Early diagnosis of diseases with minimal cost and time-consumption has become achievable due to recent advances in the development of biosensors. These devices use biorecognition elements for the selective interaction with an analyte and the signal read-out is obtained via different types of transducers. The operational characteristics of biosensors have been reported as improving substantially when a diverse range of nanomaterials is employed. This review presents the construction of electrochemical biosensors based on graphene, atomically thin 2D carbon crystals, a nanomaterial currently the subject of intensive studies. Here, the most attractive directions for graphene applications in biosensor preparation are discussed, including novel detection and amplification schemes exploiting graphene’s unique electrochemical, physical and chemical properties. There is probably a very bright future for graphene-based biosensors, but much further work is required to fulfill the high expectations.  相似文献

Genetic analysis of infectious and genetic diseases and cancer diagnostics require the development of efficient tools for fast and reliable analysis of single-nucleotide polymorphism (SNP) in targeted DNA and RNA sequences often responsible for signalling disease onset. Here, we highlight the main trends in the development of electrochemical genosensors for sensitive and selective detection of SNP that are based on hairpin DNA architectures exhibiting better SNP recognition properties compared with linear DNA probes. SNP detection by electrochemical hairpin DNA beacons is discussed, and comparative analysis of the existing SNP sensing strategies based on enzymatic and nanoparticle signal amplification schemes is presented.  相似文献

The unique chemical and physical properties of nanoparticles make them extremely suitable for designing new and improved sensing devices, especially electrochemical sensors and biosensors. Many kinds of nanoparticles, such as metal, oxide and semiconductor nanoparticles have been used for constructing electrochemical sensors and biosensors, and these nanoparticles play different roles in different sensing systems. The important functions provided by nanoparticles include the immobilization of biomolecules, the catalysis of electrochemical reactions, the enhancement of electron transfer between electrode surfaces and proteins, labeling of biomolecules and even acting as reactant. This minireview addresses recent advances in nanoparticle‐based electrochemical sensors and biosensors, and summarizes the main functions of nanoparticles in these sensor systems.  相似文献

This review addresses recent advances in carbon‐nanotubes (CNT) based electrochemical biosensors. The unique chemical and physical properties of CNT have paved the way to new and improved sensing devices, in general, and electrochemical biosensors, in particular. CNT‐based electrochemical transducers offer substantial improvements in the performance of amperometric enzyme electrodes, immunosensors and nucleic‐acid sensing devices. The greatly enhanced electrochemical reactivity of hydrogen peroxide and NADH at CNT‐modified electrodes makes these nanomaterials extremely attractive for numerous oxidase‐ and dehydrogenase‐based amperometric biosensors. Aligned CNT “forests” can act as molecular wires to allow efficient electron transfer between the underlying electrode and the redox centers of enzymes. Bioaffinity devices utilizing enzyme tags can greatly benefit from the enhanced response of the biocatalytic‐reaction product at the CNT transducer and from CNT amplification platforms carrying multiple tags. Common designs of CNT‐based biosensors are discussed, along with practical examples of such devices. The successful realization of CNT‐based biosensors requires proper control of their chemical and physical properties, as well as their functionalization and surface immobilization.  相似文献

This paper presents a theoretical study of electrochemical affinity biosensors for the detection of DNA/protein that utilize nanoparticle labels for signal amplification. This study analyzes the effects of binding and mass transport of the analytes on biosensor performance by using numerical simulations. Four cases were considered: 1) nanoparticles used to increase the loading of an electroactive species, or used as catalysts under pseudo‐first‐order conditions; 2) nanoparticles used as ultramicroelectrode arrays for the electrolysis of large concentrations of substrate; 3) nanoparticles used as seeds to deposit electrochemically detectable species; and 4) nanoparticles used to mediate the deposition of electrocatalysts. By using nanoparticle labels, high sensitivity is possible under all conditions considered. However, theoretical findings suggested that nonspecific adsorption could be more problematic in cases 2–4 due to the mismatch between the chemistry of surface binding and the principle of signal amplification that originates from the effect of mass transport. Under these conditions, any given signal would plateau at a much lower analyte concentration, well before the analyte binding had actually reached a plateau. Views on possible solutions to the above limitations are also presented.  相似文献

Cancer is still one of the leading causes of death in the world. There are over 200 types of cancers currently known according to the National Cancer Institute. However, early diagnosis continues to be an important integral part of cancer treatment even though many advances in therapeutics have been made in the past decade. Quick diagnosis and early prevention are critical for the control of the disease status. Biomarkers are commonly indicative of a particular disease process and the cancer biomarkers are also widely used in oncology to help detecting the presence of various carcinomas. The detection of cancer biomarkers plays an important role in clinical diagnoses and evaluation of treatment for patients. Many immunoassay methods are developed for detection of cancer biomarkers. As the detection devices are normally viewed with high sensitivity, simple preparation and rapid response, electrochemical biosensors are increasingly used for the detection of cancer markers. This review describes the status, the latest research and trends of electrochemical sensors in the quantitation of cancer markers in recent years. In particular, the strategy to improve the sensitivities of the electrochemical biosensors by the aid of enzymatic amplification, nanoparticle amplification, ultilization of magnetic microspheres etc. is described herein. At last, we discuss some special features and limitations associated with the described systems that summarize the application and the development prospects of electrochemical immunoassay technology.  相似文献

The design and development of modern biosensors for sensitive and selective detection of various biomarkers is important in diversified arenas including healthcare, environment, and food industries etc. The requirement of more robust and reliant biosensors lead to the development of various sensing modules. The nanomaterials having specific optical, electrical, and mechanical strength can pave the way towards development of ultrafast, robust, and miniaturized modules for biosensors. It can provide not only the point‐of‐care applicability but also has tremendous commercial as well as industrial justification. In order to improve the performance of the sensor systems, various nanostructure materials have been readily studied and applied for development of novel biosensors. In the last few years, researchers are engaged on harnessing the unique atomic and molecular properties of advance‐engineered materials including carbon nanotubes, graphene nanosheets, metal nanoparticles, metal oxide nanoparticles, and their nano‐conjugates. In view of such recent developments in nanomaterial engineering, the current review has been formulated emphasizing the role of these materials in surface engineering, biomolecule conjugation, and signal amplification for development of various ultrasensitive and robust biosensors having commercial as well as industrial viability. Attention is given on the electrochemical biosensors incorporating various nanomaterials and their conjugates. Importance of nanomaterials in the analytical performance of the various biosensor has also been discussed. To put a perceptive insights on the importance of various nanomaterials, an extended table is incorporated, which includes probe design, analyte, LOD, and dynamic range of various electrochemical biosensors.  相似文献

The emergence of nanotechnology has opened new horizons for electrochemical biosensors. This review highlights new concepts for electrochemical biosensors based on different carbon/inorganic hybrid nanoarchitectures. Particular attention will be given to hybrid nanostructures involving 1‐ or 2‐dimensional carbon nanotubes or graphene along with inorganic nanoparticles (gold, platinum, quantum dot (QD), metal oxide). Latest advances (from 2007 onwards) in electrochemical biosensors based on such hybrids of carbon/inorganic‐nanomaterial heterostructures are discussed and illustrated in connection to enzyme electrodes for blood glucose or immunoassays of cancer markers. Several strategies for using carbon/inorganic nanohybrids in such bioaffinity and biocatalytic sensing are described, including the use of hybrid nanostructures for tagging or modifying electrode transducers, use of inorganic nanomaterials as surface modifiers along with carbon nanomaterial label carriers, and carbon nanostructure‐based electrode transducers along with inorganic amplification tags. The implications of these nanoscale bioconjugated hybrid materials on the development of modern electrochemical biosensors are discussed along with future prospects and challenges.  相似文献

This review summarizes recent advances in electrochemical biosensors based on carbon nanotubes (CNTs) and carbon nanofibers (CNFs) with an emphasis on applications of CNTs. CNTs and CNFs have unique electric, electrocatalytic and mechanical properties, which make them efficient materials for developing electrochemical biosensors.We discuss functionalizing CNTs for biosensors. We review electrochemical biosensors based on CNTs and their various applications (e.g., measurement of small biological molecules and environmental pollutants, detection of DNA, and immunosensing of disease biomarkers). Moreover, we outline the development of electrochemical biosensors based on CNFs and their applications. Finally, we discuss some future applications of CNTs.  相似文献

介绍了一种利用互补核酸杂交富集金胶实现信号扩增的蛋白质生物传感器. 以凝血酶蛋白为研究对象, 利用凝血酶蛋白相对应的两段核酸适配体, 将适配体Ⅰ固定在磁性颗粒上, 用于特异性地捕获蛋白, 将适配体Ⅱ标记金胶作为检测信标. 由凝血酶蛋白和相对应的两段核酸适配体构建三明治结构的凝血酶蛋白生物传感器. 另外, 再通过信标金胶上过剩的核酸适配体链与另一段标记有金胶的互补核酸进一步杂交, 获得金胶的选择性聚集, 实现了信号扩增. 通过信号扩增, 使此传感器的灵敏度大大提高, 对凝血酶蛋白的检测下限可达到4.52×10-15 mol/L. 平行测定浓度为7.47×10-14 mol/L的凝血酶8次, 其RSD为3.0%. 该生物传感器对凝血酶蛋白有很好的特异性, 其它蛋白如溶菌酶和牛血清白蛋白的存在对于检测没有影响.  相似文献

Electrochemical biosensors are a well-known group of tools used widely in a variety of industries. Due to interactions between analytes and a surface, a measurable signal occurs which can then be processed to quantitative and/or qualitative data. The knowledge of surfaces’ electrochemical properties as well as their pKa is of great importance when it comes to the construction of biosensors based on an electrochemical signal. In our work, we focused on determining electrochemical properties as well as pKa of thiol-based SAMs – commonly used linker layers in biosensors. By applying cyclic voltammetry and electrochemical spectroscopy we were able to describe the overall resistive behaviour of SAMs synthesised with both alkanethiols and thiols with carboxyl and amine groups. Coherent use of contact angle measurements let us also describe the overall polarity of the investigated surfaces. The obtained data of pKa values differed from the ones described for bulk solutions of modifying agents, which yields important information for further modification of the surface.  相似文献

Methods based on sandwich-type immunosensors and immunoassays have been developed for detection of multivalent antigens/analytes with more than one eptiope due to the use of two matched antibodies. High-affinity antibodies and appropriate labels are usually employed for the amplification of detectable signal. Recent research has looked to develop innovative and powerful novel nanoparticle labels, controlling and tailoring their properties in a very predictable manner to meet the requirements of specific applications. This articles reviews recent advances, exploiting nanoparticle labels, in the sandwich-type immunosensors and immunoassays. Routine approaches involve noble metal nanoparticles, carbon nanomaterials, semiconductor nanoparticles, metal oxide nanostructures, and hybrid nanostructures. The enormous signal enhancement associated with the use of nanoparticle labels and with the formation of nanoparticle-antibody-antigen assemblies provides the basis for sensitive detection of disease-related proteins or biomolecules. Techniques commonly rely on the use of biofunctionalized nanoparticles, inorganic-biological hybrid nanoparticles, and signal tag-doped nanoparticles. Rather than being exhaustive, this review focuses on selected examples to illustrate novel concepts and promising applications. Approaches described include the biofunctionalized nanoparticles, inorganic-biological hybrid nanoparticles, and signal tage-doped nanoparticles. Further, promising application in electrochemical, mass-sensitive, optical and multianalyte detection are discussed in detail.  相似文献

A novel [Ru(bpy)2(dcbpy)NHS] labeling/aptamer‐based biosensor combined with gold nanoparticle amplification for the determination of lysozyme with an electrochemiluminescence (ECL) method is presented. In this work, an aptamer, an ECL probe, gold nanoparticle amplification, and competition assay are the main protocols employed in ECL detection. With all the protocols used, an original biosensor coupled with an aptamer and [Ru(bpy)2(dcbpy)NHS] has been prepared. Its high selectivity and sensitivity are the main advantages over other traditional [Ru(bpy)3]2+ biosensors. The electrochemical impedance spectroscopy (EIS) and atomic force microscopy (AFM) characterization illustrate that this biosensor is fabricated successfully. Finally, the biosensor was applied to a displacement assay in different concentrations of lysozyme solution, and an ultrasensitive ECL signal was obtained. The ECL intensity decreased proportionally to the lysozyme concentration over the range 1.0×10?13–1.0×10?8 mol L?1 with a detection limit of 1.0×10?13 mol L?1. This strategy for the aptasensor opens a rapid, selective, and sensitive route for the detection of lysozyme and potentially other proteins.  相似文献

We propose a separation/concentration‐signal‐amplification in‐one method based on electrochemical conversion (ECC) of magnetic nanoparticles (MNPs) to develop a facile and sensitive electrochemical biosensor for chloramphenicol (CAP) detection. Briefly, aptamer‐modified magnetic nanoparticles (MNPs‐Apt) was designed to capture CAP in sample, then the MNPs‐Apt composite was conjugated to Au electrode through the DNA hybridization between the unoccupied aptamer and a strand of complementary DNA. The ECC method was applied to transfer MNPs labels to electrochemically active Prussian blue (PB). The anodic and cathodic currents of PB were taken for signal readout. Comparing with conventional methods that require electrochemically active labels and related sophisticated labelling procedures, this method explored and integrated the magnetic and electrochemical properties of MNPs into one system, in turn realized magnetic capturing of CAP and signal generation without any additional conventional labels. Taking advantages of the high abundance of iron content in MNPs and the refreshing effect deriving from ECC process, the method significantly promoted the signal amplification. Therefore, the proposed biosensors exhibited linear detection range from 1 to 1000 ng mL−1 and a limit of detection down to 1 ng mL−1, which was better than or comparable with those of most analogues, as well as satisfactory specificity, storage stability and feasibility for real samples. The developed method may lead to new concept for rapid and facile biosensing in food safety, clinic diagnose/therapy and environmental monitoring fields.  相似文献

The interest in the fabrication of electrochemical biosensors with high sensitivity, selectivity and efficiency is rapidly growing. In recent years, noble metal nanoparticles (NMNPs), with extraordinary conductivity, large surface-to-volume ratio and biocompatibility, have been extensively employed for developing novel electrochemical sensing platforms and improving their performances. Through distinct surface modification strategies (e.g. self-assembly, layer-by-layer, hybridization and sol-gel technology), NMNPs provide well control over the microenvironment of biological molecules retaining their activity, and facilitate the electron transfer between the redox center of biomolecules and electrode surface. Moreover, NMNPs have been involved into biorecognition events (e.g. immunoreactions, DNA hybridization and ligand-receptor interactions) by conjugating with various biomolecules, chemical labels and other nanomaterials, achieving the signal transduction and amplification. The aim of this review is to summarize different strategies for NMNP-based signal amplification, as well as to provide a snapshot of recent advances in the design of electrochemical biosensing platforms, including enzyme/protein sensors focused on their direct electrochemistry on NMNP-modified electrode surface; immunosensors and gene sensors in which NMNPs not only participate into biorecognition, but also act as electroactive tags to enhance the signal output. In addition, NMNP alloy-based multifunctional electrochemical biosensors are briefly introduced in terms of their unique heterostructures and properties.

Heavy-metal pollution has attracted intensive attention from the public because of the severe threats of heavy metals to the ecosystem and human health. Ultralow concentration of heavy metals in aquatic environment leads to the urgent needs of sensitive approaches for heavy-metal detection. Electrochemical DNA biosensors present outstanding superiority in convenience, selectivity, and sensitivity compared with conventional methods. To achieve the ultralow detection limit, efforts have been made to implement signal enhancement strategies to develop electrochemical DNA biosensors with enhanced sensing performance. This review focuses on the recent progress in signal enhancement strategies applied to electrochemical DNA biosensors for heavy-metal-ion detection including nicking enzyme–assisted amplification, the utilization of core–shell nanoparticles, and nanocomposites modification.  相似文献
