous solution, has been developed. Detection limits of 0.7 ng ml(-1) for 2,2-bis[4-(glycidyloxy)phenyl]propane, 0.9 ng ml(-1) for bisphenol A bis(3-chloro-2-hydroxypropyl)ether, 1.1 ng ml(-1) for 2,2-bis(4-hydroxyphenyl)propane and 2.4 ng ml(-1) for bisphenol F diglycidyl ether have been achieved working in the linear range 10-500 ng ml(-1). The good analytical features achieved make the proposed method an interesting option for the direct determination of these compounds in aqueous canned food such as peas, tuna, olives, maize, artichokes or palm hearts. Both the optimization process and the results, including the analysis of real samples, are given and discussed.  相似文献

Reliable methods for the determination of tryptophan and its metabolites are vital to the monitoring of biochemical states during the initiation and progression of cardiovascular disease. In the present study, a single‐run liquid chromatography–tandem mass spectrometry (LC‐MS/MS) method was developed for the simultaneous determination of tryptophan (Trp) and its metabolites, including kynurenine (Kyn), kynurenic acid (KA), xanthurenic acid (XA) and 5‐hydroxytryptamine (5‐HT), in human plasma. The plasma samples were prepared using a protein precipitation approach, and the chromatographic separation was performed by gradient elution on a C18 column within a total analysis time of 3.5 min. The calibration ranges were 40–20,000 ng/mL for Trp, 4–2000 ng/mL for Kyn, 0.2–100 ng/mL for KA, 0.4–200 ng/mL for XA and 1–500 ng/mL for 5‐HT, and the precision and accuracy were acceptable. The evaluation of recovery and internal standard‐normalized matrix effect proved that the sample preparation approach was effective and the matrix effect could be negligible. The newly developed method was successfully applied to the analysis of plasma samples from healthy individuals and myocardial infarction patients. The findings suggested that the plasma concentrations of Trp, Kyn, 5‐HT as well as the concentration ratios of Kyn/Trp and Trp/5‐HT might serve as biomarkers for the monitoring of acute myocardial infarction.  相似文献

We present results from a comparative study of three proposed phosphorimetric methods for determination of naphazoline (NPZ) in solution. The first method is based on use of micelles to stabilize phosphorescence signals in solutions at room temperature (MS-RTP). The second is based on the use of a heavy atom salt and sodium sulfite as an oxygen scavenger to obtain room-temperature phosphorescence (HAI-RTP) in solution. The last method employs an optical sensor for NPZ based on the phosphorescent properties of the analyte on a solid sensor phase. The aim of this work was to compare time consumption, simplicity, sensitivity, selectivity, detection, and quantification limits for use of these three phosphorimetric methods to determine naphazoline in pharmaceutical preparations. The most simple, sensitive, and reproducible of the three methods for naphazoline analysis is the HAI-RTP method. Detection limits are 4.9, 1.7, and 9.4 ng mL–1, respectively, for the MS-RTP, HAI-RTP, and optosensor methods.  相似文献

Liquid phase room temperature phosphorescence (RTP) properties of melatonin were studied using heavy atom induced-room temperature phosphorescence (HAI-RTP) technique. 1.2 M potassium iodide was used as a heavy atom reagent together with 0.002 M sodium sulphite as deoxygenating agent to produce the RTP signal. The maximum phosphorescence emission and excitation wavelengths of melatonin were 290 and 457 nm, respectively. The effect of potassium iodide concentration on the RTP lifetime of melatonin was also investigated and based on the results, the rate constants for phosphorescence decay (k(p)) and radiationless deactivation through reaction with heavy atom (k(h)) were determined. Based on the obtained results, a simple and sensitive room temperature phosphorimetric method was developed for the determination of melatonin. The method allowed the determination of 10.0-200 ng ml(-1) melatonin in aqueous solution with the limits of detection and quantification of 3.6 and 12 ng ml(-1), respectively. The proposed method was satisfactorily applied to the determination of melatonin in commercial pharmaceutical formulations.  相似文献

A rapid sampling technique for the analysis of beer aroma compounds is described. The headspace (10 ml) is passed through the microcolumn filled with 5 mg of Tenax TA and thermally desorbed in a modified GC inlet (modification is described). Eight compounds (from acetaldehyde to 2-phenylethanol) in four beer samples were analyzed. The correlation coefficients (r2), repeatability (RSD) and limits of detection (LOD) were 0.9973-0.9994, 2.1-6.9% and 0.00002-0.13 mg/l, respectively. The methodology can be useful for routine beer sample analysis.  相似文献

Two biogenic amines, tryptamine and tyramine, and their precursors, tryptophan and tyrosine, were determined by a liquid chromatographic procedure. A hybrid micellar mobile phase of sodium dodecyl sulphate (SDS) and 1-propanol, a C18 column and electrochemical detection were used. A pH study in the range of 3-9 was performed and pH 3 was finally selected in accordance with resolution and analysis time. Oxidation potential was also checked in the range 0.6-0.9V: the maximum area obtained in all those potentials was at 0.8V, which was selected to carry out the analysis using a sequence of pulsed amperometric detection waveform. The four compounds were resolved using a mobile phase of 0.15M SDS-5% 1-propanol with an analysis time of 16 min. Repeatabilities and intermediate precision were evaluated at three different concentrations for each compound with RSD values lower than 2.6 and 4.8%, respectively. Limits of detection and quantification were also obtained within the 10-40 and 33-135 ng/ml ranges, respectively. Finally, the applicability of the procedure was tested in several types of wine and no matrix effect was observed. The possibility of direct sample introduction simplifies and greatly expedites the treatments with reduced cost, improving the accuracy of the procedures.  相似文献

A photochemical method for the determination of lactate using a flow-injection system is proposed. The method is based on the decomposition of lactate in the presence of UO2(2+) and Fe3+ upon irradiation with UV or visible light. The Fe2+ produced in the photochemical process was monitored by measuring the absorbance after complexation with ferrozine (lambda max = 562 nm) or the chemiluminescence (CL) intensity in a luminol system without added oxidant. The range of measurements depended on the length of the irradiation time and the detection system used. The detection limits using CL and photometric detection were 2 ng ml-1 and 50 ng ml-1, respectively. The sample throughput was 45 samples h-1. The usefulness of the method was demonstrated by determining lactate levels in blood serum, milk, yoghurt, beer and pharmaceutical preparations.  相似文献

A capillary zone electrophoresis method has been developed for the quantitation of antihypertensive drug cilazapril and its active metabolite cilazaprilat in pharmaceuticals and urine. The separation of the compounds was performed in a fused-silica capillary filled with the running electrolyte, which consisted of a 60 mM borate buffer solution at pH 9.5. Under the optimized experimental conditions, the separation took less than 5 min. The analysis of urine samples required a previous solid-phase extraction step using C8 cartridges. The method was successfully applied to the determination of the drug and its metabolite in urine samples obtained from three hypertensive patients (detection limits of 115 ng ml(-1) for cilazaprilat and 125 ng ml(-1) for cilazapril) and to pharmaceutical dosage forms. The method was validated in terms of reproducibility, linearity and accuracy.  相似文献

A method has been developed for the simultaneous extraction and determination of five of the most commonly used anticoccidial drugs (nicarbazin, lasalocid, monensin, narasin and salinomycin) from livers and eggs by LC-MS-MS. Results show good repeatability, with mean spiked recoveries for nicarbazin, lasalocid, monensin, narasin and salinomycin in poultry livers in the average range of 92-118%, and 86-110% in eggs. The detection limit is at 1 ng ml(-1) for all the named compounds and a quantitation level of 2.5 ng g(-1) has been achieved. A high throughput of samples is achievable using this method which allows the analysis of up to 40 samples by one analyst in a day.  相似文献

A novel LC-ESI-MS method for the simultaneous determination of four of the most significant estrogenic compounds naturally occurring in beer, 8-prenylnaringenin (8-PN), zearalenone (ZON), alpha-zearalenol (alpha-ZOL) and beta-zearalenol (beta-ZOL) which requires minimal sample preparation was developed using a chemometric approach. Experimental design was applied to assess the effects of the LC-ESI-MS parameters (mobile phase flow rate, drying gas flow, nebuliser pressure and capillary potential) on the obtained signal and to optimize the values in order to provide maximum sensitivity and detectability. The proposed method is simple, consisting only of degassing the beer and diluting with water (1:1, v/v) before injection. Comparison between the two internal standards used, zearalanone (ZAN) and 4,2'-dihydroxychalcone (4,2'-DHC), showed that ZAN performs better as internal standard not only for the mycotoxins but for 8-PN as well, giving lower % RSDs. Under inter-day conditions mean recoveries were 107% for ZON, 87.8% for alpha-ZOL, 72.8% for beta-ZOL, and 77.5% for 8-PN. The corresponding % RSDs ranged between 5.0 and 8.0. The method limits of detection were 1.3, 1.4, 1.0 and 0.8 ng mL(-1) for ZON, alpha-ZOL, beta-ZOL and 8-PN, respectively. The method was applied to 15 beer samples obtained from local supermarkets and the concentration of the phytoestrogen 8-PN in beer ranged between <0.8 and 38.6 ng mL(-1), while neither ZON nor its metabolites, alpha-ZOL and beta-ZOL, were detected.  相似文献
