Abstract

The newly proposed linear modulated stochastic resonance algorithm (LSRA) was used to amplify and detect the weak chromatographic peaks of thidiazuron. The output chromatographic peak is often distorted when using the traditional stochastic resonance algorithm because of the existence of strong noise. In LSRA, the distortion of the output peak can be corrected by introducing a linear force into the nonlinear system. A two‐step optimization method was proposed to give attention to both the signal‐to‐noise ratio and the peak shape of output signal. The weak chromatographic peaks of thidiazuron can be amplified significantly and the distortion of the output peaks can be corrected using LSRA. The algorithm was used to detect thidiazuron residue in water with solid phase extraction‐high performance liquid chromatography. The limit of detection and limit of quantification were improved to 2.5 ng/l and 10 ng/l, respectively.  相似文献

A sensitive and specific electrochemical immunosensor was developed with α‐fetoprotein (AFP) as the model analyte by using gold nanoparticle label for enzymatic catalytic amplification. A self‐assembled monolayer membrane of mercaptopropionic acid (MPA) was firstly formed on the electrode surface through gold‐sulfur interaction. Monoclonal mouse anti‐human AFP was covalently immobilized to serve as the capture antibody. In the presence of the target human AFP, gold nanoparticles coated with polyclonal rabbit anti‐human AFP were bound to the electrode via the formation of a sandwiched complex. With the introduction of goat anti‐rabbit IgG conjugated with alkaline phosphatase, the dentritical enzyme complex was formed through selective interaction of the secondary antibodies with the colloidal gold‐based primary antibody at the electrode, thus affording the possibility of signal amplification for AFP detection. Current response arising from the oxidation of enzymatic product was significantly amplified by the dentritical enzyme complex. The current signal was proportional to the concentration of AFP from 1.0 ng mL?1 to 500 ng mL?1 with a detection limit of 0.8 ng mL?1. This system could be extended to detect other target molecules with the corresponding antibody pairs.  相似文献

We report on a sensitive electrochemical aptasensor for the detection of human prostate specific antigen (PSA). It is based on the signal amplification of the biotin-avidin system using a sensing platform that is making use of a graphite electrode modified with gold nanoparticles that were covered with graphitized mesoporous carbon nanoparticles (AuNPs@GMCs). The AuNPs@GMCs hybrid was prepared by linking 1,6-hexanedithiol-functionalized GMCs and gold nanoparticles via Au-S groups. Then, streptavidin was immobilized on the electrode modified with the AuNPs@GMCs so to enlarge the amount of biotin-aptamer which led to enhanced detection sensitivity. If an PSA aptamer captures the target PSA on the electrode, the differential pulse voltammetric (DPV) signal of the hexacyanoferrate redox system decreases. Factors affecting the performance of the aptasensor were studied in detail. Under optimal conditions, the DPV signal changes could be used to quantitatively detect PSA in the concentration range from 0.25 to 200?ng?mL?1, with a lowest limit of detection as small as 0.25?ng?mL?1. The aptasensor is highly specific and displays acceptable precision, good stability and repeatability.  相似文献

A simple on-line method for simultaneous determination of some oestrogens including oestriol (E3), norethisterone (NORE), ethynylestradiol (EE2), D-norgestrel (NORG) and bisphenol A (BPA), in environmental liquid samples was developed by coupling in-tube solid phase microextraction (in-tube SPME) to high-performance liquid chromatography with diode array (DAD) and fluorescence (FLD) detectors. Two capillary chromatographic columns (Supel-Q? and Carboxen? 1006 porous layer open tubular) were selected to develop this method. To achieve optimum extraction performance, several parameters were investigated including number of draw/eject cycles and the sample volume for each of the columns. Reproducibility was satisfactory for inter- and intra-day precision, yielding % RSDs of less than 10% and 7.6%, respectively. Limits of detection (LODs) and quantification (LOQs) for the proposed method using a DAD detector were achieved in the ranges of 0.04–0.63?ng?mL?1 and 0.12–1.9?ng?mL?1, depending of the capillary column used. Fluorescence detection improved these parameters for E3, BPA and EE2, obtaining LODs of 0.005–0.03?ng?mL?1 and LOQs of 0.015–0.08?ng?mL?1 using Supel-Q and LODs of 0.01–0.015?ng?mL?1 and LOQs of 0.025–0.04?ng?mL?1 using Carboxen. The proposed method was successfully applied to spiked environmental waters obtaining recoveries greater than 80%.  相似文献

Abstract

A novel, selective, and sensitive magnetic-mimetic enzyme fluorescence immunoassay method for antigen detection has been developed by taking advantage of a magnetic separation process and the amplification feature of the hemin label. This method is based on a twice amplified fluorescence signal. The signal is first amplified due to the ultrasmall size and the high surface-to-volume ratio of the silica-coated magnetite nanoparticles, which enable the nanoparticles to carry much more antibodies. Second, the mimetic enzyme (hemin) as a labeling reagent catalyzes the reaction of p-hydroxyphenyl acetic acid and H2O2 can further amplify the fluorescence signal. This protocol was also evaluated for a sandwich-type immunoassay of human IgG, and the calibration graph for human IgG was linear over the range of 0–100 ng mL?1 with a detection limit of 9.8 ng mL?1. This method can easily separate magnetic nanoparticles from the solution, which simplified the process and played a promising role for various applications in immunoassay.  相似文献

A high-affinity polyclonal antibody was prepared by immunizing animals with haptens FFD and FFM. Under the optimal combination of coating antigen and antibody, an indirect competitive enzyme-linked immunosorbent assay (icELISA) for simultaneous detection of florfenicol and thiamphenicol residues in animal meat and urine samples was developed. The icELISA showed an IC50 value of 1.32 ng mL?1 for florfenicol and 2.13 ng mL?1 for thiamphenicol, respectively. The linear ranges were from 0.31 to 5.61 ng mL?1 with a limit of detection of 0.12 ng mL?1 for florfenicol, and 0.41 to 11.2 ng mL?1 with a limit of detection of 0.15 ng mL?1 for thiamphenicol, respectively. The average recoveries of florfenicol and thiamphenicol in spiked samples ranged from 77.2% to 116.0% with a relative standard deviation of less than 15%. Therefore, this proposed icELISA provided a valid detection method for florfenicol and thiamphenicol residues in animal tissue and urine samples.  相似文献

以负载Au的金属有机骨架材料(AuNPs/Cu-TPA)标记CEA抗体(Ab2)为信号探针,通过电还原的方法将氧化石墨烯还原到电极上,研制了一种捕获CEA抗体(Ab1)的电化学免疫传感器,并将其应用于癌胚抗原(CEA)检测.所合成的MOFs材料中含有大量Cu2+,且电化学信号比较稳定,因此可以通过检测MOFs材料中Cu2+的信号实现对CEA的检测.此信号探针不需要预处理和酸处理,易负载贵金属从而固定抗体,大大简化了检测步骤并缩短了检测时间.此传感器对CEA的检测灵敏度好,操作简便.在最优实验条件下,此传感器的线性范围为0.1~ 80 ng/mL,检出限为0.03 ng/mL,线性相关系数为0.9887,可用于真实样品中CEA的测定.  相似文献

A new, simple, fast and reliable solid-phase extraction method has been developed for separation/preconcentration of trace amounts of Pb(II) using dithizone/sodium dodecyl sulfate-immobilized on alumina-coated magnetite nanoparticles, and its determination by flame atomic absorption spectrometry (FAAS) and graphite furnace atomic absorption spectrometry (GFAAS) after eluting with 4.0?mol?L?1 HNO3. Optimal experimental conditions including pH, sample volume, eluent concentration and volume, and co-existing ions have been studied and established. Under the optimal experimental conditions, the preconcentration factor, detection limit, linear range and relative standard deviation of Pb(II) using FAAS technique were 280 (for 560?mL of sample solution), 0.28?ng?mL?1, 1.4?C70?ng?mL?1 and 4.6% (for 10?ng?mL?1, n?=?10), respectively. These analytical parameters using GFAAS technique were 300 (for 600?mL of sample solution), 0.002?ng?mL?1, 0.006?C13.2?ng?mL?1 and 3.1% (for 5?ng?mL?1, n?=?10), respectively. The presented procedure was successfully applied for determination of Pb(II) content in opium, heroin, lipstick, plants and water samples.  相似文献

Abstract

An indirect competitive chemiluminescent enzyme-linked immunosorbent assay (CL-ELISA) for detection of gatifloxacin residue in milk was developed in this study. Compared with conventional colorimetric ELISA using the same antibody, the developed CL immunoassay shows a significant improvement in sensitivity and detectability with an IC50 of 0.4 ng mL?1 and a detection limit of 0.001 ng mL?1 and thus is suitable to be used as a highly sensitive screening method to detect and regulate illegal use of gatifloxacin in food and food products. The test kit was applied to detect milk samples spiked by gatifloxacin, and satisfactory results were obtained.  相似文献

A rapid and novel method combining dispersive liquid–liquid microextraction and high-performance liquid chromatography coupled with fluorescence detection was developed for the determination of donepezil in human urine. Parameters affecting extraction efficiency and chromatographic determination, such as the type and volume of the extraction and disperser solvent, pH of sample for dispersive liquid–liquid microextraction, mobile-phase composition, pH, column oven temperature, and flow rate for chromatographic determination, were evaluated and optimized. Using a C18 core–shell column (7.5 × 4.6?mm, 2.7?μm), the determination of donepezil was accomplished within 5?min. Under optimum conditions, developed method was linear in the range of 0.5–25?ng?mL?1 with the correlation coefficient >0.99. Limit of detection was 0.15?ng?mL?1. The relative standard deviation at three concentration levels (2, 12.5, and 20?ng?mL?1) was less than 11% with accuracy in the range of 96.9–102.8%. The results of this study demonstrate that the use of dispersive liquid–liquid microextraction and core–shell column can be considered as a powerful tool for the analysis of donepezil in human urine.  相似文献

A competitive colloidal gold-based immunoassay in lateral-flow format for the rapid detection of medroxyprogesterone acetate (MPA) in biological materials was developed. A nitro-cellulose membrane strip was separately coated with goat anti-rabbit IgG (control line) and MPA hapten-OVA conjugate (test line). Anti-MPA polyclonal antibody labelled with colloidal gold particles was first incubated with MPA. The limit of detection for lateral flow was 5?ng?g?1 for detecting an MPA standard solution, and the limit of detection was 10?ng?mL?1 for detecting the MPA spiked in pig urine and 10?ng?g?1 for spiked in pig liver. The results were confirmed by high-performance liquid chromatography tandem mass spectrometry (HPLC/MS/MS) and indicated that there was a good agreement between both methods (R 2?=?0.976). The assay time for the test was less than 5?min, suitable for rapid testing on site.  相似文献

A continuous flow hydride generation laser-induced fluorescence (HG-LIF) spectrometry technique has been investigated for performing trace determination of germanium (Ge). Hydride generation of Ge is performed using reagent concentrations of 2?M H3PO4 and 0.5% NaBH4 and fluorescence detection is performed using tunable dye laser radiation at 253.323?nm for Ge excitation with fluorescence measured at 303.907?nm. The HG-LIF approach provides a linear response for Ge in the concentration range from 1.0 to 50?ng mL?1 and a limit of detection of 0.1?ng mL?1. Replicate measurements at 10?ng mL?1 have a relative standard deviation of 0.1% (n?=?8). Measurements of Ge in different sample matrices have demonstrated the effectiveness of thiourea and ascorbic acid as masking agents that compensate for samples containing interfering ions. The determinations of the Ge content in reference water samples, fly ash samples, and supplement capsules demonstrate that the HG-LIF approach has feasibility for measuring Ge in different sample matrices at environmentally relevant concentrations.  相似文献

Assays were developed for DNA or polymyxin B (PMB) based on enhanced resonance Rayleigh scattering (RRS) and resonance nonlinear scattering (including second order scattering and frequency doubling scattering) that result from the interaction of PMB with DNA. A minor-groove binding mechanism is suggested from the results obtained with RRS and from absorption and circular dichroism spectroscopy. The types of interaction and reasons of RRS enhancement are discussed. Linear relationships do exist over a wide range between the intensity of enhanced scattering and the concentrations of either DNA or PMB. When ctDNA is used as a probe to determine PMB, the detection limit (3σ) is 9.8 ng mL?1. When PMB is used as a probe to determine DNA, the detection limit (3σ) is in the range from 3.8 to 9.0 ng mL?1.

A column, solid phase extraction (SPE), preconcentration method was developed for determination of silver by using alumina coated with 1-((5-nitrofuran-2-yl)methylene)thiosemicarbazide and determination by flame atomic absorption spectrometry. The separation/preconcentration conditions for the quantitative recovery were investigated. At pH 2, the maximum sorption capacity of Ag+ was 7.5?mg?g?1. The linearity was maintained in the concentration range of 0.02–11.0?µg?mL?1 in the final solution or 0.14–1.10?×?104?ng?mL?1 in the original solution for silver. The preconcentration factor of 140 and relative standard deviation of ±1.4% was obtained, under optimum conditions. The limit of detection (LOD) was calculated as 0.112?ng?mL?1, based on 3σbl/m (n?=?8) in the original solutions. The proposed method was successfully applied to the determination trace amounts of silver in the environmental samples such as tea, rice and wheat flour, mint, and real water samples.  相似文献

A sensitive electrochemical immunosensor was developed for detecting fumonisin B1 (FB1) in corn using the single‐walled carbon nanotubes/chitosan. The detection mechanism of immunosensor was based on an indirect competitive binding to a fixed amount of anti‐FB1 between free FB1 and FB1‐bovine serum albumin, which was conjugated on covalently functionalized nanotubes/chitosan laid on the glass carbon electrode. The anti‐rabbit immunoglobulin G secondary antibody labeled with alkaline phosphatase was then bound to the electrode surface through reactisubstrate α‐naphthyl phosphate, which produced electrochemical signal. Under optimized conditions, this method could detect FB1 from 0.01 to 1000 ng mL?1 with a detection limit of 2 pg mL?1. This is well below the detection limit required from European Union legislation, 2–4 mg L?1. Moreover, good recoveries were obtained for the detection of spiked corn samples and actual corn samples. As the method has good sensitivity and recovery for detecting FB1, it is a practical detection method.  相似文献

Abstract

Square-wave voltammetry is a fast technique used for determination of trace amounts of acrylamide. When cobalt(II) ions were added to the acrylamide solution, a catalytic peak at about ?1.35 V vs. Ag/AgCl was observed, which was proportional to acrylamide concentration. The calibration curve showed good linearity in the range of 200–800 ng mL?1 of acrylamide with a regression coefficient of 0.9989. The limit of detection of the method was 3.52 ng mL?1, and the relative standard deviations for concentrations of 300 ng mL?1 and 700 ng mL?1 were 99.8% × 10?2 and 79.7% × 10?2, respectively.  相似文献

In this work, a novel sandwich‐type electrochemical immunosensor with electroactive nickel hexacyanoferrate nanoparticles (NiHCFNPs) as matrix was constructed for α‐fetoprotein (AFP) detection in a signal‐off manner by using FeS2?AuNPs nanocomposite catalyzed insoluble precipitation to significantly inhibit the electrochemical signal. Initially, the NiHCFNPs with excellent electrochemical property was modified on the electrodeposited nano‐Au electrode to obtain a strong initial electrochemical signal. Subsequently, another nano‐Au layer was formed for immobilization of capture antibody (Ab1). In the presence of target AFP, the prepared FeS2?AuNPs‐Ab2 bioconjugate could be specifically recognized and immobilized on electrode through the sandwich‐type immunoreaction. The FeS2 with large specific surface areas were used as scaffolds to load abundant mimicking enzyme AuNPs. With the help of hydrogen peroxide (H2O2), FeS2?AuNPs with peroxidase‐like activity accelerated the 4‐chloro‐1‐naphthol (4‐CN) oxidation with generation of insoluble precipitation on electrode, which would greatly hinder the electron transfer and thus caused the decrease of electrochemical signal for quantitative determination of AFP. This approach achieved a wide dynamic linear range from 0.0001 to 100 ng mL?1 with an ultralow limit detection of 0.028 pg mL?1. Especially, the proposed AFP immunosensor can be applied to detect human serum samples with satisfactory results, indicating a potential application in clinical monitoring of tumor biomarkers.  相似文献

In this research, a mixed immunoassay design for multiple chemical residues detection based on combined reverse competitive enzyme-linked immunosorbent assay (ELISA) procedure was developed. This method integrated two reverse ELISA reactions in one assay by labeling horseradish peroxidase to deoxynivalenol (DON) and orbifloxacin. Within this method, IC50 of the two mAbs for each analyte we produced ranged from 23?~?68 ng?mL?1 for DONs and 4.1?~?49 ng?mL?1 for quinolones (QNs). The limit of detection measured by IC10 was achieved at 0.45–1.3 ng?mL?1 for DONs and 0.59–6.9 ng?mL?1 for QNs, which was lower than the maximum residue levels. Recoveries in negative samples spiked at concentrations of 100, 200, and 500 ng?mL?1 ranged from 91.3 to 102.2 % for DONs and 88.7–98.05 % for QNs with relative standard deviation less than 9.88 and 12.67 %. The results demonstrated that this developed immunoassay was suitable for screening of low molecular weight contaminants.

A high‐sensitivity carcinoembryonic antigen immunosensor was successfully prepared via a one‐step hydrothermal method, wherein nitrogen‐doped graphene oxide (Nr GO) loaded Ag and Co3O4 nanomaterials were synthesized using ammonia as the nitrogen source. Doping nitrogen atoms into the graphene structure forms a new type of N‐type semiconductor with an increased number of graphene layers and more active sites for bonding with chemicals, thereby providing excellent in biocompatibility and good electrical conductivity. The electrical signal of the sensor is further amplified due to the good catalytic effect of Co3O4 and Ag NPs on H2O2. The signal probe requires neither pretreatment nor acid treatment, and can be easy to loaded with metal‐immobilized antibodies, which greatly simplifies the detection step not shorten the detection time. The sensor has good sensitivity to detecting carcinoembryonic antigen (CEA) and can easily operate, and requires mild reaction conditions. Under optimal experimental conditions, the linear range of the sensor is 0.001–200 ng ? mL?1, the detection limit is 0.18 pg ? mL?1, and the linear correlation coefficient is 0.991, which can be used for CEA determination of the actual sample.  相似文献

Magnetic ion-imprinted polymer nanospheres, which have core–shell structures, have been synthesized as an adsorbent for extraction of Pb2+ from real samples prior to its flame atomic absorption spectrometric determination. The prepared adsorbent has been characterized using XRD, VSM, TEM, and FTIR measurements. The optimization results revealed that the adsorbent exhibited high selectivity toward Pb2+ over other cations such as Cu2+ and Zn2+. In addition, the removal efficiency of synthesized adsorbent was considerable (qm?=?171.42?mg g?1), its calibration curve was linear (0.5?850.0?ng mL?1), and detection limit was 0.01?ng mL?1. These results suggested that the prepared nanoadsorbent is an ideal candidate for solid-phase extraction of Pb2+ ions.  相似文献
